刺五加(Acanthopanax senticosus, AS)中含有皂苷类、多糖和黄酮等多种活性成分,具有抗氧化、增强免疫功能、抗肿瘤及改善动物生产性能的作用,它作为一种中草药饲料添加剂可有效缓解仔猪断奶应激综合症。文章综述了AS的生物学功能及在畜牧生产上的应用,旨在为刺五加畜牧生产领域的应用及深入的研究提供理论依据。

摘要:
对于畜牧业来说,疫病的防控具有举足轻重的作用,新技术在畜牧兽医站的推广至关重要。通过不断应用新技术,可以使畜牧兽医站在动物疫病防控中的作用发挥到最大,不断满足畜牧业在新条件下的新需求,不仅如此,新技术的使用,可以推动畜牧养殖经济的发展,在一定程度上减少经济损失。有效控制动物疫病,畜牧业发展势头呈上升状态,也推动了国家经济的向上向好发展,由此可见新技术的应用具有不可或缺的作用。

摘要:
Porcine parvovirus (PPV) is considered one of the most important infectious agents of reproductive failure in sows. Little information, however, is available on its prevalence in healthy fattening pigs. Therefore, in the present study, 197 fecal swabs, 197 nasal swabs, 389 serum samples, and 310 lung samples were collected from pigs aged 10-25 weeks across Hunan, China, and tested for the presence of PPV. PPV DNA was amplified by polymerase chain reaction, demonstrating an overall positivity rate of 7.69%, with a particularly high infection rate of 22.90% in the lungs. A total of five PPV strains (PPV-HuNl-5) were isolated on the basis of cytopathic effects in swine testicular cells, and the near-complete genomes were sequenced and subjected to phylogenetic analysis with reference to reported PPV sequences in GenBank. The five Hunan isolates showed a close relationship with each other and predominantly with reported European PPV strains. Moreover, seven amino acid substitutions were detected within the coding region of the VP2 of PPV-HuNs when compared with that of the Chinese vaccine strain PPV-NJ. The relatively high prevalence of PPV discovered in healthy fattening pigs despite a long-term vaccination program in China, highlights the need for improved prevention, monitoring, and control of related diseases in herds. © 2020 Polskie Towarzystwo Nauk Weterynaryjnych. <正>伪狂犬病又名奥叶兹基病(Aujeszky-s disease),是一种由伪狂犬病病毒(Pseudorabies virus,P RV)感染引起的包括猪、牛、羊、猫、犬、家兔等多种家畜和野生动物的以发热、奇痒及脑脊髓炎为主要特征的一种高度接触性、急性传染病。除猪以外的其他动物都呈致死性感染,猪是伪狂犬病的唯一自然宿主[1]。

河南省猪流行性腹泻病毒流行株的遗传变异分析

摘要:
为分析河南省猪流行性腹泻病毒(PEDV)S基因的遗传变异情况,设计并合成能够扩增N基因和S基因的引物,利用RT-PCR方法对2017年1月至2018年5月河南省11个地区规模猪场的178份病料进行检测、克隆和测序,并在氨基酸水平上将本研究分离株的S基因与国内外流行毒株的S基因进行比对分析.结果 显示,河南省11个地区的178份样品,PEDV阳性样品113份,阳性率63.5%(113/178).选取29株PEDV代表株进行氨基酸序列比较,结果显示,29株分离株与经典株CV777同源性为88.4%~90.3%,与经典株CV777相比,29株代表株在S1区域存在碱基的插入、缺失和变异现象;基于S基因的遗传进化分析表明,本研究的29株分离株均属于G2群,而以CV777为代表的经典株属于G1群.通过与经典株CV777的S蛋白进行抗原表位比对分析,在5~190 aa存在较大差异.本研究结果明确了目前河南省PEDV的遗传变异情况,为河南省猪流行性腹泻(PED)的诊断和防控提供参考依据.

