摘要:
Jasmonate (JA) inhibits root growth of Arabidopsis thaliana seedlings. The mutation in COI1, that plays a central role in JA signaling, displays insensitivity to JA inhibition of root growth. To dissect JA signaling pathway, we recently isolated one mutant named psc1, which partially suppresses coi1 insensitivity to JA inhibition of root growth. As we identified the PSC1 gene as an allele of DWF4 that encodes a key enzyme in brassinosteroid (BR) biosynthesis, we hypothesized and demonstrated that BR is involved in JA signaling and negatively regulates JA inhibition of root growth. In our Plant Physiology paper, we analyzed effects of psc1 or exogenous BR on the inhibition of root growth by JA. Here we show that treatment with brassinazole (Brz), a BR biosynthesis inhibitor, increased JA sensitivity in both coi1-2 and wild type, which further confirms that BR negatively regulates JA inhibition of root growth. Since effects of psc1, Brz and exogenous BR on JA inhibition of root growth were mild, we suggests that BR negatively finely regulates JA inhibition of root growth in Arabidopsis. © 2010 Landes Bioscience.

摘要:
The chlorogenic acids of Gardeniae Fructus used traditionally as a Chinese herbal medicine (zhizi) have been investigated qualitatively by liquid chromatography/multi-stage mass spectrometry (LC/MS4). Twenty-nine chlorogenic acids were detected and twenty-five characterised to regioisomer level on the basis of their fragmentation, twenty-four for the first time from this source. Assignment to the level of individual regioisomers was possible for three caffeoylquinic acids, three dicaffeoylquinic acids, three sinapoylquinic acids, four caffeoyl-sinapoylquinic acids, two feruloyl-sinapoylquinic acids, one p-coumaroyl-sinapoylquinic acid, three (3-hydroxy, 3-methyl)glutaroyl-quinic acids, two (3-hydroxy, 3-methyl)glutaroyl-feruloylquinic acids, one (3-hydroxy, 3-methyl)glutaroyl-dicaffeoylquinic acid, and one (3-hydroxy, 3-methynglutaroyl-caffeoyl-feruloylquinic acid. Six (3-hydroxy, 3-methyl)glutaroyl-caffeoylquinic acids were detected and two were tentatively assigned as 3-caffeoyl-4-(3-hydroxy, 3-methyl)glutaroylquinic acid and 3-caffeoyl-5-(3-hydroxy, 3-methyl)glutaroylquinic acid. The (3-hydroxy, 3-methyl)glutaroyl residue modifies the mass spectral fragmentation behavior and elution sequence compared with the chlorogenic acids that contain only a cinnamic acid residue(s). Tomato chlorotic dwarf viroid (TCDVd) manually inoculated to transgenic (cv.'Desiree') potato plants containing antimicrobial cationic peptides failed to develop symptoms in above ground plant parts, but infected tubers were symptomatic. Plants from the infected tubers (second generation plants) emerged as either severely stunted (bushy stunt isolate, BSI) or tall and symptomless. Molecular characterization of BSI isolates showed TCDVd sequence variants 95 to 98% identical to TCDVd sequences from the database, while a viroid variant identical to TCDVd type isolate (acc # AF162131) was cloned from symptomless plants. The TCDVd BSI variants had novel U165C, GU177-178AA, and UCAC181-184CUUU nucleotide substitutions in the terminal right (TR) domain of the viroid molecule. The cloned viroid cDNAs of the BSI were infectious to experimental (cv. 'Sheyenne') tomato plants causing stunted plants with profuse auxiliary shoots. Visual evaluation of the susceptibility of the BSI to 18 potato and 21 tomato cultivars revealed severe symptoms in most cultivars of both species. The progeny variants accumulating in each potato and tomato cultivar exhibited the same novel TR domain in most cultivars, with only a slight variation in a few. The severity of the stunting symptoms induced by TCDVd from BSI isolates in both potato and tomato cultivars has not been noted previously with other TCDVd isolates and, as such, it is proposed that this new isolate be recognized as a distinct genotype. Emergence of this type of sequence variant in commercial fields or commercial tomato greenhouses could potentially cause relevant losses in both crops. © 2009 KNPV.

