摘要:
Daweizi pig is one of the famous native breed in China. In this work we reported the complete mitochondrial genome sequence of the Daweizi pig in Human Province for the first time. The total length of the mitogenome is 16,690 bp, with the base composition of 34.68% for A, 25.80% for T, 26.21% for C, 13.31% for G and an A + T (60.48%)-rich feature occurs in the Daweizi pig. It is made up of two ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and one non-coding control region (D-loop region). The arrangement of these genes was the same as that found in the Landrace pig. The complete mitochondrial genome sequence of the Daweizi pig provides an important data set for further study on the germplasm resources. © 2013 Informa UK Ltd.

摘要:
A recent study indicated that the JAZF1 gene was related to lipid metabolism by regulating the level of gene expression in humans and mice. In order to investigate whether JAZF1 gene expression was associated with fat deposition in pig, we cloned the full-coding region of the JAZF1 gene (GenBank accession No. KF307636) from porcine longissimus dorsi. Results showed that the open reading frame of JAZF1 covered 732 bp and encoded 243 amino acids. Multiple alignment of isoform sequences revealed that the deduced amino acid sequence of JAZF1 had a high degree of sequence similarity to other vertebrates, indicating that it was highly conserved during evolution. Bioinformatic analysis indicated that pig JAZF1 contained 23 phosphorylation sites and 19 glycosyl sites. JAZF1 was predicted to have 3 ZnF-C2H2 and 2 low-complexity domains. The JAZF1 mRNA expression pattern indicated that JAZF1 mRNA expression level in the liver was significantly different in 2 divergent breeds (P < 0.05). This article perhaps provided an important experimental basis for further research on the mechanisms of lipid metabolism and fat deposition in pigs. ©FUNPEC-RP.

摘要:
The growth arrest-specific 6 gene (GAS6) is a member of the family of plasma vitamin K-dependent proteins, which are able to bind to phospholipids using an N-terminal gamma-carboxyglutamic acid domain. A recent report has demonstrated that the GAS6 gene can promote fat deposition and is associated with an increased number of fat cells in mice. In order to investigate whether GAS6 expression is associated with meat quality in pigs, a 2382-bp cDNA sequence of the porcine GAS6 gene (GenBank accession No. KC526197) was first obtained using rapid amplification of cDNA ends from porcine longissimus dorsi tissue. One A/G single nucleotide polymorphism anchored in exon 12 was genotyped using the marker PCR-RFLP-BglI, and the G allele was dominant in the pig breeds tested. Quantitative real-time polymerase chain reaction showed that the porcine GAS6 gene was expressed in all tissues examined in weaned male Shaziling (SZL) and Yorkshire (YS) weanling piglets, and mRNA expression of GAS6 in the longissimus dorsi tissue of SZL piglets was significantly higher than that in YS piglets (P < 0.05). The GAS6 protein was likely to be a membrane protein and was detected in longissimus dorsi tissue from SZL and YS piglets using immunohistochemistry, with an abundant protein expression index (P > 0.05). The results imply that the GAS6 gene can be considered a potential candidate for meat quality trait selection and fat deposition in pigs. © FUNPEC-RP.

摘要:
为研究不同营养水平对长白×沙子岭二元杂种猪肉理化指标的影响,选择体重25 kg左右的长×沙二元杂种猪60头,随机分成3组,分别饲喂高、中、低3种不同营养水平饲粮,每组2个重复,每个重复10头猪饲养在同一栏内,进行为期122 d的饲养试验。饲养试验结束后每组屠宰4头猪测定猪肉理化指标。结果表明,肌内脂肪含量,低水平组、中水平组分别比高水平组高1.08个百分点、0.74个百分点,差异显著(P<0.05)。硬脂酸含量,高水平组和中水平组分别比低水平组高2.80个百分点和2.38个百分点,差异显著(P<0.05);肉豆蔻酸、月桂酸含量,高水平组分别比中、低水平组高0.69个百分点和0.75个百分点、0.05个百分点和0.04个百分点,差异显著(P<0.05);饱和脂肪酸含量,高水平组和中水平组分别比低水平组高3.66个百分点和2.75个百分点,差异显著(P<0.05)。高、中水平组鲜味氨基酸、必需氨基酸及总氨基酸含量高于低水平组,但差异不显著(P>0.05);粗蛋白质、水分、粗灰分及矿物元素的含量,3组差异不显著(P>0.05);3组均未检测出砷、氟,铅含量都在安全范围内。

