摘要:
掌叶半夏凝集素(Pinellia pedatisecta Agglutinin, PPA )基因属于单子叶甘露糖结合凝集素家族,具有抗虫活性。采用反转录聚合酶链式反应(RT-PCR)技术从掌叶半夏幼叶中克隆到1个新的凝集素基因,命名为PPA2,其开放阅读框长408 bp,编码135个氨基酸,有一个甘露糖保守结合域,包含3个甘露糖专一结合位点。分别构建了由35S启动子和韧皮部特异性启动子 AtPP2驱动的植物表达载体,农杆菌介导的叶盘法转化烟草后,经鉴定获得了单拷贝插入高表达量的纯合转基因烟草株系。通过 Western-blot分析,将获得的转基因纯合株系按蛋白表达水平分成高、中、低3类并进行抗蚜试验,结果表明这些转基因烟草均具有明显抗蚜效果,35S-PPA2转基因植株平均抑蚜率极高,而3种蛋白表达类型的AtPP2-PPA2转基因植株普遍低于35S启动子驱动下的转基因植株。PPA2可以作为高效抗蚜候选基因,具有重要的应用价值。

摘要:
bZIP transcription factors play key roles in plant growth, development, and stress signaling. A bZIP gene BnbZIP2 (GenBank accession number: KP642148) was cloned from ramie. BnbZIP2 has a 1416 base pair open reading frame, encoding a 471 amino acid protein containing a characteristic bZIP domain and a leucine zipper. BnbZIP2 shares high sequence similarity with bZIP factors from other plants. The BnbZIP2 protein is localized to both nuclei and cytoplasm. Transcripts of BnbZIP2 were found in various tissues in ramie, with significantly higher levels in female and male flowers. Its expression was induced by drought, high salinity, and abscisic acid treatments. Analysis of the cis-elements in promoters of BnbZIP2 identified cis-acting elements involved in growth, developmental processes, and a variety of stress responses. Transgenic Arabidopsis plants' overexpression of BnbZIP2 exhibited more sensitivity to drought and heavy metal Cd stress during seed germination, whereas more tolerance to high-salinity stress than the wild type during both seed germination and plant development. Thus, BnbZIP2 may act as a positive regulator in plants' response to high-salinity stress and be an important candidate gene for molecular breeding of salt-tolerant plants. © 2016 Mary Ann Liebert, Inc.

摘要:
中熟转基因抗虫杂交棉中棉所70具有高产优质特点,由高产且综合性状优良的抗虫棉SGK 156与品质优异的渐渗系材料901-001为亲本配制。本文对该杂交种F_1及其自交后代F_2、F_(2:3)、F_(2:4)和F_(2:5)共4个分离世代材料的皮棉产量和品质性状进行了详细调查和相关性分析。结果表明,杂种F_1纤维品质表现突出,上半部平均长度平均为33.00 mm,断裂比强度为33.3 cN·tex~(-1)。在分离群体中,各性状均存在大量的超亲变异,衣分与纤维长度及强度为极显著负相关,与马克隆值为极显著正相关,说明棉花产量与品质间的同步改良仍存在困难。此外,从F_(2:5)株系中挑选出11个优异品系,其中4个材料的皮棉产量超对照,上半部平均长度均高于31.80 mm,断裂比强度在31.0 cN·tex~(-1)以上,铃重均大于6.26 g,衣分最高为38.64 %,这表明通过高代自交是可以筛选出具有优异产量及品质性状的品系。本研究为棉花数量性状遗传、基因功能分析、基因聚合育种改良等研究储备了大量的基础材料。

