Physiological responses and metal uptake of Miscanthus under cadmium/arsenic stress

摘要:
The aim of this study is to explore the impact of cadmium (Cd) and arsenic (As) stress on physiological indexes and Cd/As uptake ability of Miscanthus, including Miscanthus sacchariflorus A0104, Miscanthus sinensis C0424 and C0640. Cd and As concentration showed significant hormesis effects on some physiological indexes, such as chlorophyll content, net photosynthetic rate, and MDA content. Compared with control group, Cd uptake can be most greatly enhanced by above fourfold with 150 mg/kg Cd treatment. As uptake of A0104 was increased by 3 ~ 33 folds with 200 mg/kg As treatment, C0424 was increased by 7–12 folds with 100 mg/kg As treatment, while C0640 was increased 1 ~ 6 folds with 250 mg/kg As treatment. The results also showed that C0640 was relatively better for the Cd remediation in the high Cd concentration (150 mg/kg) contaminated soil, while A0104 and C0424 were relatively better for the As remediation in the high As concentration (100 ~ 200 mg/kg) contaminated soil. Additionally, significantly (p < 0.05) close correlations were found among physiological indexes (except MDA content), while physiological indexes showed no significant relationship with the heavy metal contents in root, stem, and leaf. © 2018, Springer-Verlag GmbH Germany, part of Springer Nature.

Background/Aims: There are known links between inflammatory bowel disease (IBD) and changes in the microbiota of the gut and inflammation and oxidative stress. In this study, a colitis model induced by dextran sodium sulfate (DSS) in mice is used to evaluate whether the presence of bioactive peptides IRW (Ile-Arg-Trp) and IQW (Ile-Gln-Trp) peptides is advantageous. Methods: The mice were arbitrarily assigned to the following four groups: (i) control (untreated), (ii) dextran sodium sulfate (DSS) treated, (iii) IRW-DSS treated, and (iv) IQW-DSS treated. For 7 days, the control group subjects had unrestricted access to untreated drinking water, whereas the drinking water supplied to the subjects in the DSS, IRW-DSS, and IQW-DSS groups during this period consisted of 5% DSS solution. The colonic lesions were scored after hematoxylin and eosin staining. Serum antioxidant capacity was analyzed by 2,2'-azino-bis(3-ethylbenz-thiazoline-6-sulfonic acid) (ABTS) radical cation decolorization test and the microbiota in the colonic contents were sequenced by HiSeq2500 PE250. Results: The presence of DSS reduced daily weight gain, enhanced histopathology scores, and inhibited antioxidant enzyme expression. Superoxide dismutase, catalase, and glutathione peroxidase activities in the DSS-induced colitis model were significantly enhanced (P < 0.05) in the presence of dietary IRW and IQW. Furthermore, the Simpson index was significantly increased (P < 0.05) in the presence of dietary IRW and IQW compared to the control group. IRW and IQW increased the abundance of Coprococcus-1, Ruminococcaceae-UCG-014, and Desulfovibrio compared to the control group and DSS group. Furthermore, IQW decreased the abundance of Bacteroides in relation to the control group, but increased Parabacteroides. In addition, IRW increased the level of Anaerotruncus, Oscillibacter, and Ruminiclostridium-9 compared to the control group. Conclusion: This study concludes that the presence of IRW or IQW can mitigate DSS-induced oxidative stress by improving the activities of antioxidant enzymes, increasing intestinal microbial diversity and enhancing the abundance of gut microbiota, which may help maintain the homeostasis of host health and microenvironment in a DSS-induced mouse model, thus providing a potential further treatment for IBD patients. © 2018 The Author(s). 摘要:
Many laboratory and field studies have been conducted on rice field spiders in China. There are 375 species, 108 genera, and 22 families of rice field spiders distributed within the major rice growing areas and 17 dominant species. The biological and ecological characteristics of 17 rice field spider species have been reported in detail. The biology and ecology of these species show significant differences among regions, farmland habitats, and agricultural practices. Future research should focus on rice field habitat diversity, enhancing the insecticide resistance of dominant spider populations, implementing large-scale breeding of spiders and augmentative release, breeding more leaf dominant species, conducting biosafety assessment of spiders in transgenic crops.