摘要:
The purpose of this study was to analyze the correlation of the rate of Mycoplasma hyopneumoniae (M. hyo) in slaughter pigs with season, climate change and enzootic pneumonia (EP) lesions. We collected 530 slaughter pig lungs with suspected lesions from two slaughterhouses in different seasons and weather conditions from November 2014 to March 2017 in Changsha Hunan Province, China. The EP lesions of these lungs were quantified, and a PCR analysis was used to detect M. hyo in samples of bronchoalveolar lavage fluid (BALF). Twenty percent, 10%, and 9% of the lung specimens were scored 1-5,6-10, and > 11, respectively. Additionally, we found that 36% of all BALF samples tested positive for M. hyo. Among the lung specimens collected in winter, 41% scored 1 or more, and 53% tested positive for M. hyo. With respect to seasons, the lung specimens collected in summer showed the least number of EP lesions and the lowest positive testing rate for M. hyo. Of these specimens, 27% scored 1 or more, and 22% tested positive for M. hyo. Additionally, low temperature and fast temperature change (during 10 days before sampling) were associated with a higher rate of M. hyo detection in BALF. There was a positive correlation between the lung EP lesion score and the detection rate of M. hyo in the BALF of slaughter pigs. In conclusion, lung EP lesion scoring in slaughter pigs is of referential value to the evaluation of the dynamics of M. hyo infection in a swine population. It is essential to control the spread of M. hyo by careful management of swine populations, and the prevention and control of M. hyo in fattening pigs is of great significance to the eradication of the disease. © 2019 Medycyna Weterynaryjna. 摘要:
为了解猪链球菌病在湖南病猪群中不同血清型的流行情况,实验室于2015—2017年从湖南省部分病猪群中收集到疑似链球菌感染病料142份,分别进行细菌分离,并采用猪链球菌GDH特异性引物鉴定到106株猪链球菌,阳性率为74.65%(106/142)。然后采用猪链球菌33个血清型的特异性引物对这106个菌株进行PCR鉴定,结果 86.79%的菌株被鉴定到具体血清型,其中优势血清型为2型(1/2型)30.19%,其次是7型(11.32%)、8型(9.43%)和3型(6.60%)。

摘要:
Streptococcus suis is a zoonotic pathogen that harbors anti-oxidative stress genes, which have been reported to be associated with virulence. Serial passage has been widely used to obtain phenotypic variant strains to investigate the functions of important genes. In the present study, S. suis serotype 9 strain DN13 was serially passaged in mice 30 times. The virulence of a single colony from passage 10 (SS9-P10) was found to increase by at least 140-fold as indicated by LD50 values, and the increased virulence was stable for single colonies from passage 20 (SS0-P20) and 30 (SS0-P30). Compared to the parental strain, the mouse-adapted strains were more tolerant to oxidative and high temperature stress. Genome-wide analysis of nucleotide variations found that reverse mutations occurred in seven genes, as indicated by BLAST analysis. Three of the reverse mutation genes or their homologs in other bacteria were reported to be virulence-associated, including ide Ssuis in S. suis, a homolog of malR of Streptococcus pneumoniae, and a homolog of the prepilin peptidase-encoding gene in Legionella pneumophila. However, these genes were not involved in the stress response. Another gene, srtR (stress response transcriptional regulator), encoding an XRE family transcriptional regulator, which had an internal stop in the parental strain, was functionally restored in the adapted strains. Further analysis of DN13 and SS9-P10-background srtR-knock-out and complementing strains supported the contribution of this gene to stress tolerance in vitro and virulence in mice. srtR and its homologs are widely distributed in Gram-positive bacteria including several important human pathogens such as Enterococcus faecium and Clostridioides difficile, indicating similar functions in these bacteria. Taken together, our study identified the first member of the XRE family of transcriptional regulators that is involved in stress tolerance and virulence. It also provides insight into the mechanism of enhanced virulence after serial passage in experimental animals.

