山慈菇多糖对H22肝癌小鼠IL-2及p53蛋白表达的影响

摘要:
研究山慈菇多糖对H22的体内抑制作用及其机制。建立小鼠H22腹水瘤和实体瘤模型观察山慈菇多糖的抑瘤作用,Elisa检测实体瘤小鼠外周血IL-2水平,HE染色法观察小鼠肿瘤组织的细胞形态,SABC免疫组化法检测p53蛋白表达情况。能改善腹水瘤小鼠的生存状态和延长生存天数,延长率为21.4%。实体瘤小鼠低、中、高剂量组抑瘤率分别为21.58%、32.76%、40.05%,IL-2的中高剂量组相对模型组都具有极显著性(P<0.01),增强了p53蛋白的表达水平。山慈菇多糖延长了腹水瘤小鼠生存时间,解剖观察可能是减少小鼠体内炎症反应;实体瘤小鼠此次试验中呈现了一定的剂量依赖性,其机制可能与提高小鼠IL-2含量从而提高小鼠免疫力有关,p53随着肿瘤细胞分化程度下降而表达增加。

摘要:
Xuefeng black-boned chicken is one of the famous native breeds in China. In this work, we reported the complete mitochondrial genome sequence of the Xuefeng black-boned chicken for the first time, which was determined through PCR-based method. The total length of the mitogenome was 16,783 bp, with the base composition of 30.24% for A, 23.72% for T, 32.52% for C, 13.53% for G, in the order C > A > T > G feature occurring in the Xuefeng black-boned chicken. It contained the typical structure, including 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and 1 non-coding control region (D-loop region). It was similar to the gene arrangement in Silky chicken. The complete mitochondrial genome sequence of the Xuefeng black-boned chicken provided an important data forfurther study on the genetic mechanism. © 2014 Informa UK Ltd.

摘要:
Taoyuan chicken is excellent native breeds in China. This study firstly determined the complete mitochondrial genome sequence of Taoyuan chicken using PCR-based amplification and Sanger sequencing. The characteristic of the entire mitochondrial genome was analyzed in detail, with the base composition of 30.26% A, 23.79% T, 32.44% C, 13.50% G in the Taoyuan chicken (16,784 bp in length). It contained 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and a major non-coding control region (D-loop region). The complete mitochondrial genome sequence of Taoyuan chicken will be useful for the phylogenetics of poultry, and be available as basic data for the genetics and breeding. © 2014 Informa UK Ltd.

摘要:
This study was conducted to explore the effect of chitosan supplementation on mouse intestinal microbiota and interleukin-17 (IL-17) expression. Chitosan supplementation reduced mouse body weight, associating with change of intestinal microbiota (i.e. lowering the ratio of Firmicutes: Bacteroidetes). In antibiotics treated mice, chitosan supplementation had little effect on mouse body weight. Chitosan supplementation decreased expression of IL-17 in mouse jejunum and inhibited the mTOR pathway. In antibiotics treated mice, chitosan also reduced expression of IL-17 in the jejunum. Activation of mTOR signalling in mouse increased expression of IL-17 in the jejunum in the context of chitosan supplementation. We conclude that chitosan supplementation reduces mouse body weight through the intestinal microbiota, and inhibits intestinal IL-17 expression through the mTOR signalling. The discovery of this study suggests the potentials of chitosan as a functional supplement in intestinal health.

Linwu mallard is an excellent native breeds from Hunan province in China. This is the first study to determine the complete mitochondrial genome sequence of L. mallard using PCR-based amplification and Sanger sequencing. The characteristic of the entire mitochondrial genome was analyzed in detail, with the base composition of 29.19% A, 22.19% T, 32.83% C, 15.79% G in the L. mallard (16,605 bp in length). It contained 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and a major non-coding control region (D-loop region). The complete mitochondrial genome sequence of L. mallard will be useful for the phylogenetics of poultry, and be available as basic data for the genetics and breeding. © 2014 Informa UK Ltd.

