利用正交设计优化羊食道口线虫SRAP-PCR反应体系

摘要:
本研究利用正交设计L16(4)对羊食道口线虫SRAP-PCR反应体系的4因素:Taq酶、Mg2+、dNTPs、引物进行摸索。结果表明:各因素的不同水平对SRAP-PCR反应结果都有显著的影响,正交设计组合4中扩增的条带最清晰;羊食道口线虫SRAP-PCR最佳反应条件:最佳引物给合为Me1/Em7,最佳反应体系为25μL:2.5μL 10×PCR buffer、20 ng模板DNA、Mg2+(25 mmol/L)3μL、dNTPs (2.5 mmol/L/μL)3μL、引物(10 pmol/μL)4μL、rTaq DNA聚合酶(5U/μL)0.2μL、灭菌ddH20补足。本研究结果为进一步采用SRAP技术来研究食道口线虫遗传进化奠定了坚实基础。

摘要:
胃瘤线虫病是一种由寄生于鱼类、鸟类体内的线虫引起的疾病,对野生动物保护、养殖业均构成较大威胁.试验于2012年4月 2013年4月用蠕虫学剖检方法检查了58839副黄鳝内脏胃瘤线虫的感染情况,并对其感染平均丰度、虫体长度、体长与存活时间关系进行观察与分析,同时测定了黄鳝血液pH.结果显示,胃瘤线虫的平均丰度为5.45%,其中春夏季平均丰度(7.85%)大于秋冬季平均丰度(4.08%),野生黄鳝平均丰度(4.08%)明显高于养殖黄鳝平均丰度(1.10%),表明胃瘤线虫平均丰度受季节及养殖方式的影响;测量300条胃瘤线虫平均长度为49.69 mm(体长范围为26.80~70.50 mm),春夏季及秋冬季虫体平均长度分别为47.10、54.86 mm,表明胃瘤线虫秋冬季长度整体长于春夏季;虫体长度在<30、30~39.9、40~49.9、50~59.9、>60 mm的平均存活时间分别为2.5、3.6、4.4、5.4、9.0d,表明虫体存活时间与虫体长度呈一定的正相关;黄鳝血液pH无明显季节性变化.本试验结果可为湖南省胃瘤线虫病防制提供依据.

The present study examined sequence variation in three mitochondrial DNA (mtDNA) genes, namely cytochrome c oxidase subunit 3 (cox3) and NADH dehydrogenase subunits 1 and 4 (nad1 and nad4), among Ascaridia galli isolates from different geographical localities in China. A portion of cox3 (pcox3), nad1 (pnad1) and nad4 (pnad4) genes were amplified by polymerase chain reaction (PCR) separately from adult A. galli individuals and the amplicons were subjected to sequencing from both directions. The length of the sequences of pcox3, pnad1 and pnad4 were 408Â bp, 471Â bp and 333Â bp, respectively. The intraspecific sequence variations within A. galli were 0-1.7% for pcox3, 0-2.8% for pnad1 and 0-3.4% for pnad4. The A+T contents of the sequences were 67.16-67.65% (pcox3), 67.09-67.94% (pnad1) and 69.91-71.77% (pnad4). The interspecific sequence differences among members of the Ascaridida were significantly higher, being 13.2-30.9%, 12.8-29.0% and 15.1-34.1% for pcox3, pnad1 and pnad4, respectively. Phylogenetic analyses using combined sequences of pcox3, pnad1 and pnad4, with three different computational algorithms (Bayesian analysis, maximum likelihood and maximum parsimony), all revealed distinct groups with high statistical support. These findings demonstrated the existence of intraspecific variation in mitochondrial DNA (mtDNA) sequences among A. galli isolates from different geographical regions in China, and have implications for studying molecular epidemiology and population genetics of A. galli. 摘要:
本研究旨在比较不同来源的多拉菌素对猪消化道线虫和体表寄生虫的驱杀效果,为临床应用该药提供科学依据.将自然感染寄生虫的猪,随机分为高、中、低3个剂量的药物试验组和不给药物治疗对照组,将健康无寄生虫感染的猪作为对照,根据试验前和试验后7、14、28、35天猪寄生虫数量和临床变化判定疗效.结果显示海正多拉与通灭对猪只的体表寄生虫及消化道线虫均有极好的驱杀作用,临床推荐剂量以中剂量组300ug/kg体重肌肉注射为宜.

