Highly efficient degradation of thidiazuron with Ag/AgCl- activated carbon composites under LED light irradiation

摘要:
Thidiazuron (TDZ;1-phenyl-3-(1,2,3-thiadiazol-5-yl)urea) is one of the most widely used defoliant and easy to dissolve in surface water. Risk associated with the pesticide is not clearly defined, so it is important to remove/degrade TDZ with an efficient and environment friendly technology. Here, we investigated the use of Ag/AgCl-activated carbon (Ag/AgCl–AC) composites in photocatalytic degradation of TDZ under LED light. By the synergic effect of Ag/AgCl and AC, the optimum Ag/carbon weight ratio of 2:1 exhibited superior visible-light photocatalytic activity, the highest removal efficiency was close to 91% in pH 7 matrix. Different types of Ag/AgCl–AC composites were tested, all showed much faster photodegradation kinetics than bare Ag/AgCl in 210 min. The degradation products as identified by HPLC–MS revealed that the hydroxylation by hydroxyl radicals and that of oxidation by superoxide radicals as well as holes were the two main pathways for TDZ degradation. Results revealed that the adsorption concentrated TDZ molecules and the photocatalytically generated radicals rapidly degradated TDZ, the two contributions functioned together for removal of the pollutant from water. ©2017 Elsevier B.V.

摘要:
为寻找控制烟草黑胫病发生的有效措施,研究了硝态氮、铵态氮及其配比液对烟草黑胫病菌孢子萌发、菌丝生长以及烟株次生代谢物和抗病性的影响。结果表明,硝态氮、铵态氮及其混合液均可以加速烟草黑胫病游动孢子的萌发,二者混合溶液更有利于黑胫病菌丝的生长。随着溶液中铵态氮浓度的升高,烟株根系木质素、纤维素和MDA含量以及SOD、POD、 PAL活性显著升高,增强了烟株的抗病性,从而使其发病率和病情指数显著下降。相关性分析表明,烟株黑胫病发病率和病情指数与其根系木质素、MDA含量和SOD、POD、PAL活性均极显著负相关,黑胫病发病率与纤维素含量显著负相关。因此,烟株黑胫病发病率和病情指数与其次生代谢物密切相关,铵态氮可以显著降低烟草黑胫病发病率和病情指数,但硝态氮和铵态氮最适配比及田间抗病效果还需进行大量相关田间试验研究。

摘要:
bZIP transcription factors play key roles in plant growth, development, and stress signaling. A bZIP gene BnbZIP2 (GenBank accession number: KP642148) was cloned from ramie. BnbZIP2 has a 1416 base pair open reading frame, encoding a 471 amino acid protein containing a characteristic bZIP domain and a leucine zipper. BnbZIP2 shares high sequence similarity with bZIP factors from other plants. The BnbZIP2 protein is localized to both nuclei and cytoplasm. Transcripts of BnbZIP2 were found in various tissues in ramie, with significantly higher levels in female and male flowers. Its expression was induced by drought, high salinity, and abscisic acid treatments. Analysis of the cis-elements in promoters of BnbZIP2 identified cis-acting elements involved in growth, developmental processes, and a variety of stress responses. Transgenic Arabidopsis plants' overexpression of BnbZIP2 exhibited more sensitivity to drought and heavy metal Cd stress during seed germination, whereas more tolerance to high-salinity stress than the wild type during both seed germination and plant development. Thus, BnbZIP2 may act as a positive regulator in plants' response to high-salinity stress and be an important candidate gene for molecular breeding of salt-tolerant plants. © 2016 Mary Ann Liebert, Inc.

采用砂培的方式,研究了打顶后养分供应水平与叶片采收方式对烤烟体内钾素积累分配的影响。结果表明,钾素在烟株体内移动活跃,自然生长烟株的花序和打顶烟株的上部叶片作为活跃的生长点,是钾素优先分配部位。在低钾条件下,下部叶中贮存的钾素能够大量外运,以满足生长点对钾素的需要;分次采收烟株的下部叶由于离体,其内部固定有大量钾素,故保持较高的钾含量,但花序或上部叶对钾素的需求未得到满足,钾含量偏低:一次采收烟株的下部叶则能够始终充当钾源,有利于上部叶钾含量的提高。保持钾素的正常供应,能够促进烟株生长,增加钾素在烟株体内的积累。