摘要:
To provide information on reclamation of multi-heavy metal polluted soils with conception of phytostabilization, a field survey on the uptake and accumulation of potentially toxic elements such as antimony (Sb), arsenic (As), lead (Pb), cadmium (Cd), copper (Cu), and zinc (Zn) in colonized plant species around the world’s largest antimony mine area, China, was conducted. Samples including leaves and shoots (including roots and stems) of colonized plants as well as rhizospheric soils were collected from eight sampling zones in the studied area. The results showed that the contents of Cu, Zn, and Pb in rhizospheric soils below plants were comparable to the corresponding background values of Hunan province, otherwise Sb, Cd, and As contents were extremely high (17–106, 17–87, and 3–7 times of the corresponding background values). The highest concentration of Sb was found in Aster subulatus (410 mg kg −1 ); Cd, As, and Zn were in Herba bidentis bipinnatae (10.9, 264, and 265 mg kg −1 , respectively); and Cu was in Artemisia lavandulaefolia (27.1 mg kg −1 ). It also exhibited that all the contents of As in leaves were several times of those in shoots of plants, Cd and other heavy metals showed in a similar pattern in several studied species, implying that the uptake route of these heavy metals via foliar might contribute to the accumulation. With high bioconcentration factors of heavy metals (more than 1, except for Zn), together with the growth abundance, Herba bidentis bipinnatae was considered as the most suitable colonized species for phytostabilization of the multi-heavy metal pollution in soils on this antimony mine area. © 2018, Springer Science+Business Media B.V., part of Springer Nature.

In this study, zero valent iron (ZVI) activated peroxymonosulfate (PMS) as novel technique (i.e. ZVI-PMS technology) was employed to enhance sludge dewatering. In optimal sludge dewatering conditions of ZVI and KHSO5 dosages, the specific resistance to filtration (SRF) was reduced by 83.6%, which was further decreased to 90.6% after combination of ZVI-PMS with thermal treatment at 50 °C (i.e. ZVI-PMS-T technology). Subsequently, the ESR spectrum and quenching tests demonstrated that OH[rad], rather than SO4 [rad]−, was predominant radicals in ZVI-PMS conditioning. Thereafter, the variation of physicochemical properties and the distributions and compositions of extracellular polymeric substances (EPS) were further investigated to uncover the influence of these techniques on sludge bulk properties. The results indicated that sludge particles were disintegrated into smaller particles and surface charges were neutralized, sludge flowability were elevated obviously after treatments. In ZVI cycle experiment, the high dewatering efficiency was maintained by ZVI-PMS and ZVI-PMS-T pretreatment. © 2018 Elsevier Ltd

Successful floral meristem (FM) determinacy is critical for subsequent reproductive development and the plant life cycle. Although the phytohormones cytokinin and auxin interact to coregulate many aspects of plant development, whether and how cytokinin and auxin function in FM determinacy remain unclear. Here, we show that in Arabidopsis thaliana, cytokinin homeostasis is critical for FM determinacy. In this developmental context, auxin promotes the expression of AUXIN RESPONSE FACTOR3 (ARF3) to repress cytokinin activity. ARF3 directly represses the expression of ISOPENTENYLTRANSFERASE (IPT) family genes and indirectly represses LONELY GUY (LOG) family genes, both of which encode enzymes required for cytokinin biosynthesis. ARF3 also directly inhibits the expression of ARABIDOPSIS HISTIDINE KINASE4, a cytokinin receptor gene, resulting in reduced cytokinin activity. Consequently, ARF3 controls cell division by regulating cell cycle gene expression through cytokinin. In flowers, we show that AGAMOUS (AG) dynamically regulates the expression of ARF3 and IPTs, resulting in coordinated regulation of FM maintenance and termination through cell division. Moreover, genome-wide transcriptional profiling revealed both repressive and active roles for ARF3 in early flower development. Our findings establish a molecular link between AG and auxin/cytokinin and shed light on the mechanisms of stem cell maintenance and termination in the FM. © 2018 ASPB.

Chiral pharmaceuticals (CPs), including non-steroid anti-inflammatory drugs (NSAIDs), β-blockers and some herbicide and pesticides, are widely used in aquaculture, clinical treatment and many other fields. However, people are increasingly concerned about such ubiquitous pollutants, which can frequently be detected in contaminated soil and water. In large part, the significant sources of chiral pharmaceuticals stem from industrial processes, such as the direct discharge of untreated or incompletely treated wastewaters containing chiral pharmaceuticals, incorrect storage and use, animal wastes and biosolids. The main ways for human exposure to chiral pharmaceuticals are the disease treatment process and chiral pharmaceuticals contaminants. According to the results of a series of toxic studies, some diseases, even cancers, may be associated with exposure to certain chiral pharmaceuticals. Therefore, the treatment of chiral pharmaceuticals has become an important issue. The current advanced remediation techniques for chiral pharmaceuticals include the conventional method (sorption and sonolysis), biotransformation (an aerobic granular sludge-sequencing batch reactor and constructed wetland system) and advanced oxidation processes (ozonation and photocatalysis). Herein, in this review, we summarize the current status and sources of chiral pharmaceuticals, potential effects on human health, as well as the superiority, disadvantages and prospects of current advanced remediation technologies. Moreover, we also anticipate the prospect of the future research needed for chiral pharmaceuticals pollutant remediation. © 2018 Elsevier Ltd

