摘要:
In this study, the solid-state fermentation (SSF) of dark tea was carried out using Bacillus subtilis LK-1, which was isolated from Fu brick tea (FBT). The effects of SSF with B. subtilis on volatile organic compounds (VOCs), non-volatile metabolites, and antioxidant activities of dark tea was investigated. A total of 45 VOCs were identified, primarily consisting of ketones (18), hydrocarbons (8), aldehydes (7), and alcohols (6). Following fermentation, the content of key odor active substances such as linalool, β-ionone, and 3,5-octadiene-2-one significantly increased, resulting in an enhanced floral and fruity aroma of dark tea. Furthermore, new flavor substances like geranyl isovalerate and decanal were produced during SSF, enriching the aroma profile of dark tea. Non-ester catechins demonstrated a drastic increase, while ester catechins remarkably decreased after SSF. Furthermore, SSF led to a slight decrease in the total polyphenols content and antioxidant activity of dark tea. There is a close relationship between VOCs and the main non-volatile metabolites during SSF. Overall, this study highlighted the great impact of SSF with B. subtilis on the metabolites of dark tea and provided valuable insights into the role of bacteria in shaping the metabolite profile of FBT. © 2023 The Authors

Cuticle wax is closely related to fruit quality during storage. In this study, changes in epidermal wax morphology, composition, and genes regulation induced by heat shock (HT), 1-methylcyclopropene (1-MCP) or their combination (HT + 1-MCP) were investigated in jujube fruit during cold storage. HT, 1-MCP, or HT + 1-MCP caused a smoother wax layer and fewer micro-cracks compared to the control (CK) during cold storage. It was confirmed that acids and terpenoids were the main wax components by gas chromatography-mass spectrometry. HT + 1-MCP and 1-MCP treatments could significantly increase (p < 0.05) the wax content at 45 d of cold storage. The transcriptomics results indicated that HT + 1-MCP treatment up-regulated FATB, FATB, FAB2, FAD2 and CYP716A, and maintained the wax content of jujube fruit during cold storage. These results could provide new perspective for regulating the cuticle characteristics to extend the shelf life of jujube fruit. © 2022 Elsevier Ltd

In this study, a double resonator piezoelectric cytometry (DRPC) technology based on quartz crystal microbalance (QCM) was first employed to identify HeLa cell pyroptosis and apoptosis by monitoring cells’ mechanical properties in a real-time and non-invasive manner. AT and BT cut quartz crystals with the same frequency and surface conditions were used concurrently to quantify the cells-exerted surface stress (ΔS). It is the first time that cells-exerted surface stress (ΔS) and cell viscoelasticity have been monitored simultaneously during pyroptosis and apoptosis. The results showed that HeLa pyroptotic cells exerted a tensile stress on quartz crystal along with an increase in the elastic modulus (G′), viscous modulus (G″), and a decrease of the loss tangent (G″/G′), whereas apoptotic cells exerted increasing compressive stress on quartz crystal along with a decrease in G′, G″ and an increase in G″/G′. Furthermore, engineered GSDMD−/−-DEVD- HeLa cells were used to investigate drug-induced disturbance and testify the mechanical responses during the processes of pyroptosis and non-pyroptosis. These findings demonstrated that the DRPC technology can serve as a precise cytomechanical sensor capable of identifying pyroptosis and apoptosis, providing a novel method in cell death detection and paving the road for pyroptosis and apoptosis related drug evaluation and screening. © 2023 by the authors.

