替代抗生素添加剂在断奶仔猪上应用效果及机制

摘要:
为了提高母猪利用率,提高生产效率。目前规模化养殖场常采用早期断奶技术。而早期断奶会造成仔猪腹泻,免疫力降低,生长缓慢的症状。传统上使用抗生素治疗仔猪的断奶综合症,然而抗生素会造成动物机体产生抗药性,对环境产生污染。因此绿色无污染的替代抗生素添加剂成为新热点,目前主要替代抗生素产品主要有:酸化剂,植物提取物,酶制剂,益生菌,抗菌肽。本文主要针对以上添加剂在断奶仔猪上使用效果和作用机理进行综述。

植物多糖对断奶仔猪生长性能及肠道内环境的影响

摘要:
本试验旨在探讨断奶仔猪饲粮中单独或联合添加黄芪多糖、白术多糖和牛膝多糖对断奶仔猪生长性能及肠道内环境的影响。试验选取256头35日龄的杜×长×大三元杂交断奶仔猪,随机分为8个组,每组4个重复,每个重复8头猪。采用三因素两水平( 2×2×2)试验设计,饲粮中黄芪多糖、白术多糖和牛膝多糖的添加水平设置为0和800 mg /kg,试验共进行28 d。结果表明: 1)在断奶仔猪饲粮中单独或两两联合添加黄芪多糖、白术多糖和牛膝多糖有提高断奶仔猪平均日增重的趋势( P > 0.05) ,3种多糖联合使用能显著提高断奶仔猪平均日增重( P < 0.05) ,各组之间料重比无显著差异( P>0.05) 。断奶仔猪饲粮中单独添加黄芪多糖、白术多糖和牛膝多糖均能显著降低断奶仔猪的腹泻率( P<0.05) ,多糖之间存在一定互作效应。2)与对照组相比,单独或联合添加多糖组断奶仔猪回肠、直肠内容物的pH无显著变化( P>0.05) ,盲肠和结肠内容物pH显著降低( P<0.05) ,3种多糖联合添加对盲肠内容物pH有互作效应( P<0.05) 。 3)与对照组相比,单独或联合添加多糖组断奶仔猪盲肠内容物总挥发性脂肪酸含量均显著上升( P<0.05) ,三者联合的效果显著优于单独添加( P<0.05) 。4)在断奶仔猪饲粮中单独或联合添加植物多糖均能显著提高肠道内乳酸菌和双歧杆菌的数量( P<0.05) ,多糖两两组合组与单一添加组相比肠道内乳酸菌和双歧杆菌的数量有增加趋势,但差异不显著( P>0.05) ,单独或联合添加多糖能显著降低肠道中大肠杆菌的数量( P<0.05) 。由此可见,饲粮添加黄芪多糖、白术多糖和牛膝多糖可改善其肠道内环境,降低仔猪腹泻,从而改善断奶仔猪的生长性能,且以三者联合添加效果最佳。

原花青素是植物中广泛存在的一大类多酚化合物的总称,具有超强的抗氧化能力,是目前国际上公认的清除机体自由基最有效的天然抗氧化剂。已有的体内外试验表明,原花青素在改善机体功能方面具有广阔的应用前景,如能够减少炎症、改善氧化应激、抗癌和调节免疫等。本文就原花青素的化学结构、原料来源、分离提取方法、生物学功能,及其在畜禽生产中的应用现状等进行综述。

牛膝多糖是植物多糖的一种,分离提取于牛膝根中,是一种水溶性较好的小分子多糖化合物。牛膝多糖能缓解抗氧化应激、抑制肿瘤细胞以及改善机体免疫功能等。牛膝多糖在畜牧业的应用主要体现在增强免疫功能,有望成为一种安全系数高、稳定性强的抗生素替代物。同样,牛膝多糖在水产养殖和人体方面都具有很好的应用前景。文章主要从牛膝多糖对肠道营养吸收、关节炎症治疗、缓解氧化应激和改善机体免疫功能等方面作以阐述。

摘要:
High ambient temperatures affect animal production and welfare in tropical and sub-tropical regions of the world. Feed intake, growth rate, mortality, egg production, hatchability and other production traits related to the economic success of the poultry industry are adversely affected by severe heat stress. In general, heat stress induces the activity of the neuroendocrine system, resulting in activation of the hypothalamic-pituitary-adrenal (HPA) axis, and elevated corticosterone (CORT) concentrations, which affects metabolism and immune responses. These include negative regulation of metabolic hormones, antibody production and heterophil to lymphocyte (H/L) ratio. Heat stress increases mitochondrial activity, causing reactive species overproduction which disrupts the antioxidant balance, leading to oxidative stress damage of membranes, protein and DNA. Heat stress stimulates the central nervous system (CNS), which significantly reduces daily gain, feed intake and FCR in poultry. Consequently, from an animal husbandry perspective, intervention strategies to relieve heat stress conditions have been the focus of many published studies. This review describes the effect of high temperature on production, behavioural, biochemical and immune responses, including oxidative damage that occur during heat stress in poultry, in broilers and laying hens. Moreover, nutritional interventions to alleviate the negative consequence of heat stress is discussed.