Background: The identification of quantitative trait loci (QTLs) that are stable and consistent across multiple environments and populations plays an essential role in marker-assisted selection (MAS). In the present study, we used 28,861 simple sequence repeat (SSR) markers, which included 12,560 Gossypium raimondii (D genome) sequence-based SSR markers to identify polymorphism between two upland cotton strains 0-153 and sGK9708. A total of 851 polymorphic primers were finally selected and used to genotype 196 recombinant inbred lines (RIL) derived from a cross between 0 and 153 and sGK9708 and used to construct a linkage map. The RIL population was evaluated for fiber quality traits in six locations in China for five years. Stable QTLs identified in this intraspecific cross could be used in future cotton breeding program and with fewer obstacles. Results: The map covered a distance of 4,110 cM, which represents about 93.2 % of the upland cotton genome, and with an average distance of 5.2 cM between adjacent markers. We identified 165 QTLs for fiber quality traits, of which 47 QTLs were determined to be stable across multiple environments. Most of these QTLs aggregated into clusters with two or more traits. A total of 30 QTL clusters were identified which consisted of 103 QTLs. Sixteen clusters in the At sub-genome comprised 44 QTLs, whereas 14 clusters in the Dt sub-genome that included 59 QTLs for fiber quality were identified. Four chromosomes, including chromosome 4 (c4), c7, c14, and c25 were rich in clusters harboring 5, 4, 5, and 6 clusters respectively. A meta-analysis was performed using Biomercator V4.2 to integrate QTLs from 11 environmental datasets on the RIL populations of the above mentioned parents and previous QTL reports. Among the 165 identified QTLs, 90 were identified as common QTLs, whereas the remaining 75 QTLs were determined to be novel QTLs. The broad sense heritability estimates of fiber quality traits were high for fiber length (0.93), fiber strength (0.92), fiber micronaire (0.85), and fiber uniformity (0.80), but low for fiber elongation (0.27). Meta-clusters on c4, c7, c14 and c25 were identified as stable QTL clusters and were considered more valuable in MAS for the improvement of fiber quality of upland cotton.Conclusion: Multiple environmental evaluations of an intraspecific RIL population were conducted to identify stable QTLs. Meta-QTL analyses identified a common chromosomal region that plays an important role in fiber development. Therefore, QTLs identified in the present study are an ideal candidate for MAS in cotton breeding programs to improve fiber quality. © 2016 Jamshed et al.

SET (Su(var), E(z), and Trithorax) domain-containing proteins play an important role in plant development and stress responses through modifying lysine methylation status of histone. Gossypium raimondii may be the putative contributor of the D-subgenome of economical crops allotetraploid G. hirsutum and G. barbadense and therefore can potentially provide resistance genes. In this study, we identified 52 SET domain-containing genes from G. raimondii genome. Based on conserved sequences, these genes are grouped into seven classes and are predicted to catalyze the methylation of different substrates: GrKMT1 for H3K9me, GrKMT2 and GrKMT7 for H3K4me, GrKMT3 for H3K36me, GrKMT6 for H3K27me, but GrRBCMT and GrS-ET for nonhistones substrate-specific methylation. Seven pairs of GrKMT and GrRBCMT homologous genes are found to be duplicated, possibly one originating from tandem duplication and five from a large scale or whole genome duplication event. The gene structure, domain organization and expression patterns analyses suggest that these genes’ functions are diversified. A few of GrKMTs and GrRBCMTs, especially for GrKMT1A;1a, GrKMT3;3 and GrKMT6B;1 were affected by high temperature (HT) stress, demonstrating dramatically changed expression patterns. The characterization of SET domain-containing genes in G. raimondii provides useful clues for further revealing epigenetic regulation under HT and function diversification during evolution.