A This study aimed to employ three reducing sugar (fructose, galactose and glucose)-lysine mode reaction systems (FLMRS, GaLMRS and GLMRS) in examining the influences of reaction conditions, such as initial pH value, reaction temperature, and time on the Nε-(1-Carboxymethyl)-L-lysine (CML) formation. Results indicated that the initial pH value were positively correlated with the CML content in the system. However, the influence of reaction temperature and time on the CML content in the system displayed an initial increasing trend followed by a decline. A temperature of 100.58/125.24/123.24 ℃ and a heating time of 30.00/24.66/25.38 min maximized the formation of CML on FLMRS/GaLMRS/GLMRS and resulted in a CML production of 2.11/1.39/1.34 mmol/mol lysine of response surface methodology. The results of this study would provide particular theoretical support for studying the formation mechanism and control methods of CML in food processing. DNA is use to precisely control the morphology of prussian blue nanoparticles (PBNPs) via electrostatic interaction‐mediated self‐assembly of inorganic ions and protonated DNA for the first time. The flower‐like PBNPs (PB nanoflowers) mediated by 30‐mer Oligo‐C exhibit excellent photothermal properties, mimetic peroxidase activity, biorecognition ability, photo‐fenton catalytic performance and light scattering, and is expected to be widely used in biosensing, catalysis and imaging. Abstract While shapes and surface properties of nanomaterials are known to play important roles in defining their properties, it remains challenging to fine‐tune the morphologies systematically and predictably. Considering the extraordinary performance, prussian blue nanoparticles (PBNPs) are selected as the proof‐of‐concept nanomaterials. Herein, a DNA‐dependence approach to fine‐control the morphology of PBNPs via electrostatic interaction‐mediated self‐assembly of inorganic ions and protonated DNA is developed. The regulation of different DNA on the morphology of PBNPs is systematically investigated. 30‐mer Oligo‐C or ‐T (C30/T30) mediates formation of flower‐like PBNPs (PB nanoflowers), whereas cubic structure with different sizes is observed in the presence of 10‐mer oligo‐G or 30‐mer Oligo‐A (G10/A30). Detailed mechanism studies indicate that the protonation of nucleobases is the key factor for the morphological evolution. C30‐dependent PB nanoflowers are superior to PB nanocubes in photothermal properties, peroxidase mimetic activity, photo‐Fenton catalytic performance, and light scattering property, which present 1.2‐, 3.78‐, 1.58‐, 1.93‐fold improvement, respectively. Furthermore, PB nanoflowers mediated by the diblock DNA (sDNA; comprising C30 and complementary strands of the target DNA) unexpectedly acquire biorecognition capabilities. This work opens a new avenue for the systematic and predictable synthesis of PB nanoflowers, which broadens the repertoire of PBNPs for catalysis, biosensing, and imaging.

摘要:
Acteoside, an important phenylethanol glycoside, is the main active component in Osmanthus fragrans flower. Our previous study found that acteoside showed high antiaging effect but its absorption rate was low. We speculated acteoside palliated aging-related cognitive impairment before being absorbed, that was intestinal homeostasis underlie the antiaging effect of acteoside. In this study, acteoside was confirmed to palliate cognitive impairment in D-galactose induced aging mice. Acteoside treatment dramatically reduced oxidative stress, alleviated intestinal inflammation, restored intestinal mucosal barrier, rebuilt gut microbiome structure and upregulated gut microbiome metabolites short-chain fatty acids (SCFAs) and amino acids (AAs). Furthermore, antibiotic treatment revealed that the antiaging ability of acteoside was abolished in microbiota depleted mice, which offered direct evidence for the essential role of gut microbiota in the attenuation of cognitive impairment of acteoside. Together, our study indicated that acteoside palliated cognitive impairment by regulating intestinal homeostasisand acteoside intake might be a promising nutritional intervention in prevention of neurodegenerative diseases. © 2023

In order to find out the effect of salt concentration on fermented rape stalks, the physicochemical quality and volatile components was investigated using high performance liquid chromatography (HPLC) and headspace solid phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC–MS). The results showed that there were abundant kinds of free amino acids (FAAs) in all samples, mainly presenting sweet, umami and bitter taste. Through taste activity value (TAV), His, Glu, and Ala contributed significantly to the taste of the sample. 51 volatile components were identified, of which the relative contents of ketones and alcohols were high. By the relative odor activity value (ROAV) analysis, the main components that had a great impact on the flavor were phenylacetaldehyde, β-Ionone, ethyl palmitate and furanone. Adjusting the appropriate salt concentration for fermentation could improve the comprehensive quality of fermented rape stalks and promote the development and utilization of rape products. © 2023 The Author(s)

