摘要:
Muscle degeneration and cachexia caused by aging and chronic diseases seriously reduce the quality of life of patients. Catechins in tea enhance skeletal muscle performance, but the specific function and mechanism require further exploration. Myogenic differentiation is accompanied by extensive changes in cell morphology and gene expression patterns. In this study, we measured the number, length, diameter, and transcriptional levels of MyoD, MyoG, and MyHC of myotubes after C2C12 cell differentiation to assess changes in response to four different catechin monomers: epicatechin, epigallocatechin, epicatechin gallate (ECG), and epigallocatechin gallate. The differentiation-promoting effect of ECG was the strongest. Atomic force microscopy showed that ECG significantly reduced the adhesion force, stiffness, and Young's modulus of myotubes. Finally, the mechanism of ECG-enhanced C2C12 cell differentiation was explored by a LC/MS whole proteomic strategy and Ingenuity Pathway Analysis. The results provide a powerful basis for further study of catechin-enhanced skeletal muscle regeneration and repair.

摘要:
Epigallocatechin-3-gallate (EGCG) and caffeine in tea exert anti-obesity effects and induces nonalcoholic fatty liver disease (NAFLD) amelioration. However, previous studies usually performed a high-dose EGCG administration, whereas the insecurity was arisen in recent researches. In this study, we treated obese rats with an elaborate dose-40 mg/kg EGCG, 20 mg/kg caffeine, and the coadministration of them as low dose, which were similar to the daily intake; 160 mg/kg EGCG as high dose, which was the maximum safe dose had touched the contentious edge. The results suggested that the coadministration of EGCG and caffeine exerted more remarkable function on suppressing body weight gain, reducing white adipose tissue weight and decreasing the energy intake than single use. This may be due to the variation in serum lipid profile, oxidative stress, and adipose-derived and inflammatory cytokines. The pathological micrographs showed long-term high-fat diets caused severe NAFLD, but it was ameliorated at different levels by all of the administrations. In summary, low dose of EGCG or caffeine only showed a mild effect of anti-obesity and NAFLD amelioration. The coadministration of them could exert a superior curative effect as well as high dose EGCG but no anxiety regarding safety.

摘要:
Primary dark tea is used as raw material for compressed dark tea, such as Fu brick tea, Hei brick tea, Hua brick tea, and Qianliang tea. Pile-fermentation is the key process for the formation of the characteristic properties of primary dark tea, during which the microorganism plays an important role. In this study, the changes of major chemical compounds, enzyme activities, microbial diversity, and their correlations were explored during the pile-fermentation process. Our chemical and enzymatic analysis showed that the contents of the major compounds were decreased, while the activities of polyphenol oxidase, cellulase, and pectinase were increased during this process, except peroxidase activity that could not be generated from microbial communities in primary dark tea. The genera Cyberlindnera, Aspergillus, Uwebraunia, and Unclassified Pleosporales of fungus and Klebsiella, Lactobacillus of bacteria were predominant in the early stage of the process, but only Cyberlindnera and Klebsiella were still dominated in the late stage and maintained a relatively constant until the end of the process. The amino acid was identified as the important abiotic factor in shaping the microbial community structure of primary dark tea ecosystem. Network analysis revealed that the microbial taxa were grouped into five modules and seven keystone taxa were identified. Most of the dominant genera were mainly distributed into module III, which indicated that this module was important for the pile-fermentation process of primary dark tea. In addition, bidirectional orthogonal partial least squares (O2PLS) analysis revealed that the fungi made more contributions to the formation of the characteristic properties of primary dark tea than bacteria during the pile-fermentation process. Furthermore, 10 microbial genera including Cyberlindnera, Aspergillus, Eurotium, Uwebraunia, Debaryomyces, Lophiostoma, Peltaster, Klebsiella, Aurantimonas, and Methylobacterium were identified as core functional genera for the pile-fermentation of primary dark tea. This study provides useful information for improving our understanding on the formation mechanism of the characteristic properties of primary dark tea during the pile-fermentation process. © 2018 Li, Chai, Li, Huang, Luo, Xiao and Liu.

