摘要:
The various post-translational modifications (PTMs) of plant proteins have important regulatory roles in development. We therefore examined various modified proteins from strawberry stigmata and found that succinylation of lysine residues was the most abundant type of modification. We then subjected proteins from strawberry stigmata to an efficient enrichment method for succinylated peptides and identified 200 uniquely succinylated lysines (Suks) in 116 proteins. A bioinformatics analysis revealed that these proteins are involved in important biological processes, including stress responses, vesicular transport, and energy metabolism. Proteomics, combined with immunoprecipitation and immunoblotting, revealed an obvious increase in succinylation of the assembly polypeptide 2 (AP2) and clathrin from 0.5 to 2 h after pollination, suggesting that succinylation is involved in the recognition of pollen-stigma signaling substances and vesicular transport. These results suggest that AP2/clathrin-mediated vesicular transport processes are regulated by lysine succinylation during pollen recognition. Copyright © 2018 American Chemical Society.

摘要:
Fumarylacetoacetate hydrolase participates in positive regulation of salt stress in Arabidopsis. Fumarylacetoacetate hydrolase (FAH) catalyzes the hydrolysis of fumarylacetoacetate into fumarate and acetoacetate, the final step in the Tyr degradation pathway that is essential to animals. However, the Tyr degradation pathway is not well understood in plants. Previously, we found that mutation of the SHORT-DAY SENSITIVE CELL DEATH 1 (SSCD1) gene encoding FAH in Arabidopsis causes spontaneous cell death under short day, which first indicated that the Tyr degradation pathway also plays an important role in plants. In this study, we found that the SSCD1 gene was up-regulated by salt stress, and the sscd1 mutant was hypersensitive to salt stress. However, the double mutant of SSCD1 and HOMOGENTISATE DIOXYGENASE, in which intermediates of the Tyr degradation pathway could not be produced, displayed a normal response to salt stress. Furthermore, the sscd1 mutant showed more accumulation of reactive oxygen species (ROS) and less up-regulation of some ROS-scavenging genes such as ASCORBATE PEROXIDASE 2 and COPPER/ZINC SUPEROXIDE DISMUTASE 1 compared with wild type under salt stress. In addition, SSCD1 expression was also up-regulated by H2O2, and the sscd1 mutant exhibited hypersensitivity to oxidative stress compared with wild type. Taken together, we concluded that loss of FAH in sscd1 leads to the accumulation of Tyr degradation intermediates, which impairs the up-regulation of some ROS-scavenging genes under salt stress, causing more accumulation of ROS, resulting in the hypersensitivity of sscd1 to salt stress.

We report the molecular attributes of a novel bisegmented double-stranded RNA (dsRNA) virus, designated “Nigrospora oryzae partitivirus 1” (NoPV1), from a phytopathogenic fungus Nigrospora oryzae. The genome of NoPV1 contains two dsRNA segments (dsRNA1 and 2), 1875 bp and 1601 bp in length, respectively. dsRNA1 and -2 both have a single open reading frame encoding the RNA-dependent RNA polymerase and coat protein, respectively. NoPV1 has a high degree of sequence identity to members of genus Gammapartitivirus in the family Partitiviridae. This is the first report of a mycovirus in the family Partitiviridae that infects N. oryzae. © 2018, Springer-Verlag GmbH Austria, part of Springer Nature.

