摘要:
本试验旨在研究桦木酸(BA)对地塞米松(Dex)诱导氧化应激小鼠的保护作用和机制。将40只健康雄性昆明小鼠随机分为5组,即对照(NC)组、Dex组、0.25 mg / kg BA组、 0.50 mg / kg BA组、1.00 mg / kg BA组。NC组和Dex组小鼠灌服1 %的可溶性淀粉溶液,其余各组按不同剂量的BA灌服,连续14 d后,除对照组注射生理盐水外,其余4组均腹腔注射Dex(25 mg / kg BW)诱导氧化应激模型。检测各组小鼠肝脏、脾脏和胸腺总抗氧化能力(T-AOC) 、抑制羟自由基能力和过氧化物酶(POD)的活性,反转录(RT) -PCR检测脾脏和胸腺丝裂原活化蛋白激酶(MAPK)信号通路中凋亡信号调节激酶1(ASK1) 、c-Jun氨基末端激酶(JNK)和P38基因的表达量,蛋白质免疫印迹(Western Blot)法检测脾脏MAPK信号通路中ASK1、JNK和P38蛋白的表达量。结果表明:1)与NC组相比,Dex组的肝脏T-AOC、抑制羟自由基能力,脾脏POD活性,胸腺T-AOC、抑制羟自由基能力均极显著下降(P<0.01);与Dex组相比,0.50和1.00 mg / kg BA组肝脏T-AOC、抑制羟自由基能力以及POD活性均显著或极显著的升高(P < 0.05或P<0.01),0.50 mg / kg BA组脾脏T-AOC、0.50和1.00 mg / kg BA组脾脏抑制羟自由基能力、0.25和1.00 mg / kg BA组脾脏POD活性均显著或极显著的升高(P<0.05或P<0.01),0.50和1.00 mg / kg BA组胸腺T-AOC、抑制羟自由基能力和POD的活性均显著或极显著升高(P< 0.05或P<0.01) 。2)与NC组相比,Dex组脾脏和胸腺ASK1、JNK和P38 mRNA表达量均极显著升高(P < 0.01);与Dex组相比,0.50和1.00 mg / kg BA组脾脏和胸腺ASK1、JNK和P38 mRNA表达量均显著或极显著下降(P<0.05或P<0.01) 。3)与NC组相比,Dex组脾脏JNK和P38蛋白的表达量极显著升高(P<0.01);与Dex组相比,0.50 mg / kg BA组脾脏ASK1蛋白的表达量显著降低(P<0.05),0.25、0.50和1.00 mg / kg BA组脾脏JNK和P38蛋白的表达量显著或极显著降低(P<0.05或P<0.01) 。由此可见,BA预处理后,增强了Dex应激小鼠肝脏、脾脏和胸腺的T-AOC、抑制羟自由基能力和POD活性,降低了Dex应激小鼠脾脏和胸腺MAPK信号通路中ASK1、JNK和P38 mRNA表达量以及脾脏ASK1、JNK和P38蛋白的表达量。BA对Dex造成的氧化损伤具有预防性的保护作用,并且这种保护作用与JNK-P38 MAPK信号通路相关。

摘要:
为了探讨桦木酸对大鼠胃溃疡的保护作用及可能机制,采用无水乙醇灌胃法建立大鼠胃黏膜损伤模型,检测桦木酸对大鼠胃黏膜组织中丙二醛(MDA)含量、超氧化物歧化酶(SOD)与谷胱甘肽过氧化物酶(GSH-Px)活性的影响。结果表明,桦木酸能降低大鼠胃黏膜组织中MDA含量,提高SOD和GSH-Px活性。说明桦木酸对大鼠胃黏膜损伤的保护作用与增强胃黏膜抗氧化能力、抑制自由基生成密切相关。

摘要:
研究桦木酸(betulinic acid,BA)对小鼠细胞因子的影响.取健康昆明系小鼠28只,随机分为4组,即空白对照组,BA低、中、高剂量组(0.25 mg/kg、0.5 mg/kg、1 mg/kg),灌胃给药,1次/d,连续14 d.检测BA对小鼠血清白细胞介素-2(IL-2)、IL-6和IL-10以及对腹腔巨噬细胞肿瘤坏死因子(TNF-α)水平的影响.结果表明,BA可降低小鼠血清IL-2和IL-6的分泌量,提高腹腔巨噬细胞TNF-α的分泌量.说明BA在治疗机体肿瘤、感染和炎症的过程中发挥着重要作用.

