Dietary resveratrol supplementation inhibits heat stress-induced high-activated innate immunity and inflammatory response in spleen of yellow-feather broilers.

摘要:
The aim of this study was to investigate the effect of dietary resveratrol supplementation on innate immunity and inflammatory responses in the spleen of yellow-feather broilers under heat stress. A total of 288 yellow-feather broilers of 28-day-old were randomly assigned to 3 treatment groups with 6 replicates. A thermo-neutral group (TN) (24 +/- 2 degrees C) received a basal diet and another 2 heat-stressed groups (37 +/- 2 degrees C for 8 h/D and 24 +/- 2 degrees C for the remaining time) were fed the basal diet (HT) or basal diet with 500 mg/kg resveratrol (HT+Res) for 14 consecutive days. The results showed that heat stress decreased (P < 0.05) the growth index of thymus, spleen, and bursa of Fabricius, reduced (P < 0.05) the levels of complement C3 and C4 in serum. Heat stress also caused activation of inflammatory immune responses evidenced by increased (P < 0.05) the mRNA abundance of HSP (heat shock protein) 70, toll-like receptor (TLR)1, TLR4, TLR5, myeloid differentiation factor-88 (MyD88), nucleotide-binding oligomerization domain 1 (NOD1), Dectin-1, transforming growth factor-beta-activated kinase 1 (TAK1), interleukin (IL)-1, IL-4, IL-6, and tumor necrosis factor (TNF)-alpha, but decreased the mRNA abundance of interferon (IFN)-gamma, activated nuclear factor kappa B (NF-kappaB), mitogen-activated protein kinases (MAPK), and phosphoinositide-3 kinases-protein kinase B (PI3K/AKT) signaling pathways. Dietary supplementation with resveratrol improved (P < 0.05) the growth index of thymus, spleen and bursa Fabricius, and increased (P < 0.05) the serum level of complement C3 under heat stress. In addition, resveratrol reduced (P < 0.05) the mRNA abundance of HSP70, TLR4, TLR5, NOD1, Dectin-1, and TAK1, and inhibited the NF-kappaB, MAPK and PI3K/AKT signaling pathway via down-regulated the phosphorylation of p65, extracellular signal-regulated kinases 1/2, c-Jun N-terminal protein kinase and AKT, as well as decreased the inflammatory cytokines expression, including IL-1, IL-4, IL-6, and TNF-alpha in the spleen under heat stress. Collectively, dietary resveratrol could have beneficial effects to regulate innate immunity and inflammatory response, via inhibiting the activation of NF-kappaB, MAPK, and PI3K/AKT signaling pathways induced by heat stress in the spleen.

The intestinal microbiome plays a crucial role in promoting intestinal health, and perturbations to its constitution may result in chronic intestinal inflammation and lead to colorectal cancer (CRC). α-Ketoglutarate is an important intermediary in the NF-B-mediated inflammatory pathway that maintains intestinal homeostasis and prevents initiation of intestinal inflammation, a known precursor to carcinoma development. The objective of this study was to assess the potential protective effects of α-ketoglutarate intervention against CRC development, which may arise due to its known anti-inflammatory and antitumour effects. CRC was induced in C57BL/6 mice using azoxymethane (AOM) and dextran sulfate sodium (DSS). Tumour frequency, histological rating, and colonic microbiota were assessed in colonic samples. The findings demonstrated that α-ketoglutarate offered significant protection against CRC development in mice. Furthermore, α-ketoglutarate also exhibited immunomodulatory effects mediated via downregulation of interleukin (IL)-6, IL-22, tumour necrosis factor (TNF)-α, and IL-1β cytokines. Finally, intervention with α-ketoglutarate tended to minimise the frequency of opportunistic pathogens (Escherichia and Enterococcus) while increasing the populations of Akkermansia, Butyricicoccus, Clostridium, and Ruminococcus. Taken together, our findings show that dietary α-ketoglutarate intervention may protect against inflammation-related CRC. © 2019 Si Li et al.

