巴克夏猪生长发育性能测定及其相关分析

摘要:
猪只可通过品种改良而提高生长发育性能,进而提高养猪业的经济效益.巴克夏猪源于英国巴克夏郡,至今已有300多年的历史.巴克夏猪盛产雪花瘦肉,特别是肩背部的瘦肉,被称之为"极品猪肉".以我国肉质优良,色、香、味具佳的地方品种猪为母本,以巴克夏猪为父本,生产优质黑猪肉产品具有广阔的市场前景.巴克夏猪的生长发育性状是中等遗传力性状,受遗传因素影响较大,同时也受营养水平和环境较大的影响.该试验测定了巴克夏猪的主要生长发育性能,并进行了相关分析.通过对55头巴克夏猪的体长、胸围、腿臀围、体高、背膘厚和体重等6项生长发育性状的测定,计算出各性状之间的相关系数,再进行显著性检测并进行通径分析和相关系数的剖分,分析生长发育性状间的相关性,厘清生长发育性状间的真正关系,为巴克夏猪的选育及生产提供参考,并提出提高巴克夏猪生长发育性能的措施.

摘要:
A large number of microRNAs (miRNAs) have been detected from porcine testicular tissues thanks to the development of high-throughput sequencing technology. However, the regulatory roles of most identified miRNAs in swine testicular development or spermatogenesis are poorly understood. In our previous study, ULK2 (uncoordinated-51-like kinase 2) was predicted as a target gene of miR-26a. In this study, we aimed to investigate the role of miR-26a in swine Sertoli cell autophagy. The relative expression of miR-26a and ULK2 levels has a significant negative correlation (R2 =.5964, p ≤.01) in nine developmental stages of swine testicular tissue. Dual-luciferase reporter assay results show that miR-26a directly targets the 3′UTR of the ULK2 gene (position 618–624). In addition, both the mRNA and protein expression of ULK2 were downregulated by miR-26a in swine Sertoli cells. These results indicate that miR-26a targets the ULK2 gene and downregulates its expression in swine Sertoli cells. Based on the expression of marker genes (LC3, p62 and Beclin-1), overexpression of miR-26a or knock-down of ULK2 inhibits swine Sertoli cell autophagy. Taken together, these findings demonstrate that miR-26a suppresses autophagy in swine Sertoli cells by targeting ULK2. © 2018 Blackwell Verlag GmbH

摘要:
A comprehensive and systematic understanding of the roles of lncRNAs in the postnatal development of the pig testis has still not been achieved. In the present study, we obtained more than one billion clean reads and identified 15,528 lncRNA transcripts; these transcripts included 5032 known and 10,496 novel porcine lncRNA transcripts and corresponded to 10,041 lncRNA genes. Pairwise comparisons identified 449 known and 324 novel lncRNAs that showed differential expression patterns. GO and KEGG pathway enrichment analyses revealed that the targeted genes were involved in metabolic pathways regulating testis development and spermatogenesis, such as the TGF-beta pathway, the PI3K-Akt pathway, the Wnt/β-catenin pathway, and the AMPK pathway. Using this information, we predicted some lncRNAs and coding gene pairs were predicted that may function in testis development and spermatogenesis; these are listed in detail. This study has provided the most comprehensive catalog to date of lncRNAs in the postnatal pig testis and will aid our understanding of their functional roles in testis development and spermatogenesis. © 2017

microRNAs (miRNAs) and PIWI-interacting RNAs (piRNAs) execute important regulatory roles in testis development and spermatogenesis, while previous studies mainly focus on the expression profiles in immature and mature porcine testes, which may cause a bottleneck for further understanding their complex physiological processes in porcine testes development and spermatogenesis. Thus, we presented the expression and characterization of miRNAs and piRNAs in DS (60-day-old), DN (90-day-old), DT (120-day-old) and DF (150-day-old) pig testes. In total, 12,834,628, 13,359,726, 12,851,249 and 12,938,601 clean reads were generated from these libraries, respectively. 293 mature and 36 novel miRNAs as well as 4923 piRNA clusters were identified from pig testes, and they showed an age-dependent manner. GO enrichment analysis of miRNA target genes and piRNA generated genes showed that they participated widely in regulating the pig spermatogenesis process. In addition, 12 differentially expressed miRNAs were randomly selected to validate using qRT-PCR. Our results provided novel comprehensive expression profiles of miRNAs and piRNAs in pig testes at different stages of sexual maturity, which will promote our knowledge of them in regulating the pig testes development and spermatogenesis process. © 2017, The Genetics Society of Korea and Springer Science+Business Media B.V.