摘要:
Obesity, which is associated with dietary habits, has become a global social problem and causes many metabolic diseases. In China, both percentages of adult obesity and overweight are far lower compared to western countries. It was designed to increase the two levels of daily intake in human, namely 3.8% and 6.5%, which are recommendatory intake (25 g/d) and Chinese citizens' practical intake (41.4 g/d), respectively. The mice were respectively fed with feeds added with soybean oil, lard or the oil blended by both for 12 weeks. In the mice fed with diet containing 3.8% of the three oils or 6.5% blended oil, their body weight, body fat rate, cross-sectional area of adipocytes, adipogenesis and lipogenesis in adipose were decreased, whereas hydrolysis of triglyserides in adipose was increased. This study demonstrated that the oil mixture containing lard and soybean oil had a remarkable anti-obesity effect. It suggests that the traditional Chinese dietary habits using oils blended with lard and soybean oil, might be one of the factors of lower percentages of overweight and obesity in China, and that the increasing of dietary oil intake and the changing of its component resulted in the increasing of obesity rate in China over the past decades.

摘要:
IRW (Ile-Arg-Trp) is an angiotensin converting enzyme (ACE) inhibitory peptide identified from egg protein ovotransferrin. Given its role also as an antioxidant, the aims of this work were to investigate whether IRW exerts its protective effect against oxidative stress by inhibiting intracellular ROS accumulation and blocking ROS activated mitochondria-mediated cell apoptosis pathway. Adding 200 μM H2O2 to cells led to decreased cell viability while increased (ROS) formation. However, IRW significantly inhibited H2O2-induced cytotoxicity and ROS production. Flow cytometry was used to assess the cell apoptosis and the mitochondrial membrane potential. Pretreatment with IRW decreased cell apoptosis while elevated the mitochondrial membrane potential compared to H2O2 treated cells. Furthermore, IRW reduced the levels of caspase-9 and caspase-3 and thus improved cell survival. The results demonstrate that IRW can significantly protect Caco-2 cells against oxidative stress by inhibiting intracellular ROS accumulation and blocking the ROS activated mitochondria-mediated cell apoptosis pathway. © 2017 Elsevier Ltd

摘要:
目的:探讨桦木酸体外拮抗T-2毒素、减少T-2毒素对细胞的氧化应激作用.方法:选用人肝脏细胞L02作为研究对象,将含0.2、2.0、20.0 μmol/L的桦木酸和5 nmol/L T-2毒素共同暴露在L02中12h,后测定细胞中SOD、MDA、GSH、GSH-Px、CAT、ALT和AST水平以及细胞培养上清液中的LDH活性,检测细胞氧化损伤情况.结果:随着浓度的提高,桦木酸能剂量依赖地改善由T-2毒素引起的氧化损伤.结论:桦木酸能拮抗T-2毒素,从而改善由T-2毒素引起的氧化损伤.

摘要:
研究桦木酸对胃黏膜的保护作用。采用无水乙醇灌胃法建立大鼠胃黏膜损伤模型,以溃疡指数和溃疡抑制率表示大鼠胃黏膜损伤程度。结果表明,桦木酸能显著降低胃黏膜损伤模型的溃疡指数,提高其溃疡抑制率和胃内容物p H值(P〈0.01)。说明桦木酸对大鼠乙醇胃黏膜损伤具有保护作用,该作用可能与其减少胃酸分泌以及增强组织自身生长和修复能力等有关。

