摘要:
昆虫作为通过可变剪切调控性别决定的生物,拥有着保守性与多样性并存的性别决定机制。快速进化的上游性别决定初始信号使得几乎共享相同基因组的两性个体在形态、生理、行为等方面上有着巨大差异,而下游的双性基因doublesex在结构和功能上高度保守。本文综述了昆虫性别决定调控机制中初始信号的作用机理,概括了下游信号通路的响应机制,介绍了性别决定下游保守基因doublesex,以期为深入理解昆虫性别决定机制和农业害虫防治提供新思路。

6种杀菌剂对油菜菌核病防治效果研究(英文)

摘要:
为了筛选出安全高效杀菌剂防治油菜菌核病,以 6 种不同作用机理的杀菌剂为试验材料,比较其对油菜菌核病的防效.结果表明:6 种药剂对油菜均无药害;其中50%啶酰菌胺水分散粒剂、62%嘧环·咯菌清水分散粒剂和 25%吡唑醚菌酯悬浮剂的防治效果较好,防效分别为 85.6%、77.2%和70.1%;其次为 25%腐霉利水分散粒剂,防效为 64.9%;50%多菌灵和 45%咪鲜胺对油菜菌核病防效较差,仅为 38.6%和 33.3%;50%啶酰菌胺水分散粒剂的产量最高,比对照增产 53.6%,其次是 62%嘧环·咯菌清水分散粒剂和 25%吡唑醚菌酯悬浮剂.综合 6 种杀菌剂的田间防效和对产量的影响,50%啶酰菌胺水分散粒剂可作为衡阳地区油菜菌核病有效防治药剂.

摘要:
Spodoptera frugiperda (S. frugiperda) is an invasive pest that threatens global crop production and food security and poses a serious threat to maize production worldwide. Metal-organic framework (MOF) nanocarriers have great potential for agricultural pest control applications. The present study successfully prepared the chemical cross-linking of iron-based metal-organic framework nanoparticles (MIL-101(Fe)-NH(2) NPs) with sodium lignosulfonate (SL) as a pH/laccase double stimuli-responsive pesticide release system. The average particle size of the prepared chlorfenapyr (CF)-loaded nanoparticles (CF@MIL-101-SL NPs) was 161.54 nm, and the loading efficiency was 44.52%. Bioactivity assays showed that CF@MIL-101-SL NPs increased the toxicity of CF to S. frugiperda and caused the rupture of the peritrophic membrane and enlargement of the midgut. Data from 16S rRNA gene sequencing showed that CF@MIL-101-SL treatment reduced the resistance of S. frugiperda to pesticides and pathogens and affected nutrient and energy availability by remodeling the intestinal microbiota of S. frugiperda. The dysregulated microbial community interacted with the broken peritrophic membrane, which exacerbated damage to the host. Nontargeted metabolomic results showed that ABC transporters may be a potential mechanism for the enhanced toxicity of CF@MIL-101-SL to S. frugiperda. In summary, the present study provides effective strategies for toxicological studies of nanopesticides against insects.

摘要:
A chitosan-based nanoparticle was prepared using chitosan (CS) and O-carboxymethyl chitosan (O-CMCS). Our study revealed that chitosan/O-carboxymethyl chitosan/tebuconazole nanoparticles (CS/O-CMCS/TBA NPs) exhibited superior antifungal activity, foliar adhesion, and microbial target adhesion performance compared to commercial suspension concentrate (SC). The antifungal activity of CS/O-CMCS/TBA NPs against C. gloeosporioides, with a 3.13-fold increase in efficacy over TBA (SC). We also found that low concentrations of CS/O-CMCS NPs promoted the growth of C. gloeosporioides and enhanced the fungal catabolism of chitosan. Overall, the CS/O-CMCS/TBA NPs were found to possess the remarkable capability to selectively aggregate around pathogenic microorganisms and CS/O-CMCS NPs can enhance the fungal catabolism of chitosan. CS/O-CMCS/TBA NPs, as a "sugar-coated bomb", was a promising asset for effective plant disease management and pesticide utilization through the affinity of chitosan-based nanoparticles and C. gloeosporioides, enabling targeted delivery and targeted release of their encapsulated active ingredient, which was important for the development and application of biocompatible chitosan-based nanopesticides.