Two highly water-soluble amino acids,which derived fromβ-CDs,i.e.,glutamic acid-β-cyclodextrin(GluCD)and ethylene-diamine-β-cyclodextrin(EDCD),were synthesized and were examined for their effect on solubilization of anthracene(ANT),complexation of cadmium(Cd~(2+)),and elution removal of ANT and Cd~(2+)in soil.The results showed that GluCD and EDCD were powerful complexant for ANT and Cd~(2+).In the presence of 10 g/L GluCD and EDCD,the solubilization of ANT increased by 47.04 and 23.85 times compared to the control,respectively.GluCD resulted in approximately 90%complexation of Cd~(2+)while 70%complexation was observed for EDCD.Simultaneously,GluCD and EDCD could greatly enhance the elution removal of ANT and Cd~(2+)from soil.GluCD resulted in the highest elution effciency of ANT and Cd~(2+).With the addition of 10 g/L GluCD,53.5%of ANT and 85.6%of Cd~(2+)were eluted,respectively.The ANT had a negligible effect on the Cd~(2+)removal due to different complexing sites of ANT and Cd~(2+),while Cd~(2+)enhanced the ANT removal under the addition of GluCD because Cd~(2+)neutralized the-COOH group of GluCD.Adversely,the removal of ANT was decreased with Cd~(2+)under the addition of EDCD,this was due to the fact that Cd~(2+)enhanced the polarity of EDCD molecule and inhibited the complexation between ANT and EDCD.The study suggested that GluCD could be preferred and be successfully applied to remediation of heavy metals or organic compounds in contaminated soil

摘要:
The objective of this study was to explore the effects of Gynostemma pentaphyllum on Zearalenone-induced apoptosis in mouse male germ cells. Fifty Kunming male mice at 25-days-old were classified into five groups: group A was the control (10% ethanol, 0.5 ml/day); group B with 10 μg Zearalenone/day; group C with 10 μg Zearalenone and 50 mg/kg/day Gynostemma pentaphyllum; group D with 10 μg Zearalenone and 100 mg/kg/day Gynostemma pentaphyllum; and group E with 10 μg Zearalenone and 200 mg/kg/day Gynostemma pentaphyllum. It was found that Gynostemma pentaphyllum has a marked effect on protecting male germ cells against Zearalenone-induced apoptosis, as evidenced by a reduced apoptosis rate of male germ cells and Bax expression as well as an enhancement of Bcl-2 expression in Gynostemma pentaphyllum-treated groups compared to the control. In addition, Gynostemma pentaphyllum remarkably improved pathologic changes of testicular tissue, reduced the content of malondialdehyde (MDA), and increased the activity of superoxide dismutase (SOD) caused by Zearalenone. Taken together, these results suggest that Gynostemma pentaphyllum protects against toxicity caused by Zearalenone through anti-oxidation and anti-apoptosis via the regulation of Bax and Bcl-2 expression. © 2010 Informa UK Ltd.

摘要:
The objective of the present study was to evaluate the effect of dietary supplemented with copper, ferrous iron and zinc together from their proteinates or sulphates on growth performance, plasma concentration and balance of copper, ferrous iron and zinc in pigs at different growth stages. Three growth experiments and one mineral balance test were included in present study. In comparison with diet supplemented with copper, ferrous iron and zinc from sulfates together, diet supplemented with lower concentrations of copper, ferrous iron and zinc together from their proteinates significantly increased (P < 0.05) the feed efficiency in all three growth experiments and also increased (P < 0.05) the average daily gain (ADG) in Exp. 1, however, no difference of average daily feed intake (ADFI) in Exp. 1 and ADG ADFI and plasma concentrations of copper, ferrous iron and zinc were observed in Exp. 2 and 3, respectively. In mineral balance trail, pigs consumed copper, ferrous iron and zinc from their proteinates decreased copper, ferrous iron and zinc excretion by 91 % (P < 0.01 ), 45% (P < 0.01 ) and 71 % (P < 0.01 ) compared with pigs that consumed these minerals from sulfates, respectively. We concluded that dietary supplemented with copper, ferrous iron and zinc from their proteinate sources at the levels of 20, 100 and 40 mg/kg together have a growth-promoting effect on pigs at early growth stage and decrease copper, ferrous and zinc elimination into the environment.

摘要:
In this paper, the entanglement dynamics of a double two-photon Jaynes-Cummings model with Kerr-like medium is investigated. It is shown that initial entanglement has an interesting subsequent time evolution, including the so-called entanglement sudden death effect. It is also shown analytically that the Kerr-like medium can repress entanglement sudden death and enhance the degree of atom-atom entanglement. A more interesting fact is that the Kerr effect is more obvious when each of the two cavities with have the Kerr-like medium than only one of them with the Kerr-like medium.