本试验旨在研究桦木酸(BA)对地塞米松(Dex)诱导氧化应激小鼠血清指标的影响。将40只健康雄性昆明小鼠随机分为5组, 即对照(NC)组、Dex组、0.25 mg / kg BA组、 0.50 mg / kg BA组、1.00 mg / kg BA组,每组8只。NC组和Dex组灌服1%的可溶性淀粉,其余各组灌服混悬于1%可溶性淀粉溶液中不同剂量的BA,连续灌服14 d后,除NC组注射生理盐水外,其余4组均腹腔注射25 mg / kg Dex诱导氧化应激模型。15 h后,眼眶采血,收集血清。检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和碱性磷酸酶(ALP)活性,总蛋白(TP)、白蛋白(ALB)、总胆固醇(TC)、甘油三酯(TG)、总胆红素(T–Bil)、钙离子(Ca~(2+))、细胞色素C(CytC)含量,总抗氧化能力(T–AOC)、抑制羟自由基能力、超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)和丙二醛(MDA)含量。结果显示:BA预处理后,能够显著或极显著降低Dex诱导氧化应激小鼠血清ALP活性及TG、T–Bil、CytC含量(P<0.05或P<0.01),且呈量效关系;低剂量BA(0.25、0.50 mg / kg)显著或极显著降低了血清MDA含量(P<0.01或P<0.05),但高剂量BA(1.00 mg / kg)反而极显著升高了其含量(P<0.01);能够显著或极显著升高血清Ca2+ 和GSH含量、SOD的活性、T–AOC和抑制羟自由基能力(P<0.05或P<0.01),并呈量效关系;降低了血清ALT和AST活性,但影响不显著(P>0.05),对血清TP和ALB含量无显著影响(P>0.05)。由此可见,BA能够改善Dex引起的小鼠血清指标的变化,对Dex诱导的氧化损伤有预防性的保护作用。

摘要:
The study was aimed to investigate the effect of Pb toxicity on mouse Leydig cells and its molecular mechanism. The TM3 cells were cultured in vitro and exposed to Pb at different concentrations for 24 h. The effects of Pb on cell proliferation and apoptosis were analyzed with MTT and Annexin V-FITC/PI via flow cytometry, respectively. Expression levels of Fas, Fas-L and caspase-8 in TM3 cells were determined by western blot. As well as the inhibitory effect of the caspase-8 inhibitor Z-IETD-FMK on cell apoptosis. We found that Pb treatment significantly decreased the cellar viability (P < 0.05), increased the apoptosis (P < 0.01) and the Fas, FasL, and caspase-8 expression levels in Pb-treated cells as compared to the control cells (P < 0.05 or P < 0.01). Furthermore, the caspase-8 inhibitor effectively block the Pb-induced cell apoptosis. Taken together, our data suggest that Pb-induced TM3 cell toxic effect may involve in the Fas/FasL death receptor signaling pathway. © 2017

研究桦木酸(betulinic acid,BA)对淋巴细胞凋亡的影响。采用体外培养的方法,以地塞米松(Dexamethasone,Dex)诱发胸腺淋巴细胞凋亡为模型,实验分为空白对照组、Dex组和BA(5μg/m L)组,用荧光显微镜进行细胞形态学观察,通过琼脂糖凝胶电泳观察细胞凋亡过程中DNA的裂解,用流式细胞仪分析并测定细胞凋亡率。结果表明,BA可降低Dex诱导的淋巴细胞凋亡。提示BA对Dex诱导的淋巴细胞凋亡具有干预作用,在一定程度上起到预防性的保护作用。

摘要:
T-2 toxin is a highly toxic mycotoxin produced by various Fusarium species, mainly, Fusarium sporotrichoides, and has been reported to have toxic effects on reproductive system of adult male animals. This study investigated the dose-dependent cytotoxicity of T-2 toxin on reproductive cells using TM3 Leydig cells. Specifically, the cytotoxic effect of T-2 toxin was assessed by measuring cell viability; lactate dehydrogenase (LDH); malondialdehyde (MDA); antioxidant activity by measuring superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX), and DNA damage; and cell apoptosis. Results showed that T-2 toxin is highly cytotoxic on TM3 Leydig cells. However, Trolox-treated TM3 Leydig cells showed significantly reduced oxidative damage, DNA damage, and apoptosis induced by T-2 toxin. This study proves that T-2 toxin can damage the testes and thus affects the reproductive capacity of animals and humans. Furthermore, oxidative stress plays an important role in the cytotoxic effect of T-2 toxin. © 2015 Published by Elsevier Inc.