PCV2 is highly pathogenic, however, its effect on the serum amino acids profile is unknown. This study was conducted to explore the profile of amino acids in the serum in porcine circovirus type 2 (PCV2) infected mice. The serum levels of amino acids were detected with isotope dilution liquid chromatography-mass spectrometry methods at 3, 7, 10 and 14 days post infection (DPI). Meanwhile, the expression of seven amino acids transporters (SLC6A14, SLC6A20, SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A9) in the jejunum was analyzed by reverse transcription polymerase chain reaction (RT-PCR) at 3 and 7 DPI. Serum PCV2 load was also analyzed by quantitative PCR at 3, 7, 10 and 14 DPI. Serum levels of most amino acids, such as Pro, Orn, and Met, significantly (P < 0.05) increased at 3 DPI. However, most amino acids, including Asp, Sar, Arg, Hyl, Pro, Lys, Val, Ile and Leu, significantly (P < 0.05) decreased at 7 DPI. There was no significant difference for most amino acids at 10 and 14 DPI. PCV2 infection significantly (P < 0.05) decreased expression of SLC7A5 and SLC7A6 at 7 DPI. In conclusion, PCV2 infection affects the profile of amino acids in the serum and the expression of amino acids transporters in the intestine.

Nuclear Receptor subfamily 1, group H, member 4 (NR1H4) is a receptor for bile acids and has an important role in regulating energy metabolism in liver, muscle and adipose tissues in humans and animals. In this study, we cloned the full coding region of NR1H4 gene from porcine Longissimus dorsi by Rapid amplification of cDNA end (RACE). Results indicated that the open reading frame of NR1H4 covered 1461 bp encoding 486 amino acid residues and the deduced amino acid sequence was 91-94 % identical to that of Homo sapiens, Bos taurus, Macaca mulatta, Gorilla gorilla, and Ovis aries. Bioinformatic analysis indicated that NR1H4 contained 31 phosphorylation sites with 14 serine, 6 threonine and 11 tyrosine. One single nucleotide polymorphism (SNP) was detected by PCR-RFLP in 3' untranslated region of exon 9 (NR1H4) and the allele frequency analysis showed that A allele frequency was low among 396 pigs from five breeds. The NR1H4 mRNA expression pattern showed that NR1H4 gene was expressed highly in live and Longissimus dorsi. This work provided an important experimental basis for further research on mechanism of lipid metabolism and fat deposition in pigs.

摘要:
The aim of the present study was to investigate the effect of selenium (Se), polysaccharide of Atractylodes macrocephala Koidz. (PAMK), and the combination of Se and PAMK on the immune response, heat shock protein (HSP) levels under heat stress (HS) condition in chicken spleen. Two hundred chickens were randomly divided into two groups, the HS group and the control (Con) group. Then these chickens were treated with Se (0.3 mg/kg), PAMK (200 mg/kg) alone, and the combination of Se (0.3 mg/kg) and PAMK (200 mg/kg). The antioxidative enzymes, cytokines contents, and expression levels of HSP27 and HSP70 were examined in chicken spleen. The results indicated that HS induced higher levels of TNF-α, IL-4, HSP27, HSP70, and MDA levels but lower level of IFN-γ, IL-2, Gpx, and SOD in spleen (P<0.05). These responses were ameliorated by the treatment of Se, PAMK alone, and the combination of Se and PAMK (P<0.05 or not) The results showed that under common condition, Se and PAMK could improve the immune response by enhancing the levels of some cytokines to proper levels; however, under HS condition, Se and PAMK could change the abnormal levels of cytokines and oxidative damages to ameliorate the injury induced by HS. In addition, there existed synergistic effect on the modulation of these biomarkers in chicken spleen between Se and PAMK. So both Se and PAMK play important roles in regulating the immune function in chicken. Considering the synergistic effect on immune regulation of PAMK, this herb deserves further investigation to evaluate its role in improving the effect of traditional immune regulators. © 2014 Springer Science+Business Media.

