5例复发型吉兰-巴雷综合征临床特点分析

摘要:
目的研究和分析复发型吉兰-巴雷综合征(GBS)的临床特点。方法收集72例GBS患者的临床资料,排除Miller-Fisher综合征、波动型GBS以及急性发作型CIDP病例,将其余病例分为复发型吉兰-巴雷综合征组(R-GBS组)5例和非复发型吉兰-巴雷综合征组(NR-GBS组)56例,通过统计学分析比较其两组间在年龄、性别、是否伴有自身免疫性疾病、前驱感染、是否伴有脑神经受累、病情高峰时神经功能受损情况、临床发作类型方面有无差别;并将5例R-GBS组患者的临床特点进行进一步比较及归纳。结果R-GBS组首次发病年龄为31.80± 13. 37岁,NR-GBS组为44. 84 ± 13.72岁,P值<0.05,具有统计学意义;而在性别、是否伴有自身免疫性疾病、前驱感染、是否伴有脑神经受累、病情高峰时神经功能受损情况、临床发作类型方面无统计学差异。5例R-GBS患者共有6次复发,复发后病情达高峰时Hughes评分4次升高,2次无变化;病情达高峰时前一次Hughes评分为2. 50 ± 1.05分,复发时为3. 83 ±0.75分,P <0.05,两次比较有统计学差异;与前一次发作相比,临床症状类型4次无变化,2次由感觉-运动型变为纯运动型;前驱感染3次相同,3次不同。结论与NR-GBS组相比,R-GBS组患者年龄较小,而在其他主要临床症状方面无显著差异;R-GBS患者症状呈现加重趋势,前驱感染类型与临床发作类型无相关性,临床医生应该注意到这些特点,以便更好的指导治疗。

摘要:
对不同耐性烟草苗期叶片的光合特性在低温胁迫及其随后常温恢复生长条件下的变化进行了详细研究。结果表明,4℃~6℃的低温胁迫几乎完全抑制较耐冷烟草品种K326和敏感品种云烟97幼苗的生长,但在恢复至常温条件时K326的生长速度显著高于云烟97。低温胁迫降低烟草幼苗叶片的光合作用,而且胁迫时间越长,抑制效应越明显,处理6d后叶片气体交换参数净光合速率(Pn)、气孔导度(Gs) 、胞间CO_2浓度(Ci) 、蒸腾速率(Tr)、水分利用效率(WUE)、气孔限制值(Ls)以及叶绿素荧光参数最大光化学效率(Fv/Fm)、PS II量子产量或光能捕获的效率(Φ_(PSII))、光化学猝灭系数(qP)、表观电子传导率(ETR)和非光化学猝灭系数(NPQ)均与处理前的差异达到了显著水平。恢复至常温生长条件时,幼苗叶片光合作用迅速得以增强,K326提高的幅度比云烟97大;虽然恢复至第9d时这两个品种的光合作用还没有回复到处理前的水平,但较耐冷品种K326的光合作用参数与处理前的差异较小,而低温敏感品种云烟97与处理前仍存在显著性差异。

摘要:
An Fe(III) oxide-reducing bacterium designated as SgZ-3T, which could couple glucose oxidation to Fe(III) oxide reduction for energy conservation, was isolated from an electrochemical biofilm. The isolate was Gram-negative, catalase-positive and oxidase-negative, and facultative anaerobic. NaCl was not required for growth, but NaCl concentrations up to 5 % (w/v) were tolerated. Growth occurred in TSB (tryptic soy broth) at 20-40 C (optimum 30 C) and at pH 6.0-7.5 (optimum 7.0). Phototrophic growth could not be demonstrated. No vesicular photosynthetic membrane was observed. Bacteriochlorophyll α and carotenoids were absent. Biotin and thiamine were required as growth factors for the isolate. Phylogenetic analysis of the 16S rRNA gene sequence placed strain SgZ-3T within the family Rhodobacteraceae and affiliated with an phototrophic genera Rhodobacter. The G+C content of the genomic DNA was 68.6 mol%. Strain SgZ-3T contained Q-10 as the predominant quinone. The major cellular fatty acids were C 18:1 ω6c and/or C18:1 ω7c (66.9 %) and C 16:0 (9.5 %). Based on its phenotypic and phylogenetic properties, chemotaxonomic analysis and the results of physiological and biochemical tests, strain SgZ-3T (=KACC 16603T = CCTCC AB2012026T) was designated as the type strain of a new genus and a novel species of the family Rhodobacteraceae, for which the name Sinorhodobacter ferrireducens Sp. ‘湘农大棉1号’的SSR数字指纹图谱构建及纯度鉴定

