Exploring the relationship between net anthropogenic phosphorus input (NAPI) and soil available P (SAP) content could inform applied issues related to environmental quality and agronomic productivity and increase our knowledge of element biogeochemical cycles. Here, the NAPI was estimated and the SAP content determined in eight counties in subtropical China from 1980 to 2010. It is suggested that the NAPI ranging 318-924 km−2 yr−1 in 1980 had increased substantially to 865–3601 km−2 yr−1 in 2010 across the eight counties, in which the P fertilizer application was estimated to represent the largest individual source of NAPI, accounting for an average of 36.1–74.6% of the NAPI. The NAPI in agricultural land (NAPIa) was the largest component of the NAPI, and 60.7–77.1% of the NAPIa accumulated in the upper 20 cm layer of agricultural soils, which significantly increased soil total–P (TP) and SAP contents. The increases in SAP, resulting from 10,000 kg P km−2 of the NAPIa (IOPNAPI), were estimated to be 1.61–4.36 mg P kg−1 in the counties. Both the correlation and variation partitioning analyses (VPAs) suggested that the soil pH and organic matter content (SOM) were the most important factors influencing the variations of IOPNAPI (determination coefficient: 72.5%). Therefore, the contribution of soil pH and SOM should be considered in enriching soil SAP levels and implementing optimal P management strategies to improving the agronomic effectiveness of P fertilization and further reduce the environmental risk of P loss in subtropical region. © 2018 Elsevier Ltd

Successful floral meristem (FM) determinacy is critical for subsequent reproductive development and the plant life cycle. Although the phytohormones cytokinin and auxin interact to coregulate many aspects of plant development, whether and how cytokinin and auxin function in FM determinacy remain unclear. Here, we show that in Arabidopsis thaliana, cytokinin homeostasis is critical for FM determinacy. In this developmental context, auxin promotes the expression of AUXIN RESPONSE FACTOR3 (ARF3) to repress cytokinin activity. ARF3 directly represses the expression of ISOPENTENYLTRANSFERASE (IPT) family genes and indirectly represses LONELY GUY (LOG) family genes, both of which encode enzymes required for cytokinin biosynthesis. ARF3 also directly inhibits the expression of ARABIDOPSIS HISTIDINE KINASE4, a cytokinin receptor gene, resulting in reduced cytokinin activity. Consequently, ARF3 controls cell division by regulating cell cycle gene expression through cytokinin. In flowers, we show that AGAMOUS (AG) dynamically regulates the expression of ARF3 and IPTs, resulting in coordinated regulation of FM maintenance and termination through cell division. Moreover, genome-wide transcriptional profiling revealed both repressive and active roles for ARF3 in early flower development. Our findings establish a molecular link between AG and auxin/cytokinin and shed light on the mechanisms of stem cell maintenance and termination in the FM. © 2018 ASPB.

Bacterial wilt, caused by Ralstonia solanacearum, occurs occasionally during tobacco planting and potentially brings huge economic losses in affected areas. Soil microbes in different management stages play important roles in influencing bacterial wilt incidence. Studies have focused on the impacts of species diversity and composition during cropping periods on disease morbidity; however, the effects of the soil bacterial biomass, species diversity, species succession, and population interactions on morbidity remain unclear during non-cropping periods. In this study, we explored the soil bacterial communities in the non-cropping winter fallow (WF) and cropping late growing (LG) periods under consecutive monoculture systems using 16S ribosomal RNA gene sequencing and qPCR and further analyzed their effects on tobacco bacterial wilt incidence. We found that the bacterial communities in the WF period were significantly different from those in the LG period based on detrended correspondence analysis and dissimilarity tests. Crop morbidity was significantly related to bacterial community structure and to the presence of some genera during WF and LG periods. These genera, such as Arthrobacter, Pseudomonas, Acidobacteria GP6, and Pasteuria, may be potential biological control agents for bacterial wilt. Further analysis indicated that low soil bacterial diversity during the WF period, decrease of bacterial interactions from the WF to LG periods, and low soil biomass during the LG period all have the potential to increase morbidity. In conclusion, an increase of soil bacterial diversity and control of some bacterial abundances in the WF period might be an effective approach in controlling bacterial wilt incidence. © 2018, Springer-Verlag GmbH Germany, part of Springer Nature.

