Combined use of epigallocatechin-3-gallate (EGCG) and caffeine in low doses exhibits marked anti-obesity synergy through regulation of gut microbiota and bile acid metabolism

摘要:
Epigallocatechin-3-gallate (EGCG) and caffeine constitute the most effective ingredients of weight loss in tea. However, whether combination of EGCG and caffeine exhibits anti-obesity synergy remains unclear. Here, we showed low-doses of EGCG and caffeine used in combination led to synergistic anti-obesity effects equivalent to those of high-dose EGCG. Furthermore, combination treatment exhibited a synergistic effect on altering gut microbiota, including decreased Firmicutes level and increased Bifidobacterium level. Other notable effects of combination treatment included synergistic effects on: increasing fecal acetic acid, propionic acid, and total SCFAs; decreasing expression of GPR43; and increasing microbial bile salt hydrolase gene copies in the gut, facilitating generation of unconjugated BAs and enhancing fecal BA loss. Additionally, combination treatment demonstrated synergistic effects toward increasing the expression of hepatic TGR5 and decreasing the expression of intestinal FXR-FGF15, resulting in increased expression of hepatic CYP7A1. The genus Pyrus is characterized by an S-RNase-based gametophytic self-incompatibility (GSI) system, a mechanism that promotes outbreeding and prevents self-fertilization. While the S-genotype of the most widely known pear cultivars was already described, little is known on the S-allele variability within local accessions. The study was conducted on 86 accessions encompassing most of the local Sicilian varieties selected for their traits of agronomic interest and complemented with some accessions of related wild species (P. pyrifolia Nakai, P. amygdaliformis Vill.) and some national and international cultivars used as references. The employment of consensus and specific primers enabled the detection of 24 S-alleles combined in 48 S-genotypes. Results shed light on the distribution of the S-alleles among accessions, with wild species and international cultivars characterized by a high diversity and local accessions showing a more heterogeneous distribution of the S-alleles, likely reflecting a more complex history of hybridization. The S-allele distribution was largely in agreement with the genetic structure of the studied collection. In particular, the 'wild' genetic background was often characterized by the same S-alleles detected in P. pyrifolia and P. amygdaliformis. The analysis of the S-allele distribution provided novel insight into the contribution of the wild and international cultivars to the genetic background of the local Sicilian or national accessions. Big data has brought about opportunities for landscape architecture, changing the design thinking of layout optimization simulation, expanding the platform for public participation in layout optimization simulation design, reflecting social and humanistic care, and promoting the integration of discipline cooperation and data. At the same time, it also brings about challenges. The proposal of data theory, the acquisition and analysis of data, and the protection of privacy are all issues that we need to face and solve. First, build a layout optimization simulation program under the background of big data. Follow the procedures of conventional layout optimization simulation, add big data analysis technology to the preliminary analysis, layout optimization simulation design, and later evaluation management, introduce new data processing methods, and build a theoretical framework of landscape optimization simulation methods that integrate new and traditional data. Second, studying the evolution of the landscape image layout of the three-dimensional pastoral complex is different: the overall landscape image layout of the three-dimensional pastoral complex shows a trend of fragmentation and heterogeneity. Among them, the arable land is gradually fragmented, the wetland and water area are simplified in form, the woodland and grassland patches are gradually distributed, the fragmentation is reduced by 76.19%, and the connectivity index is gradually increased. From a spatial perspective, the edge area outside the Fourth Ring Road becomes the most obvious area of fragmentation tendency. Finally, by introducing big data into the landscape layout optimization simulation, a layout optimization simulation method based on big data is constructed and used in the layout optimization simulation of the landscape image layout of the three-dimensional pastoral complex to guide the participatory layout optimization simulation. To investigate the effects of l-Theanine (LTA) on intestinal mucosal immunity and the regulation of short-chain fatty acid (SCFA) metabolism under dietary fiber feeding, a 28-day feeding experiment was performed in Sprague-Dawley rats. The results show that LTA increased the proportion of Prevotella, Lachnospira, and Ruminococcus while increasing the total SCFA, acetic acid, propionic acid, and butyric acid contents in the feces. LTA also increased IgA, IgE, and IgG levels in the ileum, and increased villi height and crypt depth. Moreover, LTA upregulated the mRNA and protein expression of acetyl-CoA carboxylase 1, sterol element-binding protein 1c, fatty acid synthase, and 3-hydroxy-3-methylglutaryl coenzyme A reductase in the liver, while downregulating the expression of glucose-6-phosphatase and phosphoenolpyruvate carboxykinase 1 in the colon. Our study suggests that LTA can affect intestinal mucosal immunity by regulating SCFA metabolism under dietary fiber feeding. © 2020 The Royal Society of Chemistry.

