摘要:
在前段120℃后段90℃(120&90℃),110&80℃和100&70℃不同热风温度组合的变温干燥条件下,考察了烟丝含水率、温度等变化规律,并与恒温干燥结果进行了对比分析.结果表明:①变温干燥时,烟丝含水率及干燥速率变化曲线在变温点之前与高温热风条件下的恒温干燥曲线一致,在变温点之后则逐渐接近低温热风条件下的恒温干燥曲线,Midilli模型可较好对变温干燥动力学进行描述;②变温干燥时,烟丝干燥终点温度显著低于高温热风条件下恒温干燥终点温度;对不同变温干燥条件,在变温点附近均存在一烟丝表面温度相对稳定的脱水过程;③不同变温干燥条件下,烟丝填充值介于前后段温度热风恒温干燥之间.前段100℃后段70℃的变温干燥试验条件下,烟丝填充值则相对接近于100℃热风恒温干燥结果.

摘要:
Effects of inhibitors on uptake and assimilation of ferricyanide by different plants were investigated. Detached roots of plants were kept in a closed-dark hydroponic system with ferricyanide solution amended with various inhibitors. Dissociation of ferricyanide to free cyanide and iron in solution was negligible. The application of inhibitors affected both botanical assimilation and uptake of ferricyanide. Of the inhibitors tested, silver nitrate showed a significantly inhibitory effect on ferricyanide uptake by rice, soybean and maize (P < 0.01), while a negligible effect was found in willows spiked with the same inhibitor (P > 0.05). However, lanthanum chloride showed the most severe effect on botanical assimilation of ferricyanide by maize and rice (P < 0.01), whereas silver nitrate and tetraethylammonium chloride were the most sensitive inhibitors to soybean and willows, respectively (P < 0.01). Botanical assimilation of ferricyanide was observed positively in responses to temperatures, in which maize was more susceptible than other selected plants. In conclusion, application of inhibitors has a substantial influence on the uptake and assimilation of ferricyanide by plants, and the inhibitory efficiency is highly dependent on the species of inhibitors applied.

摘要:
Animal venoms contain a fascinating array of divergent peptide toxins that have cross-activities on different types of voltage-gated ion channels. However, the underlying mechanism remains poorly understood. Jingzhaotoxin-III (JZTX-III), a 36-residue peptide from the tarantula Chilobrachys jingzhao, is specific for Nav1.5 and Kv2.1 channels over the majority of other ion channel subtypes. JZTX-III traps the Nav1.5 DII voltage sensor at closed state by binding to the DIIS3-S4 linker. In this study, electrophysiological experiments showed that JZTX-III had no effect on five voltage-gated potassium channel subtypes (Kv1.4, Kv3.1, and Kv4.1-4.3), whereas it significantly inhibited Kv2.1 with an IC50 of 0.71 ± 0.01 μM. Mutagenesis and modeling data suggested that JZTX-III docks at the Kv2.1 voltage-sensor paddle. Alanine replacement of Phe274, Lys280, Ser281, Leu283, Gln284, and Val288 could decrease JZTX-III affinity by 7-, 9-, 34-, 12-, 9-, and 7-fold, respectively. Among them, S281 is the most crucial determinant, and the substitution with Thr only slightly reduced toxin sensitivity. In contrast, a single conversion of Ser281 to Ala, Phe, Ile, Val, or Glu increased the IC50 value by >34-fold. Alanine-scanning mutagenesis experiments indicated that the functional surface of JZTX-III bound to the Kv2.1 channel is composed of four hydrophobic residues (Trp8, Trp28, Trp30, and Val33) and three charged residues (Arg13, Lys15, and Glu34). The bioactive surfaces of JZTX-III interacting with Kv2.1 and Nav1.5 are only partially overlapping. These results strongly supported the hypothesis that animal toxins might use partially overlapping bioactive surfaces to target the voltage-sensor paddles of two different types of ion channels. Increasing our understanding of the molecular mechanisms of toxins interacting with voltage-gated sodium and potassium channels may provide new molecular insights into the design of more potent ion channel inhibitors. © 2013 American Chemical Society.

