Transcription Factor WRKY70 Displays Important but No Indispensable Roles in Jasmonate and Salicylic Acid Signaling

摘要:
The transcription factor WRKY70 was previously reported to be a common component in salicylic acid (SA) and jasmonate (JA) mediated signal pathways in Arabidopsis. Here, we present that the inactivation of the WRKY70 gene in wrky70-1 mutant does not alter the responses of both JA and SA, and that wrky70 mutation is unable to restore the coi1 mutant in JA responses. However, overexpression of WRKY70 reduces JA responses such as expression of JA-induced genes and JA-inhibitory root growth, and activates expression of SA-inducible PR1. These data indicate that the WRKY70 is important but not indispensable for JA and SA signaling, and that other regulators may display the redundant role with WRKY70 in modulation of JA and SA responses in Arabidopsis. Furthermore, we showed that JA inhibits expression of WRKY70 and PR1 by both COI1-dependent and COI1-independent pathways.

摘要:
采用红外光谱和扫描电镜等手段研究了浓硫酸处理前、后碳纸的表面结构和形貌的变化.并将这类碳纸用作全钒液流氧化还原电池电极材料,对其电化学性能进行了详细研究.结果表踢通过酸处理,碳纸表面有-COOH官能团生成,其电化学活性增强.酸处理后的碳纸电极组装成的电池在电流密度20 mA·cm~(-2)时有良好充放电性能,平均电流和电压效率达到95%和82%.

Ubiquitin ligases play a central role in determining the specificity of the ubiquitination system by selecting a myriad of appropriate candidate proteins for modification. The U-box is a recently identified, ubiquitin ligase activity-related protein domain that shows greater presence in plants than in other organisms. In this study, we identified 77 putative U-box proteins from the rice genome using a battery of whole genome analysis algorithms. Most of the U-box protein genes are expressed, as supported by the identification of their corresponding expressed sequence tags (ESTs), full-length cDNAs, or massively parallel signature sequencing (MPSS) tags. Using the same algorithms, we identified 61 U-box proteins from the Arabidopsis genome. The rice and Arabidopsis U-box proteins were classified into nine major classes based on their domain compositions. Comparison between rice and Arabidopsis U-box proteins indicates that the majority of rice and Arabidopsis U-box proteins have the same domain organizations. The inferred phylogeny established the homology between rice and Arabidopsis U-box/ARM proteins. Cell death assay using the rice protoplast system suggests that one rice U-box gene, OsPUB51, might act as a negative regulator of cell death signaling. In addition, the selected U-box proteins were found to be functional E3 ubiquitin ligases. The identification and analysis of rice U-box proteins hereby at the genomic level will help functionally characterize this class of E3 ubiquitin ligase in the future.

摘要:
The phytohormone abscisic acid (ABA) is the major player in mediating the adaptation of plants to stress. Previously developed phytohormonal biosensors usually employed indirect detection of the products of conjugated oxidase reactions. A label-free electrochemical impedance immunosensor for ABA detection was developed using an anti-ABA antibody adsorbed directly on a porous nanogold film. The film was produced electrochemically on a glassy carbon electrode in 0.008 mol/L hydrogen tetrachloroaurate solution containing 0.004 mol/L lead acetate with an applied potential of -0.5 V (versus Ag/AgCl) for 50 s. The anti-ABA antibody was immobilized onto the porous nanogold through electrostatic adsorption and covalent conjugation. Electrochemical impedance spectroscopy was used to characterize the successful construction of the porous nanogold film and the stepwise modification of the glassy carbon electrode. The concentration increase of the antigen brought about a decrease of the interfacial electron transfer, which also meant an increase of the impedance signal. The experimental parameters pH, antibody incubation time, and antibody concentration were optimized. The results showed significant linearity R=0.9942, with the content of ABA in the range 0.5-5,000 ng/mL with a detection limit at about 0.1 ng/mL. © 2008 Springer-Verlag.

