高脂对中华鳖肝脏组织中microRNA及相关基因表达的影响

摘要:
为研究高脂对中华鳖肝脏组织中micro RNA及相关基因表达的影响,选取180只中华鳖随机分成两个实验组,分别投喂脂肪质量分数为7.98%(对照组)和13.86%(高脂日粮组)的饲料.定制光周期为12 h光照vs12 h黑暗.饲养6周后,每间隔3 h采集一次肝脏样品,研究高脂对中华鳖micro RNA及相关基因节律性表达的影响.结果表明,中华鳖肝脏组织中3个重要的micro RNA均具有节律性表达特征,且与生物钟核心基因(Clock,Bmal1和Bmal2)和脂肪代谢基因(Lxrα,PPARα和Sirt1)之间具有较强的相关关系.但高脂日粮改变了mi R-1a,mi R-21和mi R-34a节律性表达特征和表达水平,从而引起其相应靶基因(生物钟核心基因和脂肪代谢基因)的昼夜节律表达模式、m RNA表达水平及他们之间的相关关系发生相应改变.该结果为micro RNA参与调控中华鳖肝脏中脂肪代谢相关基因的节律表达提供新视角.

摘要:
Schizothorax prenanti (S. prenanti) is an indigenous fish species and is popularly cultured in southwestern China. In recent years, intensive farming of S. prenanti and water quality deterioration has increased the susceptibility of this fish to various pathogens, including Aeromonas hydrophila (A. hydrophila), which has caused severe damage to S. prenanti production. However, the understanding of molecular immune response of S. prenanti to A. hydrophila infection is still lacking. In order to better comprehend the S. prenanti time series immune response process against A. hydrophila, we conducted the first transcriptomic comparison in S. prenanti spleen at 4, 24, and 48 h after the infection challenge of A. hydrophila against their control counterparts. In total, 628 million clean reads were obtained from 18 libraries and assembled into 262,745 transcripts. After eliminating sequence redundancy, 69,373 unigenes with an average length of 1476 bp were obtained. Comparative analysis revealed 1890 unigenes with significantly differential expression, including 172, 455, 589 upregulated and 27, 676, 551 unigenes downregulated genes for 4, 24, and 48 h post-infection, respectively. Differentially expressed genes (DEGs) were validated using qPCR for 15 randomly selected genes. Enrichment and pathway analysis of DEGs was carried out to understand the functions of the immune-related genes. Our results revealed that many important functional genes relating to complement and coagulation cascades, chemokine signaling pathway, toll-like receptor signaling pathway, NOD-like receptor signaling pathway and leukocyte transendothelial migration were regulated during the infection of A. hydrophila, and the expression of those genes reflected the transcriptome profiles during the challenging stages.

将初始体重(31.75±7.20)g的试验用中华鳖480只分为4组,每组设3个重复,分别投喂添加0mg/kg、50mg/kg、100mg/kg和150mg/kg的血根碱制剂制的试验饲料75d,研究血根碱对中华鳖生长和血清指标的影响。试验结果显示,50mg/kg组、100mg/kg组的成活率显著高于150mg/kg组和0mg/kg组(P<0.05)。50mg/kg组及100mg/kg组血清中GLC含量显著低于150mg/kg组及0mg/kg组(P<0.05)。50mg/kg组AKP活性显著高于0mg/kg组(P<0.05)。50mg/kg组GSH-PX活性显著高于其他三组(P<0.05)。50mg/kg组与100mg/kg组AST活性显著低于0mg/kg组。试验结果表明,基础饲料中添加50mg/kg~100mg/kg血根碱有益于中华鳖的生长。