Red mud and compost are two inexpensive and effective materials available for the in situ treatment of soil contaminated with heavy metals. In order to investigate the specific process involved, a detailed analysis of the synergistic effects of combined red mud and compost was carried out. The efficiency of three different treatment systems, namely compost (C), red mud (R), and a mixture of compost and red mud (R + C), on the movability, ecological risk, and biological effective Cu, Zn, Cd, and Pb over time of incubation in agricultural soil was evaluated. Results showed that the pH, water extractable organic carbon (WEOC) and total organic carbon (TOC) showed significant changes after addition of these amendment materials to the target soil. The combination of red mud and compost mixture as amendment material (R + C) resulted in the largest reduction in efficiency of the available heavy metals. In addition, the microbial biomass in the treated soil increased after addition of the amendment materials, highest increase was observed with R + C. The results indicate that these amendment materials, especially R + C could lower the ecological risk of heavy metals in soil. ©2017 Elsevier Ltd

Highly efficient simultaneous removal of Cu(II) and tetracycline (TET) from aqueous solution was accomplished by iron and zinc doped sawdust biochar (Fe/Zn-biochar). The mutual effects and inner mechanisms of their adsorption onto Fe/Zn-biochar were systematically investigated via sole and binary systems, sorption isotherm and adsorption kinetics models. The liquid-film diffusion step might be the rate-limiting step for tetracycline, the interaction of Cu(II) was more likely controlled by both intra particle diffusion model and liquid film diffusion model. The fitting of experimental data with kinetic models, Temkin model indicates that the adsorption process of tetracycline and Cu(II) involve chemisorption, and physico-chemical adsorption, respectively. There exists site competition and enhancement of Cu(II) and tetracycline on the sorption to Fe/Zn-biochar. The results of this study indicate that modification of biochar derived from sawdust shows great potential for simultaneous removal of Cu(II) and tetracycline from co-contaminated water. © 2017 Elsevier Ltd

In recent years, advanced oxidation processes (AOPs), especially sulfate radical based AOPs have been widely used in various fields of wastewater treatment due to their capability and adaptability in decontamination. Recently, metal-free carbon materials catalysts in sulfate radical production has been more and more concerned because these materials have been demonstrated to be promising alternatives to conventional metal-based catalysts, but the review of metal-free catalysts is rare. The present review outlines the current state of knowledge on the generation of sulfate radical using metal-free catalysts including carbon nanotubes, graphene, mesoporous carbon, activated carbon, activated carbon fiber, nanodiamond. The mechanism such as the radical pathway and non-radical pathway, and factors influencing of the activation of sulfate radical was also be revealed. Knowledge gaps and research needs have been identified, which include the perspectives on challenges related to metal-free catalyst, heterogeneous metal-free catalyst/persulfate systems and their potential in practical environmental remediation. © 2017 Elsevier Ltd

