摘要:
Rice is one of the major pathways of arsenic (As) exposure in human food chain, threatening over half of the global population. Greenhouse pot experiments were conducted to examine the effects of Si application on iron (Fe) plaque formation, As uptake and rice grain As speciation in indica and hybrid rice genotypes with different radial oxygen loss (ROL) ability. The results demonstrated that Si significantly increased root and grain biomass. Indica genotypes with higher ROL induced greater Fe plaque formation, compared to hybrid genotypes and sequestered more As in Fe plaque. Silicon applications significantly increased Fe concentrations in iron plaque of different genotypes, but it decreased As concentrations in the roots, straws and husks by 28-35%, 15-35% and 32-57% respectively. In addition, it significantly reduced DMA accumulation in rice grains but not inorganic As accumulation. Rice of indica genotypes with higher ROL accumulated lower concentrations of inorganic As in grains than hybrid genotypes with lower ROL. © 2016 Elsevier Ltd. All rights reserved.

高速滚动轴承喷油润滑油液穿透机理分析

摘要:
基于空气动力学研究高速滚动轴承环间气流特性,通过建立气相流数学模型和气液两相流数学模型研究润滑油在轴承环间的穿透过程,探究高DN值时润滑油的穿透机理.利用流体体积(VOF)模型对此状态下的空气和润滑油界面进行动态捕捉,以研究润滑油的运动过程、分布特点以及不同参数对环间油液体积分数的影响,得到了高速滚动轴承环间气相流的特性;润滑油在不同转速下进入轴承环间的运动过程;轴承环间气液两相流场的压力、速度特性;轴承环间初始气流场对润滑油进入的影响.结果表明:在轴承小端面靠近内圈附近喷油可以避免湍流对润滑油的影响和干扰,有利于润滑油进入环间;轴承环间存在有利于润滑油贴滚道运动的气流径向作用力,随着转速的增加,该力呈近似线性增加;气流的初始状态影响着轴承环间润滑油的运动状态,润滑油对气流的运动影响较小;较低转速时,轴承环间周向压力变化很小可忽略,较高转速时,其呈现周期性波动,对润滑油进入的影响不可忽视;在较低转速时通过提高喷油体积流量可以有效提高轴承环间油液体积分数,但是高转速时,通过提高喷油体积流量来提高轴承环间的油液体积分数的效果并不明显.

摘要:
DELLA proteins and phytohormone gibberellin act together to control convergence point of plant development. A gibberellin-bound nuclear receptor that interacts with the N-terminal domain of DELLA proteins is required for gibberellin induced degradation of DELLA proteins. N-terminal DELLA domain includes two conserved motifs: DELLA and VHYNP. However, their respective functions remain unclear. Meanwhile, the identification and detection of several bioactive gibberellins from the more than 100 gibberellin metabolites are overwhelmingly difficult for their similar structures. Using in vitro biochemical approach, our work demonstrates for the first time that the synthetic GAI N-terminal DELLA domain peptides have similar bioactive function as the expressed protein to interact with AtGID1 a receptor. Furthermore, our results reveal that DELLA motif is vitally important region and DELLA segment is essentially required region to recognize AtGID1a receptor. Finally, based on bioactive GA-dependent of the interaction between AtGID1a and DELLA protein, we generated a new method that could identify and detect bioactive GAs accurately and rapidly with surface plasmon resonance assays. 摘要:
Nobiletin (NOB) is major citrus flavonoid with many health-promoting benefits. We reported previously that 4′-demethylnobiletin (4DN), a major metabolite of NOB, significantly inhibited lipopolysaccharide (LPS)-stimulated inflammation in RAW 264.7 macrophages. In this study, we further studied the anti-inflammatory effects of 4DN in TPA-induced skin inflammation in mice. We demonstrated that topical application of 4DN decreased TPA-induced ear edema by >88 ± 4.77% in mice. This inhibitory effect was associated with inhibition on TPA-induced up-regulation of pro-inflammatory cytokines IL-1β, IL-6, and TNF-α. Immunoblotting results showed that 4DN resulted in profound effects on multiple proteins related with inflammation and carcinogenesis. 4DN significantly decreased the expression levels of iNOS, COX-2, and MMP-9, suppressed phosphorylation of PI3K/Akt and ERK, and increased the levels of HO-1 and NQO1 in TPA-treated mice. Overall, the results demonstrated that 4DN had strong anti-inflammatory effects in vivo, which provided a scientific basis for using NOB to inhibit inflammation-driven diseases. © 2015 American Chemical Society.