Soybean is one of most important dicotyledonous food crops and is widely planted in Hunan Province, China. However, mining activity causes contamination of the soil in which soybean grows. To assess the impact of mining-induced soil contamination on soybean plants, a geoaccumulation index (I<inf>geo</inf>) was used to evaluate 20 soil samples from the mining and smelting areas of southern Hunan Province. The results indicated that Zn ranged from uncontaminated to a moderately contaminated level (I<inf>geo</inf><1), Pb was at a strongly contaminated level (I<inf>geo</inf>>3), and Cd was at an extremely contaminated level (I<inf>geo</inf>>5) across the whole study area. All of the studied soybean plants were affected by heavy metal Pb and Cd contamination, and the mean concentrations in seeds were 13.9mg/kg and 2.95mg/kg, respectively. The estimated bioconcentration factor and translocation factor showed that the soybean roots had a strong Cd bioconcentration capability and the stems had a strong translocation capability in terms of Pb, Cd, and Zn, with preferential transference of metals to the soybean leaves. The bioavailable fraction in the soil was characterized by the exchangeable fraction of heavy metals. In the present study, the bioavailable fractions of Pb, Cd, and Zn were significantly positively correlated with the concentration of these metals in soybean tissues (roots, stems, leaves, husks, and seeds). ©2013 SETAC.

摘要:
为了考察湘南烟区气象因子对烤烟烟叶理化指标的影响,收集了湘南烟区30年主要气象资料,并与烟叶理化指标进行了描述性统计和相关性分析.结果表明:①湘南烟区3月份气温、日照时数的变异系数均高于其余月份,并且月活动积温最低,30年中3月日平均气温低于13 ℃的年份占71.2%;②4~7月活动积温为2981.40℃,有利于烟草大田生育期生长,但30年中7月平均气温超过35℃的年份占18.0%,易造成“高温逼熟”或高烟碱,进而影响烟叶品质;③3月和6月降水量、日照时数、蒸发条件与烤烟C3F单叶重、含梗率、含水率的相关性较显著.④湘南烟区气象因子与C3F等级烟叶化学指标的显著相关性(P<0.05)高于物理指标,4个主要气象因子中降水量和日照时数与化学指标的相关性更加显著(P<0.05).

为揭示植物激素调控烟草腋芽萌发及生长的生理机制,以烟草K326(腋芽生长势强品种)和云烟87(腋芽生长势弱品种)为材料,测定和分析了打顶期、打顶后5,10,15,20和25 d的烟株上第1、第3腋芽(从上至下数)长度及腋芽内生长素(IAA)、脱落酸(ABA)、赤霉素(GA3)等植物激素含量的变化。结果表明:烟株顶的存在严重影响着腋芽的萌发与生长,打顶可以促进腋芽的萌发和伸长,且越靠近顶部的腋芽,萌发能力和生长势越强。打顶后上部腋芽较下部腋芽先萌发,且K326品种的腋芽长度在各个时期均显著高于云烟87;打顶后烟株腋芽内IAA,ABA和GA3含量显著低于打顶期,总体上呈现出先减少再增加的趋势。打顶后顶端优势削除,腋芽部位IAA和GA3含量迅速减少,从而促进了腋芽的萌发与生长。

摘要:
Based on the chemical properties, sixteen combinations are obtained. By allocating these combinations to sixteen different two-component vectors respectively in the Cartesian coordinate system, we present a novel 2-D graphical representation of DNA primary sequences, which can offer more biology information. This approach is not any circuit and has one to one correspondence between DNA sequences and DNA graphs. The examination of similarities/ dissimilarities among the coding sequences of the first exon of --globin gene of different species illustrates the utility of the approach. ©2013 American Scientific Publishers. 摘要:
In this study, total 80 typical Miscanthus accessions were examined with diverse lignocellulose features, including cellulose crystallinity (CrI), degree of polymerization (DP), and mole number (MN). Correlation analysis revealed that the crude cellulose CrI and MN, as well as crystalline cellulose DP, displayed significantly negative influence on biomass enzymatic digestibility under pretreatments with NaOH or H2SO4 at three concentrations. By contrast, the comparative analysis of two Miscanthus samples with similar cellulose contents showed that crude cellulose DP and crystalline cellulose MN were positive factors on biomass saccharification, indicating cross effects among the cellulose levels and the three cellulose features. The results can provide insights into mechanism of the lignocellulose enzymatic digestion, and also suggest potential approaches for genetic engineering of bioenergy crops. © 2012 Elsevier Ltd.

摘要:
The objective of this study is to investigate the characteristics and influencing factors of phosphorus leaching. The processes of phosphorus leaching of four typical soils (red soil, alluvial soil and purple soil) in Hunan are studied through the simulated leaching method by adding CaCl<inf>2</inf> solution with phosphorus of different concentration. The phosphorus concentration in the leaching liquid increase as phosphorus concentration in the eluent increased. The process of phosphorus leaching is carried out in stages. There is a critical point on every curve;the critical values of phosphorus leaching in 6 soils fluctuate between 9.854-65.372 mg·kg<sup>-1</sup>. A big difference of critical values appears among different soils. There is a significantly negative correlation between the critical value of phosphorus leaching and Olsen-P, Fe-P, Al-P, the correlation coefficients are -0.950, -0.834 and -0.897 respectively. The characteristics of phosphorus leaching in different soils varies greatly, different evaluation standard of phosphorus leaching and agricultural management measures should be put forward in view of the different soil in order to reduce phosphorus leaching. ©2012 IEEE.

