The first complete mitochondrial genome of the agricultural pest Micromelalopha sieversi (Staudinger, 1892) (Lepidoptera: Notodontidae)

摘要:
Micromelalopha sieversi (Staudinger, 1892) is a significant pest of Poplar trees in China. In this study, we used high-throughput sequencing to sequence the whole mitochondrial genome of M. sieversi. The length of the genome was 15,373 base pairs. The nucleotide composition was 39.8%, 11.5%, 8.0%, and 40.7% for A, C, G, and T, respectively. We used the maximum-likelihood method to construct a molecular phylogenetic tree based on complete mitogenome sequences of 19 Noctuoidea species as ingroups and five Geometroidea species as outgroups. The results indicate that the genus Micromelalopha is closely related to the genus Clostera in family Notodontidae. © 2024 The Author(s). The ETHYLENE INSENSITIVE3-LIKE (EIL) family is one of the most important transcription factor (TF) families in plants and is involved in diverse plant physiological and biochemical processes. In this study, ten EIL transcription factors (CsEILs) in sweet orange were systematically characterized via whole-genome analysis. The CsEIL genes were unevenly distributed across the four sweet orange chromosomes. Putative cis-acting regulatory elements (CREs) associated with CsEIL were found to be involved in plant development, as well as responses to biotic and abiotic stress. Notably, quantitative reverse transcription polymerase chain reaction (qRT-PCR) revealed that CsEIL genes were widely expressed in different organs of sweet orange and responded to both high and low temperature, NaCl treatment, and to ethylene-dependent induction of transcription, while eight additionally responded to Xanthomonas citri pv. Citri (Xcc) infection, which causes citrus canker. Among these, CsEIL2, CsEIL5 and CsEIL10 showed pronounced upregulation. Moreover, nine genes exhibited differential expression in response to Candidatus Liberibacter asiaticus (CLas) infection, which causes Citrus Huanglongbing (HLB). The genome-wide characterization and expression profile analysis of CsEIL genes provide insights into the potential functions of the CsEIL family in disease resistance. © 2023 by the authors.

We performed the first sequencing of the complete mitogenome of Botyodes diniasalis by high-throughput sequencing. A circular DNA molecule of 15,219 bp in size, encoding 2 rRNAs, 22 tRNAs, and 13 PCGs, contains a non-coding AT-rich region. The overall nucleotide composition of the genome is A (39.5%), T (41.3%), C (11.3%), and G (7.8%). Phylogenetic analysis based on mitogenomic data suggest that the species B. diniasalis has a close evolutionary relationship with B. principalis in Margaroniini. The complete mitogenome of B. diniasalis will serve as a valuable resource for future studies on evolution, taxonomy, genetic conservation, and utilization of Botyodes. © 2023 The Author(s). 摘要:
Rice bacterial leaf blight (BLB) is the most destructive bacterial diseases caused by Xanthomonas oryzae pv. oryzae (Xoo). Phages have been proposed as a green and efficient strategy to kill bacterial pathogens in crops, however, the mechanism of action of phages in the control of phyllosphere bacterial diseases remain unclear. Here, the glasshouse pot experiment results showed that phage combination could reduce the disease index by up to 64.3%. High-throughput sequencing technology was used to analyze the characteristics of phyllosphere microbiome changes and the results showed that phage combinations restored the impact of pathogen invasion on phyllosphere communities to a certain extent, and increased the diversity of bacterial communities. In addition, the phage combination reduced the relative abundance of epiphytic and endophytic Xoo by 58.9% and 33.9%, respectively. In particular, Sphingomonas and Stenotrophomonas were more abundant. According to structural equation modeling, phage combination directly and indirectly affected the disease index by affecting pathogen Xoo biomass and phage resistance. In summary, phage combination could better decrease the disease index. These findings provide new insights into phage biological control of phyllosphere bacterial diseases, theoretical data support, and new ideas for agricultural green prevention and control of phyllosphere diseases.