摘要:
Calcium (Ca2+) plays an important role in guaranteeing the formation and structure of microbial biofilm. However, little is known regarding its influence on the structure and electron transfer properties of electroactive biofilms (EABs). In this study, advanced investigative techniques such as microscopy and electrochemistry are employed to characterize biofilm structure and electron transfer properties following growth at different Ca2+ concentrations in a bioelectrochemical system (BES). As revealed by electrochemical measurements, a lower current is obtained from the EABs formed in the presence of higher Ca2+ concentrations. The decline of the current is due to the dominance of non-active cells and non-exoelectrogens in the biofilms formed at high Ca2+ concentrations. The results of this study provide a better understanding of the effect of Ca2+ on EABs structure and thus electricity generation in BESs. (C) 2015 Elsevier B.V. All rights reserved.

摘要:
Convenient detection methods for bioactive food compounds and their metabolites in biological samples are needed to better understand their mechanism of actions. Herein, we developed a novel approach to directly monitor and visualize the distribution of 5,3′,4′-tridemethylnobiletin (TDN), a unique polymethoxyflavone metabolite derived from citrus polymethoxyflavone, in biological samples such as cultured cells and mouse colonic tissues. Our results showed that a fluorescent conjugate could be formed between TDN and 2-aminoethyl diphenyl borate (DPBA) under simple reaction conditions, which was confirmed by both Raman spectroscopy and mass spectroscopy. We further demonstrated the application of DPBA-based conjugation reaction in the characterization of TDN in different biological samples including floating cells, adherent cells, and animal tissues. This is the first report demonstrating direct fluorescent detection of polymethoxyflavone in biological samples. ©2015 American Chemical Society.

Obesity, considered as a consequence of overnutrition, sustains a low-degree inflammatory state and results in insulin-resistance and type 2 diabetes. Here, we investigated the anti-inflammatory effects of 5-caffeoylquinic acid (5-CQA) in high-fat diet-induced obese rats. Serum interleukin (IL)-6, monocyte chemotactic protein 1 (MCP-1), tumor necrosis factor-alpha (TNF-α), total cholesterol (TC), triglyceride (TG), and free fatty acid (FFA) levels were determined. Expression of genes related to TG metabolism, macrophage biomarkers, and inflammation was assessed by real-time PCR. Protein expression of NF-κB, PPARγ2, and phosphorylated IκBα was evaluated by western blotting, and the histology of adipose tissue was examined. Supplementation of the rat diet with 5-CQA reduced obesity development, macrophage infiltration, and steatosis. Additionally, 5-CQA decreased the expression of NF-κB and downstream inflammatory cytokines, but increased the expression of PPARγ2, in a dose-dependent manner. Thus, 5-CQA improved obesity and obesity-related metabolic disturbances via PPARγ2 and the NF-κB signaling pathway. © The Royal Society of Chemistry.

为了揭示根区温度对烤烟烟碱积累的影响机制,通过盆栽试验研究了根区温度(10,18,26和35℃)与氮素形态(100%NH_4~+-N,50%NH_4~+-N+50%NO_3~--N和100%NO_3~--N)互作对烤烟生物量和烟碱积累的影响。结果表明:适当提高根区温度可增加烟株生物量,NO_3~--N比NH_4~+-N更有利于烟株的生长。当根区温度为35℃时,供应100%NH_4~+-N和50%NH_4~+-N+50%NO_3~--N的烟株生长受到抑制,而供应100%NO_3~--N的烟株生长旺盛。氮素形态对烟株生物量的影响大于根区温度。烟株的烟碱积累量在根区温度为18℃时达到最大,供应100%NO_3~--N的烟株烟碱积累量是供应100%NH_4~+-N烟株的1.64倍,施用NO_3~--N有利于烟碱的合成。当根区温度为35℃时,烟株的烟碱合成受到抑制。根区温度对烟碱积累的影响大于氮素形态。

