摘要:
综述了烟草香味物质提取新技术,包括超临界CO2萃取、微波萃取、双水相萃取、超声波提取、酶法提取等,分离纯化技术包括分子蒸馏技术、膜分离技术、大孔吸附树脂分离法、高速逆流色谱法、分子印迹技术.通过多种提取分离方法的例举阐述了烟草制品用烟草香味物质的研究现状,并对烟草香味物质提取与分离纯化技术的发展前景进行了展望.

摘要:
Identifying important regulative elements and pathways related to dwarfism in cotton is a major challenge in cotton breeding. Isobaric tags for relative and absolute quantitation-based proteomics of stem terminal buds from upland cotton (Gossypium hirsutum L.) dwarf line LA-1 and high near-isogenic line LH-1 was performed. Moreover, transcriptional expression of differentially-expressed proteins (DEPs) belonging to phytohormone signal transduction, hormone biosynthesis, and ubiquitin system were analysed using quantitative real-time polymerase chain reaction. A total of 4 849 proteins were identified from LA-1 and LH-1, 697 of which showed differential accumulations. Most of the DEPs have catalytic, binding, and transporter activity and are involved in metabolism- and protein processing-related pathways. In particular, seven DEPs, including two gibberellin (GA) receptors, three cytokinin (CK) receptors, CKoxidase, and CK-N-glucosyltransferase were up-regulated in LA-1, and GA20-oxidase was down-regulated in LH-1. Our results suggest that the DELLA-independent GAsignalling pathway is the primary cause of dwarfism in LA-1 and indicate that CKresponse element 1-2, GA-insensitive dwarf, CKoxidase, and GA3-β-dioxygenase are potential indicators of dwarf cotton. The profiling of DEPs may offer a valuable resource for cotton breeding. © 2017, Springer Science+Business Media Dordrecht.

摘要:
Sinapis alba possesses a number of desirable traits that can be employed to broaden genetic variability in Brassica oleracea or B. napus. Using transgenic B. oleracea (CC, 2n = 18) as paternal plants and non-transgenic S. alba (SS, 2n = 24) as maternal plants, 11 hybrid plants were successfully obtained by using a combination of ovary culture and embryo rescue, while 2 seedlings of the reciprocal cross were generated but lost during the propagation. The hybridity of these plants was confirmed by flower color and other agronomic characteristics, chromosome counting, and pollen viability. Most of the intergeneric F1 plants showed only one set each of S and C chromosome (SC, 2n = 21) and were male sterile, while a few plants were semi-fertile and had one set of S chromosomes but two sets of C chromosomes (CCS, 2n = 30). PCR, genomic Southern blotting and qRT-PCR for bar gene revealed that among the F1 hybrid plants only 9.1% was bar positive and could be forwarded to F2 and F3 generation. The majority of F3 plants obtained sufficient resistance to Alternaria brassicae. Thus, the generation of disease (A. brassicae) and herbicide (Bastar) resistant intergeneric hybrids was of importance for breeding program. The existence of bar gene in these intermediate materials will facilitate the identification of hybridity and transfer of S.alba traits into target genetic background. © 2017, The Author(s).

摘要:
With the massive data generated by the Human Microbiome Project, how to transform such data into useful information and knowledge remains challenging. Here, with currently available sequencing information (reference genomes and metagenomes), we have developed a comprehensive microarray, HuMiChip2, for strain-level identification and functional characterization of human microbiomes. HuMiChip2 was composed of 29,467 strain-specific probes targeting 2063 microbial strains/species and 133,924 sequence- and group-specific probes targeting 157 key functional gene families involved in various metabolic pathways and host-microbiome interaction processes. Computational evaluation of strain-specific probes suggested that they were not only specific to mock communities of sequenced microorganisms and metagenomes from different human body sites but also to non-sequenced microbial strains. Experimental evaluation of strain-specific probes using single strains/species and mock communities suggested a high specificity of these probes with their corresponding targets. Application of HuMiChip2 to human gut microbiome samples showed the patient microbiomes of alcoholic liver cirrhosis significantly (p < 0.05) shifted their functional structure from the healthy individuals, and the relative abundance of 21 gene families significantly (p < 0.1) differed between the liver cirrhosis patients and healthy individuals. At the strain level, five Bacteroides strains were significantly (p < 0.1) and more frequently detected in liver cirrhosis patients. These results suggest that the developed HuMiChip2 is a useful microbial ecological microarray for both strain-level identification and functional profiling of human microbiomes.

