摘要:
Fusarium spp. are significantly important plant pathogens, and some of them are ethanol-producing strains. During infection and/or ethanol production, Fusarium requires a plenty of energy that is mainly provided by mitochondria. Here we report the first mitogenome from a selected Fusarium oxysporum strain mh2-2 that produces ethanol from glucose and xylose. The size of this mitogenome, 46 kb, is different from the size of any reported Fusarium mitogenome. Our results provide insight into the functions and evolution of mitochondrial genes and genomes. © 2017, © 2017 The Author(s). Published by Informa UK Limited, trading as Taylor & Francis Group.

摘要:
为了研究蔗糖在马铃薯试管苗离体繁殖中的作用,本文以2个马铃薯品种为试验材料,在(25±1)℃培养条件下添加不同蔗糖浓度对马铃薯试管苗生长发育情况以及表型特征进行观察与分析。结果表明,蔗糖处理2个马铃薯试管苗后其植株高度、根系长度、节数、叶片数目、第2节节长、第2节节粗、根系鲜重、根系干重、地上部鲜重和地上部干重处理间差异明显;蔗糖浓度显著影响马铃薯试管苗离体培养根系的生长和地上部形态的建成;无蔗糖

摘要:
以登海661为参试材料,研究不同播期及不同种植模式对秋玉米生长及产量的影响,探索秋玉米的适宜播期及高产高效种植模式。结果表明:(1)不同播期的秋玉米物候期和植物学特性差异不显著,但其产量、穗粒数、单穗重和千粒重差异明显,产量随播期延后呈递减趋势,7月3日的产量最高,为13 080.5 kg/hm~2,其穗粒数最多,单穗重最大;7月13日播种的千粒重最大。因此,秋玉米的播期以7月上旬为佳,能获得较高产量。(2)种植密度越大,截光率越大;HH模式株高最高,HH、H与FP、N0之间株高、叶面积指数及穗长差异显著;N0的生物量最小;FP、HH与H、N0之间穗粒差异显著,各处理间单穗重差异不显著;HH的产量最高,为12 604.5 kg/hm2,比FP增产62.7%,H的产量为10 684.5kg/hm~2,比FP增产37.9%,N0的产量最低,为7 087.5 kg/hm~2。因此,HH的种植模式是适合湖南的秋玉米高产高效种植模式。

摘要:
While plants produce reactive oxygen species (ROS) for stress signaling and pathogen defense, they need to remove excessive ROS induced during stress responses in order to minimize oxidative damage. How can plants fine-tune this balance and meet such conflicting needs? Here, we show that XANTHINE DEHYDROGENASE1 (XDH1) in Arabidopsis thaliana appears to play spatially opposite roles to serve this purpose. Through a large-scale genetic screen, we identified three missense mutations in XDH1 that impair XDH1’s enzymatic functions and consequently affect the powdery mildew resistance mediated by RESISTANCE TO POWDERY MILDEW8 (RPW8) in epidermal cells and formation of xanthine-enriched autofluorescent objects in mesophyll cells. Further analyses revealed that in leaf epidermal cells, XDH1 likely functions as an oxidase, along with the NADPH oxidases RbohD and RbohF, to generate superoxide, which is dismutated into H2O2. The resulting enrichment of H2O2 in the fungal haustorial complex within infected epidermal cells helps to constrain the haustorium, thereby contributing to RPW8-dependent and RPW8-independent powdery mildew resistance. By contrast, in leaf mesophyll cells, XDH1 carries out xanthine dehydrogenase activity to produce uric acid in local and systemic tissues to scavenge H2O2 from stressed chloroplasts, thereby protecting plants from stress-induced oxidative damage. Thus, XDH1 plays spatially specified dual and opposing roles in modulation of ROS metabolism during defense responses in Arabidopsis. © 2016 American Society of Plant Biologists. All rights reserved.

