摘要:
Testicular development and spermatogenesis are strictly regulated with the complex interactions between several cell types in testis. However, the limited availability of genomic and molecular information limits our understanding of this complex physiological process. In this study, we characterized the transcriptome between immature (30-day-old) and mature (180-day-old) pig testes using RNA-seq technology. 24,469 known coding gene transcripts corresponding to 20,566 genes were mapped in these two developmental stages, 3,328 genes were differentially expressed, and numerous novel transcripts and alternative splicing events were also identified. Ten differentially expressed genes were validated by measuring the relative expression using real-time quantitative polymerase chain reaction (PCR). 125 Gene Ontology (GO) terms were significantly enriched, and most of them involved in GO terms related to male reproduction, testicular development, and spermatogenesis. In addition, this study also represented the prediction of 10,000 circRNAs, as well as the validation of six pig circRNAs using Find_cric algorithm. Our study substantially expanded our knowledge about the transcriptomic profile of immature and mature pig testes, and provided a useful resource to study the mechanisms of pig testis development and spermatogenesis at the molecular level. © 2017, The Genetics Society of Korea and Springer Science+Business Media B.V.

摘要:
为探讨雪峰乌骨鸡体重、体尺性状及产肉性能的相关关系.选取同批孵化、体重基本一致的1日龄健康雪峰乌骨鸡300羽,126日龄时随机选取60羽(公母各半),测定公母鸡的体重、体尺性状及产肉性能,并对各指标进行相关性分析,同时,比较126日龄雪峰乌骨鸡公母鸡体重与屠宰性能指标.结果表明:公鸡的胸深与体重呈极显著正相关(P<0.01),公鸡体重与各屠宰性能指标之间均呈极显著正相关(P<0.01);公鸡的体重与腿肌、胸肌均呈极显著正相关(P<0.01),而母鸡的体重仅与腿肌呈极显著正相关(P<0.01),和胸肌相关性不显著(P>0.05);雪峰乌骨鸡公鸡体重显著高于母鸡(P<0.05),母鸡的腹脂率极显著高于公鸡(P<0.01),而公鸡的腿肌率、瘦肉率显著高于母鸡(P<0.05).结果显示,可通过胸深这一体尺指标预测雪峰乌骨鸡公鸡的屠宰性能;通过体重来选择种公母鸡的效果会不同;雪峰乌骨鸡公鸡的屠宰性能优于母鸡.

沙子岭猪GSG1基因克隆、生物信息学分析及组织表达

摘要:
采用RACE技术克隆猪GSG1(Germ Cell-specific gene l)基因cDNA1全长并进行生物信息学分析,利用实时荧光定量PCR对GSG1基因在沙子岭猪睾丸组织8个不同发育阶段(E90、D1、D30、D60、D90、D120、D150和D180)和30日龄时9个不同组织(睾丸、肌肉、小肠、肺、心脏、肾脏、肝脏、脾脏和脂肪)的表达量.结果表明:GSG1基因cDNA全长为1 257 bp,含1个长993 bp的开放阅读框,编码330个氨基酸.实时荧光定量PCR结果显示,GSG1基因在睾丸组织中的表达量最高,显著高于其余8个组织(P<0.05),且其在肌肉、小肠、肺脏、心脏、肾脏和脂肪中的表达水平极低;GSG1基因在睾丸组织的E90、D1、D30和D60这4个发育时期的表达量较低,显著低于D90、D120、D150和D180这4个时期(P<0.01).本试验首次克隆了猪GSG1基因cDNA全长并检测了其在不同组织和不同发育阶段睾丸组织的表达,为进一步研究其调控猪睾丸发育和精子生成的机制奠定了理论基础.