Excessive UDPG resulting from the mutation of UAP1 causes programmed cell death by triggering reactive oxygen species accumulation and caspase-like activity in rice

Lesion mimic mutants are valuable to unravel the mechanisms governing the programmed cell death (PCD) process. Uridine 5′-diphosphoglucose-glucose (UDPG) functions as a signaling molecule activating multiple pathways in animals, but little is known about its function in plants. Two novel allelic mutants of spl29 with typical PCD characters and reduced pollen viability were obtained by ethane methyl sulfonate mutagenesis in rice cv Kitaake. The enzymatic analyses showed that UDP-N-acetylglucosamine pyrophosphorylase 1 (UAP1) irreversibly catalyzed the decomposition of UDPG. Its activity was severely destroyed and caused excessive UDPG accumulation, with the lesion occurrence associated with the enhanced caspase-like activities in spl29-2. At the transcriptional level, several key genes involved in endoplasmic reticulum stress and the unfolded protein response were abnormally expressed. Moreover, exogenous UDPG could aggravate lesion initiation and development in spl29-2. Importantly, exogenous UDPG and its derivative UDP-N-acetylglucosamine could induce reactive oxygen species (ROS) accumulation and lesion mimics in Kitaake seedlings. These results suggest that the excessive accumulation of UDPG, caused by the mutation of UAP1, was a key biochemical event resulting in the lesion mimics in spl29-2. Thus, our findings revealed that UDPG might be an important component involved in ROS accumulation, PCD execution and lesion mimicking in rice, which also provided new clues for investigating the connection between sugar metabolism and PCD process. © 2017 The Authors. 与稻田土壤碳周转密切相关的酶活性是评价土壤肥力和肥料管理的重要指标.本研究选取秸秆还田(ST) 、化肥(NPK)和不施肥(CK)的长期定位试验田,以10 cm的间距分段采集土壤剖面0 ~ 40 cm范围内的新鲜土样,利用96微孔酶标板荧光分析法,测定参与土壤碳氮转化过程关键酶β-1,4-葡萄糖苷酶(BG)和β-1,4 -N-乙酰氨基葡萄糖苷酶(NAG)活性,探讨不同施肥措施对不同深层土壤酶活的影响.结果表明,相比不施肥的土壤,施用化肥和秸秆土壤的BG酶活性分别提高了35% ~ 118%和55% ~ 342%; NAG酶活性分别提高了9% ~ 30%和102% ~ 484%.同时,随着土层深度的增加,土壤酶活性逐渐降低,0 ~ 20 cm深层土壤酶活性显著高于20 ~ 40 cm深层土壤.在不同施肥措施中秸秆还田可高程度影响稻田深层土壤. RDA分析表明土壤碳氮含量主要与0 ~ 20 cm的土壤酶活性有显著的正相关关系,与20 ~ 40 cm的土壤酶活性呈负相关关系.综上所述,随着土壤深度增加土壤微生物量和土壤酶活性显著降低.长期施肥显著提高了不同深层土壤生物量和土壤酶活性,其中秸秆还田作用尤为突出.因此,合理的秸杆还田有利于改善稻田深层土壤肥力,优化农田土壤养分循环,为作物生长提供良好的土壤环境.

