水稻Pi9基因序列标记的开发及其抗瘟育种应用

摘要:
利用广谱抗稻瘟病基因Pi9的3′UTR序列设计特异DNA标记pi9utr,通过分子标记辅助选择和连续回交育种实践,改良7份籼稻亲本(316B、金23B、R207、R228、R288、R389、明恢86)的稻瘟病抗性。结果表明:pi9utr是一个高效显性分子标记,除R207外,在其余6份受体亲本与Pi9供体亲本75-1-127间均存在明显而稳定的多态;在室内接种后的316B ×75-1-127 BC4F1群体和R288×775-1-127 BC6F1群体中随机取样,进行稻瘟病抗性表型和基因型鉴定及 Pi9基因表达分析,证明 pi9 utr对两个组合回交后代个体的抗病性辅助选择效率均为100%,且在所有抗病单株中均能检测到Pi9基因的高效表达,在所有感病单株中均没有检测到Pi9基因的表达。利用pi9utr在回交世代中的连续辅助选择,获得了3个组合的BC4F1及3个组合的BC6F2代群体,为选育抗稻瘟病新品种打下了基础。

摘要:
本文研究了斜纹夜蛾长距姬小蜂的生物学特性,结果表明,实验室条件下,斜纹夜蛾长距姬小蜂单雌最大产卵量为139粒,平均产卵量为73.69粒;卵主要产在斜纹夜蛾幼虫的1~4腹节背面,少部分产在侧面,不产于寄主腹面;单寄主上通常产卵1~5粒,有时为6~9粒;喜寄生2、3龄斜纹夜蛾幼虫,偶尔寄生1龄幼虫,但绝大部分不能完成其生命历期;当寄主斜纹夜蛾2龄、3龄初、3龄末幼虫同时存在时,其偏爱寄生3龄初幼虫,偏好系数为0.65、发育历期为9.93±0.19 d。

摘要:
A chiral selector of cellulose-2,3-bis(3,5-dimethylphenylcarbamate) (CBDMPC) was synthesized by reacting 3,5-dimethylphenyl isocyanate with microcrystalline cellulose dissolved in an ionic liquid of 1-allyl-3-methyl-imidazolium chloride (AMIMCl). The obtained chiral selector was effectively characterized by infrared spectroscopy, elemental analysis and 1H NMR. The selector was reacted with 3-aminopropylsilanized silica gel and the CBDMPC bonded chiral stationary phase (CSP) was obtained. Chromatographic evaluation of the prepared CSPs was conducted by high performance liquid chromatographic (HPLC) and baseline separation of three typical fungicides including hexaconazole, metalaxyl and myclobutanil was achieved using n-hexane/isopropanol as the mobile phase with a flow rate 1.0 mL/min. Experimental results also showed that AMIMCl could be recycled easily and reused in the preparation of CSPs as an effective reaction media. © 2014 by the authors; licensee MDPI, Basel, Switzerland.

坡柳皂苷对大菜粉蝶生长发育的抑制活性

摘要:
采用浸渍叶碟饲喂法测定了坡柳皂苷对大菜粉蝶的生物活性.结果表明:用500、1 625、2 750、3 875和5 000 μg/mL的坡柳皂苷处理24 h后,对大菜粉蝶4龄幼虫生长发育的抑制率分别为14.47%、31.54%、49.70%、62.48%和79.94%,处理48 h后的抑制率分别为2.72%、39.26%、54.82%、70.53%和86.39%;坡柳皂苷对大菜粉蝶4龄幼虫具有显著的拒食活性,供试5种浓度下处理24 h后,拒食率分别为30.21%、40.62%、54.33%、72.47%和84.84%,48 h后的拒食率为4.4% ~ 86.67%;5 000 μg/mL处理14 d后,幼虫化蛹率为32%,显著低于对照组(72%);供试5种浓度坡柳皂苷处理23 d后,蛹的羽化率分别为56%、44%、44%、36%和32%,均低于对照组(64%).研究表明,坡柳皂苷对大菜粉蝶生长发育、取食及化蛹均有明显的抑制作用.