Cold stress is a major adverse environmental factor that affects plant growth, development, productivity and quality. In the present study, comparative genome-wide transcriptome analysis on two tobacco (Nicotiana tobacum L.) cultivars, cold-tolerant NC567 and cold-sensitive Taiyan8, was performed using RNA-seq technology. After the first assembly, total length of unigenes is from 101,308,644 to 123,781,795 bp, the N50 length is from 1357 to 1475 bp, and 152,688 unigenes in NC567 and 144,160 unigenes in Taiyan8 were identified, respectively. Functional classification of cold-responsive (COR) genes showed that the genes involved in cell wall metabolism, transcription factors, ubiquitin-proteasome system (UPS) and signaling are over-represented, and the COR genes are specifically induced during cold stress in NC567. Pathway analysis revealed the significant enrichment of the COR genes in plant circadian clock. Taken together, the present study suggested the positive roles of the highly induced expression of the COR genes and the conserved mechanism of circadian clock related genes in tobacco response to cold stress, and provided some valuable genes for crop improvement to cope with cold stress. © 2015 Elsevier Inc. Background: Verticillium wilt (VW) caused by Verticillium dahliae (Kleb) is one of the most destructive diseases of cotton. The identification of highly resistant QTLs or genes in the whole cotton genome is quite important for developing a VW-resistant variety and for further molecular design breeding. Results: In the present study, BC1F1, BC1S1, and BC2F1 populations derived from an interspecific backcross between the highly resistant line Hai1 (Gossypium barbadense L.) and the susceptible variety CCRI36 (G. hirsutum L.) as the recurrent parent were constructed. Quantitative trait loci (QTL) related to VW resistance were detected in the whole cotton genome using a high-density simple sequence repeat (SSR) genetic linkage map from the BC1F1 population, with 2292 loci covering 5115.16 centiMorgan (cM) of the cotton (AD) genome, and the data concerning VW resistance that were obtained from four dates of BC2F1 in the artificial disease nursery and one date of BC1S1 and BC2F1 in the field. A total of 48 QTLs for VW resistance were identified, and 37 of these QTLs had positive additive effects, which indicated that the G. barbadense alleles increased resistance to VW and decreased the disease index (DI) by about 2.2-10.7. These QTLs were located on 19 chromosomes, in which 33 in the A subgenome and 15 QTLs in the D subgenome. The 6 QTLs were found to be stable. The 6 QTLs were consistent with those identified previously, and another 42 were new, unreported QTLs, of which 31 QTLs were from G. barbadense. By meta-analysis, 17 QTL hotspot regions were identified and 10 of them were new, unreported hotspot regions. 29 QTLs in this paper were in 12 hotspot regions and were all from G. barbadense. Conclusions: These stable or consensus QTL regions warrant further investigation to better understand the genetics and molecular mechanisms underlying VW resistance. This study provides useful information for further comparative analysis and marker-assisted selection in the breeding of disease-resistant cotton. It may also lay an important foundation for gene cloning and further molecular design breeding for the entire cotton genome. © 2016 The Author(s).

摘要:
Thermosensitive genic male sterile (TGMS) rice line has made great economical contributions in rice production. However, the fertility of TGMS rice line during hybrid seed production is frequently influenced by low temperature, thus leading to its fertility/sterility alteration and hybrid seed production failure. To understand the mechanism of fertility alternation under low temperature inducement, the extracted proteins from young panicles of two TGMS rice lines at the fertility alternation sensitivity stage were analyzed by 2DE. Eighty-three protein spots were found to be significantly changed in abundance, and identified by MALDI-TOF-TOF MS. The identified proteins were involved in 16 metabolic pathways and cellular processes. The young panicles of TGMS rice line Zhu 1S possessed the lower ROS-scavenging, indole-3-acetic acid level, soluble protein, and sugar contents as well as the faster anther wall disintegration than those of TGMS rice line Zhun S. All these major differences might result in that the former is more stable in fertility than the latter. Based on the majority of the 83 identified proteins, together with microstructural, physiological, and biochemical results, a possible fertile alteration mechanism in the young panicles of TGMS rice line under low temperature inducement was proposed. Such a result will help us in breeding TGMS rice lines and production of hybrid seed.