摘要:
为探究叶黄素对中华鳖体色、生长及血清指标的影响,分别在饲料中添加不同比例叶黄素,饲养初始体重(37.44±0.18)g的中华鳖幼鳖12周,结果显示:0.2%组增重率和特定增长率显著高于其他各组(P<0.05)。中华鳖腹部色值均大于背部,各组腹部L*值无显著差异,背部L*值随叶黄素增加先降低后增加;添加叶黄素能提高腹部b*值。各组背、腹皮肤类胡萝卜含量差异不显著,添加叶黄素可增加背部和肝脏类胡萝卜素含量,而降低腹部含量,其中0.2%组和0.3%组肝脏显著高于对照组(P<0.05);肌肉类胡萝卜素含量先增加后降低,0.2%组显著高于对照组。叶黄素对血清TG、T-CHO、ALT及SOD无显著影响(P>0.05);随叶黄素添加量的增加,AST活性降低;AKP和ACP活性随叶黄素的增加先增加后降低,GSH-Px则与之相反。综上,饲料中添加叶黄素能提高中华鳖生长性能、改善体色、提高肌肉和肝脏中类胡萝卜素含量,适宜的添加量为0.2%。

Zhushuqiao Reservoir is one of the main sources of drinking water for Changsha City, which is the capital of Hunan province in central China. Understanding phytoplankton functional groups and their relationships with the environment is critical for the sustainable management of the reservoir for the supply of drinking water. Therefore, phytoplankton and water samples were collected and analyzed seasonally in 2016 to detect these relationships, with a total of 23 functional groups of phytoplankton identified. The highest diversity of functional groups was observed in spring, while the lowest was in winter. Due to differences in the geographical position, as well as the hydrological and environmental conditions, upstream phytoplankton-community structures are different from those downstream, with the dominant functional groups having a clear seasonal succession. The succession dynamics of phytoplankton functional groups from upstream to downstream in spring were M (Microcystis aeruginosa) + Y (Cryptomonas sp.) → S1 (Pseudanabaena limnetica) + L M (Gomphosphaeria sp.) → S1 + L M + X1 (Chlorella sp.) → X1 + X2 (chlamydomonas sp.) + F (Oocystis sp.) → F + X1 + X2; X1, F and Lo (Peridinium sp.) were the co-dominant groups from summer to autumn, and group C (Melosira ambigua) was dominant in winter. The upstream assemblages of X1 + X2, X1 + F, S1 + L M + M in spring and L O in summer can adapt to the rapid growth in eutrophic conditions with potential bloom threats. Water temperature, light and phosphorus were the primary factors regulating the dominant functional groups, and the phytoplankton succession was seasonally dependent. These results may have implications for the management of future water resources in the Zhushuqiao Reservoir, such as the determination of the intake depth of drinking water, and the prevention of nutrient pollution. © 2018 E. 摘要:
采用四引物扩增受阻突变体系PCR法对中华鳖的2个SNP位点进行验证,并统计其基因型频率。采用方差分析法分析2个SNP位点与中华鳖6个生长性状(体质量、背甲长、背甲宽、腹甲长、体高和裙边宽度)的差异。结果显示,只有A–1609G位点上的体高和裙边宽度有显著差异;这2个SNP位点的不同基因型共组成6种双倍型(去掉频率小于3%的双倍型D1、D6和D9),ANOVA分析结果表明这6种双倍型的6个生长性状间均存在显著差异,D4双倍型6个生长性状的均值最大,属于生长优势双倍型。筛选到的IGF2基因上的2个SNP位点具有用作中华鳖生长性状分子标记辅助育种的潜力。