摘要:
Increased nitrogen (N) concentrations in water bodies have highlighted issues regarding nutrient pollution in agricultural catchments. In this study, the ammonium-N (NH4
+-N), nitrate-N (NO3
−-N), and total N (TN) concentrations were observed in the stream water and groundwater of two contrasting catchments (named Tuojia and Jianshan) in subtropical central China from 2010 to 2014, to determine the rice agriculture impacts on the hydrographic patterns, and N export characteristics of the catchments. The results suggested that greater amounts of stream flow (523.0 vs. 434.7 mm year−1) and base flow (237.6 vs. 142.8 mm year−1) were produced in Tuojia than in Jianshan, and a greater base flow contribution to stream flow and higher frequencies of high-base flow days were observed during the fallow season than during the rice-growing season, indicating that intensive rice agriculture strongly influences the catchment hydrographic pattern. Rice agriculture resulted in moderate N pollution in the stream water and groundwater, particularly in Tuojia. Primarily, rice agriculture increased the NH4
+-N concentration in the stream water; however, it increased the NO3
−-N concentrations in the groundwater, suggesting that the different N species in the paddy fields migrated out of the catchments through distinct hydrological pathways. The average TN loading via stream flow and base flow was greater in Tuojia than in Jianshan (1.72 and 0.58 vs. 0.72 and 0.15 kg N ha−1 month−1, respectively). Greater TN loading via stream flow was observed during the fallow season in Tuojia and during the rice-growing season in Jianshan, and these different results were most likely a result of the higher base flow contribution to TN loading (33.5 vs. 21.3%) and greater base flow enrichment ratio (1.062 vs. 0.876) in Tuojia than in Jianshan. Therefore, the impact of rice agriculture on catchment eco-hydrological processes should be considered when performing water quality protection and treatment in subtropical central China.

Temperature and energy resources (e.g., iron, sulfur and organic matter) usually undergo dynamic changes, and play important roles during industrial bioleaching process. Thus, it is essential to investigate their synergistic effects and the changes of their independent effects with simultaneous actions of multi-factors. In this study, we explored the synergistic effects of temperature and original mineral compositions (OMCs, energy resources) on the sphalerite bioleaching process. The microbial community structure was monitored by 16S rRNA gene sequencing technology and showed clear segregation along temperature gradients and Shannon diversity decreased at high temperature. On the contrary, the physicochemical parameters (pH and [Fe3+]) in the leachate were significantly affected by the OMCs. Interestingly, the influence of temperature on zinc recovery was greater at relatively simpler OMCs level, whereas the influence of OMCs was stronger at lower temperature. In addition, using [Fe3+], pH, relative abundances of dominant OTUs of microbial community and temperature as variable parameters, several models were constructed to predict zinc leaching efficiency, providing a possibility to predict the metal recovery efficiency under temperature change and variable energy resources. © 2017, The Author(s).

APETALA2 (AP2) is best known for its function in the outer two floral whorls, where it specifies the identities of sepals and petals by restricting the expression of AGAMOUS (AG) to the inner two whorls in Arabidopsis thaliana. Here, we describe a role of AP2 in promoting the maintenance of floral stem cell fate, not by repressing AG transcription, but by antagonizing AG activity in the center of the flower. We performed a genetic screen with ag-10 plants, which exhibit a weak floral determinacy defect, and isolated a mutant with a strong floral determinacy defect. This mutant was found to harbor another mutation in AG and was named ag-11. We performed a genetic screen in the ag-11 background to isolate mutations that suppress the floral determinacy defect. Two suppressor mutants were found to harbor mutations in AP2. While AG is known to shut down the expression of the stem cell maintenance gene WUSCHEL (WUS) to terminate floral stem cell fate, AP2 promotes the expression of WUS. AP2 does not repress the transcription of AG in the inner two whorls, but instead counteracts AG activity. © 2016 The Authors. New Phytologist © 2016 New Phytologist Trust

Background: As an Agrobacterium tumefaciens T-DNA oncogene, T-6b induces the development of tumors and the enation syndrome in vegetative tissues of transgenic plants. Most of these effects are related to increases in soluble sugar contents. To verify the potential roles of T-6b in the distribution of carbon in developing seeds, not in vegetative tissues, we fused an endosperm-specific promoter to the T-6b gene for expression in transgenic Arabidopsis thaliana plants. Results: The expression of T-6b in reproductive organs did not induce the development of the enation syndrome, and moreover, promoted endosperm expansion, which increased the total seed biomass by more than 10%. Additionally, T-6b also increased oil content in mature seeds by more than 10% accompanied with the decrease of starch and mucilage content at the same time. Conclusions: T-6b enhances seed biomass and helps oil biosynthesis but not polysaccharides in reproductive organs without disturbing vegetative growth and development. Our findings suggest T-6b may be very useful for increasing oil production in biodiesel plants. © 2017 The Author(s).