Potato virus Y (PVY) undergoes continuous nucleotide mutation and genome recombination, leading to novel strains with varying pathogenicity on host plants. Little is known about how tomato plants respond to infections with different PVY strains, especially the emerging strains such as PVYNTN and PVYN:O, and in combination with other viral species such as Potato virus X (PVX). In this study, the response of tomato (cultivar Rutgers) plants to single and mixed infections with PVY and PVX was investigated. Plants infected singly with PVYO, PVYN:O, PVYN or PVYNTN developed mosaic symptoms and leaf deformation; whereas plants infected with PVX developed mild local lesion and varying degrees of mosaic and leaf deformation symptoms. Mixed PVX+PVY infections induced more severe symptoms, which include severe local and systemic leaf necrosis, leaf drop and severe leaf deformation, compared to single infections with PVX or PVY, indicating PVX-PVY synergism. The level of PVX-PVY synergism was affected significantly by PVY strains, with PVYNTN being the greatest effector. Quantitative analysis of viral titre indicated that mixed PVX+PVY infections elevated PVX level and reduced PVY level. A comparative transcriptional analysis of 46 key genes involved in signal molecule synthesis and signal transduction using NanoString technology revealed diverse gene expression profiles in both single and mixed infections. Two hierarchical expression clusters, one up-regulated and the other down-regulated, were observed during the infections. In general, mixed infections led to more genes being differentially regulated than single infections. The expression alteration of genes involved in oxidative stress and salicylic acid (SA)-pathways is particularly noteworthy, as the mixed infections, especially PVX+PVYNTN infections, led to a further elevation of reactive oxygen species (ROS) and SA producing abilities and a reduction of ROS scavenging ability over single infections. © 2015, UK Crown.

马铃薯是世界第三大粮食作物,目前我国的种植面积和产量均居世界第一。晚疫病是马铃薯生产上的毁灭性病害,每年造成的损失巨大,严重影响马铃薯生产。种植抗病品种是防治晚疫病最经济、环保、有效的途径,可以从根本上解决农药残留和环境污染的问题。为促进马铃薯晚疫病的抗病育种研究,本文总结了不同育种方法在育种工作中的贡献及其存在的问题,并对基因工程育种将来在晚疫病抗性育种中的作用进行了展望,有助于马铃薯晚疫病抗性未来的晚疫病抗性育种工作。

摘要:
[目的]运用HPLC法比较分析11种甘薯不同部位绿原酸含量,为甘薯尤其是地上部分茎叶的开发利用提供参考。[方法]样品通过质量为200倍的甲醇超声提取20 min,乙腈-0.2%磷酸为流动相,梯度洗脱,对绿原酸、4,5-O-咖啡酰基奎尼酸、3,5-O-咖啡酰基奎尼酸、3,4-O-咖啡酰基奎尼酸形成良好分离,加标回收率分别为:94.74%、102.63%、100.01%和101.05%。[结果]不同甘薯品种间及同一品种不同部位间咖啡酰奎尼酸类化合物含量显著差异;地上部分以双咖啡酰奎尼酸含量为主,块薯中以绿原酸含量最高;各组织部位咖啡酰奎尼酸类化合物总量分别为:茎尖9.77~40.86 mg/g,成熟叶2.68~13.97 mg/g,茎干0.56~7.90 mg/g,块薯0.69~4.33 mg/g。[结论]甘薯茎尖和叶片可以作为绿原酸提取材料加于开发利用,其中紫甘薯系列优于非紫甘薯系列,试验中两个11ZY紫薯材料表现尤为突出。

以南方红豆杉的枝、叶、果实为材料,建立了高效液相色谱法同时测定6种紫杉烷类化合物(紫杉醇、三尖杉宁碱、7–表–10–去乙酰紫杉醇、7–表–紫杉醇、巴卡亭III、10–去乙酰巴卡亭III)含量的方法。结果表明:以甲醇–乙腈–水(体积比为25∶36∶39)为流动相,等度洗脱,检测波长227 nm,6种紫杉烷类化合物在35 min内完全分离;南方红豆杉的枝、叶、果实中6种紫杉烷类化合物含量存在差异,其中,紫杉醇和7–表–10–去乙酰紫杉醇在种胚中含量最高,分别为866.47、722.50 μg/g,10–去乙酰巴卡亭III、巴卡亭III和7–表–紫杉醇在叶中的含量最高,分别为301.20、234.08、11.74 μg/g,三尖杉宁碱含量最高的部位是枝,为392.69 μg/g。

The xyloglucan endotransglycosylase gene RcXET of China Rose (Rosa chinensis Jacq.) and the MtDREB1C gene of Medicago truncatula Gaertn. were pyramided into the plant expression vector pBin438 and transformed into China Rose. Southern blot and Northern blot analyses showed that the heterologous gene MtDREB1C was integrated into the genome of surviving transgenic rose plants and expressed at different levels. Real-time PCR analysis demonstrated that robust expression of the congenetic gene RcXET was activated in the five surviving transgenic rose plants. The performance of the five transgenic lines under freezing and drought stress was superior to that of non-transformed controls. Thus, pyramiding of the genes MtDREB1C and RcXET in China Rose was more effective to enhance freezing and drought tolerance than untransformed controls. A positive correlation was observed between the expression of RcXET and the growth rate in contrast to the non-transgenic plants. The physiological assay showed that co-expression had greater effects on EC%, contents of proline, soluble sugar, photosynthesis rate, negative water potential, and turgor loss point than activities of POD and SOD under stress. The study also highlights the utility of a simple and rapid approach to express two or even more genes in one expression vector.

