Epidermal growth factor containing culture supernatant enhances intestine development of early-weaned pigs in vivo: Potential mechanisms involved

摘要:
We have previously generated epidermal factor expressing Lactococcus lactis (EGF-LL) using a bioengineering approach, and shown that EGF-LL fermentation supernatant enhanced newly weaned pigs growth. The objective of the current study was to further understand the mechanisms behind this improved performance. Sixty-four piglets were weaned at 3 weeks of age and then fed ad libitum according to a 2-phase feeding program. Four pens with 8 pigs per pen were assigned to each of two treatments for 3 weeks: (1) EGF containing supernatant from EGF-LL culture (SuperEGF) or (2) blank M17GE media (Control). Consistent with previous findings, SuperEGF pigs had an increased average daily gain during week 3 post-weaning (433.4. ±. 10.86 vs 388.7. ±. 7.76. g;P<. 0.05) and overall gain:feed ratio (0.757. ±. 0.03 vs 0.677. ±. 0.01. kg/kg, P<. 0.05). Moreover, jejunal structure development was enhanced, and inflammation index was minimized in SuperEGF pigs as indicated by increased villi height ( P<. 0.05), decreased lamina propria width ( P<. 0.05), and higher expression of anti-inflammatory cytokine, IL-13 ( P<. 0.05). Further, goblet cell numbers and Muc2 levels were increased in SuperEGF pigs. Interestingly, the weaning-induced decrease of glucose cotransporter sodium-glucose linked transporter 1 (SGLT1) and glucagon-like peptide-2 (GLP2) levels was reversed by SuperEGF supplementation. Our findings add to our understanding of the mechanisms behind enhancing piglet performance by EGF containing fermentation product. ©2015 Elsevier B.V.

摘要:
Mastitis is the inflammation of the mammary gland. Recent research has shown that Angiopoietin-like protein 2 (ANGPTL2) is a key inflammatory mediator. In the present study, we tested whether there is a correlation between increased ANGPTL2 expression and inflammation in response to Staphylococcus aureus in murine mastitis and the mechanisms involved. Thirty mice were divided into two groups: blank control group, challenged group. The entire infused mammary glands were removed to observe the changes of histopathology, myeloperoxidase (MPO) activity, production of tumour necrosis factor-alpha (TNF-alpha) and interleukin (IL)-6, and genes expression of ANGPTL2, TNF-alpha and IL-6. In challenged group, the structure of mammary glands was damaged and the large areas of cell fragments were observed. The MPO activity, IL-6 and TNF-alpha concentrations, ANGPTL2, IL-6, and TNF-alpha mRNA levels were significantly elevated in challenged group compared with blank control group. The present findings indicate ANGPTL2 may mediate the inflammation in murine mastitis through the activation of IL-6 and TNF-alpha.

摘要:
The Chinese perch (Siniperca chuatsi) is one of the most commercially important carnivorous fish species in aquaculture with its large-scale culture in China. Increasing evidence suggests that microRNAs (miRNAs) play an important role in muscle cell proliferation and differentiation. However, the knowledge of the identity of myogenic miRNAs and the effect of nutrient status on miRNA expression in teleost remains limited. In the present study, among the 21 miRNAs identified with high abundance in the fast muscle of adult Chinese perch, 19 miRNAs were differentially expressed in the adults and juveniles. The postprandial changes in the transcript abundance were determined for the 21 miRNAs following a single satiating meal in the juveniles after fasting for 1 week. The results showed that the seven miRNAs (miR-10c, miR-107a, miR-133a-3p, miR-140-3p, miR-181a-5p, miR-206, and miR-214) were sharply upregulated or downregulated within 1 h after refeeding. These miRNAs may be the promising candidate miRNAs involved in a fast-response signaling system that regulates fish skeletal muscle growth. Target prediction and expressional analysis suggested that four miRNAs (miR-10c, miR-107a, miR-140-3p, and miR-181a-5p) might play a role in regulating the translation of target gene transcripts such as myostatin following acute anabolic stimuli.

PCV2 is highly pathogenic, however, its effect on the serum amino acids profile is unknown. This study was conducted to explore the profile of amino acids in the serum in porcine circovirus type 2 (PCV2) infected mice. The serum levels of amino acids were detected with isotope dilution liquid chromatography-mass spectrometry methods at 3, 7, 10 and 14 days post infection (DPI). Meanwhile, the expression of seven amino acids transporters (SLC6A14, SLC6A20, SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A9) in the jejunum was analyzed by reverse transcription polymerase chain reaction (RT-PCR) at 3 and 7 DPI. Serum PCV2 load was also analyzed by quantitative PCR at 3, 7, 10 and 14 DPI. Serum levels of most amino acids, such as Pro, Orn, and Met, significantly (P < 0.05) increased at 3 DPI. However, most amino acids, including Asp, Sar, Arg, Hyl, Pro, Lys, Val, Ile and Leu, significantly (P < 0.05) decreased at 7 DPI. There was no significant difference for most amino acids at 10 and 14 DPI. PCV2 infection significantly (P < 0.05) decreased expression of SLC7A5 and SLC7A6 at 7 DPI. In conclusion, PCV2 infection affects the profile of amino acids in the serum and the expression of amino acids transporters in the intestine.