摘要:
为了快速而准确地鉴定杂交棉种子的纯度和真伪,根据亲本材料来源和被推荐使用的SSR核心引物,挑选了33对引物对‘湘农大棉1号’及其亲本进行多态性筛选。结果表明,其中8对SSR引物在父母本及其杂交种扩增出清晰、稳定的多态性条带,且这些位点在F1中均表现为共显性标记。获得的共显性标记都能用于该杂交种的纯度鉴定,可以用来区别其中混杂的母本、父本及其他种子。将这8对引物在3个材料中扩增得到的01(二进制)数据转换成十进制数据,构建了‘湘农大棉1号’及其亲本的数字指纹图谱。采用多对引物构建的杂交棉数字指纹图谱,能为杂交种的真伪鉴定、纯度检测及亲本提纯等工作提供更加准确的技术指导。

摘要:
Abscisic acid (ABA) regulates many aspects of plant development, including seed dormancy and germination, root growth and stomatal closure. Plant SKP1 proteins are subunits of the SCF complex E3 ligases, which regulate several phytohormone signalling pathways through protein degradation. However, little is known about SKP1 proteins participating in ABA signalling. Here, we report that the overexpression of Triticum aestivum SKP1-like 1 (TSK1) in Arabidopsis thaliana (Arabidopsis) resulted in delayed seed germination and hypersensitivity to ABA. The opening of stomatal guard cells and the transcription of several ABA-responsive genes were affected in transgenic plants. In contrast, Arabidopsis skp1-like 1 (ask1)/ask1 ASK2/ask2 seedlings exhibited reduced ABA sensitivity. Furthermore, the transcription of ASK1 and ASK2 was down-regulated in abi1-1 and abi5-1 mutants compared with that in wild type. ASK1 or ASK2 overexpression could rescue or partially rescue the ABA insensitivity of abi5-1 mutants, respectively. Our work demonstrates that SKP1 is involved in ABA signalling and that SKP1-like genes may positively regulate ABA signalling by SCF-mediated protein degradation.

摘要:
针对混合交通特征表达和分类识别的问题,提出了基于边缘偏心率向量的混合交通视频检测算法。将利用上下文比对获取的边缘信息与图像重心相结合构建混合交通的边缘偏心率向量,对混合交通前景进行了特征表达。再结合极限学习机建立了快速学习机制,实现了快速分类识别,克服了采用支持向量机训练难以达到实时检测的问题。试验结果表明:本文算法中各个类别的混合交通边缘偏心率特征区分明显,识别准确率可达93%以上,且处理速度快,能够满足实时检测的需求。

棉花花药中高温胁迫响应基因的克隆与分析

摘要:
以耐高温和敏感型陆地棉品系HLY11与TS18为材料,采用抑制差减杂交技术(SSH)构建高温胁迫下花药的正向和反向差减文库。共获得了2880个阳性克隆,随机挑取400个克隆进行PCR验证,经测序和序列拼接分析,共获得342条有效EST序列,聚类分析得到278条非冗余的EST(unigenes)。BlastN分析发现,有211条unigenes能在GenBank数据库中找到同源序列,其中164条与已知功能蛋白有较高同源性,47条与未知功能或假定蛋白有较高相似性,其余67条没有同源匹配。KEGG分析将152条unigenes定位到46条代谢途径中,而P<0.05的代谢途径有14条,包含了31个unigenes。初步分析发现,棉花花药中响应高温逆境胁迫的基因涉及热激蛋白、抗氧化酶类、碳水化合物代谢、转录因子和跨膜转运蛋白等过程,其中淀粉和糖代谢、谷胱甘肽代谢、植物病原生物相互作用和氧化磷酸化作用等可能与棉花花药高温逆境耐性的相关性较大。