氯虫苯甲酰胺与甲维盐复配对玉米田草地贪夜蛾的防效

摘要:
草地贪夜蛾是一种重要的外来入侵害虫,已对我国玉米产业造成巨大的经济损失。化学农药对草地贪夜蛾的速效性好,但大量使用会导致其产生抗药性;生物农药对其速效性较差,不易产生抗药性且环境兼容性好。为此,于室内利用浸虫法测定了氯虫苯甲酰胺与甲氨基阿维菌素苯甲酸盐(简称甲维盐)复配对玉米草地贪夜蛾的联合毒力,并于2021年采用田间小区试验评价了5%氯虫苯甲酰胺悬浮剂、5%甲维盐微乳剂及其增效组合物叶面喷雾处理对玉米田草地贪夜蛾的防治效果。结果表明:氯虫苯甲酰胺与甲维盐复配剂以7∶1的质量比复配对草地贪夜蛾2龄幼虫具有最高的增效作用,其共毒系数为136%;氯虫苯甲酰胺悬浮剂与5%甲维盐微乳剂现混现用时,以40+5.5 mL/667m~2处理对玉米田草地贪夜蛾的防治效果最佳,药后1~14 d的防治效果高达87.43%~98.10%。这说明氯虫苯甲酰胺与甲维盐复配不仅可实现化学农药减量使用的目标,而且还能有效控制玉米草地贪夜蛾危害。

摘要:
Heat shock proteins are molecular chaperones that are involved in numerous normal cellular processes and stress responses, and heat shock factors are transcriptional activators of heat shock proteins. Heat shock factors and heat shock proteins are coordinated in various biological processes. The regulatory function of heat shock factors in the expression of genes encoding heat shock proteins (Hsps) has been documented in some model insects, however, the role of transcription factors in modulating Hsps in other insects is still limited. In this study, one heat shock factor gene (AhHsf) was isolated and its two potential target genes (AhHsp70 and AhsHsp21) were confirmed from Agasicles hygrophila. AhHsf sequence analysis indicated that it belongs to the Hsfs gene family. RT-qPCR showed that expression levels of heat shock factors and of two heat shock proteins significantly increased under heat stress. Injection with double-stranded Hsf RNA in freshly emerged adult beetles significantly inhibited expression of AhHsp70 and AhsHsp21, shortened the adult survival, drastically reduced egg production, and ultimately led to a decrease in fecundity. RNA interference (RNAi)-mediated suppression of AhHsp70 or AhsHsp21 expression also significantly affected expression of AhHsf. Our findings revealed a potential transcriptional function of AhHsf to regulate expression of AhHsp70 and AhsHsp21, which may play a key role in A. hygrophila thermotolerance. 摘要:
为明确饥饿情况下蠋蝽种内残杀行为的发生时期和影响因素,指导蠋蝽规模化繁育和释放,本文研究了不同发育时期和密度下的蠋蝽种内残杀行为.研究发现:蠋蝽对卵粒的种内互残行为主要发生在饥饿24 h以内,雌性蠋蝽对卵粒的取食行为显著强于雄性蠋蝽,取食比例是雄性的2.80~3.25倍、取食量是雄性的18.75~21.20倍,雌性取食下的卵粒羽化率为70.66%,显著低于雄性和对照的86.59%和87.88%.发育时期和饲养密度均显著影响蠋蝽的种内残杀行为,主要发生在饥饿后的19.63~36.00 h,并且1、2、3龄若虫没有表现出种内残杀行为,4龄以后种内残杀行为显著;低密度下蠋蝽种内互残行为不显著,但高密度会增强种内互残行为,且随密度的增加,种内互残行为显著增强.

摘要:
The predatory insect Arma chinensis (Hemiptera: Pentatomidae) is widely distributed in China, where it is also used to control many agricultural and forest pests. The chemosensory genes expressed in its antennae play crucial roles in food-seeking and mating behaviors. To better understand the olfaction of A. chinensis antennae, we identified the genes related to food-seeking and mating. Sequencing of the antennal transcriptomes of full and hungry male and female A. chinensis revealed 38 odorant-binding proteins (OBPs), 1 chemosensory protein (CSP), 1 Niemann–Pick C2 protein (NPC2), 3 odorant receptors (ORs), 12 ionotropic receptors (IRs), 2 gustatory receptors (GRs), and 3 sensory neuron membrane proteins (SNMPs). These results were used to construct phylogenetic trees. A quantitative real-time PCR (qRT-PCR) analysis showed that the relative transcript levels of AchiGR1, AchiGR2, and AchiOBP28 were higher in female than in male antennae in both full and hungry insects, but that the expression of AchiOBP13 and AchiOBP16 was higher only in full A. chinensis females. Thus, the latter genes may encode proteins involved in oviposition selection behavior. AchiGRs (1 and 2), AchiIR6, and AchiOBPs (6–8, 12, 20–22, 28, and 34) were highly expressed only in the antennae of full males, indicating the participation of these genes in mate-searching or male pheromone recognition. The expression of AchiOBP31 in the antennae of starved males, AchiOBPs (15, 18, and 29) in the antennae of starved females, and AchiOBPs (3, 4, and 24) in the antennae of starved males and females suggested that these genes encode food-seeking functions. Our identification of chemosensory genes in A. chinensis antennae and their differential expression in full and hungry insects provides the basis for further functional studies on the chemoreception system of A. chinensis and the sex hormones of predatory insects. © Copyright © 2020 Wu, Deng, Li, Xiao, Li, Teng, Xiao, Li, Zhou and Li.