摘要:
对湖南省棉花主产区棉田杂草的种类、发生规律及化学除草情况进行了调查。结果发现,湖南省棉田常见杂草包括12科,28种,其中禾本科杂草占25.00%,阔叶杂草占67.86%,莎草科占7.14%;一年生杂草占78.57%,多年生杂草占21.43%。相对多度10%以上的杂草有7种,其中马唐、牛筋草、千金子、铁苋菜、小飞蓬5种杂草为湖南省棉田杂草的优势种,相对多度分别为37.79%、33.64%、29.15%、26.23%、23.83%。杂草发生呈现3个高峰期,分别为5月下旬,6月中下旬,7月下旬至8月初,杂草防除主要采用苗前土壤处理和苗后茎叶处理相结合的化学防除方法。

摘要:
2012年8月湖南新田县蔬菜基地发生茄子果实腐烂病,损失严重。从感病茄果上分离到一种真菌,镜检表明为茄褐纹拟茎点霉,此菌rDNA-ITS序列亦与茄褐纹拟茎点霉高度同源。因当地反映百菌清、多菌灵、代森锰锌等药剂均无明显效果,故尝试了室内实验以筛选出有效防治该病的药剂。采用菌丝生长速率法,用11种杀菌剂做了室内毒力测定。咪鲜胺对茄褐纹病菌的抑制效果最好,EC50值为0.021 mg/L;其次是甲基硫菌灵和苯醚甲环唑,EC50值是0.252和0.434 mg/L;百菌清与醚菌酯的抑制效果最差,EC50值分别为44.400和54.470 mg/L。建议在病区选用咪鲜胺防治茄褐纹病。

摘要:
In this paper, a novel monolithic stir bar based on molecularly imprinted polymer (MIP) was firstly developed by filling modified neodymium magnet (Nd2Fe14B) powders into a glass tube (60 × 4 mm), followed by the imprinted grafting with bisphenol A (BPA) as the template molecule by thermal polymerization. It has been successfully used for the stir bar sorptive extraction (SBSE) and its extraction performance illustrated that the MIP-encapsulated stir bar had stronger affinity to the template molecule, compared with the stir bar based on the non-imprinted molecularly polymer (NIP). Under the optimal extraction conditions, a simple method based on the coupling of MIP-SBSE with high performance liquid chromatography (HPLC) was used for the selective determination of the model mixtures of BPA, 4-phenylphenol (PP) and phenol (P) in bottled water. The recoveries of BPA, PP and P were in the range of 88.5-96.1%, 78.2-89.7%, 81.3-89.5% at three spiked levels, respectively, demonstrating that higher extraction and the specific absorption occurred between the template molecule and the prepared MIP stir bar. The monolithic stir bars with enough stiffness and magnetic force were successfully synthesized by thermal polymerization in the glass tube using the Nd2Fe14B magnetic particles as the starting materials. The modification was carried out by three steps including the enwrapping of SiO2, the induction of vinyl end groups and the form of MIPs on the surface of the magnetic particles. 摘要:
As part of a long-term sloped land use experiment established in 1995 at Taoyuan Agro-ecosystem Research Station (111°26′ E, 28°55′ N) in China, soil samples were collected from three land use types, including cropland (CL), natural forest, and tea plantation. Quantitative polymerase chain reaction and terminal restriction fragment length polymorphism were used to determine the abundance and community composition of amoA-containing bacteria (AOB) and archaea (AOA). The results indicate that land use type induced significant changes in soil potential nitrification rate and community composition, diversity, and abundance of AOB and AOA. Both AOB and AOA community compositions were generally similar between upper and lower slope positions (UP and LP), except within CL. The LP soils had significantly (p < 0.05) higher diversity and abundance of both AOB and AOA than in the UP. Potential nitrification rate was significantly correlated (p < 0.05) with diversity and abundance of AOA, but not with AOB. Among land use types, the NO3- and amoA-containing AOA runoff loss was greatest in CL. Nitrate-N runoff loss was significantly correlated (p < 0.05) with the loss of AOA amoA copies in the runoff water. Furthermore, relationships between NO3--N runoff loss and abundance of AOA but not of AOB at both slope positions were significantly correlated (p < 0.05). These findings suggest that AOA are more important than AOB in nitrification and NO3--N runoff loss in acidic soils across sloped land use types. © 2012 Springer-Verlag Berlin Heidelberg.