As a class of bioactive and toxic compounds widely present in foodstuffs, the health effects of dietary exposure to β-carboline heterocyclic amines (HAs) have not been elucidated. Based on our previous research that a typical β-carboline HA (harmane) affects blood glucose metabolism and organ dysfunction, the present study mainly focused on the health effects of dietary exposure to harmane in diabetic Goto-Kakizaki (GK) rats. Twenty-four GK rats were administered daily with harmane (0.1 mg per kg body weight) for eight weeks. A comprehensive evaluation of the health effects of harmane was conducted on serum biochemistry, histopathology, and GC-TOF-MS-based metabolomics. The results showed that harmane exerts non-significant effects on the blood glucose metabolism of GK rats. However, it did cause pathological damage to gastrocnemius nerves and showed adverse effects on brain neurons by significantly activating astrocytes and downregulating brain-derived neurotrophic factor (BDNF), which are potential mechanisms related to the disruption of the normal glutamine-glutamate/γ-aminobutyric acid cycle. Moreover, an increased value of AST and urea, alterations in the amino acid, carbohydrate, purine, pyrimidine, and gut microbiota metabolism as well as the tricarboxylic acid (TCA) cycle could be associated with kidney, liver, and gut dysfunction. Our results suggest that given the role of harmane in nerve injury in GK rats, reducing the production and consumption of β-carboline heterocyclic amines in our daily diets should be considered. © 2023 The Royal Society of Chemistry.

Stable isotope and multi-element analytical techniques with chemometrics were developed to trace the origin authenticity of rice in China market. In the long-term study from 2017 to 2020, a total of 115 batches of rice samples from 8 main producing areas of 7 Asian countries were determined 5 stable isotope ratios and 18 elemental contents. One-way analysis of variance (ANOVA) and various multivariate modeling methods were performed for the origin discrimination. Supervised multivariate modeling including partial least squares discriminant analysis (PLS-DA) and linear discriminant analysis (LDA) can realize more satisfactory identification of 8 rice origins than ANOVA comparison and unsupervised methods, their leave-one-out cross-validation accuracies approach 85.0 % and 90.9 %, respectively. δ2H, δ13C, Ba, Al, Mg, δ34S, Pb and δ18O were screened as the most important variables for rice origin traceability (VIP > 1 or AUC > 0.5). This analytical strategy combining maybe promising to ensure the origin authenticity and combat illegal mislabeling in rice trade. © 2023 Elsevier Ltd

Mulberry leaf (ML) and mulberry leaf extract (MLE) have numerous biological properties, such as regulating sugar and lipid metabolism, reducing blood glucose, and increasing insulin secretion. The aim of this study was to perform a systematic review and meta-analysis of randomized clinical trials to examine the effect of ML/MLE supplementation on glycemic traits in adults, including fasting blood glucose (FBG), glycosylated hemoglobin (HbA1c), and fasting plasma insulin (FPI). Twelve clinical trials (615 participants) fulfilled the eligibility criteria for the present meta-analysis, which included sensitivity analysis and GRADE (grading of recommendations assessment, development, and evaluation) certainty. Based on the heterogeneity between included studies, a random effects model was applied in the meta-analysis, and the results are expressed as WMD (weighted mean differences) with 95% CI (confidence intervals). Meta-analysis showed that ML/MLE supplementation resulted in a significant reduction in FBG by -0.47 mmol L-1, HbA1c by -2.92 mmol mol-1, and FPI by -0.58 µIU mL-1. In addition, subgroup analysis indicated that long-term supplementation of ML/MLE (=8 weeks) was more effective for regulation of the glycemic traits in the non-healthy and baseline FPG >6.1 mmol L-1 subgroups. Glycemic regulation by ML/MLE may be attributed to the phytochemicals they contain, which are mainly 1-deoxynojirimycin, flavonoids, phenolics, and polysaccharides. © 2023 The Royal Society of Chemistry.