摘要:
RanGTPase, one family of small G protein superfamily, has been widely demonstrated to be involved in transport system between cytoplasm and nucleus. However the knowledge about the function of RanGTPase in immunity remains limited. In this report, Ran gene (named LycRan) cDNA was cloned from the large yellow croaker, Pseudosciaena crocea, a marine fish. The full-length cDNA of LycRan was of 1033 bp, including a 5'-terminal untranslated region (UTR) of 43 bp, 3'-terminal UTR of 338 bp and an open reading frame (ORF) of 648 bp encoding a polypeptide of 216 amino acids. The deduced protein is highly homologous, it shares 90.74%, 88.89%, 89.35% and 85.20% identities with those of salmon, frog, human and fruit fly respectively. RT-PCR analysis indicated that LycRan gene was constitutively expressed in 9 tissues examined, including kidney, liver, gill, muscle, spleen, skin, heart, intestine and blood. The result of quantitative Real-Time RT-PCR analysis revealed the highest expression in kidney and the weakest expression in skin. Time course analysis showed that LycRan expression was obviously up-regulated in kidney, blood and spleen after immunization with either poly I:C or formalin-inactive Gram-negative bacterium Vibrio parahaemolyticus. It indicated that the highest expression was 2.8 times (at 48 h) as much as that in the control in the kidney (p < 0.05) challenged by poly I:C and 3.2 times (at 24 h) in the blood (p < 0.05) challenged by bacteria. These results suggested that LycRan might play an important role in large yellow croaker defense against the pathogen infection. Our study, therefore, might provide a clue to elucidate the large yellow croaker innate immunity. © 2010.

摘要:
A method for simultaneous concentration of multiple bioactive components (MBCs) in Danggui-Buxue Decoction (DBD) was comprehensively investigated in this work. The column chromatography with macroporous resin ADS-7 as an absorbent was used for concentration of the MBCs in DBD under the guidance of liquid chromatography (LC). After one-run treatment with the resin, the contents of major flavonoids reached 18.3 and 13.9% in the eluting fraction of 50 and 70% ethanol, which were 177.2 and 134.6 times higher than those in DBD crude extract. The contents of main saponins reached 8.7 and 9.9% in the eluting fractions of 10 and 30% ethanol, which were 32.8 and 37.3 times higher than those in DBD crude extract; and ferulic acid and ligustilide were also enriched in the eluting fractions of 10 and 70% ethanol. © 2010 Vieweg+Teubner Verlag | Springer Fachmedien Wiesbaden GmbH.

摘要:
An efficient method for the simultaneous extraction and quantification of both jasmonic acid (JA) and methyl jasmonate (MeJA) from a single plant sample was developed. Plant tissues were firstly extracted by using 100% cold methanol, then the Sep-pak C18 solid-phase extraction (SPE) cartridges were adopted for the purification of sample extract, and finally JA and MeJA were determined by the liquid chromatography tandem mass spectrometry (LC-MS-MS) system. It was found that the accuracy was improved by using the extra standard for the estimation and correction of JA and MeJA losses in the extraction and purification process. The detection limits for JA and MeJA were 0.03 and 0.075 ng mL-1, respectively, and the average recovery rate of JA and MeJA was 92.48% and 94.30%. The method was found to be reproducible and selective, yielding well isolated and easily detectable peaks for both JA and MeJA and simplifying the time-consuming extraction and purification. High-performance liquid chromatography (HPLC) system was employed as the control detection method for the LC-MS-MS system. The self-diffusion of single Pt adatom on the surface of cuboctahedral and truncated decahedral clusters with 561-10179 atoms are studied within the context of the many-body potentials obtained via the embedded atom method. The minimum energy diffusion path and the corresponding energy barrier for adatom diffusion on the cuboctahedral and truncated decahedral clusters surfaces are determined through a combination of the quenched molecular dynamics and the nudged elastic band method. The calculated energy barriers are consistent with the available experimental data. The dependence of energy barrier for adatom diffusion across the step edge on the cluster size is found. For the larger cuboctahedral and truncated decahedral clusters, the simulations show that the movement of the adatom is confined to a central region, and the adatom may escape from the center region only at elevated temperatures. In addition, we also find that the truncated decahedral structure is more favorable over the cuboctahedral structure for smaller clusters. Background: MicroRNAs (miRNAs) are considered to be very important in regulating the growth, development, behavior and stress response in animals and plants in post-transcriptional gene regulation. Pinewood nematode, Bursaphelenchus xylophilus, is an important invasive plant parasitic nematode in Asia. To have a comprehensive knowledge about miRNAs of the nematode is necessary for further in-depth study on roles of miRNAs in the ecological adaptation of the invasive species. Methods and Findings: Five small RNA libraries were constructed and sequenced by Illumina/Solexa deep-sequencing technology. A total of 810 miRNA candidates (49 conserved and 761 novel) were predicted by a computational pipeline, of which 57 miRNAs (20 conserved and 37 novel) encoded by 53 miRNA precursors were identified by experimental methods. Ten novel miRNAs were considered to be species-specific miRNAs of B. xylophilus. Comparison of expression profiles of miRNAs in the five small RNA libraries showed that many miRNAs exhibited obviously different expression levels in the third-stage dispersal juvenile and at a cold-stressed status. Most of the miRNAs exhibited obviously down-regulated expression in the dispersal stage. But differences among the three geographic libraries were not prominent. A total of 979 genes were predicted to be targets of these authentic miRNAs. Among them, seven heat shock protein genes were targeted by 14 miRNAs, and six FMRFamide-like neuropeptides genes were targeted by 17 miRNAs. A real-time quantitative polymerase chain reaction was used to quantify the mRNA expression levels of target genes. Conclusions: Basing on the fact that a negative correlation existed between the expression profiles of miRNAs and the mRNA expression profiles of their target genes (hsp, flp) by comparing those of the nematodes at a cold stressed status and a normal status, we suggested that miRNAs might participate in ecological adaptation and behavior regulation of the nematode. This is the first description of miRNAs in plant parasitic nematodes. The results provide a useful resource for further in-depth study on molecular regulation and evolution of miRNAs in plant parasitic nematodes. © 2010 Huang et al.