摘要:
A taxonomic revision of the subgenus Cosmiomorpha (Cosmiomorpha) Saunders is presented. Seven species are recognized, including four described herein, C. fortis new species, C. nigripedis new species, C. maolanensis new species, and C. cheni new species all from China. Cosmiomorpha baryi Bourgoin and C. squamulosa Schürhoff are placed as junior synonyms of C. decliva Janson, and C. angulosa Fairmaire as a synonym of C. decliva is also confirmed. Lectotypes are designated for C. decliva Janson, C. angulosa Fairmaire, and C. squamulosa Schürhoff. Color photographs and diagnoses of all species are provided, with comments on intraspecific variations. A key to males is also presented. Localities of "Siáo-Lóu" and "Se Pin-Lou Chan" are discussed with a map. © 2013 Magnolia Press.

麦蛾柔茧蜂作为石河子地区田间常见寄生蜂,对当地玉米的保产保质起到了非常重要的作用,为更好地利用麦蛾柔茧蜂防治玉米螟,本文就其寄生行为、寄主喜好性等方面展开研究。结果表明:多数羽化成蜂当日交尾,雄蜂可多次交尾,雌蜂一般交尾一次;多数雌蜂在4min内找到寄主,约4min内处理完寄主;寄主喜好性测定显示对玉米螟的寄生效果最好。

在湖南省湘潭县易俗河镇杨溪村农业科技园,运用5种不同波长的佳多牌高压电网式太阳能杀虫灯对其诱杀菜田害虫的综合效果进行了试验,历时25天试验结果显示,高压电网式太阳能杀虫灯的诱虫谱有9个目,30个科,45个种,总诱虫量为25 543头,对鞘翅目和鳞翅目的诱虫效果最佳,分别占害虫总量的66.7%和21.9%,平均单灯累计诱杀1 702.9头,5种不同波长杀虫灯的天敌与害虫之比分别为:1:35.71,1:24.58,1:8.67,1:15.43和1:11.87。经分析得出:试验所用的5种不同波长的高压电网式杀虫灯中只有1号和5号波长的杀虫灯对蔬菜害虫的诱集效果最好。本研究积累了大量的田间观察数据,为诱虫灯在农业防治上的推广提供参考。

摘要:
The sweet potato whitefly, Bemisia tabaci (Gennadius) (Hemiptera:Aleyrodidae), is an invasive and damaging pest of field crops worldwide. The neonicotinoid insecticide imidacloprid has been widely used to control this pest. We assessed the species composition (B vs. Q), imidacloprid resistance, and association between imidacloprid resistance and the expression of five P450 genes for 14-17 B. tabaci populations in 12 provinces in China. Fifteen of 17 populations contained only B. tabaci Q and two populations contained both B and Q. Seven of 17 populations exhibited moderate to high resistance to imidacloprid, and eight populations exhibited low resistance to imidacloprid, compared with the most susceptible field WHHB population. In a study of 14 of the populations, resistance level was correlated with the expression of the P450 genes CYP6CM1 and CYP4C64 but not with the expression of CYP6CX1, CYP6CX4, or CYP6DZ7. This study indicates that B. tabaci Q has a wider distribution in China than previously reported. Resistance to imidacloprid in field populations of B. tabaci is associated with the increased expression of two cytochrome P450 genes (CYP6CM1 and CYP4C64). 摘要:
Morophaga parabucephala Ponomarenko & Park, 1996 closely related to Morophaga bucephala (Snellen, 1884) is newly recorded from Japan and China. The adults of the two species are redescribed with figures of male and female genitalia. The morphological characteristics are compared with the related species M. bucephala (Snellen). The female genitalia of M. parabucephala are described for the first time.