摘要:
采用湖南桂阳和桑植烟区1981-2010年气温和降水量观测资料,以线性趋势函数和Mann-Kendall突变检测方法分析不同烟区烤烟生育期气温和降水变化特征。结果表明:(1)桂阳烟区各生育期平均气温呈上升趋势,尤其是苗期和伸根期上升速率最为明显,分别为0.863℃/10a和0.596℃/10a,并且在烤烟苗期、伸根期和旺长期气温都出现了突变,突变均发生在20世纪90年代和21世纪初期,均是由寒转暖的趋势;桑植烟区各生育期平均气温总体高于桂阳烟区,其中苗期上升速率最快,为0.547℃/10a,并且20世纪90年代苗期出现了突变,表明桂阳烟区烤烟苗期低温是影响烟苗培育的主要气候障碍因素。(2)桂阳烟区各生育期降水量变化趋势呈现伸根期下降、成熟期上升趋势,并且在20世纪90年代伸根期和成熟期出现突变点。桑植烟区各生育期平均年降水量比桂阳低,年度间变化小,并且不存在突变点,表明桑植烟区烤烟大田生长期的降水量是影响其烟叶产质量的主要气候障碍因素。

采用盆栽试验,研究了根部不同温度和不同氮素形态的互作对于烤烟生长和钾素积累的影响。结果表明:提高根部温度和供应NO_3-N比例,能够显著促进烟株的生长,根系活力和钾素积累增加。根部温度为10℃和18℃时,供应的NH_4-N烟株根系对钾素的吸收能力较弱;当根部温度为35℃时,供应100%NO_3-N的烟株生长旺盛,但供应NH_4-N的烟株生长受到强烈抑制,根部生物量下降,并有大量的钾素外泌。通过分析可知,氮素形态对烟株生长和钾素积累的影响大于根部温度。提高根部温度和供应硝态氮有利于烟株的生长和钾素的吸收;当根部温度较高时,氨态氮不利于烟株的生长。

The effects of root oxidization ability and P fertilization on As mobility in soils, and subsequently As uptake, translocation and speciation in rice plants were investigated. Results show that root oxidation significantly influences As mobility in rhizoshphere. Genotype TD71 with higher radial oxygen loss (ROL) induces more Fe plaque formation and sequesters more As and P in iron plaque and rhizoshphere soil, leading to the reduction of As accumulation in rice plants. Additionally, P addition mobilizes As in soil solution, and increases As accumulation in rice plants. Arsenic speciation results show that the majority of As species in husks detected is inorganic As, accounting for 82%–93% of the total As, while in grains the majority of As is inorganic As and dimethyl arsenic (DMA), with DMA accounting for 33%–64% of the total As. The fraction of inorganic As decreases while fraction of DMA increases, with increasing As and P concentrations. The study further elucidates the mechanisms involved in effects of ROL on As tolerance and accumulation in rice.

本试验采用顶空固相萃取结合气质联用的技术检测由米曲霉、黑曲霉、根霉、毛霉4种不同单一菌种发酵的蚕豆酱在后发酵15d和40d两个时期酱醅中的香气成分。数据表明各菌种发酵的蚕豆酱在后发酵15d至40d酱醅中的挥发性成分变化较大,主要特征风味物质种类不断丰富。在后发酵前期阶段蚕豆酱中醇类和酸类物质占主要地位,随着发酵时间的延长,蚕豆酱中的醛类、酯类和杂环类物质含量会上升,而酮类及酸类物质减小。由米曲霉、黑曲霉、根霉及毛霉纯种发酵制作的4种酱中主要的特征风味物质有2,3-丁二醇、3-甲基丁醇、2-甲基丁醇、芳樟醇、苯乙醇、苯甲醛、苯乙醛、琥珀酸-3,4-二甲基苯基庚基酯、三甲基吡嗪等,且由米曲霉发酵的蚕豆酱风味物质较丰富,该菌种是制作蚕豆酱的良好发酵剂。

