5-Caffeoylquinic acid decreases diet-induced obesity in rats by modulating PPAR and LXR transcription

Background: Chlorogenic acids (CGAs) are widely distributed in plant material, including foods and beverages. 5-Caffeoylquinic acid (5-CQA) is the most studied CGA, but the mechanism of its hypolipidaemic effect remains unclear. This study aimed to determine the effect of 5-CQA on lipid metabolism in the liver of Sprague-Dawley rats fed a high-fat diet (HFD). Results: 5-CQA suppressed HFD-induced increases in body weight and visceral fat-pad weight, serum lipid levels, and serum and hepatic free fatty acids in a dose-dependent manner. Real-time polymerase chain reaction revealed that 5-CQA altered the mRNA expression of the transcription factors peroxisome proliferator-activated receptor α (PPARα) and liver X receptor α (LXRα) and target genes involved in hepatic fatty acid uptake, β-oxidation, fatty acid synthesis, and cholesterol synthesis. Moreover, hepatic tissue sections from HFD-fed rats showed many empty vacuoles, suggesting that liver cells were filled with more fat droplets. However, 5-CQA significantly ameliorated this effect. Conclusion: 5-CQA may improve lipid metabolism disorders by altering the expression of PPARα and LXRα, which are involved in multiple intracellular signalling pathways. © 2014 Society of Chemical Industry.

摘要:
目的研究绿原酸对高脂饲喂SD大鼠骨骼肌糖代谢的调节机制。方法40只雄性SD大鼠随机分成5组:对照组、模型组,低、高剂量(20、90 mg/kg)绿原酸组,罗格列酮(阳性对照)组,其中对照组饲喂普通饲料,其余各组大鼠饲喂高脂饲料。给药12周后处死大鼠,取骨骼肌,用实时荧光定量-PCR方法测定骨骼肌糖代谢过程中相关基因蛋白激酶B(PKB,又名Akt)、磷脂酰肌醇-3激酶(PI3K)、胰岛素受体底物-1(IRS-1)、葡萄糖转运蛋白4(GLUT-4)、蛋白激酶A(PKA)、 AMP依赖的蛋白激酶(AMPK-α2)、过氧化物酶体增殖物激活受体(PPARα、PPARβ、PPARγ)的表达。结果与对照组比较,模型组大鼠骨骼肌中GLUT-4、AMPK-α2和PPARβ mRNA表达量显著下降(P<0.05、0.01);与模型组相比,绿原酸干预能提高糖代谢途径中相关基因的表达,其中绿原酸高剂量组Akt、PI3K、IRS-1、GLUT-4、PKA、AMPK-α2、PPARα、 PPARβ mRNA的表达量显著上调(P<0.05、0.01)。结论高剂量绿原酸能显著减缓高脂饲喂SD大鼠体质量增加,并通过调节PI3K/Akt途径、AMPK途径和胰岛素敏感性,改善骨骼肌中糖代谢。

摘要:
MAX2 (for MORE AXILLARY GROWTH2) has been shown to regulate diverse biological processes, including plant architecture, photomorphogenesis, senescence, and karrikin signaling. Although karrikin is a smoke-derived abiotic signal, a role for MAX2 in abiotic stress response pathways is least investigated. Here, we show that the max2 mutant is strongly hypersensitive to drought stress compared with wild-type Arabidopsis (Arabidopsis thaliana). Stomatal closure of max2 was less sensitive to abscisic acid (ABA) than that of the wild type. Cuticle thickness of max2 was significantly thinner than that of the wild type. Both of these phenotypes of max2 mutant plants correlate with the increased water loss and drought-sensitive phenotype. Quantitative real-time reverse transcription-polymerase chain reaction analyses showed that the expression of stress-responsive genes and ABA biosynthesis, catabolism, transport, and signaling genes was impaired in max2 compared with wild-type seedlings in response to drought stress. Double mutant analysis of max2 with the ABA-insensitive mutants abi3 and abi5 indicated that MAX2 may function upstream of these genes. The expression of ABA-regulated genes was enhanced in imbibed max2 seeds. In addition, max2 mutant seedlings were hypersensitive to ABA and osmotic stress, including NaCl, mannitol, and glucose. Interestingly, ABA, osmotic stress, and drought-sensitive phenotypes were restricted to max2, and the strigolactone biosynthetic pathway mutants max1, max3, and max4 did not display any defects in these responses. Taken together, these results uncover an important role for MAX2 in plant responses to abiotic stress conditions.