The Clinterocera jucunda species group of the genus Clinterocera Motschulsky, 1858 is revised. Fourteen species are recognized within this group, including five new species described herein: C. brevifasciata Xu & Qiu, new species from northern Vietnam and China; C. krikkeni Xu & Qiu, new species from southwestern China; C. sinensis Xu & Qiu, new species from southern China; C. velutina Xu & Qiu, new species from Hainan, China; and C. vietnamensis Xu & Qiu, new species from central Vietnam and Laos. Clinterocera nigra (Kano, 1931) is elevated to the species level. Clinterocera donckieri (Bourgoin, 1924) revised status is revalidated from the synonymy with C. davidis (Fairmaire, 1878). Two new synonyms are proposed based on examination of types and additional specimens: C. humeralis (Moser, 1902) new synonym and C. cervenkai Krajčík, 2012 new synonym are placed as junior synonyms of C. jucunda (Westwood, 1873) and C. donckieri, respectively. Lectotypes are designated for C. davidis and C. humeralis. Diagnoses and colored illustrations of most species are provided, including other four rare species, C. bicolor (Nonfried, 1893), C. ishikawai (Kurosawa, 1973), C. raui (Paulian, 1961), and C. yunnana (Moser, 1911). A key and a detailed description for the species group are presented. Male genitalia, intraspecific variations, new distribution records, and natural history are given for most species. The Bemisia tabaci (Gennadius) (Hemiptera:Aleyrodidae) species complex includes invasive and destructive pests of field crops, and the sibling species MEAM1 and MED are its two most damaging members. Previous research indicated that the replacement of Middle East-Minor Asia 1 (MEAM1) by Mediterranean (MED) as the dominant B. tabaci species in China can be mainly attributed to MED's greater tolerance to insecticides. Glutathione S-transferases (GSTs) play important roles in the detoxification of hydrophobic toxic compounds. To increase our understanding of differences in insecticide resistance between B. tabaci MEAM1 and MED, we searched the genomic and transcriptomic databases and identified 23 putative GSTs in both B. tabaci MEAM1 and MED. Through measuring mRNA levels of 18 of the GSTs after B. tabaci MEAM1 and MED adults were exposed to the insecticide imidacloprid, we found that the expression levels were increased more in B. tabaci MED than in MEAM1 (in particular, the expression level of GST-d7 was increased by 4.39-fold relative to the control). Knockdown of GST-d7 in B. tabaci MED but not in B. tabaci MEAM1 resulted in a substantial increase in the mortality of imidacloprid-treated adults. These results indicate that differences in GST-d7 may help explain why insecticide tolerance is greater in B. tabaci MED than in B. tabaci MEAM1. © 2017 The Royal Entomological Society

摘要:
Controlled release system has been widely developed and utilised in agriculture to optimise efficacy and minimise environmental pollution. Here, the azoxystrobin and difenoconazole compound pesticide microsphere was constructed by the solvent evaporation method. Blending poly (butylene succinate) (PBS) and poly (lactic acid) (PLA) as shells, in order to increase its entrapment efficiency and match the goal of complex synergism. The resulting microsphere processes high entrapment efficiency with a mean diameter of 7.2 μm. Acute toxicity suggested microsphere on zebrafish was moderate toxicity. Comparative studies revealed that the microsphere had a longer period of sustained release than difenoconazole–azoxystrobin (5:8) 32.5% w/v suspension concentrate (SC). In addition, the concentration ratio of active ingredients closed to the optimum compound stoichiometry, which can achieve the objective of improving the efficacy of the pesticides. These results demonstrated that such a compound pesticide microsphere delivery system may be a considerable potential for further exploration. © 2018, © 2018 Informa UK Limited, trading as Taylor & Francis Group.

Invasive species often cause enormous economic and ecological damage, and this is especially true for invasive plants in the Asteraceae family. Arbuscular mycorrhizal fungi (AMF) play an important role in the successful invasion by exotic plant species because of their ability to promote growth and influence interspecific competition. However, few studies have evaluated the effects of invasive Asteraceae species on AMF diversity and how feedback mechanisms during competition with native species subsequently affect the accumulation of nutrient resources. Two exotic Asteraceae, Ambrosia artemisiifolia and Bidens pilosa, were monitored during competition with a native grass species, Setaria viridis, which is being replaced by these exotic species in natural areas around the study site. From these species continuously maintained in a field plot for 5 years, we collected the rhizosphere soil and cloned and identified soil AMF. Furthermore, AM fungal spores were isolated from rhizosphere soil of the two invasive species and used as inoculum in greenhouse experiments, to compare growth and nutrient accumulation during competition. The results indicate that although the AMF diversity in the rhizosphere soil of A. artemisiifolia and B. pilosa differed, the three most abundant species (Septoglomus viscosum, Septoglomus constrictum, Glomus perpusillum) were identical. The addition of AMF inoculum changed the competition between the plants, increasing the competitive ability of the invasives and decreasing that of the native. The results show a similar AMF community composition between A. artemisiifolia and B. pilosa, increased AMF root colonization of the invasive species during competition, AMF-enhanced N accumulation, and AMF-facilitated competitive growth of the invasive species.

