摘要:
Zearalenone (ZEA) is a mycotoxin with estrogen-like biological activity, which widely present in feed and raw materials, with strong reproductive system toxicity and a major threat to animal reproduction. Betulinic acid (BA) is a natural plant compound with antioxidant, anti-inflammatory and other pharmacological activities. However, the mechanism of ZEA-induced uterine injury and the protective effect of BA have not been reported. Our results show that ZEA could cause uterine histopathological damage and cellular ultrastructural damage, affecting the secretion of sex hormones, such as estradiol (E2) and progesterone (P4), and increase the mRNA and protein expression of estrogen receptor α (ERα). ZEA could inhibit the activities of catalase (CAT) and superoxide dismutase (SOD), increase the production of malondialdehyde (MDA) and reactive oxygen species (ROS), and cause uterine oxidative stress. Furthermore, ZEA affected the homeostasis of uterine cell proliferation and death by regulating the expression of proliferating cell nuclear antigen (PCNA) and activating the mitochondrial apoptotic pathway. ZEA-induced uterine injury might be related to the activation of p38/ERK MAPK signaling pathway. However, the regulatory effect of ZEA on the uterus was reversed after BA treatment. In conclusion, the uterus is an important target organ attacked by ZEA, and BA showed a good therapeutic effect. © 2022

摘要:
L-glutamate (L-Glu) has gained much attention owing to its contribution to the umami taste and it plays important roles in the central nervous system. Herein, an enzyme-free amperometric biosensor based on a peptide possessing an electroactive ferrocene linker as ferrocene-Gly-Gly-Gly-Gly-Ile- Pro-Val-Tyr-Cys-Gly-Leu-Ile-Gly-Gly-Gly-Gly-Lys-(CH2)4- thioctic acid self- assembled on gold electrode was designed and fabricated for specific determination of L-Glu. The biosensor was characterised via cyclic voltammetry, electrochemical impedance spectroscopy, atomic force microscopy and scanning electron microscopy. The biosensor showed optimum response within 200 s at 0.10 V in phosphate-buffered saline. Moreover, the biosensor exhibited excellent sensitivity and a low detection limit of 1.00 × 10−10 M. The sensitivity at an L-Glu concentration of 1.00 × 10−7 M – 1.00 × 10−3 M was 0.1572 μA/M, and that at an L-Glu concentration of 1.00 × 10−10 M – 1.00 × 10−7 M was 0.0293 μA/M. The peptide-based biosensor had excellent specificity and a wider linear range. The relative standard deviation of the L-Glu concentrations measured by the biosensor in a hundred-fold dilution of mouse serum samples was less than 5.00% compared with the high-performance liquid chromatography results, and the recovery rate of L-Glu was from 93.32% to 105.15%. © 2022 Elsevier B.V.

Endoplasmic reticulum stress (ERS) plays a vital role in the pathogenesis of the alcoholic liver disease (ALD). Betulinic acid (BA) has been reported to be effective in the attenuation of ALD; however, its role in ERS and associated stress-signaling pathways remains elusive. Here, we found that the BA pretreatment significantly reduced the alcohol-induced liver injury by decreasing the activities of serum alanine aminotransferase and aspartate aminotransferase, alleviating fat deposition and rupturing the ER in hepatocytes. Moreover, the protective effect of BA on ALD was associated with the inhibition of reactive oxygen species accumulation and ERS, accompanied by the downregulation of glucose-regulated protein 78 (Grp78), Grp94, phosphorylation-inositol-requiring enzyme 1α (p-IRE1α), and phosphorylation-protein kinase R-like endoplasmic reticulum kinase (p-PERK), activating the transcription factor 6 (ATF6) and C/EBP homologous protein (CHOP). Moreover, the alcohol-induced hepatocyte apoptosis was reduced, along with the downregulation of the mitogen-activated protein kinase pathway, caspase-12, caspase-3, and caspase-7, following BA administration. Additionally, the BA-mediated mitigation of alcohol-induced liver injury and deactivation of the ER pathways were the same with 4-PBA, an inhibitor of ERS, indicating that the protective effect of BA on ALD may be regulated by ERS-associated pathways. Collectively, BA is a potentially desirable agent for the ALD, which may reduce hepatocyte apoptosis by suppressing excessive ERS in the liver. © 2022 The Royal Society of Chemistry.

