This study demonstrates that GT79 based nontarget‐site resistance likely confers resistance to cyhalofop‐butyl in an Echinochloa crus‐galli population. Abstract BACKGROUND Echinochloa crus‐galli is the most troublesome and widespread weed of most rice‐growing regions of the world. Cyhalofop‐butyl, a herbicide within the acetyl‐CoA carboxylase (ACCase) chemical group, has been extensively used to control barnyardgrass in rice. The repeated exposure to cyhalofop‐butyl has led to resistance evolution in E. crus‐galli populations. RESULTS In this study, we identified a population of E. crus‐galli (R‐HN) in a rice field in Hunan, China, that developed resistance to cyhalofop‐butyl at 4.49‐fold the recommended field dose. No known target mutation was detected in the ACCase gene of the R‐HN population by ACCase sequencing compared to sensitive populations. Both cytochrome P450 (CYP450) and glutathione S‐transferase (GST) inhibitors could not significantly reverse the resistance to cyhalofop‐butyl. The nontarget‐site resistance (NTSR) mechanism was investigated by transcriptome sequencing. Validation of the screened candidate genes by quantitative real‐time (qRT)‐PCR revealed that six glycosyltransferases (GTs) and four ATP‐binding cassette (ABC) transporter genes were consistently upregulated in the R‐HN population. Five GTs and one ABC transporter genes were constitutively upregulated after cyhalofop‐butyl treatment in the R‐HN population. Molecular docking results showed that the significant binding energy of GT79, GT75L6 and GT74E among all candidate genes. CONCLUSION Thus, the GT genes appear to be directly implicated in NTSR to cyhalofop‐butyl in the R‐HN populations through metabolic enhancement, but their functional characterization needs to be studied. © 2024 Society of Chemical Industry.

稻田稗草种群对氰氟草酯的抗性水平和抗性机理初探

摘要:
为明确来自湖南和江西省稻田稗草种群对氰氟草酯的抗性水平和抗性机理,采用整株生物测定法、基因测序、代谢酶抑制剂联用试验进行了研究。结果显示:R1、R2、R3种群对氰氟草酯的抗性指数分别为3.08、2.19、3.83,均为中抗水平;R4种群抗性指数为1.45,为低抗水平。与敏感种群相比,R1种群存在Ile-2041-Val突变,R2种群中存在Ile-2041-Val、Asp-2078-Gly突变,且细胞色素P450酶抑制剂马拉硫磷和谷胱甘肽硫转移酶抑制剂NBD-Cl均能提高R1和R2种群对氰氟草酯的敏感性;R3种群中存在Trp-2027-Arg突变,且NBD-Cl可以提高其对氰氟草酯的敏感性;R4种群中存在Cys-2088-Tyr突变,马拉硫磷和NBD-Cl不能逆转其对氰氟草酯的抗性。研究表明,稗草种群R1、R2、R3对氰氟草酯的中等水平抗性是由靶标抗性和非靶标抗性共同介导,种群R4对氰氟草酯的低水平抗性主要是由靶标抗性介导。

Background: Leptochloa chinensis (L.) Nees is a troublesome weed across China in rice fields, and a suspected L. chinensis resistant population (R) that has survived the recommended field dose of cyhalofop-butyl was collected in a rice field of Hunan Province, China. In this study, we aimed to determine the acetyl-CoA carboxylase-inhibiting herbicide resistance profile of this R population and to investigate its mechanisms of resistance to cyhalofop-butyl. Results: Compared with the susceptible population (S), the R population was confirmed to be 18.9-, 3.2-, 4.1-, 3.6- and 5.8- fold resistant to the APP herbicides cyhalofop-butyl, haloxyfop-P-methyl, clodinafop-propargyl, metamifop and fenoxaprop-P-ethyl, respectively. ACCase gene sequencing analysis revealed no known resistance mutations for TSR in the R population. Pretreatment with the glutathione S-transferase (GST) inhibitor 4-chloro-7-nitrobenzoxadiazole (NBD-Cl) and cytochrome P450 (CYP450) inhibitor malathion reversed resistance to cyhalofop-butyl. The GST gene GSTU1 and CYP450 gene CYP707A5 were constitutively upregulated in the R population according to RNA-seq analysis and RT-qPCR verification. The molecular docking results indicated a good affinity of the active site for five APP herbicides with GSTU1 and CYP707A5. Conclusion: This study shows that the GSTU1 and CYP707A5 genes expressed highly in the R population may be responsible for cyhalofop-butyl resistance in L. chinensis. © 2023