摘要:
为了研究猪粪直接燃烧供能的可行性,该文测定了干清粪方式收集的玉米-豆粕日粮型猪粪的含水率、纤维素、半纤维素和木质素含量以及发热量;采用热重法测定了猪粪的热重曲线(thermogravimetric curves,TG curves)、微分热重曲线(diffenrential thermogravimetric curves,DTG curves),分析了猪粪的可燃性指数、燃烧稳定性指数、综合燃烧特性指数。结果表明:新鲜猪粪含水率在74.28%~76.75%;高位发热量为16.72~17.65 MJ/kg,低位发热量为14.24~15.34 MJ/kg;热重分析表明猪粪可燃性指数为(1.552~1.652)×215;10-5 mg/(min×183;℃2),燃烧稳定性指数为(1.393~1.466)×215;10-6 mg/(min×183;℃2),综合燃烧特性指数为(6.480~7.305)×215;10-9 mg2/(min2×183;℃3)。猪粪球能在锅炉内良好燃烧,呈现明显的挥发分燃烧状态。研究的结果为猪粪直接燃烧供能提供了依据和参考。

摘要:
Density gradient ultracentrifuge separation was employed to study the phase transition of Yb3+/Er3+ co-doped NaYF4 from α-phase to β-phase. It was found that the cubic α-phased NaYF4 was rich in Y, exhibited predominantly red luminescence, whilst the newly formed hexagonal β-phased NaYF4 had relatively higher Yb and Er atomic ratios, exhibited green luminescence most strongly. © 2013 The Royal Society of Chemistry.

The Musmusculus myosin-18A gene is expressed as two alternatively spliced isoforms, αand β, with reported roles in Golgi localization, in maintenance of cytoskeleton, and as receptors for immunological surfactant proteins. Both myosin-18A isoforms feature a myosin motor domain, a single predicted IQ motif, and a long coiled-coil reminiscent of myosin-2. The myosin-18Aαisoform, additionally, has an N-terminal PDZ domain. Recombinant heavy meromyosin- and subfragment-1 (S1)-like constructs for both myosin-18Aαand -18βspecies were purified from the baculovirus/Sf9 cell expression system. These constructs bound both essential and regulatory light chains, indicating an additional noncanonical light chain binding site in the neck. Myosin-18Aα-S1 and -18Aβ-S1 molecules bound actin weakly with Kd values of 4.9 and 54 μM, respectively. The actin binding data could be modeled by assuming an equilibrium between two myosin conformations, a competent and an incompetent form to bind actin. Actin binding was unchanged by presence of nucleotide. Both myosin-18A isoforms bound N-methylanthraniloyl- nucleotides, but the rate of ATP hydrolysis was very slow (<0.002 s <sup>-1</sup>) and not significantly enhanced by actin. Phosphorylation of the regulatory light chain had no effect on ATP hydrolysis, and neither did the addition of tropomyosin or of GOLPH3, a myosin-18A binding partner. Electron microscopy of myosin-18A-S1 showed that the lever is strongly angled with respect to the long axis of the motor domain, suggesting a pre-power stroke conformation regardless of the presence of ATP. These data lead us to conclude that myosin-18A does not operate as a traditional molecular motor in cells. ©2013 by The American Society for Biochemistry and Molecular Biology, Inc.

Metabolism studies play an important role at various stages of drug discovery and development. Liquid chromatography combined with mass spectrometry (LC/MS) has become a most powerful and widely used analytical tool for identifying drug metabolites. The suitability of different types of mass spectrometers for metabolite profiling differs widely, and therefore, the data quality and reliability of the results also depend on which instrumentation is used. As one of the latest LC/MS instrumentation designs, hybrid ion trap/time-of-flight MS coupled with LC (LC-IT-TOF-MS) has successfully integrated ease of operation, compatibility with LC flow rates and data-dependent MS<sup>n</sup> with high mass accuracy and mass resolving power. The MS<sup>n</sup> and accurate mass capabilities are routinely utilized to rapidly confirm the identification of expected metabolites or to elucidate the structures of uncommon or unexpected metabolites. These features make the LC-IT-TOF-MS a very powerful analytical tool for metabolite identification. This paper begins with a brief introduction to some basic principles and main properties of a hybrid IT-TOF instrument. Then, a general workflow for metabolite profiling using LC-IT-TOF-MS, starting from sample collection and preparation to final identification of the metabolite structures, is discussed in detail. The data extraction and mining techniques to find and confirm metabolites are discussed and illustrated with some examples. This paper is directed to readers with no prior experience with LC-IT-TOF-MS and will provide a broad understanding of the development and utility of this instrument for drug metabolism studies. ©2012 John Wiley &Sons, Ltd.