Background: Dexamethasone (Dex), a synthetic glucocorticoid, is strictly controlled for use due to its serious side effects, including immune suppression. Betulinic acid (BA), an antioxidant prepared from the white birch, exhibits immunomodulation properties. To assess the implications and investigate the mechanisms of BA-elicited immunomodulation, we hypothesized that Dex induced thymocyte apoptosis via oxidative stress could be lessened by BA. Methods: Mice were given oral doses of BA (0.25, 0.5, and 1.0 mg/kg) daily for 14 days, and induced oxidative stress by giving a single dose of Dex intraperitoneal at the dosage of 25 mg/kg body weight 8 h after the last administration of BA. Results: Dex administration alone significantly decreased antioxidant enzyme activities, while significantly increased reactive oxygen species (ROS) production, lipid peroxidation, mitochondrial dysfunctions, caspase-3 activation and cellular apoptosis. However, pretreatment with BA dose dependently mitigated Dex-induced oxidative damage after 14 days of feeding. In addition to ROS scavenging activity in Dex-induced thymocyte, BA administration decreased lipid peroxidation, up regulated antioxidant enzymes, restored mitochondrial function, increased Bcl-2 expression but reduced Bax expression, inhibited caspase-3 activation, and improved cell survival. Conclusions: These findings reveal a protective capability of BA against Dex-induced cell death by reducing oxidative stress via mitochondrial mediated signal pathway which could be the potential mechanism underlying BA-elicited immunomodulation. 摘要:
目的:观察γ-谷维素对过氧化氢诱导人肝细胞(L02)氧化损伤的增殖和保护作用的影响。方法:建立过氧化氢氧化损伤模型,采用不同浓度的γ-谷维素对L02细胞进行处理,检测L02细胞增殖率、丙二醛和微量谷胱甘肽含量、超氧化物歧化酶和过氧化氢酶活力及活性氧水平的变化。结果:γ-谷维素浓度在0.1~0.4 mmol/L对H_2O_2引起的细胞损伤具有修复作用,同时降低丙二醛含量和活性氧水平,提高超氧化物歧化酶和过氧化氢活性以及谷胱甘肽含量,且呈量效关系。结论:L02细胞经0.1~0.4 mmol/L γ-谷维素预处理后,可以缓解H_2O_2造成的氧化损伤,维持正常的生理形态。

桦木酸对Dex致小鼠氧化损伤的保护作用

摘要:
为了研究桦木酸(betulinic acid,BA)对地塞米松(dexamethasone,Dex)致氧化损伤的保护作用。将35只健康雄性KM小鼠随机分为5组,即对照组、Dex组、BA低、中、高剂量(0.25、0.5、1mg/kg)+Dex组。对照组和Dex组灌服1%的可溶性淀粉,其余各组灌服混悬于可溶性淀粉中不同剂量的BA,1次/d,连续14d后,除对照组注射生理盐水外,其余4组均腹腔注射地塞米松(25 mg/kg)诱导氧化应激模型。15h后,处死小鼠,收集淋巴器官和肝脏。检测小鼠淋巴细胞的活性以及活性氧(ROS)水平,肝脏、脾脏和胸腺的超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性以及丙二醛(MDA)含量。结果显示,BA预处理后,能显著降低Dex诱导氧化损伤小鼠ROS水平和MDA含量,提高脾淋巴细胞和胸腺淋巴细胞的活性,增强小鼠SOD和GSH-Px活性,且呈量效关系。表明BA能加速细胞清除活性氧自由基,增强小鼠抵抗氧化损伤的能力,对Dex诱导的氧化损伤有一定的保护作用。