烟芽夜蛾囊泡病毒3h株(Heliothis virescens ascovirus 3h, HvAV-3h)作为一种极具生防潜力的环状双链DNA昆虫病毒,自被分离后,对其基因的特性和相关蛋白功能研究从未间断。本研究通过生物信息学预测3H-38蛋白序列发现其131~150氨基酸位置有一段由胞外向胞内的跨膜区序列,N端23~121氨基酸位置有一个BRO家族结构域,和其同源蛋白的序列对比发现3H-38与烟芽夜蛾囊泡病毒3i株(Heliothis virescens ascovirus 3i, HvAV-3i)和烟芽夜蛾囊泡病毒3j株(Heliothis virescens ascovirus 3j, HvAV-3j)编码的同源蛋白3I-40和3J-43相似性高达85%以上。进一步通过RT-PCR克隆、构建pET-28a-38原核表达载体、IPTG诱导表达、Ni2+-NTA亲和层析柱纯化蛋白等方法获得了His-tag融合的3H-38重组蛋白,并制备了该蛋白的兔多克隆抗体。通过检测3h-38基因在HvAV-3h感染的甜菜夜蛾Spodoptera exigua(Hübner)幼虫中的转录和表达时相,本研究发现3h-38从感染后3 h开始转录,从感染后36 h开始表达,即3h-38是一个早期转录、晚期表达的基因。3h-38基因的生物学信息分析和转录表达时相检测为进一步研究该蛋白的功能和特性奠定了基础。

摘要:
The complete mitochondrial genome (mitogenome) of Lyssa zampa was first reported. It is 15,314 bp in length (GenBank accession number: MW435592) and consists of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes. The nucleotide composition is A (41.5%), C (11.1%), G (7.4%), and T (40.0%). Based on the sequences of complete mitogenome from 12 geometroid species and three drepanoid species as ingroups, and two noctuoid species as outgroups, the phylogenetic tree was constructed. The results showed that the closest relationship between Uraniidae and Epicopeiidae was strongly supported by Bayesian posterior probabilities values of 0.99. © 2021 The Author(s). 摘要:
为了解八大公山国家级自然保护区内的蝴蝶群落组成及海拔分布,作者于2016–2018年间对该区域不同生境的蝴蝶群落进行了系统调查和多样性分析,并持续观测其群落的月动态变化。本研究共记录蝴蝶6,164只,隶属于5科107属191种。低海拔(250–450 m)生境记录到的蝴蝶以凤蝶亚科为主,高海拔(1,200–1,400 m)生境以粉蝶亚科为主,而中海拔(700–900 m)生境无明显的蝴蝶优势类群。相似性分析结果表明,各生境存在很高比例的共有种(59%以上),这些共有种在不同生境的个体数分布存在很大的差异。季节动态分析结果表明,八大公山蝴蝶群落的物种数、个体数及多样性指数在4–6月逐渐上升, 7月达到峰值, 8–9月逐渐下降。总体来说,八大公山的蝴蝶群落组成沿海拔梯度呈现垂直分布差异,高海拔与低海拔存在明显的异质性,中海拔则呈现出一种过渡状态;且八大公山不同海拔的蝴蝶群落表现出较为一致的季节动态,都具有很高比例的物种更替;相对于低海拔,高海拔生境蝴蝶群落的活跃期更短,物种的时间周转率更高。