摘要:
A traditional genomewide association study (GWAS) detects genotype–phenotype associations by the vast number of genotyped individuals. This method requires large-scale samples and considerable sequencing costs. Extreme phenotypic sampling proposes make GWAS more cost-efficient and are applied more widely. With extreme phenotypic sampling, we performed a GWAS for n-3 highly unsaturated fatty acids (HUFA) and eviscerated weight (EW) traits in the large yellow croaker population. Of the 32,249 and 29,748 detected SNPs for the two traits, three candidate regions were found in each trait. Three candidate regions associated with HUFA were known near genes on chromosomes 4 and 11, and three candidate regions were on chromosome 6, and 15 for the EW trait. By combing through our GWAS results and the biological functional analysis of the genes, we suggest that the FABP, DGAT, ATP8B1, FAF2 and CERS2 genes, as well as the IGF2, BORA, CYP1A1, GRTP1 and HOX genes are promising candidate genes for n-3 HUFA and EW, respectively, in the large yellow croaker. Moreover, compared with the different numbers of the extreme phenotypic sampling, we conclude that 60% of the extreme phenotypic subsample can obtain a similar result as GWAS with whole phenotypes. Thus, extreme phenotypic sampling could save 40% of the cost for genotyping and DNA extraction without loss of the candidate regions and functional genes. Our study may provide a basis for further genomic breeding and a reference for others who want to perform GWAS with extreme phenotypes. © 2018, Indian Academy of Sciences.

p>The functional groups approach is an efficient way to analyze seasonal changes in phytoplankton biomass as it is based on the physiological, morphological, and ecological attributes of the species. In this study, we identified the functional groups and driving factors behind short-term succession in phytoplankton communities. We analyzed physical, chemical, and biological factors of the Maixi River in Baihua Reservoir (BHR) from August to September, 2013 (summer, phase I) and March to May, 2014 (late spring and early summer, phase II). The 226 samples collected were divided into 23 functional groups. In phase I, phytoplankton biomass ranged from 4.88 to 30.59 mg·L<sup>-1</sup>, and the group S1 ( Pseudanabaena limnetica ) had the greatest biomass. In phase II, phytoplankton biomass ranged from 2.22 to 50.61 mg·L<sup>-1</sup>, and groups Y ( Cryptomonas sp.) and S1 ( P. limnetica ) had the greatest biomass. Dominant functional groups in the Maixi River changed from S1 + D + Y + Lo in phase I to Y + S1 in summer. Changes in the phytoplankton community varied between 0 and 0.144 day<sup>-1 </sup>in phase I and between 0.008 and 0.389 day<sup>-1 </sup>in later spring and early summer. This showed a steady-state phytoplankton community during the two phases, in which the functional groups S1 ( P. limnetica ) and Y ( Cryptomonas sp.) were dominant. Pseudanabaena limnetica , Synedra sp., Peridinium sp., and Cryptomonas sp. were dominant during summer, contributing to 70% of the total biomass in the steady-state community, and P. limnetica , Synedra sp., Cryptomonas sp., and Chlamydomonas sp. were dominant during later spring and early summer, contributing to 60% of the total biomass in the community. Groups S1, D, and Y formed easily in the Maixi River, but P. limnetica was the dominant species in the eutrophic conditions of the Maixi River. According to biotic and abiotic factors, we concluded that the Maixi River is hypertrophic, and water resource management should take blooms of P. limnetica occurring in May into account. Temperature and dissolved oxygen were the critical factors affecting the steady-state of the phytoplankton community in late spring and early summer and summer, respectively. Because the Maixi River is an important source in the BHR, its phytoplankton functional groups directly affect the ecological balance of the water environment.</p

摘要:
为研究蚕蛹替代不同水平白鱼粉对中华鳖幼鳖表观消化率、消化酶及血液生化指标的影响。设计54%白鱼粉含量为对照组饲料,以5%、10%蚕蛹分别替代对照饲料组中2.7%、5.4%的白鱼粉,配制成3种等氮饲料,投喂平均体质量3.93 g的中华鳖幼鳖56 d。试验共设3个处理组,每组3个重复,每个重复20只中华鳖幼鳖。结果表明,用5%蚕蛹替代鱼粉对中华鳖幼鳖营养成分表观消化率无显著影响(P>0.05),而用10%蚕蛹替代白鱼粉时,干物质和粗脂肪的表观消化率显著降低;各替代组的消化酶活性无显著性差异(P>0.05)。同时,5.0%蚕蛹替代组对中华鳖的血液生化指标无显著影响,但是10%蚕蛹替代组的谷丙转氨酶、谷草转氨酶活性有上升的趋势(P>0.05)。说明5.0%蚕蛹替代2.7%白鱼粉没有降低中华鳖幼鳖对饲料的消化、内源酶的分泌及血液代谢。