为了解猪肺炎支原体流行菌株遗传变异情况,选取2014- 2017年不同时间、地点收集的17头猪肺炎支原体阳性屠宰猪肺脏灌洗液DNA为模板,进行猪肺炎支原体P36(编码具有强免疫原性的胞浆蛋白)、P46(编码具有强免疫原性的表面蛋白)、P97 R1区、P146高变区(猪肺炎支原体黏附气管纤毛细胞的重要功能区域)基因的PCR扩增、测序。结合GenBank登录的猪肺炎支原体相关序列进行氨基酸序列的比对分析。结果表明,比对的序列之间, P36、P46蛋白氨基酸序列同源性均在98.9%以上,但P97功能区R1区、P146多变区在菌株间存在较大差异。11个屠宰猪样本和5个GenBank登录菌株P97 R1区重复氨基酸序列数量达到14个以上,而GenBank登录的3个疫苗相关菌株(168株、168-L株、J株)及另外4个菌株、4个屠宰猪样本重复序列为9~ 11个不等; P146高变区氨基酸序列的差异主要集中在2个重复氨基酸序列区(R1、R2),GenBank登录的168株、168-L株、J株以及另外2个菌株,1个屠宰猪样本在R1区出现较多氨基酸缺失。168株、168-L株,6个屠宰猪样本及另外3个GenBank登录菌株在R2区出现了3~ 9个不等的氨基酸缺失。利用DNAStar进行抗原表位预测发现,菌株间P97 R1、P146 R1、P146 R2区氨基酸序列的差异,导致了抗原表位出现明显的变化。提示开展P97、P146等黏附因子的筛选和研究或许能进一步提升疫苗阻止猪肺炎支原体黏附气管纤毛细胞的作用,从而进一步改善疫苗的免疫效果。

一例竹鼠感冒的诊治

竹鼠,又名竹根鼠、茅根鼠、竹狸、冬芒狸、竹纯、竹馏、稚子等。其肉质鲜美,倍受老百姓喜爱。野外竹鼠一般感冒很少发生,但人工圈养时就偶有感冒发生,笔者在当地一小规模竹鼠养殖场发现一年四季时不时会有竹鼠感冒发生,现就感冒发生的病因、临床症状、病理变化、预防与治疗措施进行分析。竹鼠感冒是由受寒冷刺激而引起的发热和上呼吸道黏膜发热的一种急性、全身性感染疾病,若不及时治疗,很容易继发支气管炎、肺炎和其他恶性疾病。

目的探讨立显蛋白染色液在SDS-PAGE中检测微量蛋白的应用效果。方法制备SDS-PAGE胶,采用立显蛋白染色液及普通考马斯亮蓝(Coomassie brilliant blue,CBB)染色液,检测定量的牛血清白蛋白(bovine serum albumin, BSA)的敏感度及猪细小病毒VP2蛋白、猪蓝耳病病毒N蛋白、猪丹毒杆菌Fe蛋白和猪伪狂犬病病毒gB蛋白基因工程纯化样品的纯度。结果采用立显蛋白染色液染色可识别蛋白质含量为2. 5 ng的蛋白条带,敏感性高于普通CBB染色40倍。4种蛋白纯化样品经CBB染色后检测纯度均较高,但经立显蛋白染色液染色后,4种纯化蛋白样品均检测出杂蛋白,测定的纯度均有所下降,其中VP2及gB蛋白的纯度有较大幅度下降(分别由83. 33%、100%降至51. 8%、85. 6%)。结论立显蛋白染色液染色SDS-PAGE胶中的蛋白条带敏感性高,快速简便,适用于微量蛋白质的检测。

转移因子对猪丹毒杆菌SpaA抗原免疫效果的影响

摘要:
为了探寻转移因子(Transfer factor,TF)对猪丹毒杆菌SpaA(Surface protective antigen A)蛋白抗原免疫效果的影响,将重组SpaA蛋白与双相佐剂乳化后(称SpaA抗原)免疫小鼠,取免疫小鼠和未免疫小鼠脾脏,以超滤法分别制备特异性TF和普通TF,并利用BCA法测定所得TF的质量分数,经过无菌及安全性检测后,将2种TF分别腹腔接种至小鼠体内,24 h后,对2组TF处理后的小鼠均免疫SpaA抗原,并以未接种TF的免疫小鼠作为对照(仅免疫组)。分别于免疫后12 d、21 d和30 d对小鼠进行采血,用间接ELISA法对小鼠血清进行抗体检测。结果表明,利用原核表达系统表达的重组SpaA蛋白分子质量60 ku且具有可溶性;以超滤法制备的特异性TF质量浓度为2.56 mg/mL,普通TF质量浓度为2.40 mg/mL,且2种TF安全无毒;抗体检测数据显示,2种TF处理组在免疫后21、30 d的抗体水平均高于仅免疫组,且差异显著,而2种TF处理组之间抗体水平差异不显著。因此,TF能诱导SpaA抗原快速产生抗体,提高SpaA抗原的免疫效果,但与TF的特异性关系不大,表明TF可作为猪丹毒杆菌基因工程亚单位疫苗的免疫增效剂,提高免疫动物的抗体水平。