摘要:
To introgress the good fiber quality and yield from Gossypium barbadense into a commercial Upland cotton variety, a high-density simple sequence repeat (SSR) genetic linkage map was developed from a BC1F1 population of Gossypium hirsutum×Gossypium barbadense. The map comprised 2,292 loci and covered 5115.16 centiMorgan (cM) of the cotton AD genome, with an average marker interval of 2.23 cM. Of the marker order for 1,577 common loci on this new map, 90.36% agrees well with the marker order on the D genome sequence genetic map. Compared with five published high-density SSR genetic maps, 53.14% of marker loci were newly discovered in this map. Twenty-six quantitative trait loci (QTLs) for lint percentage (LP) were identified on nine chromosomes. Nine stable or common QTLs could be used for marker-assisted selection. Fifty percent of the QTLs were from G. barbadense and increased LP by 1.07%-2.41%. These results indicated that the map could be used for screening chromosome substitution segments from G. barbadense in the Upland cotton background, identifying QTLs or genes from G. barbadense, and further developing the gene pyramiding effect for improving fiber yield and quality. © 2014 Institute of Botany, Chinese Academy of Sciences.

摘要:
Class I alpha-mannosidases play an important role in co- and post-translational N-glycosylation modification of proteins, and also in glycoprotein glycan hydrolysis. Herein, we investigated a protein named Man-41, from liquid exudate droplets secreted on the surface of developing sclerotia by Sclerotinia sclerotiorum. The protein was identified by MALDI-TOF mass spectrometry to be a alpha-mannosidase. The full-length open reading frame of Man-41 consists of 1581 bp, encoding 526 amino acid residues and containing a putative signal peptide at amino acid residues 1-20, and a conserved sequence at residues 50-521. Man-41 was classified into glycoside hydrolase family 47 (GH47) by clustering analysis. The catalytic residues include Glu(125), Arg(129), Asp(270), Ser(271), Glu(274), Arg(420), Glu(422), Glu(425), Glu(485), Thr(514), and Glu(515), which are conserved in all Class I alpha-1,2-mannosidases. Recombinant Man-41 protein had 26.67 +/- A 2.18 U/mg of alpha-mannosidase activity, about one-half of intracellular mannosidase activity of sclerotia. In conclusion, this is the first time that mannosidase has been identified in an extracellular fluid and Man-41 is also a new member of GH47 with Ca2+-dependent characteristics. This work lays the foundation for further study of the function of Man-41 in sclerotial development.

摘要:
为了提高烟草单倍体技术中的染色体加倍率,以K394 ×云烟87的F1 代花药组培单倍体烟苗为供试材料,研究了不同浓度秋水仙素处理对烟草单倍体苗的成苗率、大田移栽成活率和染色体加倍率的影响;以浸苗法为对照,在植物无根的情况下,研究了秋水仙素对烟草单倍体苗的成苗率、大田移栽成活率和染色体加倍率的影响.结果表明:秋水仙素浓度降低,成苗率和大田移栽成活率会升高而染色体加倍率会降低;无根系烟苗在加倍时染色体加倍率普遍比有根的烟苗低.

摘要:
Degradation of rhamnolipid-solubilized hexadecane and mass hexadecane as a separate phase by Pseudomonas aeruginosa CCTCC AB93066 treated with rhamnolipid biosurfactant was studied for better understanding on the roles of rhamnolipid in hydrocarbon biodegradation. The results of hexadecane solubilization experiment showed that solubility of hexadecane was linearly related to the concentration of rhamnolipid below or above its critical micelle concentration (CMC), and the ability of monorhamnolipid (monoRL) to solubilize hexadecane was stronger at concentration below CMC than above CMC. MonoRL was then used for treating cells in degradation experiment. Results showed that 75μM (1 CMC) monoRL treatment had a small inhibitory effect on cell growth on glucose or mass hexadecane, however 750μM (10 CMC) monoRL treatment accelerated degradation of mass hexadecane by reducing the lag phase of cell growth for 36h; this effect was not caused by initial cell surface hydrophobicity enhancement. No degradation of hexadecane solubilized by 750μM monoRL was observed for the cells treated with or without monoRL, indicating that the pseudo-solubilized hexadecane is not available to cells. It is inferred from the data that the effectiveness of rhamnolipid to accelerate degradation of hydrocarbons by enhancing solubilization of the hydrocarbons may not always be guaranteed, which is of importance for evaluation of rhamnolipid biosurfactant application to hydrocarbon-contaminated sites during remediation. © 2014 Elsevier Ltd.