摘要:
With the aim of developing an effective protocol for virus elimination from potato (Solanum tuberosum L.) plantlets, thermotherapy coupled with isolating the first nodal cuttings by in vitro culture was successful to potato virus A (PVA) elimination. The survival ratio of potato plantlets was affective by thermotherapy temperatures and durations. The optimal thermotherapy temperature was 36±1 oC with highest survival ratio and effective elimination. The results of RT-PCR indicated that the regenerated plantlets obtained from the first cycle (four weeks) of thermotherapy in daytime at 36±1 oC with light intensity 40 μmole/m/s for 12 hr, and 20±1 oC in darkness for 12 hr had PVA infected. While isolated the first nodal cuttings and followed by thermotherapy at the first cycle conditions for another two weeks, the PVA could be eliminated. Thermotherapy was given by culturing the nodal cutting from the infected of PVA for six weeks in total on MS medium, and the PVA-free plantlets were obtained. In concluded that the protocol of thermotherapy coupled with isolating the first nodal cuttings by in vitro culture in the study can be effectively used for virus free plantlets in potato, and probably also for other vegetable propagated plant species.

摘要:
在植物病害研究中,观察寄主植物被病原菌入侵的过程非常重要。Solophenyl Flavine 7GFE染料可附着于菌丝,在波长为330~380 nm的激发光下被激发出蓝色荧光。为了更好的观察寄主植物中病原真菌的侵染情况,本实验以Solophenyl Flavine 7GFE为染剂,对寄主植物中病原真菌的侵染情况进行了观察研究。结果显示,用0.002%(M/V)Solophenyl Flavine 7GFE溶于0.1 mol/L Tris-HCl (pH 8.5)配制的染色液染色5 min的效果最佳;使用95%乙醇溶液替代0.15%三氯乙酸(W/V)酒精溶液∶氯仿(V/V)(4∶1)对寄主植物叶片脱色的方法操作简便、毒害较低;染色时省略了番红预染步骤。将改良的染色方法用于晚疫病菌入侵的马铃薯叶片观察取得了良好的效果。该技术是一种改良的、快速有效、安全无毒的观察真菌菌丝入侵植物的荧光染色方法,也适用于观察其他真菌入侵寄主植物组织的过程。

Obtainment and characterization of the novel endosperm callus of Taxus chinensis Rehd. var. mairei are valuable for haploid breeding, genome, and functional genome in Taxus. Callus was obtained by hydropriming with sterile water for 3 days and suitable medium composition. The highest callus induction (70.89 %) and lower browning ratio (7.95 %) were obtained from Gamborg (B5) medium supplemented with 30 g l−1 of sucrose, 2.5 mg l−1 of 2,4-dichlorophenoxyacetic (2,4-D), 0.5 mg l−1 of 6-benzylademine (6-BA) and 7 g l−1 of agar under dark conditions. The auxin of 2,4-D had a better efficiency of callus induction than naphthylacetic acid, and over 1 mg l−1 of 6-BA was inhibitory to the callogensis of endosperm. The endosperm callus was haploid which was detectable by the flow cytometry. The genome block of homozygosity of callus was homozygous which was indicated by PCR-based SNP marks. The homozygous haploid of endosperm callus in vitro culture may be useful tools for taxoid-metabolism of gene engineering and bio-fermentation engineering. © 2016, The Author(s).