Pilot-scale three-stage surface flow constructed wetlands (SFCWs) planted with Myriophyllum aquaticum were constructed to study the organics removal performance from lagoon-pretreated swine wastewater. The removal performance of organics in the SFCWs was evaluated using deterministic and probabilistic methods and the results were consistent. The SFCWs achieved a relatively high removal efficiency (79.0-82.7%) for a wide influent COD concentration range (456-1010 mg L-1). No significant difference (p&gt; 0.05) of COD removal efficiency and first-order removal rate constant among the various strengths of influent suggested that the present loading rates (2.74-6.06 g m(-2)d(-1)) have not yet reached the maximum removal capacity of the SFCWs. The mean emission fluxes of methane from the SFCW units fed with different strengths of wastewater were 25-1210 mg m(-2)d(-1). A significantly positive correlation (p &lt; 0.01) between methane emission fluxes and COD loading rates indicated that the anaerobic digestion of organics was an important process for organics removal in the SFCWs. No significant organics accumulation in the sediment over time suggested that plant harvest could be in favor of reducing the organics accumulation in the substrate and should be considered important during management of constructed wetlands. Rosiglitazone (RG) is a well-known activator of peroxisome proliferator-activated receptor-gamma (PPARγ) and used to treat hyperglycemia and type 2 diabetes; however, its clinical application has been confounded by adverse side effects. Here, we assessed the roles of chlorogenic acid (CGA), a phenolic secondary metabolite found in many fruits and vegetables, on the differentiation and lipolysis of mouse 3T3-L1 preadipocytes. The results showed that CGA promoted differentiation in vitro according to oil red O staining and quantitative polymerase chain reaction assays. As a potential molecular mechanism, CGA downregulated mRNA levels of the adipocyte differentiation-inhibitor gene Pref1 and upregulated those of major adipogenic transcriptional factors (Cebpb and Srebp1). Additionally, CGA upregulated the expression of the differentiation-related transcriptional factor PPARγ2 at both the mRNA and protein levels. However, following CGA intervention, the accumulation of intracellular triacylglycerides following preadipocyte differentiation was significantly lower than that in the RG group. Consistent with this, our data indicated that CGA treatment significantly upregulated the expression of lipogenic pathway-related genes Plin and Srebp1 during the differentiation stage, although the influence of CGA was weaker than that of RG. Notably, CGA upregulated the expression of the lipolysis-related gene Hsl, whereas it did not increase the expression of the lipid synthesis-related gene Dgat1. These results demonstrated that CGA might function as a potential PPARγ agonist similar to RG; however, the impact of CGA on lipolysis in 3T3-L1 preadipocytes differed from that of RG. © 2018 Shu-guang Peng et al.

Maternal dietary supplementation during gestation and lactation improves the health of piglets. The purpose of this study was to determine the effect of different levels of methionine (Met) supplementation in the sows' diet during late gestation and lactation on piglets. Thirty sows were randomly divided into three groups and fed the following diets from day 90 of gestation to day 21 of lactation: (a) control group (a basal diet containing 0.36% Met), (b) 0.48% Met group (a basal diet with additional 0.12% Met), and (c) 0.60% Met group (a basal diet with additional 0.24% Met). On day 21 after farrowing, piglets of average body weight (n = 10 per group) were selected for sample collection. The results showed that the 0.48% Met and 0.60% Met diets significantly lowered the malondialdehyde content in the piglets' serum (P < 0.05). In addition, the glutathione peroxidase content was significantly increased in the 0.48% Met group (P < 0.05) and the total glutathione content was significantly reduced in the 0.60% Met group (P < 0.05) compared to the control group. Furthermore, Met supplementation of the sows' diet was associated with alterations in 37 plasma metabolites in the piglets. In the piglets' intestinal microbiota, the relative abundances of Phascolarctobacterium and Bacteroidetes in the 0.48% Met group were higher than those in the other two groups (P < 0.05). Our results suggest that a diet including 0.48% Met during late gestation and lactation can maintain the health of piglets by increasing the antioxidant capacity and changing the intestinal microbiota composition, but a higher level of Met supplementation may increase the potential risk to piglets. © 2018 The Royal Society of Chemistry.

A series of three-stage pilot-scale surface flow constructed wetlands (CWs) planted with Myriophyllum aquaticum were fed with three strengths of lagoon-pretreated swine wastewater to study nitrogen (N) removal and recovery under sustainable plant harvesting management. The CWs had mean removal efficiency of 87.7–97.9% for NH4+-N and 85.4–96.1% for total N (TN). The recovered TN mass via multiple harvests of M. aquaticum was greatest (120–222 g N m−2 yr−1) when TN concentrations were 21.8–282 mg L−1. The harvested TN mass accounted for 0.85–100% of the total removal in the different CW units. Based on mass balance estimation, plant uptake, sediment storage, and microbial removal accounted for 13.0–55.0%, 4.9–8.0%, and 33.0–67.5% of TN loading mass, respectively. The results of this study confirm that M. aquaticum is appropriate for the removal and recovery of nutrients in CW systems designed for treating swine wastewater in conjunction with sustainable plant harvesting strategies. © 2018 Elsevier Ltd