Eleusine indica has become a global nuisance weed and has evolved resistance to glufosinate. The involvement of target-site resistance (TSR) in glufosinate resistance in E. indica has been elucidated, while the role of nontarget-site resistance (NTSR) remains unclear. Here, we identified a glufosinate-resistant (R) population that is highly resistant to glufosinate, with a resistance index of 13.5-fold. Molecular analysis indicated that the resistance mechanism of this R population does not involve TSR. In addition, pretreatment with two known metabolic enzyme inhibitors, the cytochrome P450 (CYP450) inhibitor malathion and the glutathione S-transferase (GST) inhibitor 4-chloro-7-nitrobenzoxadiazole (NBD-Cl), increased the sensitivity of the R population to glufosinate. The results of subsequent RNA sequencing (RNA-seq) and quantitative real-time PCR (RT-qPCR) suggested that the constitutive overexpression of a GST gene (GSTU3) and three CYP450 genes (CYP94s and CYP71) may play an important role in glufosinate resistance. This study provides new insights into the resistance mechanism of E. indica. © 2023 American Chemical Society

【目的】筛选防治马唐(Digitariasanguinalis)的生防真菌,挖掘其应用于杂草生物防治的潜力。【方法】采用组织分离法,从自然发病的马唐植株中分离出对马唐具有较强致病力的生防菌株,通过形态学观察、ITS、β-...展开更多 【目的】筛选防治马唐(Digitariasanguinalis)的生防真菌,挖掘其应用于杂草生物防治的潜力。【方法】采用组织分离法,从自然发病的马唐植株中分离出对马唐具有较强致病力的生防菌株,通过形态学观察、ITS、β-Tubulin、EF-1α基因序列同源性比对及其系统进化分析确定其分类地位;利用单因素试验研究其生物学特性及整株生物测定法测定马唐生防真菌对几种常见杂草的侵染效果及对常见作物的安全性。【结果】分离得到的马唐生防真菌MA4被鉴定为藤仓镰刀菌(Fusarium fujikuroi);最适合MA4生长的培养基为PDA和PSA;最适合MA4生长和产孢的培养条件为:温度28~30℃,p H值6.0~8.0,黑暗培养;MA4对马唐鲜重防效较好,达到91.4%,对看麦娘、狗尾草和钻形紫菀的鲜重抑制率分别为63.8%、80.6%、69.4%;MA4对双子叶作物相对安全。【结论】藤仓镰刀菌MA4菌株对马唐等禾本科杂草防治效果较好,对双子叶作物较安全,具有开发成生物除草剂的巨大潜力。收起

为了评价4种水稻田新除草剂对后茬烟草的安全性,通过室内盆栽和田间小区试验评估了4种水稻田新除草剂对烟草生长的影响,并采用QuEChERS前处理方法结合高效液相色谱-串联三重四极杆质谱(HPLC-MS/MS)联用技术检测了...展开更多 为了评价4种水稻田新除草剂对后茬烟草的安全性,通过室内盆栽和田间小区试验评估了4种水稻田新除草剂对烟草生长的影响,并采用QuEChERS前处理方法结合高效液相色谱-串联三重四极杆质谱(HPLC-MS/MS)联用技术检测了二氯喹啉草酮在烟田土壤中的残留水平。结果表明,氯氟吡啶酯、氟酮·呋喃酮和硝磺草酮对烟草生长没有明显影响。在750~2 250 g a.i./hm2剂量下,二氯喹啉草酮对盆栽和大田试验中烟株叶长、叶宽、株高的抑制率分别为48.27%~69.00%、66.30%~78.08%、58.76%~75.83%和24.89%~43.79%、35.47%~49.81%、28.87%~48.75%,二氯喹啉草酮对后茬烟草造成了严重药害。次年移栽烟苗时土壤中未检出二氯喹啉草酮(低于LOQ,0.002mg/kg)。因此,水稻田除草剂二氯喹啉草酮对后茬烟草存在较大药害风险,在烟稻轮作产区应谨慎使用。收起