摘要:
2013年8月至2014年7月, 采用组织学和实验生态学方法对钝缀锦蛤(Tapes conspersus)的性腺发育、生殖周期、肥满度、胚胎和幼虫发育进行了研究。结果显示, 钝缀锦蛤性腺发育以1年为1个周期, 性腺发育过程可分为增殖期、生长期、成熟期、排放期和休止期5个时期; 繁殖期在每年10月至翌年3月, 繁殖盛期为3月、10月和11月(水温20.3~24.6℃), 分批产卵。8月肥满度最高, 为33.43%; 4月最低, 为18.31%。钝缀锦蛤卵径为70~80 μm; 在水温24~ 26℃、盐度28~32条件下, 受精卵经16 h左右发育为D形幼虫; 初孵D形幼虫大小为90~110 μm, 浮游幼虫经7~8 d培育进入附着变态期, 此时壳长为210~230 μm, 再经4~5 d发育变态为稚贝。本研究为钝缀锦蛤的人工繁育及种质资源保护和利用提供了基础资料。

<正>近年来,随着大鲵养殖高密度集约化程度的提高和大鲵人工养殖年限的延长,大鲵病毒病的发病率和死亡率都呈现上升趋势,给大鲵养殖带来的危害越来越大,如大鲵虹彩病毒病、大鲵蛙病毒病等,由于大鲵病毒病目前无有效的药物治疗,也没有预防病毒病的相应有效疫苗制剂,所以给大鲵的人工集约化养殖带来的危害和损失非常大。本文主要介绍大鲵病毒病防控中存在的问题和实践中对大鲵病毒病的防控体会,希望给广大大鲵养殖者带来有益的参考。

摘要:
2013年8月至2014年7月, 采用组织学和实验生态学方法对钝缀锦蛤(Tapes conspersus)的性腺发育、生殖周期、肥满度、胚胎和幼虫发育进行了研究。结果显示, 钝缀锦蛤性腺发育以1年为1个周期, 性腺发育过程可分为增殖期、生长期、成熟期、排放期和休止期5个时期; 繁殖期在每年10月至翌年3月, 繁殖盛期为3月、10月和11月(水温20.3~24.6℃), 分批产卵。8月肥满度最高, 为33.43%; 4月最低, 为18.31%。钝缀锦蛤卵径为70~80 μm; 在水温24~ 26℃、盐度28~32条件下, 受精卵经16 h左右发育为D形幼虫; 初孵D形幼虫大小为90~110 μm, 浮游幼虫经7~8 d培育进入附着变态期, 此时壳长为210~230 μm, 再经4~5 d发育变态为稚贝。本研究为钝缀锦蛤的人工繁育及种质资源保护和利用提供了基础资料。

摘要:
为了研究嗜水气单胞菌(Aeromonas hydrophila)对齐口裂腹鱼(Schizothorax prenanti)血液生理生化指标的影响,将180尾齐口裂腹鱼平均分成6组(4、24和48 h对照组; 4、24和48 h感染组)。对照组腹腔注射0.2 mL 0.65%的无菌生理盐水,感染组腹腔注射0.2 mL 2.5 × 10~7 CFU/mL的嗜水气单胞菌液。分别测定4、24和48 h对照组和感染组齐口裂腹鱼各项血液生理生化指标,并对肾脏、肝脏和肌肉组织进行病理学观察。结果显示:谷丙转氨酶、白蛋白、总胆固醇、葡萄糖、乳酸脱氢酶、钾、钠、氯、钙和磷含量均有显著变化。谷草转氨酶、碱性磷酸酶、总蛋白、球蛋白、甘油三酯则没有受到显著影响。由此可见,嗜水气单胞菌感染后对齐口裂腹鱼血液指标产生了显著的影响。对病鱼的病理学观察发现病鱼体表多处出现充血出血,溃疡和腹水,尤其是肝脏、肾脏和肌肉组织均出现不同程度的损伤,变性和局部坏死。