摘要:
Little information is available about prevalence of helminthes in pigs in Hunan Province. In the present study, the prevalence of helminths in adult pigs was investigated in Hunan Province, People's Republic of China between 2009 and 2012. A total of 690 pigs slaughtered in local abattoirs from ten representative administrative regions in Hunan Province were examined for the presence of helminths using traditional helminthological approach. The worms were examined, counted and identified to species according to existing keys and descriptions. A total of eight species of helminths were found in pigs which represent three classes. The results of the present investigation have important implications for the control of helminth infections in pigs in Hunan Province, China. © Medwell Journals, 2013.

摘要:
为探究胃瘤线虫幼虫体外存活影响因素,观察了黄鳝源胃瘤线虫幼虫在不同温度、溶液、pH值条件下的存活情况。结果表明:胃瘤线虫幼虫在-36℃、-20℃、-7℃的存活时间分别为(2±0.71) h、(5±1.15)h、(5±1.15)d;在4℃、16℃~28℃、32℃及68℃的存活时间分别为(55±5.29)d、9 d以上、3 d及(5±2) s。胃瘤线虫幼虫在pH=7.4、pH=7、pH≥8.87或pH≤5.10溶液中,存活时间分别为(15±2)d、(8±1)d、(2±1)d。胃瘤线虫幼虫在2%葡萄糖溶液、任氏液、0.9%生理盐水、蒸馏水、高浓度(≥6.5%)NaCl溶液中存活时间分别为(14±2)d、(11.67±1.53)d、(10.33±3.21)d、(8±1)d、(4.33±0.58)h。本试验结果可为胃瘤线虫幼虫的实验室保存及肉品安全参数提供依据。

通过对动物医学院大学生问卷调查分析,得出动物医学院大部分学生对创业感兴趣,且对创业的认识比较理智;动物医学院学生创业方向主要集中在养殖场、宠物医院、相关产品代售。然而,也存在着大部分学生对创业环境不太满意,认为创业有资金问题、经验不足等障碍;半数以上的学生自身创业知识不足,对国家创业相关政策了解不多;也有部分学生从未参加创业活动。建议学校加强创业指导的力度,提供有利的条件和平台,营造良好的创业氛围;学院拓展创业教育指导方式和途径,利用好“校创业孵化基地”及“创业讲坛”,采取案例分析讨论、提供创业项目、开展运行好企业班和科创课题兴趣小组等多种方式开展创业教育。

摘要:
本研究旨在阐明黄鳝胃瘤线虫湖南分离株的核糖体DNA(rDNA)内转录间隔区(ITS)及5.8SrDNA序列的遗传变异情况,并用ITS序列重构胃瘤线虫与其它线虫的种群遗传关系。利用聚合酶链反应(PCR)扩增胃瘤线虫rDNA的ITS-1、5.8S及ITS-2片段,将PCR扩增出的片段纯化后克隆至pGEM-T Easy载体,重组质粒通过菌落PCR鉴定后,对阳性菌落进行序列测定并进行序列分析。结果显示所获得的胃瘤线虫ITS及5.8SrDNA序列总长存在一定差异(922~927bp),其中包含部分的18S、28S及全部的ITS-1(350~351bp)、5.8S(102bp)及ITS-2(340~344bp)序列。本研究系国内首次报道胃瘤线虫的ITS序列,其结果为黄鳝胃瘤线虫的分类鉴定以及进一步的分子流行病学调查和群体遗传研究奠定了基础。

为研究蛇钩口线虫长沙分离株的核糖体DNA (rDNA)内转录间隔区(ITS)序列的遗传变异情况,并利用ITS序列构建蛇钩口线虫与其它线虫的种群遗传关系,本研究利用PCR扩增蛇钩口线虫rDNA的ITS片段,并克隆至pGEM-T载体中进行序列测定及分析.结果显示,长沙市各分离株的ITS序列长度均为734 bp,与其它线虫的同源性均低于83.9%,而与十二指肠钩口线虫的同源性较高.本研究为蛇钩口线虫的分类鉴定以及进一步的分子流行病学调查和群体遗传研究奠定了基础.