摘要:
为筛选高温淀粉酶高产菌株,利用淀粉酶水解圈作筛选模型,从稻田中采集土样,筛选得到产淀粉酶能力较高的菌株E10。为提高菌株产酶能力,进行了碳源种类及浓度,氮源种类及浓度,无机盐种类及浓度,pH值,种龄及接种量、温度、时间的单因素试验,并对发酵培养基及发酵条件进行了L9(34)正交优化试验。结果表明:最佳产酶发酵培养基为2.0%麸皮、0.1%硝酸钾、0.25%氯化钙,最适初始pH值为6.0。最适产酶条件为种龄24 h,接种量7 mL,于36益下培养48 h。在优化条件下,菌株产酶活力可达180.94 IU/mL。

摘要:
A feather-degrading bacterium was isolated from the gut of the tarantula Chilobrachys guangxiensis, and was classified as Bacillus subtilis (named Bacillus subtilis CH-1) according to both the phenotypic characteristics and 16S rRNA profile. The improved culture conditions for feather-degrading were 10.0gl<sup>-1</sup> mannitol, 10.0gl<sup>-1</sup> tryptone, 0.1gl<sup>-1</sup> MgCl<inf>2</inf>, 0.4gl<sup>-1</sup> KH<inf>2</inf>PO<inf>4</inf>, 0.3gl<sup>-1</sup> K<inf>2</inf>HPO<inf>4</inf>, 0.5gl<sup>-1</sup> NaCl, and 2.0gl<sup>-1</sup> intact feather, with pH 8.5 and 37°C. In the optimized medium, the intact black feather was completely degraded by Bacillus subtilis CH-1 in 24h. Furthermore, four kinds of enzymes which include extracellular protease Vpr, peptidase T, γ-glutamyl transpeptidase and glyoxalmethylglyoxal reductase were identified as having principal roles. Simultaneously, the relationship between the disulfide bond reducing activity (DRT) and the keratinase activity (KT) in B.subtilis CH-1 fermentation system was discussed. This is the first report for a feather-degrading enteric bacterium from tarantula. The identification of the enzymes shines a light on further understanding the molecular mechanism of feather-degrading by microbes. ©2014 Elsevier Ltd.

目的:研究灵芝菌发酵紫甘薯渣产物对STZ诱导的糖尿病大鼠的血糖和抗氧化水平的影响。方法:30只SD雌性大鼠随机分成3组:正常对照组(NC组)、模型组(DM组)、发酵产物干预组(L组)。测定空腹血糖(FBG)、耐糖量(OGTT)、丙二醛(MDA)、谷胱甘肽(GSH)水平及超氧化物歧化酶(SOD)活性。结果:发酵产物可显著性提高糖尿病大鼠对葡萄糖的耐受能力(P<0.05),极显著地降低FBG、MDA水平(P<0.01),极显著提高GSH水平以及SOD活性(P<0.01)。结论:灵芝菌发酵紫薯渣产物能够通过降低大鼠空腹血糖水平,提高大鼠机体抗氧化能力,较好地缓解链脲佐菌素(STZ)诱导的糖尿病症状。

多酚类物质,包括白藜芦醇、绿原酸、儿茶素等广泛存在于多种食物中,其普遍具有抗氧化、清除体内自由基等生物活性,对心血管疾病、肿瘤、癌症、糖尿病等疾病有一定的预防作用。越来越多的证据表明,食物多酚能在多个层面上影响糖代谢,如调节餐后血糖、刺激肠道激素分泌、提高胰岛素敏感性等。本文结合国内外研究结果,从肠道消化酶、肠道葡萄糖转运、微生物菌群生态及肠道激素分泌四个方面,讨论了食物多酚对肠道糖吸收与代谢的影响,以期为后期研究提供理论依据。