Juvenile hormone (JH) plays a pivotal role in insect reproduction. The Krüppel-homolog 1 (Kr-h1) is a JH-inducible zinc finger transcription factor that has also been found to play a role in insect reproduction, however, its function varies across species. In this study, we cloned SfKr-h1 from Sogatella furcifera and investigated its role in ovarian development. The open reading frame of SfKr-h1 is 1 800bp encoding 599 amino acids. The putative amino acid sequence of SfKr-h1 contains eight putative C2H2-type zinc finger domains and is highly homologous with the Kr-h1s of other hemipteran species. Expression of SfKr-h1 peaked 96h after adult emergence and was highest in the ovary. RNA interference (RNAi) knockdown of SfKr-h1 substantially reduced the transcription of SfVg, and arrested ovarian development. These results suggest that SfKr-h1 is critical for normal ovarian development in S. furcifera. © 2019, Springer Nature B.V.

摘要:
从灰毛豆叶片中分离出内生真菌-淡紫拟青霉菌Purureocillium lilacinum (TPL04),对其发酵产物采用硅胶柱色谱、SephadexLH-20凝胶色谱、制备薄层色谱、中压柱色谱及制备液相色谱等方法进行分离纯化,共获得8个单体化合物.通过波谱学方法并结合相关文献数据比对,化合物结构鉴定为5-羟基己酸-4-内酯(1)、(R)-4-苄基-2-噁唑烷酮(2)、2'-脱氧胸苷(3)、对羟基苯甲酸甲酯(4)、光色素(5)、β-胡萝卜苷(6)、尿嘧啶核苷(7)、己六醇(8).其中化合物1、2、4是首次从淡紫拟青霉菌中分离得到,抑菌实验表明化合物4对水稻纹枯菌(Pellicularia sasakii)、辣椒疫霉菌(Phytophthora capsici)、草莓灰霉菌(Botrytis cinerea)、油菜菌核菌(Sclerotinia sclerotiorum)四种植物病原菌生长有抑制作用.

The Southern rice black-streaked dwarf virus (SRBSDV), a novel Fijivirus, poses a major threat to rice production in East Asia. Analysis of the gene expression of SRBSDV-infected rice plants may reveal the molecular basis of interactions between the virus, its vector and rice plants. Reliable reference genes are required for accurate qRT-PCR analysis. However, no reliable, valid reference genes for examining gene expression in SRBSDV-infected rice plants have so far been identified. We examined the expression of eight candidate reference genes in the leaves of SRBSDV-infected, and healthy, rice plants at different points in time after virus inoculation. We used four dedicated algorithms, geNorm, BestKeeper, NormFinder and RefFinder, to evaluate the performance of these candidate genes. The RefFinder results indicate that 18S, EF1 alpha and UBQ10 are suitable reference genes. In addition, we used these three reference genes to analyze the expression of key genes involved in the isoprenoid metabolic pathway in rice leaves after infection by SRBSDV. The results of this analysis reveal that SRBSDV may suppress the production of the rice plant volatiles that attract natural enemies of its vector Sogatella furcifera, thereby increasing the likelihood of pathogen transmission.

