摘要:
An amperometric immunosensor for phytohormone abscisic acid was developed based on in situ chemical reductive growth of gold nanoparticles on glassy carbon electrode. First, an approximate 10 nm gold layer was sputtered uniformly onto the electrode surface, and then gold nanoparticles were grown directly on the gold layer for antibody adsorption by immersing the electrode into the H2AuCl4 solution. Determination was based on an enzyme-linked competitive immunoreaction between free and enzyme-labeled abscisic acid to bind on immobilized antibody on electrode. The linear response was from 10 ng/ml to 10 μg/ml with a detection limit of 5 ng/ml. © Taylor & Francis Group, LLC.

摘要:
Treatment of potato (Solanum tuberosum L.) with chlorocholine chloride (CCC) applied twice as a foliar spray 25 and 30 days after planting has shown to decrease shoot and stolon growth but increase tuber yield. However, the regulatory role of CCC on translocation of recently fixed photoassimilates into different parts of potato plants has not been fully illustrated. In this study, C-14-isotope labelling technique was used to estimate the photosynthetic capacity and photoassimilate partitioning among leaves, stems, roots + stolons, and tubers of potted potatoes treated with 1.5 g l(-1) CCC. CCC treatment significantly increased tuber dry mass but reduced leaf dry mass. CCC-treated leaves had significantly higher chlorophyll and carotenoid contents and assimilated 22.0% more (CO2)-C-14 per leaf dry mass than the controls. Compared with the control, CCC treatment reduced the translocation of C-14-photoassimilates into leaves, steins and roots + stolons but increased that into tubers. CCC-treated leaves exported 14.6% more C-14-photoassimilates into other parts of the plants. In addition, CCC treatment reduced C-14-Soluble sugarand C-14-starch accumulation in leaves and stems but enhanced them in tubers and roots + stolons. Collectively, the results indicate that CCC treatment significantly improves the photosynthetic capacity of potato leaves and promotes photoassimilates partitioning into tubers thereby enhancing tuber growth. 摘要:
Effects of chlorocholine chloride (CCC) on phytohormones and photosynthetic characteristics of Zhongshu 3, a potato (Solanum tuberosum L.) variety widely cultivated in south China, were studied by foliar CCC application on 24 and 28 days after emergence, that is, at the tuber initiation stage. It was found that on 42 days after emergence, that is, at the tuber bulking stage, spraying CCC increased indolacetic-3-acid (IAA) and zeatin (Z) contents but decreased abscisic acid (ABA) content in leaves. The content ratios of IAA/Z, IAA/ABA, Z/ABA, and (IAA + Z)/ABA in leaves treated with CCC were higher than those of the control. CCC plays a prominent regulating role in the photosynthesis of Zhongshu 3. The net photosynthetic rate (P
n), stomatal conductance (G
s), intercellular CO2 concentration (C
i), and transpiration rate (T
r) of treated leaves were superior to those of controls at the tuber bulking stage. CCC markedly increased tuber yield and quality. The contents of sucrose and starch in tubers treated with CCC increased at the end of the vegetation period, whereas the contents of reducing sugars and solanine decreased. CCC at 2.0 g L−1 was found to be the most effective concentration. Collectively, the results of this research identify phytohomonal metabolism and photosynthetic physiology of potato leaves as processes affected early after application of CCC resulting in significantly improved increases in tuber yield and quality.

摘要:
Precise coordination between stamen and pistil development is essential to make a fertile flower. Mutations impairing stamen filament elongation, pollen maturation, or anther dehiscence will cause male sterility. Deficiency in plant hormone gibberellin (GA) causes male sterility due to accumulation of DELLA proteins, and GA triggers DELLA degradation to promote stamen development. Deficiency in plant hormone jasmonate (JA) also causes male sterility. However, little is known about the relationship between GA and JA in controlling stamen development. Here, we show that MYB21, MYB24, and MYB57 are GA-dependent stamen-enriched genes. Loss-of-function of two DELLAs RGA and RGL2 restores the expression of these three MYB genes together with restoration of stamen filament growth in GA-deficient plants. Genetic analysis showed that the myb21-t1 myb24-t1 myb57-t1 triple mutant confers a short stamen phenotype leading to male sterility. Further genetic and molecular studies demonstrate that GA suppresses DELLAs to mobilize the expression of the key JA biosynthesis gene DAD1, and this is consistent with the observation that the JA content in the young flower buds of the GA-deficient quadruple mutant ga1-3 gai-t6 rga-t2 rgl1-1 is much lower than that in the WT. We conclude that GA promotes JA biosynthesis to control the expression of MYB21, MYB24, and MYB57. Therefore, we have established a hierarchical relationship between GA and JA in that modulation of JA pathway by GA is one of the prerequisites for GA to regulate the normal stamen development in Arabidopsis. © 2009 Cheng et al. 摘要:
为进一步研究超级稻的生长素结合蛋白,采用生物信息学方法获得其全长cDNA序列,并进行分析;对全长序列进行了全长验证。从数据库搜索获得了一条水稻cDNA序列,含一个推测的206个氨基酸的开放阅读框,与其它植物的ABP1cDNA序列和ABP1蛋白质序列具有明显的同源性,并且其推测编码的蛋白质的各种性质,类似于其它植物的ABP1。结果显示,该序列为水稻的ABP1cDNA。以该序列为指导,利用RT-PCR扩增超级稻ABP1cDNA,并克隆测序,其序列与电子搜索结果一致。