The complete genome comprising three genomic RNAs of three Canadian and two Chinese isolates of Potato mop-top virus were sequenced and analysed. Two open reading frames (ORFs) were found in RNA1 of 6.1 kb, encoding a readthrough RNA-dependent RNA polymerase (RdRp). A coat protein (CP)-readthrough protein was encoded by RNA2 of 3.1 kb. Four ORFs that encoded the triple gene block proteins (TGBps) and a cysteine-rich protein were found in RNA3 (2.9 kb) of the Chinese isolate ‘Yunnan’; whereas in the remaining isolates (three Canadian isolates and the Chinese isolate ‘Guangdong’), only three ORFs encoding TGBps were observed in RNA3. A single nucleotide mutation of A2462 to G2462 abolished the start codon ‘AUG’ for the fourth putative ORF in RNA3 of these isolates. Based on phylogenetic and sequence similarity analysis of these isolates as well as those reported by others at the complete RNA sequence level, each of RNA1, RNA2 and RNA3 could be divided into at least two groups. In Canadian isolates ‘Ch9’, ‘Ch10’ and ‘Ch20’ and Chinese isolate ‘Guangdong’, all genomic RNAs belonged to group A; and in Chinese isolate ‘Yunnan’, all of its RNA belonged to group B. Interestingly, in Swedish isolate ‘Sw’, RNA1 and RNA2 belonged to group A while RNA3 belonged to group B. A duplex RT-PCR for differentiating groups A and B of RNA3 was developed and evaluated. All PMTV samples collected in Guangdong, China, and New Brunswick, Canada, possessed a RNA3 belonging to group A; whereas the samples collected in Yunnan, China, possessed a RNA3 belonging to group B. 摘要:
The water environmental capacity is not only the basic foundation in objective management of water environment, but also the constraint condition for the planning of water environment and its surrounding environment. Dongjiang Lake is the location of strategic water resources reserves in Xiangjiang River Basin urban agglomeration, and the Dongjiang Lake Scenic Area is a national scenic area that combines the ecological and tourism functions. Based on the statistics of water environment pollution in Dongjiang Lake Scenic Area, this paper conducts quantitative research and analysis of its water environmental capacity, hoping to conduct phased control of target volume of pollution load into Dongjiang Lake Scenic Area and provide data support to various follow-up special plans in the scenic area.

本文在冬闲稻田研究了农民种植模式(FP)、高产种植模式(H)和超高产种植模式(SH)对冬种马铃薯生长、产量、品质和植株氮磷钾积累、氮素利用效率的影响。结果表明,SH模式株高最高,主茎数最多,单位面积的生物量积累最大,叶面积指数最大,植株积累的氮、磷、钾最多;各处理各部位氮、钾含量差异较大,叶片中最高,块茎中最低,磷含量差异较小;SH模式产量最高,为2 936 kg/667 m~2,其次为H模式,S

摘要:
以两个耐热性差异较大的多年生黑麦草品种首相(Premier,耐热性强)和顶峰(Pinnacle,耐热性弱)为试验材料,分析了人工模拟昼夜温度为适温(25 ℃/20 ℃)和高温(35 ℃/30 ℃)两种温度条件下,高温胁迫对黑麦草生长、叶片细胞膜稳定性、叶绿素含量、光合作用、内源激素(ABA、IAA,GA_3和ZR)以及碳水化合物(可溶性糖、淀粉、果聚糖)代谢的影响。结果表明,高温胁迫条件下黑麦草生长减缓,叶片电导率升高,叶绿素含量和光合作用显著下降,内源激素脱落酸(ABA)含量升高,而生长素(IAA)、赤霉素(GA_3)和细胞分裂素(ZR)含量均下降,但是在耐热性强的品种中的变化程度均小于耐热性弱的品种;高温胁迫下碳水化合物中的可溶性糖含量随处理时间的延长呈先升高随后下降的趋势,淀粉含量在两个品种中均呈不同程度的下降,果聚糖含量在耐热性强的品种中升高,在耐热性弱的品种中先升高随后下降。高温胁迫下黑麦草叶片中积累或维持较高的内源激素(ABA、GA_3、IAA、ZR)和碳水化合物(可溶性糖和果聚糖)可能是耐热性较强的主要原因,这些内源激素或碳水化合物的积累或维持有助于黑麦草细胞内渗透平衡和细胞膜稳定性,保护光合器官免受损害以维持光合能力,延缓因高温和呼吸增强导致的叶片的枯黄衰老,从而提高了黑麦草的耐高温能力。

研制一种2-2型圆管压电复合材料。将压电陶瓷圆管沿轴向方向均匀切割,把环氧树脂浇注于切槽,经打磨和蒸镀电极,制成2-2型压电复合材料圆管。对压电复合材料圆管的压电和介电性能进行测试,结果为谐振频率388 k Hz,带宽11.2 k Hz,声阻抗18.05 Pa·s/m^3,相对介电常数859,声速3 200 m/s,d33常数480 p C/N,振动位移89.5 pm。该压电复合材料圆管适合做水平全向宽带换能器。
