摘要:
Cucumber is an important vegetable crop and a model crop for the study of sex expression in plants. However, the genomic resources and tools for functional genomics studies in cucumber are still limited. In this paper, we conducted ethyl methyl sulfone (EMS) mutagenesis in the northern China ecotype cucumber inbred line 406 to construct a mutant library. We optimized the conditions of EMS mutagenesis on inbred line 406 which included treatment of seeds at 1.5% EMS for 12 h. We obtained a number of mutant lines showing inheritable morphological changes in plant architecture, leaves, floral organs, fruits and other traits through M_1, M_2 and M_3 generations. The F_2 segregating populations were constructed and analyzed. We found that a short fruit mutant and a yellow-green fruit peel mutant were both under the control of a single recessive gene, respectively. These results provide valuable germplasm resources for the improvement of cucumber genetics and functional genomic research.

摘要:
南瓜白粉病是一种危害严重的真菌病害,研究不同病情等级下细菌群落组成的异同对进一步研究南瓜白粉病的生物防治具有重要意义。采集来自3个不同发病等级的南瓜白粉病叶片(R1、R2和R3),运用高通量测序方法分析比较了不同发病等级的南瓜叶片的细菌群落结构和多样性,发现三个不同病情等级下细菌群落结构存在显著差异。在门水平上,三个病情等级下优势门均为Actinobacteria(放线菌门) 、Bacteroidetes(拟杆菌门) 、Proteobacteria(变形菌门)和Firmicutes(厚壁菌门);在属水平上,R1病情等级下优势属是Methylobacterium、Sphingomonas和Acinetobacter,R2病情等级下为Acinetobacter、Kluyvera和Pantoea,R3病情等级下为Methylobacterium、Kluyvera和Pantoea。南瓜白粉病菌可以改变南瓜叶际细菌群落结构,影响细菌群落多样性。Kluyvera、Pantoea和Erwinia的相对丰度与病情等级呈正相关,随着病情严重发生,丰度得到增强,表明它们可能与白粉菌存在协同作用。这些结果对进一步研究南瓜叶际微生态从而防控白粉病提供了科学参考依据。

The degenerin/epithelial sodium channel (DEG/ENaC) superfamily of ion channels contains subfamilies with diverse functions that are fundamental to many physiological and pathological processes, ranging from synaptic transmission to epileptogenesis. The absence in mammals of some DEG/ENaCs subfamily orthologues such as FMRFamide peptide-activated sodium channels (FaNaCs), which have been identified only in mollusks, indicates that the various subfamilies diverged early in evolution. We recently reported that the nonproton agonist 2-guanidine-4-methylquinazoline (GMQ) activates acid-sensing ion channels (ASICs), a DEG/ENaC subfamily mainly in mammals, in the absence of acidosis. Here, we show that GMQ also could directly activate the mollusk-specific FaNaCs. Differences in ion selectivity and unitary conductance and effects of substitutions at key residues revealed that GMQ and FMRFamide activate FaNaCs via distinct mechanisms. The presence of two activation mechanisms in the FaNaC subfamily diverging early in the evolution of DEG/ENaCs suggested that dual gating is an ancient feature in this superfamily. Notably, the GMQgating mode is still preserved in the mammalian ASIC subfamily, whereas FMRFamide-mediated channel gating was lost during evolution. This implied that GMQ activation may be essential for the functions of mammalian DEG/ENaCs. Our findings provide new insights into the evolution of DEG/ENaCs and may facilitate the discovery and characterization of their endogenous agonists.<br/> ©2017 by The American Society for Biochemistry and Molecular Biology, Inc.