【目的】通过盆栽试验探讨了在施加不同水平的生石灰时,酸性土壤pH值及微生物群落功能多样性的变化量。【方法】本实验采用Biolog-ECO检测法,探究了在4种不同生石灰施用量CK、11、T2、T3(0、1125、2250、3375 kg/hm~2)下酸性土壤微生物群落碳源代谢的多样性变化。【结果】①在240 h的培养期内,施加不同量生石灰条件下微生物群落单孔平均颜色变化率AWCD值从高到低依次是T2>T1>T3>CK。②土壤pH值与AWCD值相关分析表明,两者呈中度正相关。③土壤微生物群落功能多样性指数结果显示,T1、T2、T3处理的Simpson指数显著高于CK处理,而Shannon指数与McIntosh指数在各处理之间的差异不显著。④主成分分析结果表明,T3、CK处理土壤微生物碳源利用方式相似,而T1、T2处理有不同的碳源利用方式。【结论】由此可见,每公顷施加2250 kg的生石灰(T2处理)能提高土壤pH值、土壤AWCD值、物种优势度指数与微生物群落碳源利用能力,对微生物多样性的提高有较大的促进作用。

摘要:
As China requires more cultivated land to feed its increasing population, the sustainable development of cultivated land in its intensive agricultural production system has become a major issue for government and society. We firstly reviewed the progress of paddy-upland rotation (PUR) and conservation tillage, found that PUR was improve significantly soil properties, including redox potential of soil, the mineralization of soil organic matter, and the abundance of beneficial microorganisms, suggested that conservation tillage was an effective measurement for remedying the deficiency under the PUR. Meanwhile, we also systemically reviewed the statues of water resource infrastructure and policy level. Consequently, with current agriculture technology and the increasing capacity of adjusted artificial water resources, our results highlight the development of national-scale PUR as an effective adaptation strategy to protect and improve soil fertility and ensure agricultural security under climate change. In other words, from national strategy scale, national-scale PUR was a good choice in the major grain-producing areas of northern China in the coming 50-100 years. Lastly, further research in the few aspects was needed to better provide theoretical and technical support for promoting effectively the PUR. © 2016 Elsevier B.V.

为探讨生物炭作为土壤改良剂对植烟土壤养分含量的影响,以湖南烟区主要栽培品种云烟87为试验材料,通过大田试验研究了不同的生物炭用量(CK: 0 kg /hm~2; T1: 3 000 kg /hm~2; T2: 3 750 kg /hm~2; T3: 4 500 kg /hm~2)下植烟土壤中的养分含量。结果表明: 施用生物炭能够显著提高植烟土壤有机质、碱解氮、铵态氮及速效钾含量,且随着生物炭施用量的增加呈先升高后降低的趋势,从改良土壤肥力效果来看,生物炭施用量在3 750 kg /hm~2的水平下较为适宜。

摘要:
为了探明不同生物炭用量与氮肥配施对烤烟生长及烟叶主要化学成分的影响,以云烟87为试材,通过田间小区试验研究了9种不同的生物炭与氮肥配施比例对烤烟生长及烟叶主要化学成分的影响。结果表明:生物炭与氮肥配施能够调控烤烟的生长,促进烟株的生长发育,加快出叶速度,但施炭量与对烤烟生长的促进作用不呈正相关,当施炭量超过3 750 kg/hm~2、施氮量超过112.5 kg/hm~2时对烤烟生长的促进作用逐渐下降。生物炭与氮肥配施能够在一定程度上促进根系的发育,提高烤烟干物质积累量。生物炭与氮肥配施能够调节烤后烟叶的总糖与还原糖含量至最适范围内,降低烟叶淀粉含量,提高烟叶钾含量,有利于提高烟叶燃烧性。同时合理配施生物炭与氮肥也能改善烟叶的糖碱比、糖氮比、钾氯比使之趋近于适宜值,但当配施比例较低时即施炭量为3 000 kg/hm~2、施氮量为75.0 kg/hm~2的配施比例不利于改善烤烟糖氮比以及钾氯比。综合来看,以施炭量3 750 kg/hm~2,施氮量112.5 kg/hm~2的配施比例适宜,烟株的生长发育较快,烤后烟叶的主要化学成分含量适宜。