摘要:
Salinity stress is one of the major abiotic stresses that limit agricultural yield. To understand salt-responsive protein networks in soybean seedling, the extracted proteins from seedling roots of two different genotypes (Lee 68 and Jackson) were analyzed under salt stress by two-dimensional polyacrylamide gel electrophoresis. Sixty-eight differentially expressed proteins were detected and identified. The identified proteins were involved in 13 metabolic pathways and cellular processes. Proteins correlated to brassinosteroid and gilbberellin signalings were significantly increased only in the genotype Lee 68 under salt stress; abscisic acid content was positively correlated with this genotype; proteins that can be correlated to Ca2+ signaling were more strongly enhanced by salt stress in the seedling roots of genotype Lee 68 than in those of genotype Jackson; moreover, genotype Lee 68 had stronger capability of reactive oxygen species scavenging and cell K+/Na+ homeostasis maintaining in seedling roots than genotype Jackson under salt stress. Since the genotype Lee 68 has been described in literature as being tolerant and Jackson as sensitive, we hypothesize that these major differences in the genotype Lee 68 might contribute to salt tolerance. Combined with our previous comparative proteomics analysis on seedling leaves, the similarities and differences between the salt-responsive protein networks found in the seedling leaves and roots of both the genotypes were discussed. Such a result will be helpful in breeding of salt-tolerant soybean cultivars. © 2014 The Authors.

摘要:
Cold stress adversely affects rice (Oryza sativa L.) growth and productivity, and has so far determined its geographical distribution. Dissecting cold stress-mediated physiological changes and understanding their genetic causes will facilitate the breeding of rice for cold tolerance. Here, we review recent progress in research on cold stress-mediated physiological traits and metabolites, and indicate their roles in the cold-response network and cold-tolerance evaluation. We also discuss criteria for evaluating cold tolerance and evaluate the scope and shortcomings of each application. Moreover, we summarize research on quantitative trait loci (QTL) related to cold stress at the germination, seedling, and reproductive stages that should provide useful information to accelerate progress in breeding cold-tolerant rice.

高剂量绿原酸对脂多糖诱导肝脏炎症因子mRNA表达的影响

目的 研究不同剂量绿原酸(5-CQA)在非炎症及脂多糖(LPS)诱导急性炎症条件下,对肝组织炎症反应的影响.方法 雄性C57BL/6小鼠48只,随机分为空白对照(NC)、LPS诱导模型(LM)、绿原酸+LPS干预[5-CQA(mg/kg·d)低(LA50)、中(LA100)、高剂量(LA200)]、5-CQA (mg/kg·d)低、中、高剂量(A50,A100,A200)共8组, NC和LM组灌胃蒸馏水,其余组灌胃不同剂量绿原酸.饲喂10d后处死,取血清检测谷草转氨酶(AST)和谷丙转氨酶(ALT)活性;采集肝脏制备HE染色切片观察病理情况,实时荧光定量PCR方法测定肝脏组织肿瘤坏死因子(TNF-α)、白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、一氧化氮合成酶(iNOS)、环氧合酶(COX-2)mRNA的表达.结果 LA200组显著提高LPS诱导的急性炎症小鼠血清AST和ALT活性.同时,可显著促进急性炎症小鼠肝脏TNF-α、IL-6、IL-1β、iNOS、COX-2 mRNA表达.切片结果显示LA200组肝组织发生了巨噬细胞聚集,细胞核退化和细胞水肿现象.结论 LPS诱导急性炎症下,前期5-CQA的摄入并不能降低血清ASL、ALT活性、减少炎症因子表达,5-CQA 200mg/kg·d甚至会促进炎症因子表达,加强LPS诱导的肝脏急性炎症反应.