Dimethoate, a systemic insecticide, has been used extensively in vegetable production. Insecticide residues in treated vegetables, however, pose a potential risk to consumers. Photocatalytic degradation is a new alternative to managing pesticide residues. In this study, the degradation of dimethoate in Bok choy was investigated under the field conditions using cerium-doped nano titanium dioxide (TiO2/Ce) hydrosol as a photocatalyst. The results show that TiO2/Ce hydrosol can accelerate the degradation of dimethoate in Bok choy. Specifically, the application of TiO2/Ce hydrosol significantly increased the reactive oxygen species (ROS) contents in the treated Bok choy, which speeds up the degradation of dimethoate. Ultra-performance liquid chromatography coupled with mass spectrometry (UPLC-MS) analysis detected three major degradation products, including omethoate, O,O,S-trimethyl thiophosphorothioate, and 1,2-Bis (acetyl-N-methyl-) methane disulfide. Two potential photodegradation pathways have been proposed based on the intermediate products. To understand the relationship between photodegradation and the molecular structure of target insecticides, we investigated the bond length, Mulliken atomic charge and frontier electron density of dimethoate using ab initio quantum analysis. These results suggest the P = S, P-S and S-C of dimethoate are the initiation sites for the photocatalytic reaction in Bok choy, which is consistent with our empirical data.

Ascoviruses are circular double-stranded DNA viruses that infect insects. Herein we sequenced and analyzed the genome of the previously unrecorded ascovirus isolate Heliothis virescens ascovirus 3i (HvAV-3i). The genome size is 185,650 bp with 181 hypothetical open reading frames (ORFs). Additionally, definition based on ascovirus repeated ORFs (aros) is proposed; whereby the 29 aros from all sequenced Ascoviridae genomes are divided into six distinct groups. The topological relationship among the isolates of Heliothis virescens ascovirus 3a is (HvAV-3f, {HvAV-3h, [HvAV-3e, (HvAV-3g, HvAV-3i)]}) with every clade well supported by a Bayesian posterior probability of 1.00 and a Bootstrap value of 100%.

Ascoviruses are a recently described family and the traditional plaque assay and end-point PCR assay have been used for their titration. However, these two methods are time-consuming and inaccurate to titrate ascoviruses. In the present study, a quick method for the determination of the titer of ascovirus stocks was developed based on ascovirus-induced apoptosis in infected insect cells. Briefly, cells infected with serial dilutions of virus (10 −2 –10 −10 ) for 24 h were stained with trypan blue. The stained cells were counted, and the percentage of nonviable cells was calculated. The stained cell rate was compared between virus-infected and control cells. The minimum-dilution group that had a significant difference compared with control and the maximum-dilution group that had no significant difference were selected and then compared each well of the two groups with the average stained cell rate of control. The well was marked as positive well if the stained cell rate was higher than the average stained cell rate of control wells; otherwise, the well was marked as negative wells. The percentage of positive wells were calculated according to the number of positive. Subsequently, the virus titer was calculated through the method of Reed and Muench. This novel method is rapid, simple, reproducible, accurate, and less material-consuming and eliminates the subjectivity of the other procedures for titrating ascoviruses. © 2018 Elsevier B.V.