花生生育早期耐涝性室内鉴定对大田期的意义

摘要:
为探索花生不同品种生育早期耐涝性差异及与大田耐涝性的关系,对8个品种的种子进行室内180h浅、深水处理,对幼苗进行10d淹涝和大田短(10d)、长期(88d)湿涝,测定发芽能力、幼苗生长发育和乙醇脱氢酶(ADH)活性、大田湿涝产量(WY_(10)、WY_(88))和产量耐性系数(WTC_(10)、WTC_(88))。结果表明,(1)花生对淹涝较敏感,其耐涝性因淹涝时期、程度(水深)和品种而异。浅涝时种子先露尖而后发芽;深涝时只露尖而不能形成幼芽。不同品种发芽期耐涝性差异较大。幼苗的根部受害程度远甚于地上部,根色、根重是不同品种耐性差异的主要表征指标。(2)花生幼苗根系ADH活性受淹涝即厌氧诱导的效应极为明显,其中淹涝3~10d的ADH活性与耐涝性负相关,尤其是淹涝10d后ADH活性高的品种,植株发育不良。(3)与大田所有产量耐涝性指标的相关性,ADH活性、根色均为负相关,单株根重、单株地上部重、单株总重则为正相关,尤其是根重与WY_(10)两年均呈显著正相关。(4)耐涝性鉴定时间,在发芽期浅水和深水中分别以180h和120h为宜,幼苗期以3~10d为宜。因此,花生生育早期耐涝性室内鉴定对大田期有重要参考价值。

超级杂交稻乙烯反应元件结合蛋白cDNA的生物信息学分析与克隆

目的:从超级杂交稻中克隆乙烯反应元件结合蛋白(EREBP)的cDNA.方法:利用模式植物拟南芥中编码乙烯反应元件结合蛋白的cDNA,对现有的水稻基因组数据库进行搜索,获得一条高同源的未知序列.对这条未知序列的核酸序列和蛋白质序列及其结构、性质、功能等进行生物信息学分析后,以超级杂交稻为材料,用未知序列设计一对简并引物,用RT-PCR技术扩增后进行T-A克隆.结果:生物信息学分析结果表明,这个未知序列应为水稻中编码EREBP的cDNA;克隆后经测序获得一条915 bp的cDNA,BLAST表明这条cDNA序列与未知序列的部分核酸序列的同源性达到了99%;提交NCBI的GenBank后被接受,登录号为EF507537.结论:以生物信息学分析为基础,结合RT-PCR和T-A克隆技术,成功地从超级杂交稻中克隆了EREBP cDNA.

摘要:
The phytohormone abscisic acid (ABA) is the major player in mediating the adaptation of plants to stress. Previously developed phytohormonal biosensors usually employed indirect detection of the products of conjugated oxidase reactions. A label-free electrochemical impedance immunosensor for ABA detection was developed using an anti-ABA antibody adsorbed directly on a porous nanogold film. The film was produced electrochemically on a glassy carbon electrode in 0.008 mol/L hydrogen tetrachloroaurate solution containing 0.004 mol/L lead acetate with an applied potential of -0.5 V (versus Ag/AgCl) for 50 s. The anti-ABA antibody was immobilized onto the porous nanogold through electrostatic adsorption and covalent conjugation. Electrochemical impedance spectroscopy was used to characterize the successful construction of the porous nanogold film and the stepwise modification of the glassy carbon electrode. The concentration increase of the antigen brought about a decrease of the interfacial electron transfer, which also meant an increase of the impedance signal. The experimental parameters pH, antibody incubation time, and antibody concentration were optimized. The results showed significant linearity R=0.9942, with the content of ABA in the range 0.5-5,000 ng/mL with a detection limit at about 0.1 ng/mL. © 2008 Springer-Verlag.