Klebsiella pneumoniae (K. pneumonia, KpI) is a predominate inducement of bovine mastitis, which is associated with high mortality and milk yield reduction. However, data is lacking on the molecular characteristics of bovine K. pneumoniae, limiting the risk assessment of its transmission through the food chain. Herein, we investigated the prevalence of K. pneumoniae in 6301 clinical mastitis (CM) milk samples from dairy cattle in northern area of China. In total, 183 K. pneumoniae isolates were recovered, with detection rates of 3.0% and 2.8% in 2018 and 2019, respectively. Like human clinical K. pneumoniae, all CM K. pneumoniae isolates belonged to one of three phylogroups: KpI (n = 143), Klebsiella. quasipneumoniae subsp. similipneumoniae (KpII-B) (n = 37), and Klebsiella variicola (KpIII) (n = 3). We detected the extended-spectrum β-lactamase-encoding genes blaSHV-2a, blaCTX-M-14, and blaCTX-M-15, as well as clpC, lpfA, lacI, lacZ, lacY, and the fecABDEIR operon in the KpI isolates, which may contribute to their pathogenicity and host adaptability in cows. The high prevalence of KpI in dairy farms may be problematic, as it showed relatively higher rates of antibiotic resistance and virulence gene carriage than the KpII-B and KpIII isolates. Furthermore, we observed distinct differences in population structure between CM- and human infection-associated KpI isolates, with the genes associated with invasive infection in humans rarely being observed in bovine isolates, indicating that few CM-associated K. pneumoniae isolates pose a threat to human health. Nevertheless, bovine KpII-B isolates shared a high level of nucleotide sequence identity with isolates from human infections and frequently carried the nitrogen-fixation gene nif, suggesting an association between KpII-B isolates from cattle and humans, and plant-derived bacteria. © 2022

Bacillus Thuringiensis (Bt) protein has a strong ability to complex with metal ions, which may increase the transport of metal ions in the soil multi-media system. In this study, the interactions between Cry1Ac protein and metal ions (Zn2+ and Cd2+) were investigated through spectroscopies and molecular docking methods. The spectra results showed that both Zn2+ and Cd2+ quenched the fluorescence intensity of Cry1Ac protein through the static quenching. The binding constants with 4–5 orders of magnitude also indicated the interactions between the ions and the Cry1Ac protein. The thermodynamic analysis showed that hydrogen bonds and van der Waals forces were predominant during the processes. In terms of the Förster non-radiation energy transfer theory, the binding distances between metal ions and Cry1Ac protein were approximately 0.21–0.24 nm, indicating the existence of a non-radiative energy transfer between them. Furthermore, molecular docking revealed that the metal ions participated in ligand binding with the Cry1Ac at the locations Asp569, Thr560, Asn564 and Gln566. The present work provided reasonable models helping us further understand the transport effect of heavy metals in the presence of Cry1Ac. The results could provide mechanistic insights into the nature of metal ions-Cry1Ac interactions and offer important information on the toxicity risk of metal ions-Cry1Ac binding interactions. © 2022 Elsevier Ltd

Zearalenone (ZEA) is an estrogenic toxin produced by Fusarium strains, that is widely present in crops, and endangers the reproductive system of animals. Tannic acid (TA) is a natural polyphenolic substance that is widespread in the roots, stems, and leaves of plants, and has special pharmacological activity. This study was designed to investigate the therapeutic effect of TA on ZEA-induced ovarian damage in mice and to explore the molecular mechanism involved. Ninety healthy Kunming female mice were divided into six equal groups. All the groups but the control group were administered daily with ZEA [10 mg/kg body weight (bw)] orally, for 7 days, to induce damage to the reproductive system. Some groups were also administered with TA (50, 100, and 200 mg/bw) for 7 days. Mice were euthanized 24 h later to allow for collection of serum and ovaries. TA can effectively alleviate the appearance of congestion and redness of the ovary, caused by ZEA, and increase the number of healthy growing follicles. Moreover, the estrogen content and the levels of MDA and ROS in the ovaries can be effectively reduced by TA. It can also reduce the apoptosis of ovarian cells, decreases the protein expression of the estrogen receptor, Fas, Fasl, caspase-3, caspase-8, caspase-9, and Bax, and increases the protein expression of Bcl-2. Our study indicates that TA reduces the strong estrogen and oxidative damage induced by ZEA, and these therapeutic effects may be partially mediated by the death receptor and mitochondrial apoptosis signaling pathway. © 2021