Although hundreds of genetic male sterility (GMS) mutants have been identified in maize, few are commercially used due to a lack of effective methods to produce large quantities of pure male-sterile seeds. Here, we develop a multicontrol sterility (MCS) system based on the maize male sterility 7 (ms7) mutant and its wild-type Zea mays Male sterility 7 (ZmMs7) gene via a transgenic strategy, leading to the utilization of GMS in hybrid seed production. ZmMs7 is isolated by a map-based cloning approach and encodes a PHD-finger transcription factor orthologous to rice PTC1 and Arabidopsis MS1. The MCS transgenic maintainer lines are developed based on the ms7-6007 mutant transformed with MCS constructs containing the (i) ZmMs7 gene to restore fertility, (ii) α-amylase gene ZmAA and/or (iii) DNA adenine methylase gene Dam to devitalize transgenic pollen, (iv) red fluorescence protein gene DsRed2 or mCherry to mark transgenic seeds and (v) herbicide-resistant gene Bar for transgenic seed selection. Self-pollination of the MCS transgenic maintainer line produces transgenic red fluorescent seeds and nontransgenic normal colour seeds at a 1:1 ratio. Among them, all the fluorescent seeds are male fertile, but the seeds with a normal colour are male sterile. Cross-pollination of the transgenic plants to male-sterile plants propagates male-sterile seeds with high purity. Moreover, the transgene transmission rate through pollen of transgenic plants harbouring two pollen-disrupted genes is lower than that containing one pollen-disrupted gene. The MCS system has great potential to enhance the efficiency of maize male-sterile line propagation and commercial hybrid seed production. © 2017 The Authors. Genetically modified Bacillus thuringiensis (Bt) crops, which have been widely used in agricultural transgenic plants, express insecticidal Cry proteins and release the toxin into soils. Taking into consideration the environmental risk of Cry proteins, biosurfactant—rhamnolipids were applied to desorb Cry proteins from soil environment, which has not been elucidated before. Quartz crystal microbalance with dissipation (QCM-D) was used in this article to investigate the adsorption and desorption behaviors of Cry1Ac on SiO<inf>2</inf>surface (model soil). Results showed that patch-controlled electrostatic attraction (PCEA) governed Cry1Ac adsorption to SiO<inf>2,</inf>and the solution pH or ionic strength can affect PCEA. The adsorption kinetics could be fitted by the pseudo-second-order model, and the adsorption isotherm was fitted to Langmuir model with correlation coefficients higher than 0.999. The desorption characteristics of Cry1Ac from SiO<inf>2</inf>were assessed in the presence of mono-rhamnolipid, di-rhamnolipid or complex-rhamnolipid. Mono-rhamnolipid exhibited the most significant positive effect on desorption performance. With a complete removal of Cry1Ac reached when mono-rhamnolipid concentration was up to 50 mg L<sup>−1</sup>. Additionally, the desorption was enhanced at alkaline pH range, and Cry1Ac can be completely and rapidly desorbed by rhamnolipids from SiO<inf>2</inf>at ionic strength of 5 × 10<sup>−2</sup> M. ©2017 Elsevier B.V.

The global incidence of inflammatory bowel disease (IBD), a group of chronic gastrointestinal disorders, has been rising. The preponderance of evidence demonstrates that oxidative stress (OS) performs a critical function in the onset of IBD and the manner of its development. The purpose of this review is to outline the generation of reactive oxygen species and antioxidant defense mechanisms in the gastrointestinal tract and the role played by OS in marking the onset and development of IBD. Furthermore, the review demonstrates the various ways through which OS is related to genetic susceptibility and the mucosal immune response. The experimental results suggest that certain therapeutic regimens for IBD could have a favorable impact by scavenging free radicals, reducing cytokine and prooxidative enzyme concentrations, and improving the antioxidative capabilities of cells. However, antioxidative activity characterized by a high level of specificity may be fundamental for the development of clinical therapies and for relapsing IBD patients. Therefore, additional research is required to clarify the ways through which OS is related to the pathogenesis and progression of IBD. © 2018 Guiping Guan and Shile Lan.