Reverse transcription quantitative polymerase chain reaction (PCR) has become an invaluable technique for analyzing gene expression in many insects. However, this approach requires the use of stable reference genes to normalize the data. Chlorops oryzae causes significant economic damage to rice crops throughout Asia. The lack of suitable reference genes has hindered research on the molecular mechanisms underlying many physiological processes of this species. In this study, we used quantitative real-time PCR to evaluate the expression of eight C. oryzae housekeeping genes glyceraldehyde-3-phosphate dehydrogenase (GAPDH), beta-actin (betaACT), beta-tubulin (betaTUB), Delta Elongation factor-1 (EF1delta), ribosomal protein S11 (RPS11), RPS15, C-terminal-Binding Protein (CtBP), and ribosomal protein 49 (RP49) in different developmental stages and tissues in both larvae and adults. We analyzed the data with four different software packages: geNorm, NormFinder, BestKeeper, and RefFinder and compared the results obtained with each method. The results indicate that PRS15 and RP49 can be used as stable reference genes for quantifying gene expression in different developmental stages and larval tissues. GAPDH and betaACT, which have been considered stable reference genes by previous studies, were the least stable of the candidate genes with respect to larval tissues. GAPDH was, however, the most stable reference gene for adult tissues. We verified the candidate reference genes identified and found that the expression levels of Cadherins (Cads) changed when different reference genes were used to normalize gene expression. This study provides a valuable foundation for future research on gene function, and investigating the molecular basis of physiological processes, in C. oryzae.

油茶象Curculio chinensis Chevrolat是我国特有木本油料树种--油茶Camellia spp.的专性蛀果害虫,常导致其大量落果。本文旨在明确油茶象幼体(卵+幼虫)种群动态、空间格局及其关键影响因子,以便为调查取样、长期监测和科学防控等提供理论依据。2017年在湖南常宁普通油茶林,2016年-2017年在湖南益阳小果油茶林于雌虫产卵季节进行了调查,解剖收集的果实并记录其所含幼体数。用6种聚集度指标(m*/m、I_δ、C、I、C_a、k)和Taylor幂法则、Iwao回归模型分析油茶象幼体的空间格局,并探讨其关键影响因子。调查和分析表明,雌虫于6月上、中旬开始产卵,幼体种群密度呈单峰变化,2016年和2017年高峰期分别在7月25日和7月18日。小果油茶林2016年、2017年高峰期种群密度分别为17.95±1.53和16.27±1.06幼体/样株,高于普通油茶林,分别是普通油茶林种群密度(8.16±1.23幼体/样株)的2.20倍和1.99倍,差异极显著(P<0.001)。聚集度指标分析显示,油茶象在普通油茶林均呈聚集分布,在小果油茶林呈聚集分布或随机分布。回归模型表明,该虫在两种油茶林均呈聚集分布。Blackith种群聚集均数λ>2,表明油茶象幼体聚集是由亲代雌虫本身的产卵行为与寄主果实特征综合作用的结果。给出了最适理论抽样数公式并绘制了3种抽样精度下最适抽样数随种群密度变化的曲线图。综上,初步明确了油茶象幼体在普通油茶林和小果油茶林的空间分布格局和种群动态,并探讨了其关键影响因子。试验结果可为该虫调查取样、长期监测和科学防控等提供理论依据。

随着全球贸易的加速发展,入侵物种对农林业、生态环境及人类健康的威胁日益严重。基因组学研究为阐明外来有害生物入侵的分子机制与生态适应性过程以及研发新型防控技术提供了新手段、新平台与大数据。本文综述了入侵昆虫基因组学研究的发展现状,系统总结了基因/基因家族、转座子/重复序列等基因组信息在决定昆虫入侵性中的重要作用,着力探讨了基因组学研究在助力害虫RNAi、昆虫不育技术(SIT)、化学生态防治和物理防治等防控新技术/新产品开发方面的潜力,并展望了基因组学研究应用于入侵昆虫综合防控的前景。

Phytochemical investigation of the aerial parts of Berchemiella wilsonii (Schneid.) Nakai (Rhamnaceae) led to the isolation of four flavonoids (1–4), three phenolic acids (5–7), two megastigmane derivatives (8–9) and one triterpene (10). The structures of these compounds were elucidated as taxifolin (1), (−)-epicatechin (2), quercetin 3-O-a-L-arabinopyranoside (3), vitexin (4), methyl p-hydroxycinnamat (5), 3,4-dihydroxybenzoic acid (6), 2-hydroxy-4-methoxy-3,6-dimethyl benzoic acid (7), (3S,5R,6R,7E,9S)-3,5,6,9-tetrahydroxy-7-en-megastigmane (8), (6S,9R)-roseoside (9) and lupeol (10) on the basis of NMR spectral data and comparison with literature values. These results are the first chemical constituent data of the genus Berchemiella, and the chemotaxonomic significance of these compounds is discussed. © 2019