An increasing number of people are processing transactions online and the numbers are likely to increase rapidly in the near future. It is important to analyze the relationship between customer values of social network service and Customer Loyalty. The purpose of this paper is to answer the following questions. First, what are key components of customer value in social network service? Second, what influences the relationship between customer value and Customer Loyalty? Data collected from 160 respondents who participate in social network service were used to test a research model. Several managerial implications were derived from the analysis and further studies were suggested. ©2012 Springer Science+Business Media Dordrecht.

摘要:
The objective of this study was to investigate the protective effect of soluble fiber extracted from Undaria pinnatifida (UP) on vascular endothelium and the mechanisms involved. The experiments were performed in aorta and plasma from C57BL/6J control and apolipoprotein E deficient (ApoE<sup>-/-</sup>) mice treated or not with UP soluble fiber (5 or 10%) for 8 weeks. UP soluble fiber treatment improved endothelium-dependent vasorelaxation, increased the maximal relaxation value, and decreased the half-maximum effective concentration concomitantly with a decrease in asymmetric dimethylarginine (ADMA) and malondialdehyde plasma concentrations, and increase in the expression as well as activity of dimethylarginine dimethylaminohydrolase II in ApoE<sup>-/-</sup> mice. Treatment with UP soluble fiber also significantly decreased reactive oxygen species production in mice aorta. The present results suggest that the protective effect of UP soluble fiber on vascular endothelium may be associated with a decrease in ADMA level in mice. PRACTICAL APPLICATIONS: Soluble fiber have complex beneficial effects on cardiovascular system such as lowering serum cholesterol, inhibiting lipid peroxidation, reducing blood pressure, increasing insulin sensitivity, modulating plasma glucose, improving homocysteine levels, protecting against intima-media thickness progression of the common carotid arteries, and decreasing the serum levels of markers of inflammation. The present work reports that the protective effect of Undaria pinnatifida soluble fiber on vascular endothelium may be associated with a decrease in ADMA level. It complements previous work. This is important because the mechanism responsible for the protective effect of fiber on endothelial function is defined. Furthermore, it provides information for the consumer about the potential effects of fiber and Undaria pinnatifida. ©2011 Wiley Periodicals, Inc.

A new strategy using hybrid ion trap/time-of-flight mass spectrometry coupled with high-performance liquid chromatography and post-acquisition data mining techniques was developed and applied to the detection and characterization of degradation products of danofloxacin. The degradation products formed under different forced conditions were separated using an ODS-C18 column with gradient elution. Accurate full-scan MS data were acquired in the first run and processed with the combination of extracted ion chromatograms and LC-UV chromatograms. These processes were able to find accurate molecular masses of possible degradation products. Then, the accurate MS/MS data acquired through data-dependent analysis mode in another run facilitated the structural elucidations of degradation products. As a result, a total of 11 degradation products of danofloxacin were detected and characterized using the developed method. Overall, this analytical strategy enables the acquisition of accurate-mass LC/MS data, search of a variety of degradation products through the post-acquisition processes, and effective structural characterization based on elemental compositions of degradation product molecules and their product ions. The ability to measure degradation products via tandem mass spectrometry coupled with accurate mass measurement, all in only two experimental runs, is one of the most attractive features of this methodology. The results demonstrate that use of the LC/MS-IT-TOF approach appears to be rapid, efficient and reliable in structural characterization of drug degradation products. A simple technique, REMI (restriction enzyme-mediated integration), was used to construct transformants of Trichoderma atroviride with improved capability of degrading organophosphate pesticide dichlorvos. Linearized DNA of plasmid pV2 bearing the hygromycin B phosphotransferase (hph) gene was inserted into chromosomes of wild strain T23 and transformation was confirmed by PCR and Southern blot analysis, respectively. Of 247 transformants, 76% showed improved dichlorvos degradation ability as compared to the parent strain T23 based on the least significant difference (LSD) test at p = 0.01. Among them, 8 transformants exhibited 30% higher in degradation rate than the parent isolate. The highest dichlorvos degradation rate of the transformants was up to 96%. This study provided an effective approach for improving organophosphate pesticide-degrading capability of T. atroviride. © 2008 Elsevier Ltd. All rights reserved.

蜱类系统分类学研究已经进入分子阶段,与传统的形态学、生物学鉴定相比,无疑更加客观准确,建立系统进化树更能全面反映其亲缘关系,极大地提高了鉴定效率与可信度。论文总结了近年来应用分子生物学技术鉴定硬蜱的主要方法,包括核酸序列分析、随机扩增DNA多态性分析、限制性片段长度多态性分析。随着分子生物学和现代统计学的广泛应用,蜱类的鉴定将更加依赖现代分类学方法。