摘要:
试验采用等蛋白替代方式,研究了蚕蛹粉替代白鱼粉对中华鳖幼鳖形体指标、生长性能及体成分的影响。试验配置了含54%白鱼粉的对照组饲料、含51.3%白鱼粉(5%蚕蛹替代组)和含48.6%白鱼粉(10%蚕蛹替代组)等氮饲料。结果表明:蚕蛹粉替代鱼粉对中华鳖幼鳖形体指标有显著影响,5%替代组的体比系数与10%替代组较接近,均显著低于对照组(P<0.05);5%替代组的脂体指数最低,同时生长性能下降;蚕蛹粉替代鱼粉对中华鳖幼鳖体成分影响不显著(P>0.05)。综合形体指标、生长性能及体成分分析表明,5%和10%蚕蛹粉替代白鱼粉降低了中华鳖幼鳖对饲料的利用率,且由于脂肪增加导致中华鳖幼鳖肉品质下降,因此,在中华鳖幼鳖配合饲料中不宜添加未脱脂的蚕蛹粉。

摘要:
In the present study, we sought to investigate the influence of high fat diet on the core clock genes and the muscle functional genes daily expression in the skeletal muscle of Chinese soft-shelled turtle. The turtles were fed by two diets including a control fat diet (the CON treatment, 7.98% lipid) and a high fat diet (the HFD treatment, 13.86% lipid) for six weeks and administrated by the photophase regimen of 24 h light/dark (12L:12D) cycle. After the feeding trial experiment, we measured the daily expression levels of 17 core clock genes (Clock, Bmal1/2, NPAS2, Tim, Cry1/2, Per1/2, DBP, AANAT, NIFL3, BHLHE40, NR1D2, RORA, RORB, RORC) and 12 muscle functional genes (FBXO32, MBNL1, MSTN, Myf5, Myf6, MyoD, MyoG, MyoM1, PPARa, PDK4, Trim63, UCP3) in the skeletal muscle of the two treatments. The results showed that except for Bmal1, NPAS2, Per2 and RORB, the expression of the other 13 core clock genes exhibited circadian oscillation in the CON treatment. Among the 12 muscle functional genes, MBNL1, PDK4 and MyoM1 did not exhibit circadian oscillation in the CON treatment. In the HFD treatment, the circadian rhythms expressional patterns of the 8 core clock genes (Clock, Bmal2, Cry2, Per1, DBP, NFIL3, BHLHE40 and RORA) and 6 muscle functional genes (MSTN, Myf5, MyoD, MyoG, PPARa and Trim63) were disrupted. In addition, compared with the CON treatment, the circadian expression of the 5 core clock genes (Tim, Cry1, AANAT, NR1D2, RORC) and the 3 muscle functional genes (FBXO32, Myf6, UCP3) showed the advanced or delayed expression peaks in the HFD treatment. In CON treatment, the circadian expression of the MyoG, MyoD, Myf6, FBXO32 and PPARa showed positive or negative correlation with the transcription pattern of Clock, Bmal2, Cry1/2, Per1/2. However, only the FBXO32 and Myf6 presented positive or negative correlation with the circadian expression of Cry1, RORB, AANAT and Tim in HFD treatment. In summary, these results demonstrate that the disruption of the circadian rhythm of the core clock genes and muscle functional genes in the skeletal muscle is closely associated with feeding high fat diet to Chinese soft-shelled turtle. © 2017 Elsevier Inc.