In this study, steam explosion pretreatment was performed in cotton stalks, leading to 5-6 folds enhancements on biomass enzymatic saccharification distinctive in Gossypium barbadense and Gossypium hirsutum species. Sequential 1% H2SO4 pretreatment could further increase biomass digestibility of the steam-exploded stalks, and also cause the highest sugar-ethanol conversion rates probably by releasing less inhibitor to yeast fermentation. By comparison, extremely high concentration alkali (16% NaOH) pretreatment with raw stalks resulted in the highest hexoses yields, but it had the lowest sugar-ethanol conversion rates. Characterization of wall polymer features indicated that biomass saccharification was enhanced with steam explosion by largely reducing cellulose DP and extracting hemicelluloses. It also showed that cellulose crystallinity and arabinose substitution degree of xylans were the major factors on biomass digestibility in cotton stalks. Hence, this study has provided the insights into cell wall modification and biomass process technology in cotton stalks and beyond. © 2015 Elsevier Ltd.

Variable (wavelength or feature) selection techniques have become a critical step for the analysis of datasets with high number of variables and relatively few samples. In this study, a novel variable selection strategy, variable combination population analysis (VCPA), was proposed. This strategy consists of two crucial procedures. First, the exponentially decreasing function (EDF), which is the simple and effective principle of 'survival of the fittest' from Darwin's natural evolution theory, is employed to determine the number of variables to keep and continuously shrink the variable space. Second, in each EDF run, binary matrix sampling (BMS) strategy that gives each variable the same chance to be selected and generates different variable combinations, is used to produce a population of subsets to construct a population of sub-models. Then, model population analysis (MPA) is employed to find the variable subsets with the lower root mean squares error of cross validation (RMSECV). The frequency of each variable appearing in the best 10% sub-models is computed. The higher the frequency is, the more important the variable is. The performance of the proposed procedure was investigated using three real NIR datasets. The results indicate that VCPA is a good variable selection strategy when compared with four high performing variable selection methods: genetic algorithm-partial least squares (GA-PLS), Monte Carlo uninformative variable elimination by PLS (MC-UVE-PLS), competitive adaptive reweighted sampling (CARS) and iteratively retains informative variables (IRIV). Inorganic nanomaterials have been increasingly utilized in many consumer products, which has led to concerns about their potential toxicity. At present, there is limited knowledge about the gastrointestinal fate and cytotoxicity of ingested inorganic nanoparticles. This study determined the influence of particle size and concentration of gold nanoparticles (AuNPs) on their absorption, accumulation, and cytotoxicity in model intestinal epithelial cells. As the mean particle diameter of the AuNPs decreased (from 100 to 50 to 15 nm), their rate of absorption by the intestinal epithelium cells increased, but their cellular accumulation in the epithelial cells decreased. Moreover, accumulation of AuNPs caused cytotoxicity in the intestinal epithelial cells, which was evidenced by depolarization of mitochondria membranes. These results provide important insights into the relationship between the dimensions of AuNPs and their gastrointestinal uptake and potential cytotoxicity. ©2015 American Chemical Society.