选取茶叶中三种主要生物碱咖啡碱(Caffeine)、可可碱(Theobromine)、茶碱(Theophylline)作为构建单体-冠突散囊菌共培养液态发酵体系(10 d)的添加底物,考察了冠突散囊菌对茶叶中主要生物碱的发酵特性。结果表明,与对照组相比,咖啡碱(0.2mg/mL)的添加可刺激冠突散囊菌的生长与繁殖,发酵液中葡萄糖的含量下降更为迅速,pH值随着发酵时间的增加持续下降直至第9 d趋于稳定,可溶性蛋白含量呈现先增加后下降的趋势,但在发酵过程中,冠突散囊菌既不能将其作为碳源、氮源消耗来维持生长,也不能代谢分解咖啡碱;可可碱与茶碱(0.08 mg/mL)在发酵体系中的发酵情况与咖啡碱基本相似,两者均能刺激冠突散囊菌的生长与繁殖,但均不能将其作为碳、氮源消耗,且发酵结束后在各自发酵体系中分别检测到0.00091 mg/mL、0.00109 mg/mL的咖啡碱,这说明冠突散囊菌可能具有以可可碱、茶碱作为前体合成咖啡碱的能力。

The green tea polyphenol epigallocatechin-3-gallate (EGCG) is widely consumed as a dietary supplement. Its potential properties include slowing aging and extending lifespan, although how exactly this is achieved remains unclear. Here, we report that EGCG promoted healthy lifespan in Caenorhabditis elegans when administered throughout or only at early-to-mid adulthood. Specifically, EGCG extended lifespan in an inverted U-shaped dose-response manner. The life-extending mechanism was stimulated by EGCG-induced production of reactive oxygen species (ROS). Additionally, EGCG triggered mitochondrial biogenesis to restore mitochondrial function. The EGCG-induced increase in lifespan depends on known energy sensors such as AMPK/AAK-2, as well as SIRT1/SIR-2.1 and FOXO/DAF-16. Interestingly, aging decreased the response to EGCG and progressively neutralized its beneficial effects on longevity. Collectively, our findings link EGCG to the process of mitohormesis and suggest an inducible, AMPK/SIRT1/FOXO-dependent redox signaling module that could be invoked in different contexts to extend healthy lifespan. Its effectiveness is higher in younger adults and declines with age.

对不同年份的27个普洱生茶样品按照国家审评标准从外形、内质对其进行感官评价,并对其汤色的色差值进行测定。结果表明,随着陈化年份的延长,普洱生茶茶饼不同程度地出现疏松,茶叶条索逐渐松散,干茶色泽呈黄绿—褐黄—黄褐—棕褐的变化趋势;茶汤色泽逐渐变深,呈橙黄—黄橙—橙红—棕红的变化趋势;香气中的花香逐渐淡化,由带陈到陈香显露;滋味中的收敛性和粗涩感逐步减弱;色差值a-*值、b-*值增大,茶汤颜色变红变深。普洱生茶感官品质随储藏年份的增加发生一定趋势的变化,但变化的程度及方向受到原料、加工工艺、贮藏环境等影响。试验通过对跨度近20年的不同厂家普洱生茶感官品质进行探讨,为进一步了解普洱茶的陈化过程中品质变化规律提供参考。

Dark tea has a significant effect on the prevention and treatment of age-related degenerative diseases. At present, further exploration of its functional mechanisms is delayed because of the complexity of post-fermentation microbial metabolites during the production phase. In this study, new isolated microbial metabolites extracted from dark tea were used to explore the neuroprotective effects, and they also helped allow further exploration of the mechanism of dark tea. Taking senescence-accelerated mouse prone 8 (SAMP8) mice as a biological model, we examined the protective effect on brain neurons of post-fermentation microbial metabolites which were extracted from dark tea. The 4-month-old mice were given treatments of the same concentration (10 mg kg−1 d−1) which were l-theanine, 3,3′-azanediylbis(4-hydroxybenzoic acid) (CDT-1) and one of the 8-C N-ethyl-2-pyrrolidinone substituted flavan-3-ols (CDT-2) by gavage for 14 weeks. Relative measurements such as RT-PCR, ELISA, western blotting, and section staining (HE, Nissl and myelin) were carried out. The results showed that l-theanine, CDT-1 and CDT-2 could inhibit the decrease in body weight, and down-regulate the formation of 4-HNE and ubiquitinated protein aggregates and the Aβ metabolic pathway. They could also increase endogenous antioxidant capacity, relieve cell hypoxia, and reduce the rate of neuronal apoptosis. This means that their protective activity regarding SAMP8 neurons was excellent and the activity of CDT-2 was the most significant. © The Royal Society of Chemistry.

