摘要:
The aim of the present study was to investigate the effect of selenium (Se), polysaccharide of Atractylodes macrocephala Koidz. (PAMK), and the combination of Se and PAMK on the immune response, heat shock protein (HSP) levels under heat stress (HS) condition in chicken spleen. Two hundred chickens were randomly divided into two groups, the HS group and the control (Con) group. Then these chickens were treated with Se (0.3 mg/kg), PAMK (200 mg/kg) alone, and the combination of Se (0.3 mg/kg) and PAMK (200 mg/kg). The antioxidative enzymes, cytokines contents, and expression levels of HSP27 and HSP70 were examined in chicken spleen. The results indicated that HS induced higher levels of TNF-α, IL-4, HSP27, HSP70, and MDA levels but lower level of IFN-γ, IL-2, Gpx, and SOD in spleen (P<0.05). These responses were ameliorated by the treatment of Se, PAMK alone, and the combination of Se and PAMK (P<0.05 or not) The results showed that under common condition, Se and PAMK could improve the immune response by enhancing the levels of some cytokines to proper levels; however, under HS condition, Se and PAMK could change the abnormal levels of cytokines and oxidative damages to ameliorate the injury induced by HS. In addition, there existed synergistic effect on the modulation of these biomarkers in chicken spleen between Se and PAMK. So both Se and PAMK play important roles in regulating the immune function in chicken. Considering the synergistic effect on immune regulation of PAMK, this herb deserves further investigation to evaluate its role in improving the effect of traditional immune regulators. © 2014 Springer Science+Business Media.

摘要:
采用单因子随机分组设计,选取1日龄灵山土鸡540只,随机分为6组,每组3个重复,每个重复30只鸡,分别采用不同的光照时间和光照强度,研究不同光照制度对灵山土鸡冠头性状及体重的影响。前60日龄6个组均采用自然光照+12 h的人工光照,60日龄至上市1、2、3组仍采用自然光照+12 h的人工光照,光照强度依次为0.8 W/m^2、2.5 W/m^2和3.75 W/m^2;4、5、6组采用自然光照+4 h的人工光照,光照强度依次为0.8 W/m^2、2.5W/m^2和3.75 W/m^2。试验鸡采用地面平养方式,饲养周期为110 d,试验结束时测量鸡冠头性状和体重。结果表明,4、5、6组冠高、冠长和冠面积均优于人工1、2、3组,说明光照时间以自然光照+4 h的人工光照对冠头发育更优,且光照时间大于光照强度对冠头发育的影响。1、2、3组体重有高于4、5、6组的趋势,说明延长光照时间有利于鸡的增重。总之,光照时间对土鸡冠头发育的刺激作用大于光照强度,其中以每天补充4 h、强度为3.75 W/m^2的人工光照效果最佳。

茶氨酸(Thenine,N-gamma-ethyl-L-glutamine),是一种非蛋白质氨基酸,最早发现于绿茶(Camellia sinensis)叶中,目前,除在蘑菇、油茶、红山茶等植物中已检测出微量存在外,在其它植物中尚未检测到,其CAS登录号(CAS Registry Number)为3081-61-6,分子式C_7H_(14)N_2O_3,分子量为174.1977。茶氨酸是茶叶的主要呈味物质,约占茶叶干重的1.0%~2.0%,约占茶叶中游离氨基酸总量的50%,自然存在的茶氨酸均为L型。L-茶氨酸具有抗肿瘤、神经保护、延缓衰老、提高动物认知能力等作用,但对其是否具有调控机体免疫机能的作用鲜见报道。本研究旨在探讨L-茶氨酸对小鼠巨噬细胞系RAW264.7炎症模型细胞因子分泌的影响,为调节脂多糖(LPS)诱导的炎症反应提供理论依据。

摘要:
Garlic (Allium sativum L.) has long been used both for culinary and medicinal purposes by many cultures. Population and preclinical investigations have suggested that dietary garlic intake has health benefits, such as lowering the risk of esophageal, stomach and prostate cancers. Extensive studies from laboratory and animal models have revealed that garlic has a wide range of biological activities, and garlic organosulfur compounds (OSCs) are responsible for the biological activities. However, the presence and potency of garlic OSCs vary with respect to the mode of garlic preparation and extraction. Cooked or processed garlic products showed different kinds of garlic OSCs, some of which are highly unstable and instantly decomposed. These facts, possibly gave paradoxical results on the garlic effects. In this review, we first summarized the biotransformation processes of garlic alliin into different garlic OSCs as well as the garlic OSCs compositions from different garlic preparations. Next, we reviewed the chemopreventive functions and molecular mechanisms focusing on the anti-inflammation, antioxidation, anti-diabetes and anticancer activity behind different garlic OSCs.