摘要:
Dark tea is a unique fermented tea produced by solid-state fermentation of tea leaves (Camellia sinensis). It includes ripe Pu-erh tea, Fu brick tea, Liupao tea, and other teas. Microbial fermentation is considered to be the key factor controlling the quality of dark tea. It involves a series of reactions that modify the chemical constituents of tea leaves. These chemical conversions during microbial fermentation of dark tea are associated with a variety of functional core microorganisms. Further, Multi-omics approaches have been used to reveal the microbial impact on the conversion of the chemical components in dark tea. In the present review, we provide an overview of the most recent advances in the knowledge of the microbial bioconversion of the chemical components in dark tea, including the chemical composition of dark tea, microbial community composition and dynamics during the fermentation process, and the role of microorganisms in biotransformation of chemical constituents.

l-Theanine is an important component found in tea and has positive effects on nutrient absorption and transport. However, whether l-theanine can regulate glucose, lipid, and protein metabolism remains unknown. This study aims to investigate the effects of l-theanine on glucose, lipid, and protein metabolism in male Sprague-Dawley rats and characterize the underlying mechanisms. Compared to the control group, l-theanine increased the contents of hepatic and muscle glycogen, serum total protein (TP), and albumin (Alb), lowered the serum low-density lipoprotein cholesterol (LDL-C) level, decreased the activity of acetyl-CoA carboxylase (ACC), and enhanced carnitine palmitoyl transferase-1 (CPT-1) activity in the liver. Additionally, l-theanine upregulated the mRNA expression of phosphofructokinase (PFKL), CPT-1, insulin receptor (INSR), insulin receptor substrate (IRS), and liver kinase B1 (LKB1) and downregulated the mRNA expression of phosphoenolpyruvate carboxykinase 1 (PCK1), glucose-6-phosphatase catalytic subunit (G6PC), fatty acid synthase (FAS), and 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR). Moreover, l-theanine upregulated the expression of PFKL, glycogen synthase 2 (GYS2), ribosomal protein S6 (S6), INSR, IRS, and phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K) proteins; downregulated the expression of FAS, sterol regulatory element binding protein-1c (SREBP-1c), and HMGCR proteins; enhanced the phosphorylation of the mammal target of rapamycin (mTOR), ribosomal protein S6 kinase (p70S6K), protein kinase B (AKT), and AMP-activated protein kinase (AMPK); and decreased the phosphorylation of glycogen synthase kinase 3β (GSK-3β) and ACC1. Furthermore, 100 mg kg-1l-theanine was more effective at eliciting these effects than 200 and 400 mg kg-1l-theanine. In conclusion, l-theanine can regulate glucose, lipid, and protein metabolism via insulin and AMPK and their downstream signaling pathways. © The Royal Society of Chemistry 2020.

In this study, the crude exopolysaccharides (CEPSs) from fungus Aspergillus cristatus (MK346334, NCBI) isolated from Fuzhuan brick tea and its main purified fraction (EPSs-2) were investigated. Using the RAW264.7 cell model, EPSs-2 exhibited an excellent immunomodulatory effect in vitro. Then, the regulating effects of EPSs on immune function and gut microbiota were evaluated using a cyclophosphamide (Cy)-induced mice model. It was found that both CEPSs and EPSs-2 improved the body weight loss, immune organ indexes as well as the levels of TNF-α, IL-1β, IFN-γ and IgA, exhibiting potent immunoregulatory activity. Moreover, CEPSs and EPSs-2 not only attenuated the intestinal tissue damage, but also promoted the production of short-chain fatty acids and modulated the microbial composition by increasing the growth of Muribaculaceae, Prevotellaceae_UCG-001, Bacteroides, Parabacteroides and Tidjanibacter, while decreasing the relative abundances of Helicobacter, Bilophila, Mucispirillum, Lachnospiraceae, Ruminococcaceae and Clostridiales. These results indicated that the EPSs, especially EPSs-2, exhibited immunomodulatory activity associated with the modulation of gut microbiota to maintain gut homeostasis, which provided evidence for the development of novel potential prebiotics and immunomodulators. © The Royal Society of Chemistry.