植物多糖对断奶仔猪小肠黏膜形态及肠黏膜屏障功能的影响

摘要:
旨在探讨断奶仔猪饲粮中单独或联合添加黄芪多糖、牛膝多糖及白术多糖对其小肠黏膜形态及肠黏膜屏障功能的影响。选取256头35日龄的杜长大三元杂交断奶仔猪,随机分为8个组,每组4个重复,每个重复8头猪,采用三因素两水平(2×2×2)试验设计,饲粮中黄芪多糖、白术多糖和牛膝多糖的添加水平均为0,800mg/kg,试验共进行28d。结果显示:植物多糖能显著提高断奶仔猪十二指肠、空肠和回肠的黏膜绒毛高度,提高绒腺比(P<0.05),3种多糖联用效果最适。与对照组相比,多糖组断奶仔猪回肠V/C比值显著提高(P<0.05),各单一多糖组之间差异不显著(P>0.05),3种多糖联合添加与两两添加及单一添加相比显著提高了V/C比值(P<0.05)。多糖组断奶仔猪血清中二胺氧化酶(DAO)的活性、血清中内毒素(ET)含量及D-乳糖的含量均显著降低(P<0.05),3种多糖联用效果最适。多糖能显著提高断奶仔猪肠道黏膜Occludin、Claudin和ZO-1的基因表达量(P<0.05)。单一添加黄芪多糖、白术多糖及牛膝多糖能显著提高断奶仔猪肠道钠-葡萄糖共转运载体1(SGLT1)的mRNA相对表达量(P<0.05),对葡萄糖转运载体2(GLUT2)mRNA相对表达量有提高的趋势,但差异不显著(P>0.05)。多糖两两联用及3种多糖联合添加能显著提高断奶仔猪肠道SGLT1和GLUT2mRNA相对表达量(P<0.05)。结果表明,在断奶仔猪饲粮中添加植物多糖可显著改善断奶仔猪肠道黏膜形态结构和肠道的通透性,其机制可能与植物多糖改善断奶仔猪肠黏膜屏障功能作用有关。

血根碱和牛膝多糖对黄羽肉鸡生长性能及免疫性能的影响

摘要:
本试验旨在研究血根碱和牛膝多糖单独或联合添加对黄羽肉鸡生长性能及免疫性能的影响.试验选用1日龄的黄羽肉鸡300只, 随机分成5组, 每组6个重复, 每个重复10只.对照组饲喂基础饲粮, 血根碱组在基础饲粮中添加0.75 mg/kg血根碱, 牛膝多糖组在基础饲粮中添加400 mg/kg牛膝多糖, 血根碱+牛膝多糖组在基础饲粮中添加0.75 mg/kg血根碱和400 mg/kg牛膝多糖, 抗生素组在基础饲粮中添加300 mg/kg杆菌肽锌.试验期56 d, 分为2个阶段 (1~28日龄、29~56日龄) 进行.结果表明:1) 试验处理对黄羽肉鸡的1~28日龄、29~56日龄平均日采食量、平均日增重以及1~28日龄料重比均有显著影响 (P<0.05) .29~56日龄阶段, 血根碱组、牛膝多糖组和血根碱+牛膝多糖组的平均日采食量和平均日增重均显著高于对照组和抗生素组 (P<0.05) .血根碱和牛膝多糖对1~28日龄、29~56日龄的平均日采食量均有显著的互作效应 (P<0.05) .2) 28日龄时, 血根碱组、牛膝多糖组和血根碱+牛膝多糖组外周血淋巴细胞增殖率均显著高于对照组和抗生素组 (P<0.05) ; 56日龄时, 血根碱+牛膝多糖组外周血淋巴细胞增殖率最高, 与牛膝多糖组差异不显著 (P>0.05) , 但显著高于对照组、血根碱组和抗生素组 (P<0.05) .血根碱和牛膝多糖对28日龄外周血淋巴细胞增殖率存在显著的互作效应 (P<0.05) .3) 28日龄时, 与对照组相比, 添加血根碱和/或牛膝多糖均在一定程度上提高了黄羽肉鸡的免疫器官指数 (P>0.05) , 但血根碱和牛膝多糖对黄羽肉鸡免疫器官指数没有显著的互作效应 (P>0.05) .由此得出, 血根碱和牛膝多糖对黄羽肉鸡有促生长和提高机体免疫力的作用, 且两者对生长性能和外周血淋巴细胞增殖率都有一定的互作效应.