摘要:
The present study was conducted to investigate the effects of dietary Achyranthes bidentata polysaccharide (ABPS) supplementation on performance, immune response, intestinal mucosal morphology, and gut microbiome in yellow-feathered broilers challenged with Escherichia coli K88. A 2 × 2 factorial design was used for the trial. Two hundred and forty one-day-old female broilers were randomly assigned to four treatments: (1) negative-control broilers were fed by a basal diet and saline (NG); (2) positive-control broilers were fed by a basal diet and orally challenged with 108 CFU E. coli K88 (CNG); (3) ABP group broilers were fed by a basal diet containing ABPS (500 mg/kg of feed) and saline; (4) CABP group broilers were fed by a basal diet containing ABPS (500 mg/kg of feed) and orally challenged with 108 CFU E. coli K88. Growth performance, serum biochemical indexes, immune responses, intestinal mucosal morphology, and cecal microbial community structure were evaluated. The ABP group had greatest body weight (BW), average daily body weight gain (ADG), and intestinal villus height compared to other treatments (p < 0.05). The CABP group had a higher villus height/crypt depth ratio (V/C) compared with other treatments (p < 0.05). The expression levels of NF-κB were lower in the ABP group. The CNG group had higher Escherichia coli and Enterococcus contents in cecal samples compared to other treatments (p < 0.05). Serum glucose, uric acid, TNF-α, and Secretory Immunoglobulin A (S-IgA) levels were higher in broilers challenged with E. coli (p < 0.001) than that with saline. Broilers challenged with E. coli had reduced taxa richness in the cecal samples. Sequencing of 16S rRNA genes in cecal samples revealed that a lower proportion of Firmicutes and a higher proportion of Proteobacteria were detected in the broilers challenged with E. coli. Compared with the controls, dietary ABPS supplementation increased serum total protein, albumin, and S-IgA levels, but decreased serum glucose, uric acid, and TNF-α levels in broilers (p < 0.05). Diet supplemented with ABPS increased the Firmicutes/Bacteroidetes ratio and the abundance of Ruminococcaceae and Lachnospiraceae, and altered cecal microbiota community structure. These results suggest that ABPS can promote growth performance and improve intestinal morphology and microbiota community structure in broilers challenged with E. coli K88. © 2018 by the authors.

摘要:
研究旨在对宁乡猪和杜长大杂种猪的生长性能、胴体性状和肉质性状进行评估.选取体重、胎次、日龄相近的宁乡猪和杜长大分为两组,每组24头,分为4个重复,每个重复6头,在同一饲养标准下饲养.结果表明:1)杜长大的日增重是宁乡猪的1.5倍,差异极显著(P<0.01);料重比杜长大比宁乡猪低27.25%,差异显著(P<0.05).2)杜长大的屠宰率、瘦肉率、眼肌面积、胴体斜长和胴体直长等分别比宁乡猪高5.31%、67.99%、248.36%、19.74%、20.23%;宁乡猪的脂肪率和腹脂率分别为40.43%、2.87%,杜长大分别为17.53%、0.49%,杜长大比宁乡猪分别低56.64%、82.93%,均差异极显著(P<0.01);宁乡猪背膘厚、皮厚高于杜长大,差异不显著(P>0.05).3)宁乡猪的大理石纹评分和在24 h的肉色红度极显著大于杜长大(P<0.01);宁乡猪的pH1和pH2均大于杜长大,两猪种的熟肉率、失水率、肉色亮度和pH均差异不显著(P>0.05).综上所述,杜长大杂种猪的生长性能和胴体性状较好,宁乡猪肉质较好,这主要是脂肪沉积能力决定的.