本文以蚜茧蜂Lysiphlebia japonica Ashmead与棉蚜Aphis gossypii Glover系统为研究对象,以内禀增长率r_m、作为适合度指标,通过开顶式动态CO_2气室(OTC),重点比较了目前大气CO_2浓度与未来加倍的大气CO_2浓度下(375 μL/L VS 750 μL/L)直接放入蚜茧蜂、笼罩间接放入蚜茧蜂的胁迫作用对不同世代和3个连续世代棉蚜适合度的影响。结果表明:CO_2浓度变化、世代之间、寄生胁迫方式对棉蚜净繁殖率R_0、平均世代时间r和内禀增长率r_m均存在着极显著的影响;CO_2浓度变化和寄生胁迫方式之间、CO_2浓度变化和棉蚜发生世代之间的交互作用对棉蚜净繁殖率R_0和内禀增长率r_m也存在着极显著的影响。结果提示:大气CO_2浓度将通过影响寄生蜂的间接干扰作用,从而影响棉蚜的适合度和种群动态,使棉蚜种群有下降的趋势。

灰毛豆种子中化学成分的鉴定及其对黑翅土白蚁的杀虫活性

摘要:
为探明灰毛豆Tephrosia purpurea种子中的杀虫成分及其对黑翅土白蚁Odontotermes formosanus工蚁成虫的杀虫活性,对其甲醇提取物采用多种色谱技术分离纯化,结合核磁共振图谱和质谱方法鉴定化合物的结构,并测定了各化合物对黑翅土白蚁工蚁成虫的触杀、忌避和传毒活性。从灰毛豆种子甲醇提取物中分离鉴定了10个化合物,即Obovatachalcone,Obovatin,异合生果素,鱼藤酮,Pongachin,12α-羟基鱼藤酮,6a,12a-脱氢灰叶酚,灰叶素,Penduletin和棉子糖。5个化合物(异合生果素,灰叶素,Obovatin,12α-羟基鱼藤酮和鱼藤酮)表现出触杀活性,处理后24 h的LD_(50)值分别为每虫93.9,123.5,209.1,259.4和370.8μg;3个化合物(Obovatin,鱼藤酮和12α-羟基鱼藤酮)表现出传毒活性,处理后56 h累积中毒率达24.3,38.7和100%;2个化合物(异合生果素和灰叶素)在处理浓度为300 mg·L~(-1)时表现出较弱的忌避活性。结果表明,化合物12α-羟基鱼藤酮对黑翅土白蚁工蚁成虫无忌避性,有一定的触杀活性,在群体中的毒性传播能力强,可进一步研究用于白蚁防治。

沉默cadherin基因对小菜蛾敏感性的影响

钙粘蛋白(cadherin)是许多鳞翅目昆虫中苏云金芽孢杆菌(Bt)毒素的受体。利用RNAi技术对小菜蛾中肠cadherin基因进行沉默处理,明确了cadherin基因沉默对小菜蛾对Cry1Ac毒素敏感性的影响。结果表明,注射目标dsRNA后的第1天至第5天,Cry1Ac敏感及抗性小菜蛾品系cadherin的基因表达量均显著减少,尤以第2天的减少量最为明显;Cry1Ac毒力测定结果表明,沉默cadherin基因能在一定程度上提高幼虫的成活率,即可使小菜蛾对Cry1Ac毒素的敏感性降低。研究初步表明,cadherin基因的功能与小菜蛾对Cry1Ac毒素的抗性有关。

重要气象因子对农田灯光诱虫效率的影响

摘要:
为提高农田灯光诱虫效率以及农田灯光诱虫装置的自动化开关灯条件的合理设计提供理论依据,实验分别于湖南南部、中部及北部3地的烟地或菜地设置若干台太阳能诱虫灯,每日收集各诱虫灯下的重要害虫和天敌,并进行鉴定、计数或烘干称重,然后比较同一地点或不同地点诱虫灯之间诱虫量的差异,并用相关-回归法分析每日诱虫量与每日重要气象因子之间的各种复杂关系。结果表明:诱虫量与大气日平均气温呈显著正相关关系(即灯光诱虫量随气温的升高而显著增加),而与气压、降雨和相对湿度则呈显著的负相关关系(即灯光诱虫量随气压或湿度的升高以及降雨的发生而显著减少),即高温、低湿、低气压和少雨天气有利于提高农田灯光诱虫的效率。