Theasinensin A (TSA) and theasinensin B (TSB), dimers of tea catechins produced during the processing of oolong tea and black tea, had superior inhibitory effects on α-glucosidase. However, the potential inhibitory mechanisms on α-glucosidase are still unclear. In the present study, TSA and TSB were chemically synthesized and purified, and their inhibitory effects on α-glucosidase and potential mechanisms were investigated. The results showed that TSA and TSB could inhibit the activity of α-glucosidase in a reversible and noncompetitive manner with IC50 values of 6.342 and 24.464 μg mL-1, respectively, which were much lower than that of acarbose. The fluorescence and circular dichroism spectra revealed that TSA and TSB could alter the microenvironment and the secondary structure of α-glucosidase, thereby decreasing the α-glucosidase activity. Molecular docking results indicated that both TSA and TSB had a strong binding affinity to α-glucosidase by hydrophobic interactions and hydrogen bonds. Moreover, the stronger inhibition of TSA on α-glucosidase might be related to the closer binding site to the active site pocket of α-glucosidase. © The Royal Society of Chemistry.

Muscle-related disorders, such as sarcopenia and cachexia, caused by aging and chronic diseases can lead to the loss of muscle mass and strength to different degrees, severely affecting human health. Globally, tea is one of the three most popular beverages, and its major active ingredient catechins have been reported to delay muscular atrophy and enhance movement. However, currently, there is no systematic review to elaborate its roles and the associated mechanisms. This article reviews the (1) functions and mechanisms of catechins in the differentiation of myogenic stem cells, biogenesis of mitochondria, synthesis and degradation of proteins, regulation of glucose level, and metabolism of lipids in muscle cells; and (2) effect of catechins on the blood vessels, bones, and nerves that are closely related to the skeletal muscles. Catechins could prevent, mitigate, delay, and even treat muscle-related disorders caused by aging and diseases. © 2019, © 2019 Taylor & Francis Group, LLC.

The human purinergic P2Y1 receptor (P2Y1R) belongs to the family of G-protein coupled receptors (GPCRs). Two stable cell lines, hP2Y1R-TGFP/U2OS and hP2Y1R-β-AR2-TGFP/U2OS, were generated with steady expression of fluorescent tGFP tagged P2Y1R or tGFP tagged β-Arrestin2. The High Content Screening (HCS) technique was used to investigate P2Y1R mediated signal transduction processes based on the distribution of fluorescent receptors or the β-Arrestin2/receptor complex. All procedures including aggregation, endolysis and resensitization that were involved in the interaction between the P2Y1 receptor and its ligand can be visualized using the tGFP labeled receptor combined with HCS technology in this study. ADP activated P2Y1R to induce a distinguished pattern of fluorescent vesicle formation inside both cell types. These two cell lines produced different peak fluorescence signals when activated by ADP at 100 μM, and the time to form the maximal vesicles was detected as 15 and 20 min respectively. The EC50 values of the concentration-response curves of ADP induced formation of fluorescent vesicles were 35.7 nM and 71.8 nM respectively. The ADP effects could be blocked by a P2Y1R specific antagonist MRS2179. The P2Y1R are coupled to Gq protein to activate PLC first, PLC cleave PIP2 into IP3 and DAG. And then DAG and Ca2+ activate PKC pathway, and finally the phospholipase D initiates β-Arrestin2 mediated signal pathway. It took 30 and 45 min to complete P2Y1R re-sensitization in these two constructs respectively. Our results demonstrated that P2Y1R triggered downstream responses not only via heterotrimeric Gq proteins but also through β-Arrestin2. © The Royal Society of Chemistry.

摘要:
1,3-Dioleoyl-2-palmitoylglycerol (OPO), an important structured triacylglycerol, is generally supplemented into infant and toddler formulas as a nutrient fortifier. However, its role on fecal microbiota has not received attention in the last decades. In this study, anaerobic fermentation in vitro and high-throughput sequencing were conducted to investigate effects of standard OPO alone (OPO-A) and OPO-enriched plant-oil formula (OPO-P) on toddler fecal microbiota. The results showed OPO could be efficiently utilized by toddler fecal microbiota. OPO supplementation enhanced the production of short-chain fatty acids and lactic acid by microbiota. Furthermore, OPO promoted the growth of Bifidobacterium whereas inhibited the growth of Enterobacter at different fermentation time when compared with the control group. Besides, OPO supplementation enhanced bacterial diversity. Thus, this study indicated that OPO could modulate the fecal microbiota. The results provided useful information for further exploring the physiological role of OPO on colon, and provided new ideas for application of OPO. © 2019, © 2019 The Author(s). Twenty-seven representative Hunan fuzhuan brick teas were collected to develop a terminology lexicon and a quantitative descriptive analysis (QDA) method suitable for the sensory evaluation of Hunan fuzhuan brick tea infusion. Ten trained panelists developed a terminology lexicon comprised of eleven aroma and six taste attributes and evaluated the intensities of sensory attributes of each sample by conducting the QDA method. The QDA results showed that seventeen attributes listed in the final lexicon can be used to evaluate the quality of Hunan fuzhuan brick tea infusion properly, among which five aroma attributes, overall aroma, smoky, floral, fermented, and sweet (fruit), and one taste attribute, bitter, were the characteristic attributes to distinguish the differences in the sample qualities. Another panel made up of four professional cuppers evaluated samples by the cupping method to analyze the applicability and accuracy of the lexicon and the QDA method. The results showed that both the cupping method and QDA can be effectively used to evaluate Hunan fuzhuan brick tea quality, and their evaluation results showed high consistency and mutual complementation. This information will be beneficial for developing a sensory evaluation method and quality control for Hunan fuzhuan brick tea. © 2019