Plant height is a key morphological trait of rapeseed. In this study, we measured plant height of a rapeseed population across six environments. This population contains 476 inbred lines representing the major Chinese rapeseed genepool and 44 lines from other countries. The 60K Brassica Infinium® SNP array was utilized to genotype the association panel. A genome-wide association study (GWAS) was performed via three methods, including a robust, novel, nonparametric Anderson-Darling (A-D) test. Consequently, 68 loci were identified as significantly associated with plant height (P < 5.22 × 10−5), and more than 70% of the loci (48) overlapped the confidence intervals of reported QTLs from nine mapping populations. Moreover, 24 GWAS loci were detected with selective sweep signals, which reflected the signatures of historical semi-dwarf breeding. In the linkage disequilibrium (LD) decay range up—and downstream of 65 loci (r2 > 0.1), we found plausible candidates orthologous to the documented Arabidopsis genes involved in height regulation. One significant association found by GWAS colocalized with the established height locus BnRGA in rapeseed. 为了改良土壤容重,筛选优化生物炭施用量,以湖南烟区主要栽培品种云烟87为试验材料,通过大田试验研究3种不同的生物炭用量对田间土壤改良及烤烟生长的影响。结果表明,施用生物炭能够显著降低土壤容重,提高田间持水量,土壤容重与施炭量呈负相关,田间持水量与施炭量呈正相关。生物炭用量在3 750kg·hm~(-2)水平下,烤烟的植物学性状优良,干物质积累量大,烟株根系发达,根冠比协调,且氮肥利用率显著提高,但随着施炭量继续升高,烤烟的生长发育又相对受到抑制。综上所述,生物炭施用量为3 750kg·hm~(-2)较为适宜。本研究结果对利用生物炭改良植烟土壤,促进优质烟叶生产有一定的理论指导意义。

长沙烟稻轮作区烤烟正常与推迟采收条件下的适宜晚稻品种筛选

摘要:
为了筛选出适应长沙烟稻轮作区不同烟叶采收期的晚稻品种,模拟烤烟正常采收与推迟采收2种条件,以15个晚稻品种为材料开展了大田品种比较试验。结果表明:(1)从全生育期角度,6月18日播种条件下,所有品种均适合作烟后晚稻种植,而6月28日播种条件下仅‘湘丰优103’、‘隆香优130’、‘深优9520’、‘湘丰优9号’、‘金优284’、‘丰源优299’、‘丰源优272’能够正常成熟。(2)从产量角度,6月18日播种条件下,‘两优527’、‘丰源优272’和‘隆香优130’产量可达8000 kg/hm^2左右,是烟稻轮作区烟叶正常采收条件下烟后晚稻品种的首选;6月28日播种条件下,‘丰源优272’、‘湘丰优103’、‘深优9520’和‘湘丰优9号’是较佳选择,产量可达7600 kg/hm^2以上。(3)相关分析表明,产量与每穗粒数呈显著正相关,与经济系数呈正相关,因此选择穗型较大、经济系数较高的晚稻品种易获得高产。结果对长沙烟区烟后晚稻品种选择具有一定指导作用。

为给浏阳烟区烤烟生产提供施肥技术指导,通过研究施氮量对烤烟经济性状与烟叶化学成分的影响,探讨了浏阳烟稻轮作区烤烟的适宜施氮量。结果表明:(1)施氮120 kg/hm2不能满足烟株生长发育的需要,增大施氮量可显著改善烟株农艺性状,提高烟株物质积累量;(2)各处理经济性状差异显著,施氮150~210 kg/hm2处理产量与产值较施氮120 kg/hm2处理显著提高,其中施氮150 kg/hm^2处理上中等烟比例与均价最高,产量与品质协调较好;(3)上部和下部烟叶以施氮150 kg/hm2处理的总氮和烟碱含量较高,化学成分协调性较好;而中部烟叶以施氮180 kg/hm^2处理的总氮和烟碱含量相对较高。综合考虑烤烟生长发育、烟叶产量与品质,浏阳烟稻轮作区烤烟的适宜施氮量为150 kg/hm2。