摘要:
P2X receptors are ATP-gated trimeric channels with important roles in diverse pathophysiological functions. A detailed understanding of the mechanism underlying the gating process of these receptors is thus fundamentally important and may open new therapeutic avenues. The left flipper (LF) domain of the P2X receptors is a flexible loop structure, and its coordinated motions together with the dorsal fin (DF) domain are crucial for the channel gating of the P2X receptors. However, the mechanism underlying the crucial role of the LF domain in the channel gating remains obscure. Here, we propose that the ATP-induced allosteric changes of the LF domain enable it to foster intersubunit physical couplings among the DF and two lower body domains, which are pivotal for the channel gating of P2X4 receptors. Metadynamics analysis indicated that these newly established intersubunit couplings correlate well with the ATP-bound open state of the receptors. Moreover, weakening or strengthening these physical interactions with engineered intersubunit metal bridges remarkably decreased or increased the open probability of the receptors, respectively. Further disulfide cross-linking and covalent modification confirmed that the intersubunit physical couplings among the DF and two lower body domains fostered by the LF domain at the open state act as an integrated structural element that is stringently required for the channel gating of P2X4 receptors. Our observations provide new mechanistic insights into P2X receptor activation and will stimulate development of new allosteric modulators of P2X receptors.

Photosynthetic bacteria (PSB) have been extensively used in agriculture to promote plant growth and to improve crop quality. Their potential application in plant disease management, however, is largely overlooked. In this study, the PSB strain Rhodopseudomonas palustris GJ-22 was investigated for its ability to induce resistance against a plant virus while promoting plant growth. In the field, a foliar spray of GJ-22 suspension protected tobacco plants against tobacco mosaic virus (TMV). Under axenic conditions, GJ-22 colonized the plant phyllosphere and induced resistance against TMV. Additionally, GJ-22 produced two phytohormones, indole-3-acetic acid and 5-aminolevulinic acid, which promote growth and germination in tobacco. Furthermore, GJ-22-inoculated plants elevated their immune response under subsequent TMV infection. This research may give rise to a novel biological agent with a dual function in disease management while promoting plant growth.

摘要:
采用PMA和ox–LDL构建THP–1巨噬细胞源性泡沫细胞模型,观察不同浓度(5、10、15 nmol/L)仙人掌多糖(ODP–Ia)对泡沫细胞内脂质、胆固醇蓄积以及介导胆固醇流出的ABCA1、ABCG1、SR–BI基因表达的影响。结果显示:THP–1巨噬细胞泡沫化后胞内蓄积了大量脂质,高剂量(15 nmol/L)ODP–Ia能够显著减少胞内脂质、胆固醇蓄积,并增强ABCA1、ABCG1、SR–BI mRNA及蛋白表达(与单独ox–LDL处理组比较,P<0.05),但效果均次于阳性对照依折麦布组(P<0.05),低、中剂量(5、10 nmol/L)ODP–Ia作用效果均不显著。本研究结果表明, ODP–Ia可通过增强ABCA1、ABCG1、SR–BI基因表达来减少胞内胆固醇蓄积。

【目的】探究不同饲料饲养黄牛瘤胃厌氧真菌多样性的差异,了解芒草单一饲料饲养时瘤胃中特殊的真菌种类。【方法】通过构建芒草单一喂养、混合饲料饲养的黄牛瘤胃微生物18S rRNA基因文库,来对比分析单一饲喂芒草对瘤胃厌氧真菌多样性的影响。【结果】发现持续喂养18个月后,混合饲料饲养牛瘤胃真菌包含的种类较分散,无明显的优势类别,而芒草单一喂养牛瘤胃真菌包含的种类相对集中,主要集中在Orpinomyces sp. ,Piromyces sp. ,Neocallimastix frontalis,Microdochium sp.几种。【结论】使用芒草单一饲养确实改变了黄牛瘤胃中真菌的组成,并且其中优势真菌类群很可能与芒草的降解有密切关系,为今后开发瘤胃微生物资源在生物能源方面的利用奠定基础。

Enterobacter sp. strain ODB01, which was isolated from the Changqing oil field, can degrade crude oil efficiently and use crude oil as its sole source of carbon and energy. We report the complete genome sequence of ODB01. The results promote its application in the remediation of petroleum contaminants. © 2017 Lan et al.