摘要:
为了评价几种除草剂对烟草田杂草的防除效果及对烟草的安全性,在浏阳市烟草基地开展了田间试验.结果表明,供试的除草剂对烟草安全性较好.土壤处理剂中,80%异·异丙甲乳油、50%仲灵·异松乳油、45%甲戊·异丙甲乳油这3种复配除草剂对烟草田杂草的防治效果较好,在药后30 d对杂草的株防效为89.20%~93.92%,药后60 d对杂草的株防效为83.38%~88.53%,鲜重防效为86.53%~90.36%,均要显著优于常规对照药剂960 g/L精异丙甲草胺.行间定向喷雾茎叶处理剂中,250 g/L氟磺胺草醚AS+10%精喹禾灵EC、10%乙羧氟草醚EC+10%精喹禾灵EC、200 g/L敌草快AS+10%精喹禾灵EC等3种复配除草剂的防效较好,药后10 d对烟草田杂草的株防效为94.98%~95.75%,药后20 d对烟草田杂草的株防效为93.86%~96.03%,鲜重防效为94.98% ~96.41%,均要显著优于烟草田常规使用药剂25%砜嘧磺隆WG+10%精喹禾灵EC.在供试除草剂中,除二甲戊灵对烟草生长表现轻微抑制作用外,其余除草剂对烟草农艺性状均表现为一定的促进作用,但效果不显著.试验表明,上述除草剂及复配剂可以用于烟草田进行合理轮换使用,延缓烟草田杂草抗药性产生并提高烟草田杂草防治水平.

摘要:
为了明确湖南省烟田杂草种群与分布,采用对角线五点取样法在全省烟田进行了系统调查。结果表明,烟田杂草共有30科77种,其中禾本科杂草占比14.29%,阔叶杂草占比81.82%,莎草科杂草占比3.89%;一年生杂草占比75.32%,多年生杂草占比24.68%。相对多度大于3%的杂草有铁苋菜、酸模叶蓼、稗、碎米荠、马唐、繁缕、喜旱莲子草、看麦娘、通泉草、鸭跖草、水苦荬等11种,其中铁苋菜、酸模叶蓼、稗为湖南省烟田优势杂草种群,碎米荠、马唐、繁缕、喜旱莲子草、看麦娘、通泉草、鸭跖草、水苦荬为湖南省烟田区域性优势杂草种群。湖南省烟田杂草防除主要采用苗前土壤处理和苗后茎叶处理相结合的化学防除方法,主要使用的化学除草剂是精异丙甲草胺和砜嘧磺隆。本研究结果表明,湖南省烟田杂草存在区域性差异,应因地制宜制定合适的化学防治方案。

摘要:
Cereal cyst nematodes are sedentary biotrophic endoparasites that secrete effector proteins into plant tissues to transit normal cells into specialized feeding sites and suppress plant defenses. To understand the function of nematode effectors in Heterodera avenae, here, we identified a calreticulin protein HaCRT1, which could suppress the cell death induced by Bax when expressed in Nicotiana benthamiana. HaCRT1 is synthetized in the subventral gland cells of pre-parasitic second-stage nematodes. Real-time PCR assays indicated that the expression of HaCRT1 was highest in parasitic second-stage juveniles. The expression of an HaCRT1-RFP fusion in N. benthamiana revealed that it was localized in the endoplasmic reticulum of the plant cell. The ability of H. avenae infecting plants was significantly reduced when HaCRT1 was knocked down by RNA interference in vitro. Arabidopsis thaliana plants expressing HaCRT1 were more susceptible than wild-type plants to Pseudomonas syringae. The induction of defense-related genes, PAD4, WRKY33, FRK1, and WRKY29, after treatment with flg22 was suppressed in HaCRT1-transgenic plants. Also, the ROS accumulation induced by flg22 was reduced in the HaCRT1-transgenic plants compared to wild-type plants. HaCRT1 overexpression increased the cytosolic Ca2+ concentration in A. thaliana. These data suggested that HaCRT1 may contribute to the pathogenicity of H. avenae by suppressing host basal defense.