摘要:
γ-谷维素(γ-oryzanol)是米糠油中存在的微量生物活性物质,是环木菠萝醇类阿魏酸酯和甾醇类阿魏酸酯的混合物。γ-谷维素具有很多生理功能,如抗氧化、抗衰老、降血脂、降血糖等。γ-谷维素作为一种天然抗氧化剂,主要通过清除自由基、抑制脂质过氧化等方面发挥抗氧化作用。本文重点综述动物机体内自由基的产生和清除以及γ-谷维素的抗氧化作用机制。

摘要:
研究桦木酸对脾淋巴细胞凋亡的影响.以地塞米松建立小鼠体外脾淋巴细胞凋亡模型,利用倒置显微镜、荧光显微镜和电子透射显微镜观察细胞形态和亚细胞结构,琼脂糖凝胶电泳观察DNA的裂解,流式细胞术测定细胞凋亡率,检测华木酸对Dex介导的小鼠脾淋巴细胞凋亡的影响.结果表明,2.5 μmol Dex可使脾淋巴细胞凋亡明显增加,染色质发生边聚、浓缩、凋亡小体等典型的凋亡特征,桦木酸可明显降低地塞米松引起的脾淋巴细胞凋亡.提示桦木酸对地塞米松诱导的淋巴细胞凋亡具有预防性保护作用.

摘要:
Koumine is a kind of alkaloid extracted from Gelsemium elegans (G. elegans). Benth, which has shown promise as an anti-Tumor, anxiolytic, and analgesic agent. In our present study, the effect of koumine on lipopolysaccharide (LPS)-mediated RAW 264.7 cell apoptosis was evaluated. MTT assays showed that koumine obviously increased cell viability in LPS-mediated RAW 264.7 macrophages. Preincubation with koumine ameliorated LPS-medicated apoptosis by decreasing reactive oxygen species (ROS) production, which resulted in a significant decrease in the levels of nitric oxide (NO) and inducible nitric oxide synthase (iNOS). In addition, koumine-pretreated RAW 264.7 macrophages exhibited reduction of LPS-induced levels of TNF-α, IL-1β, and IL-6 mRNA. Furthermore, pretreatment with koumine suppressed LPS-mediated p53 activation, loss of mitochondrial membrane potential, caspase-3 activation, decrease of Bcl-2 expression, and elevation of Bax and caspase-3 expressions, suggesting that koumine might act directly on RAW 264.7 cells to inhibit LPS-induced apoptosis. It seems as though the mechanism that koumine possesses is the anti-Apoptotic effect mediated by suppressing production of ROS, activation of p53, and mitochondrial apoptotic pathways in RAW 264 cells. Koumine could potentially serve as a protective effect against LPS-induced apoptosis. © 2016 by the authors; licensee MDPI, Basel, Switzerland.

桦木酸(betulinic acid,BA)属于羽扇豆烷型五环三萜类物质,广泛分布于多种植物中。BA具有抗炎、抗肿瘤、抗微生物、抗寄生虫、抗溃疡、抗艾滋病毒和抗疟原虫等多种生物活性,并且其毒性低,安全指数高,具有广泛的应用前景。炎症是许多常见疾病的基本病理过程,调节炎症可以干预许多疾病的发生和发展。本文就BA的抗炎活性及其抗炎机制进行综述,为BA的研究和临床应用提供科学依据。

氧化应激为机体内活性氧的产生与抗氧化系统之间平衡失调,造成体内生物大分子(脂质、蛋白质以及DNA)损伤,导致细胞凋亡,从而诱发各种疾病的发生.抗氧化剂能有效调控机体的氧化应激与细胞凋亡,保障机体细胞内外环境的稳定和生理机能正常.氧化应激在高度集约化的现代养殖业中的危害已大量显现,减少或缓解畜禽的氧化应激已成为现代养殖业的重大课题.深入开展氧化应激与细胞凋亡以及新抗氧化剂的相关研究,对促进动物健康养殖和提高畜产品质量具有重要意义.笔者等在概述有关氧化应激与细胞凋亡的产生、反馈机制及其信号通道的基础上,综述了抗氧化剂应用的最新研究进展.