To date, two-dimensional (2D) and three-dimensional (3D) metal organic frameworks (MOFs) have been promising materials for applications in electrocatalysis, separation, and sensing. However, the exploration of a simple method for simultaneous fabrication of 2D/3D MOFs on a surface remains challenging. Herein, a one-step and in situ electrosynthesis strategy for fabrication of 2D Hemin-bridged MOF sheets (Hemin-MOFs) or 2D/3D Zn(II)-MOF hybrid nanocomposites on an electrode is reported. It exhibits varied morphologies at different electrodeposition times and attains a 2D/3D complex morphology by adding 1,3,5-benzenetricarboxylic acid (H3BTC) as an organic ligand. The morphology and size of 2D Hemin-MOFs are important factors that influence their performance. Since Pt nanoparticles (PtNPs) are grown on 2D Hemin-MOF sheets, this composite can serve as the peroxidase mimics and PtNPs can act as an anchor to capture the antibody. Therefore, this hybrid nanosheet-modified electrode is used as an electrochemical sensing platform for ultrasensitive pig immunoglobulin G (IgG) and the surface-protective antigen (Spa) protein ofErysipelothrix rhusiopathiaeimmunodetection. Moreover, this work provides a new avenue for the electrochemical synthesis of 2D/3D MOF hybrid nanocomposites with a high surface area and biomimetic catalysts. © 2021 American Chemical Society

To simultaneously impart excellent biological activity and antibacterial function to titanium-based metal materials, rubidium-doped titanium surfaces were prepared via alkali heat treatment, subsequent hydrothermal treatment and final heat treatment. The alkali heat treatment was employed to fabricate an amorphous sodium titanate hydrogel layer on titanium substrates. Thereafter, rubidium was introduced through the hydrothermal process. After final heat treatment, crystallized rubidium titanate and sodium titanate were obtained on titanium surfaces. The viability of MC3T3-E1 cells was inhibited on rubidium-doped titanium surfaces for short-term (day 1). With prolonged duration, the viability and alkali phosphatase (ALP) activity were comparable for various surfaces with different amounts of rubidium (day 5). With further increased culture duration, the collagen synthesis (day 10) and in vitro mineralization of osteoblasts were found to be significantly enhanced on rubidium-doped titanium surfaces. The Rb-doped Ti surfaces showed antibacterial capacity against Staphylococcus aureus at both 12 and 24 h. The results indicate that doping rubidium into titanium surfaces could simultaneously endow materials with favorable osteogenic and antibacterial capacity. © 2019 IOP Publishing Ltd.

The present paper describes a novel two-dimensional liquid chromatography (2D-LC) system, which is comprised of a first-dimensional ion exchange chromatography (IEX1) column, trap column, and second-dimensional reversed-phase chromatography (RP2) column system. The biological sample is separated by the first-dimensional LC using an IEX column to remove interferences. The analytes are transferred to the trap column after heart-cutting. Then, the analytes are transferred to the second-dimensional LC using an RP2 column for further separation and ultraviolet detection. This 2D-LC system can offer a large injection volume to provide sufficient sensitivity and exhibits a strong capacity for removing interferences. Here, the determination of three monoterpene indole alkaloids (MIAs; gelsemine, koumine, and humantenmine) from Gelsemium in biological matrices (plasma, tissue, and urine) was used this 2D-LC system. After a rapid and easy sample preparation method based on protein precipitation, the sample was injected into the 2D-LC. The method was developed and validated in terms of the selectivity, LOD, LOQ, linearity, precision, accuracy, and stability. The sample preparation time for the three MIAs was 15min. The LOD for these compounds was 10ng/mL, which was lower than the developed HPLC methods. The results showed that this method had good quantitation performance and allowed the determination of gelsemine, koumine, and humantenmine in biological matrices. The method is rapid, exhibits high selectivity, has good sensitivity, and is low-cost, thus making it well-suited for application in the pharmaceutical and toxicological analysis of Gelsemium. 以芘作为外源荧光探针,采用牛血清蛋白作为参照,考察了转基因抗虫蛋白Cry1Ac与4种表面活性剂相互作用的荧光光谱特征.结果表明,鼠李糖脂和Tween 80体系的芘荧光行为有相似的变化规律,可与蛋白质发生稳定的缔合过程.Cry1Ac蛋白的亲水性和分子极性较强,在表面活性剂浓度低时,可导致芘的I1/I3值较高.在阴离子、非离子表面活性剂胶团形成后,2种蛋白质的存在均不改变芘与表面活性剂的结合位点.十六烷基三甲基溴化铵对芘的荧光有一定猝灭作用,且在Cry1Ac蛋白介入下不能形成稳定的胶束.