摘要:
Xylans are the major hemicelluloses in grasses, but their effects on biomass saccharification remain unclear. In this study, we examined the 79 representative Miscanthus accessions that displayed a diverse cell wall composition and varied biomass digestibility. Correlation analysis showed that hemicelluloses level has a strong positive effect on lignocellulose enzymatic digestion after NaOH or H2SO4 pretreatment. Characterization of the monosaccharide compositions in the KOH-extractable and non-KOH-extractable hemicelluloses indicated that arabinose substitution degree of xylan is the key factor that positively affects biomass saccharification. The xylose/arabinose ratio after individual enzyme digestion revealed that the arabinose in xylan is partially associated with cellulose in the amorphous regions, which negatively affects cellulose crystallinity for high biomass digestibility. The results provide insights into the mechanism of lignocellulose enzymatic digestion upon pretreatment, and also suggest a goal for the genetic modification of hemicelluloses towards the bioenergy crop breeding of Miscanthus and grasses. © 2012 Elsevier Ltd.

摘要:
Near infrared spectroscopy (NIRS) has been broadly applied as a quick assay for biological component and property analysis. However, NIRS remains unavailable for in-depth analysis of biomass digestibility in plants. In this study, NIRS was used to determine biomass enzymatic digestibility using 199 Miscanthus samples, which represents a rich germplasm resource and provides for a stable calibration model. The intensive evaluation indicates that the calibration and validation sets are comparable. Using the modified partial least squares method, seven optimal equations were generated with high determination coefficient on calibration (R2) at 0.75-0.89, cross-validation (R2cv) at 0.69-0.87, and the ratio performance deviation (RPD) at 1.80-2.74, which provide multiple options for NIRS prediction of biomass digestibility under different pretreatments. As biomass digestibility is a crucial parameter for biofuel processing, NIRS is a powerful tool for the high-throughput screening of biomass samples in plants. © 2012 Elsevier Ltd.

摘要:
Biosurfactant-based reversed micellar extraction (RME) is an innovative method for separation and purification of biomolecules. In this study, rhamnolipid (RL), a kind of biosurfactant, was firstly adopted to form a novel reversed micellar system for extracting and purifying laccase from Coriolus versicolor crude extract. Several significant factors affecting both forward and backward extraction processes were studied. The appropriate conditions for forward extraction process were: 3.3 mM RL, 50 mM KCl, pH 5.5 and extracting time 40 min. As regards backward extraction process, 250 mM KCl, pH 7.0 and extracting time 40 min were suggested. The corresponding activity recovery (AR) and purification fold (PF) were 92.7% and 4.79, respectively. Electrophoresis analysis indicated that the laccase was successfully purified. After this reversed micellar system was reused three times, the AR and PF declined to 70.8% and 4.35, respectively, indicating that the reversed micellar system could be reused. Comparisons results of synthetic surfactant-based RME and RL-based RME further verified the superiority of RL. ©2012 Elsevier Ltd. All rights reserved.