摘要:
采用砂培方式,研究枸溶性钾肥与硫酸钾配施(质量比1∶3、1∶1和3∶1)对烤烟根系性状、根系活力及烟株钾素吸收积累的影响。结果表明,枸溶性钾肥与硫酸钾配施,可促进烤烟根系生长发育,优于单施硫酸钾和枸溶性钾肥;枸溶性钾肥与硫酸钾配施促进烟株对钾素的吸收和分配,烟株的上、中、下3部位烟叶钾素含量较单施硫酸钾的提高7.60%、23.24%和39.95%,总叶钾含量分配率分别提高8.41%、20.91%和42.03%;以75%枸溶性钾肥与25%硫酸钾配施处理效果最佳。

为了揭示根区温度对烤烟烟碱积累的影响机制,通过盆栽试验研究了根区温度(10,18,26和35℃)与氮素形态(100%NH_4~+-N,50%NH_4~+-N+50%NO_3~--N和100%NO_3~--N)互作对烤烟生物量和烟碱积累的影响。结果表明:适当提高根区温度可增加烟株生物量,NO_3~--N比NH_4~+-N更有利于烟株的生长。当根区温度为35℃时,供应100%NH_4~+-N和50%NH_4~+-N+50%NO_3~--N的烟株生长受到抑制,而供应100%NO_3~--N的烟株生长旺盛。氮素形态对烟株生物量的影响大于根区温度。烟株的烟碱积累量在根区温度为18℃时达到最大,供应100%NO_3~--N的烟株烟碱积累量是供应100%NH_4~+-N烟株的1.64倍,施用NO_3~--N有利于烟碱的合成。当根区温度为35℃时,烟株的烟碱合成受到抑制。根区温度对烟碱积累的影响大于氮素形态。

摘要:
以K326为供试品种,利用盆栽试验研究不同硫肥施用量对烟草农艺性状、叶绿素(叶绿素a、b)含量及硝酸还原酶活性的影响。结果表明:相比于施硫处理,不施硫处理烟株生长状况、烟叶叶绿素含量、硝酸还原酶(NR)活性受到明显的抑制;每株施硫量为1.5 g的中等施硫处理烟株长势最好,移栽前期叶绿素含量及NR活性较高,到成熟期时叶绿素含量及NR活性均降低;每株施硫量为3.0 g的高硫处理,烟株能够完成生育期,但是叶片横向生长受到限制,最大叶宽仅为20.67 cm。高硫处理对烟叶叶绿素含量及NR活性具有显著的促进作用,但是烟株生长后期烟叶中叶绿素含量及NR活性持续偏高,分别为1.10 mg/g和63.10μmol/g·h,不利于烟株的正常成熟落黄及内在品质的形成。

采用盆栽试验,研究了根部不同温度和不同氮素形态的互作对于烤烟生长和钾素积累的影响。结果表明:提高根部温度和供应NO_3-N比例,能够显著促进烟株的生长,根系活力和钾素积累增加。根部温度为10℃和18℃时,供应的NH_4-N烟株根系对钾素的吸收能力较弱;当根部温度为35℃时,供应100%NO_3-N的烟株生长旺盛,但供应NH_4-N的烟株生长受到强烈抑制,根部生物量下降,并有大量的钾素外泌。通过分析可知,氮素形态对烟株生长和钾素积累的影响大于根部温度。提高根部温度和供应硝态氮有利于烟株的生长和钾素的吸收;当根部温度较高时,氨态氮不利于烟株的生长。

摘要:
为研究烤烟根系分泌物对烤烟幼苗生长的影响,在烤烟幼苗生长过程中施加不同浓度与烤烟根系分泌物主要组成成分相同的3种外源性化学物质(苯甲酸、水杨酸和烟碱),模拟烤烟根系分泌物对烤烟幼苗生长的影响,对比三者的影响差异。结果表明,水杨酸、苯甲酸和烟碱3种物质促进烤烟幼苗根系生长和幼苗光合特性的浓度范围主要集中在0 ~ 200、0 ~ 400、0 ~ 800μmol·L ~(- 1)(其中根表面积达到1 000μmol·L~(- 1),气孔导度和蒸腾速率达到600μmol·L ~(- 1)),促进烤烟幼苗叶绿素荧光参数的浓度范围主要集中在0 ~ 200、0 ~ 800、0 ~ 800μmol·L~(- 1),而3种外源物质的PS Ⅱ实际光合效率在200μmol·L - 1时均达到最大。由此可知,在烤烟幼苗生长过程中,3种外源性化学物质对幼苗的根系形态结构、光合特性和叶绿素荧光参数表现为低浓度促进、高浓度抑制,且浓度越高抑制程度越大,但不同组分对烤烟幼苗生长影响不同,相同浓度下3种物质的促进作用表现为烟碱> 苯甲酸> 水杨酸。本研究可为解决烤烟连作障碍提供理论依据。