湖南浓香型烟叶化学成分与物理性状相关性分析

摘要:
为探索烤烟烟叶质量评价指标间的关系,于2011-2013年间采集湖南257个烟叶样品,通过简单相关和典型相关研究化学成分与物理性状的相关性。结果表明:湖南烤烟化学成分多数指标间呈极显著相关;物理性状除叶长、平衡含水率与含梗率之间无显著相关外,其余指标间均呈显著或极显著相关;化学成分与物理性状之间存在显著相关,主要由总糖与叶长(正相关)、叶宽(正相关)、单叶重(负相关),总氮与单叶重(正相关)、叶面积质量(正相关),烟碱与单叶重(正相关)、叶面积质量(正相关)、叶长(正相关)、叶宽(负相关)的相关性决定。研究结果为丰富烟叶质量评价体系提供了一定的参考。

长沙烟区烟后晚稻适宜氮肥施用量与运筹方式研究

摘要:
为给长沙烟区烟后晚稻高产高效栽培提供科学依据,以超级杂交稻品种深优9586为材料,于2011~2012年在长沙市宁乡烟区研究了施氮量与氮肥运筹方式对烟后晚稻产量形成与氮素吸收利用的影响。结果表明:(1)在施氮0~120 kg/hm~2范围内,晚稻产量随施氮量增加呈先增后降趋势,施氮90 kg/hm~2条件下产量最高,过量施氮导致减产的原因在于有效穗数与千粒重下降;(2)氮肥运筹方式影响晚稻产量,施氮90 kg/hm~2时W3处理(基蘖肥∶穗肥∶粒肥比例为0.5∶0.25∶0.25)产量最高,施氮30~60 kg/hm~2时以W4处理(0.25∶0.5∶0.25)产量最高,增产的原因在于氮肥后移提高了烟后晚稻齐穗期叶面积指数、灌浆中期叶面积指数和SPAD值;(3)氮肥用量增大,烟后晚稻氮积累量提高,但氮农学利用率、氮肥吸收利用率、氮生理利用率、氮素干物质生产效率、氮素稻谷生产效率下降;氮肥运筹方式影响氮素利用效率,施氮量较低时,氮肥后移有利于氮素利用效率的提高,而施氮量偏高时(90 kg/hm~2)后期施用氮肥比例不宜过高。可见,为提高长沙烟区烟后晚稻产量和氮素利用效率,施氮量在60 kg/hm~2以内可采取氮肥后移策略,而施氮90 kg/hm~2时后期施氮比例不宜过高。

摘要:
为明确长沙烟区烟稻复种体系烟后晚稻的适宜施氮量和种植密度,以丰源优299为材料,于2013-2014年在湖南省长沙市浏阳市开展了烟后晚稻的肥密互作效应研究。结果表明:烟后晚稻全生育期因施氮延长2d,与密度无明显关系;水稻LAI整体随施氮量和密度增大而提高,但是施氮量和密度间存在互作效应,且两年的互作效应有差异;烟后晚稻叶片SPAD值因施氮而明显提高,与密度关系不大;水稻产量构成因素中,千粒重较稳定,不因密度与施氮量变化而发生显著变化;密度与施氮量对烟后晚稻产量构成因素有明显互作效应,较低密度(152 000-200 000穴/hm^2)条件下,有效穗数、穗粒数和产量随密度和施氮量增大而明显提高,而较高密度(210000-300000穴/hm^2)条件下,结实率随施氮量增大而增大,有效穗数、穗粒数和产量随施氮量增大表现先升后降趋势,密度300 000穴/hm^2、施氮90kg/hm^2处理表现最好。