Dietary supplementation of glutamine prevents intestinal dysfunction and atrophy in weanling piglets, but the underlying mechanism(s) are largely unknown. This study was conducted to test the hypothesis that weaning or glutamine may modulate expression of genes that are crucial for intestinal metabolism and function. In Expt. 1, we obtained small intestine from 28-d-old pigs weaned at 21 d of age and from age-matched suckling piglets. In Expt. 2, piglets were weaned at 21 d of age and then had free access to diets supplemented with 1% L-glutamine (wt:wt) or isonitrogenous L-alanine (control). At d 28, we collected small intestine for biochemical and morphological measurements and microarray analysis of gene expression using the Operon Porcine Genome Oligo set. Early weaning resulted in increased (52-346%) expression of genes related to oxidative stress and immune activation but decreased (35-77%) expression of genes related to macronutrient metabolism and cell proliferation in the gut. Dietary glutamine supplementation increased intestinal expression (120-124%) of genes that are necessary for cell growth and removal of oxidants, while reducing (34-75%) expression of genes that promote oxidative stress and immune activation. Functionally, the glutamine treatment enhanced intestinal oxidative-defense capacity (indicated by a 29% increase in glutathione concentration), prevented jejunal atrophy, and promoted small intestine growth (112%) and body weight gain (119%) in weaned piglets. These findings reveal coordinate alterations of gene expression in response to weaning and aid in providing molecular mechanisms for the beneficial effect of dietary glutamine supplementation to improve nutrition status in young mammals. © 2008 American Society for Nutrition.

Based on the atomic local structure, a nanocrystalline material is separated into two parts: inner grains and grain boundaries. Molecular dynamics simulations have been performed to study the difference in their vibrational properties in nanocrystalline nickels. The results obtained reveal that a similar phonon frequency softening as in the perfect lattice mainly focuses on grains in nanocrystalline materials, and for the grain boundary part there is no obvious change in the vibrational density of states in the temperature range 300-900 K, especially in the lower frequency range. Comparing with conventional crystals, the higher specific heat and vibrational entropy, and lower vibrational free energy with decreasing mean grain size, result from the high proportion of grain boundaries in nanocrystals. In addition, the composite model provides a good description of the contributions of grain and grain boundary phases to thermodynamic quantities. Supposing the nanocrystalline material can be treated as a composite composed of grain and grain boundary network, the vibrational thermodynamic quantities of the system can be well determined from the proportion of grain boundaries and the corresponding thermodynamic quantities of grain and grain boundary parts. 摘要:
A novel method of estimating conformation of protein secondary structure within Fusarium cells under the influence of growth cultures and bodies was studies by Fourier-Transform Infrared (FT-IR) spectroscopy combined with a deconvolution procedure of amide I band. After infrared data collection and curve-fitting process, the percentages of secondary structure components (a-helix, β-sheet, turn, random coil and β-antiparallel) were obtained. It showed structural diversity among different Fusarium species and variations caused by growth cultures and bodies. The method provides a rapidly quantitative technique to study cell structure of fungi.

A central composite design was used to study the effect of glycerol, rate of stirring, air aeration and pH on the synthesis of 1,3-propanediol (1,3-PD) by Klebsiella pneumoniae AC 15. Among the four variables, glycerol and rate of stirring significantly affected 1,3-PD productivity, whereas air aeration and pH were not effective. A quadratic polynomial equation was obtained for 1,3-PD productivity by multiple regression analysis using response surface methodology. The validation experimental confirmed with the predicted model. The optimum combinations for 1,3-PD productivity was glycerol, rate of stirring, air aeration, and pH of 50 g/l, 318 rpm, 0.6 vvm, 6.48, respectively. The subsequent fed batch experiments produced 1,3-PD of 70 g/l at a fermentation of 30 h. © 2007 Elsevier Ltd. All rights reserved.

摘要:
SnO<sub>2</sub> hollow nanospheres have been synthesized by heat treatment of a mixture composed of SnCl<sub>4</sub> and carbon microspheres. The SnO<sub>2</sub> nanospheres are 100 nm in size and composed of large amount of SnO<sub>2</sub> nanocrystals with diameters of ~ 6 nm, which is close to two times of the Debye length of SnO<sub>2</sub>. It was found that the sensitivity to 1000 ppm ethanol obtained with SnO<sub>2</sub> hollow nanosphere-based sensors is as high as 75. The response and recovery time are only 4 and 10 s, respectively. Such excellent sensing properties can be ascribed to the small-size nanocrystals and porous structure of SnO<sub>2</sub> hollow nanospheres, which lead to large diffusion constant. ©2008 Elsevier B.V. 摘要:
A pot experiment was conducted to study the effect of an anionic surfactant linear alkylbenzene sulfonate (LAS) application on cadmium (Cd) fractions in soils and Cd distribution in different tissues of soybean (Glycine max)plants as well as soil pH. Soil samples were treated with three levels of Cd (0, 5, and 10 mg kg -1 ) and five levels of LAS (0, 5, 15, 50, and 100 mg L -1 ). Results indicated that compared to the control soils (no Cd and no LAS treatment), soil pH increased and available Cd decreased in the soil treated with external Cd and watered with LAS solutions. Meanwhile, soil exchangeable Cd and Cd bound to carbonates decreased; Cd bound to amorphous iron and manganese oxides and Cd bound to organic matter increased. In addition, LAS application could reduce enrichment of Cd in soybean plants, resulting in decreased Cd in the soybean plants. Thus, suitable LAS application could decrease bioavailability and mobility of soil Cd. © 2008 Soil Science Society of China.