Rhodopseudomonas palustris strain JSC-3b isolated from a water canal adjacent to a vegetable field produces a protein that was purified by bioactivity-guided fractionation based on ammonium sulfate precipitation, ion-exchange absorption and size exclusion. The protein was further identified as an endoribonuclease L-PSP (Liver-Perchloric acid-soluble protein) by shotgun mass spectrometry analysis and gene identification, and it is member of YER057c/YjgF/UK114 protein family. Herein, this protein is designated Rhp-PSP. Rhp-PSP exhibited significant inhibitory activities against tobacco mosaic virus (TMV) in vivo and in vitro. To our knowledge, this represents the first report on the antiviral activity of a protein of the YER057c/YjgF/UK114 family and also the first antiviral protein isolated from R. palustris. Our research provides insight into the potential of photosynthetic bacterial resources in biological control of plant virus diseases and sustainable agriculture.

Ustilaginoidea virens (Cooke) Takah is an ascomycetous fungus that causes rice false smut, a devastating emerging disease worldwide. Here we report a 39.4a €‰Mb draft genome sequence of U. virens that encodes 8,426 predicted genes. The genome has ∼25% repetitive sequences that have been affected by repeat-induced point mutations. Evolutionarily, U. virens is close to the entomopathogenic Metarhizium spp., suggesting potential host jumping across kingdoms. U. virens possesses reduced gene inventories for polysaccharide degradation, nutrient uptake and secondary metabolism, which may result from adaptations to the specific floret infection and biotrophic lifestyles. Consistent with their potential roles in pathogenicity, genes for secreted proteins and secondary metabolism and the pathogen-host interaction database genes are highly enriched in the transcriptome during early infection. We further show that 18 candidate effectors can suppress plant hypersensitive responses. Together, our analyses offer new insights into molecular mechanisms of evolution, biotrophy and pathogenesis of U. virens. © 2014 Macmillan Publishers Limited. All rights reserved.

为建立不同产区及风格烤烟烟叶与化学指标之间的关系,对2005~2009年湖南省、河南省、福建省和云南省等4省份共计1040份烟叶样品的21种化学成分进行检测,并利用Mining Tree模型进行分类-回归决策树(C&RT analysis)数据分析。结果表明:(1)从21个烟叶化学成分中,共筛选出14个特征化学指标,其中钾和硝酸根为4省烟叶共有特征化学指标;(2)湖南、河南、福建和云南4省份识别概率最高的特征化学指标分段组合(Segment)分别为:还原糖(≤24.93%)、钾(〉1.98%)、p H值(≤5.37)(p=0.658);钾(≤1.98%)、灰分(〉11.03%)、水溶性灰份碱度(≤0.49)、硝酸根(≤0.06%)(p=0.776);还原糖(〉24.93%)、钾(〉1.98%)、硝酸根(≤0.06%)、蛋白质(〉5.01%)(p=0.914);钾(≤1.98%)、灰分(≤11.03%)、水溶性总糖(〉28.94%)、硫酸根(≤1.43%)(p=0.957)。分类-回归决策树方法在建立烤烟化学成分识别模型中具有重要的应用价值。