摘要:
电活性生物膜形成、 胞外电子传递机制以及环境效应研究已成为环境领域关注的热点. 通过在碳毡上富集形成生物膜,采用电化学方法对生物膜的电活性进行了表征,对电活性生物膜直接介导Cu~(2+)的还原转化进行了初探. 利用扫描电子显微镜(SEM)、 能谱分析仪(EDS)、 X射线光电子能谱(XPS)对Cu元素的形貌、 含量以及价态进行分析,结果表明电活性生物膜能够利用乙酸作为电子供体,Cu~(2+)作为电子受体,将Cu~(2+)还原转化成Cu或Cu~+. 同时,通过激光共聚焦显微镜(LSCM)观察Cu~(2+)对微生物的影响,结果显示Cu~(2+)对微生物有明显的毒害作用,所以电活性生物膜介导Cu~(2+)的转化只能在较低的铜离子浓度下进行. 本研究结果将为电活性生物膜应用于环境中铜离子的固定和回收提供理论依据.

摘要:
Cotton is an important fiber plant, and it's attractive to elucidate the molecular mechanism of anther development due to the close relationship between the anther fertility and boll-setting, and also fiber yield. In the present paper, 47.2. million paired-end reads with average length of 82.87. bp from the anthers of TM-1 (. Gossypium hirsutum L.), a genetic standard line, were generated through transcriptome sequencing, and 210,965 unigenes of more than 100. bp were obtained. BLAST, KEGG, COG, and GO analyses showed that the genes were enriched in the processes of transcription, translation, and post-translation as well as hormone signal transduction, the transcription factor families, and cell wall-related genes mainly participating in cell expansion and carbohydrate metabolism. Further analysis identified 11,153 potential SSRs. A suit of 5122 primer pair sequences were designed, and 82 of 300 randomly selected primer pairs produced reproducible amplicons that were polymorphic among 22 cotton accessions from G. hirsutum, Gossypium barbadense and Gossypium arboreum. The UPGMA clustering analysis further confirmed high quality and effectiveness of these novel SSR markers. The present study provided insights into the transcriptome profile of the cotton and established a public information platform for functional genomics and molecular breeding. © 2014 Elsevier B.V.All rights reserved.

摘要:
为进一步了解棉花主要性状的遗传规律和杂种优势,为选育高产、优质、多抗的棉花新品种提供参考依据,试验以封花自交的陆地棉岱字15号,湘棉10号,湘棉15号、和3个自育稳定的新品系为材料,按完全双列杂交配置30个棉花杂交新组合;将6个亲本与组合采用顺序排列,重复3次进行田间小区试验;对其主要农艺、经济性状进行相关性研究。结果表明:株高和果枝数的遗传主要受加性效应控制;单铃重和成铃数遗传变异来源于加性效应和母体效应;籽棉和皮棉的遗传主要受到加性效应控制,非加性效应的作用相应较小;衣分的遗传受加性效应和非加性效应共同控制,但主要受加性效应的影响。