As a widely used deoxyribonuclease, DNase I is involved in many physiological processes including tumor cell proliferation, metastasis and apoptosis. Furthermore, the level of this enzyme in serum can act as a functional biomarker for the therapeutic monitoring of systemic lupus erythematosus and other diseases. We report here a low cost and sensitive DNase I detecting system based on the single-stranded fluorogenic substrate and nanographene oxide (NGO) and use it for DNase-targeted natural product screening. The system with a detection limit of 0.005 U was then used to evaluate the effect of external factors on DNase I. The results show that Hg2+, As2+, Pb2+, Cd2+ and Cu2+ can inhibit DNase I activity in a concentration-dependent manner with IC50 values of 0.37 mM (Hg2+), 2.7 mM (As2+), 5 mM (Pb2+), 5.3 mM (Cd2+) and 7.8 mM (Cu2+), respectively. Meanwhile, 10 natural compounds isolated from Cyclocarya paliurus leaves were screened as DNase I inhibitors, while 5 compounds were identified as activators. Finally, the system was used to discriminate DNase activity of serum samples with and without HBV. The results showed that HBV infection significantly decreased the level of DNase I in serum samples. In summary, these data indicate that this method with the advantages of rapidity, low cost and high sensitivity is hopeful for DNase assay in biological samples as well as compound screening in vitro.

Plant meristems are responsible for the generation of all plant tissues and organs. Here we show that the transcription factor (TF) FAR-RED ELONGATED HYPOCOTYL3 (FHY3) plays an important role in both floral meristem (FM) determinacy and shoot apical meristem maintenance in Arabidopsis, in addition to its well-known multifaceted roles in plant growth and development during the vegetative stage. Through genetic analyses, we show that WUSCHEL (WUS) and CLAVATA3 (CLV3), two central players in the establishment and maintenance of meristems, are epistatic to FHY3. Using genome-wide ChIP-seq and RNA-seq data, we identify hundreds of FHY3 target genes in flowers and find that FHY3 mainly acts as a transcriptional repressor in flower development, in contrast to its transcriptional activator role in seedlings. Binding motif-enrichment analyses indicate that FHY3 may coregulate flower development with three flower-specific MADS-domain TFs and four basic helix–loop–helix TFs that are involved in photomorphogenesis. We further demonstrate that CLV3, SEPALLATA1 (SEP1), and SEP2 are FHY3 target genes. In shoot apical meristem, FHY3 directly represses CLV3, which consequently regulates WUS to maintain the stem cell pool. Intriguingly, CLV3 expression did not change significantly in fhy3 and phytochrome B mutants before and after light treatment, indicating that FHY3 and phytochrome B are involved in light-regulated meristem activity. In FM, FHY3 directly represses CLV3, but activates SEP2, to ultimately promote FM determinacy. Taken together, our results reveal insights into the mechanisms of meristem maintenance and determinacy, and illustrate how the roles of a single TF may vary in different organs and developmental stages.

It has been perplexing that DNA topoisomerases, enzymes that release DNA supercoils, play specific roles in development. In this study, using a floral stem cell model in Arabidopsis thaliana, we uncovered a role for TOPOISOMERASE1α (TOP1α) in Polycomb Group (PcG) protein-mediated histone 3 lysine 27 trimethylation (H3K27me3) at, and transcriptional repression of, the stem cell maintenance gene WUSCHEL (WUS). We demonstrated that H3K27me3 deposition at other PcG targets also requires TOP1α. Intriguingly, the repression of some, as well as the expression of many, PcG target genes requires TOP1α. The mechanism that unifies the opposing effects of TOP1α appears to lie in its role in decreasing nucleosome density, which probably allows the binding of factors that either recruit PcG, as we demonstrated for AGAMOUS at the WUS locus, or counteract PcG-mediated regulation. Although TOP1α reduces nucleosome density at all genes, the lack of a 5′ nucleosome-free region is a feature that distinguishes PcG targets from nontargets and may condition the requirement for TOP1α for their expression. This study uncovers a connection between TOP1α and PcG, which explains the specific developmental functions of TOP1α. © 2014 American Society of Plant Biologists. All rights reserved.