The existing form of an ionizable organic compound can simultaneously affect its soil adsorption and plant bioactivity. In this experiment, the adsorption and bioactivity of two weak acid herbicides (WAHs), imazethapyr and 2,4-D, were studied to explore the predominant mechanism by which the soil pH and the addition of biochar can influence the phytotoxicity of WAHs in soil. Then, the WAH concentration extracted by hollow fiber-based liquid-phase microextraction (C HF-LPME ), the in situ pore water concentration (C IPW ) and the added concentration (C AC ) were employed to estimate the phytotoxicity. The results showed that with increased pH from 5.5 to 8.5, the phytotoxicity of the WAHs to rice increased about 1-fold in the soil, but decreased in aqueous solutions, the IC 50 values for imazethapyr and 2,4-D at pH 5.0 were 3- and 2-fold higher than that at pH 8.0. In addition, the soil adsorption decreased, indicating that the adsorption process was the dominant factor for the variation of the phytotoxicity of the WAHs in the tested soil instead of the decreasing bioactivity. The concentration that inhibits plant growth by 50% (IC 50 ) calculated by the C AC in different pH and biochar soils ranged from 0.619 to 3.826 mg/kg for imazethapyr and 1.871–72.83 mg/kg for 2,4-D. The coefficient of variation (CV) of the IC 50 values reached 65.61% for imazethapyr and 130.0% for 2,4-D. However, when IC 50 was calculated by C IPW and C HF-LPME , the CVs of the IC 50 values decreased to 23.51% and 36.23% for imazethapyr and 40.21% and 50.93% for 2,4-D, respectively. These results suggested that C IPW and C HF-LPME may be more appropriate than C AC for estimating the phytotoxicity of WAHs. © 2018 Elsevier Inc.

Rosiglitazone (RG) is a well-known activator of peroxisome proliferator-activated receptor-gamma (PPARγ) and used to treat hyperglycemia and type 2 diabetes; however, its clinical application has been confounded by adverse side effects. Here, we assessed the roles of chlorogenic acid (CGA), a phenolic secondary metabolite found in many fruits and vegetables, on the differentiation and lipolysis of mouse 3T3-L1 preadipocytes. The results showed that CGA promoted differentiation in vitro according to oil red O staining and quantitative polymerase chain reaction assays. As a potential molecular mechanism, CGA downregulated mRNA levels of the adipocyte differentiation-inhibitor gene Pref1 and upregulated those of major adipogenic transcriptional factors (Cebpb and Srebp1). Additionally, CGA upregulated the expression of the differentiation-related transcriptional factor PPARγ2 at both the mRNA and protein levels. However, following CGA intervention, the accumulation of intracellular triacylglycerides following preadipocyte differentiation was significantly lower than that in the RG group. Consistent with this, our data indicated that CGA treatment significantly upregulated the expression of lipogenic pathway-related genes Plin and Srebp1 during the differentiation stage, although the influence of CGA was weaker than that of RG. Notably, CGA upregulated the expression of the lipolysis-related gene Hsl, whereas it did not increase the expression of the lipid synthesis-related gene Dgat1. These results demonstrated that CGA might function as a potential PPARγ agonist similar to RG; however, the impact of CGA on lipolysis in 3T3-L1 preadipocytes differed from that of RG. © 2018 Shu-guang Peng et al.

Background: Chemosensory proteins are a family of insect-specific chemical sensors that sense specific chemical cues and regulate insect behavior. Chemosensory proteins have been identified and analyzed in many insect species, such as Drosophila melanogaster, Bactrocera dorsalis and Calliphora stygia. This research has revealed that these proteins play a crucial role in insect orientation, predation and oviposition. However, little is known about the chemosensory proteins of Chlorops oryzae, a major pest of rice crops throughout Asia. Results: Comparative transcription analysis of the genes of Chlorops oryzae larvae, pupae and adults identified a total of 104 chemosensory genes, including 25 odorant receptors (ORs), 26 odorant-binding proteins (OBPs), 19 ionotropic receptors (IRs), 23 gustatory receptors (GRs) and 11 sensory neuron membrane proteins (SNMPs). The sequences of these candidate chemosensory genes were confirmed and used to construct phylogenetic trees. Quantitative real-time PCR (qRT-PCR) confirmed that the expression of candidate OR genes in different developmental stages was consistent with the fragments per kilobase per million fragments (FPKM) values of differentially expressed genes (DEGs). Conclusions: The identification of chemosensory genes in C. oryzae provides a foundation for the investigation of the function of chemosensory proteins in this species, which, in turn, could allow the development of new, improved methods of controlling this pest. © 2018 The Author(s).