摘要:
A novel thermophilic, Gram-staining positive bacterium, designated DX-2T, was isolated from the anode biofilm of a microbial fuel cell. Cells of the strain were oxidase positive, catalase positive, facultative anaerobic, motile rods. The isolate grew at 30-60°C (optimum 50°C) and pH 5-9 (optimum pH 8-8.5). The pairwise 16S rRNA gene sequence similarities showed that strain DX-2T was most closely related to Bacillus fumarioli LMG 17489T (96.2 %), B. firmus JCM 2512T (96.0 %) and B. foraminis DSM 19613T (95.7 %). Phylogenetic analysis based on 16S rRNA gene sequences showed that strain DX-2T formed a cluster with B. smithii (95.5 %) and B. infernus (94.9 %). The genomic G+C content of DX-2T was 43.7 mol%. The predominant respiratory quinone was MK-7. The polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and unknown phospholipids. The major cellular fatty acid was iso-C16:0. Based on its phenotypic characteristics, chemotaxonomic features, and results of phylogenetic analysis, the strain was identified to represent a distinct novel species in the genus Bacillus, and the name proposed is B. thermophilum sp. nov. The type strain is DX-2T (=CCTCC AB2012194T = KCTC 33128T). © 2014 Springer-Verlag.

普遍且频繁发生的蚜虫危害给农业生产造成严重损失。就抗蚜虫基因工程研究取得的进展进行综述,分析目前克隆的抗蚜基因及其获得的转基因作物,并讨论转基因抗蚜中存在的问题,探讨通过新的毒杀基因克隆、基因定点突变、增加表达调控元件及利用凝集素蛋白作为运输载体等方法来提高抗蚜效果,为今后抗蚜转基因作物育种研究提供参考。

干湿交替条件下不同晶型铁氧化物对水稻土甲烷排放的影响

摘要:
以典型华南水稻土为研究对象,通过室内恒温培养试验,分析了不同水分条件下[干湿交替(DW)和持续淹水(CF)]不同晶型铁氧化物[水铁矿(SF)和赤铁矿(SH)]对水稻土 CH_4平均排放速率及产CH_4潜力的影响.结果表明,与CF相比,DW处理的CH_4平均排放速率平均减少了61%以上;与没有添加铁氧化物的处理相比,铁氧化物抑制了53%的CH_4排放. DW与铁氧化物的耦合抑制了65% 的产CH_4平均排放速率及57% ~93%的产CH_4潜力,抑制效果明显高于DW或铁氧化物的单独作用.其中DW耦合晶型较弱的水铁矿(SF-DW)所产生的抑制效应(94%)明显高于DW耦合晶型较好的赤铁矿(SH-DW, 65%).统计分析结果表明Fe(Ⅲ)浓度与水稻土 CH_4排放速率存在显著负相关性(R2=-0. 98,P < 0. 01).且实验结果表明,DW可以加速Fe(Ⅲ)的循环再生,而晶型较弱的SF易还原性使其Fe(III)的循环再生更易受到DW的影响.上述结果说明,DW可以促进铁氧化物对水稻土CH_4排放及产.CH_4潜力的抑制效应,且干湿交替与晶型较弱的水铁矿的耦合效应比干湿交替与晶型较高的赤铁矿的耦合效应更为显著。

烟草耐冷性的苗期鉴定新方法

摘要:
针对中国早春季节烟草移栽后低温逆境频繁发生,易致幼苗前期生长缓慢、出现早花,并严重引起烟叶产量和品质的波动等问题,笔者提出了一种新的烟草耐冷性的苗期鉴定方法,用于烟草低温敏感性的评价及耐冷烟草资源材料的筛选与鉴定。通过漂浮育苗3?4叶期幼苗假植至花盆、6?8叶期常温驯化2?3天、6℃?8℃持续低温胁迫7天及随后常温恢复生长10天的处理程序,根据幼苗在低温胁迫处理下的形态反应特征和常温恢复生长情况进行综合评价,将耐冷程度分为低温敏感、较耐冷和耐冷3种类型。对包括烤烟、晒烟和野生种的36份烟草资源材料进行苗期耐冷性鉴定,持续低温处理6天对植株幼叶形态的危害程度可以直观地分为4种类型,随后的常温恢复生长过程也能将不同材料的表现简单地分成3种类型,依据这些材料在低温胁迫及随后的常温恢复过程中的形态表现类型,共筛选出5份耐冷、 7份较耐冷的资源材料。