本研究利用以SGK9708为母本,0-153为父本构建的196个陆地棉重组自交系(F_(6:8))构建了包含 186个标记,总长827.84 cM,标记间平均距离4.45 cM,覆盖棉花基因组18.6%的遗传连锁图谱,并对7个环境下的铃重和衣分性状进行QTL定位和上位性互作分析。利用两种分析软件(WinQTLcart2.5和QTLNetwork2.0)共同定位了多个环境下稳定表达的5个主效QTLs(qBW-1-1,qBW-1-2,qLP-2-1,qLP-2-2和qLP-4-2)。利用QTLNetwork2.0分别检测到4对铃重上位性互作QTLs和7对衣分上位性互作QTLs,以背景位点间的互作和加性效应位点与背景位点间的互作为主。除主效QTL外,上位性效应也是陆地棉铃重和衣分性状的重要遗传基础。本研究定位的5个主效QTLs为选择高铃重、高衣分品种的棉花分子标记辅助育种提供了重要依据。

摘要:
本研究利用中棉所36和海1配制杂交组合,并用中棉所36为轮回亲本构建高代回交群体.利用3 223对SSR引物筛选亲本和F1,筛选到294对含有海1显性带的引物,占筛选引物总数的9.12%.最终对其中的267对引物进行了群体扩增,获得277个SSR标记差异位点.其中,217个标记位点连锁,构建44个连锁群,平均每个连锁群包含4.93个标记位点.图谱覆盖1 908.67 cM,占棉花基因组的42.89%,平均每个连锁群覆盖43.38 cM,标记间平均相距8.80 cM.利用陆海杂种BC_1F_1、BC_2F_1和BC_1S_1的表型数据,通过复合区间作图,共检测到9个QTL,解释表型变异6.90%~19.17%.其中3个增效基因来自海岛棉亲本海1,6个增效基因来自陆地棉亲本中棉所36.此外,控制衣分的2个QTL在3个世代都能够检测到,分别解释6.90%和19.17%的表型变异,效应稳定,为进一步分子标记辅助选择奠定了基础.

摘要:
This study demonstrated the feasibility to produce Bacillus thuringiensis subsp. kurstaki (Btk) based biopesticides using wastewater sludge as raw materials under solid-state fermentation (SSF). More than 1010CFU/g viable cells of Btk were obtained using sludge or its mixture with agricultural wastes. This study well considered the effect of heavy metals on Btk growth and their changes of chemical speciation caused by SSF. The IC50 (concentration causing 50% inhibition in total cell biomass) for Pb(II), Cu(II), Cd(II) and Cr(III) on Btk were determined to be 227, 82, 15 and 263mg/L, respectively. Exposure to 150mg/L of Cu(II) severely reduced the amount and size of toxin crystals, which decreased the endotoxin synthesis and entomotoxicity potency of Btk cells. Using Tessier's sequential extraction procedure, the exchangeable heavy metals in sludge were shown to be transformed into residual fractions after SSF, and thus significantly reduced their bioavailability and potential environmental risks. © 2010 Elsevier Ltd.