摘要:
为探讨植物生长调节剂对大豆Cd胁迫伤害的缓解效应,采用营养液培养试验方法,研究了喷施萘乙酸(NAA)和加Zn处理对3个Cd胁迫大豆幼苗的影响。结果表明,加Zn和喷施NAA均可降低Cd胁迫大豆幼苗叶片丙二醛(MDA)和脯氨酸(PRO)含量,可减轻膜脂的过氧化作用及蛋白质的水解;喷施NAA还可降低Cd胁迫大豆幼苗POD活性,提高硝酸还原酶(NR)活性;加Zn对Cd胁迫大豆幼苗NR活性的降低缺乏抑制作用,但降低沔1101叶片中POD活性,提高湘04-6和特早熟毛豆的POD活性,品种之间表现明显的差异性。

摘要:
通过比较盐、冷、热激三种不同胁迫预处理后超级稻幼苗在低温伤害条件下含水量、过氧化氢酶(catalase,CAT酶)活性、同工酶谱、内源激素的变化,初步研究超级稻的适应性机制。结果表明,水稻幼苗经过不同胁迫预处理后,与未处理的对照苗相比,含水量有所提高;热激预处理苗CAT酶活性高于对照,而盐迫和低温则稍低于对照;同工酶谱变化各有异同,也有其独特性;冷处理苗的脱落酸(abscisic acid,ABA)含量高于对照,而热激处理苗ABA含量稍低于对照,三种预处理苗的赤霉素(gibberellin,GA 3)、玉米素(zeatin,Z)、吲哚乙酸(indole-3-acetic acid,IAA)含量均高于未处理苗。

摘要:
为了明确内生枯草芽孢杆菌B-001对烟草的促生机制和对烟草青枯病的防治效果,采用温室盆栽试验、内源激素检测及处理烟草植株B-001菌量追踪测定等方法研究内生枯草芽孢杆菌B-001的作用机制及其自身在烟草体内的群体数量动态等。结果表明:施用B-001菌株后,烟草幼苗鲜重增加了43.01%~57.70%;烟草中吲哚乙酸、赤霉素和玉米素核苷的含量增加,而脱落酸的含量降低;苗期至旺长期的烟草K326中B-001数量大幅增加,但到成熟期数量大幅下降;1×10^7、1×10^8、1×10^9、1×10^10 cfu/mL的菌悬浮液对烟草青枯病的防治效果分别为50.3%、62.8%、70.2%、70.3%。

土壤Cd处理对大豆叶片激素含量的影响

摘要:
采用土壤盆栽试验与高效液相色谱法(HPLC)分析测试技术,研究了不同浓度Cd处理对大豆生长及叶片生长素(IAA)、赤霉素(GA3)、玉米素(Z)和脱落酸(A)含量变化的影响。结果表明,当Cd处理浓度≤0.50mg·kg~(-1)时,对大豆株高及幼苗期地上部生物产量无明显影响,Cd浓度≥2.50mg·kg~(-1)时,则表现明显的抑制作用。Cd浓度≥0.50mg·kg~(-1)时,对大豆地下部及花荚期地上部生物产量即表现明显的抑制作用。Cd胁迫抑制大豆叶片IAA和Z的合成,当Cd处理浓度为0.25mg·kg~(-1)时,大豆幼苗期和花荚期叶片IAA和Z含量分别为对照的43.1%、75.5%和60.6%、32.6%,Cd浓度升高,抑制作用加强。Cd胁迫对A的合成则表现明显的促进作用,当Cd处理浓度为2.50mg·kg~(-1)时,幼苗期和花荚期叶片A含量分别为对照的13.6倍和3.86倍。对GA3合成的影响则表现为低浓度下的促进作用和高浓度下的抑制作用,即当Cd处理浓度为0.25mg·kg~(-1)时,幼苗期和花荚期叶片GA3含量均高于对照,分别为对照的150.2%和109.7%,Cd浓度增至2.50mg·kg~(-1)时,其含量则分别为对照的36.4%和75.2%。大豆幼苗期和花荚期叶片A/IAA、A/GA3及A/Z与Cd处理浓度之间均达到了显著或极显著的相关性,表明Cd胁迫不仅影响植物激素的含量,同时也影响激素间的比例和平衡。

本研究针对南方花生产区春夏湿涝害,通过盆栽试验研究了湿涝及其营养调控对花生营养吸收的影响.结果表明,1)湿涝时花生对N、P、K和Ca、Mg的吸收和积累严重受抑.N、P、K、Ca总体以叶片下降最明显,其次是根系,茎秆相对下降较小.Mg以茎秆下降最多,其次是叶片,根系相对下降最小.湿涝对花269主要营养元素积累总量的影响大于豫花15号;2)几种营养调控措施对受湿涝后花生的N、P、K和Ca、Mg的吸收和积累几乎均有一定程度的提高.其中含有氮组分的调控处理效果明显,追施磷肥后期可较好的促进花生营养吸收,但效应较迟缓,其余调控处理提高幅度较小.花生叶片中N的积累量和幼果中Ca的积累量与产量之间显著正相关.