摘要:
The capacity of Kaempferia galanga L. (KG) and kaempferol to mitigate the formation of free and bound heterocyclic amines (HAs) and advanced glycation end products (AGEs) in roast beef patties was explored. Electron paramagnetic resonance (EPR) and density functional theory (DFT) were used to reveal the possible mechanisms involved in quenching the free radicals. KG (0.5%, 1.0%, 1.5%) and kaempferol (0.005%, 0.010%, 0.015%) reduced HAs and AGEs in a dose-dependent manner. Alkyl free radical, HOO·, and 1O2 were critical to the formation of HAs, and 1O2 was pivotal to AGEs. They were quenched by KG and kaempferol in a dose-dependent manner. DFT indicated that the 3-OH group of kaempferol was most pivotal and quenched the HOO· mainly via H-atom transfer. The active carbonyl intermediates phenylacetaldehyde, glyoxal, and methylglyoxal can also be reduced by KG and kaempferol in a dose-dependent manner, which may be result from the quenching of free radicals. © 2022 Elsevier Ltd

近年来, 假冒伪劣食品已日益成为广大消费者密切关注的问题, 食品真实性评估是缓解这一问题、 保护公众健康的有力手段。 在仪器设备和样品处理的高要求下, 现代检测技术通常需要大量时间和金钱的成本消耗, 而如今食品掺假手段不断变换, 花样日益翻新, 使得这类检测技术存在一定的局限性。 为促进食品安全质量监管的效率和水平提高, 为监管工作提供有力的科学技术支撑和保障, 需要寻求新型检测技术。 光谱分析技术, 以操作简单、 快速无损的优势近年来被广泛应用, 作为一种间接分析技术, 结合数据统计学中的分类方法建立模型后更能有效进行真假鉴别。 在分类方法中, 由于现实生活中五花八门的掺假类型以及在真假样本数量差异大的情况下, 常用的分类方法效果可能出现偏差。 但单类分类方法(one-class classification)是一种只针对一类实例建模分析, 以特定的置信水平固定目标样本类的边界, 对新样本的类别进行判定的方法, 利用这一特点能有效区分不同于真实样本的数据, 大大减少了检测的工作量, 在食品掺假检测应用领域有一定的发展潜力。 对近年来模式识别中的分类方法——单类分类方法进行了综述。 通过阐述光谱分析结合分类方法用于食品掺假检测的必要性, 比较在同一情形下多类分类方法和单类分类方法的判别率, 简介单类分类方法的特点, 并重点介绍几种常见的单类分类方法如数据驱动的簇类独立软模式(DD-SIMCA)、 单类偏最小二乘(OCPLS)、 单类支持向量机(OCSVM)以及单类随机森林(OCRF), 论述单类分类方法在食品真实性鉴别中的应用, 具体在食用油, 乳制品, 饮料, 保健品, 香辛料及谷物方面进行了阐述, 还分析了当前单类分类方法存在的问题, 最后对该技术的应用前景进行展望, 为食品认证分析提供了一定的理论依据。

Phytochemicals self-stabilized Pickering emulsion is expected to be a highly desirable oral self-delivery system. In this study, ursolic acid (UA), a representative pentacyclic triterpene, was reported for the first time that can be formed self-stabilized Pickering emulsion. That is, it is experimentally and theoretically proved that water-in-oil Pickering emulsion could be stabilized solely by UA. First, the morphology, wettability and interfacial properties of the UA particles were characterized. Then, the influence of variation with UA particle concentration (c) and water fraction (ϕ) on the droplet size and coalescence (and sedimentation) stability of UA stabilized emulsions were also studied. It was found that the formed emulsions performed excellent sediment stability except that under conditions where the UA particle concentration is low (c=1 wt%) or at low water fraction(ϕ=0.1). The viscoelastic properties of the emulsions could be improved with the increasing c or ϕ. Finally, The molecular dynamics simulation further verified that the UA particles are located at the oil-water interface and incompletely wrapped the water droplets to form a stable W/O Pickering emulsion. This work enriches the family of sustainable W/O Pickering stabilizers and provides a new strategy for the construction of a Carrier-Free Delivery System for pentacyclic triterpenes. © 2021 Elsevier B.V.

Dual-mode sensing with a two-signal read-out is conducive to the improvement of detection accuracy. Herein, a fluorescent and scattering dual-mode chemosensor for tetracycline (TC) is proposed based on a carbon dot@cerium-guanosine monophosphate (CD@GMP-Ce) coordination polymer network. The inexpensive CD@GMP-Ce was prepared by exploiting the adaptive inclusion capability of coordination polymers and possessed remarkable fluorescence and strong Rayleigh scattering. The functional CD@GMP-Ce demonstrated fluorescence and scattering, the two optical-signal responses to TC simultaneously. Based on TC-specific fluorescence and scattering decline, the dual-mode detection of TC was established and the probe's detection limits were 43 nM in the fluorescence mode and 77 nM in the scattering mode, respectively. Furthermore, the potential application of the dual-mode sensor was verified by measuring TC in milk and tap-water samples. The study not only provides a new perspective for the development of assay methods for TC but also expands the applications of cerium coordination polymers. © 2022 The Royal Society of Chemistry.