将雄性SD大鼠24只随机分为4组,对照组自由取食普通饲料和水,模型组自由取食高糖高脂饲料和水,这2组每天灌胃灭菌水,绿原酸高、低剂量组自由取食普通饲料和水,且每天按照90 mg/kg,20 mg/kg剂量分别灌胃绿原酸,12周后测肝脏组织中总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)和游离脂肪酸(NEFA)含量,并采用RT-PCR方法检测肝脏SOD与GSH-PX mRNA的表达量。结果表明:与模型组相比,绿原酸低剂量组肝脏中NEFA和MDA显著下降(P<0.01,P<0.05),绿原酸高剂量组肝脏NEFA和MDA显著下降(P<0.01)。与模型组相比,绿原酸低、高剂量组肝脏T-SOD显著上升(P<0.01),高剂量组肝脏CAT指标显著上升(P<0.01)。低、高剂量组SOD与GSH-PX mRNA表达水平高于模型组且呈剂量依赖性,但没有显著性差异。绿原酸提高了高脂高糖饲喂的SD大鼠肝脏抗氧化能力。

目的 探讨绿原酸(Chlorogenic acid,CGA)是否具有缓减或抑制高脂乳剂诱导小鼠产生胰岛素抵抗(Insulin Resistance,IR)的作用及其机制.方法 SPF级雌性昆明小鼠随机分为3组:对照组(Control)、高脂乳剂组(high fat emulsion,HFE)灌胃高脂乳剂、绿原酸干预组(CGA+HFE)同时灌胃高脂乳剂和绿原酸.用自动血糖仪检测口服糖耐量(OGTT).用酶联免疫法(Elisa)测空腹血清胰岛素(FINS)水平,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR).用全自动生化仪和相应的试剂盒检测血清中的甘油三酯(TC)、总胆固醇(TG)、高密度胆固醇(HDL-C)、低密度胆固醇(LDL-C)的含量.用RT-PCR法检测各组小鼠肝脏组织6-磷酸葡萄糖酶基因(glucose-6-phosphatase,G-6-Pase)和骨骼肌中葡萄糖转运体4基因(Glucose transporter 4,GLUT-4)的mRNA表达水平.并用HE染色法观察肾脏和胰腺的组织形态变化,通过免疫组化分析β细胞在胰腺中的分布与数量变化.结果 高脂乳剂组小鼠出现了明显的IR,绿原酸干预组小鼠糖耐量能力有明显的改善、ISI升高、HOMA-IR降低、血清中TG、TC、和LDL-C的含量降低、肝脏G-6-Pase mRNA水平下调、骨骼肌GLUT-4mRNA水平上调.在组织水平上,绿原酸减少了高脂乳剂对肾脏和胰腺的损伤.结论 绿原酸具有降血糖和降血脂作用,有效缓解高脂乳诱导的小鼠形成IR.

摘要:
绿原酸是一类广泛存在于自然界中的酚酸化合物,常见于人类植物源饮食中,也是很多中药材的主要组分.研究表明,绿原酸具有调节肠道激素分泌、抑制胰岛素抵抗、减缓肝脏糖异生等作用.近年来,有关绿原酸调节糖代谢研究进一步深入,例如绿原酸可以抑制胰淀粉酶活性、抑制人胰岛淀粉样多肽形成、抑制醛糖还原酶和糖基化终末产物形成等.本文综述了绿原酸在调节糖脂代谢方面的作用机制和研究进展.

摘要:
Revegetation in the water-level-fluctuation zone (WLFZ) could stabilize riverbanks, maintain local biodiversity, and improve reservoir water quality in the Three Gorges Reservoir Region (TGRR). However, submergence and cadmium (Cd) may seriously affect the survival of transplantations. Bermuda grass (Cynodon dactylon) is a stoloniferous and rhizomatous prostrate weed displaying high growth rate. A previous study has demonstrated that Bermuda grass can tolerate deep submergence and Cd stress, respectively. In the present study, we further analyzed physiological responses of Bermuda grass induced by Cd-and-submergence stress. The ultimate goal was to explore the possibility of using Bermuda grass for revegetation in the WLFZ of China's TGRR and other riparian areas. The Cd-and-submergence-treated plants had higher malondialdehyde contents and peroxidase than control, and both increased with the Cd concentration increase. All treated plants catalase activity increased with the experimental duration increases, and their superoxide dismutase also gradually increased with the Cd concentration from 1 day to 15 days. Total biomass of the same Cd-and-submergence plants increased along the experimental duration as well. Plants exposed to Cd-and-submergence stress showed shoot elongation. The heights of all treated plants were taller than those of the control. Leaf chlorophyll contents, maximum leaf length, and soluble sugars contents of all the Cd-and-submergence-treated plants were more than those of the untreated control. Although Cd inhibits plants growth, decreases chlorophyll and biomass content, and with the submergence induced the leaf and shoot elongation, more part of the Cd-and-submergence stress plants appeared in the air, exhibited fast growth with maintenance of leaf color, which guaranteed the plants' photosynthesis, and ensured the total biomass and carbohydrate sustainability, further promoting Cd-and-submergence tolerance. The results imply that the negative effects of cadmium on Bermuda grass growth might be offset by submergence. © 2013 Springer-Verlag Berlin Heidelberg.