白藜芦醇是一种含有茋类结构的天然非黄酮类多酚类化合物,具有多种生物学功能:可抑制和清除体内过量的自由基,激活机体抗氧化酶系统,发挥抗氧化功能;通过调控一系列炎症或抗炎通路,降低机体内炎性因子表达,降低炎症反应。白藜芦醇还具有调节糖脂代谢、保护心血管、抗癌、抗病毒等重要生物学功能,且具有安全高效、无残留、不产生耐药性的优点,在动物生产中的应用具有广阔的应用前景。动物试验表明,白藜芦醇能提高肉鸡平均日增重、平均采食量,降低料重比;提高育肥猪肉品质,增加背最长肌pH 24 h、红度、粗蛋白质和肌红蛋白含量,还可降低24 h后肌肉亮度、剪切力、滴水损失、背膘厚度;提高动物机体内抗氧化酶活性;降低炎性因子的产生,增强机体免疫;改善肠道菌群组成和肠道黏膜形态;抑制癌细胞的生成。本试验针对白藜芦醇生物学特性、生理功能及其在畜禽生产中的应用现状进行综述,旨在为其在畜禽生产中的应用提供参考依据。

摘要:
Heat stress is a non-specific physiological response of the body when exposed to high ambient temperatures, which can break the balance of body redox and result in oxidative stress that affects growth performance as well as the health of poultry species. Polyphenols have attracted much attention in recent years due to their antioxidant ability and thus, can be an effective attenuator of heat stress. In this paper, the potential mechanisms underlying the inhibitory effect of polyphenols on heat stress in poultry has been reviewed to provide a reference and ideas for future studies related to polyphenols and poultry production.

摘要:
断奶应激会给养猪生产带来许多问题,抗生素是解决仔猪断奶应激问题的手段之一。长期使用抗生素会引起耐药菌产生、降低机体免疫力,故我国决定在2020年开始全面禁止抗生素在饲料中的使用。而短链脂肪酸是一种具有多种生理功能的添加剂,并且没有毒副作用,是一种绿色无污染的抗生素替代品,或将在养猪生产中起到重要作用。本文综述了短链脂肪酸在动物机体内的吸收代谢过程及其参与能量代谢、改善消化道微生态和提高机体免疫等生理功能,并介绍了部分短链脂肪酸在仔猪生产上的应用效果,旨在响应国家禁抗政策的同时,为生产中提高仔猪生长性能和替代抗生素的使用提供新思路。

摘要:
This study investigated the effect of dietary resveratrol supplementation on growth performance, rectal temperature, and serum parameters of yellow-feather broilers under heat stress. A total of 480 yellow-feather broilers (28-day-old) were randomly allotted to five groups with six replicates. A thermoneutral group (TN) (24 ± 2°C) received a basal diet and another four heat-stressed groups (37 ± 2°C for 8 hr/day and 24 ± 2°C for the remaining time) were fed the basal diet or basal diet with 200, 350, and 500 mg/kg resveratrol for 14 consecutive days. The results revealed that resveratrol supplementation improved average daily gain (p = 0.001), and decreased (p < 0.05) rectal temperature from d 3 when compared with heat-stressed group without resveratrol. In addition, supplementation with resveratrol at 350 or 500 mg/kg lowered (p < 0.05) the contents of corticosterone, adrenocorticotropic hormone, cholesterol, triglycerides, uric acid, malonaldehyde, and activities of aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase, increased (p < 0.05) the levels of triiodothyronine, the ratio of triiodothyronine to thyroxine, total protein, glutathione, and activities of alkaline phosphatase, total superoxide dismutase, catalase, and glutathione peroxidase, though with few fluctuation. In conclusion, supplementation with resveratrol can improve the growth performance by positively regulating serum metabolic parameters and alleviating tissue oxidant damage of broilers under heat stress. © 2019 Japanese Society of Animal Science