沼气工程对畜禽粪便污染环境成本的控制效果

摘要:
沼气工程是治理畜禽粪便污染的主要方法,可提供清洁能源,减少温室气体排放,减轻环境污染,具有良好的环境效益。本研究以湖南省洞庭湖区为例,利用《气候变化框架公约》清洁机制执行理事会批准的方法(ACM0010)和环境成本估算方法,分析了沼气工程对畜禽粪便污染的环境成本的控制潜力。结果表明,在湖南省洞庭湖区的畜禽粪便均采用沼气项目进行污染治理条件下,年出栏5 000头以上的生猪规模养殖场均进行沼气发电,其他畜禽养殖场的沼气全部用于畜禽养殖场及周边居民的供热和照明使用,则2006年可减排温室气体2 891 614 t CO2当量,减少温室气体排放环境成本为4.52亿元;同时,可减排水体污染、土壤污染、微生物污染控制的环境成本分别为4.56亿元、1.69亿元、5.32亿元。因此,要支持和鼓励养殖户利用沼气工程处理畜禽粪便,实现畜禽粪便污染治理环境效益、经济效益和社会效益的有机统一。

Sequence variability in two mitochondrial DNA (mtDNA) regions, namely cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase subunit 4 (nad4), and internal transcribed spacer (ITS) of rDNA among and within three cestodes, Spirometra erinaceieuropaei, Taenia multiceps and Taenia hydatigena, from different geographical origins in China was examined. A portion of the cox1 (pcox1), nad4 genes (pnad4) and the ITS (ITS1+5.8S rDNA+ITS2) were amplified separately from individual cestodes by polymerase chain reaction (PCR). Representative amplicons were subjected to sequencing in order to estimate sequence variability. While the intra-specific sequence variations within each of the tapeworm species were 0-0.7% for pcox1, 0-1.7% for pnad4 and 0.1-3.6% for ITS, the inter-specific sequence differences were significantly higher, being 12.1-17.6%, 18.7-26.2% and 31-75.5% for pcox1, pnad4 and ITS, respectively. Phylogenetic analyses based on the pcox1 sequence data revealed that T. multiceps and T. hydatigena were more closely related to the other members of the Taenia genus, and S. erinaceieuropaei was more closely related to the other members of the Spirometra genus. These findings demonstrated clearly the usefulness of mtDNA and rDNA sequences for population genetic studies of these cestodes of socio-economic importance. © 2011 Cambridge University Press.

为了解郴州市黑斑蛙裂头蚴的感染情况,控制该虫在郴州市的流行,从郴州五个地方的菜市场购买428只黑斑蛙,利用寄生虫学调查方法对郴州市黑斑蛙裂头蚴感染情况进行调查,统计感染强度,计算感染率.检测结果显示,有126只感染裂头蚴,感染率为29.44%.其中,郴州安仁黑斑蛙裂头蚴感染率最高为43.75%,感染强度为1~15条.

Representative amplicons were subjected to sequencing in order to estimate sequence variability. The sequences of pcox3, pnad1 and pnad4 were 541, 607 and 847'bp in length, respectively. The A+T contents of the sequences were 68.39-68.76% (pcox3), 63.76-64.91% (pnad1) and 67.18-67.77% (pnad4), respectively, while the intra-specific sequence variations within each of the S. erinaceieuropaei spargana were 0-1.5% for pcox3, 0-2.8% for pnad1 and 0-2.7% for pnad4. Phylogenetic analysis using neighbour joining (NJ), maximum likelihood (ML) and maximum parsimony (MP) methods, indicated that all the spargana isolates in Hunan Province represented S. erinaceieuropaei. These findings demonstrated clearly the usefulness of the three mtDNA sequences for population genetics studies of S. erinaceieuropaei spargana of human and animal health significance. © 2011 Cambridge University Press.

