摘要:
肥胖症是由多种因素共同作用引起的营养性代谢障碍性疾病,越来越多的研究表明茶叶具有一定的减肥作用。茶叶减肥作用的活性成分主要为儿茶素、咖啡因,这些成分可能通过增加机体能量消耗、改变机体脂类代谢、调节食欲等机理而起作用。本文综述了近年来国内外关于茶叶减肥作用的研究,并简要阐述了其减肥作用可能的成分及机理。

建立了配体交换色谱手性流动相法拆分茶氨酸对映体的方法.采用Polaris C_(18)柱;流动相为L-脯氨酸:Cu~(2+)(摩尔比)为2:1,Cu~(2+)浓度为0.5 mmol/L,pH6.8,甲醇的加入体积比为2%;波长254 nm;流速0.9 ml/min;柱温30℃.L-茶氨酸的进样量在0.09542 μg~4.241 μg范围内峰面积与进样量之间线性关系良好,回收率在97.45%~100.4%之间;D-茶氨酸的进样量在0.08486 μg~4.243 μg范围峰面积与进样量之间线性关系良好,回收率在97.07%~100.1%之间.该方法通用,准确.

摘要:
Analysis of the membrane proteins, particularly the integral membrane proteins, is limited by the inherent membrane hydrophobicity. Sodium dodecyl sulfate (SDS) is one of the most efficient reagents used for the extraction of membrane proteins, but its presence in samples interferes with LC-MS-based proteomic analyses because it affects RP-LC separations and electrospray ionization. In this paper, we present an improved sample preparation strategy based on SOS-assisted digestion and peptide-level SDS-removal using an optimized potassium dodecyl sulfate (KDS) precipitation method (SSDP method) for shotgun analysis of the membrane proteome. This method utilizes a high concentration of SDS (1.0%) to lyse the membranes and to solubilize the hydrophobic membrane proteins, resulting in a more complete protein digestion in the diluted SDS buffer (0.1% SOS), and a high efficiency of SOS removal and peptide recovery by the optimized KDS precipitation for protein identification. The SSDP method provides evidence that proteins can be efficiently digested, and the SDS can be decreased to <0.01% allowing >95% peptide recovery. Compared to other sample preparation methods commonly used in shotgun membrane proteomics, the newly developed method not only increased the identified number of the total proteins, membrane proteins and integral membrane proteins by an average of 33.1%, 37.2% and 40.5%, respectively, but also leading to the identification of highest number of matching peptides. All the results showed that the method yielded better recovery and reliability in the identification of the proteins especially the highly hydrophobic integral membrane proteins, and thus providing a promising tool for the shotgun analysis of membrane proteome. 摘要:
对不同陈化时间六堡茶茶叶粉末和提取物进行傅里叶红外光谱分析,考察陈化时间与红外光谱特征关系。结果表明:六堡茶陈化时间不同,其红外光谱不同;茶叶粉末与提取物的红外光谱差异较大;除了共同红外光谱特征,还具有因陈化时间变化引起成分差异的特征红外光谱;同一厂家不同陈化时间的六堡茶样品根据红外光谱的峰数、波数、峰形、相对强度及二阶导数谱的差异考察存在差异。利用傅里叶红外光谱法分析六堡茶,该方法快速、简便、直观,茶叶无需特殊处理,对同一厂家不同陈化时间六堡茶的分析鉴别有一定作用。