摘要:
To assess the safety of transgenic rice expressing Cry1Ab protein to vertebrates, the effect of Cry1Ab rice on broad health indicators in blood and various organs of Swiss rats were analyzed. The 30 and 90 day safety studies of Cry1Ab rice on female Swiss rats revealed that Cry1Ab rice had no significant effect on the several elements of blood lymph including hemogram, calcium ion concentration and apoptosis rate of lymphocytes, indicating that Cry1Ab protein could not affect the blood lymph of Swiss rat. Similarly, Cry1Ab rice had no effect on enzyme activities in a variety of organs of Swiss rat. However, Cry1Ab rice did have significant effects on the blood biochemistry indexes including urea, triglyceride (TG), glutamic oxalacetic transaminase (AST) and alkaline phosphatase (ALP) after the rats were fed with Cry1Ab rice for 30 days, but not after 90 days, indicating that Cry1Ab protein may influence blood metabolism for a short duration. Quantitative real-time PCR (qPCR) analysis of the 6 genes encoding enzymes responsible for the major detoxification functions of liver revealed that Cry1Ab rice exerted no influences on the levels of these transcripts in liver of Swiss rat, indicating that significant differences registered in part of the blood biochemical parameters in the 30 day study might result from other untested organs or tissues in response to the stress of exogenous Cry1Ab protein. The results suggest that Cry1Ab protein has no significant long-term (90 day) effects on female Swiss rat.

摘要:
目的探讨膳食多酚[绿原酸、表没食子儿茶素没食子酸酯(EGCG)、槲皮素]对链脲佐菌素(STZ)诱导的糖尿病大鼠血糖、血脂、肝脏中葡萄糖-6-磷酸酶(G-6-pase)和骨胳肌组织中葡萄糖转运体4 (GLUT4)表达的影响。方法单次腹腔注射链脲佐菌素(STZ,35 mg/kg)结合高脂饮食建立糖尿病大鼠模型,将成模大鼠分成5组[糖尿病模型组(DM)、糖尿病模型+二甲双胍组(S)、糖尿病模型+绿原酸组(CA)、糖尿病模型+EGCG组(E)、糖尿病模型+槲皮素组(Q)],另设正常对照组(NC),分别灌喂二甲双胍、绿原酸、EGCG和槲皮素4w后,测定其糖耐量、空腹胰岛素、甘油三酯、胆固醇、G-6-pase和GLUT4 mRNA的表达。结果绿原酸、EGCG、槲皮素均表现出改善STZ诱导的糖尿病大鼠血糖、甘油三酯(TG)和胆固醇(TC)的含量,并能改善糖尿病模型大鼠的胰岛素敏感性,抑制肝脏G-6-pase mRNA的表达,且提高了骨胳肌GLUT4 mRNA的表达,其中以绿原酸效果最佳。仅仅其糖耐量改善弱于槲皮素作用,但均弱于阳性对照组二甲双胍的作用。结论绿原酸、EGCG、槲皮素均能有效改善STZ诱导的SD糖尿病大鼠的糖代射、脂代射、胰岛素敏感生及肝脏G-6-pase mRNA和骨胳肌GLUT4 mRNA的表达,绿原酸的效果最佳。[营养学报,2012,34(6)_:572-575,581]