摘要:
In an effort to further understand of the differences of meat flavor and texture between Linwu ducks and Pekin ducks at market age, we investigated the meat metabolite composition of the two breeds of ducks using 600 MHz 1H nuclear magnetic resonance (NMR) spectroscopy. Comprehensive multivariate data analysis including principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA), and orthogonal projection to latent structure-discriminant analysis (OPLS-DA) were applied to analyze the 1H-NMR profiling data to identify the distinguishing metabolites of breast meat between two breeds of ducks. Compared with 42-d-old Pekin duck meat, breast from 72-d-old Linwu duck has higher concentration of anserine, carnosine, homocarnosine, and nicotinamide, but significantly lower concentration of succinate, creatine, and myo-inositol. These results contribute to a better understanding of the differences in meat metabolite composition between 72-d-old Linwu and 42-d-old Pekin ducks, which could be used to help assess the quality of duck meat as a food. © 2016 Poultry Science Association Inc.

摘要:
猪多杀性巴氏杆菌(Pasteurella multocidain, Pm)感染可引起猪(Sus scrofa)产生猪肺疫、猪出血性败血症和猪萎缩性鼻炎等。目前应用的灭活全菌苗交叉保护效果差且免疫保护期短,重组疫苗尚不能提供高的保护率。为研究猪多杀性巴氏杆菌重组甲硫氨酸转运体Q(recombinant methionine transporter Q, rMetQ)的免疫保护性,本研究根据GenBank中登录的Pm metQ基因序列,根据去除影响表达的长片段疏水区的相应核苷酸序列设计引物对,以Pm的基因组为模板PCR扩增metQ基因,测序进行生物信息学分析。结果表明,猪Pm metQ基因片段含768 bp核苷酸,编码255个氨基酸的蛋白质,其理论分子量为28.14 kD,是与细菌的毒性有关的胞外结合蛋白,不同血清型的该蛋白同源型达99%以上。将Pm metQ亚克隆到pET-28a(+)构建重组质粒pET-metQ并转化至大肠杆菌(Escherichia coli) BL21 (DE3),用异丙基-β-D-硫代半乳糖苷(isopropylβ-D-1-thiogalactopyranoside, IPTG)诱导表达Pm rMetQ,经十二烷基硫酸钠聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis, SDS-PAGE)检测,结果表明,在28 kD处出现特异性条带。动物保护实验采用昆明小鼠(Mus musculus)进行,免疫球蛋白G (immunoglobulin G, IgG)抗体滴度采用酶联免疫吸附测定法(enzyme-linked immuno sorbent assay, ELISA)检测。结果表明,3次免疫后第14天,铝佐剂+rMetQ组小鼠血清中的IgG抗体水平显著升高,明显高于对照组和低剂量组,对照组和低剂量组小鼠攻毒后3 d存活率为20%,7 d后全部死亡。铝佐剂+30 μg rMetQ免疫组,10只免疫组小鼠3 d后,6只死亡,存活率为40%;7 d后,又有3只死亡,存活率为10%。铝佐剂+50 μg rMetQ组3 d后存活率达80%,7 d后存活率40%。死亡小鼠剖检发现肝脏为主要病变器官,呈现有不同程度的出血点甚至坏死。可见,Pm rMetQ皮下免疫注射可诱导小鼠产生免疫应答和较高的抗攻击感染的免疫保护力。该研究为进一步筛选猪Pm分子疫苗积累了资料。

固有免疫是机体免疫系统防御微生物感染的第1道防线。模式识别受体(PRRs)中的Toll样受体(TLRs)、核苷酸寡聚结合域样受体(NLRs)和视黄酸诱导基因1样受体(RLRs)等识别不同或重叠的微生物组成成分,形成相应的信号转导产生免疫应答。近些年对于固有免疫相关PRRs的研究也取得了较大的进展,为免疫相关疾病的治疗提供了思路。本文对固有免疫相关PRRs的特征及不同PRRs的信号传导途径之间相互作用进行综述。

摘要:
Magnolol rich in Magnolia officinalis is a bioactive polyphenolic compound. The aim of this study was to examine the effects of magnolol additive (MA) on growth performance, expression levels of antioxidant-related genes, and intestinal mucosal morphology of Linwu ducks aged from 49 to 70 days, comparing with that of an antibiotic additive (colistin sulfate [CS]). A total of 275, 49-day-old ducks were assigned to 5 groups with 5 cages of 11 ducks each and fed diets supplemented with 0, 100, 200 and 300 mg of MA/kg and 300 mg of CS/kg for 3 weeks, respectively. The results showed that the average daily body weight gain (ADG) was increased significantly in MA-fed groups (200 and 300 mg/kg), compared with the basal diet (BD) group (P < 0.05). The mRNA levels of superoxide dismutase-1 (SOD1), manganese superoxide dismutase-2 (MnSOD2) and catalase (CAT) were also increased significantly in MA groups (P < 0.05). In addition, hematoxylin and eosin staining revealed that Linwu ducks fed the diets with MA had more intact intestinal mucosa than those fed the BD and CS diets. In addition, ileal villus height, ileal villus height/crypt depth ratio (V/C) and duodenal V/C were also improved significantly (P < 0.05). Taken together, these data demonstrated that MA is an effective feed additive to enhance the growth performance of the Linwu ducks by improving the antioxidant and intestinal mucosal status, suggesting that MA will be a potential additive to replace antibiotic (CS). © 2017 Chinese Association of Animal Science and Veterinary Medicine