摘要:
The water extraction and composition of pu-erh tea, as well as the hypoglycemic effect of the water extract of pu-erh tea (WEPT) in vivo and in vitro, are reported to investigate its hypoglycemic effect on diabetes. High-performance liquid chromatography and colorimetric methods are used to analyze the tea catechins, caffeine, polyphenols, amino acids, and polysaccharides of the WEPT. The effect of the WEPT on glucose uptake by cultured HepG2 cells and the inhibition effect of rat intestinal sucrase, maltase, and porcine pancreatic amylase are determined in vitro. Then, the blood glucose and insulin levels of intragastrically administered WEPT on fasting and oral glucose tolerance test (OGTT) using type 2 diabetic db/db (BKS.Cg-m +/+ Lepr<sup>db</sup>/J) mice are determined in vivo. The results showed that the WEPT dose-dependently and significantly increased glucose uptake by HepG2 cells and inhibited rat intestinal sucrase, maltase, and porcine pancreatic amylase activity. The WEPT intragastrically given for 4 weeks suppressed the increase in blood insulin and glucose levels of db/db mice fasted overnight. In OGTT, the WEPT improved impaired glucose tolerance and ameliorated retarded insulin response at 60 and 120 min in db/db mice. These results suggest that the WEPT has beneficial effects on glucose homeostasis in type 2 diabetes and in amendment of insulin resistance. ©2012 American Chemical Society.

在对中国古典美学重要理论和方法进行梳理的基础上,结合历代茶人在茶审美中的经典茶诗词,归纳提炼出“物我观照”、“净静虚明”、“妙悟自然”三种茶审美方法。“物我观照”源于自然的某些物征可以象征人高尚的道德品质,在经常的茶事实践中,茶人从茶的清香和内蕴的品质中观照出“茶佳人”、“茶君子”的形象。“净静虚明”发端于道家老子“涤除玄鉴”及“致虚极,守静笃”的悟道理念,在空灵虚静的心境中,即使是最简单不过的茶事活动仍洋溢着诗性之美。“妙悟自然”取材于禅宗、老庄及后来文艺美学理论,指茶人在品茶过程中通过长期的亲身体验和凝神遐想,终致随心所欲而不逾矩的人生境界。

茯茶中“金花”孢子粉提取物对体外诱导的非酒精性脂肪肝细胞内甘油三酯代谢的影响

摘要:
建立非酒精性脂肪肝细胞模型,研究茯茶中"金花"孢子粉提取物对非酒精性脂肪肝细胞甘油三酯代谢的影响,筛选出具有降脂功效的活性组分,为茯茶的深度开发及"金花"孢子粉降脂成分的分离纯化提供一定的指导。以正常人肝细胞LO2为实验对象,采用脂肪乳剂加入细胞培养液培养肝细胞,建立非酒精性脂肪肝细胞模型;以MTT法筛选出提取物最佳作用浓度;通过检测细胞内甘油三酯来评价药物的降脂效果。结果表明,用含1.0 mL/L 20%脂肪乳剂的培养液培养LO2肝细胞48 h,肝细胞内甘油三酯与对照组比较呈现极显著升高(P<0.01)且肝细胞ALT、AST泄漏量比对照组极显著加大(P<0.01),因此本方法能建立非酒精性脂肪肝细胞模型;"金花"孢子粉提取物作用脂肪肝细胞最佳质量浓度分别为150μg/mL和5μg/mL,当醇提物和萃取的水层部分分别作用脂肪肝细胞24、48 h时都能使细胞内甘油三酯含量降低且与阳性药物辛伐他汀和血脂康效果相当,因此本实验筛选出"金花"孢子粉醇提物和萃取的水层具有较好的降脂效果。

摘要:
采用高速逆流色谱法分离纯化紫甘薯花色苷。以正丁醇-乙酸乙酯-0.5%乙酸(3∶1∶4,V/V)为溶剂体系,上相为固定相,下相为流动相,流速2 mL/min,进样量300 mg,分离得到两种花色苷的混合物;混合物再以0.2%三氟乙酸-正丁醇-甲基叔丁基醚-乙腈(6∶5∶2∶1,V/V)为溶剂体系,上相为固定相,下相为流动相,流速1.5 mL/min,进样量100 mg,分离得到纯度分别为98.5%和96.7%的两个花色苷单体组分。通过紫外-可见光谱、质谱、核磁等技术进行结构鉴定,确定组分1为3-O-{6-O-(E)-咖啡酰-2-O-[6-O-(E)-对羟基苯甲酰-β-D-吡喃葡糖基]-β-D-吡喃葡糖苷}-[5-O-(β-D-吡喃葡糖苷)]芍药素,组分2为3-O-{6-O-(E)-咖啡酰-2-O-[6-O-(E)-阿魏酰-β-D-吡喃葡糖基]-β-D-吡喃葡糖苷}-[5-O-(β-D-吡喃葡糖苷)]芍药素。实验从450 g紫甘薯中获得63 mg组分1和48 mg组分2,为紫甘薯花色苷分离提供了高效、稳定、可靠的方法。