摘要:
Urban heat island (UHI) could have significant impacts on building energy consumption by increasing space cooling demand and decreasing space heating demand. However, the impacts of UHI on building energy consumption were understudied due to challenges associated with quantifying UHI-induced temperature change and evaluating building energy consumption. In this study, we reviewed existing literature for improving the understanding of UHI impacts on building energy consumption. It was found that UHI could result in a median increase of 19.0% in cooling energy consumption and a median decrease of 18.7% in heating energy consumption. The reported UHI impacts showed strong intercity variations with an increase of cooling energy consumption from 10% to 120% and a decrease of heating energy consumption from 3% to 45%. The UHI impacts also showed clear intra-city variations with stronger impacts in urban center than that in urban periphery. There were significant differences in the method and the data used to evaluate the UHI impacts in previous studies. Four future research focuses were recommended to better understand the UHI impacts on building energy consumption. © 2019 Elsevier Ltd

摘要:
High spatial resolution urban population dataset is increasingly required for sustainable urban planning and management. Dasymetric mapping is an effective approach to create such dataset. However, the created gridded total population datasets usually have limitation for urban analysis in developing countries as they usually underestimate urban population because of the strong urban-rural difference. In this study, we aimed to create a dataset of gridded urban population with 1 km resolution in China in year 2000 and 2010. We proposed an index of urban nighttime light (UNTL) by integrating radiance corrected DMSP nighttime light (RcNTL) and urban land, which is then used as weight to disaggregate county-level urban population. The validation using township population in Beijing as references shows reasonable accuracy with a mean relative error of 38% and a R-2 of 68%. Using only two widely available datasets (RcNTL and urban land), the proposed method is simple and computing efficient compared with methods using multiple geospatial data (e.g., land use and land cover, distance to city center, slope) and that combined with remote sensing imagery. As the used two auxiliary datasets are accessible globally, the method has great potential to produce similar urban population dataset for other developing countries where fine scale census population datasets are scarce. The produced urban population dataset is valuable for enriching our understanding of the urbanization process and designing sustainable urban planning and management strategies in China. An increasing amount of evidence suggests that the gut microbiota composition and structure contribute to the pathophysiology of metabolic syndrome (MS), which has been put forward as a new target in the treatment of diet-induced MS. In this work, we aimed to investigate effects of Fuzhuan brick tea polysaccharides (FBTPS) on MS and gut microbiota dysbiosis in high-fat diet (HFD) fed mice and to further investigate whether its attenuation of MS is related to the modulation of gut microbiota. The results showed that FBTPS intervention could significantly attenuate metabolic syndrome in HFD-induced mice. Based on results of sequencing, FBTPS treatment could increase the phylogenetic diversity of HFD-induced microbiota. FBTPS intervention could significantly restore the HFD-induced increases in relative abundances of Erysipelotrichaceae, Coriobacteriaceae, and Streptococcaceae. Spearman's correlation analysis showed that 44 key OTUs were negatively or positively associated with MS. Our results suggested that FBTPS could serve as a novel candidate for prevention of MS in association with the modulation of gut microbiota.