摘要:
The intensive production system for broiler chicken is characterised by the provision of a suitable micro-climatic condition such as temperature, airflow, relative humidity and light for proper bird's management which always, together with appropriate feeding and nutrition, favours the full growth and production potentials of the birds. Lighting, amongst other factors, is a potent and critical micro-climatic component in broiler houses as it influences many behavioural, physiological and metabolic processes in birds. To optimise the intensive system for broiler production, various lighting programmes (regarding light duration and its distribution, light colour/wavelength and light intensity) have been explored. This review compares the effects of different elements of lighting regimen on the growth performance, health, and welfare and carcass characteristics of broilers. Considering this, various degrees of intermittent photo-period (i.e. mixing photo- A nd scoto-periods within 24 hours) rather than one continuous photoperiod have been proven to significantly improve broilers' weight gain by 3.4-5.8%, feed to gain ratio up to 7.3%, mobility up to 46.5%, decrease mortality rate ranging between 0.43% and 0.72%, and finally, increase carcass yield. Short wavelength lights and light intensity of ≥5 lux after the initial brooding period are said to stimulate birds' metabolism and growth thereby, enhancing the production system. In conclusion, the lighting programme, apart from improving broiler productivity, could reduce cost expended on energy in an intensive production system. © 2018 World's Poultry Science Association.

旨在研究血根碱和/或牛膝多糖对黄羽肉鸡血液生化指标及免疫性能的影响.试验选用1日龄(d)黄羽肉鸡300只,随机分成5个处理,每个处理6个重复,每个重复10只.试验分为两个阶段(1~28 d,28~56 d)进行.结果 显示,血根碱和/或牛膝多糖在一定程度上能够降低血液生化指标.1~28 d,血根碱和牛膝多糖对TC有显著的互作效应(P<0.05).28~56 d,血根碱和牛膝多糖对血液生化指标无显著互作效应(P>0.05);血根碱和牛膝多糖联合添加显著提高了脾小结直径和动脉周围淋巴鞘面积(P<0.05),两者对脾小结直径没有显著互作效应(P>0.05),对动脉周围淋巴鞘有极显著互作效应(P<0.01).血根碱和牛膝多糖在一定程度上降低了NF-κB、ERK1/2的平均光密度,两者对NF-κB、ERK1/2在脾脏中表达无显著影响或无显著互作效应(P>0.05).上述结果表明,血根碱和牛膝多糖有提高肉鸡免疫性能的作用,和抗生素效果相当.

摘要:
Ruminant production, especially in the tropics and developing countries suffers a setback when compared with the temperate and developed countries, which is attributable to the kinds of available feed resources in the region of production. In the tropics, ruminants are restricted to grazing on low-quality forages, crop residues and agro-industrial by-products with very little or no concentrate diets, which adversely affect the animals in exhibiting their full production potential. Considering this fact, there is an increasing interest in improving the digestibility of these feed resources. In recent years, researchers have explored several methods to enhance the functions of rumen microflora, improve digestion and fermentation processes, as well as increase bioavailability and utilization of nutrients through feed supplementation. This review aims to explore the positive effects of supplementation of ruminant diets with probiotics or botanical extracts and their metabolites on the productivity of the animals. Moreover, the functions of these non-pathogenic and non-toxic live microorganisms (probiotics) and plant biologically active compounds (botanical extract) are explored because of the ban on non-therapeutic use of antibiotics as growth promoters coupled with the critical preference of consumers to high quality and safe animal products. It has been reported that these alternative supplemental products have a beneficial impact on both animal health and productivity, which is affecting stabilization of rumen environment, inhibition of pathogenic bacteria proliferation in gastro-intestinal tract, modulation of immune response, increase in fibre degradation and fermentation, nutrients availability and utilization, animal growth performance and milk production, among others. However, long-term in vivo studies are still required to determine the synergetic effects of these 2 safe supplemental products. © 2018 Chinese Association of Animal Science and Veterinary Medicine