Three monomers of G. elegans indole alkaloids (gelsemine, koumine and humantenmine) were simultaneously detected in porcine plasma for the first time with the development and validation of a sensitive and reliable LC-ESI-MS/MS method. Using a gradient mobile phase at a constant flow rate of 0.2 mL/min via electrospray ionization (positive ion mode) in a multiple reaction monitoring (MRM) scan, gelsemine, koumine and humantenmine were eluted, separated and detected at an appropriate retention time. The porcine plasma was prepared using protein precipitation with 1% formic acid-acetonitrile: methanol (2:1, v/v). Using matrix-matched calibration curves and weighted least squares linear regression, a good linearity (r<sup>2</sup>>0.99) was achieved with a concentration range of 0.1–200 μg/L for gelsemine, koumine and humantenmine;estimated LOD and LOQ values were 0.10 μg/L and 0.2 μg/L, respectively. The mean of the recoveries was in the range of 82.68–100.35% of porcine plasma at four different levels, and the intra-day and inter-day precision (CV) were lower than 15% with a range of 2.46–8.76% and 2.73–10.83%, respectively. The proposed method has proved to be suitable for accurate, quantitative determination of gelsemine, koumine and humantenmine in porcine plasma.<br/> ©2018

Exploring the diffusion dynamics of a viral capsid proteins (VCP)-functionalized nanocarrier on a living cell membrane could provide much kinetic information for the better understanding of their biological functionality. Gold nanoparticles are an excellent core material of nanocarriers because of the good biocompatibility as well as versatile surface chemistry. However, due to the strong scattering background from subcellular organelles, it is a grand challenge to selectively image an individual nanocarrier on a living cell membrane. In this work, we demonstrated a convenient strategy to effectively screen the scattering background from living cells for single-particle imaging with a polarization-resolved dual-channel imaging module. By taking advantage of the polarization of anisotropic gold nanoparticles (gold nanorods, GNRs), the signals from cell components could be counteracted after subtracting the sequential images one by one, while those transiently rotating GNRs on the cell membrane still exist in the processed image. In contrast to the previously reported methods, this method does not require a complicated optical setup alignment and sophisticated digital image analysis process. According to the single-particle imaging results, the majority of VCP-GNRs were anchoring on the cell membrane with confined diffusion. Interestingly, on further inspection of the diffusion trajectories, the particles displayed anomalous confined diffusion with randomly distributed large walking steps during the whole track. Non-Gaussian step distribution was noted, indicating heterogeneous binding and desorption processes on the cell membrane. As a consequence of the robust background screening capability, this approach would find broad applications for single-particle imaging under a noisy environment, e.g., living cells. © 2017 American Chemical Society.