摘要:
为利用分光光度计原理测定鸡精子密度,对黑丝羽乌骨鸡及其杂交公鸡的精液,分别用吸光度法和计数板法进行精子密度的测定,得出722型分光光度计和手持精子密度仪所测定吸光度与鸡精子密度之间的回归方程。结果表明:鸡精子密度与722型分光光度计和精子密度仪吸光值之间均呈幂函数回归关系,回归方程分别为Y=19.959xX^0.975(R^2=0.928)、Y=10.834xX^0.970(R^2=O.940),与精子密度仪相关性更好。试验结果为将此回归方程植入简单且性价比高的国产手持精子密度仪,开发专用的鸡精子密度仪奠定了基础。

摘要:
试验旨在研究放牧饲养条件下桃源鸡的屠宰性能及肌肉常规品质.选取同一批饲养的24只(公母各半)7月龄平均体重为(1.98±0.20)kg、符合品种标准的桃源鸡进行屠宰和肉品质测定.结果:①桃源鸡屠宰率、全净膛率、胸肌率、腿肌率分别为91.39%、71.54%、12.24%、18.17%;公鸡全净膛率、腿肌率极显著高于母鸡(P<0.01).②桃源鸡肉粗蛋白、粗脂肪、粗灰分、水分含量平均分别为23.43%、2.50%、1.22%、72.34%;公鸡胸肌粗脂肪含量显著低于母鸡(P<0.05),腿肌粗脂肪含量极显著低于母鸡(P<0.01).(③桃源鸡公鸡剪切力显著高于母鸡(P<0.05);④桃源鸡胸肌、腿肌的肌纤维直径平均分别为23.58、22.58 μn,肌纤维密度分别为830.17、878.16根/mm2,公鸡腿肌肌纤维密度显著高于母鸡(P<0.05).由此可知,桃源鸡具有屠宰性能好、肉质细嫩、营养丰富、肉品质好等特点,是一个优良的地方鸡种.

摘要:
可变剪接作为一种增加基因组和蛋白质组多样性的重要机制, 广泛发生于真核生物基因转录的过程中.本文采用Illumina Hiseq 2500测序平台对猪睾丸组织4个不同发育时期(60胚龄、90胚龄、30日龄和180日龄)进行转录组测序, 以猪基因组数据为参考, 鉴定和分析了猪基因组可变剪接事件. 从猪基因组中鉴定出20398个基因(80.6%)对应的92738个可变剪接事件.在不同的可变剪接类型中, 以第一个外显子可变剪切(alternative 5′ first exon, TSS)、最后一个外显子可变剪切(alternative 3′ last exon, TTS)、单外显子跳跃(skipped exon, SKIP)和可变5′或3′端剪切(alternative exon ends, AE) 4种类型为主,分别占所有可变剪接事件的41.0%, 32.9%, 7.6%和7.4%. GO功能富集分析显示, 发生可变剪接的基因主要富集于物质合成、物质结合及酶活性相关的GO项中, 而各发育时期特异的可变剪接基因与发育时期的生理状态密切相关, 60胚龄时主要与酶活性和组织形成相关, 30日龄时主要与抗环境应激和离子通道活性相关, 180日龄时则主要与循环系统相关. 此外,在筛选出64个与睾丸素代谢相关的基因中, 63个基因发生可变剪接, 且以TSS和TTS为主,表明这两种可变剪接类型与睾丸素合成和分泌密切相关.通过对猪基因组可变剪接的分析,为深入研究可变剪接生物学功能及进一步开展分子育种工作提供理论依据.

摘要:
Abstract Jintang black goat (Capra hircus) is an indigenous breed of Sichuan province of China. It is the first time that the complete mitochondrial genome sequence of Jintang black goat is reported in this work, which is determined through the PCR-based method. The total length of the mitogenome is 16,813 bp, which contains two ribosomal RNA genes, 22 tRNA genes, 13 PCGs, and a control region (D-loop region). The total base composition of Jintang black goat mitochondrial genome is 33.52% A, 13.13% G, 27.35% T, and 26.00% C, and in the order A > T > C > G. The complete mitochondrial genome of Jintang black goat provides an important data in genetic mechanism and the evolution genomes.

