Phylogenetic analysis of endosymbionts associated with Bemisia tabaci (Hemiptera: Aleyrodidae)

摘要:
Endosymbionts include primary endosymbionts and secondary endosymbionts, accreting with insects. Endosymbionts evolve with insects and benefit from each other. A report was given on the difference of endosymbionts found in Bemisia tabaci (Gennadius 1889) and Trialeurodes vaporariorum (Westwood 1856) before. However, the phylogentic analysis of primary and secondary bacterial symbionts associated with the whitefly B tabaci B biotype are poorly known. Here are presented the results of cloning and sequencing 16S rDNA of primary and secondary endosymbionts in B tabaci living for extended periods on different host plants in Beijing. Molecular phylogenetic trees, including about 1 000bp 16S rDNA of primary endosymbionts, and about 1 250bp 16S rDNA of secondary endosymbionts in B tabaci were constructed. The results indicate that primary endosymbionts in B tabaci were Proteobacteria, Gammaproteobacteria, Oceanospirillales, Halomonadaceae, Zymobacter and Candidatus Portiera aleyrodidarum. Secondary endosymbionts in B tabaci were Proteobacteria, Gammaproteobacteria, Enterobacteriales and Enterobacteriaceae. The primary and secondary endosymbionts in B tabaci from different host plants or other countries might belong to different ecological types from one species specifically. B tabaci independently acquired various endosymbionts when living on different host plants, and these endosymbionts played important roles for B tabaci's accomodation in different environments. The phylogentic trees based on 16S rDNA of primary endosymbionts could be explained by geographical origin more reasonably than those of the secondary endosymbionts, and the phylogentic trees based on 16S rDNA of secondary endosymbionts can be explained by varying host resources more reasonably than those of primary endosymbionts. B tabaci may have been introduced to China via the importation of Poinsettia plants. © 2007 E. Schweizerbart'sche Verlagsbuchhandlung.

摘要:
以张家界国家森林公园为例,分别设背景、缓冲和活动3个试验区,探讨了旅游活动对土壤理化性质、土壤酶活性和土壤微生物分布的影响.结果表明:土壤酶和土壤微生物的分布特征呈现背景区最高,活动区最低,缓冲区居中的变化规律;对土壤酶活性的影响在0~5 cm土层最为显著,其中纤维素酶活性在活动区分别比缓冲区和背景区降低了92.8%和80.0%;其次,旅游活动显著影响了0~5 cm土壤土层中的细菌、放线菌、真菌及总微生物数量,其中背景区明显高于缓冲区和活动区,缓冲区又明显高于活动区.旅游活动已对张家界国家森林公园土壤生态系统产生了较大影响.

从工厂排污口废水中分离.纯化并筛选出一株降解十二烷基聚氧乙烯醚(Brij-30)的菌株,鉴定为伯克氏菌属(Pandoraca sp),命名为B30.当温度为30℃,pH5~8,底物浓度低于0.2g/L,且以蛋白胨做氮源时,菌株B30降解效果最好;Zn~(2+)Ca~(2+)、Al~(3+)、Fe~(3+)对菌株B30的生长及其降解性能具有较大的促进作用.在低浓度区(0.05-0.20g/L),菌株B30生长最快,降解活力性最强,12h内能将底物基本降解;在中等浓度区(0.40g,L左右),72h内能降解50%的底物;在高浓度区(0.80-1.50g/L),72h内底物的降解率在30%以下.

摘要:
Three nematode-endoparasitic, Hirsutella rhossiliensis (18 isolates), Hirsutella minnesotensis (8 isolates) and Hirsutella vermicola (3 isolates) were studied for their variability in morphology, nematode parasitism, and DNA sequences of internal transcribed spacer (ITS) of ribosomal DNA (rDNA) and mitogen-activated protein kinase (MAPK) gene fragments. There were differences in length and width of conidiogenous cells and conidia among the three Hirsutella species and among isolates within species of H. rhossiliensis and H. minnesotensis. Most isolates of H. rhossiliensis and H. minnesotensis parasitized higher percentages of the cyst nematodes, Heterodera glycines and Heterodera avenae, than the four non-cyst nematodes, Meloidogyne hapla, Bursaphelenchus xylophilus, Heterorhabditis bacteriophora, and Steinernema carpocapsae. H. vermicola had no or weak parasitism on the six assayed nematode species. The phylogenetic trees of ITS region, MAPK gene fragments and their combination revealed that there was genetic variability among species and isolates, but there was no apparent relationship between the genetic variability and their host or geographic origin. © 2007 Elsevier Inc. 摘要:
The phylogeny of nematode-endoparasitic Hirsutella species was studied based on morphology and DNA sequence analysis. Hirsutella vermicola is proposed as a new species that preys on bacteria-feeding nematodes and nonpathogenic or weakly pathogenic to plant-parasitic nematodes. It is distinguished from H. rhossiliensis by having shorter conidiogenous cells with a swollen base, helical necks and slightly wider conidia. Phylogenetic trees based on sequences of the ITS region, MAPK gene fragment, and the combined data revealed two clusters corresponding to these two species and thus supported the establishment of this new species.