采用组织分离法分离了油茶炭疽病病原菌YCTJ,通过柯赫氏法则、形态学和分子生物学特征以及系统发育树对病原菌进行了鉴定,并采用生长速率法测定了400 g/L氟硅唑乳油等10种杀菌剂对油茶炭疽病菌的室内药效.结果表明:经鉴定,证实引起油茶炭疽病的病原菌为胶孢炭疽菌;供试的10种杀菌剂中,400 g/L氟硅唑乳油、10% 苯醚甲环唑微乳剂、80% 戊唑醇可湿性粉剂、30% 肟菌 · 戊唑醇悬浮剂和43% 氟菌 · 肟菌酯悬浮剂这5种杀菌剂对油茶炭疽病菌的抑制效果在98.41%~100% 之间,可以作为油茶炭疽病田间防治的备选药剂.

The root-knot nematode Meloidogyne graminicola is an important pathogen in rice, causing huge yield losses annually worldwide. Details of the interaction between rice and M. graminicola and the resistance genes in rice still remain unclear. In this study, proteome-wide analyses of the compatible interaction of the japonica rice cultivar “Nipponbare” (NPB) with M. graminicola were performed. In total, 6072 proteins were identified in NPB roots with and without infection of M. graminicola by label-free quantitative mass spectrometry. Of these, 513 specifically or significantly differentially expressed proteins were identified to be uniquely caused by nematode infection. Among these unique proteins, 99 proteins were enriched on seven Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. By comparison of protein expression and gene transcription, LOC_Os01g06600 (ACX, a glutaryl-CoA dehydrogenase), LOC_Os09g23560 (CAD, a cinnamyl-alcohol dehydrogenase), LOC_Os03g39850 (GST, a glutathione S-transferase) and LOC_Os11g11960 (RPM1, a disease resistance protein) on the alpha-linolenic acid metabolism, phenylpropanoid biosynthesis, glutathione metabolism and plant–pathogen interaction pathways, respectively, were all associated with disease defense and identified to be significantly down-regulated in the compatible interaction of NPB with nematodes, while the corresponding genes were remarkably up-regulated in the roots of a resistant rice accession “Khao Pahk Maw” with infection of nematodes. These four genes likely played important roles in the compatible interaction of rice with M. graminicola. Conversely, these disease defense-related genes were hypothesized to be likely involved in the resistance of resistant rice lines to this nematode. 摘要:
采用组织分离法,从水稻健叶分离得到一株对水稻纹枯病有较强拮抗作用的内生拮抗细菌WK1,经16S rDNA序列同源性和进化树分析,表明WK1菌株为贝莱斯芽孢杆菌(Bacillus velezensis),在此基础上采用混药平板法测定WK1对水稻纹枯病菌菌丝的抑制率,结果为86.67%,进而进行田间防效试验.结果 表明:WK1菌株对水稻纹枯病的防治效果为52.23%,与5%井冈霉素水剂防效相当.

摘要:
The existing form of ionizable organic contaminants (IOCs) could affect their adsorption characteristics to soil and biochar. In this study, 2 IOCs, namely, sulfadiazine and imazalil, were selected to study their adsorption by rice straw-derived biochar-amended soils, as well as the effect of pH and gallic acid on their adsorption. The results showed that the soil adsorption isotherms of the two ionizable organic contaminants could be fitted well by a linear equation and the Freundlich equation, and r(2) was more than 0.80. The adsorption coefficient (Kd) in the three kinds of soil ranged from 0.262 to 4.07Lkg(-1) for sulfadiazine and from 3.11 to 96.5Lkg(-1) for imazalil. After the addition of biochar, the adsorption of sulfadiazine and imazalil in the soil increased. The adsorption of sulfadiazine by biochar gradually decreased with the increase in pH; the adsorption of imazalil increased when the pH increased from 2 to 5 and then gradually decreased with increasing pH. Gallic acid enhanced the adsorption of the two IOCs to pure soil and biochar-amended soil.