摘要:
The objective of the study was to determine the role of vitamin D3(VD3) in regulating adaptation and mechanism of rats to low-phosphorus(P) diets. Rats were assigned to 4 diets containing 0.2%, 0.4%, 0.6%,or 0.8% P consisting of 5 replicate cages with 6 rats per replicate cage and fed for 7 days. Four rats from each replicate cage were treated with ethane-1-hydroxy-1,1-diphosphonicacid, tetrasodium salt(EHDP)and 2 rats remained untreated. Twelve hours prior to preparation on d 7, two of the EHDP-treated rats received an intraperitoneal injection of VD3 [1,25-(OH)2 D3] at 600 ng per kg body weight, while two rats did not receive the injection. Rats that did not receive VD3 injection had decreased(P < 0.001) P absorption, but injection of VD3 resulted in increased(P < 0.001) absorption. The effect of VD3 injection was greater(P < 0.001) for rats fed 0.2% P diet than rats fed 0.8% P diet in ileum. Sodium dependent phosphate cotransporter type IIb(Na/Pi-IIb) and 25-hydroxyvitamin D 1-α hydroxylase(CYP27 B1) mRNA level showed the same trend with P absorption. Serum concentration of VD3 and la-hydroxylase activity in rats fed 0.2% P diet were lower than those fed 0.8% P diet. The injection of VD3 increased(P < 0.001)serum concentration of VD3 and la-hydroxylase activity. Thus, VD3 increased Na/Pi-Ⅱb and CYP27 B1 mRNA level and improved serum concentration of VD3 and la-hydroxylase activity in rats fed low-P diets.

摘要:
研究山慈菇多糖对H22肝癌实体瘤小鼠的抗肿瘤作用及其机制。制作小鼠H22实体瘤肝癌模型,通过脾脏指数、胸腺指数分析其对小鼠免疫器官的影响,Elisa检测血清白介素-2(IL-2)含量、肿瘤坏死因子(TNFα)含量,免疫组化法观察(Bcl-2)表达水平。山慈菇多糖的低、中、高剂量组抑瘤率分别为43.74%、37.57%、30.76%,;能增强血清中IL2、TNFα活性;抗凋亡因子Bcl-2的表达量不同程度地减少。山慈菇多糖对H22肝癌小鼠具有一定的肿瘤抑制作用,本次实验不体现剂量依赖性,显示山慈菇多糖无毒副作用,能增强机体的抗肿瘤能力,降低肿瘤的持续增殖能力。

摘要:
试验旨在研究复合添加剂对围产期奶牛生产性能的影响。试验按照年龄、胎次、体重、产犊时间接近的的原则,选择健康的围产前期中国荷斯坦奶牛16头,随机分为两组,分别为对照组和试验组。试验组奶牛每天按照精料量的1.0%添加复合添加剂,对照组不添加;试验期40 d,包括10 d预试期。结果表明:在奶牛产后的第10天,试验组奶牛产奶量比对照组高7.18 kg(P<0.05);在产后的第15天,试验组奶牛泌乳量比对照组高7.75 kg(P<0.05);试验组乳糖率和去脂干物质分别比对照组提高了0.31个百分点和0.59个百分点(P<0.05),其他乳成分二者间均无显著差异(P>0.05)。研究显示,在围产期奶牛日粮中添加此种复合添加剂可以提高围产期奶牛的生产性能。

植酸酶对肉鸭饲用原料钙与磷及其养分真利用率的影响

摘要:
在18种常用肉鸭饲料原料中添加750 U/kg的植酸酶,通过鸭的强饲代谢试验来探究植酸酶对钙、磷及干物质、粗蛋白、粗脂肪、能量等真利用率的改善程度。结果表明:与加酶前比较,加酶后供试原料的总磷真利用率的提高幅度为0.92%~17.29%,其中,对次粉2、麦麸、米糠、玉米总磷的提高幅度差异达到显著或极显著水平,分别为11.79%(P<0.05)、8.46%(P<0.05)、17.29%(P<0.05)、16.33%(P<0.01),其余原料的总磷真利用率均有不同程度的提高,但差异不显著(P>0.05);对钙真利用率的提高幅度为2.74%~22.12%,其中,对碎米(早)、玉米皮、小麦、麦麸、棉粕的钙真利用率的提高作用显著(P<0.05),分别为16.37%、17.12%、30.75%、21.40%、22.12%,对糙米(晚)的提高幅度最小,仅为2.74%,对其余原料的提高幅度均在5%以上;加酶后供试原料的干物质、粗蛋白、粗脂肪、能量真利用率的提高幅度分别为0.03%~14.60%,0.71%~27.05%,2.00%~37.69%、0.17%~10.07%;植酸酶对原料有效磷的改进值范围为0.02~2.60 g/kg。综合分析,在肉鸭饲料原料中添加750 U/kg的植酸酶,对钙、磷及干物质、粗蛋白、粗脂肪真利用率的改善效果较明显。