某猪场猪伪狂犬病免疫程序优化对比试验

摘要:
为了探究保育猪不同伪狂犬病免疫程序的免疫效果,笔者采取4种不同免疫程序进行试验,即4组猪群于35日龄首免和70日龄二免:A组分别免疫接种伪狂犬病弱毒疫苗和灭活疫苗;B组分别接种伪狂犬病灭活疫苗和弱毒疫苗;C组均接种伪狂犬病灭活疫苗;D组分别接种伪狂犬病弱毒疫苗,接种方式和剂量均按照说明书进行。于试验前(25日龄)和试验后(100日龄)应用ELISA法分别检测不同组别的猪群伪狂犬病毒g E和g B抗体水平水平,确定阳性率差异。结果显示:免疫程序A和B可以均能有效控制猪群伪狂犬病野毒的进一步流行,降低猪群的死亡率,但免疫程序B无法使猪群伪狂犬病得到净化,而免疫程序C和D均无法抑制伪狂犬病毒野毒的流行。因此,选择免疫程序A对于防控与净化伪狂犬病毒的发生与流行效果最好。

为评估猪细小病毒7型(porcine parvovirus 7,PPV7)在猪群中的流行,根据GenBank 中PPV7全基因组序列设计1对扩增PPV7 NS1基因的特异性引物,并建立PCR方法.对该引物的退火温度进行优化,且分析其特异性和敏感性.结果显示,在52~72 ℃退火温度范围内该对引物均能特异性扩增出目的基因片段,最低检测限为3.6×102 copies / L.对不同来源的282份组织样品和来自湖南省4个不同猪场的226份血清样品进行检测,结果,组织样品中PPV7检出率为55.0%(155/ 282),血清中PPV7检出率为10.2%(23/226).其中,屠宰猪组织样品中PPV7检出率最高(71.4%),而PPV7感染对猪的健康是否存在影响还不能确定.本研究结果为PPV7流行病学的研究奠定了一定的基础.

Porcine teschovirus (PTV) comprises at least 13 genotypes (PTV 1–13). Here, the genotypes of field strains prevalent among pig populations in Hunan Province, China, were identified. Multiple PTV genotypes, including all genotypes except PTV 7 and 8, were found co-circulating in the pig populations, reflecting a high genetic diversity. Moreover, we identified nine novel PTV genotypes, provisionally designated as PTV 14–22. PTV 21-HuN41 and PTV 21-HuN42 were successfully isolated, and their nearly complete genomes were sequenced. Homology comparison of the polyprotein genes of PTV 21-HuN41–42 to those of other known PTVs revealed low identities, ranging from 70.1 to 71.9 % (nucleotide identity) and 75.4 to 77.6 % (amino acid identity). Moreover, PTV 21-HuN41–42 were identified as a novel teschovirus species (tentatively Teschovirus B), based on the analyses of phylogenetics and evolutionary divergence. The findings of this study are expected to greatly enrich our knowledge of PTV genotypes. © 2018 The Authors.