Here we identified that BTE (black tea extract), within the studied concentration range, is more effective than GTE (green tea extract) in protecting C. elegans against hypertonic stress, by enhancing survival after exposure to various salts, and alleviating suffered motility loss and body shrinkage. The mechanism of such protection may be due to the ability of black tea to induce the conserved WNK/GCK signaling pathway and down-regulation of the expression levels of nlp-29. Intriguingly, black tea does not relieve hypertonicity-induced protein damage. The findings implicate the potential health benefits of black tea consumed worldwide. © The Royal Society of Chemistry.

以大红袍、红茶与茯砖茶原料为供试原材料,利用超高效液相色谱电喷雾四级杆飞行时间串联质谱(UPLC-ESI-Q-TOF-MS)技术的代谢组学对不同茶叶原料茯茶的“发花”前后样品的整体成分组成差异进行了分析.结果表明,不同原料“发花”前、后间差异成分数量均有不同,各组差异代谢化合物个数分别为大红袍49个、红茶40个、茯砖茶原料49个;综合3组茶叶的具体差异代谢化合物,发现其从类别上表现出相似的变化规律,即酯型儿茶素下降的极为显著,而简单儿茶素除没食子儿茶素(GC)变化无明显差异外,表儿茶素(EC)、儿茶素(DL-C)、表没食子儿茶素(EGC)的含量较原料都有所升高,原花青素和咖啡碱、可可碱、茶碱、黄嘌呤与次黄嘌呤也有相同的变化;黄酮类化合物槲皮素、山奈酚、牡荆素和芦丁的含量均有不同程度的升高;而大黄素则是在各原料“发花”后均新增的蒽醌类化合物.

摘要:
Tea O-methylated catechins, especially (-)-epigallocatechin 3-O-(3-O-methyl)gallate (EGCG3″Me), have been attracting much attention as a result of their positive health effects. The transcription regulators of O-methylated catechin biosynthesis remain elusive. In this study, the expression pattern of genes related to O-methylated catechin biosynthesis, including CsLAR, CsANS, CsDFR, CsANR, and CCoAOMT, in three tea cultivars with different contents of EGCG3″Me was investigated. Two WRKY transcription factors (TFs), designated as CsWRKY31 and CsWRKY48, belonging to groups IIb and IIc of the WRKY family, respectively, were further identified. CsWRKY31 and CsWRKY48 were nuclear-localized proteins and possessed transcriptional repression ability. Furthermore, expression of CsWRKY31 and CsWRKY48 showed negative correlation with CsLAR, CsDFR, and CCoAOMT during EGCG3″Me accumulation in tea leaves. More importantly, W-box (C/T)TGAC(T/C) elements were located in the promoter of CsLAR, CsDFR, and CCoAOMT, and further assays revealed that CsWRKY31 and CsWRKY48 were capable of repressing the transcription of CsLAR, CsDFR, and CCoAOMT via the attachment of their promoters to the W-box elements. Collectively, our findings identify two novel negative regulators of O-methylated catechin biosynthesis in tea plants, which might provide a potential strategy to breed high-quality tea cultivar. 代谢组学是系统生物学研究的重要组成部分,研究生物体在不同条件下所产生的小分子代谢产物的变化,从而揭示生物体的生理状态.近年来代谢组学技术在茶叶相关研究领域得到广泛应用,着重论述了代谢组学在茶叶品质(产地环境、采摘时间、加工工艺对品质的影响以及品质评价等)与药理(茶叶的降脂减肥、延缓衰老、抗辐射、生物利用度与生物转化等)等领域的应用现状,代谢组学技术将在茶叶品质形成及评价、茶叶功效等方面发挥不可替代的作用.

摘要:
Chemical vapour deposited graphene on Cu foil should be transferred to target dielectric substrate before being used in various applications. This route often involves preparation of a supporting material on graphene before the transfer process and removal of the material after graphene is transferred to a foreign substrate. Herein, we demonstrate a simple process combining graphene transfer and subsequent graphene-based sensor preparation in one route. By applying a thin flexible polyethylene (PE) film onto graphene and subsequently chemical etching Cu foil, this process enables the high-quality transfer of graphene on PE film. A flexible strain sensor prepared from the as-transferred graphene/PE film shows good detection sensitivity. Therefore, this process could greatly reduce the cost of transfer and device fabrication for mass production.