Testicular development and spermatogenesis are strictly regulated with the complex interactions between several cell types in testis. However, the limited availability of genomic and molecular information limits our understanding of this complex physiological process. In this study, we characterized the transcriptome between immature (30-day-old) and mature (180-day-old) pig testes using RNA-seq technology. 24,469 known coding gene transcripts corresponding to 20,566 genes were mapped in these two developmental stages, 3,328 genes were differentially expressed, and numerous novel transcripts and alternative splicing events were also identified. Ten differentially expressed genes were validated by measuring the relative expression using real-time quantitative polymerase chain reaction (PCR). 125 Gene Ontology (GO) terms were significantly enriched, and most of them involved in GO terms related to male reproduction, testicular development, and spermatogenesis. In addition, this study also represented the prediction of 10,000 circRNAs, as well as the validation of six pig circRNAs using Find_cric algorithm. Our study substantially expanded our knowledge about the transcriptomic profile of immature and mature pig testes, and provided a useful resource to study the mechanisms of pig testis development and spermatogenesis at the molecular level. © 2017, The Genetics Society of Korea and Springer Science+Business Media B.V.

采用RACE技术克隆猪GSG1(Germ Cell-specific gene l)基因cDNA1全长并进行生物信息学分析,利用实时荧光定量PCR对GSG1基因在沙子岭猪睾丸组织8个不同发育阶段(E90、D1、D30、D60、D90、D120、D150和D180)和30日龄时9个不同组织(睾丸、肌肉、小肠、肺、心脏、肾脏、肝脏、脾脏和脂肪)的表达量.结果表明:GSG1基因cDNA全长为1 257 bp,含1个长993 bp的开放阅读框,编码330个氨基酸.实时荧光定量PCR结果显示,GSG1基因在睾丸组织中的表达量最高,显著高于其余8个组织(P<0.05),且其在肌肉、小肠、肺脏、心脏、肾脏和脂肪中的表达水平极低;GSG1基因在睾丸组织的E90、D1、D30和D60这4个发育时期的表达量较低,显著低于D90、D120、D150和D180这4个时期(P<0.01).本试验首次克隆了猪GSG1基因cDNA全长并检测了其在不同组织和不同发育阶段睾丸组织的表达,为进一步研究其调控猪睾丸发育和精子生成的机制奠定了理论基础.

miR-221/222基因家族进化分析及其在猪睾丸组织中的表达变化

运用生物信息学方法在miRBase中搜索动物miR-221/222基因家族的序列,利用Ensembl数据库信息确定miR-221/222在基因组中的位置,采用MEGA 5.0构建miR-221/222系统进化树;并利用RT-qPCR技术定量检测猪睾丸组织中不同发育时期miR-221/222的表达变化.结果表明:在34种动物中共搜索到59条miR-221/222序列,所有的miR-221/222基因都存在于基因间隔区.进化分析表明,各物种miR-221/222基因家族可能以不同的方式和速度进化而来.RT-qPCR检测结果显示,miR-221/222在猪睾丸组织中不同发育时期表现出相同趋势的表达规律,即出生后其表达水平先升高,生长发育时期降低,随着性成熟又呈现升高趋势.研究结果为深入理解miR-221/222基因家族的作用机制奠定基础,也为后续研究提供参考.

摘要:
【目的】采用实时荧光定量PCR技术(qRT-PCR)对基因表达进行分析时,选择适当的内参基因是获得准确分析结果的关键。在猪睾丸分子生物学研究中,表达稳定的蛋白编码内参基因和microRNA (miRNA)内参基因均有哪些目前尚不清楚。本研究旨在筛选出合适的内参基因,为准确定量不同时期猪睾丸组织中目的基因的表达提供可靠依据。【方法】选用8个不同发育时期(E90、 Dl、D 30、 D 60、D 90、D 120、D 150、 DM)的猪睾丸组织为材料,采用Trizol裂解法提取各样品的总RNA,利用NanDrop ND-2000分光光度计检测总RNA的浓度和纯度。设计并合成已报道的猪其他组织中表达相对稳定的5个编码内参基因(GAPDH、TBP、 β -actin. SDHA和B2M)以及5个miRNA内参基因(U6、ssc-miR-17-5p、 ssc-miR-26a、ssc-miR-27a和ssc-miR-103)的特异性引物序列,逆转录得到cDNA模板后,按1:10梯度稀释为7个浓度梯度进行qRT-PCR反应以构建标准曲线。并对10个候选内参基因在各时期睾丸组织中的表达情况进行实时荧光定量全面检测,采用GeNorm法对定量结果进行综合分析,最后根据内参基因稳定性值(M值)的大小筛选出最稳定的参考基因;M值越小,表明内参基因的稳定性越好,反之则越差。【结果】对GAPDH、TBP、β-actin、 SDHA、B2M、 U6、ssc-miR-17-5p、ssc-miR-26a、ssc-miR-27a和ssc-miR-103 10个候选内参基因的熔解曲线进行分析发现,均无非特异扩增及引物二聚体,说明各内参基因特异性良好,qRT-PCR反应的专一性高;以Ct值为纵坐标,相对拷贝数的对数为横坐标进行标准曲线分析可知,各内参基因在系列稀释的浓度梯度内具有良好的线性关系;通过对10个候选内参基因在不同时期猪睾丸组织中的表达稳定性分析发现,蛋白编码基因中,最稳定的为TBP,最不稳定的是GAPDH; miRNA候选内参中,最稳定的为U6,最不稳定的是ssc-miR-26a。【结论】成功筛选出猪睾丸组织基因表达分析中稳定表达的内参基因TBP和U6,可作为猪睾丸组织基因表达研究中最佳内参基因候选者。