生物农药的使用极大地减少了对环境的污染,探究生物农药对致病菌的细菌多样性及群落分布将为后续研究生物农药对致病菌的微生态调控提供理论依据.本研究运用Illumina MiSeq高通量测序技术分析比较辣椒健康植株与疫病发病植株根际土壤微生物多样性,以及研究沼泽红假单胞菌PSB06对植株根际土壤的微生物多样性影响,探究沼泽红假单胞菌PSB06对辣椒疫病的微生态调控机制.结果显示,在第7 d和第14 d的来自同一处理的根际土壤细菌群落多样性变化没有显著差异,辣椒疫病发病植株根际土壤微生物多样性均小于健康植株根际土壤微生物多样性且喷洒沼泽红假单胞菌PSB06发酵液的土样微生物多样性最高;辣椒疫病发病植株根际土壤中放线菌丰度均小于健康植株且喷洒沼泽红假单胞菌菌剂PSB06的土壤中放线菌的丰度最高.辣椒发病植株与健康植株根际土壤中微生物多样性存在显著差异. 施用沼泽红假单胞菌可以改善土壤微生物区系,提高土壤微生物群落丰富性以及土壤中放线菌所占的丰度.

摘要:
A short blossom period is a major obstacle for the tree peony industry. In order to preferentially isolate genes associated with flower senescence in the tree peony variety ‘Lu He Hong’, forward and reverse suppression subtractive hybridization cDNA libraries were constructed using senescent and blooming petals as testers, respectively. A total of 712 high-quality expressed sequence tags, representing 211 unigenes, were obtained, and 181 unigenes were annotated on the basis of sequence similarity with data in the National Center for Biotechnology Information’s non-redundant database. In addition, 168 unigenes were functionally annotated using Kyoto Encyclopedia of Genes and Genomes databases, and 44 unigenes were classified into 14 functional categories using the Clusters of Orthologous Groups. Most of the unigenes appear to be involved in cellular components, biological processes, and molecular functions, according to Gene Ontology analysis. Analysis of 23 chosen genes using quantitative reverse transcription PCR confirmed that their transcription levels were consistent with the results of the suppression subtractive hybridization libraries. © 2016 The Journal of Horticultural Science & Biotechnology Trust.

摘要:
Camellia oleifera Abel, a woody oil plant of major economic value, has strong ability for stress resistance. However, insufficient genetic and genomic information hinders the research into the mechanisms of its stress response. In this work, Illumina Genome sequencing platform was used for de novo assembling the transcriptomes of leaves from C. oleifera seedlings grown under optimal (control) and drought conditions. A total of 66570 unigenes with a mean length of 659.78 bp were assembled, amongst which 35259 unigenes could be annotated using the NCBI nr database, Swiss-Prot protein database, Cluster of Orthologous Groups of protein (COG) and Kyoto Encyclopedia of Genes and Genomes (KEGG) database. In addition, 10869 simple sequence repeats (SSRs) were mined in the leaf transcriptome of C. oleifera. In a comparative transcriptome analysis, when large numbers of differentially expressed unigenes (DEUs) were detected at different stages of drought stress, most unigenes were downregulated under the stress. In the KEGG pathway enrichment analysis, some important KEGG metabolic pathways of C. oleifera were discovered, such as circadian rhythm, flavone and flavonol biosynthesis, and ribosomal structure. Our studies provide a comprehensive map of physiological and molecular responses of C. oleifera to drought stress.

A novel substrate of Ca<inf>14</inf>Ga<inf>10-m</inf>Al<inf>m</inf>Zn<inf>6</inf>O<inf>35</inf>(CGAZO) was successfully synthesized via a high-temperature solid-state reaction method and co-doped with Dy<sup>3+</sup>and Mn<sup>4+</sup>to form a new deep red phosphor (CGAZO:Dy<sup>3+</sup>,Mn<sup>4+</sup>) for plant growth LED lighting. This extraordinary phosphor can exhibit strong far-red emission with a maximum peak at 715 nm between 650 and 750 nm, and other four shoulder peaks at 680, 689, 698 and 706 nm, which can be ascribed to 2E<inf>g</inf>→2A<inf>g</inf>spin-forbidden transition of Mn<sup>4+</sup>. X-ray diffraction (XRD) patterns clarified that Ga<sup>3+</sup>sites in the host were partly replaced by Al<sup>3+</sup>ions. Moreover, an effective energy transfer from Dy<sup>3+</sup>to Mn<sup>4+</sup>in the CGAZO host had been verified since linear reduction of emission intensities and decay times of the Dy<sup>3+</sup>emission of the phosphors CGAZO:Dy<sup>3+</sup>,Mn<sup>4+</sup>with increasing concentration of Mn<sup>4+</sup>was observed. Finally, a LED device was fabricated using a 460 nm blue-chip, silicon gel, and the as-obtained far-red emitting CGAZO:Dy<sup>3+</sup>,Mn<sup>4+</sup>phosphors. The results of the tobacco plant cultivation lighting by this LED device indicate that the as-prepared phosphor has a potential application in the agricultural industry. ©2017 The Royal Society of Chemistry.