摘要:
大造桥虫Ascotis selenaria Schiffermuller et Denis属鳞翅目Lepidoptera尺蛾科Geometridae,又名尺蠖、步曲、棉大造桥虫等,是一种间歇性、爆发性、局部为害严重的世界性害虫,广泛分布于亚洲、欧洲和非洲。在国内主要分布于东北、华北、华东、华中和西南等地区。大造桥虫食性复杂,主要寄主为辣椒、茄子以及十字花科、豆科等经济作物,也可危害多种果树。主要以幼虫取食植物叶片,严重时取食整片叶,只留下枝干,极大影响了植物的光合作用,降低了作物及果树的经济价值和观赏价值。为此,本文以采于贵州省安顺市西秀区七眼桥镇山岚村试验田大造桥虫幼虫为供试虫源,将幼虫置于培养皿(直径10 cm)饲养,每天更换新鲜辣椒叶片,记录(拍照)各虫态的发育、形态(如体型、颜色、体表特征等)、行为习性与辣椒叶片受害症状,可为当地辣椒大造桥虫危害与防治提供参考依据。(1)形态特征:该虫幼虫(图1-A~B)躯体柔软,呈椭圆形,胸足3对,腹足2对。低龄幼虫黄白色,有白色横纹,第二腹节背面有两个不规则黑斑。高龄幼虫黑棕色,覆有体毛,头尾部居多;体表有多条棕色纵纹和黑斑;两对腹足距离缩短,气门线较低龄更明显。其蛹(图1-C)纺锤形,体长为14 mm左右。初为浅褐色,逐渐变为深褐色,有光泽。头顶有疣状突起,尾端尖,臀棘2根;成虫(图1-D)体长15~20 mm,翅展38~45 mm,体为浅灰褐色。翅面横线和斑纹均为暗褐色,前翅亚基线和外横线锯齿状,其间为灰黄色。雌触角丝状,雄羽状,淡黄色。(2)行为习性:初孵幼虫受到惊吓时,会吐丝下垂,随风飘荡到其它植物上继续取食。幼虫曲弓爬行,爬行时弯腰如拱桥;静止时靠2对腹足固定身体,斜向伸直如一个干枯的小树枝(图1-E)。1~3龄幼虫常昼夜取食,4~5龄夜晚活动取食。高龄幼虫有拟态和假死性,可用来躲避敌害。幼虫老熟后白天吐丝下垂或直接掉在地面,入松土化蛹。化蛹后不食不动,不存在排便行为。触碰尾部会转圈晃动,多个蛹距离近时会出现连锁反应一起晃动尾部。成虫多在傍晚羽化,羽化后1~3天开始交尾,交尾后1~2天开始产卵,产卵多在夜间,产于叶背、叶柄、树缝等处。(3)受害症状:大造桥虫幼虫咀嚼式口器,取食叶片,沿叶脉、叶缘将叶片咬成孔洞、缺刻,甚至将叶片吃光(仅剩主叶脉,如图1-F)。初孵幼虫会先集中在顶部嫩叶处,将顶部嫩叶咬成圆形网状或小孔洞。3龄幼虫开始分散取食,将叶片啃食成缺刻状。4~5龄幼虫取食量剧增,取食整片叶片,仅剩主叶脉,6龄幼虫取食量最大。被为害的辣椒叶片呈弧形,里圈为小锯齿状形状。幼虫先沿叶片的最周边开始进行取食,咬成圆弧状,后沿叶片向下进行取食,在叶片的周围形成一个个弧状,最后弧形直至相通,只剩叶脉。危害严重时穿插在叶肉间的大叶脉也被取食。近年来随着当地辣椒栽培面积的不断扩大,辣椒的虫害也日渐严重。大造桥虫主要取食辣椒叶片,特点是食量大,爆发性强,是辣椒虫害中比较严重的害虫之一。不仔细观察或防治不及时,会在短时间内将植株叶片吃光,后借助抽丝下垂转移到其他植株为害,给辣椒的栽培造成极大困扰。本次实验观察到大造桥虫幼虫期虫体颜色较浅,存在吐丝和排便行为。取食辣椒叶片,严重时整片叶尽食。爬行过程中,虫体依靠后半部分的两对腹足抓附在固定物上,曲弓前行。受到侵扰时,幼虫会出现"假死"行为;侵扰解除后,幼虫恢复活动。进入化蛹时期,不食不动,当触动尾部时,左右晃动。随着进入化蛹的时间越长,外壳颜色会逐渐加深,尾部体节数逐渐明显,可清楚观察到大造桥虫的头部、两翅的位置及尾部的结构。观察造桥虫形态特征、行为特性等,充分了解其危害辣椒的症状,以对症下药、科学防治。针对不同时期的虫态采取不同的方法,如人工防治、灯光诱杀、化学防治等,对其达到高效的防治,提前控制预防害虫大发生,提高辣椒品质及产量,降低农民的经济损失。

Although the cotton bollworm Helicoverpa armigera has traditionally been controlled by application of chemical pesticides, chemical control selects for resistance, pollutes the environment, and endangers human health. New methods for controlling H. armigera are therefore needed. Heliothis virescens ascovirus 3i (HvAV-3i) is a recently identified virus of the lepidopteran larvae. We tested the effects of HvAV-3i on H. armigera larvae following oral ingestion of HvAV-3i-containing hemolymph (about 1.0 × 1010 virus genome copies per larvae) and following injection of HvAV-3i-containing hemolymph by insertion of a needle. Following oral ingestion, first-instar to fifth-instar larvae grew and developed normally. Following needle injection, in contrast, the corrected mortality of third and fourth instars was 88.9 ± 2.1 and 93.7 ± 3.4%, respectively. Food intake was significantly lower for larvae injected with virus-containing hemolymph than with virus-free hemolymph. Larvae injected with virus-containing hemolymph had extended survival times and could not complete the pre-pupal stage. These results indicate that inoculation of HvAV-3i via needle injection, but not via oral ingestion, significantly reduced the growth and development of H. armigera larvae. © Copyright © 2020 Chen, Liu, Mo, Hu, Liu, Bustos-Segura, Xue and Wang.