目的:模拟中国人饮食习惯,以小鼠为实验对象,探讨不同玉米油与猪油1:1调和油(后称为调和油)摄入量对其血脂含量、肝功能以及肝脏抗氧化能力的影响。方法:将24只8周龄雄性昆明小鼠随机平均分为空白组、推荐摄入量组和实际摄入量组,每组分别在基础日粮中添加0%、3.8%和6.5% (质量分数)的调和油,饲喂8周后称小鼠体质量,解剖,取外周血清检测血脂和肝功能指标,取完整肝脏测量肝指数、进行组织形态观测和组织切片镜检,对肝脏进行组织匀浆检测氧化损伤指标。结果:小鼠的肝指数与饲料中所添加的调和油剂量呈正相关,空白组小鼠肝脏碱性磷酸酶、谷丙转氨酶和谷草转氨酶活性均明显高于摄入油脂组,推荐摄入量组的小鼠肝脏总抗氧化能力显著高于空白组和实际摄入量组,空白组的肝脏氧化损伤程度显著高于推荐摄入量组和实际摄入量组。结论:不食用油脂对小鼠肝功能有一定的影响;推荐摄入量的玉米油与猪油1:1调和油能提高小鼠肝脏抗氧化能力,降低氧化损伤。

This study reports an applicable analytical strategy of comprehensive identification and structure characterization of target components from Gelsemium elegans by using high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QqTOF MS) based on the use of accurate mass databases combined with MS/MS spectra. The databases created included accurate masses and elemental compositions of 204 components from Gelsemium and their structural data. The accurate MS and MS/MS spectra were acquired through data-dependent auto MS/MS mode followed by an extraction of the potential compounds from the LC-QqTOF MS raw data of the sample. The same was matched using the databases to search for targeted components in the sample. The structures for detected components were tentatively characterized by manually interpreting the accurate MS/MS spectra for the first time. A total of 57 components have been successfully detected and structurally characterized from the crude extracts of G. elegans, but has failed to differentiate some isomers. This analytical strategy is generic and efficient, avoids isolation and purification procedures, enables a comprehensive structure characterization of target components of Gelsemium and would be widely applicable for complicated mixtures that are derived from Gelsemium preparations. Baclofen was illegally used in veterinary clinical medicine as a growth-promoting agent. To date, few methods have been developed for the monitoring of baclofen in animal tissues. In this study, a sensitive and efficient liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to identify and quantify baclofen in the muscle, liver, kidney, and fat of swine was developed and validated. Baclofen was extracted from tissues with ammonium acetate buffer (pH 5.2) and isolated with isopropyl-ethyl acetate (4:6, v/v). Then, a solid phase extraction using MCX cartridge was used to clean up the extracts. The elution was evaporated to dryness and reconstituted with water/methanol (90:10 v/v). All samples were determined by LC-MS/MS system through positive ionization in a multiple reaction monitoring (MRM) mode. The proposed method was validated by evaluation of specificity, linearity, recovery, accuracy, precision, LOD, and LOQ values according to Commission Decision 2002/657/EC. Estimated limit of quantification for baclofen in the muscle, liver, kidney, and fat of this method was 1.00 μg/kg, respectively. The mean intra- and inter-day assay accuracies fell within a range 88.5–93.9% and 86.2–93.2%, respectively. The mean intra- and inter-day precisions were 1.78 and 4.95% (RSD < 15%), respectively. Blebbistatin is a potent and specific inhibitor of the motor functions of class II myosins, including striated muscle myosin and nonmuscle myosin-2 (NM2). However, the blebbistatin inhibition of NM2c has not been assessed and remains controversial with respect to its efficacy with smooth muscle myosin (SmM), which is highly homologous to NM2. To clarify these issues, we analyzed the effects of blebbistatin on the motor activities of recombinant SmM and three NM2s (NM2a, -2b, and -2c). We found that blebbistatin potently inhibits the actin-activated ATPase activities of SmM and NM2s with following IC50 values: 6.47 μM for SmM, 3.58 μM for NM2a, 2.30 μM for NM2b, and 1.57 μM for NM2c. To identify the blebbistatin-resistant myosin-2 mutant, we performed mutagenesis analysis of the conserved residues in the blebbistatin-binding site of SmM and NM2s. We found that the A456F mutation renders SmM and NM2s resistant to blebbistatin without greatly altering their motor activities or phosphorylation-dependent regulation, making A456F a useful mutant for investigating the cellular function of NM2s. © 2017 American Chemical Society.