摘要:
利用实时荧光定量PCR(Real-time PCR)检测miR-126-5p在沙子岭猪睾丸组织8个不同发育时期(E90、D1、D30 、D60、D90、D120、D150、D180)表达量,以了解其在动物睾丸发育及精子生成中的作用.利用miRanda、TargetScan和mirWalk 3个数据库预测miR-126-5p靶基因并取交集,利用David在线软件分析miR-126-5p靶基因的生物功能.结果表明,miR-126-5p在胚胎期(E90-D1)睾丸组织中高表达,而在生长期(D1-D90)表达量下调,但在沙子岭猪性成熟(D120)后表达量又上调,其中D1表达量最高,D90表达量最低,且二者与其他7个时期均差异极显著(P<0.01);D30、E90、D180相互之间差异不显著(P>0.05),且分别与其余各时期差异极显著(P<0.01);D120与D30差异显著(P<0.05),与其余各时期差异极显著(P<0.01);D150与D60之间差异不显著(P>0.05),二者分别与其余各时期差异极显著(P<0.01).miR-126-5p靶基因参与细胞增殖、凋亡、分化、细胞通讯、迁移及蛋白质转运等生物学过程(P<0.001);靶基因分子功能包括蛋白二聚体活性、配体依赖性核受体活性(P<0.001);KEGG分析显示miR-126-5p靶基因参与TGF-β、MAPK、细胞黏附等信号通路(P<0.01).结合miR-126-5p在睾丸组织不同发育时期表达差异结果及其靶基因功能预测结果,miR-126-5p可能参与沙子岭猪生殖细胞分化及精子生成调控,本研究为miR-126-5p在动物精子生中的作用及其调控机制的研究提供依据.

通过miRBase数据库检索miR-24基因家族序列,利用Ensembl数据库信息确定miR-24家族在不同物种中的基因组定位,采用MEGA 5.0构建miR-24基因家族的系统进化树,并利用RT-qPCR检测miR-24-1及miR-24-2在沙子岭猪睾丸组织不同发育期(E90、D1、D30、D60、D90、D120、D150、D180)的表达变化。在miRBase数据库检索32个物种共66条序列,且各序列均具有高度保守性。基因组定位显示大部分物种的miR-24基因位于常染色体上。RT-qPCR结果显示,miR-24基因家族2个成员(miR-24-1、miR-24-2)在沙子岭猪睾丸不同发育阶段呈现出协同表达的规律。本研究对miR-24基因家族进行系统进化分析并检测其在沙子岭猪睾丸组织中表达变化规律,为进一步探明miR-24基因家族的功能及作用机制奠定基础。

转化生长因子β(transforming growth factorβ,TGF-β)超家族是一类结构、功能相关的多肽,能够调节动物细胞的增殖、分化、凋亡,影响动物的胚胎发育、器官形成以及细胞外基质合成及稳态。此外,TGF-β超家族对胚胎原始生殖细胞形成和迁移及定植、胚胎性别分化和睾丸发育、生殖细胞增殖和静止及分化、动物出生后睾丸正常功能维持、生精干细胞自我维持更新、精子生成等过程发挥着重要的生理调控作用。本文综述了TGF-β超家族在动物睾丸发育和精子生成过程中的信号传递及调节机制,以期为后续的相关研究提供参考。

摘要:
Lantang pig is a native breed of Guangzhou Province in China. It is the first time that the complete mitochondrial genome sequence of Lantang pig is reported in this work, which is determined through the PCR-based method. The total length of the mitognome is 16,709 bp, which contains 2 ribosomal RNA genes, 22 tRNA genes, 13 PCGs and 1 conntrol region (D-loop region, Table 1). The total base composition of Lantang pig mitochondrial genome is 34.69% for A, 26.18% for C, 25.82% for T and 13.31% for G, in the order A>C>T>G. The complete mitochondrial genome of Lantang pig provides an important data in genetic mechanism and the evolution genomes. © 2014 Informa UK Ltd.