Garlic (Allium sativum L.) has long been used both for culinary and medicinal purposes by many cultures. Population and preclinical investigations have suggested that dietary garlic intake has health benefits, such as lowering the risk of esophageal, stomach and prostate cancers. Extensive studies from laboratory and animal models have revealed that garlic has a wide range of biological activities, and garlic organosulfur compounds (OSCs) are responsible for the biological activities. However, the presence and potency of garlic OSCs vary with respect to the mode of garlic preparation and extraction. Cooked or processed garlic products showed different kinds of garlic OSCs, some of which are highly unstable and instantly decomposed. These facts, possibly gave paradoxical results on the garlic effects. In this review, we first summarized the biotransformation processes of garlic alliin into different garlic OSCs as well as the garlic OSCs compositions from different garlic preparations. Next, we reviewed the chemopreventive functions and molecular mechanisms focusing on the anti-inflammation, antioxidation, anti-diabetes and anticancer activity behind different garlic OSCs.

摘要:
本试验旨在研究酵母硒和纳米硒对鹌鹑产蛋后期生产性能、蛋品质、蛋中硒含量及血清抗氧化指标的影响。选用300只300日龄健康产蛋鹌鹑,随机分成5组,每组6个重复,每个重复10只。对照组饲喂基础饲粮,试验组分别在基础饲粮中添加0.2、0.4 mg/kg 酵母硒和0.2、0.4 mg/kg纳米硒。预试期7 d,正试期42 d。结果表明:1)与对照组相比,0.2 mg/kg酵母硒组料蛋比显著降低(P<0.05),0.4 mg/kg酵母硒组产蛋率显著提高(P<0.05);0.2 mg/kg纳米硒组产蛋率和平均蛋重显著提高(P<0.05),料蛋比显著降低(P<0.05);0.4 mg/kg纳米硒组产蛋率显著提高(P<0.05)。2)2种硒源对鹌鹑产蛋后期蛋形指数、蛋黄指数、蛋壳厚度无显著影响(P>0.05),但能极显著延缓鹌鹑蛋在储藏过程中哈氏单位的下降(P<0.01)。与对照组相比,0.4 mg/kg酵母硒和纳米硒组鹌鹑蛋的蛋黄色泽和哈氏单位均极显著提高( P <0.01)。3)与对照组相比,试验第14、28和42天,酵母硒和纳米硒组蛋中硒含量均极显著提高(P<0.01)。4)与对照组相比,酵母硒和纳米硒组血清谷胱甘肽过氧化物酶、总超氧化物歧化酶活性及总抗氧化能力均极显著提高(P<0.01),丙二醛含量极显著降低(P<0.01),纳米硒的抗氧化效果高于酵母硒。由此可见,鹌鹑产蛋后期饲喂纳米硒效果优于酵母硒,且以添加0.2 mg/kg的纳米硒为宜。

溆浦鹅肉营养成分与品质分析

摘要:
参照国家食品相关标准分析的测定方法对溆浦鹅胸肌和腿肌肉营养成分和肉品质进行了比较分析和营养品质评价.结果表明:溆浦鹅屠宰率、半净膛率、全净膛率分别为90.11%、80.45%、68.62%,脂肪率为4.93%,肝重100.75 g;屠宰24 h后pH在5.71~5.94之间,胸肌肌肉失水率27.54%,腿肌失水率20.58%,粗蛋白、粗脂肪、粗灰分、水分分别为22.64%、2.72%、2.30%、70.17%;肉中铁、铜、锌、镁、钙、硒平均含量分别为6.97 mg/100g、0.41 mg/100g、2.57 mg/100g、39.94 mg/100g、13.84 mg/100g、18.71 μg/100g;肌纤维直径39.52 μm,肌纤维密度630.1根/mm2;17种氨基酸总量(干样)为568.53 mg/g,必需氨基酸、甜味氨基酸、鲜味氨基酸平均比例分别为46.74%、26.12%、34.22%,根据氨基酸评分,溆浦鹅的第一限制性氨基酸为蛋氨酸+胱氨酸,第二限制性氨基酸为缬氨酸;肌苷酸1.435 mg/g;饱和脂肪酸32.45%,单不饱和脂肪酸46.6%,多不饱和脂肪酸21.24%,胆固醇55.94 mg/100g.与常规禽肉对比,溆浦鹅肉蛋白高、脂肪低、产肉性能好,含较丰富的硒、钙、镁、铁等矿物元素,且胆固醇含量低、肌苷酸含量高,有较好的食用价值与保健作用.

