摘要:
To introgress elite QTL alleles of Gossypium barbadense L.for fiber yield and quality and resistance to Verticillium wilt into G.hirsutum L.,enlarge the genetic base of G.hirsutum,and provide new germplasm resources for the variety development,the cultivars Zhongmiansuo 36,G.hirsutum,and Hai 1,G.barbadense,were used as recurrent and donor parent,respectively,to produce BC1 F1,BC1S1,and BC2F1 populations.QTL analyses of fiber quality,yield-related,Key words: interspecific backcrosss; AB-QTL; fiber quality; yield; Verticillium wilt resistance

特棉S-1雄性不育发生的细胞学基础及可育花粉对温度的反应

摘要:
特棉S-1是一个新发现的高温敏感型雄性不育材料.体外花粉培养试验表明,在15,20,25,30,35,40 ℃处理中,特棉S-1花粉萌发率和花粉管长度均低于对照苏棉16;特棉S-1的花粉萌发率和花粉管长度最高值为37%和450 μm,而对照苏棉16分别为51.8%和682 μm,差异达极显著水平.细胞学观察发现,受到高温的影响,花粉母细胞不能形成和减数分裂发生异常不能发育成正常四分体是特棉S-1雄性不育发生的主要原因.

摘要:
本研究利用以SGK9708为母本,0-153为父本构建的196个陆地棉重组自交系(F_(6:8))构建了包含 186个标记,总长827.84 cM,标记间平均距离4.45 cM,覆盖棉花基因组18.6%的遗传连锁图谱,并对7个环境下的铃重和衣分性状进行QTL定位和上位性互作分析。利用两种分析软件(WinQTLcart2.5和QTLNetwork2.0)共同定位了多个环境下稳定表达的5个主效QTLs(qBW-1-1,qBW-1-2,qLP-2-1,qLP-2-2和qLP-4-2)。利用QTLNetwork2.0分别检测到4对铃重上位性互作QTLs和7对衣分上位性互作QTLs,以背景位点间的互作和加性效应位点与背景位点间的互作为主。除主效QTL外,上位性效应也是陆地棉铃重和衣分性状的重要遗传基础。本研究定位的5个主效QTLs为选择高铃重、高衣分品种的棉花分子标记辅助育种提供了重要依据。

摘要:
The cell culture in a production medium in the dark is capable of producing a large quantity of shikonin and its derivatives, which were completely inhibited when the cell cultures were irradiated with continuous white light and blue light and partially repressed under continuous red light. The expressions of PAL1, 4CL1, and CYP98A6 were induced under continuous red, blue and white light. Transcript levels of HMGR and LDI2 gradually were decreased in the dark and under continuous red light, while no transcriptional product of LDI2 was detected under blue and white light. C4H2 and LePGT1 genes were constantly expressed irrespective of different light conditions. Among the genes studied, the expression of LDI2 was critical for light-regulated shikonin formation. The inducible expression of PAL1, 4CL1, and CYP98A6 as well as the inhibitory transcription of LDI2 mediated by continuous red light irradiation were likely accounted for the reduced accumulation of shikonin.

摘要:
With radical global climate change and global warming, high temperature stress has become one of major factors exerting a major influence on crop production. In the cotton (Gossypium hirsutum L.)-growing areas of China, especially in the Yangtze River valley, unexpected periodic episodes of extreme heat stress usually occur in July and August, the peak time of cotton flowering and boll loading, resulting in lower boll set and lint yield. Breeding programs for screening high temperature-tolerant cotton germplasm and cultivars are urgent in order to stabilize yield in the current and future warmer weather conditions. In the present study, 14 cotton cultivars were quantified for in vitro pollen germination and pollen tube growth in response to temperatures ranging from 10 to 50 °C at 5 °C intervals. Different cotton genotypes varied in their in vitro pollen germination and pollen tube length responses to the different temperatures. Maximum pollen germination and pollen tube length ranged from 25.2% to 56.2% and from 414 to 682 μm, respectively. The average cardinal temperatures (Tmin, Topt, and Tmax) also varied among the 14 cultivars and were 11.8, 27.3, and 42.7 °C for pollen germination and 11.8, 27.8, and 44.1 °C for maximum pollen tube length. Variations in boll retention and boll numbers per plant in field experiments were found for the 14 cotton cultivars and the boll retention and boll retained per plant on 20 August varied considerably in different years according to weather conditions. Boll retention on 20 August was highly correlated with maximum pollen germination (R2 = 0.84) and pollen tube length (R2 = 0.64). A screening method based on principle component analysis of the combination of pollen characteristics in an in vitro experiment and boll retention testing in the field environment was used in the present study and, as a result, the 14 cotton cultivars could be classified as tolerant, moderately tolerant, moderately susceptible and susceptible to high temperature. © 2006 Institute of Botany, Chinese Academy of Sciences.

以带顶芽或不定芽的杜鹃兰假鳞茎切块为外植体进行试管培养,可在短时间内获得大量优质幼苗.经试验筛选出各培养阶段的最适培养基为:(1)原球茎诱导和增殖培养基:MS+6-BA 2.0mg/L(单位下同)+2,4-D0.5+杜鹃兰共生真菌提取物10.0+PVP500.0;(2)芽和根分化及试管苗生长培养基:1/2MS+NAA0.5+IBA0.5+共生真菌提取物10.0+PVP500.0.上述2种培养基均加入3.0%蔗糖和0.8%琼脂,pH5.8.培养温度(25±1)℃,光照12h/d,光照度2000lx.

在对水分胁迫下甘薯形态、生长势、生理生化和产量性状等指标与品种抗旱性进行相关分析的基础上,采用抗旱系数法、聚类分析法、隶属函数法对品种抗旱适应性予以综合评价.综合评价的结果,将15个供试甘薯品种按抗旱水平由弱到强划分为不抗(农大红、漳薯1号、8908-383)、低抗(宿芋1号、9318-58、渝苏303、89-1、95-411-153、西蒙1号)、中抗(92-111-107、渝薯34、南薯88、92-103-30)和高抗(Y3、潮薯1号)四级.上述几种关于甘薯品种抗旱适应性综合评价的方法具有较好的一致性.