Plants have evolved exquisite molecular mechanisms to adapt to diverse abiotic stresses. MicroRNAs play an important role in stress response in plants. However, whether the other small RNAs (sRNAs) possess stress-related roles remains elusive. In this study, thousands of sRNAs responsive to cold, drought and salt stresses were identified in rice seedlings and panicles by using high-throughput sequencing data. These sRNAs were classified into 12 categories, including "Panicle-Cold-Down", "Panicle-Cold-Up", "Panicle-Drought-Down", "Panicle-Drought-Up", "Panicle-Salt-Down", "Panicle-Salt-Up", "Seedling-Cold-Down", "Seedling-Cold-Up", "Seedling-Drought-Down", "Seedling-Drought-Up", "Seedling-Salt-Down" and "Seedling-Salt-Up". The stress-responsive sRNAs enriched in Argonaute 1 were extracted for target prediction and degradome sequencing data-based validation, which enabled network construction. Within certain subnetworks, some target genes were further supported by microarray data. Literature mining indicated that certain targets were potentially involved in stress response. These results demonstrate that the established networks are biologically meaningful. We discovered that in some cases, one sRNA sequence could be assigned to two or more categories. Moreover, within certain target-centered subnetworks, one transcript was regulated by several stress-responsive sRNAs assigned to different categories. It implies that these subnetworks are potentially implicated in stress signal crosstalk. Together, our results could advance the current understanding of the biological role of plant sRNAs in stress signaling. © 2015 Elsevier Ltd. All rights reserved.

In this study, bioleaching was coupled with electrokinetics (BE) to remove heavy metals (Cu, Zn, Cr and Pb) from contaminated soil. For comparison, bioleaching (BL), electrokinetics (EK), and the chemical extraction method were also applied alone to remove the metals. The results showed that the BE method removed more heavy metals from the contaminated soil than the BL method or the EK method alone. The BE method was able to achieve metal solubilization rates of more than 70 % for Cu, Zn and Cr and of more than 40 % for Pb. Within the range of low current densities (<1 mA cm<sup>-2</sup>), higher current density led to more metal removal. However, the metal solubilization rates did not increase with increasing current density when the current density was higher than 1 mA cm<sup>-2</sup>. Therefore, it is suggested that bioleaching coupled with electrokinetics can effectively remediate heavy metal-contaminated soils and that preliminary tests should be conducted before field operation to detect the lowest current density for the greatest metal removal. ©2015 Springer Science+Business Media New York.

A method for rapid quantitative analysis of epimedin A, B, C and icariin in Epimedium was developed based on Fourier transform near infrared (FT-NIR) spectroscopy, and by adopting high performance liquid chromatography-diode array detection (HPLC-DAD) as the reference method. Multivariate calibrations models were built by partial least squares regression (PLSR) based on the full absorbance spectra (10 000-4000 cm<sup>-1</sup>) or only the most informative key variables selected by the competitive adaptive reweighted sampling (CARS) method. In comparison, the accuracy of the CARS-PLSR method was apparently higher than full spectrum-PLSR for four kinds of investigated flavonoids. For CARS-PLSR, the coefficients of determination (R<sup>2</sup>) for prediction were 0.8969, 0.8810, 0.9273 and 0.9325 and the root mean square errors of prediction (RMSEP) were 0.1789, 0.2572, 1.2872 and 0.3615 for epimedin A, B, C and icariin, respectively. The good performance indicates that the combination of NIR spectroscopy with CARS-PLSR is an effective method for determination of epimedin A, B, C and icariin in Epimedium with fast, economic and nondestructive advantages compared to traditional chemical methods. ©The Royal Society of Chemistry 2015.

Plant architecture, a complex of the important agronomic traits that determine grain yield, is a primary target of artificial selection of rice domestication and improvement. Some important genes affecting plant architecture and grain yield have been isolated and characterized in recent decades; however, their underlying mechanism remains to be elucidated. Here, we report genetic identification and functional analysis of the PLANT ARCHITECTURE AND YIELD 1 (PAY1) gene in rice, which affects plant architecture and grain yield in rice. Transgenic plants over-expressing PAY1 had twice the number of grains per panicle and consequently produced nearly 38% more grain yield per plant than control plants. Mechanistically, PAY1 could improve plant architecture via affecting polar auxin transport activity and altering endogenous indole-3-acetic acid distribution. Furthermore, introgression of PAY1 into elite rice cultivars, using marker-assisted background selection, dramatically increased grain yield compared with the recipient parents. Overall, these results demonstrated that PAY1 could be a new beneficial genetic resource for shaping ideal plant architecture and breeding high-yielding rice varieties.