Scope: Garlic is used for both culinary and medicinal purposes by many cultures. The garlic organosulfur compounds (GOSCs) are thought to be bioactive components. This study aims to clarify the antiinflammatory effects and molecular mechanisms of GOSCs in both cell and animal models. Methods and results: RAW264.7 cells were treated with six kinds of GOSCs to screen their influence on cyclooxygenase-2 and inducible nitric oxide synthase expression by Western blotting. Prostaglandin E2 and nitrite were measured by ELISA and Griess reaction, respectively. Cytokines in culture medium were assayed by the multiplex technology. Proteins were detected by Western blotting. Mouse paw edema was induced by LPS. The results revealed that diallyl trisulfide (DATS) was a strongest inhibitor for cyclooxygenase and inducible nitric oxide synthase among GOSCs, and reduced the levels of LPS-induced IL-6, IL-10, IL-12(p70), KC, MCP-1, and TNF-α. Cellular signaling analysis revealed that DATS downregulated AKT1/TGF-β-activated kinase-mediated mitogen-activated protein kinase (MAPK) and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) pathways. Furthermore, DATS activated Nrf2-mediated expression of HO-1 and NQO1 and reduced LPS-induced intracellular reactive oxygen species, which may contribute to suppress inflammatory mediator production. Finally, in vivo data demonstrated that DATS attenuated LPS-induced mouse paw edema. Conclusion: DATS as a potential inhibitor revealed antiinflammatory effect in both cell and animal models by downregulating AKT1/TGF-β-activated kinase-mediated NFκB and MAPK signaling pathways. © 2013 WILEY-VCH Verlag GmbH & Co. 摘要:
PPAR is one cluster of the most important genes that related with lipid metabolism. And it can used as the index of goose strain selection. In this study, we collected 282 blood samples of 75 days Dongting goose to detect the SNPsof PPAPα gene by PCR-SSCP. The results showed that in the gene sequence of 215bp had a mutation of C→T and the gene frequency of A,B were 0.393,0.607, respectively, the genotype frequency of AA, AB, BB were 0.126, 0.533, 0.341, respectively. So we can concluded the polymorphism of goose PPAPα gene and the genotype frequency of heterozygote was higher in the test samples. After genotype detection of all samples, 35 AA and 96 BB genotype obtained. The abdominal fat weight of AA and BB genotype were 23.14±5.08 and 43.69±8.04, the abdominal fat ratio and liver weight ratio of AA and BB genotype were 0.80±0.30, 2.74±0.82 and 1.58±0.55, 2.87±0.69, respectively. By statistics of variance, the PPAPa genotype was highly significantly associated with abdominal fat weight, abdominal fat ratio and liver weight ratio (p<0.05). In general, the abdominal fat weight, abdominal fat ratio and liver weight ratio showed a tendency of BB>AA, indicating that the common allele was the least favorable for abdominal fat deposition. Thus, there is an enormous opportunity to decrease abdominal fat deposition in case this allele is confirmed in other more studies. © 2013 Academic Journals Inc.