摘要:
研究对黔邵花猪的胴体性状、肉品质及背最长肌中的营养组分进行分析测定。试验随机选取体重相近的黔邵花猪(公)3头,禁食24 h进行屠宰测定,分离留取100 g左右背最长肌肉进行营养组分测定。结果显示,黔邵花猪的屠宰率较高为76.06%,瘦肉率较低为41.56%,背膘较厚为3.51 cm,脂率较高为33.64%,大理石纹评分较高为4.33,贮存损失较低为3.62%。营养组分上,背最长肌中粗蛋白质和粗脂肪含量较高分别为64.69%和18.75%,风味氨基酸和不饱和脂肪酸含量较高分别为23.03%和61.94%,此外,微量矿物质元素含量较丰富。由此可见,黔邵花猪具有较好的胴体性能和肉质特性。

摘要:
为测定湘西黑猪的胴体性状、肉品质及背最长肌中的营养组分,随机选取体重相近的湘西黑猪母猪3头,禁食24 h进行屠宰测定,分离留取100 g左右背最长肌进行营养组分分析。结果显示,湘西黑猪屠宰率较高为74.88%,瘦肉率适中为46.58%,背膘较厚为3.37 cm,脂率较高为30.93%,贮存损失较低为2.96%。营养组分上,背最长肌中粗蛋白质和粗脂肪含量较高分别为62.10%和14.70%,风味氨基酸和不饱和脂肪酸含量较高分别为30.67%和58.83%。此外,肌肉中矿质元素含量较丰富,肌纤维多而纤细。由此可见,湘西黑猪产肉适中,体脂较高,肉鲜香耐贮,肉质细嫩营养丰富,具备肉质优势。

摘要:
Thousands of long noncoding RNAs (lncRNAs) have been identified in mouse, rat, and human testes, some of which play important roles in testis development and spermatogenesis. However, systematic analysis of lncRNAs expressed in postnatal pig testes has not been reported. Thus, in this study, we present the expression and characterization of lncRNAs in immature (30-day-old [D30]) and mature (180-day-old [D180]) pig testes. A total of 90 440 168 (85.75%) and 97 001 700 (95.35%) 150-base-pair paired-end clean reads were generated in D30 and D180 cDNA libraries, respectively, using the Illumina HiSeq 4000 platform; 36 727 transcripts were assembled in those two libraries, 777 lncRNA transcripts from 752 lncRNA gene loci were identified using the highly stringent pipeline, and 101 of those lncRNA transcripts were significantly differentially expressed. Those lncRNAs shared some characteristics with other mammals, including fewer exons, shorter length and exon length, and lower expression level compared with those of protein-coding genes; 402 protein-coding genes (<10 kb) were found as nearest neighbors of 294 out of 752 lncRNA genes, and gene ontology enrichment showed that they were enriched in transcription- and development-related processes. Fifteen differentially expressed and 10 novel lncRNAs were randomly selected and validated by quantitative polymerase chain reaction (PCR) and reverse transcriptase PCR. In addition, one of the 10 novel lncRNAs was further confirmed using RACE clone technology. This study provides a catalog of porcine testes lncRNAs for further understanding their regulation roles in pig testis development and spermatogenesis. © 2016 by the Society for the Study of Reproduction, Inc.

本试验选择体重79kg 左右的大巴沙三元杂交猪78 头, 随机分成三个组(对照组、学府鲜组、自配调理剂组),每组26头.对照组饲喂基础日粮, 学府鲜组在基础日粮中添加1%的学府鲜调理剂, 自配调理剂组在基础日粮中添加1%的中草药微生物发酵添加剂.试验期3 1 天,饲养试验结束后,每组随机抽取6耀8 头进行屠宰测定.结果表明: 日增重方面,学府鲜调理剂与自配调理剂分别比对照组提高13 . 42 % ( P〈0 . 05 ) 和9 . 73%% ( P〈0 . 0 5 ) ; 料重比均比对照组降低15 . 76 % .同时, 日粮中添加学府鲜调理剂和自配调理剂有改善试验猪胴体品质的趋势,且胴体品质优良.据此结果, 笔者认为课题组研制的中草药微生物发酵添加剂用于生猪生产是可行的.