摘要:
The synchronous mitigative effects of Alpinia officinarum Hance (AOH) and curcumin on the generation of methylimidazole and acrylamide in cookies were investigated. Possible mechanisms related to quenching free radicals, reducing lipid oxidation and eliminating carbonyl intermediates were explored by electron paramagnetic resonance (EPR) and HPLC. The total methylimidazole and acrylamide contents raised with an increase in heating temperature and time, and reached a maximum at 200 °C for 11 min. AOH and curcumin reduced methylimidazole and acrylamide simultaneously; the maximum inhibition rates for methylimidazole and acrylamide were 51.55% (0.015% curcumin) and 73.66% (1.5% AOH). Alkyl free radicals and HO· were proved to be the critical free radicals for methylimidazole and acrylamide, AOH and curcumin quenched these radicals in a dose-dependent manner. The lipid oxidation, active carbonyl intermediates glyoxal, methylglyoxal, and acrylaldehyde were also reduced by AOH and curcumin simultaneously, which may be resulted from the quenching of free radicals. © 2022 Elsevier Ltd

Understanding the formation mechanism of the flavor compounds in stinky tofu brine is crucial for controlling the flavor quality of Changsha stinky tofu. Dynamic changes in associated bacteria, enzymes, and differential metabolites in the metabolic pathway of aromatic amino acids in brine were investigated. Results showed that phenol (0.39~89.96 μg/mL), p-cresol (0.19~389.62 μg/mL), indole (1.14~242.97 μg/mL), 3-methylindole (0.14~3.00 μg/mL) were the key flavor substances of brine. The main associated bacteria Clostridiales bacterium SYSU GA17129, Aneurinibacillus aneurinilyticus, and Anaerosalibacter massiliensis were significantly positively correlated with key flavor substances (P < 0.05). The main associated enzymes were transaminase, decarboxylase, and lyase. In summary, phenol and p-cresol were formed by the metabolism of tyrosine and phenylalanine through five reaction chains, and indole and 3-methylindole were formed by the metabolism of tryptophan through one and three reaction chains, respectively. © 2022 Elsevier Ltd

In this study, we report a top-down approach to fabricate starch nanoplatelets (SNPs) based on a deep eutectic solvent (DES) comprised of choline chloride and oxalic acid dihydrate. When subjecting waxy maize starch (WMS) to 2 h of DES treatment, the SNPs of oxalate half-ester were successfully fabricated. The formation mechanism of SNPs was studied by monitoring the changes in nanoplatelet morphology, amylopectin chain distribution, long-range crystallinity, and semi-crystalline lamellar structure of the DES-treated WMS at various treatment times. During the DES treatment, relative crystallinity values of WMS gradually decreased from 28.7 to 25.2%. With increasing DES treatment time from 0 to 1.5 h, the thickness of crystalline lamellae decreased from 6.38 to 5.57 nm, whereas the opposite trend was observed for the thickness of amorphous lamellae. The method developed in this work offers a green and efficient route to prepare non-toxic starch nanomaterials. © 2022 Elsevier Ltd