为研究猪多杀性巴氏杆菌(Pm)重组抗菌肽ABC转运体ATP结合蛋白的免疫保护性,本研究采用 PCR方法扩增该蛋白基因,并将其克隆于pET-28a(+)中构建重组质粒pET-ABP,转化大肠杆菌BL21(DE3),IPTG诱导重组蛋白表达。分析表明,猪Pm抗菌肽ABC转运体ATP结合蛋白基因为696 bp,编码231 aa,不同血清型的该蛋白基因推导氨基酸序列同源性达99%以上。表达的蛋白分子量约为27 ku。动物实验表明,实验组小鼠三免后第14 d其血清IgG抗体水平显著升高,明显高于对照组。对照组攻毒后3 d存活率为20%, 7d全部死亡;免疫组小鼠攻毒后3 d存活率为80%, 7d存活率为40%。该研究为进一步研制Pm的混合疫苗奠定了基础。

为了研究猪多杀性巴氏杆菌IbeB重组蛋白(rIbeB)的免疫保护性,根据G enBank中登录的多杀性巴氏杆菌ibeB基因序列设计1对引物扩增ibeB基因,并重组至pET-28a(+)构建重组表达质粒pET- ibeB,转化到大肠杆菌BL21 (D E3),经IPTG诱导获得rIbeB。生物信息学分析表明,猪多杀性巴氏杆菌ibeB基因长1 392 bp,编码蛋白质含463个氨基酸。该蛋白质的理论分子质量为51.98 ku,具有2个外排蛋白结构域,属外膜外排蛋白超家族。蛋白序列比对发现,不同血清型的多杀性巴氏杆菌IbeB的同源性达99%以上。rIbeB诱导小鼠的免疫保护结果表明,第3次免疫后第14天小鼠血清中IgG抗体水平显著升高,明显高于佐剂对照组。攻毒后,对照组小鼠第3天的存活率为20%,7 d后全部死亡;免疫组第3天的存活率达70%,第7天的存活率为40%。结果表明,rIbeB蛋白具有很好的免疫原性,可诱导小鼠产生部分免疫保护力。

摘要:
本实验旨在探究酵母培养物对哺乳母猪及仔猪生产性能的影响。试验采用单因素试验设计,试验选取品种一致、体重均匀、胎次接近的健康PIC哺乳母猪24头.随机分为A、B、C3个处理,每个处理8个重复,每个重复1头母猪。A组为基础日粮,B组使用酵母培养物替换麦麸,c组使用酵母培养物替代部分玉米及豆粕(成本与A相当3)。结果表明:1)哺乳仔猪在1~3d日增重有显著差异,但在4~lOd及11—18d阶段平均日增重无显著差异(P〉0.05),但0—18日全程日增重有差异趋势,B组平均日增重分别比A、C组高出34.8g/d,以及7g/d。2)各组母猪哺乳期采食总量无显著差异(P〉0.05),母猪泌乳量在产后3~10d及11~18d均无显著差异。但0-18日全程泌乳量有差异的趋势。B组的泌乳量最好,比A、C组分别高出22kg以及10kg。3)日粮B对单头泌乳母猪的经济效益贡献值最大.分别比日粮A和C提高了252.31元和69.0l元;日粮C对单头泌乳母猪的利润也提高了183.3元。综上所述.在PIC哺乳母猪日粮中添加酵母培养物能有效增加哺乳母猪及仔猪的生产性能.提高经济效益。