武冈铜鹅与其他3种鹅肉品质特性的比较

摘要:
本试验旨在测定武冈铜鹅、四川白鹅、武川鹅以及天府肉鹅的常规肉品质和矿物质元素、氨基酸、风味脂肪酸、肌苷酸含量及肌纤维直径与密度,并进行比较。选取1日龄体格健壮的武冈铜鹅、四川白鹅、武川鹅和天府肉鹅各30只,分为4组,每组5个重复,每个重复6只鹅,公母各占1/2,饲喂相同饲粮。试验期为70 d。结果表明:1)武冈铜鹅胸肌粗蛋白质含量显著高于其他3种鹅(P<0.05),胸肌粗脂肪含量显著高于四川白鹅(P<0.05);而腿肌粗脂肪含量与其他3种鹅差异不显著(P>0.05),腿肌贮存损失率显著低于其他3种鹅(P<0.05)。2)武冈铜鹅肌肉铁、镁、钾含量均显著高于四川白鹅和天府肉鹅(P<0.05)。3)武冈铜鹅肌肉中天门冬氨酸、谷氨酸、丙氨酸、缬氨酸、赖氨酸的含量极显著高于武川鹅和四川白鹅(P<0.01),甘氨酸和脯氨酸含量极显著高于其他3种鹅(P<0.01),而胱氨酸和苯丙氨酸与其他3种鹅差异不显著(P>0.05)。4)武冈铜鹅肌肉中肉豆蔻酸含量极显著高于其他3种鹅(P<0.01);十六烯酸含量极显著高于武川鹅(P<0.01);亚麻酸含量显著高于武川鹅(P<0.05);二十烯酸含量极显著高于天府肉鹅(P<0.01)。5)武冈铜鹅胸肌肌纤维密度显著高于四川白鹅和武川鹅(P<0.05),而各组肌纤维直径没有显著差异(P>0.05)。由此可见,武冈铜鹅肌肉品质以及风味优于武川鹅、四川白鹅以及天府肉鹅。

摘要:
Enterococcus faecalis was used as probiotics and in food fermentation; however, it had became one of the leading causes of nosocomial bacteremias, surgical wound, tissue, intra-abdominal, pelvic and urinary tract infections and endocarditis. To make matters worse, E. faecalis was reservoir and vehicle of antibiotic resistance, performed resistance against many commonly used antimicrobial agents such as aminoglycosides, penicillins, tetracycline, chloramphenicol and vancomycin. Thus, E. faecalis infection has an economic and epidemiological impact on human and animal disease research worldwide. From this study in mouse model, researchers concluded that dietary arginine and glutamine supplementation ameliorated the cytokines profile and blood parameters, enhanced the clearance against E. faecal is, eventually decreased the mortality caused by E. faecalis. © Medwell Journals, 2012.

摘要:
Intestinal phosphate (Pi) absorption across the apical membrane of small intestinal epithelial cells is mainly mediated by the type IIb Na-coupled phosphate co-transporter (NaPi-IIb), but its expression and regulation in the chicken remain unclear. In the present study, we investigated the mRNA and protein levels of NaPi-IIb in three regions of chicken small intestine, and related their expression levels to the rate of net phosphate absorption. Our results showed that maximal phosphate absorption occurs in the jejunum, however the highest expression levels of NaPi-IIb mRNA and protein occurs in the duodenum. In response to a low-Pi diet (TP 0.2%), there is an adaptive response restricted to the duodenum, with increased brush border membrane (BBM) Na-Pi transport activity and NaPi-IIb protein and mRNA abundance. However, when switched from a low- (TP 0.2%) to a normal diet (TP 0.6%) for 4 h, there is an increase in BBM NaPi-IIb protein abundance in the jejunum, but no changes in BBM NaPi-IIb mRNA. Therefore, our study indicates that Na-Pi transport activity and NaPi-IIb protein expression are differentially regulated in the duodenum vs the jejunum in the chicken.

以ATP2B1基因作为仔猪腹泻相关的候选基因,在2个品种63头猪的小肠组织中,采用实时荧光定量PCR技术对肠毒素大肠杆菌(ETEC)F4受体不同黏附表型个体进行表达量的比较分析,以验证其基因功能和作为仔猪腹泻相关候选基因的可能性。结果表明:ATP2B1基因在不同黏附表型间的表达量有显著的差异,可望作为F4ab受体的相关侯选基因。

猪F4型肠毒素大肠杆菌(ETEC)是仔猪泻痢的主要病原菌之一。仔猪是否泻痢,关键的一个决定因素是,仔猪肠上皮刷状缘上是否存在ETEC F4受体。ETEC F4受体由猪基因组上的基因所控制。用δ差异显示法已筛选到ETEC F4受体相关EST序列42条,其中11条EST片段在GenBank中无同源基因。本研究用5’RACE法对这些EST片段进行了扩增,以进一步了解这些EST所在基因的特性,从而揭示仔猪泻痢的分子遗传机制,为抗泻痢猪的选育奠定基础。