In the present study, sequence-related amplification polymorphism (SRAP) was utilized to study the genetic variability among Schistosoma japonicum isolates from different provinces in China, using Schistosoma mansoni from Puerto Rico for comparison. Five out of ten tested SRAP primer combinations displayed significant polymorphisms among S. japonicum isolates from China, namely ME2/EM1, ME4/EM1, ME4/EM6, ME5/EM4 and ME5/EM5. Analysis of the 61 S. japonicum samples from China with five SRAP primer combinations identified a total of 83 reproducible polymorphic fragments. The number of fragments using each primer combination ranged from 14 to 19, with an average of 16 polymorphic bands per primer pair, and the size of fragment ranged approximately from 100 to 1000bp. Representative-specific SRAP fragments were excised from the gels, and confirmed by PCR amplification of genomic DNA using primers designed and based on the sequences of these SRAP fragments. Based on SRAP profiles, unweighted pair-group method with arithmetic averages (UPGMA) dendrogram was constructed. UPGMA clustering algorithm categorized S. japonicum isolates from China into nine clades and two lineages (representing the mountainous and lake/marshland regions). These results indicate the usefulness of the SRAP technique for revealing genetic variability among S. japonicum isolates from China, and the SRAP technique should be applicable to other living organisms. © 2011 WILEY-VCH Verlag GmbH & Co. 长沙市蛇寄生钩虫感染情况调查

摘要:
为了解湖南省长沙市蛇寄生钩虫感染情况,2009年3月~2010年1月,在长沙市2个主要的蛇来源市场随机抽取180条蛇进行寄生钩虫感染情况调查。结果显示,124条蛇体内检测到钩虫,感染率为68.89%;2个市场的感染率分别为75%、58.82%。可见,长沙市钩虫感染比较严重,开展钩虫病的预防工作十分必要。

摘要:
Mitochondrial (mt) genome sequences provide useful markers for investigating population genetic structures, systematics and phylogenetics of organisms. Although Taenia multiceps, T. hydatigena, and T. taeniaeformis are common taeniid tapeworms of ruminants, pigs, dogs, or cats, causing significant economic losses, no published study on their mt genomes is available. The complete mt genomes of T. multiceps, T. hydatigena, and T. taeniaeformis were amplified in two overlapping fragments and then sequenced. The sizes of the entire mt genome were 13700 bp for T. multiceps, 13489 bp for T. hydatigena, and 13647 bp for T. taeniaeformis. Each of the three genomes contains 36 genes, consisting of 12 genes for proteins, 2 genes for rRNA, and 22 genes for tRNA, which are the same as the mt genomes of all other cestode species studied to date. All genes are transcribed in the same direction and have a nucleotide composition high in A and T. The contents of A+T of the complete genomes are 71.3% for T. multiceps, 70.8% for T. hydatigena, and 73.0% for T. taeniaeformis. The AT bias had a significant effect on both the codon usage pattern and amino acid composition of proteins. T. multiceps and T. hydatigena had two noncoding regions, but T. taeniaeformis had only one. Phylogenetic analyses based on concatenated amino acid sequences of 12 protein-coding genes revealed that T. multiceps, T. hydatigena, and T. taeniaeformis were more closely related to the other members of the Taenia genus, consistent with results of previous morphological and molecular studies. The present study determined the complete mt genome sequences for three Taenia species of animal and human health significance, providing useful markers for studying the systematics, population genetics, and molecular epidemiology of these cestode parasites of animals and humans.

湖南省畜禽粪便排放的时空分布特征

本文根据湖南的畜禽养殖数量、养殖结构对其粪便的时空分布进行了分析.结果表明:湖南省畜禽粪便排放量逐年增长.从年际间的变化情况看,1996-2006年的10年间,湖南省畜禽粪尿总量、BOD5、COD、NH3-N的年排放总量变化分别在12 251.49万~17 517.54万t、277.98万~411.21万t、313.19万~461.17万t、29.21万~43.70万t之间,年际间逐年增长,但变化幅度不大.单位养殖场的养殖规模越大,其粪便排放量越大.不同区域的畜禽粪便猪粪当量最大值与最小值之间相差1倍多,各地间的畜禽粪便年猪粪当量相差较大,分布不均.