博落回生物碱对猪肠上皮细胞增殖的影响

本试验旨在研究博落回中血根碱、白屈菜红碱、原阿片碱和别隐品碱在不同浓度、不同时间段对猪肠上皮细胞(IPEC-1)增殖的影响。IPEC-1预培养24h后,在培养基中分别加入不同浓度(4、2、1、0.5、0.25、0.1、0.05、0.025、0.0125、0.00625μg/mL的血根碱或白屈菜红碱;100、50、25、12.5、6.25、3.125、1.5625μg/mL的原阿片碱或别隐品碱,各组不同浓度梯度均设8个重复)的博落回生物碱继续培养12或24h,4-甲基偶氮唑盐(MTT)染色法检测细胞增殖。结果表明:0.25~4μg/mL的血根碱、0.25~4μg/mL的白屈菜红碱、100μg/mL的原阿片碱或100μg/mL的别隐品碱对IPEC-1的增殖有显著的抑制作用(P<0.05);而0.00625~0.1μg/mL的血根碱、0.00625~0.1μg/mL的白屈菜红碱、3.125~25μg/mL的原阿片碱或1.5625~25μg/mL的别隐品碱对IPEC-1增殖具有显著促进作用(P<0.05),随着博落回生物碱浓度的不断下降,这种促进作用又缓慢减弱;此外,延长博落回生物碱处理时间至24h,发现仅0.1μg/mL白屈菜红碱、25μg/mL原阿片碱和25μg/mL别隐品碱对IPEC-1增殖具有显著促进作用(P<0.05),其余各试验组与对照组相比均无显著性差异(P>0.05)。由此可见,博落回生物碱在一定浓度范围内可促进IPEC-1的增殖。这说明博落回生物碱提高动物生长性能可能是通过促进小肠细胞增殖分化更新,从而增强小肠消化、吸收、免疫屏障和应激反应等功能的途径实现的。

在养猪生产中,常用日增重来衡量断奶仔猪的健康和生长性能,而影响断奶仔猪日增重的主要因素是断奶时所面临的大量应激(饲粮改变、母仔分离、环境变化等)。断奶应激会使仔猪肠道发生组织形态学和生理学上的变化,如肠绒毛萎缩、隐窝增生、肠黏膜通透性增强等,这些变化会引起仔猪消化和吸收功能下降或免疫机能降低,最终导致腹泻和生长性能下降。本文针对早期断奶应激引起的仔猪肠黏膜结构和屏障功能的变化作一简要综述,总结了一些可以作为评价肠黏膜结构和功能的一些标记物,并列举了饲粮中可用来缓解断奶应激损伤的一些饲料原料或添加剂,以期为仔猪生产和断奶应激的研究提供参考。

摘要:
This study investigated the effects of dietary resveratrol at 0, 200, 400, or 600 mg/kg of diet on the performance, immune organ growth index, serum parameters, and expression levels of heat shock protein (Hsp) 27, Hsp70, and Hsp90 mRNA in the bursa of Fabricius, thymus, and spleen of 42-d-old female blackboned chickens exposed to heat stress at 37 ± 2°C for 15 d. The results showed that heat stress reduced daily feed intake and BW gain; decreased serum glutathione (GSH), growth hormone, and insulin-like growth factor- 1 levels; and inhibited GSH peroxidase (GSH-Px), superoxide dismutase (SoD), and catalase (CAT) activities compared with birds subjected to thermo-neutral circumstances. Chickens that were fed diets supplemented with resveratrol exhibited a linear increase in feed intake and BW gain (P < 0.001); serum GSH, growth hormone, and insulin-like growth factor-1 levels (P ≤ 0.01); and GSH-Px, SoD, and CAT activities (P < 0.001) compared with chickens that were fed diets without resveratrol during heat stress. In contrast, serum malonaldehyde concentrations were decreased (P < 0.001) in the chickens fed a resveratrol-supplemented diet. Heat stress also reduced (P < 0.05) the growth index of the bursa of Fabricus and spleen; however, it had no effect on the growth index of the thymus. The growth index of the bursa of Fabricius and spleen increased (P < 0.05) upon heat stress and coincided with an increase in supplemental resveratrol levels. The expression of Hsp27, Hsp70, and Hsp90 mRNA in the bursa of Fabricius and spleen were increased (P < 0.01), but those of Hsp27 and Hsp90 mRNA in thymus were decreased (P < 0.01) under heat stress compared with no heat stress. Resveratrol attenuated the heat stress-induced overexpression of Hsp27, Hsp70, and Hsp90 mRNA in the bursa of Fabricius and spleen and increased the low expression of Hsp27 and Hsp90 mRNA in thymus upon heat stress. The results suggest that supplemental resveratrol improves growth performance and reduces oxidative stress in heat-stressed black-boned chickens by increasing serum growth hormone concentrations and modulating the expression of heat shock genes in organs of the immune system. © 2014 Poultry Science Association Inc.