By using combinatorics, we give a new proof for the recurrence relations of the characteristic polynomial coefficients, and we further obtain an explicit expression for the generic term of the coefficient sequence, which yields the trace formulae of the Cayley-Hamilton's theorem with all coefficients explicitly given. This implies a byproduct, a complete expression for the determinant of any finite-dimensional matrix in terms of the traces of its successive powers. And we discuss some of their applications to chiral perturbation theory and general relativity. © 2008 Chinese Physical Society.

摘要:
The large yellow croaker (Pseudosciaena crocea) is an economically important marine fish in China. Inheritance of 22 heterozygous microsatellite loci was examined in normal crossed diploid families and meio-gynogenetic families in P. crocea. Two gynogenetic families were produced via inhibition of the second polar body in eggs fertilized with UV-irradiated sperm. The ratio of gynogenesis was proven to be 100% and 96.9% in the two families, respectively. Of the 22 examined loci, 4 showed a segregation distortion in both control and gynogenetic families. Microsatellite-centromere (M-C) map distances were examined using 18 loci with normal Mendelian segregation. Estimated recombination rates ranged between 0 and 1.0 under the assumption of complete interference. High recombinant frequencies between heterozygous markers and the centromere were found in large yellow croaker, as in other teleosts. The average recombination frequency was 0.586. Ten loci showed high M-C recombination with frequency greater than 0.67. M-C distances provide useful information for gene mapping in large yellow croaker. ©2007 Springer Science+Business Media, LLC.

摘要:
The contributions to S, T and U from heavy scalars in any irreducible representation of the electroweak gauge group SU(2)L × U(1)Y are obtained. We find that in the case of a heavy scalar doublet, there is a slight difference between the S parameter we have obtained and that in previous works. © 2008 World Scientific Publishing Company.

摘要:
By constructing a suitable Lyapunov function and using some analysis techniques, rather than employing the continuation theorem of coincidence degree theory as in other literature, a sufficient criterion is obtained to ensure the existence and global exponential stability of periodic solution for the bidirectional associative memory neural network with periodic coefficients and continuously distributed delays. The obtained result is less restrictive to the BAM neural network than the previously known criteria. And it can be applied to the BAM neural network in which signal transfer functions are neither bounded nor differentiable. In addition, an example and its numerical simulation are given to illustrate the effectiveness of the obtained result. © 2007 Elsevier Ltd. All rights reserved.

摘要:
为深入认识含氟农药生物活性与其结构之间的关系,建立了理想的QSAR模型,从化合物油水分配系数等7个分子结构描述符出发,基于支持向量回归(SVR)和MSE最小原则,经自动寻找最优核函数和非线性筛选描述符,构建了多个K-最近邻(KNN)预测子模型.再经非线性筛选获得保留子模型,以保留子模型实施组合预测(Multi—KNN—SVR).33种含氟化合物对5种不同病害生物活性的留一法组合预测结果表明,采用非线性筛选描述符和KNN子模型能有效地提高预测精度,基于多个KNN子模型的非线性组合能进一步提高预测性能.Multi—KNN—SVR组合预测在QSAR以及其它相关预测研究中具有广泛应用前景.

摘要:
The aim of this study was to investigate the effect of capsaicin on hypoxia-reoxygenation (H/R)-induced apoptosis in primary rat hippocampal neurons. Three hours of hypoxia (1% O2) and subsequent reoxygenation for 24 h significantly increased the apoptotic death of hippocampal neurons, as evidenced by increases in both TUNEL-positive cell number and caspase-3 activity. Pretreatment with capsaicin (3-30 micromol/L) or the caspase-3-specific inhibitor acetyl-DEVD-CHO (100 micromol/L) markedly attenuated H/R-induced apoptosis in hippocampal neurons. Capsaicin also markedly induced the phosphorylation of Akt. The phosphoinositide 3-kinase (PI3K) inhibitor LY294002 (10 micromol/L) prevented any capsaicin-induced survival effect in hippocampal neurons. Intracellular levels of reactive oxygen species (ROS), which were greatly increased after H/R, were significantly inhibited by capsaicin, pyrrolidine dithiocarbamate (PDTC) (50 micromol/L), and LY294002. Taken together, these data suggest that capsaicin protects against H/R-induced apoptosis of hippocampal neurons via the PI3K/Akt-mediated signaling pathway, which is related to the inhibition of oxidative stress and caspase-3 activation.