The experiments reported in this research communication aimed to plot the expression pattern of Sirt3, a master regulator of energy metabolism and antioxidation defence, in the liver of dairy goats during perinatal period. Ten healthy dairy goats in late pregnancy were chosen, and needle biopsy was applied to collect liver samples at 1-week intervals. Protein levels of hepatic Sirt3 were analysed by western-blotting. Serum enzyme activities of manganese superoxide dismutase (Mn-SOD) and non-esterified fatty acids (NEFA) levels were measured, and their correlation with Sirt3 mRNA levels was also estimated. Compared with >3-week before parturition (BP), Sirt3 proteins were significantly reduced at 1-week after parturition (AP) and 2-week AP (P < 0·05), but increased on the day of parturition (P < 0·01). Correlation analysis revealed a positive association between hepatic Sirt3 mRNA levels and serum enzyme activity of Mn-SOD (r = 0·46), but a negative association between that and serum NEFA levels (r = -0·41). These data indicate that the decreased hepatic expression of Sirt3 might be one of the reasons that dairy goats undergo oxidative stress after parturition. Background: Our previous study suggested that short-term lysine restriction improved feed intake and gut microbiota in piglets. Thus, in this study, we further used proteomics technology to investigate the potential mechanism associated with long-term lysine restriction in pigs. In addition, blood biochemical parameters, meat quality, and muscle nutrient transporters were also evaluated in lysine restricted pigs. Results: The results showed that 131 proteins in the liver were markedly altered in response to dietary lysine restriction, with 54 being downregulated and 77 being upregulated, which mainly involved in neurodegenerative diseases, oxidative phosphorylation, and metabolic pathways. In addition, lysine restriction markedly reduced serum BUN, CHOL, and HDL abundances. Dietary lysine restriction enhanced meat color lightness and upregulated SLC7A2 expression. Conclusion: Lysine restriction affected meat quality, blood biochemical parameters, and liver global protein expressions. This study is the first attempt to evaluate the liver protein profile of lysine restricted pigs, and the current results may provide valuable information with respect to better understanding the host response to lysine restriction in pigs.

为探讨饲粮中添加含丁酸梭菌的复合益生菌对断奶仔猪饲粮中的抗生素替代效果,试验选取日龄基本一致,体重为(8.5±0.5)kg经过免疫的健康PIC断奶仔猪224头,随机分为4个组,每组4个重复,每个重复14头.对照组饲喂无抗饲粮;试验组分别用含丁酸梭菌的复合益生菌替代0%、50%、100%的抗生素(金霉素75 mg/kg+杆菌肽锌40 mg/kg+吉他霉素50 mg/kg).预试验4 d,正式试验25 d.结果表明:(1)各组生长性能均无显著差异(P>0.05),但随着益生菌的替代比例加大,腹泻率呈线性下降(P=0.04). (2)不同组间的碱性磷酸酶(ALP)、总蛋白(TP)及白蛋白(ALB)均无显著差异(P > 0.05),但血清尿素氮有显著差异(P < 0.05),50%替代组的血清尿氮素与比无抗组及100%替代组分别高出51.19%(P<0.05). (3)各组粪便微生物菌群结构有显著差异(P<0.05),与无抗组相比,100%替代组沙门氏菌降低了79.11%(P<0.01),50%替代组的产气荚膜菌降低94.29%(P<0.01).无抗组的大肠杆菌数量极显著高于其他组(P<0.01). (4)随着时间的推移,50%替代组及100%替代组的沙门氏菌、产气荚膜梭菌以及100%替代组的大肠杆菌持续下降,且差异极显著(P<0.01).利用复合益生菌替代50%的抗生素能有效减低料肉比,但可能增加肾负担.而100%替代抗生素其生长性能与抗生素组效果相当,而且能降低腹泻率.综上所述,在规模化养猪生产中,可以用丁酸梭菌复合益生菌替代抗生素的使用,且以100%替代效果更好.

丁酸梭菌(Clostridium butyricum)又名丁酸梭状芽孢杆菌、酪酸梭菌,是一种产丁酸、乳酸和乙酸的厌氧异养的革兰氏阳性芽孢杆菌,具有耐高温、耐酸和耐多种抗生素等生物学特性。丁酸梭菌定植于畜禽肠道,可抑制肠道有害菌的生长,同时促进有益菌的增殖,有效提高畜禽对营养物质的消化吸收及其抵抗力,进而提高畜禽养殖经济效益。丁酸梭菌存在于土壤或健康动物的粪便中,具有绿色、无残留、安全性好,以及稳定性及耐药性强等特点,是一种具有广泛开发潜力的绿色添加剂。作者主要介绍了丁酸梭菌的特性,简述了其对畜禽肠道形态、肠道屏障、肠道菌群、免疫等方面的影响,回顾了近十年来丁酸梭菌在畜禽生产中的应用进展,并探讨了其在生产应用中的问题,以期为丁酸梭菌的进一步应用提供理论依据。