摘要:
This is the first time to describe the diversity of denitrifying bacteria in Sanjiangyuan natural reserve in Tibet Plateau by investigating the molecular diversity and phylogenetic analysis of nirK and nosZ genes using PCR-RFLP and sequencing. Four soil samples were collected from alpine meadow communities from over an altitude of 4600 m which had different physicochemical properties by principal component analysis (PCA). For the genes fragment of nirK and nosZ, the diverse PCR products were characterized by cloning, restriction fragment length polymorphism (RFLP) analysis and sequenced. A total of 253 nirK clones and 283 nosZ clones were received in four samples, and 78 operational taxonomic units (OTUSs) of nirK and 120 OTUs of nosZ by the restriction enzymes Mspl and Rsal digested. The analysis of environmental factors showed that altitude and C/N ratio in soil may be the key factors to the denitrifying bacteria community. 36 nirK clones and 17 nosZ clones were sequenced, and their levels of nucleotide identity were from 69% to 98% and 57% to 97%, respectively. The phylogenetic tree was constructed using the Clustal W and Mega softwares, and all the sequenced clones could be subdivided into 4 groups. Some of clone sequences were related to the nirK and nosZ genes belonging to three phylogenetic subdivisions (α-, β-and γ subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. The sequence distributions were not clear relating to the sample sites in the tree. © Science in China Press 2006.

To understand the composition and structure of nitrogen-fixing bacterial communities from the Sanjiangyuan Nature Reserve on the Tibetan Plateau, the molecular diversity of nifH genes from soil obtained at six sites was examined using a PCR-based cloning approach. Six samples were collected from different regions at an altitude of 3907-4824 m above sea level, and a principal component analysis (PCA) showed that they had different biogeochemical properties. A total of 446 clones and 162 unique RFLP patterns were found. PCA of the RFLP patterns and their biogeochemical parameters showed that the content of soil organic carbon (C), total nitrogen (N) and altitude were the most important factors affecting the nitrogen-fixing bacteria community. Fifty-nine nifH clones were sequenced and their nucleotide identity varied from 64% to 98%, subdivisible into four groups in our phylogenetic tree. Some of the clone sequences were related to nifH genes belonging to four phylogenetic subdivisions (α, β, γ and δ subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. The tree also showed that the sequence distributions were not clearly related to the sample sites. © 2006 Federation of European Microbiological Societies Published by Blackwell Publishing Ltd. The host range of Bemisia tabaci (Westwood 1856) and Trialeurodes vaporariorum (Gennadius 1889) and their kind of host damage are reported briefly. So far, the bacterial symbionts, associated with the biological characters of whitefly species, and their particular influence on the evolution of the whitefly host, are not yet studied sufficiently. Here, the composition and genomes of prokaryotic symbionts associated with the whitefly species B tabaci (B biotype) and T vaporariorum were examined from material collected on corresponding host plant species and from the same geographical locations. The ultrastructures of the endosymbionts in B tabaci and T vaporariorum were examined by using transmission electron microscopy: There are differences in morphology and relative number of endosymbionts between B tabaci and T vaporariorum. B tabaci contains two morphological types of microorganisms housed within the mycetocyte cells, but T vaporariorum contains only one morphological type of microorganisms housed within the mycetocyte cells. The 16S rDNA and groEL of the endosymbionts in B tabaci and T vaporariorum were obtained by polymerase chain reaction (PCR) amplification. The 16S rDNAs and groEL of the endosymbionts in B tabaci and T vaporariorum are found to be different: Primary endosymbionts in B tabaci are Proteobacteria: Gammaproteobacteria: Oceanospirillales: Halomonadaceae: Zymobacter-group: Candidatus portiera aleyrodidarum, and secondary endosymbionts in B tabaci are Proteobacteria: Gammaproteobacteria: Enterobacteriales: Enterobacteriaceae. The endosymbionts have important effects on B tabaci associated with its ecological distribution and the plant viral disease transmission. GroEL is very conservative. There is little difference among groEL from endosymbionts of B tabaci from different geographical origin. Amplification and comparison of 16S rDNA sequences revealed that a primary-like endosymbiont is associated with all whitefly samples examined.