Objective: This study investigated the effect of microbial phytase supplementation on growth performance, tibia ash, plasma parameters, apparent ileal digestibility (AID) of amino acid (AA) and apparent digestibility of nutrients in Youxian Sheldrakes fed with low-phosphorus (P) corn-soybean diets. Methods: A total of 350 Youxian Sheldrakes (7d old) were randomly divided into 5 treatment groups: positive control (PC) group has adequate available P diet (0.42% and 0.38%, starter and grower), negative control (NC) group were deficient in available P (0.32% and 0.28%, starter and grower) and NC diet was supplemented with 3 levels of microbial phytase (500, 750, and 1,000 U/kg). Results: Dietary supplementation of phytase in NC diet improved the average daily gain, increased the levels of serum calcium (Ca), tibia Ca and P, AID of AA and apparent digestibility of energy and Ca in starter stage (p<0.05). There was an increased (p<0.001) in the utilization of P from 17.3% to 23.9%. Phytase supplementation (1,000 U/kg) has shown that the AID of His, Thr, Val, indispensable AA, Glu, Pro, and dispensable AA was higher (p<0.05) than that of NC. Moreover, phytase supplementation improved (p<0.05) serum and tibia Ca and P, AID of AA and apparent digestibility of dry matter, crude protein, energy, P and Ca, and reduced (p<0.05) feed to gain ratio (F/G) and the levels of serum alkaline phosphatase in grower stage. Likewise, an increase (p<0.001) in the utilization of P was noticed from 12.6% to 17.2%. Supplement phytase at 750 U/kg improved the AID of His, Thr, Asp, Cys, Pro, and Ser (p<0.05). Conclusion: The microbial phytase supplement could improve growth performance, AID of some AA and apparent utilization of other nutrients in Youxian Sheldrakes, and reduce excreta P load to environment. 摘要:
The aim of this study was to investigate whether supplementation with resveratrol could alleviate intestinal injuries and to explore how resveratrol regulates heat shock protein (HSP)70, HSP90, nuclear factor kappa B (NF-κB) and epidermal growth factor (EGF) expression in the jejunal mucosa of black-boned chickens under circular heat stress. A total of 300 black-boned chicks of 42-d-old were randomly assigned to five treatment groups. The positive control chickens were kept in a normal-temperature (NT, 24 ±2 °C) chamber and fed with a basal diet. The other four groups were kept in a circular high-temperature (HT, 37 ±2 °C) chamber for 8 h and fed a basal diet supplemented with 0, 200, 400, or 600 mg per kg of resveratrol for 15 days. The results showed that the heat-stress responses damaged the villus structures of the jejunum and ileum, resulting in shortened intestinal villi, deepened crypts, and a reduced villus height to crypt depth (V/C) ratio and decreased the numbers of goblet cells and lymphocytes. Heat stress also caused increased mRNA and protein expression of HSP70, HSP90 and NF-κB, and reduced EGF in the jejunal mucosa. Dietary supplementation with 400 mg per kg of resveratrol improved the villus morphology, increased the numbers of goblet cells and lymphocytes, attenuated the mRNA overexpression of HSP70, HSP90 and NF-κB on the 6<sup>th</sup>, 10<sup>th</sup>and 15<sup>th</sup>day of heat stress (P <0.05), and activated the expression of EGF (P <0.05) in the jejunal mucosa. Resveratrol reduced protein expression of HSP70, HSP90 and NF-κB in the jejunal villi after 15 days of heat stress, and increased EGF expression from the lamina propria toward the epithelial cells of the villi. These results suggest that dietary resveratrol offers a potential nutritional strategy to improve the intestinal morphology and alleviate jejunum mucosa injuries by modulating the mRNA and protein expression of HSPs, and the epithelial growth factor and transcription factor in black-boned chickens subjected to circular heat stress. ©The Royal Society of Chemistry 2016.

Youxian sheldrake is excellent native breeds in Hunan province in China. The complete mitochondrial (mt) genome sequence plays an important role in the accurate determination of phylogenetic relationships among metazoans. This is the first study to determine the complete mitochondrial genome sequence of Youxian sheldrake using PCR-based amplification and Sanger sequencing. The characteristic of the entire mitochondrial genome was analyzed in detail, the total length of the mitogenome is 16,605 bp, with the base composition of 29.21% A, 22.18% T, 32.84% C, 15.77% G in the Youxian sheldrake. It contained 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and a major non-coding control region (D-loop region). The complete mitochondrial genome sequence of Youxian sheldrake provided an important data for further study of the phylogenetics of poultry, and available data for the genetics and breeding. © 2014 Informa UK Ltd.

Huang Lang chicken is the native breed of Hunan province in China. The complete mitochondrial (mt) genome sequence plays an important role in the accurate determination of phylogenetic relationships among metazoans. It is the first time that the complete mt genome sequence of the Huang Lang chicken was reported in this work, which was determined through the polymerase chain reaction-based method. The total length of the mitogenome is 16,786 bp, with the base composition of 30.25% for A, 23.71% for T, 32.53% for C and 13.51% for G, in the order C > A > T > G feature occurs in the Huang Lang chicken. It contains the typical structure, including two ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes and 1 non-coding control region (D-loop region). The complete mt genome sequence of the Huang Lang chicken provided an important data for further study on the genetic mechanism. © 2014 Informa UK Ltd.