摘要:
The dissipation and residue behavior of hexaflumuron in Chinese cabbage (Brassica pekinensis) under different treatments were investigated at two main cabbage-growing areas in China. The dissipation rates of hexaflumuron in cabbages and soils fit a first-order decay process very well. The dissipation times for 50% of the hexaflumuron in cabbage and soil were 3.37 and 3.01 days from Hunan province and 5.58 and 3.68 days in Tianjin, respectively, when Chinese cabbage was treated with 180 g of active ingredient (ai)/ha. Hexaflumuron residual times in cabbage and soil were influenced strongly by the application rate and frequency. The application regimen of hexaflumuron in the cabbage field, a rate of 120 g of ai/ha with two applications at a 7 day interval and a 21 day preharvest interval, was recommended from the point of view of ensuring food safety. The residual levels of hexaflumuron in cabbages were lower than the standards of the "Positive List System for Agricultural Chemical Residues in Foods" based on the recommended application regimen. © 2010 American Chemical Society.

Tillering ability is one of the most important traits of the rice plant because it plays a major role in determining grain yield. However, it is not clear whether rice yield potential could be further increased if unproductive tillers were reduced or eliminated. Two field experiments were conducted to determine the effects of tiller reduction on plant growth and grain yield in two varieties with different tillering ability. Two methods were used to reduce tillers: the first was the removal of tillers manually at different crop growth stages and the second was to grow rice plants through holes in styrofoam slats, which provided a physical restriction on tiller emergence. Tiller number declined by 12-57% with the first method and by 18-47% with the second method compared with the control. Despite the significant reduction in tiller number and unproductive tiller percentage, none of the treatments with manual tiller removal and physical restriction of tillering resulted in a significant increase in grain yield compared with the control. Decreases in leaf area and biomass at heading and panicle number at maturity were observed when tiller number declined significantly, but grain yield was not necessarily reduced because it was compensated for by more biomass production after heading and increased spikelet number per panicle. The two varieties with different tillering ability had similar responses to tiller reduction. Based on the two methods of tiller reduction used in this study, we conclude that a further increase in rice yield potential may not be possible by reducing or eliminating unproductive tillers. © 2009 Elsevier B.V. 摘要:
On the basis of morphological examination, field investigation, observation of cultivated accessions and statistical analysis, the genus Miscanthus s.l. (Poaceae) from China was taxonomically revised. Two subgenera (Miscanthus subgenus Miscanthus and Miscanthus subgenus Diandranthus), two sections [Miscanthus subgenus Miscanthus section Miscanthus and Miscanthus subgenus Miscanthus section Triarrhena (Maximowicz) Honda], six species, two subspecies and four varieties in this genus were recognized in this report. Miscanthus sacchariflorus ssp. lutarioriparius and Miscanthus nudipes var. yunnanensis were also recognized. A key to the taxa of Miscanthus from China is provided. A morphological description, distribution, and habit and phenology are summarized for each species. High levels of polyunsaturated fatty acids (PUFA) in pork are beneficial and saturated fatty acids (SFA) detrimental to human health. Inclusion of PUFA in diet may decrease the amount of intramuscular fat (IMF) due to inhibition of de nova synthesis of fatty acids, which has a negative effect on meat eating quality. Diet energy level is a leading factor for lipid deposition in IMF. We hypothesize that increasing dietary energy level by including PUFA will increase its content without decreasing IMF in meat, thus increasing meat's nutritional value without lowering meat eating quality. The results of this study showed that increasing dietary energy level by inclusion of soy oil decreased average daily feed intake (ADFI) (P<0.01) and feed:gain ratio (P<0.01), increased average daily weight gain ADG (P<0.05) and serum triglyceride concentration (P<0.05) and decreased serum urea concentration (P<0.05). Carcass characteristics, meat quality and serum concentrations of hormones [insulin, growth hormone (GH), insulin-like growth factor-I (IGF-I), glucagon and tumor necrosis factor-alpha (TNF-alpha)] were unaffected. Percentages of SFA and monounsaturated fatty acids (MUFA) were decreased while percentages of PUFA were increased (P<0.05) in longissimus dorsi (LD) muscle as dietary energy level increased. The results of this study suggested that increasing dietary energy level by inclusion of soy oil could be a suitable way to improve growth performance as well as nutritional value of pork without lowering eating quality.