烟青虫是一种世界性害虫,主要以幼虫为害作物的各生长期及不同生长部位,严重影响作物的生长发育和农产品的品质。目前对烟青虫的防治仍以化学防治为主,新型的防治技术相对较少且不被广大农户所接受。本文详细总结了辣椒烟青虫各类防治对策的研究进展,展望了未来的发展趋势,以期为今后烟青虫的防治应用提供理论参考。

摘要:
Abstract In this study, the complete mitochondrial genome of Hydrelia parvulata (Staudinger, 1897) was firstly sequenced by high-throughput sequencing. As a circular DNA molecule, it is 15,407 bp in length (GenBank accession number: MN962739), which consists of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and an AT-rich region. The nucleotide composition is A (39.8%), C (11.6%), G (8.2%), and T (40.4%). Based on the sequences of complete mitogenome from 10 geometrid species as ingroups and 3 bombycid species as outgroups, the phylogenetic trees were constructed using maximum likelihood (ML) method. The results showed that H. parvulata has a close evolutionary relationship with Operophtera brumata in the subfamily Larentiinae, which is a clade strongly supported by the bootstrap value of 100% and a sister-group with the subfamily Ennominae.

不同温度对草地贪夜蛾幼虫和蛹生长发育的影响

摘要:
为明确温度对草地贪夜蛾生长发育的影响,测定了6个温度(18,21,24,27,30和33℃)条件下草地贪夜蛾幼虫和蛹的发育历期、蛹重和存活率等指标,计算出各虫态的发育起点温度、有效积温和各温度条件下的发育速率。结果表明:在18~33℃范围内,草地贪夜蛾幼虫和蛹的发育历期均随着温度的升高而逐渐缩短,发育速率逐渐加快,其生长发育的最适宜温度范围为24~33℃;幼虫和蛹的发育起点温度分别为10.61±3.19℃和13.29±1.34℃,有效积温分别为183.19±9.96 d·℃和134.83±13.71 d·℃。该虫的蛹重随着温度的升高呈先升高后降低的变化趋势,在21℃条件下,其蛹重达到最大值251.91±11.19 mg。

桦蛾科Endromidae隶属于昆虫纲Insects鳞翅目Lepidoptera,全世界性分布,已知16属70种。桦蛾是一类中型蛾子,幼虫多取食植物叶片,有些种类为农林业、园艺园林业的重要害虫。因此,对其开展系统学研究,进行准确的种类鉴定对于农林业生产具有重要的研究价值和经济意义。目前世界范围内已开展的桦蛾科研究均为区域性工作,但仍缺少全面的区系研究和系统的分类修订工作。另外,桦蛾科是鳞翅目昆虫系统树上的关键点之一,对其进行深入的形态、分子研究,重建其系统发育以修订完善鳞翅目高级阶元分类系统,也将对中国鳞翅目系统分类研究提供参考,对农林害虫的防治工作提供借鉴。

中国苜蓿害虫及其天敌功能团多样性分析

摘要:
苜蓿是当今世界分布最广的栽培牧草,苜蓿害虫种类繁多,严重威胁着畜牧养殖业。经过实地调查并结合相关文献对我国苜蓿害虫及其天敌物种多样性和功能团分析,将苜蓿害虫划分为6个功能团,将天敌划分为2个功能团。结果显示:我国苜蓿害虫有301种,隶属于7目50科205属,其中以鳞翅目和鞘翅目种类最多,而功能团中则以食茎叶类害虫种类最丰富;苜蓿害虫自然天敌有68种,隶属于8目26科48属,以鞘翅目为主的捕食性天敌种类最多。本文总结了苜蓿上的害虫及其天敌数据并按功能团进行分析,旨在为苜蓿害虫的综合防治提供一定的参考依据。