Amino acids are important nutrients that function as both tissue building blocks and metabolic regulators. How amino acid balance is vital for the development and metabolism. Amino acid sensors, serials of particular functional molecules, possess the function of sensing amino acids has been reported in recent years. Plant nucleotide-binding leucine-rich repeat (NB-LRR) proteins serve as intracellular sensors to detect pathogen effectors and trigger immune responses. Transcription of the NB-LRR-encoding Resistance (R) genes needs to be tightly controlled to avoid inappropriate defense activation. How the expression of the NB-LRR R genes is regulated is poorly understood. The Arabidopsis (Arabidopsis thaliana) suppressor of npr1-1, constitutive 1 (snc1) mutant carries a gain-of-function mutation in a Toll/Interleukin1 receptor-like (TIR)-NB-LRR-encoding gene, resulting in the constitutive activation of plant defense responses. A snc1 suppressor screen identified modifier of snc1,9 (mos9), which partially suppresses the autoimmune phenotypes of snc1. Positional cloning revealed that MOS9 encodes a plant-specific protein of unknown function. Expression analysis showed that MOS9 is required for the full expression of TIR-NB-LRR protein-encoding RECOGNITION OF PERONOSPORA PARASITICA 4 (RPP4) and SNC1, both of which reside in the RPP4 cluster. Coimmunoprecipitation and mass spectrometry analyses revealed that MOS9 associates with the Set1 class lysine 4 of histone 3 (H3K4) methyltransferase Arabidopsis Trithorax-Related7 (ATXR7). Like MOS9, ATXR7 is also required for the full expression of SNC1 and the autoimmune phenotypes in the snc1 mutant. In atxr7 mutant plants, the expression of RPP4 is similarly reduced, and resistance against Hyaloperonospora arabidopsidis Emwa1 is compromised. Consistent with the attenuated expression of SNC1 and RPP4, trimethylated H3K4marks are reduced around the promoters of SNC1 and RPP4 in mos9 plants. Our data suggest that MOS9 functions together with ATXR7 to regulate the expression of SNC1 and RPP4 through H3K4 methylation, which plays an important role in fine-tuning their transcription levels and functions in plant defense. © 2013 American Society of Plant Biologists. Grain size, number and starch content are important determinants of grain yield and quality. One of the most important biological processes that determine these components is the carbon partitioning during the early grain filling, which requires the function of cell wall invertase. Here, we showed the constitutive expression of cell wall invertase-encoding gene from Arabidopsis, rice (Oryza sativa) or maize (Zea mays), driven by the cauliflower mosaic virus (CaMV) 35S promoter, all increased cell wall invertase activities in different tissues and organs, including leaves and developing seeds, and substantially improved grain yield up to 145.3% in transgenic maize plants as compared to the wild-type plants, an effect that was reproduced in our 2-year field trials at different locations. The dramatically increased grain yield is due to the enlarged ears with both enhanced grain size and grain number. Constitutive expression of the invertase-encoding gene also increased total starch content up to 20% in the transgenic kernels. Our results suggest that cell wall invertase gene can be genetically engineered to improve both grain yield and grain quality in crop plants. © 2013 Society for Experimental Biology, Association of Applied Biologists and John Wiley & Sons Ltd.

In this paper, we embed an Eu(III)-based chemosensor into a silica matrix of MCM-41, which is identified and confirmed by IR and small-angle X-ray diffraction spectra. The photophysical property of this composite material is analyzed. It is found that the emission coming from the Eu(III)-based chemosensor is immune to environmental influence due to the unique f-f radiative transitions. The sensing performance of the composite material is then investigated in detail. Data suggest that the composite material exhibits a good linear relationship with increasing Hg(II) concentrations from 1 to 12 μM, with K SV of 1.733 × 10 5 L/mol and linearly dependent coefficient of 0.9942. In addition, the emission of the composite material shows no obvious response towards interference from other metal ions and various pH values, which means that it is a promising chemo-sensing material for Hg(II) detection. © 2012 Elsevier B.V. 摘要:
The fatty acid composition in the seed oil was significantly modified following the introduction of transgenes. To further enhance the desirable characteristics of rapeseed oil, it would be beneficial to develop a new approach for the simultaneous silencing of two or more target genes. Our goals in the current study were to (1) increase oleic acid to more than 75%, (2) reduce polyunsaturated fatty acids (PUFA) to about 10% and erucic acid to zero, and (3) accomplish these changes in a single-transformation event. In a single transformation, two fragments amplified from the fatty acid Δ12-desaturase 2 (BnaFAD2) and fatty acid elongase 1 (BnaFAE1) genes of Brassica napus were linked together to form a fusion fragment. The fusion fragment was then used to assemble unique intron-spliced hairpin interfering constructs. In the transgenic plant FFRP4-4, the expression of BnaFAD2 and BnaFAE1 genes was completely inhibited. The composition of oleic acid in FFRP4-4 rose to 85%, PUFA dropped to 10% and erucic acid was undetectable. All hybrid F1 seeds obtained from the reciprocal crossing of FFRP4-4 and GX-parents (with different genetic backgrounds) contained more than 80% oleic acid, about 10% PUFA and very low, or undetectable, erucic acid. The results confirmed that the fusion fragment silencing construct can simultaneously and effectively silence the target genes on a consistent basis. The strategy provides a useful tool for detecting gene function and advancing genetic engineering techniques for the improvement of agricultural crops. © Springer-Verlag 2010.