Conventional cardiac physiology experiments investigate in vitro beat frequency using cells isolated from adult or neonatal rat hearts. Recently, there is an increased interest in measuring the mechanical properties of cardiomyocytes including their contraction force and viscoelasticity. In this study, we used a quartz crystal microbalance (QCM) to study the viscoelasticity of populations of H9C2 cardiomyoblasts under different stimuli. First, we observed the effects of osmotic pressure and found that H9C2 cardiomyoblasts became stiffer under hypertonic conditions and softer under hypotonic conditions. Then we compared the effects of two inotropic cardiovascular drugs, and found that the positive inotropic drug, isoprenaline (ISO), made H9C2 cells stiffer and the negative inotropic drug, verapamil (VRP), made cells softer. For the dosage ranges tested for ISO (12.5-125 nM) and VRP (12.5-250 nM), we also observed that the higher the ISO concentration, the stiffer were the cells; and the higher the VRP concentration, the softer were the cells. Although H9C2 cardiomyoblasts do not have the beating function, our results showed that they still have the ability to contract and expand. Moreover, they are homogeneous and immortal, making them a potentially suitable cell model for the study of contraction and relaxation characteristics of cardiomyocytes, and for pre-assessment and toxicity testing of cardiovascular drugs where high-throughput and repetitions are required. In addition, we demonstrated that the QCM is a potentially useful tool for assessing the contractility of populations of cardiomyocytes. © 2016 The Royal Society of Chemistry.

目的:观察γ-谷维素对过氧化氢诱导人肝细胞(L02)氧化损伤的增殖和保护作用的影响。方法:建立过氧化氢氧化损伤模型,采用不同浓度的γ-谷维素对L02细胞进行处理,检测L02细胞增殖率、丙二醛和微量谷胱甘肽含量、超氧化物歧化酶和过氧化氢酶活力及活性氧水平的变化。结果:γ-谷维素浓度在0.1~0.4 mmol/L对H_2O_2引起的细胞损伤具有修复作用,同时降低丙二醛含量和活性氧水平,提高超氧化物歧化酶和过氧化氢活性以及谷胱甘肽含量,且呈量效关系。结论:L02细胞经0.1~0.4 mmol/L γ-谷维素预处理后,可以缓解H_2O_2造成的氧化损伤,维持正常的生理形态。

摘要:
Resveratrol (RES), a natural polyphenol that is abundant in grapes, exerts anti-inflammatory, anti-oxidative, and antiviral bioactive effects. Protective effects of RES against pseudorabies virus (PrV)- induced reproductive failure were investigated in a mice model. Injection of PrV partially induced stillbirth and abortion, and caused poor growth of progeny. Treatment with RES attenuated the reproductive failure induced by the virus with recovery of the serum progesterone level. RES improved the growth performance of newborn mice. RES can attenuate the reproductive failure induced by PrV in mice. © 2016, The Korean Society of Food Science and Technology and Springer Science+Business Media Dordrecht.

摘要:
Flunixin is approved for use in veterinary medicine as a nonsteroidal anti-inflammatory agent. There is a need for the control of residues and development of methods to monitor its compliance with legislation. However, few methods have been reported for the analysis of flunixin in the animal liver, kidney, and fat. Therefore, the aim of this work was to develop a relatively rapid sample preparation and sensitive using liquid chromatography–tandem mass (LC-MS/MS) confirmatory method to detect flunixin residues in the liver, kidney, muscle, and fat of swine and chicken. After acid hydrolysis, the sample was extracted with ethyl acetate. The extract was finally evaporated to dryness and reconstituted in a water/methanol mixture, and determination was carried out by LC-MS/MS. Flunixin was detected using positive electrospray ionization in selected reaction monitoring (SRM). Estimated limit of quantification of the method was 0.5, 0.5, 0.25, and 0.05 μg/kg for the liver, kidney, muscle, and fat, respectively. The method was validated in animal tissues in terms of selectivity, linearity, trueness, precision, decision limit (CCα), and detection capability (CCβ). All the trueness values fell within a range between 73.6 and 84.8 %. Precision values for all levels of concentration tested showed excellent relative standard deviation (RSD <15 %). The CCαand CCβvalues have been established for each tissue. A relatively rapid and sensitive LC-MS/MS method for the quantitative determination of flunixin in different animal tissues was developed and validated.