摘要:
Ethylene response factor (ERF) proteins regulate a variety of stress responses in plant. JERF1, a tomato ERF protein, can be induced by abscisic acid (ABA). Overexpression of JERF1 enhanced the tolerance of transgenic tobacco to high salt concentration, osmotic stress, and low temperature by regulating the expression of stress-responsive genes by binding to DRE/CRT and GCC-box cis-elements. In this research, we further report that overexpression of JERF1 significantly enhanced drought tolerance of transgenic rice. The overexpression activated the expression of stress-responsive genes and increased the synthesis of the osmolyte proline by regulating the expression of OsP5CS, encoding the proline biosynthesis key enzyme deltal-pyrroline-5-carboxylate synthetase. JERF1 also activated the expression of two ABA biosynthesis key enzyme genes, OsABA2 and Os03g0810800, and increased the synthesis of ABA in rice. Analysis of cis-elements of JERF1-targeted genes pointed to the existence of DRE/CRT and/or GCC box in their promoters, indicating that JERF1 could activate the expression of related genes in rice by binding to these cis-elements. Unlike some other ERF proteins, constructive overexpression of JERF1 did not change the growth and development of transgenic rice, which makes JEFR1 a potentially useful source in breeding for greater tolerance to abiotic stress.

Pure olivine LiFePO<inf>4</inf>/C composite compounds are synthesized by carbothermal reduction (CTR) method using trigonal anhydrous FePO<inf>4</inf> raw material. The physical and electrochemical properties of the prepared samples are investigated in comparison with those of a sample obtained from incompletely crystallized hydrous FePO<inf>4</inf>•2H<inf>2</inf>O. The olivine LiFePO<inf>4</inf>/C prepared with trigonal anhydrous FePO<inf>4</inf> has a high discharge capacity of 142 mAh g<sup>- 1</sup> at 0.1 C. The composite also displays a better rate capability, a higher discharge capacity and a more stable cycle-life than that synthesized with incompletely crystallized hydrous FePO<inf>4</inf>•2H<inf>2</inf>O. The improved electrochemical performance of trigonal anhydrous FePO<inf>4</inf> synthesis samples originates mainly from the single-phase structure and small particle size. ©2010 Elsevier B.V. Stamen is the male reproductive organ of rice (Oryza sativa L.), and the development is a considerably significant stage during the sexual reproduction. It is very important to reveal what is the molecular mechanisms which controlling the development of stamen, and will be helpful in producing hybrid seeds by manipulating the male sterility. This article reviews the progress on the molecular mechanisms of male reproductive organogenesis in rice, which would facilitate the further studies on male sterile genes in rice.

In order to explore the mechanism of acute toxicity for pyrene to cyanobacterial organisms, the responses of Synechocystis sp. PCC 6803 photosystem II (PS II) under pyrene stress were studied. The results showed there was no significant difference about the oxygen evolution under 0.125 mg/L pyrene stress when compared with control, but it was significantly lower than control at 0.625 mg/L pyrene. Polyphasic chlorophyll-a fluorescence transients in cells of Synechocystis sp. PCC 6803 exhibited a typical increase including O, J, I, and P phases. Fluorescence yield at phases J, I and P declined slightly at 0.125 and 0.625 mg/L pyrene, and significantly lower than control at 3.125 mg/L. According to the parameters deviated from JIP-test, no modification was induced by pyrene both at the donor side and at the acceptor side of PS II, and the reaction centre of PS II is the primary damaging target. Based on the expressing of four key genes (psbA, psbB, psbC and psbO) of PS II, only psbA showed significant difference at 3.125 mg/L pyrene when compared with control.