The wolf spider Lycosa singoriensis is a hunting spider with a widespread distribution in northwest China. The venom gland of spiders, which is a vent specialized secretory tissue, can secrete abundant and complex toxin components. To extensively examine the transcripts expressed in the venom glands of L. singoriensis, we generated 833 expressed sequence tags (ESTs) from a directional cDNA library. Toxin-like sequences account for 69.1% of these ESTs, 17.3% are similar to cellular transcripts and 13.6% have no significant similarity to any known sequences. Here, we identified 223 novel toxin-like sequences, which can be classified into six different. superfamilies; that means a novel potential source of ligands for varied ion channels was discovered. With the aid of Gene Ontology terms and homology to eukaryotic orthologous groups, the annotation of cellular transcripts revealed some cellular processes important for the toxin secretion of venom glands including protein synthesis, protein folding, tuned post-translational processing and trafficking, etc. (C) 2009 Elsevier GmbH. Systemic acquired resistance (SAR) is a plant immune response induced by local necrotizing pathogen infections. Expression of SAR in Arabidopsis (Arabidopsis thaliana) plants correlates with accumulation of salicylic acid (SA) and up-regulation of Pathogenesis-Related (PR) genes. SA is an essential and sufficient signal for SAR. In a genetic screen to search for negative regulators of PR gene expression and SAR, we found a new mutant that is hypersensitive to SA and exhibits enhanced induction of PR genes and resistance against the virulent oomycete Hyaloperonospora arabidopsidis Noco2. The enhanced pathogen resistance in the mutant is Nonexpressor of PR genes1 independent. The mutant gene was identified by map-based cloning, and it encodes a protein with high homology to Replication Factor C Subunit3 (RFC3) of yeast and other eukaryotes; thus, the mutant was named rfc3-1. rfc3-1 mutant plants are smaller than wild-type plants and have narrower leaves and petals. On the epidermis of true leaves, there are fewer cells in rfc3-1 compared with the wild type. Cell production rate is reduced in rfc3-1 mutant roots, indicating that the mutated RFC3 slows down cell proliferation. As Replication Factor C is involved in replication-coupled chromatin assembly, our data suggest that chromatin assembly and remodeling may play important roles in the negative control of PR gene expression and SAR.

Jingzhaotoxin-34 (JZTX-34) is a 35-residue polypeptide from the venom of Chinese tarantula Chilobrachys jingzhao. Our previous work reported its full-length cDNA sequence encoding a precursor with 87 residues. In this study we report the protein expression and biological function characterization. The toxin was efficiently expressed by the secretary pathway in yeast. Under whole-cell patch-clamp mode, the expressed JZTX-34 was able to inhibit tetrodotoxin-sensitive (TTX-S) sodium currents (IC50 similar to 85 nM) while having no significant effects on tetrodotoxin-resistant (TTX-R) sodium currents on rat dorsal root ganglion neurons. The inhibition of TTX-S sodium channels was completely reversed by strong depolarization (+120 mV). Toxin treatment altered neither channel activation and inactivation kinetics nor recovery rate from inactivation. However, it is interesting to note that in contrast to huwentoxin-IV, a recently identified receptor site-4 toxin from Ornithoctonus huwena venom, 100 nM JZTX-34 caused a negative shift of steady-state inactivation curve of TTX-S sodium channels by approximately 10 mV. The results indicated that JZTX-34 might inhibit mammalian sensory neuronal sodium channels through a mechanism similar to HWTX-IV by trapping the 1154 voltage sensor in the resting conformation, but their binding sites should not overlay completely.