摘要:
Background: Lignocellulose is the most abundant biomass on earth. However, biomass recalcitrance has become amajor factor affecting biofuel production. Although cellulose crystallinity significantly influences biomasssaccharification, little is known about the impact of three major wall polymers on cellulose crystallization. In thisstudy, we selected six typical pairs of Miscanthus samples that presented different cell wall compositions, and thencompared their cellulose crystallinity and biomass digestibility after various chemical pretreatments.Results: A Miscanthus sample with a high hemicelluloses level was determined to have a relatively low cellulosecrystallinity index (CrI) and enhanced biomass digestibility at similar rates after pretreatments of NaOH and H2SO4with three concentrations. By contrast, a Miscanthus sample with a high cellulose or lignin level showed increasedCrI and low biomass saccharification, particularly after H2SO4 pretreatment. Correlation analysis revealed that thecellulose CrI negatively affected biomass digestion. Increased hemicelluloses level by 25% or decreased celluloseand lignin contents by 31% and 37% were also found to result in increased hexose yields by 1.3-times to 2.2-timesreleased from enzymatic hydrolysis after NaOH or H2SO4 pretreatments. The findings indicated that hemicelluloseswere the dominant and positive factor, whereas cellulose and lignin had synergistic and negative effects onbiomass digestibility.Conclusions: Using six pairs of Miscanthus samples with different cell wall compositions, hemicelluloses wererevealed to be the dominant factor that positively determined biomass digestibility after pretreatments with NaOHor H2SO4 by negatively affecting cellulose crystallinity. Ancient pyrometallurgical zinc generated large amounts of slags. Natural leaching from ancient pyrometallurgical zinc sites has contaminated the soil closed to the deposits. Electrokinetic removal technology was adopted to treat this slags. But the generation of metal hydroxide precipitation near the cathode during the electrokinetic removal process could be the most serious factor that influenced the removal rate. In this study, cation exchange membrane was used in electrokinetic removal process to overcome this major drawbacks. The final results showed that cation exchange membrane could effectively condition pH of the whole electrokinetic removal system resulted in the dissolution of the precipitated heavy metals near the cathode. In other words, the enhanced treatment have achieved positive effect to some extent. But the leak tightness of experiment setup might not be perfect. Moreover,some operation parameters of the electrokinetic removal technique must be further optimized.

摘要:
为解决现有往复式切割器收割龙须草的问题,拟对龙须草茎秆进行切割间隙、切割速度、切割刀片组合形式、切割茎秆部位、切割茎秆数量等因素进行了单因素试验,在单因素试验的基础上选取了正交试验的因素水平,完成了茎秆最大剪切力的正交试验,实验结果表明切割间隙和切割刀片组合形式对剪切力影响较大,切割速度对剪切力影响相对较小。最后通过选取斜齿刀-斜光刀组合模式与斜光刀-斜光刀传统刀片组合进行田间试验对比,结果表明斜齿刀-斜光刀组合的切断率提高了23%,刀片缠草率降低了94%,极大地提高了龙须草切割作业质量和作业效率。研究结果表明斜齿刀-斜光刀组合刀片用于龙须草收割是可行的。

摘要:
The study researched on the biochemical methane potential (BMP) of banana stalks by anaerobic digestion which was carried out by automatic methane potential test system (AMPTS). The results showed that after 42 days of anerobic digestion at the mesophilic condition (37°C), BMP of 20 mm lengths' banana stalks was 241.2 ml·g<sup>1</sup> (standard temperature and pressure) VS, and BMP of 5 mm rose to 247.7 ml·g<sup>-1</sup> (standard temperature and pressure) VS, which has not significant difference. However, the smaller size of banana stalks the faster speed of digestion and biogas production rate. Banana stalks of 5 mm would get 90% of biogas yield only after 14 days' anaerobic digestion, which shorted 7 days compared with the reference group, and the average daily gas production increased by 54.4%. The statistics also show that the result of BMP test carried out by AMPTS has high reproducibility and repeatability, which means AMPTS is a reliable equipment for BMP test.

摘要:
针对混合交通特征表达和分类识别的问题,提出了基于边缘偏心率向量的混合交通视频检测算法。将利用上下文比对获取的边缘信息与图像重心相结合构建混合交通的边缘偏心率向量,对混合交通前景进行了特征表达。再结合极限学习机建立了快速学习机制,实现了快速分类识别,克服了采用支持向量机训练难以达到实时检测的问题。试验结果表明:本文算法中各个类别的混合交通边缘偏心率特征区分明显,识别准确率可达93%以上,且处理速度快,能够满足实时检测的需求。

Ammonia volatilization and the distribution of Cu and Zn were investigated in two types of soil treated with coated and uncoated urea. The rate of ammonia volatilization in two weeks after fertilizing with coated urea was 8% in soil 1 (soil derived from river alluvial deposits in Dongting Lake Plain) and 5.15% in soil 2 (red soil derived from quaternary red clay), about half the rates observed when fertilizing with common urea, implying that the hydrolysis speed of the coated urea was lower than for common urea, and that the coated urea can increase nitrogen use efficacy. As for the availability of Cu and Zn, their concentrations decreased in the first week after fertilization, and then increased, which was contrary to the effect of treatment on soil pH. For example, when the pH was 7.99, there was 0.79 mg/kg exchangeable Cu and 0.85 mg/kg exchangeable Zn in the soil derived from river alluvial deposits in Dongting Lake Plain. However, the concentrations of exchangeable Cu and Zn were generally lower for the common urea treatments than those with the coated urea because the peak pH for the common urea treatment was greater. The concentrations of these elements correlated well with pH in the range 4-8 in second order polynomial fits.