摘要:
从长庆油田被原油污染土壤中筛选出1株以原油为唯一碳源的菌株ODB01。经菌株生理生化特征及16S rDNA序列分析,鉴定为肠杆菌属(Enterobacter sp.)细菌;运用响应曲面法优化细菌ODB01对原油的降解条件为pH值为8.91,w(NaCl)为1.19%,油菌比为1∶4.12,温度为36.78 ℃,在该条件下细菌ODB01对原油的降解率为34.6%;分别添加w=0.05%土温80(Tween 80)和w=0.05%辛基苯基醚(Triton X-100)后,细菌ODB01对原油降解率分别达到42.5%和46.1%。结果表明,细菌ODB01对原油具有一定的降解能力,有望作为一种微生物修复剂进行开发。

摘要:
The acidic environment has been confirmed to have significant influence on the biochar products through hydrothermal carbonization. To investigate the effect of H<inf>3</inf>PO<inf>4</inf>on the surface properties and adsorption mechanisms of the biochar, fresh banana peels were used as feedstock and transformed into hydrochars under the catalysis of H<inf>3</inf>PO<inf>4</inf>with concentration ranges from 0% to 50%wt. The addition of H<inf>3</inf>PO<inf>4</inf>greatly impacted the physicochemical properties of the as-obtained hydrochars, such as carbonization degrees, pH values, amount of surface acidic functional groups and then further affected the adsorption mechanisms of lead ions on those hydrochars. Hydrochars catalyzed by H<inf>3</inf>PO<inf>4</inf>with higher concentration showed higher degree of carbonization, lower value of pH and fewer amount of acidic functional groups, while lower content of H<inf>3</inf>PO<inf>4</inf>could not completely catalyze the degradation reactions and resulted in larger quantity of intermolecular locked oxygen containing groups that could not be participated into the adsorption process. Among all six samples, hydrochar generated in 30%wt H<inf>3</inf>PO<inf>4</inf>exhibited the best adsorption property of 241 mg g<sup>−1</sup>, possibly due to the largest concentration of the acidic functional groups on the surface rather than in the intermolecular structure of the carbon. It had been confirmed that the adsorption mechanisms were the combination of surface complexation, cationic exchange and part of precipitation. Hydrochars derived from fresh banana peel via hydrothermal carbonization catalyzed by phosphoric acid could be excellent adsorbents for lead removal in aqueous environments. ©2017 Elsevier Ltd

摘要:
Fresh and dehydrated banana peels were used as biomass feedstock to produce highly effective sorbent biochars through a facile one-step hydrothermal carbonization approach with 20%vol phosphoric acid as the reaction medium. The elemental ratio of oxygen content of the two as-prepared biochars were about 20%, and the FT-IR analysis confirmed the existence of abundant surface functional groups such as hydroxyl and carboxyl which greatly enhanced the adsorption performance. The sorbents showed excellent lead clarification capability of 359mg·g−1and 193mg·g−1for dehydrated and fresh banana peels based biochars, respectively. The change of the C[dbnd]O/O–C[dbnd]O and the appearance of Pb–O/Pb–O–C on the surface after adsorption confirmed that the ion exchange might be the dominant mechanism. The dehydration and pulverization pre-treatment and the addition of phosphoric acid can benefit the formation of those functional groups and hydrothermal carbonization can be a promising method to transfer biomass like fruit peels into biochars with excellent adsorption performance. © 2017 Elsevier Ltd