摘要:
对湖南省浓香型烟区2011—2012年111个烤烟样品的化学成分和外观质量进行了基本统计特征分析,并采用灰色关联分析研究了外观质量与常规化学指标之间的关系。结果表明,烟叶化学成分各项指标都是右偏态分布,外观质量除颜色和成熟度指标外,其它各指标是左偏态分布,烟叶总氮、总糖、烟碱含量与烟叶外观质量各项指标关系最密切,氯含量与烟叶外观质量关系最远。

摘要:
以烤烟品种G80为材料,在浏阳烟区研究了施肥量与种植密度对烤烟农艺性状和经济性状的耦合效应。结果表明:烤烟全生育期因施氮量增加而延长5~7 d,随密度增加而略有缩短3~4 d,即低氮高密处理全生育期最短;烤烟农艺性状因施氮量增大与密度降低而有所改善;试验条件下,以施氮量180 kg/hm2和密度16500~18000株/hm2的耦合效应最佳,产量分别为2383.5与2364.0 kg/hm2,增产12.9%~52.3%与12.0%~51.0%;产值分别为63043.5与62622.3元/hm2,增值11.7%~53.7%与10.9%~52.7%。

摘要:
为研究湖南浓香型烟叶产区烤炯化学成分变化规律,于2011年和2012年采集炯叶样品111个,对不同年份、不同等级、不同品种进行比较分析.结果表明:2012年的总糖、还原糖、糖碱比以及钾氯比与2011年相比有所增长,2012年的总氮、烟碱、钾、氯、淀粉以及氮碱比与201 1年相比有所下降.各化学成分指标在不同等级中差异显著或极显著,且2011年与2012年的等级变化规律基本一致.总糖、还原糖、钾、糖碱比、氮碱比、钾氯比变化为:X2F>C3F>B2F;总氮、烟碱、氯的变化为:B2F>C3F>X2F.同一化学成分在不同品种之间的变化规律不明显.

摘要:
To introgress the good fiber quality and yield from Gossypium barbadense into a commercial Upland cotton variety, a high-density simple sequence repeat (SSR) genetic linkage map was developed from a BC1F1 population of Gossypium hirsutum×Gossypium barbadense. The map comprised 2,292 loci and covered 5115.16 centiMorgan (cM) of the cotton AD genome, with an average marker interval of 2.23 cM. Of the marker order for 1,577 common loci on this new map, 90.36% agrees well with the marker order on the D genome sequence genetic map. Compared with five published high-density SSR genetic maps, 53.14% of marker loci were newly discovered in this map. Twenty-six quantitative trait loci (QTLs) for lint percentage (LP) were identified on nine chromosomes. Nine stable or common QTLs could be used for marker-assisted selection. Fifty percent of the QTLs were from G. barbadense and increased LP by 1.07%-2.41%. These results indicated that the map could be used for screening chromosome substitution segments from G. barbadense in the Upland cotton background, identifying QTLs or genes from G. barbadense, and further developing the gene pyramiding effect for improving fiber yield and quality. © 2014 Institute of Botany, Chinese Academy of Sciences.

摘要:
Flowering time adaptation is a major breeding goal in the allopolyploid species Brassica napus. To investigate the genetic architecture of flowering time, a genome-wide association study (GWAS) of flowering time was conducted with a diversity panel comprising 523 B. napus cultivars and inbred lines grown in eight different environments. Genotyping was performed with a Brassica 60K Illumina Infinium SNP array. A total of 41 single-nucleotide polymorphisms (SNPs) distributed on 14 chromosomes were found to be associated with flowering time, and 12 SNPs located in the confidence intervals of quantitative trait loci (QTL) identified in previous researches based on linkage analyses. Twenty-five candidate genes were orthologous to Arabidopsis thaliana flowering genes. To further our understanding of the genetic factors influencing flowering time in different environments, GWAS was performed on two derived traits, environment sensitivity and temperature sensitivity. The most significant SNPs were found near Bn-scaff_16362_1-p380982, just 13 kb away from BnaC09g41990D, which is orthologous to A. thaliana CONSTANS (CO), an important gene in the photoperiod flowering pathway. These results provide new insights into the genetic control of flowering time in B. napus and indicate that GWAS is an effective method by which to reveal natural variations of complex traits in B. napus.