Early instar larvae of the tobacco cutworm Spodoptera litura (Lepidoptera: Noctuidae) and the beet armyworm Spodoptera exigua (Lepidoptera: Noctuidae) are recognized hosts of the parasitic wasp Microplitis prodeniae Rao and Kurian (Hymenoptera: Braconidae), although M. prodeniae has previously been regarded as monophagous. In this study, we found the immature period and longevity of M. prodeniae developing in S. exigua was similar to that in S. litura. It was shown that the development time of M. prodeniae in S. exigua was 15.1 ± 0.3 d, not significantly different from 15.0 ± 0.2 d in S. litura. The parasitism rate of M. prodeniae attacking S. exigua was significantly lower than on S. litura (65.48 ± 2.29 and 43.83 ± 2.20%, respectively), whilst the female ratio of the wasp’s offspring was not significantly different when developing on the two species. M. prodeniae females prefer to oviposit on the second- and third-instar host larvae of S. exigua, rather than other instars. The effects of development of M. prodeniae on two important lepidopterous pests are discussed.

BACKGROUND: Ambrosia artemisiifolia, an invasive weed in Europe and Asia, is highly toxic to the golden apple snail (GAS; Pomacea canaliculata) in laboratory tests. However, little is known about the chemical components of A. artemisiifolia associated with the molluscicidal activity or about its potential application for GAS control in rice fields. This study evaluated the molluscicidal activities of powders, methanol extracts, and individual compounds from A. artemisiifolia against GAS in rice fields and under laboratory conditions. RESULTS: Ambrosia artemisiifolia powders did not negatively affect the growth and development of rice but they reduced damage to rice caused by GAS. Extracts had moderate acute toxicity but potent chronic toxicity. The 24-h 50% lethal concentration (LC50) of the extracts against GAS was 194.0 mg L–1, while the weights, lengths and widths of GAS were significantly affected by exposure to a sublethal concentration (100 mg/mL). Psilostachyin, psilostachyin B, and axillaxin were identified as the most active molluscicide components in the aerial parts of A. artemisiifolia, and the 24-h LC50 values of these purified compounds were 15.9, 27.0, and 97.0 mg/L, respectively. CONCLUSION: The results indicate that chemical compounds produced by A. artemisiifolia may be useful for population management of GAS in rice fields. © 2017 Society of Chemical Industry. © 2017 Society of Chemical Industry

Termite gut microbiome is a rich reservoir for glycoside hydrolases, a suite of enzymes critical for the degradation of lignocellulosic biomass. To search for hemicellulases, we screened 12,000 clones from a fosmid gut library of a higher termite, Globitermes brachycerastes. As a common Southeastern Asian genus, Globitermes distributes predominantly in tropical rain forests and relies on the lignocellulases from themselves and bacterial symbionts to digest wood. In total, 22 positive clones with β-xylosidase activity were isolated, in which 11 representing different restriction fragment length polymorphism (RFLP) patterns were pooled and subjected to 454 pyrosequencing. As a result, eight putative β-xylosidases were cloned and heterologously expressed in Escherichia coli BL21 competent cells. After purification using Ni-NTA affinity chromatography, recombinant G. brachycerastes symbiotic β-xylosidases were characterized enzymatically, including their pH and temperature optimum. In addition to β-xylosidase activity, four of them also exhibited either β-glucosidase or α-arabinosidases activities, suggesting the existence of bifunctional hemicellulases in the gut microbiome of G. brachycerastes. In comparison to multimeric protein engineering, the involvement of naturally occurring multifunctional biocatalysts streamlines the genetic modification procedures and simplifies the overall production processes. Alternatively, these multimeric enzymes could serve as the substitutes for β-glucosidase, β-xylosidase and α-arabinosidase to facilitate a wide range of industrial applications, including food processing, animal feed, environment and waste management, and biomass conversion. © Ivyspring International Publisher.