摘要:
对不同耐高温棉花品系叶片的光合特性及其在盛花期高温胁迫下的变化进行了比较研究。结果表明,与敏感品系XYM 68和TS 18相比,耐高温品系HLY从盛蕾期到结铃盛期的净光合速率、水分利用率、最大净光合速率和表观量子率显著高于其它2个品系,同时具有较低的光补偿点和较高的光饱和点,而在气孔导度、胞间CO_2浓度、蒸腾速率和气孔限制值上没有显著差异。盛花期高温胁迫降低了净光合速率、气孔导度和水分利用效率,增强了蒸腾速率和气孔限制值,而对胞间CO_2浓度没有明显影响;与处理前相比,敏感品系XYM 68和TS 18的净光合速率下降显著,气孔导度的降低幅度显著大于HLY,而蒸腾速率增加的强度却显著低于HLY。高温胁迫下,光系统II的最大光化学效率、量子产量和光化学猝灭系数下降,而非光化学猝灭系数增加;3个不同耐高温品系中,TS 18的最大光化学效率减少显著,XYM 68和TS 18的量子产量、光化学猝灭系数降幅显著高于HLY,而非光化学猝灭系数的增加量却显著低于HLY。

Phyton (Horn, Austria) 51 (1): 103-113, with 3 figures. The involvement of calcium homeostasis and calmodulin in shikonin accumulation in Onosma paniculata BUR. & FRANCH cell cultures was studied by using pharmacological method. The results showed that the addition of excess Ca2+ and chelator, EGTA, to M-9 production medium resulted in a reduction of shikonin formation, and shikonin production was completely suppressed by EGTA at 6 mM. Ca2+ channel blockers, verapamil and nifedipine, and Ca2+ ionophore A23187, exhibited a partial inhibition of shikonin accumulation. Furthermore, the addition of CaM antagonist, W-7 at 80 mu M, completely inhibited shikonin accumulation. In contrast to W-7, the relatively inactive analog W-5 at equivalent concentration did not apparently influence shikonin production. These results indicated that calcium homeostasis and CaM-mediated Ca2+ signal sensing might be involved in shikonin accumulation in O. paniculata cell cultures.

摘要:
为明确烤烟胞质雄性不育系在杂种优势利用上应用潜力,以3个烤烟胞质雄性不育系和4个国内推广烤烟品种为材料,配成12个杂种,利用简单加性—显性遗传模型对烤烟产量、产值、均价、上等烟比例、上中等烟比例和级指经济性状进行了遗传分析。结果表明,6个经济性状均有较高的广义遗传率和较低的狭义遗传率,它们的表型变异主要决定于遗传变异;6个经济性状的主要遗传效应为显性主效应和显性×环境互作效应,显性效应明显高于加性效应;在经济性状上显性效应大的亲本,其杂种F1中也同样表现出较高的显性效应,因而具有较大的杂种优势利用潜力。遗传分析显示了新选育的三个烤烟雄性不育系,MsYZ203-6、MsYZ216-1和MsYZ206-9在经济性状的杂种优势利用上具有较大的应用价值。

采用刚果红羧甲基纤维素钠培养基法、滤纸条降解法和胞外酶活性测定法从长白山原始森林的腐烂枯木和落叶腐质土中筛选一株高产纤维素酶的真菌,结合菌落形态学和分子生物学方法鉴定该菌株为产黄青霉,命名为CBS-1.滤纸崩解实验说明在24 h该菌对滤纸就具有明显的崩解能力,6 d后滤纸的失重率达到24.1%;以稻草粉为底物发酵120 h后,其胞外纤维素酶活性达到33.6 mkat/L,其活性高于已经报道的菌株,是一株具有研究开发潜力的纤维素酶生产菌株.