Effect of dietary supply of cysteamine on sites of digestion, small intestinal amino acid disappearance in growing goats. J. Appl. Anim. Res., 34: 71–76. Four Liuyang Black growing goats were used to examine the effect of dietary supply of cysteamine (CS) on feed intake, site and extent of digestion and apparent intestinal amino acid (AA) digestibility in a 4x4 Latin square design. The goats were weighed initially at 11.8 kg (SD 1.6), fitted with the permanent ruminal, duodenal and ileal fistulae and fed ad libitum in individual pens. Four treatments consisted of a basal diet only (control) and dietary supply of CS at levels of 20, 40 and 60 mg/kg dry matter, respectively, to the basal diet. The experimental period lasted for 14 d for each treatment. Dietary supply of CS had no effect (P>0.05) on intake of nutrients. Ruminal digestion of ADF and NDF decreased (P=0.01) on higher level of CS supplementation. The small intestinal digestibilities of alanine, methionine, threonine and valine decreased (P<0.05) with the inclusion of CS in the diets. The results imply that CS supplementation did not improve the digestibility and absorption of nutrients in the gastro-intestinal tract of goats.

OBJECTIVE:To study the kidney tonic components of Rhizoma Drynariae.METHODS:The herbal material was extracted with 50% ethanol.Silica gel and sephadex LH-20 coluuln chromatography isolated the chemical components using the Opossurn Kidney cell for the living model.The structures were identified on the basis of chemical and spectroscopic data.RESULTS.Six monocases were isolated from the EtOH extract of in Rhizomes of Drynaria fortunei J.Sm..Four structures were identified as naringin(I),neocriocitrin(Ⅱ),kaempferol(Ⅲ),and naringenin-7-O-β-D-glucoside(Ⅳ),respectively,on the basis of its spectral and physicochemical evidence.The kaempferol call promote the proliferation of the Opossum Kidney cell tested by Promega’B CellTiter96 assay in vitr0.Others are in identifying.CONCLUSION:Compound In and IV are firstly isolated from this plant,and the later may hefurther developed into a new effective drug for treatingm rural failure.

1,3-Propanediol (1,3-PD) production is favored by high amount of glycerol which also contributes to the lethal 3-hydroxypropionaldehyde (3-HPA) accumulation. The influences of three culture conditions including initial glycerol concentration, stirring rate, and oxidoreduction potential (ORP) regulation on 3-HPA accumulation and 1,3-PD production were investigated. 3-HPA detoxification was available at initial glycerol concentration, aeration rate and stirring rate of up to 40 g l-1, 0.5 vvm air flow and 250 rpm, respectively. Alternatively, the stirring rate of 300 rpm was sufficient to fulfill the fermentation with initial 50 g glycerol l-1. In addition, 3-HPA accumulation could be avoided at the initial 50 g glycerol l-1 under the regulated reducing conditions (-200 and -400 mV). However, ORP regulation was not convenient and feasible for the industrial production in that more carbon was distributed to weak organic acids. Consequently, two-stage fed-batch strategy was presented to achieve 3-HPA detoxification and maintain the expected high productivity of 1,3-PD, in which the batch culture stage was conducted at initial glycerol concentration and stirring rate of 40 g l-1 and 250 rpm, respectively, and the feeding culture stage was performed at stirring rate of 300 rpm. 74.07 g 1,3-PD l-1 was produced in 24 h with the yield and productivity of 0.62 mol mol-1 and 3.08 g l-1 h-1. © 2007 Elsevier B.V. The effects of aqueous extracts of Kuding tea (Ilex latifolia Thunb.) and the large-leaf form of Camellia sinensis var. sinensis), chrysanthemum (Dendranthema morifolium Ramat), honeysuckle flower (Lonicera japonica Thunb.), and purple sweet potato (Ipomoea batatas) stem on glucose absorption were investigated using Caco-2 cells. Glucose absorption by Caco-2 cells was significantly inhibited by aqueous extract of Kuding tea, chrysanthemum and purple sweet potato stem under both Na+-dependent conditions and Na+-free conditions indicating effects on SGLT1 and GLUT transporters. Analysis of the (poly)phenols in these aqueous extracts suggested that dicaffeoylquinic acids and flavanols may be particularly important in producing these effects. Kuding tea extract was the most effective, suggesting that this merits evaluation in a clinical study. © 2007 Elsevier Ltd. All rights reserved.