摘要:
Duroc (male symbol) x [Landrace (male symbol) x Yorshire (female symbol)] (D x LY) pig is the popular hybrid pigs in order to make the most use of the heterosis in the world. In this study, the complete nucleotide sequence of D x LY pig mitochondrial genome was determined for the first time. Sequence analysis showed that the genome structure was in accordance with other pig breeds. It contained 22 tRNA genes, 2 ribosomal RNA genes, 13 protein-coding genes and 1 control region (D-loop region). The complete mitochondrial genome sequence of the D x LY pig provides an important data set for further study in genetic mechanism.

The complete mitochondrial genome sequence of the Qianshao spotted pig enriches data resource for further study in genetic mechanism.

In this work, we reported the complete mitochondrial genome sequence of the Ningxiang pig (Human Province), which was determined through PCR-based method. The total length of the mitogenome is 16,690 bp. The overall composition of the mitogenome was estimated to be 34.70% for A, 25.81% for T, 26.18% for C, 13.30% for G, respectively, indicating that an A + T(60.52%)-rich feature occurs in the Ningxiang pig. It contains the typical structure, including 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and 1 non-coding control region (D-loop region). The arrangement of these genes was the same as that found in the Landrace pig. All the protein initiation codons are ATG, except for ND2. ND3 and ND5 are ATA, ND4L is GTG. The complete mitochondrial genome sequence of the Ningxiang pig in Human Province provides an important data for further study about genetic mechanism. © 2013 Informa UK Ltd.

摘要:
Taoyuan Black pig is the native breed of Hunan province in China. In this study, the complete nucleotide sequence of Taoyuan Black pig mitochondrial genome was first determined. Sequence analysis showed that the genome structure was in accordance with other pig breeds. It contained 22 tRNA genes, 2 ribosomal RNA genes, 13 protein-coding genes and 1 control region (D-loop region). The complete mitochondrial genome sequence of the Taoyuan Black pig provides an important data set for further study on genetic mechanism. © 2014 Informa UK Ltd.

Daweizi pig is one of the famous native breed in China. In this work we reported the complete mitochondrial genome sequence of the Daweizi pig in Human Province for the first time. The total length of the mitogenome is 16,690 bp, with the base composition of 34.68% for A, 25.80% for T, 26.21% for C, 13.31% for G and an A + T (60.48%)-rich feature occurs in the Daweizi pig. 摘要:
A recent study indicated that the JAZF1 gene was related to lipid metabolism by regulating the level of gene expression in humans and mice. In order to investigate whether JAZF1 gene expression was associated with fat deposition in pig, we cloned the full-coding region of the JAZF1 gene (GenBank accession No. KF307636) from porcine longissimus dorsi. Results showed that the open reading frame of JAZF1 covered 732 bp and encoded 243 amino acids. Multiple alignment of isoform sequences revealed that the deduced amino acid sequence of JAZF1 had a high degree of sequence similarity to other vertebrates, indicating that it was highly conserved during evolution. Bioinformatic analysis indicated that pig JAZF1 contained 23 phosphorylation sites and 19 glycosyl sites. JAZF1 was predicted to have 3 ZnF-C2H2 and 2 low-complexity domains. The JAZF1 mRNA expression pattern indicated that JAZF1 mRNA expression level in the liver was significantly different in 2 divergent breeds (P < 0.05). This article perhaps provided an important experimental basis for further research on the mechanisms of lipid metabolism and fat deposition in pigs. ©FUNPEC-RP.

为探讨肉质性状候选原癌基因(c-FOS)在国内外不同猪种群体间的遗传变异,本研究以大白猪、杜洛克猪、长白猪、宁乡猪、桃源黑猪、沙子岭猪和大围子猪7个猪种为研究对象,以PCR-RFLP方法检测该基因G2910A位点的遗传变异。结果表明,大白猪、长白猪和杜洛克猪3个外来品种的优势基因型均为BB型,A等位基因是宁乡猪、大围子猪和沙子岭猪3个地方品种的优势等位基因,大围子猪、杜洛克猪和大白猪B是优势等位基因,长白猪全是B等位基因。这与中外不同猪种肉质性状差异较大相一致,本研究为猪肉质性状分子标记辅助选择奠定基础。