摘要:
利用SDS高盐法和变性剂加SDS高盐法对土壤微生物总DNA进行了提取,然后通过电泳加树脂柱回收和连续2次树脂柱回收方法进行了纯化.结果表明,变性剂加SDS高盐法的DNA提取效率明显高于前者,电泳加树脂柱法的纯化效果更好.通过PCR扩增表明,经过纯化后的DNA,都可以进行16SrDNA扩增和nirK、nosZ、nifH等功能基因的扩增.因此,变性剂加SDS高盐法是一种更为高效、可靠且适合于环境微生物分子生态学研究的DNA提取方法.

三江源高寒草甸土固氮基因(nifH)的多样性和系统发育研究

首次利用PCR-RFLP和测序分析对位于青藏高原腹地的青海三江源保护区高寒草甸土壤微生物固氮基因(nifH)的多样性和系统发育进行了探讨.在2个样地中,共得到143个阳性克隆,用限制性内切酶MspⅠ和RsaⅠ进行RFLP分析后得到35个不同的RFLP谱带型,多样性为24.5%,其中ZD样品中获得82个阳性克隆和21个不同的RFLP谱带型,多样性为25.6%,而YS样品中获得61个阳性克隆和19个不同的RFLP谱带型,多样性为31.1%,2个样地中有5个相同的RFLP谱带型.在各样地都发现一个明显的优势种群,ZD样地的明显优势种群占克隆数的29.3%,YS的优势种群占克隆数的32.8%.对21个克隆进行了部分序列的测定,序列的相似性在71%~98%之间,在GenBank数据库中没有发现完全匹配的序列,因此这些序列可能代表着新的固氮生物株系.最后利用Clustal W与Mega软件和已有序列构建了系统发育树,结果发现,21个序列分为4个不同的簇,大部分的克隆与Proteobacteria的3个系统发育亚簇(α、β和γ)具有较高的相似性,其中主要的序列都落在第一和第二簇内.YS样地中的优势种群与α-Proteobacteria中的Rhodobacter sphaeroides具有较高的相似性,而ZD样地中的优势种群则与β-Proteobac-teria中的Delftia tsuruhatensis具有较高的相似性.

利用PCR-RFLP和测序分析法对位于青藏高原腹地三江源自然保护区的高寒草甸、高寒草原和高山森林等不同植被类型的土壤固氮微生物的群落组成进行了探讨.经过PCR-RFLP分析固氮基因nifH,在3个样品中共得到233个克隆和99个可操作分类单元(OTUs),NQ-1样地具有最多的克隆数和OTUs,多样性为49.74%,在所有样品中分别具有1~2个明显的优势种群(占总克隆数>15%),并且具有4个共同的OTUs.选取了26个克隆进行基因测序分析,通过DNAMAN比较表明,这些序列间具有66%~98%的相似性,并且在GenBank数据库中没有发现完全匹配的序列,因此这些序列可能代表着新的固氮生物株系.最后利用Clustal W与Mega软件构建了系统发育树,结果发现,这些序列被分为4个不同的簇,部分序列与属于蛋白细菌(Proteobacteria)的已知细菌具有近的亲缘关系,但是更多的序列与已知细菌具有较远的亲缘关系,而且nifH基因序列的分布在样地间没有明显的聚类.

烟粉虱内共生菌groEL基因编码一种63KD的GroEL蛋白,属于分子伴侣cpn60家族成员 ,与烟粉虱传播植物病毒病关系密切.通过PCR对长期生活在甘蓝上的烟粉虱体内groEL基因进行了PCR扩增,序列测定表明其长度为1668bp,编码555个氨基酸,与 Genebank中烟粉虱内共生菌groEL(AF130421)的核苷酸同源性为99.82%,氨基酸同源性为99.64%.

香蕉细菌性枯萎病是香蕉上的一种毁灭性维管束病害(Salanoubat et al.,2002).美国、墨西哥、中南美洲、以及亚洲一些国家都有此病发生,目前我国尚无分布的报道,因此它是被我国列为禁止入境的二类危险性有害生物.我国入世后,香蕉的进口量呈明显上升趋势,而这种病原细菌在与我国有贸易关系的国家(如菲律宾等国家)中发生极为普遍,一旦这种病原细菌传入我国,将会给我国的香蕉种植业造成不可估量的损失.本研究克隆了香蕉细菌性枯萎病菌的16S rDNA片段并对其进行了同源性分析,为设计特异引物或探针鉴定香蕉细菌性枯萎病菌提供基本资料.

