A method for separation and determination of chlorogenic acid and rutin in flue-cured tobacco leaves by high performance liquid chromatography(HPLC) was developed. The sample was extracted by petroleum ethers whose boiling point ranged from 60 -90 ℃, and the mobile phase was potassium dihydrogen phosphate buffer solution and methanol on gradient elution, which make to violate and measure the chlorogenic acid and rutin.