摘要:
微生物在重金属污染治理中具有重要作用.采用微生物纯培养技术从镉污染土壤中分离获得一株在液体培养基和固体培养基中能够分别耐受45 mmol·L~(-1)和50 mmol·L~(-1)Cd~(2+)的细菌,通过形态学、生理生化及分子鉴定,初步鉴定该菌株属于链霉菌(Streptomyces sp.),命名为 Streptomyces sp. strain CdTB01.进一步实验结果表明,耐镉链霉菌Streptomyces sp. strain CdTB01的优化培养条件为:马铃薯蔗糖培养基,在250 mL的三角瓶中装液量为90 mL,培养温度30 ℃,初始pH= 6.0,接种量6.0%,NaCl质量分数0%;湿菌体吸附Cd~(2+)的优化条件为:温度30 ℃,pH= 8.0,NaCl质量分数1.0%,Cd~(2+)浓度500 mg·L~(-1),吸附率为44.78%;干菌粉吸附Cd~(2+)的优化条件为:温度30 ℃,pH= 9.0,NaCl质量分数0%, Cd~(2+)浓度100 mg·L~(-1),吸附率达到70.45%.说明链霉菌Streptomyces sp. strain CdTB01在重金属污水处理中具有广阔的应用前景.

摘要:
In this paper, LiTi<inf>2</inf>(PO<inf>4</inf>)<inf>3</inf>@carbon composite as anode for aqueous lithium ion battery has been successfully synthesized by in-situ carbon coating method using oxalic acid and glucose as double carbon sources. XRD results confirm the rhombohedral NASICON-type LiTi<inf>2</inf>(PO<inf>4</inf>)<inf>3</inf>structure of the as-prepared composite. The LiTi<inf>2</inf>(PO<inf>4</inf>)<inf>3</inf>@carbon composite using double carbon sources (LTP-OG-10) disperses well, and the coated carbon layer has the appropriate thickness of 3–7 nm, which result in proper balance of the good electrical conductivity and the fast kinetics of diffusion of lithium ions. LTP-OG-10 exhibits better electrochemical performance compared with those using oxalic acid (LTP-O) or glucose (LTP-G) single carbon source. LTP-OG-10 anode delivers the discharge capacity of 112.5 mA h g<sup>−1</sup>, 96.8 mA h g<sup>−1</sup>, and 54.2 mA h g<sup>−1</sup>at 0.2 C, 5 C and 20 C, respectively, much larger than that of LTP-O and LTP-G. Moreover, LTP-OG-10 demonstrated excellent cycling performance, with the capacity retention of 71.1% after 400 cycles at 0.2 C, 22.9% higher than that of LTP-G. The good electrochemical properties of LTP-OG-10 is mainly ascribed to the increase of defects, high-quality and appropriate thickness of carbon layer. Our study reveals that preparing LiTi<inf>2</inf>(PO<inf>4</inf>)<inf>3</inf>@carbon composite using double carbon sources is an efficient approach to enhance its electrochemical properties. ©2017 Elsevier Ltd and Techna Group S.r.l.

摘要:
Conventional chemically oxidative polymerization with gradually added oxidant procedure primarily produces the agglomerated products ofpoly(m-phenylenediamine)(PmPD) due to secondary growth. This research developed a simple route to improve the conventional polymerization by just adjusting temperature to synthesize poly(m-phenylenediamine) nano-particles without catalyst or template. Morphology, molecular structure and properties of the polymer were investigated. Results showed that temperature of synthesis has a significant influence in PmPD micromorphology. Higher temperature would potentially lead to PmPD nano-particles. The possible formation mechanism of nano-particles has been proposed. The obtained PmPD nano-particles with high thermal stability have larger specific surface area, as compared with the micro-particles obtained by conventional method. Moreover, poly(m-phenylenediamine) nano-particles exhibit a much stronger sulfate ions adsorption capacity (93.5 mg g-1) than the micro-particles (71.2 mg g-1), which is also apparently superior to the performance reported in previous literatures(23-30). © Prints Publications Pvt. 摘要:
瞬时受体电位香草酸亚型2(TRPV2)属于瞬时受体电位离子通道家族,是一种非选择性阳离子配体门控通道。现有研究表明TRPV2在某些癌症类型和肿瘤细胞系中表达过量,在一些癌细胞中,通过TRPV2的激活剂(生长因子、激素、大麻素)激活TRPV2,发现TRPV2参与肿瘤细胞生长、细胞死亡和转移,因此TRPV2对肿瘤的发生具有重要作用。我们就TRPV2在膀胱肿瘤、前列腺肿瘤和神经胶质瘤中的影响做简要综述。