摘要:
Abstract Dahuabai pig is one of the most important indigenous breed of the Guangzhou province of China. It is the first time that the complete mitochondrial genome sequence of Dahuabai pig is reported in this work, which is determined through the PCR-based method. The total length of the mitognome is 16,709 bp, which contains a control region (D-loop region), 2 ribosomal RNA genes, 13 protein-coding genes and 22 tRNA genes. The total base composition of Dahuabai pig mitochondrial genome is 34.68% for A, 26.20% for C, 25.81% for T and 13.32% for G, in the order A > C > T > G. The complete mitochondrial genome of Dahuabai pig provides an important data in studying mitochondrial DNA's role in the process of metabolism and programmed cell death.

摘要:
Abstract Xiangdong black goat (Capra hircus) is a native breed of Hunan province in China. It is the first time that the complete mitochondrial genome sequence of Xiangdong black goat is reported in this work, which is determined through the PCR-based method. The total length of the mitognome is 16,641 bp, which contains 2 ribosomal RNA genes, 22 tRNA genes, 13 PCGs and 1 conntrol region (D-loop region). The total base composition of Lantang pig mitochondrial genome is 33.53% A, 13.12% G, 27.25% T and 26.09% C, in the order A > T > C > G. The complete mitochondrial genome of Xiangdong black goat provides an important data in genetic mechanism and the evolution genomes.

摘要:
In this study, we cloned and sequenced the complete mitochondrial genome DNA of Chinese pig, the Meishan pig. The sample was taken from Yencheng City, Jiangsu province in China. The complete genome DNA is 16 708 bp in length. We also performed a comparative analysis of the Meishan pig mitochondrial to the mitogenome sequences of 21 pig breeds which have been deposited in GenBank. Phylogenetic analysis using neighbour-joining computational algorithms showed that the analyzed species are divided into four major clades; the results can be subsequently used to provide information for pig phylogenetic and insights into the evolution of genomes.

摘要:
可变剪接作为一种增加基因组和蛋白质组多样性的重要机制, 广泛发生于真核生物基因转录的过程中.本文采用Illumina Hiseq 2500测序平台对猪睾丸组织4个不同发育时期(60胚龄、90胚龄、30日龄和180日龄)进行转录组测序, 以猪基因组数据为参考, 鉴定和分析了猪基因组可变剪接事件. 从猪基因组中鉴定出20398个基因(80.6%)对应的92738个可变剪接事件.在不同的可变剪接类型中, 以第一个外显子可变剪切(alternative 5′ first exon, TSS)、最后一个外显子可变剪切(alternative 3′ last exon, TTS)、单外显子跳跃(skipped exon, SKIP)和可变5′或3′端剪切(alternative exon ends, AE) 4种类型为主,分别占所有可变剪接事件的41.0%, 32.9%, 7.6%和7.4%. GO功能富集分析显示, 发生可变剪接的基因主要富集于物质合成、物质结合及酶活性相关的GO项中, 而各发育时期特异的可变剪接基因与发育时期的生理状态密切相关, 60胚龄时主要与酶活性和组织形成相关, 30日龄时主要与抗环境应激和离子通道活性相关, 180日龄时则主要与循环系统相关. 此外,在筛选出64个与睾丸素代谢相关的基因中, 63个基因发生可变剪接, 且以TSS和TTS为主,表明这两种可变剪接类型与睾丸素合成和分泌密切相关.通过对猪基因组可变剪接的分析,为深入研究可变剪接生物学功能及进一步开展分子育种工作提供理论依据.

摘要:
克隆猪胰岛素诱导1(INSIG1)基因,并对其蛋白质序列进行分析。根据人INSIG1基因的保守序列设计1对引物,以猪总RNA为模板,经RT–PCR获得INSIG1基因序列,通过相对荧光定量PCR分析该基因在不同组织中的表达量;将INSIG1基因序列连接到pMD19–T Simple载体上,用PCR检测阳性克隆后测序,并对克隆得到的基因进行生物信息学分析。结果表明,INSIG1基因在肺中的表达量最高,其次是在肝脏,再次是在脾脏,在心脏中的表达量最低;通过克隆,获得了一段999 bp的序列,其中831 bp的开放阅读框编码276个氨基酸;该蛋白不含信号肽序列,与其他动物INSIG1基因氨基酸序列的一致性在71%以上。

Luchuan pig is one of the famous native breeds in China. The complete mitochondrial genome of Luchuan pig provides an important data in genetic mechanism and the evolution genomes.