摘要:
以黄瓜愈伤组织为材料,比较二甲基亚砜(dimethyl sulfoxide,DMS0)、VK3和CaCl2相同浓度诱导细胞凋亡的差别.将愈伤组织悬浮培养并分离单细胞后进行活性检测和荧光检测,并对其DNA进行琼脂糖凝胶电泳检测,结果表明,3种诱导剂都能诱导黄瓜愈伤组织凋亡,诱导能力VK3>DMSO>CaCl2.

云南省马铃薯晚疫病菌生理小种的组成与分布

摘要:
利用11个分别含有单抗性基因R1~R11及1个不含抗性基因的1套鉴别寄主,对2003~2005年采自云南省10个县市26个采集点的117个马铃薯晚疫病菌的菌株进行生理小种鉴定.结果表明:共鉴定出26个菌株的生理小种,其中优势小种为3.4.6.8.10.11,占所测菌株的28.69%,主要分布在寻甸、丽江、昆明;其次是3.4.10,发生频率为13.11%,主要分布在镇雄;然后是3.10,发生频率为10.66%,主要分布在宣威.

摘要:
为了解花生不同种质和生育期的耐涝性差异及与大田产量耐涝性关系,对8个种质的种子进行180h浅、深水处理,对其幼苗进行10d淹涝和大田短(10d)、长期(88d)湿涝,测定发芽能力、幼苗生长发育和乙醇脱氢酶(ADH)活性、大田湿涝产量(WY<,10>、WY<,88>)和产量耐性系数(WTC<,10>、WTC<,88>)。结果表明:
(1)花生对淹涝较敏感,且因淹涝时期、程度(水深)和种质而异。浅涝时种子先露尖而后发芽;深涝时只露尖而不能形成幼芽。不同种质发芽期耐涝性差异较大。幼苗的根部受害程度远甚于地上部,根色、根重是不同种质耐性差异的主要表征指标。
(2)花生幼苗根系ADH活性受淹涝即厌氧诱导的效应极为明显,其中淹涝3~10d的ADH活性与耐涝性为负相关,尤其是淹涝10d后的ADH活性高的种质,其植株发育不良。
(3)与大田所有产量耐涝性指标的相关性,ADH活性、根色均为负相关,单株根重、单株地上部重、单株总重则为正相关,尤其是根重与WY<,10>两年均呈显著正相关,与WY<,88>相关度也较高。
(4)耐涝性鉴定时间,在发芽期浅、深水中分别以180h、120h为宜,幼苗期以3~10d为宜。

摘要:
To investigate the effects of formulated fertilizer synergist on the drought tolerance in rice, pot experiment was conducted to analyze the photosynthetic characteristics and the accumulation of abscisic acid (ABA) and proline in middle-season rice variety Peiliangyou 93. The synergist could improve the net photosynthetic rate, and coordination between the water loss and the CO2 absorption as well as reduce the harmful effect on photosynthetic process under drought conditions. Under drought, the ABA accumulated massively both in roots and leaves, while the ABA content in roots was far higher than that in leaves. The results indicate that synergist could increase the ABA accumulation, but reduce the proline accumulation in rice plant under drought.

采用室内培养试验,通过测定植株生理生化指标,研究了不同浓度Cd^2+处理对大豆幼苗生长及激素含量的影响.结果表明:①不同浓度Cd^2+处理对大豆幼苗地下部生长素(IAA)和赤霉素(GA3)的合成有抑制效应,对地上部生长素(IAA)和赤霉素(GA3)的合成具有低浓度下的刺激效应和高浓度下的抑制效应;对地下部、地上部玉米素(Z)和脱落酸(ABA)的合成均有刺激效应.②Cd^2+处理84h对大豆幼苗根系活力、叶绿素a和叶绿素b的合成有刺激效应,对类胡萝卜素的合成无影响.③Cd^2+处理对过氧化物酶(POD)活性和丙二醛(MDA)含量具有低浓度下的抑制效应和高浓度下的激活效应,且随着Cd^2+浓度的增加激活效应逐渐加强.④短时间、低浓度Cd^2+胁迫可刺激大豆幼苗的生理活性,以Cd^2+处理浓度为0.50mg/L时,大豆幼苗根系活力最大,IAA和GA3含量最高,POD活性最低,植物生理活性最强.