High prevalence of PCV2d in Hunan province, China: a retrospective analysis of samples collected from 2006 to 2016

Porcine circovirus 2 (PCV2) has been widely prevailing in China since the first report in 2001, causing huge economic losses to the pig industry. In the present study, 674 samples were collected from 2006 to 2016 in Hunan province, and 62% were positive for PCV2. An increase was observed from 2006 to 2011 (72.1%-89.1%), and a decrease was observed from 2012 to 2016 (78.9%-36.8%). The prevalence of genotype PCV2a, PCV2b, and PCV2d was 0, 44.7% and 67%, respectively. During 2006-2007, PCV2b was the main genotype circulating in Hunan, while, in 2008, PCV2d became the predominant one. Coinfection with PCV2b and PCV2d was observed frequently, and the positive rates of coinfection ranged from 6.3% to 18.9% during 2006-2016. The complete genome was sequenced for 54 positive samples, and four were identified as PCV2b-1, 22 as PCV2b-2, four as PCV2d-1 and 24 as PCV2d-2, based on phylogenetic analysis of the complete genome and ORF2 region. Recombination analysis using the complete genome sequences of these isolates revealed a high recombination rate of 27.7% (17/54), and showed that recombination occurred mainly in the ORF1 region. This shows that the prevalence of PCV2 has clearly decreased in recent years and that PCV2d has become a predominant genotype since 2008. In addition, frequent recombination events were observed in the PCV2 isolates from Hunan, China.

摘要:
以纯化的猪传染性胃肠炎病毒(TGEV)重组N蛋白免疫BALB/c小鼠,按照常规方法融合、筛选,最终获得2株抗TGEV N蛋白的杂交瘤细胞系,分别命名为2G7和2C2 。2株单抗亚类均为IgG1,腹水的间接ELISA效价均为1 ∶ 10~8。2株杂交瘤细胞连续培养20代,各代次的上清效价保持不变,均为1 ∶ 12 800,说明2株细胞分泌抗体的能力稳定。通过间接免疫荧光发现,制备的2株单克隆抗体能与感染TGEV的细胞发生特异性反应,在胞浆和细胞膜上有亮绿色荧光。结果表明,所制备的2株TGEV N蛋白的特异性单克隆抗体细胞株遗传稳定,分泌的抗体敏感性高,为TGEV病原检测奠定了基础。

哺乳仔猪腹泻为猪场常见病,病因复杂,影响仔猪生长发育以及猪场经济效益。河南某猪场2017年夏季哺乳仔猪腹泻发病较多,根据仔猪腹泻的发病情况以及临床症状等表现,初步判断多为非病理性腹泻。对仔猪进行补液、抗菌等治疗,对母猪采取补充营养等措施,仔猪腹泻情况有明显好转。

2017年9月,湖南省郴州市某猪场的怀孕母猪突然发生流产、死亡。为确诊病因,对该猪场病死猪脏器进行了细菌分离和PCR检测,结合临床症状最终确诊病原为猪丹毒丝菌。为了解整个猪场猪群的感染情况,随机抽取流产母猪和同栏舍健康母猪进行猪丹毒丝菌IgG抗体ELISA检测,发现流产母猪阳性率为90%,远高于同栏舍健康母猪(10%)。本次疫情共导致该猪场104头母猪死亡和94头母猪流产。本案例表明,猪丹毒不仅可以引起母猪死亡,也可引起母猪流产,应引起养殖场高度重视,建议把猪丹毒列入免疫计划。

生猪乙型脑炎抗体消长规律及蚊子携带乙脑病毒调查

摘要:
为探寻猪场仔猪乙型脑炎抗体消长与蚊子携带乙型脑炎病毒的之间的关系,4月份选取一批约15日龄仔猪共24头,每隔20 d采血1次,至7月共采血6次,用间接ELISA方法检测血清乙型脑炎抗体,且不同时间在跟踪猪舍共捕获约3720只蚊子,用RT-PCR方法鉴定乙型脑炎病毒核酸;结果表明,该批猪乙型脑炎抗体阳性率总体呈现先下降后升高规律,阳性率先后为83.3%、66.7%、41.6%、25%、50%、91.6%,生猪体内母源抗体水平在75日龄(6月20日)达到最低,阳性率降至25%,然后呈现明显的上升趋势,115日龄(7月30日)检测阳性率高达91.6%;37份蚊子样品,JEV核酸阳性样品16份,阳性率高达43.2%,其中7月份所采集14份蚊子样品检出7份,乙型脑类病毒(JEV)阳性检出率最高达50%,表明在乙型脑炎流行季节,受蚊子携带病毒及其叮咬影响,当仔猪JEV抗体下降最低时JEV易感,后期感染抗体升高。