In this work, a label-free fluorescence biosensor for ultrasensitive and simple detection of the mecA gene of Staphylococcus aureus was proposed by using an exonuclease III (Exo III)-assisted cascade signal amplification strategy. The 3' end-extruding hairpin probe (HP) acted as the target recognition element and the caged G-quadruplex was used as the signal reporter. Without the mecA gene, the HP probe cannot be digested by Exo III, as the G-rich sequences are blocked in the stem of the HP probe. In the presence of the mecA gene, the hybridization of the mecA gene with the 3' end-extruding HP probe triggers the digestion reaction of Exo III, liberating the mecA gene and the mecA gene analogue. Both the released mecA gene and the mecA gene analogue can hybridize with other HP probes and activate another round of the cleavage reaction. Consequently, the released free G-quadruplex is "lit up" by N-methylmesoporphyrin IX (NMM), displaying a dramatically enhanced fluorescence intensity. This sensing platform showed a high sensitivity towards the mecA gene with a detection limit as low as 2.4 fM without any labelling, immobilization, or washing steps. The designed sensing system also exhibits excellent selectivity for the mecA gene in the presence of other interfering DNA sequences. Furthermore, the presented biosensor is robust and has been successfully applied for the detection of the mecA gene in a real food sample with satisfactory results. Owing to its simplicity, cost-effectiveness and ultrasensitivity, our proposed sensing strategy provides a promising platform for the detection of other genes by substituting the target-recognition element.

摘要:
Recently, the onset of type 1 diabetes (T1D) has increased rapidly and became a major public health concern worldwide. Various factors are associated with the development of T1D, such as diet, genome, and intestinal microbiota. The gastrointestinal (GI) tract harbors a complex and dynamic population of microorganisms, the gut microbiota, which exert a marked influence on the host homeostasis and metabolic diseases. Recent evidence shows that altered gut bacterial composition (dysbiosis) is highly associated with the pathogenesis of insulin dysfunction and T1D and, thus, targeting gut microbiota may serve as a therapeutic potential for T1D patients. In this study, we updated the effect of gut microbiota on T1D and potential mechanisms were discussed.

Cadmium (Cd) is a heavy metal that can cause irreversible toxicity to animals, and is an environmental pollutant in farmlands. Spiders are considered to be an excellent model for investigating the impacts of heavy metals on the environment. To date, the changes at the molecular level in the cerebral ganglia of spiders are poorly understood. Cd exposure leads to strong damage in the nervous system, such as apoptosis and necrosis of nerve cells, therefore we conducted a transcriptomic analysis of Pardosa pseudoannulata cerebral ganglia under Cd stress to profile differential gene expression (DGE). We obtained a total of 123,328 assembled unigenes, and 1441 Cd stress-associated DEGs between the Cd-treated and control groups. Expression profile analysis demonstrated that many genes involved in calcium signaling, cGMP—PKG signaling, tyrosine metabolism, phototransduction–fly, melanogenesis and isoquinoline alkaloid biosynthesis were up-regulated under Cd stress, whereas oxidative phosphorylation-related, nervous disease-associated, non-alcoholic fatty liver disease-associated, and ribosomal-associated genes were down-regulated. Here, we provide a comprehensive set of DEGs influenced by Cd stress, and heavy metal stress, and provide new information for elucidating the neurotoxic mechanisms of Cd stress in spiders. © 2018, The Author(s).

Bacterial wilt, caused by Ralstonia solanacearum, occurs occasionally during tobacco planting and potentially brings huge economic losses in affected areas. Soil microbes in different management stages play important roles in influencing bacterial wilt incidence. Studies have focused on the impacts of species diversity and composition during cropping periods on disease morbidity; however, the effects of the soil bacterial biomass, species diversity, species succession, and population interactions on morbidity remain unclear during non-cropping periods. In this study, we explored the soil bacterial communities in the non-cropping winter fallow (WF) and cropping late growing (LG) periods under consecutive monoculture systems using 16S ribosomal RNA gene sequencing and qPCR and further analyzed their effects on tobacco bacterial wilt incidence. We found that the bacterial communities in the WF period were significantly different from those in the LG period based on detrended correspondence analysis and dissimilarity tests. Crop morbidity was significantly related to bacterial community structure and to the presence of some genera during WF and LG periods. These genera, such as Arthrobacter, Pseudomonas, Acidobacteria GP6, and Pasteuria, may be potential biological control agents for bacterial wilt. Further analysis indicated that low soil bacterial diversity during the WF period, decrease of bacterial interactions from the WF to LG periods, and low soil biomass during the LG period all have the potential to increase morbidity. In conclusion, an increase of soil bacterial diversity and control of some bacterial abundances in the WF period might be an effective approach in controlling bacterial wilt incidence. © 2018, Springer-Verlag GmbH Germany, part of Springer Nature.