摘要:
The aim of this study was to investigate whether supplementation with chitosan (COS) could reduce diarrhea and to explore how COS alleviates intestinal inflammation in weaned pigs. Thirty pigs (Duroc×Landrace×Yorkshire, initial BW of 5.65±0.27) weaned at age 21 d were challenged with enterotoxigenic Escherichia coli during a preliminary trial period, and then divided into three treatment groups. Pigs in individual pens were fed a corn-soybean meal diet, that contained either 0 (control), 50 mg/kg chlortetracycline, or 300 mg/kg COS for 21 days. The post-weaning diarrhea frequency, calprotectin levels and TLR4 protein expression were decreased (P<0.05) in both the COS and chlortetracycline groups compared with control. Simultaneously, supplemental COS and chlortetracycline had no effect on the mRNA expression of TNF-α in the jejunal mucosa, or on the concentrations of IL-1β, IL-6 and TNF-α in serum. However, COS supplementation improved (P<0.05) the mRNA expression of IL-1β and IL-6 in the jejunal mucosa. The results indicate that supplementation with COS at 300 mg/kg was effective for alleviating intestinal inflammation and enhancing the cell-mediated immune response. As feed additives, chitosan and chlortetracycline may influence different mechanisms for alleviating inflammation in piglets.

摘要:
目的:研究米糠植物甾醇对非酒精性脂肪性肝炎(non-alcoholicsteatohepatitis,NASH)大鼠的干预治疗作用。方法:实验选用50只健康SD雄性大鼠,随机分为正常对照组、NASH模型组和米糠甾醇低、中、高3个剂量组(65、130、195mg/kg)。高脂饲料饲喂造模,各实验组同时进行米糠甾醇溶液灌胃干预。13周后观察各组大鼠肝组织病理学特点,测定血清中血脂总胆固醇、甘油三酯、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇含量和谷丙转氨酶、谷草转氨酶、一氧化氮合酶和超氧化物歧化酶活性,并比较各组肝脏指数和脾脏指数。结果:肝脏组织切片观察到NASH模型组有明显的肝细胞脂肪变性和炎症聚集浸润,米糠甾醇各治疗组肝组织病变呈现不同程度减轻。NASH模型组血清高密度脂蛋白胆固醇含量、超氧化物歧化酶、一氧化氮合酶、谷丙转氨酶和谷草转氨酶活性与正常对照组相比,均表现出明显异常;米糠甾醇各剂量组与NASH模型组比较,血清谷丙转氨酶显著降低(P<0.001)、谷草转氨酶/谷丙转氨酶比值显著升高(P<0.01);低剂量组血清超氧化物歧化酶(P<0.05)和一氧化氮合酶(P<0.001)显著降低;中剂量组血清高密度脂蛋白胆固醇显著升高(P<0.05)。结论:米糠甾醇对大鼠非酒精性脂肪性肝炎有预防与治疗作用。

摘要:
目的 确定制备复方黄连注射剂的最佳脱色工艺方法和条件.方法 以复方黄连注射剂未脱色药液为原料,以脱色率、工艺成本、脱色后药液的气味变化及对耐药金黄色葡萄球菌的最低抑菌浓度(MIC)等为评价指标,对活性炭吸附法、铅盐沉法、氧化铝吸附法、石灰乳沉淀法、硅胶吸附法5种方法进行综合评价.结果 经过综合分析,确定最佳脱色方法为硅胶吸附法.通过正交试验,确立硅胶吸附法的最佳工艺条件为硅胶用量35%,温度40℃,时间30 min,在该条件下脱色率为35.98%,复方黄连注射剂对耐药金黄色葡萄球菌的MIC值为7.8 mg/mL.结论 用正交设计确定的硅胶吸附法最适脱色条件可实现复方黄连注射剂的有效脱色.

摘要:
In modern pork production industry, various prophylactic measures (such as anti-infective drugs, vaccines, sanitation, vector control, the optimisation of housing management, nutrition regulation) are used to prevent and treat infections, however, many measures are facing failures and side effects in some infectious diseases, such as porcine circovirus associated diseases (infected by porcine circovirus), piglet edema disease (infected by shiga toxin type 2e (Stx2e)-expressing Escherichia coli), and Glässer's disease (infected by Haemophilus parasuis). Compelling evidences indicate that arginine supplementation provided a useful adjuvant treatment in these infections; these findings have important implications for preventive medicine and animal production. This article reviewed arginine metabolism in mammals and arginine profile in infections, and provided the metabolic evidence for the beneficial role of arginine supplementation in infectious diseases.