摘要:
以乌龙茶(大红袍)、红茶、绿茶、黑茶(天尖原料、金湘益茯砖茶原料)5种茶样为原料,运用“散茶发花”技术制得不同茶类散装茯茶,基于各成品茶中“金花”菌间存在一定的形态差异,运用微生物手段对各茯茶制品中“金花”菌进行差异分析,同时从形态与DNA序列分析角度对其进行了系统的鉴定,并运用高效液相色谱-三重四极杆串联质谱对其是否具备产黄曲霉毒素的能力进行检测分析。结果表明:不同茶类发花茯茶成品中的“金花”菌实属同一种真菌,无异于原接种供试“金花”菌,该菌被鉴定为真菌门(Eumycota;Mycobionta)、子囊菌纲(Ascomycetes)、真子囊菌亚纲(Euascomycetes)、曲霉目(Eurotiales)、曲霉科(Eurotiaceae)、散子囊菌属(Eurotium)、冠突散囊菌(Eurotium cristatum),无性型为针刺曲霉(Aspergillus spiculosus Blaser);此外,质谱数据显示未检测出黄曲霉毒素B1、黄曲霉毒素B2、黄曲霉毒素G1和黄曲霉毒素G2,表明该分离“金花”菌不具备产黄曲霉毒素的能力。

摘要:
为研究茶叶提取物表没食子儿茶素没食子酸酯(epigallocatechin gallate, EGCG)、没食子儿茶素没食子酸酯(gallocatechingallate, GCG)和表儿茶素没食子酸酯(epicatechingallate, ECG)对体外培养B16小鼠黑色素瘤细胞黑色素生成及酪氨酸酶活性的影响,分别用不同浓度的EGCG、GCG、ECG和熊果苷(Arbutin,AR)处理细胞,观察效应物对细胞形态的影响,用溴化二苯四偶氮法(MTT法)测定茶叶提取物对细胞增殖率的影响,以左旋多巴(L–DOPA)为底物,测定细胞内酪氨酸酶的活性,采用氢氧化钠裂解法测定细胞内黑色素的含量。结果显示,EGCG、 ECG、GCG能显著抑制B16细胞的黑色素生成和酪氨酸酶的活性,且呈剂量依赖关系;当浓度为60 μg/mL时,细胞增殖率均低于50%,酪氨酸酶活性与对照组相比分别降低了26.67%、27.27%和32.71%,黑色素生成抑制率均在30%以上。结果表明,茶叶提取物能通过多种途径抑制黑色素生成,且GCG的作用效果最优,ECG的作用效果次之,三者的作用效果均优于熊果苷的作用效果。

摘要:
The antibacterial effects of tea polyphenol epigallocatechin gallate (EGCG), a common phytochemical with a number of potential health benefits, are well known. However, the mechanism of its bactericidal action remains unclear. Using E. coli as a model organism, it is argued here that H2O2 synthesis by EGCG is not attributed to its inhibitory effects. In contrast, the bactericidal action of EGCG was a result of increased intracellular reactive oxygen species and blunted adaptive oxidative stress response in E. coli due to the co-administration of antioxidant N-acetylcysteine, and not on account of exogenous catalase. Furthermore, we noted a synergistic bactericidal effect for EGCG when combined with paraquat. However, under anaerobic conditions, the inhibitory effect of EGCG was prevented. In conclusion, EGCG caused an increase in endogenous oxidative stress in E. coli, thereby inhibiting its growth, and hence the use of EGCG as a prooxidant is supported by this study. © 2016 Elsevier Ltd

摘要:
建立低密度脂蛋白受体(Low density lipoprotein receptor,LDLR)激动剂药物筛选模型,并用该模型筛选出具有上调LDLR功能的黑茶品类。合成LDLR启动子片端(189bp),克隆入含有萤火虫荧光素梅报告基因的p GL3-basic载体中,构建了重组报告基因质粒p GL3-LDLR,以p RL-TK为内参质粒,瞬时转染人肝癌细胞Hep G2;以降脂药物辛伐他汀作为阳性对照物,优化反应条件,再应用该模型对8种黑茶水提取物进行了初步筛选。结果表明,辛伐他汀对荧光素酶的表达具有较强的诱导作用,且呈一定剂量的依赖性,Z′因子为0.72,表明该模型具有很好的灵敏性和稳定性,成功地建立了靶向LDLR转录活性的双报告基因筛选体系,并发现待测黑茶均有一定的激动活性,其中金花天成茶对模型的激活值最高,激活值达1.761。