PCV2 is highly pathogenic, however, its effect on the serum amino acids profile is unknown. This study was conducted to explore the profile of amino acids in the serum in porcine circovirus type 2 (PCV2) infected mice. The serum levels of amino acids were detected with isotope dilution liquid chromatography-mass spectrometry methods at 3, 7, 10 and 14 days post infection (DPI). Meanwhile, the expression of seven amino acids transporters (SLC6A14, SLC6A20, SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A9) in the jejunum was analyzed by reverse transcription polymerase chain reaction (RT-PCR) at 3 and 7 DPI. Serum PCV2 load was also analyzed by quantitative PCR at 3, 7, 10 and 14 DPI. Serum levels of most amino acids, such as Pro, Orn, and Met, significantly (P < 0.05) increased at 3 DPI. However, most amino acids, including Asp, Sar, Arg, Hyl, Pro, Lys, Val, Ile and Leu, significantly (P < 0.05) decreased at 7 DPI. There was no significant difference for most amino acids at 10 and 14 DPI. PCV2 infection significantly (P < 0.05) decreased expression of SLC7A5 and SLC7A6 at 7 DPI. In conclusion, PCV2 infection affects the profile of amino acids in the serum and the expression of amino acids transporters in the intestine.

目的探讨米糠甾醇(rice bran sterol, RBS)对大鼠免疫功能的调节作用。方法健康SD大鼠160只分为正常对照组(CN,灌胃生理盐水)和RBS低、中、高剂量(RBS-L,RBS-M,RBS-H,分别灌胃RBS乳化液30、75和150 mg/(kg·bw·d)组,15d后,测定免疫器官指数、碳廓清指数、血清溶血素、淋巴细胞增殖能力和腹腔巨噬细胞功能等免疫指标。结果与CN组比较,RBS-L和RBS-H组可提高脾脏指数(P<0.05或P<0.01),但RBS-M组差异不显著(P>0.05);RBS各剂量组对大鼠胸腺指数无明显影响(P>0.05);RBS-L和RBS-M组能提高大鼠碳廓清指数(P<0.05);RBS-M组能促进血清溶血素的释放(P<0.05);RBS各剂量组能显著提高大鼠淋巴细胞的增殖能力(P<0.05或P<0.01)和巨噬细胞的吞噬百分率(P<0.05或P<0.01)与吞噬指数(P<0.01)。结论RBS可增强大鼠的免疫功能。

摘要:
试验旨在研究一种复合添加剂对高产奶牛生产性能的影响。根据产奶量、胎次、体重等相近的原则选择4f)头中国荷斯坦牛,随机分成4个处理组,即对照组、试验Ⅰ组、试验Ⅱ组、试验Ⅲ组,对照组饲喂基础日粮,试验Ⅰ、Ⅱ、Ⅲ组在此基础上,分别按每头牛每天精料量的0.5%、0.8%、1.0%添加复合饲料添加剂。、试验期45d,其中预试期10d,正试期35d,测定产奶量、乳成分等指标。结果表明,在高产奶牛日粮中添加复合添加剂对乳成分基本无影响,但可显著提高奶牛产奶量,结合成本考虑,以精料的0.5%添加量效果最好、