摘要:
Scope: Garlic is used for both culinary and medicinal purposes by many cultures. The garlic organosulfur compounds (GOSCs) are thought to be bioactive components. This study aims to clarify the antiinflammatory effects and molecular mechanisms of GOSCs in both cell and animal models. Methods and results: RAW264.7 cells were treated with six kinds of GOSCs to screen their influence on cyclooxygenase-2 and inducible nitric oxide synthase expression by Western blotting. Prostaglandin E2 and nitrite were measured by ELISA and Griess reaction, respectively. Cytokines in culture medium were assayed by the multiplex technology. Proteins were detected by Western blotting. Mouse paw edema was induced by LPS. The results revealed that diallyl trisulfide (DATS) was a strongest inhibitor for cyclooxygenase and inducible nitric oxide synthase among GOSCs, and reduced the levels of LPS-induced IL-6, IL-10, IL-12(p70), KC, MCP-1, and TNF-α. Cellular signaling analysis revealed that DATS downregulated AKT1/TGF-β-activated kinase-mediated mitogen-activated protein kinase (MAPK) and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) pathways. Furthermore, DATS activated Nrf2-mediated expression of HO-1 and NQO1 and reduced LPS-induced intracellular reactive oxygen species, which may contribute to suppress inflammatory mediator production. Finally, in vivo data demonstrated that DATS attenuated LPS-induced mouse paw edema. Conclusion: DATS as a potential inhibitor revealed antiinflammatory effect in both cell and animal models by downregulating AKT1/TGF-β-activated kinase-mediated NFκB and MAPK signaling pathways. © 2013 WILEY-VCH Verlag GmbH & Co. 湘村黑猪各阶段营养需求参数研究

本研究旨在通过研究日粮蛋白质、赖氨酸及磷相对于湘村黑猪各生长阶段的生长性能指标的定量关系,以确定湘村黑猪适宜的蛋白质、赖氨酸及磷的需要量.试验3个阶段分别选用起始体重为(10.08±0.49)、(30.07±0.73)和(59.96±0.83)kg的湘村黑猪阉公猪各48头,随机分为8个处理组,每个处理6个重复,每个重复1头猪,根据U8·(85)均匀设计表设计3因子8水平的试验.对均匀设计试验结果偏最小二乘回归方程寻优表明,除了30~60和60~90 kg阶段的磷需求量稍高于中国瘦肉猪饲养标准,湘村黑猪的3个因子的需要量和生长性能较中国瘦肉猪饲养标准低,具有地方猪种的特性;湘村黑猪10~30 kg阶段3个因子最优组合为蛋白质17.18%、赖氨酸0.90%、磷0.56%,日增重0.48 kg·d-1,饲料转换率2.58;30~60 kg阶段3个因子最优组合为蛋白质15.84%、赖氨酸0.79%、磷0.52%,日增重0.68 kg· d-1,饲料转换率2.97;60~90 kg阶段3个因素最优组合为蛋白质13.74%、赖氨酸0.68%、磷0.47%,日增重0.77 kg·d-1,饲料转换率3.60.3个因子中,日粮蛋白质水平对湘村黑猪平均日增重的影响较大,日粮赖氨酸水平对饲料转化率的影响较大.均匀设计能用于猪营养需要的研究,湘村黑猪的营养需求具有自身的特点,不宜照搬其它猪的营养需要标准,对其其余营养素需要研究还有待进一步加强.

炎热气候条件下母猪妊娠后期不同组织中GRα mRNA的表达规律研究

本研究主要揭示炎热气候条件下母猪妊娠后期各组织器官GRα mRNA的表达规律,以期为GRα在母猪的热应激环境下的分子生理机制研究提供参考.试验选用12头妊娠母猪,根据妊娠日龄不同分为2组,每组6头,分别是妊娠90、110天.按照常规方法屠宰母猪,迅速采集脑、卵巢、心脏、肝脏、脾脏、肺脏和肾脏,液氮速冻后-80℃保存,提取RNA,反转录,利用实时荧光定量PCR(qRT-PCR)方法检测各组织中GRα mRNA的相对表达量.结果显示:妊娠90天和110天时,GRα的mRNA表达丰度依次分别表现为:90天丰度,肺脏>脾脏>肝脏>肾脏>脑>卵巢>心脏;110天丰度,脾脏>肺脏>肝脏>肾脏>卵巢>心脏>脑.其次,妊娠110天与90天相比,GRα mRNA的相对表达量除了在肺脏和脑中极显著降低(P<0.01),在肝脏中显著降低之外(氏0.05),其他组织则未表现出显著性变化(p0.05).结果表明:炎热气候条件下,母猪体内GRα mRNA优先在肺脏和脾脏中表达,其次是肝脏和肾脏,在脑、卵巢和心脏中的表达量最低,这种变化随妊娠时间延长更为明显,但同时表现出较明显的组织特异性.