摘要:
为了对湖南省衡山国家级自然保护区的蝶类物种多样性保护状况进行评估。本研究于2018年4—9月,在该保护区选取5条不同生境的样线逐月进行蝶类多样性调查。共记录蝶类1 260只,隶属于5科59属84种。其中蛱蝶科的物种最为丰富(38种,占45.24%),其后依次是凤蝶科、灰蝶科、粉蝶科、弄蝶科。不同生境的蝴蝶群落存在一定差异,其中位于保护区核心区生境b(常绿阔叶林)的多样性指数最高(3.059),而位于保护区试验区e(农田)的多样性指数最低(2.559),说明保护区对于当地蝴蝶多样性的保护发挥了积极的作用。

In this study, the complete mitochondrial genome (mitogenome) of Gibbovalva kobusi was at first sequenced by high-throughput sequencing. As a circular DNA molecule, the complete mitogenome is 15,717 bp in length (GeneBank accession number: MK956103) and consists of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and an AT-rich region. The nucleotide composition is A (41.0%), C (11.6%), G (7.9%), and T (39.5%). Based on the sequences of complete mitogenome from 11 species as ingroups and 3 superfamily Tineoidea species as outgroups, the phylogenetic trees were constructed. The family Gracilariidae as a monophyletic clade is strongly supported by the bootstrap value of 100%. © 2019, © 2019 The Author(s). Microbial antagonists and their bioactive metabolites provide one of the best alternatives to chemical pesticides to control crop disease for sustainable agriculture and global food security. The rice endophyte Streptomyces hygroscopicus OsiSh-2, with remarkable antagonistic activity towards the rice blast fungus Magnaporthe oryzae, was reported in our previous study. The present study deciphered the possible direct interaction mode of OsiSh-2 against M. oryzae. An in vitro antibiotic assay for OsiSh-2 culture filtrate revealed strong suppression of mycelial growth, conidial germination and appressorial formation of M. oryzae. Meanwhile, severe morphological and internal abnormalities in M. oryzae hyphae were observed under a scanning electron microscope and transmission electron microscope. Foliar treatment of rice seedlings by OsiSh-2 culture filtrate in the greenhouse and in the field showed 23.5% and 28.3% disease reduction, respectively. Correspondingly, OsiSh-2 culture filtrate could induce disorganized chitin deposition in the cell wall and lowered ergosterol content in the cell membrane of M. oryzae. Additionally, cell wall integrity pathway activation, large cell electrolytes release, reactive oxygen species accumulation and tricarboxylic acid cycle-related enzyme activity changes were found in M. oryzae. All these results suggested that the direct antagonistic activity of OsiSh-2 against M. oryzae may be attributed to damaging the integrity of the cell wall and membrane and disrupting mitochondrial function in the pathogen. © 2019 Elsevier Inc.

The family Bombycidae (sensu Minet, 1994) is a diverse group of species belonging to the superfamily Bombycoidea. It is an economically important group of moth species, containing well-known silk-producing insects, as well as many pests of agriculture and forestry. The morphology-based hypothesis of Minet (1994) on the composition of Bombycidae is in conflict with subsequent phylogenetic hypotheses for the superfamily based on nuclear genes. In this paper, the complete mitochondrial genomes of nine species of Bombycidae are presented for the first time. Based on these genomes, four dataset partitions and three gblocks parameter settings, phylogenetic relationships among Bombycidae were reconstructed using maximum likelihood and Bayesian inference methods. Bombycidae was confirmed as a polyphyletic group, with the traditional subfamilies Prismostictinae and Oberthueriinae forming a single well-supported clade that is distant to Bombycinae. The phylogenetic relationships within Bombycoidea were supported as ((((Bombycinae, Sphingidae), Saturniidae), (Prismostictinae, Oberthueriinae)), Eupterotidae).

草地贪夜蛾入侵我国后快速扩散,发现和保护利用本土天敌是有效抑制其暴发为害的重要途径。本研究以长沙地区农田常见的斯氏侧沟茧蜂为研究对象,系统观察了其对不同龄期草地贪夜蛾幼虫的寄生行为,测定了被寄生草地贪夜蛾幼虫体重和取食量的变化。结果表明,斯氏侧沟茧蜂能寄生2~4龄草地贪夜蛾幼虫,可显著抑制其体重增量和取食量,有效阻滞其进入暴食期,并最终致其死亡。说明斯氏侧沟茧蜂可有效寄生草地贪夜蛾幼虫,并抑制其生长和取食,对其具有一定生物防控能力,是值得保护和利用的本土寄生蜂。
