The Optimization of Methods for DNA Extraction of Total Microorganisms from Cow＇s Feces

采用试剂盒提取法、冻融-CTAB法、CTAB-SDSⅠ法与CTAB-SDSⅡ法四种方法提取牛粪便总DNA,并从纯度、颜色与提取量等方面进行评价分析.实验表明,CTAB-SDSⅡ法提取的总DNA颜色透明、DNA纯度（A260/A280）为1.930±0.433、DNA提取量（μg/g）为19.870±0.433.因此,CTAB-SDSⅡ法是一种比较适合牛粪便微生物总DNA的提取方法.

The metagenomics is an important approach to develop and utilize the uncultured microorganisms.Meanwhile,the quality of DNA from microorganisms is an important guarantee to construct the metagenomic library.Four different methods including DNA isolation kit method,freeze thawing-CTAB method,CTAB-SDSⅠ method and CTAB-SDSⅡ method were used to extract DNA from cow＇s feces.Through the analysis of the purity,color and amount of DNA extraction,the results of CTAB-SDSⅡ method were as follows：the color was clear,the purity（A260/A280）of DNA was 1...