菊花为菊科植物菊的头状花序,滁菊、贡菊、杭菊和亳菊是常见的几类药用品种菊花。不同品种菊花在外观上具有极大的相似性,非专业人员仅凭肉眼难以对其进行准确鉴别分析。常规仪器分析法检测成本较高,分析时间较长,且需要对样品进行破坏性处理,影响了产品的二次销售。近红外光谱技术作为近年来快速发展起来的一种绿色、简单、快速的新型检测技术,在中药鉴别领域取得了很大的进展。基于便携式近红外光谱仪结合化学计量学方法建立了一种菊花品种无损鉴别方法。利用便携式近红外光谱仪采集了滁菊、贡菊、杭菊和亳菊完整以及粉末状两种物理形态样品的光谱,采用单一以及组合光谱预处理方法消除光谱中存在的干扰,结合不同模式识别方法(主成分分析法、软独立模式分类法和Fisher线性判别分析法)分别构建了不同品种菊花的鉴别模型。结果表明:由于仪器的限制及样品物理性状的原因,光谱中存在较为明显的背景、基线漂移以及噪声的干扰,完整样品由于物理性状的原因,基线漂移干扰尤为严重;采用主成分分析法结合光谱预处理方法无法实现不同品种菊花的准确鉴别,完整样品最佳鉴别正确率仅为8.33%,粉末样品最佳鉴别正确率为52.38%;通过软独立模式分类法结合预处理方法可以得到较为准确的鉴别结果,完整样品光谱数据经一阶导数+多元散射校正优化后鉴别正确率为95%,粉末状样品数据采用原始数据的鉴别正确率为92.5%;Fisher线性判别分析方法结果最佳,完整样品数据经连续小波变换优化后可以得到97.5%的鉴别正确率,粉末状样品采用原始光谱便可得到100%鉴别正确率。以上结果表明,当采用合适的预处理和建模方法,完整样品和粉末状样品鉴别结果较为一致,基于便携式近红外光谱仪结合化学计量学可实现对不同品种菊花的准确无损鉴别分析,为食药同源产品的无损鉴别分析提供了新途径。

Endoplasmic reticulum stress (ERS) plays a vital role in the pathogenesis of the alcoholic liver disease (ALD). Betulinic acid (BA) has been reported to be effective in the attenuation of ALD; however, its role in ERS and associated stress-signaling pathways remains elusive. Here, we found that the BA pretreatment significantly reduced the alcohol-induced liver injury by decreasing the activities of serum alanine aminotransferase and aspartate aminotransferase, alleviating fat deposition and rupturing the ER in hepatocytes. Moreover, the protective effect of BA on ALD was associated with the inhibition of reactive oxygen species accumulation and ERS, accompanied by the downregulation of glucose-regulated protein 78 (Grp78), Grp94, phosphorylation-inositol-requiring enzyme 1α (p-IRE1α), and phosphorylation-protein kinase R-like endoplasmic reticulum kinase (p-PERK), activating the transcription factor 6 (ATF6) and C/EBP homologous protein (CHOP). Moreover, the alcohol-induced hepatocyte apoptosis was reduced, along with the downregulation of the mitogen-activated protein kinase pathway, caspase-12, caspase-3, and caspase-7, following BA administration. Additionally, the BA-mediated mitigation of alcohol-induced liver injury and deactivation of the ER pathways were the same with 4-PBA, an inhibitor of ERS, indicating that the protective effect of BA on ALD may be regulated by ERS-associated pathways. Collectively, BA is a potentially desirable agent for the ALD, which may reduce hepatocyte apoptosis by suppressing excessive ERS in the liver. © 2022 The Royal Society of Chemistry.

This study aimed to investigate the absorption and transport of myofibrillar protein-bound Nɛ-(carboxymethyl)lysine (MP-bound CML) in Caco-2 cells after simulated gastrointestinal digestion. Four kinds of MP-bound CML hydrolysates with molecular weights (MWs) less than 1 kDa, 1–3 kDa, 3–5 kDa and greater than 5 kDa, were obtained by ultrafiltration; their absorption and transport were studied in Caco-2 cells. Peptide-bound CML in hydrolysates with MWs less than 1 kDa was absorbed by 6.58 % and might transport across Caco-2 cells monolayer through paracellular pathway; peptide-bound CML in hydrolysates with MWs 1–3 kDa was absorbed by 12.8 % and might transport across Caco-2 cells monolayer through paracellular pathway and transcytosis route; peptide-bound CML in hydrolysates with MWs 3–5 kDa was absorbed by 14.66 % and might be through active route via PepT-1 transport across Caco-2 cells monolayer; whereas protein-bound CML in hydrolysates with MWs greater than 5 kDa was only absorbed by 1.02 %, which was hardly transported into Caco-2 cells. In conclusion, MP-bound CML could be absorbed by 35.06 % into Caco-2 cells after simulated gastrointestinal digestion and is transported across Caco-2 cells through paracellular pathway, transcytosis route and active route via PepT-1. © 2022 Elsevier Ltd