The long period of seed dormancy is one of the most important reasons leading to the low fertility of dove tree (Davidia involucrata Baill., Davidia hereinafter). During the fruit development process of Davidia, the endocarps will become particularly compact and hard, which was considered as a critical reason for the long seed dormancy of Davidia. However, the biological significance and regulatory mechanism of this process remain unclear. In this study, we identified a CCoAOMT gene, a key member of the lignin biosynthesis pathway, from Davidia endocarp. The gene named DiCCoAOMT1 has an ORF (open reading frame) of 744 bp and encodes a predicted protein of 239 amino acids. The expression of DiCCoAOMT1 is endocarp-specific and the expression levels increased along with the fruit development process. The encoding protein of DiCCoAOMT1 demonstrated a relatively high O-methyltransferase activity when using caffeic acid as a substrate in vitro. The tissue-specific CCoAOMT gene identified in this study might play a key role in specifically regulating the rapid lignification process in Davidia endocarp. © 2018, © 2018 The Author(s). The aim of present study was to examine whether the digestive system (saliva, simulated gastric and small intestinal conditions) could break down and large intestinal microbiota could utilize the polysaccharides from Fuzhuan brick tea (FBTPS). The results showed that there was no change in molecular weight, monosaccharide content and content of reducing sugars before and after saliva, simulated gastric and small intestinal digestion, indicating that FBTPS could pass through the digestive system without being broken down and reach the large intestine safely. The content of carbohydrate was significantly decreased by fermentation in vitro of gut microbiota, suggesting that FBTPS could be broken down and utilized by gut microbiota. FBTPS could significantly modulate the composition and abundance of gut microbiota. Furthermore, the contents of short-chain fatty acids were significantly increased. Therefore, FBTPS is expected to be a functional food to improve human health and prevent disease through promoting the gut health. © 2017 Elsevier Ltd

摘要:
为了探讨黑毛茶的陈化及成品茶的安全贮存特性,本研究通过静态称质量法测定了湖南黑毛茶在20、30、 40 ℃,水分活度为0.010 3~0.981 8条件下的吸湿特性,建立不同温度下黑毛茶的吸湿曲面响应模型及等温吸湿曲线;对等温吸湿曲线进行了非线性回归拟合,并在温度25 ℃和相对湿度85%、87%、90%的条件下进行模拟仓储。结果显示:湖南黑毛茶等温吸湿曲线为Ⅱ类曲线,不同温度(20、30、40 ℃)条件下,黑毛茶吸湿平衡时间随着温度的升高而缩短,分别为14、10、6 d,并随着温度的升高而呈现平衡含水率降低的规律;3个温度条件下黑毛茶的曲面响应模型呈现一定的规律特性,GAB及Osiwin模型较优地拟合了黑毛茶的等温吸湿曲线,特定条件下模拟仓储10 d,相对湿度85%的条件下未见生霉现象,相对湿度87%的条件下出现局部霉变,相对湿度90%的条件下霉变严重。

Dark tea has a significant effect on the prevention and treatment of age-related degenerative diseases. At present, further exploration of its functional mechanisms is delayed because of the complexity of post-fermentation microbial metabolites during the production phase. In this study, new isolated microbial metabolites extracted from dark tea were used to explore the neuroprotective effects, and they also helped allow further exploration of the mechanism of dark tea. Taking senescence-accelerated mouse prone 8 (SAMP8) mice as a biological model, we examined the protective effect on brain neurons of post-fermentation microbial metabolites which were extracted from dark tea. The 4-month-old mice were given treatments of the same concentration (10 mg kg−1 d−1) which were l-theanine, 3,3′-azanediylbis(4-hydroxybenzoic acid) (CDT-1) and one of the 8-C N-ethyl-2-pyrrolidinone substituted flavan-3-ols (CDT-2) by gavage for 14 weeks. Relative measurements such as RT-PCR, ELISA, western blotting, and section staining (HE, Nissl and myelin) were carried out. The results showed that l-theanine, CDT-1 and CDT-2 could inhibit the decrease in body weight, and down-regulate the formation of 4-HNE and ubiquitinated protein aggregates and the Aβ metabolic pathway. They could also increase endogenous antioxidant capacity, relieve cell hypoxia, and reduce the rate of neuronal apoptosis. This means that their protective activity regarding SAMP8 neurons was excellent and the activity of CDT-2 was the most significant. © The Royal Society of Chemistry.

Here we identified that BTE (black tea extract), within the studied concentration range, is more effective than GTE (green tea extract) in protecting C. elegans against hypertonic stress, by enhancing survival after exposure to various salts, and alleviating suffered motility loss and body shrinkage. The mechanism of such protection may be due to the ability of black tea to induce the conserved WNK/GCK signaling pathway and down-regulation of the expression levels of nlp-29. Intriguingly, black tea does not relieve hypertonicity-induced protein damage. The findings implicate the potential health benefits of black tea consumed worldwide. © The Royal Society of Chemistry.