Purpose: The evaluation of the electron transfer capacities (ETC) of DOM is important to understand their roles in microbial activity, pollution degradation, and metal mobility. Those currently used methods to quantify ETC, such as Zn and Fe3+ assays, are normally time consuming and usually require experience and skills to achieve reproducible results. The aim of this paper is to develop a rapid and simple approach to accurately and directly quantify the ETC of DOM. Materials and methods: DOM was extracted from sewage sludge compost. Cycle voltammetry (CV) was used to investigate the redox behavior of DOM derived from sludge compost. Chronoamperometry (CA) was employed to study the electron-accepting capacities (EAC) and electron-donating capacities (EDC) by applying fixed positive or negative potentials to a working electrode in a conventional three-electrode cell. For comparison, the EAC and EDC of DOM were also determined by chemical methods using Zn as the reductant and Fe3+ as the oxidant. The reversible electron transfer of DOM was studied electrochemically by CA with a multi-potential step technique. Results and discussion: The CV of sludge DOM displayed that a couple of quasi-reversible redox peaks with a formal potential of -0.866 V (vs. Ag/AgCl), demonstrating that the DOM was capable of reversibly transferring electrons. The value of EAC was determined by CA to be 361.6 μmole- (g C)-1 at a potential of -0.6 V (vs. Ag/AgCl), and the value of initial EDC was 5.0 μmole- (g C)-1 at a potential of +0.4 V (vs. Ag/AgCl). Both the values of EAC and EDC depended on the applied potentials on the working electrode. The results determined by the proposed method were comparable with those by using chemical methods. Conclusions: A rapid electrochemical approach was employed to investigate the EAC and EDC of DOM extracted from sewage sludge compost. The acquired values of EAC and EDC were comparable with those measured by chemical methods using zinc as the reductant and ferric iron as the oxidant. The determination could be completed in tens of seconds, which was faster and more direct than conventional chemical methods. © 2011 Springer-Verlag.

Anthocyanidin synthase (ANS) is a key enzyme of the proanthocyanidin biosynthetic pathway in Arabidopsis. In this study, we report cloning and characterization of an ANS homolog, BjANS, from Brassica juncea. The BjANS cDNA sequence is 1,377 bp long and has a 1,077-bp open reading frame encoding a deduced polypeptide of 358 amino acids with a predicted molecular weight of 40,862 Da and an estimated isoelectric point of 5.18. The putative BjANS protein shows 91-99% amino acid sequence identity with Arabidopsis thaliana, Matthiola incana, B. rapa and B. oleracea ANS proteins. The expression of BjANS was analyzed in yellow- and black-seeded B. juncea accessions to determine its physiological function. RT-PCR analysis showed that BjANS expressed in the seed coats of the black-seeded lines and in embryos of the all lines, but not in the yellow-seeded seed coats. Proanthocyanidins were also detected by histochemical analysis in the seed coat of black-seeded accessions and in embryos of all the lines, but not the yellow-seeded seed coat. Both transcriptional and histochemical analyses confirmed that BjANS was involved in the biosynthesis of proanthocyanidins and in seed coat color formation of B. juncea. Lack of BjANS expression blocked biosynthesis of proanthocyanidins in the yellow seed coat, and therefore seeds appeared yellow because of transparent testa. © 2010 Springer Science+Business Media B.V.

The present study describes a rapid and sensitive HPLC method for the quantification of huperzine A (HupA) in Huperzia crispata (Huperziaceae). The sample extraction and preparation involved a simple, time-saving, single-solvent extraction, with each sample being analyzed within 12 min. The mobile phase was ammonium acetate (0.1 M, pH 6.0)-methanol (64 + 36, v/v) at a flow rate of 1.0 mL/min. Detection was at 308 nm. The calibration curve was linear from 0.049 to 7.84 μg (R<sup>2</sup> = 0.9997), with intraday and interday precision RSD of less than 2%. The extraction recovery rate was over 98.49%. Quantification of HupA was performed using this modified method, and the content of HupA was 1.86 times higher in the whole plant of H. crispata (218.17 ±1.55 μg/g) than in that of H. serrata (117.03 ±2.97 μg/g). In the whole plant of H. crispata, HupA mainly accumulated in the actively growing shoot tips, the apical bud, and the 10 youngest leaves, reaching 455.23 ±2.97 μg/g. The content of HupA in the samples from sunshine-sheltered sites was 3.45 times higher than in that from sunshine-abundant sites. The satisfactory results indicate that this modified method can be applied in the quality control of large-scale Huperziaceae plant extracts and that changes should be made in the cultivation of H. crispata so as to maximize the production of HupA.