Southern rice black-streaked dwarf virus, transmitted by the white-backed planthopper (Sogatella furcifera [Horváth]) (Hemiptera: Delphacidae) was first found in Guangdong Province, China. A previous study has demonstrated that the host plant preferences of S. furcifera are altered by infection with the virus, with virus-free S. furcifera preferring virus-infected rice plants over healthy rice plants, whereas viruliferous S. furcifera prefer uninfected plants. To test the hypothesis that odorant-binding proteins (OBPs) are involved in the preference of S. furcifera for virus-infected rice plants, we first compared the expression levels of SfurOBP2 and SfurOBP11 in virus-free and viruliferous S. furcifera. The results show that mRNA transcript of these 2 genes were significantly reduced in viruliferous S. furcifera. We then used RNAi-mediated gene silencing to confirm the function of these 2 odorant-binding proteins in host selection of S. furcifera. The results showed that silencing of the SfurOBP2 gene caused virus-free S. furcifera to no longer prefer virus-infected rice plants over uninfected rice plants, but the ability to locate host plants was maintained. These results indicate that SfurOBP2 appears to play a crucial role in the preference of S. furcifera for virus-infected rice plants. © 2019 Florida Entomological Society. All rights reserved.

Juvenile hormone esterase (JHE) is a key enzyme for insects, playing an important role in the regulation of insect growth, development, diapause and reproduction. We identified a complete putative JHE of Cydia pomonella (CpJHE-like) which is comprised of a 1 761 bp coding sequence (CDS) encoding 587 amino acid residues from the transcriptome data. The deduced protein sequence of CpJHE-like showed the highest identity of 60.44% with the Adoxophyes honmai JHE (AhJHE) and the minimal identity of 25.81% with Aedes aegypti JHE (AaJHE). CpJHE-like exhibited all the seven typical motifs of the functional JHEs and had the highly consistent tertiary structure with Manduca sexta JHE (MsJHE). Phylogenetic analysis showed that the CpJHE-like was close to two JHEs from the family Tortricidae. The CpJHE-like transcript level take a leap in the 3-day-old fifth instar larva, increased about 300-fold compared to the basal level. Tissue-specific expression profile showed that the CpJHE-like transcript was expressed mainly in the fat body. This study indicates that the CpJHE-like is the functional JHE, which may play vital roles in the development and reproduction of C. pomonella. © 2019 CAAS. Publishing services by Elsevier B.V

Plant volatiles play an important role in regulating insect behavior. Odorant binding proteins (OBPs) are involved in the first step of the olfactory signal transduction pathway and plant volatiles recognition. Sogatella furcifera is one of the most destructive pests of rice crops. Understanding the functions of S. furcifera OBPs (SfurOBPs) in the host plant location and the behavioral responses of S. furcifera to rice plant volatiles could lead to improved, more environmentally-friendly, methods for controlling this pest. We found that SfurOBP1 displayed only weak binding with all the tested volatiles. SfurOBP2, SfurOBP3 and SfurOBP11 had different binding affinities to beta-ionone. SfurOBP2 and SfurOBP11 had strong binding affinities to beta-caryophyllene (Ki = 2.23 microM) and plant alcohol (Ki = 2.98 microM), respectively. The results of Y-olfactometer experiments indicate that S. furcifera was significantly repelled by octanal and n-octane but strongly attracted by (+)-limonene, acetophenone, 2-heptanone, n-hendecane, alpha-farnesene and beta-ionone. Furthermore, the dsRNA-mediated gene silencing of SfurOBP2, SfurOBP3 and SfurOBP11 shifted the olfactory behavior of S. furcifera for beta-ionone, alpha-farnesene and plant alcohol, respectively. These results suggest that the SfurOBPs are involved in the recognition of rice plant volatiles, and several potential repellants and lures for controlling this pest.

Juvenile hormone (JH) is responsible for repressing larval metamorphosis and inducing vitellogenesis and egg production in insects. Methoprene-tolerant (Met) is known to be an intracellular receptor and transducer of JH. We examined the role of Met in ovarian development in the rice pest Sogatella furcifera (Horváth). We first cloned and sequenced S. furcifera Met (SfMet). The SfMet protein belongs to the basic helix-loop-helix/Per-Arnt-Sim (bHLH-PAS) family with a bHLH domain and two PAS domains (PAS-A and PAS-B). SfMet was expressed in all developmental stages and tissues but was most highly expressed in the ovaries of adult females. Furthermore, RNA interference (RNAi) mediated silencing of SfMet substantially reduced the expression of SfVg, decreased yolk protein deposition and blocked oocyte maturation and ovarian development. These results demonstrate that SfMet plays a key role in female reproduction in S. furcifera and suggest that targeting this gene could be an effective way of controlling this pest. © The Author(s) 2019.