Termination of the stem cells in the floral meristem (also known as floral determinacy) is critical for the reproductive success of plants, and the molecular activities regulating floral determinacy are precisely orchestrated during the course of floral development. In Arabidopsis thaliana, regulators of floral determinacy include several transcription factor genes, such as APETALA2 (AP2), AGAMOUS (AG), SUPERMAN (SUP), and CRABSCLAW (CRC), as well as a microRNA (miRNA), miR172, which targets AP2. How the transcription factor and miRNA genes are coordinately regulated to achieve floral determinacy is unknown. A mutation in POWERDRESS (PWR), a previously uncharacterized gene encoding a SANT-domain-containing protein, was isolated in this study as an enhancer of the weakly indeterminate ag-10 allele. PWR was found to promote the transcription of CRC, MIR172a, b, and c and/or enhance Pol II occupancy at their promoters, without affecting MIR172d or e. A mutation in mature miR172d was additionally found to enhance the determinacy defects of ag-10 in an AP2-dependent manner, providing direct evidence that miR172d is functional in repressing AP2 and thereby contributes to floral determinacy. Thus, while PWR promotes floral determinacy by enhancing the expression of three of the five MIR172 members as well as CRC, MIR172d, whose expression is PWR-independent, also functions in floral stem cell termination. Taken together, these findings demonstrate how transcriptional diversification and functional redundancy of a miRNA family along with PWR-mediated co-regulation of miRNA and transcription factor genes contribute to the robustness of the floral determinacy network. © 2013 Yumul et al.

The indica rice cultivar Xiangzi 3150 (XZ3150) confers a high level of resistance to 95% of the isolates of Magnaporthe oryzae (the agent of rice blast disease) collected in Hunan Province, China. To identify the resistance (R) gene(s) controlling the high level of resistance in this cultivar, we developed 286 F 9 recombinant inbred lines (RILs) from a cross between XZ3150 and the highly susceptible cultivar CO39. Inoculation of the RILs and an F 2 population from a cross between the two cultivars with the avirulent isolate 193-1-1 in the growth chamber indicated the presence of two dominant R genes in XZ3150. A linkage map with 134 polymorphic simple sequence repeat and single feature polymorphism markers was constructed with the genotype data of the 286 RILs. Composite interval mapping (CIM) using the results of 193-1-1 inoculation showed that two major R genes, designated Pi47 and Pi48, were located between RM206 and RM224 on chromosome 11, and between RM5364 and RM7102 on chromosome 12, respectively. Interestingly, the CIM analysis of the four resistant components of the RILs to the field blast population revealed that Pi47 and Pi48 were also the major genetic factors responsible for the field resistance in XZ3150. The DNA markers linked to the new R genes identified in this study should be useful for further fine mapping, gene cloning, and marker-aided breeding of blastresistant rice cultivars. © 2011 The American Phytopathological Society.