摘要:
本试验旨在研究牛膝多糖(Achyranthes bidentata polysaccharides,ABP)、白术多糖(Atractylodes macrocephalaon polysaccharides,AMP)、黄芪多糖(Astragalus polysaccharides,AP)单独或联合使用对断奶仔猪淋巴细胞增殖作用和细胞因子分泌的影响,探讨ABP、AMP、AP免疫调节作用机制.选用256头28日龄的社长大(杜洛克×长白×大白)断奶仔猪,采用2 ×2 ×2因子设计,随机分为8个处理,每个处理4个重复,每个重复8头猪,试验第14天和第28天每个重复随机选取1头仔猪进行前腔静脉采血,测定仔猪外周血淋巴细胞转化率和血清中细胞因子肿瘤坏死因子(TNF)、白细胞介素-1β(IL-1β)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、干扰素-γ(IFN-γ)的含量.结果表明:1)单独或联合添加多糖组的断奶仔猪外周血淋巴细胞转化率较对照组都显著提高(P<0.05).3种多糖合用时淋巴细胞转化率显著高于单独添加1种多糖(P<0.05),较两两合用有不同程度提高(P>0.05).2)分别添加3种多糖组断奶仔猪血清的细胞因子IL-2、IL-1β、TNF、IFN-γ的含量与对照组相比都显著提高了(P<0.05),IL-6含量有一定程度提高,但差异不显著(P>0.05).3种多糖两两合用或三者合用较对照组显著提高了细胞因子IL-2、IL-1β、IL-6、IFN-γ、TNF的含量(P<0.05).由此可知,ABP、AMP、AP单独或合并使用可促进断奶仔猪外周血淋巴细胞转化率和血清细胞因子TNF、IL-1β、IL-2、IL-6、IFN-γ的分泌.

摘要:
This study was conducted to investigate the effect of Achyranthes Bidentata Polysaccharide (ABP) on growth performance, inflammatory responses and hormones in weaned piglets after Escherichia coli Lipopolysaccharide (LPS) challenges. A total of 48 crossbred (Duroc x Large; White x Landrace) male pigs weaned at 28 days of age (8.45±0.14 kg) were randomly allotted to one of four treatments by initial BW in a 2×2 factorial design that included a dietary addition of ABP (0 or 500 mg kg-1) and immunological challenge (with LPS or saline). On day 14 and 21, pigs were injected intraperitoneally with either 100 μg kg-1 BW of LPS or an equivalent amount of sterile saline. Blood samples were obtained 3 h after injection for analysis of Tumor Necrosis Factor-α (TNF-α), Prostaglandin E2(PGE2), cortisol, Growth Hormone (GH) and Insulin-like Growth Factor (IGF)-I. The results showed that LPS challenge decreased Average Daily Feed Intake (ADFI) (p<0.05) from 14-21 and day 21 "28. ABP increased ADG (p<0.05) and ADFI (p<0.05) from day 21 "28. An interaction (p<0.05) between LPS challenge and diet was observed for the plasma concentration of TNF-α, PGE2, cortisol, and IGF-I after both LPS challenges such that among LPS-treated pigs, pigs fed the ABP diet were lower for TNF-α (p<0.05) and higher IGF-I (p<0.05) than those receiving the control diet. These results indicated that ABP alters the release of inflammatoiy cytokines that may lead to improved pig performance during an immunological stress. © Medwell Journals, 2014.

精氨酸对断奶仔猪生产性能及血清指标的影响

为研究精氨酸在断奶仔猪日粮中的添加效果,选取270头21d体重为6.20±0.3 1kg的杜×长×大健康仔猪,随机分为5组,A组为对照组,饲喂基础日粮+2.05%的丙氨酸;B、C、D、E组在基础日粮的基础上分别添加0.5%、1.0%、1.5%和2.0%的精氨酸,试验期为7d.测定仔猪的生产性能、血清生化指标及激素水平.结果表明:C、D、E组的平均日采食量显著提高(P<0.05),而料内比显著降低(P<0.05);D、E组的平均日增重显著高于对照组(P<0.05);C、D组断奶仔猪血清中的三碘甲状腺原氨酸、皮质醇的含量均显著低于对照组(P<0.05),而胰高血糖素的含量、四碘甲状腺原氨酸、胰岛素的浓度高于对照组,差异不显著(P>0.05).说明一定水平的精氨酸能有效改善断奶仔猪的生产性能,并能对血清生化指标和血清激素产生一定影响,以1.5%水平的添加效果最好.