The existing form of an ionizable organic compound can simultaneously affect its soil adsorption and plant bioactivity. In this experiment, the adsorption and bioactivity of two weak acid herbicides (WAHs), imazethapyr and 2,4-D, were studied to explore the predominant mechanism by which the soil pH and the addition of biochar can influence the phytotoxicity of WAHs in soil. Then, the WAH concentration extracted by hollow fiber-based liquid-phase microextraction (CHF-LPME), the in situ pore water concentration (CIPW) and the added concentration (CAC) were employed to estimate the phytotoxicity. The results showed that with increased pH from 5.5 to 8.5, the phytotoxicity of the WAHs to rice increased about 1-fold in the soil, but decreased in aqueous solutions, the IC50 values for imazethapyr and 2,4-D at pH 5.0 were 3- and 2-fold higher than that at pH 8.0. In addition, the soil adsorption decreased, indicating that the adsorption process was the dominant factor for the variation of the phytotoxicity of the WAHs in the tested soil instead of the decreasing bioactivity. The concentration that inhibits plant growth by 50% (IC50) calculated by the CAC in different pH and biochar soils ranged from 0.619 to 3.826 mg/kg for imazethapyr and 1.871–72.83 mg/kg for 2,4-D. The coefficient of variation (CV) of the IC50 values reached 65.61% for imazethapyr and 130.0% for 2,4-D. However, when IC50 was calculated by CIPW and CHF-LPME, the CVs of the IC50 values decreased to 23.51% and 36.23% for imazethapyr and 40.21% and 50.93% for 2,4-D, respectively. These results suggested that CIPW and CHF-LPME may be more appropriate than CAC for estimating the phytotoxicity of WAHs. © 2018 Elsevier Inc.

<jats:title>Abstract</jats:title><jats:p>The toxicity of ionizable organic compounds to organisms depends on the pH, which therefore affects risk assessments of these compounds. However, there is not a direct chemical method to predict the toxicity of ionizable organic compounds. To determine whether hollow-fiber liquid-phase microextraction (HF-LPME) is applicable for this purpose, a three-phase HF-LPME was used to measure sulfadiazine and estimate its toxicity to <jats:italic>Daphnia magna</jats:italic> in solutions of different pH. The result indicated that the sulfadiazine concentrations measured by HF-LPME decreased with increasing pH, which is consistent with the decreased toxicity. The concentration immobilize 50% of the daphnids (EC50) in 48 h calculated from nominal concentrations increased from 11.93 to 273.5 mg L<jats:sup>−1</jats:sup> as the pH increased from 6.0 to 8.5, and the coefficient of variation (CV) of the EC50 values reached 104.6%. When calculated from the concentrations measured by HF-LPME (pH 12 acceptor phase), the EC50 ranged from 223.4 to 394.6 mg L<jats:sup>−1</jats:sup>, and the CV decreased to 27.60%, suggesting that the concentrations measured by HF-LPME can be used to estimate the toxicity of sulfadiazine irrespective of the solution pH.</jats:p>

The genome sequence of pepper vein yellows virus (PeVYV) (PeVYV-HN, accession number KP326573), isolated from pepper plants (Capsicum annuum L.) grown at the Hunan Vegetables Institute (Changsha, Hunan, China), was determined by deep sequencing of small RNAs. The PeVYV-HN genome consists of 6244 nucleotides, contains six open reading frames (ORFs), and is similar to that of an isolate (AB594828) from Japan. Its genomic organization is similar to that of members of the genus Polerovirus. Sequence analysis revealed that PeVYV-HN shared 92 % sequence identity with the Japanese PeVYV genome at both the nucleotide and amino acid levels. Evolutionary analysis based on the coat protein (CP), movement protein (MP), and RNA-dependent RNA polymerase (RdRP) showed that PeVYV could be divided into two major lineages corresponding to their geographical origins. The Asian isolates have a higher population expansion frequency than the African isolates. Negative selection and genetic drift (founder effect) were found to be the potential drivers of the molecular evolution of PeVYV. Moreover, recombination was not the distinct cause of PeVYV evolution. This is the first report of a complete genomic sequence of PeVYV in China.