Anthocyanidin synthase (ANS) is a key enzyme of the proanthocyanidin biosynthetic pathway in Arabidopsis. In this study, we report cloning and characterization of an ANS homolog, BjANS, from Brassica juncea. The BjANS cDNA sequence is 1,377 bp long and has a 1,077-bp open reading frame encoding a deduced polypeptide of 358 amino acids with a predicted molecular weight of 40,862 Da and an estimated isoelectric point of 5.18. The putative BjANS protein shows 91-99% amino acid sequence identity with Arabidopsis thaliana, Matthiola incana, B. rapa and B. oleracea ANS proteins. The expression of BjANS was analyzed in yellow- and black-seeded B. juncea accessions to determine its physiological function. RT-PCR analysis showed that BjANS expressed in the seed coats of the black-seeded lines and in embryos of the all lines, but not in the yellow-seeded seed coats. Proanthocyanidins were also detected by histochemical analysis in the seed coat of black-seeded accessions and in embryos of all the lines, but not the yellow-seeded seed coat. Both transcriptional and histochemical analyses confirmed that BjANS was involved in the biosynthesis of proanthocyanidins and in seed coat color formation of B. juncea. Lack of BjANS expression blocked biosynthesis of proanthocyanidins in the yellow seed coat, and therefore seeds appeared yellow because of transparent testa. © 2010 Springer Science+Business Media B.V.

Cucumber (Cucumis sativus L.), which is a vegetable crop, has served as the model system for sex expression in flowering plants, and the inheritance of sex expression in cucumber is well documented. However, the genetics of subgynoecism expression in cucumber had rarely been described. In this study, we investigated the inheritance of subgynoecious traits in cucumber plants with the inbred cucumber lines of subgynoecious (C. sativus L var sativus cv 97-17 and S-2-98) as the materials. Genetic analysis had showed the two subgynoecious inbred lines were controlled by one pair of recessive gene and one pair of incompletely dominant gene, which were designated presently as mod-F2 and Mod-F1, respectively. Furthermore, the mod-F2 and Mod-F1 loci, which enhance the intensity of femaleness, also inherited independently with F and M genes. (C) 2010 Elsevier B.V. All rights reserved.

摘要:
Double B-box 1a (DBB1a) belongs to the zinc-finger family proteins in Arabidopsis thaliana. Transcriptional analysis uncovered that the DBB1a gene expression was blue light-dependently regulated, and the transcript level of DBB1a in cry1cry2 was decreased but not in phyAphyB compared to wild type under blue light conditions. Transgenic plants containing pDBB1a:GUS (β-glucuronidase) displayed GUS activity in the vascular system of leaves and petioles. Green fluorescent protein (GFP)-fused DDB1a (DBB1a-GFP) protein was found in the nucleus in transient transformation assays with onion epidermal cells as well as in stable transgenic Arabidopsis plants. To investigate the function of DBB1a, we generated DBB1a over-expressing and under-expressing transgenic Arabidopsis plants. Analysis of hypocotyl growth of these lines indicated that DBB1a promoted hypocotyl elongation under blue light condition. The phenotype of transgenic plants with DBB1a over-expression could be impaired by a gibberellin (GA)-biosynthesis inhibitor. Moreover, the expression analysis of GA metabolic and catabolic genes in DBB1a transgenic lines indicated that the DBB1a suppressed GA2-oxidase1 (GA2ox1) and GA2-oxidase8 (GA2ox8) expression, but induced GA3β-hydroxygenase1 (GA3ox1) and GA20-oxidase1 (GA20ox1) expression under blue light. Taken together, we concluded that DBB1a promotes hypocotyl elongation under blue light condition through an increase in bioactive GA levels in Arabidopsis.

摘要:
Weak organic acids including tartaric and citric acids were applied to a typical paddy soil in southern China to test their effects on rice in the field for three years. Their effects were also compared with top dressings of the two acids, basal application of diammonium acid phosphate, calcium carbonate, and the control. The results showed that rice grain yield of the citric acid treatment was significantly higher than that of the control and the same difference was also observed for the filled spikelets per panicle. There were no significant difference among all the treatments and the control on the accumulation of above ground dry matter. However the root dry matter, length, peroxidase activity, and white root number of citric acid treatment were significantly higher than that of the control, while no significant differences among other treatments were observed. Basal application of citric acid played a positive role on this soil.