Back pain is a common clinical symptom. Degeneration of intervertebral discs is one of the most important factors leading to back pain, namely, discogenic back pain. However, at present, the understanding of lumbar intervertebral discs causing back pain is confined to biomechanical and histological studies. The neuropathological mechanism related to discogenic back pain is still not well understood. Many studies have found that as an intervertebral disc degenerates, the peripheral nerve tissues have corresponding structural reorganization, and a series of nerve cells become involved in progression of discogenic back pain. Therefore, study of neural mechanisms that are involved in progression of discogenic back pain will provide additional assistance for treatment of its symptoms. We review the anatomical structure of intervertebral discs and the related neural mechanisms involved in discogenic back pain. We also discuss the current view of neural mechanisms underlying discogenic back pain.

The fijivirus southern rice black-streaked dwarf virus (SRBSDV) causes one of the most serious viral diseases of rice in China and Vietnam. To better understand the molecular basis of SRBSDV infection, a yeast two-hybrid screen of a rice cDNA library was carried out using P8, a minor core protein of SRBSDV, as the bait. A rice Cys2His2-type zinc finger protein (OsZFP) was found to interact with SRBSDV P8. A strong interaction between SRBSDV P8 and OsZFP was then confirmed by pull-down assays, and bimolecular fluorescence complementation assays showed that the in vivo interaction was specifically localized in the nucleus of plant cells. Using a series of deletion mutants, it was shown that both the NTP-binding region of P8 and the first two zinc fingers of OsZFP were crucial for their interaction in plant cells. The localization in the nucleus and activation of transcription in yeast supports the notion that OsZFP is a transcription factor. SRBSDV P8 may play an important role in fijiviral infection and symptom development by interfering with the host transcription activity of OsZFP.

The whitefly (Bemisia tabaci) is a cosmopolitan and devastating pest of agricultural crops and ornamentals. B. tabaci causes extensive damage by feeding on phloem and by transmitting plant viruses. Like many other organisms, insects depend on amino acid transporters (AATs) to transport amino acids into and out of its cells. We present a genome-wide and transcriptome-wide investigation of the following two families of AATs in B. tabaci biotype B: amino acid/auxin permease (AAAP) and amino acid/polyamine/organocation (APC). A total of 14 putative APCs and 25 putative AAAPs were identified, and a 10-paralog B. tabaci-specific expansion of AAAPs was found by maximum likelihood phylogeny. Detailed gene structure information revealed that 9 members of the B. tabaci-specific AAAP family expansion closely situated on a same scaffold. Expression profiling of the B. tabaci B APC and AAAP genes as affected by stage and plant host showed diverse expression patterns. The analysis of evolutionary rates indicated that purifying selection can explain the B. tabaci-specific AAAP expansion. RNA interference (RNAi)-mediated suppression of two AAAP genes (BtAAAP15 and BtAAAP21) significantly increased the mortality of B. tabaci B adults. The results provide a foundation for future functional analysis of APC and AAAP genes in B. tabaci. © Ivyspring International Publisher.

Parapoynx crisonalis (Walker, 1859) (Lepidoptera: Crambidae) is a major pest of aquatic vegetables and aquatic landscape plants. It has been responsible for causing considerable economic damage to water chestnut (Trapa natans) plants. In the Changsha vicinity of China, P. crisonalis has five generations a year. Populations of P. crisonalis were relatively low in April and began to rapidly rise at the beginning of May. At the end of July and early August, the population dropped dramatically. A rebound occurred at the end of August and early September, which was referred to as the second population peak. From then, until early November, the P. crisonalis population steadily diminished in preparation for overwintering. The primary factors influencing the seasonal dynamics of P. crisonalis were the climatic conditions, especially the temperature, and secondarily precipitation. Between May and October, the P. crisonalis adults were evenly distributed in the pond. In May and June, the eggs of P. crisonalis were present in an aggregate distribution, due to the effects of environmental heterogeneity. In July and August, however, they were found to be in a uniform distribution.