饲养东北虎的微卫星变异研究

摘要:
东北虎是世界上濒危动物之一,具有极其重要的研究价值和保护意义.该研究利用10个在东北虎基因组中表现多态性的微卫星基因座(Fca005,Fca075,Fca094,Fca152,Fca161,Fca294,Pti002,Pti003,Pti007和Pti010)对113只饲养东北虎进行了遗传多样性检测.用非变性聚丙烯酰胺凝胶电泳检测微卫星的PCR扩增产物,计算了10个微卫星基因座的等位基因频率、基因杂合度、多态信息含量和有效等位基因数.在113只东北虎样品中,10个基因座的等位基因数为3~6个,其中Fca152最多;等位基因频率处于0.009~0.767之间.基因杂合度值在0.385~0.707间,平均为0.616,多态信息含量值在0.353~0.658间,平均为0.558,有效等位基因数处于1.629~3.409之间,平均为2.784,表明所选用的10个微卫星基因座在研究样品中均为中高度多态性基因座,具有比较明显的遗传变异.113只样品中包括75只毛发样品,23只血液样品和15只组织样品,不同样品的结果比较表明,毛发、血液和组织样品均可以得到清晰的扩增结果.所以,微卫星基因座与非损伤性DNA分析方法可以成功地应用于濒危珍稀动物的遗传多样性研究.

烟粉虱与温室白粉虱内共生菌的分子比较研究

摘要:
利用PCR技术对烟粉虱与温室白粉虱内共生菌进行分子比较研究,筛选出C、D、G和H等4对引物,分别能够扩增出烟粉虱内共生菌的groEL基因、初生内共生菌与次生内共生菌的16SrDNA基因,与GenBank中相应序列的同源性分别为99%、99.58%、98%和98%,而不能从温室白粉虱内共生菌中扩增出该类基因,表明烟粉虱与温室白粉虱内共生菌存在分子差异.

摘要:
对5年连续饲养在不同种寄主植物上的B型烟粉虱北京种群的内共生菌16S rDNA基因进行了PCR扩增和测序.结合已知序列,构建了不同寄主植物烟粉虱初生内共生菌约1000bp的16S rDNA及次生内共生菌约1250bp的16S rDNA的分子系统树.结果表明,中国北京不同寄主植物的B型烟粉虱内共生菌及世界其它地区烟粉虱内共生菌可能是同一种的不同生态型,内共生菌在其宿主分化后进行了选择,之后与其宿主长期共同进化、共同适应,为宿主对不同生境的适应提供了一定的基础.

基因芯片因其具有高密度、高灵敏度、快速(实时)检测、经济、自动化和低背景水平等特点,而广泛应用于不同的研究领域.目前,应用于环境微生物研究的基因芯片主要有功能基因芯片(FGAs)、系统发育的寡核苷酸芯片(POAs)和群落基因组芯片(CGAs).综述了基因芯片在环境微生物研究中的应用,包括自然环境中微生物的基因表达分析、比较基因组分析和混合微生物群落的分析等.讨论了基因芯片面临的挑战和前景展望.

昆虫传播的植物病毒种类多、危害大,其传病毒的能力与昆虫体内共生菌产生的GroEL蛋白关系密切,该蛋白属于分子伴侣hsp60家族的成员,对病毒进入昆虫血体腔免遭破坏起着保护作用,也与昆虫传病毒的专一性有关。本文对昆虫内共生菌及其产生的GroEL进行了综述,并分析了研究内共生菌及其产生的蛋白质的科学意义与发展趋势,为植物病毒病的防治研究提供了新的思路。

烟粉虱内共生菌groEL基因编码一种63 kD的GroEL蛋白,利用一对特异性引物,通过PCR对长期培养在黄瓜上的烟粉虱体内groEL基因进行了扩增,序列测定表明其长度为1 668 bp,编码555个氨基酸,与GenBank中烟粉虱内共生菌groEL(AFl30421)的核苷酸同源性为99.58%,仅7个核苷酸发生了变异,二者氨基酸序列同源性为99.28%,仅4个氨基酸有别.构建了一个原核表达载体,表达得到了76 kD的融合蛋白.
