Ligand exchange chromatography with chiral mobile phases was established for the separation of theanine enantiomers.The optimum condition was :Polaris C18 column, mobile phase: L-proline: Cu^2+=2:1, the concentration of Cu^2+ was 0.5 mmol/L, pH6.8, the volume of methanol is 2%, wavelength: 254 nm;flow rate:0.9 ml/min, column temperature:30℃.