摘要:
建立了同时测定咖啡豆中6种酚酸类化合物(咖啡酸、3-咖啡酰奎尼酸、4-咖啡酰奎尼酸、5-咖啡酰奎尼酸、 3,5-二咖啡酰奎尼酸、4,5-二咖啡酰奎尼酸)的反相高效液相色谱测定方法。采用Kro masil C_(18)柱(200 mm×4.6 mm,5μm),以乙腈和0.1%甲酸水溶液为流动相进行梯度洗脱,二极管阵列检测器检测,45 min内可对6种目标物进行同时检测,且各化合物都能达到基线分离。经测定,样品中6种酚酸类化合物的加标回收率为90.76%~ 104.73%,相对标准偏差为0.7%~3.9%。该法简便、快速、灵敏度高,适用于咖啡豆中6种酚酸类化合物的同时分析以及咖啡豆原料与制品的质量控制和综合评价。

摘要:
Emerging evidence supports health-promoting properties of post-fermented Chinese Brick Tea. Fu Zhuan Tea, fermented with the fungus, Erotium cristatum, contains a unique phytochemical profile attributed to its unique method of processing. Fu Zhuan Tea has been shown to activate pancreatic enzymes and regulate blood lipids in laboratory models. Regulation of blood lipid levels by Fu Zhuan Tea consumption was examined in an observational pilot study of volunteers with elevated LDL cholesterol that were not taking any prescription lipid lowering medications. Significant changes in blood lipids were detected after 120. days of daily consumption. Fu Zhuan Tea fractionation led to the investigation of six compounds for regulation of transcription factors involved in lipid metabolism, including Farnesoid X receptor (FXR), Liver X-activated Receptor (LXR) and two isoforms of the Peroxisome Proliferator-Activated Receptors (PPARγ and PPARδ). Reporter gene assays with liver cells revealed dose dependent differences in regulation of transcription factor activation. These data reveal potential therapeutic efficacy and candidate molecular targets for Fu Zhuan Tea, and provide rationale for chemical characterization of bioactive fractions and investigation of therapeutic efficacy in cardiovascular disease and type 2 diabetes. © 2011 Elsevier Ltd.

为了使玉竹种茎安全贮藏过冬并在次春快速整齐萌发,以湖南玉竹主栽品种猪屎尾参为试材,研究了河沙层积、即收即播与3个不同催芽温度处理对玉竹种茎萌发特性的影响.结果表明,层积处理能显著提早玉竹种茎萌发,使出苗整齐.利用层积处理,结合人工加温催芽,既能保证玉竹种茎安全过冬,又能显著促进玉竹种茎萌发出苗,且出苗整齐.适宜玉竹种茎催芽的温度为9~12℃.

The growth inhibition and cytotoxic effects of theaflavins mixture and pure theaflavins [theaflavin, theaflavins-3-gallate, theaflavins-3′-gallate and theaflavin-3, 3′-digallate] from black tea on human liver cancer HepG2 cells, gastric cancer SGC-7901 cells, lung cancer A549 cells and acute promyelocytic leukemia K562 cells were investigated. The four major theaflavins were obtained from enzymatically oxidized catechins mixture using preparative high-performance liquid chromatography (HPLC). The purity and the structures of the isolated compounds were confirmed by <sup>1</sup>H nuclear magnetic resonance, Liquid chromatography-mass spectrometry (LC/MS), Fourier-transform infrared, HPLC with diode array detection and thin-layer chromatography analyses. Theaflavin monomers and mixture exhibited strong inhibitory effects on SGC-7901 and A549 cells. The obtained TF2A exhibited high inhibition activity against the growth of all four cancer cells examined. It is concluded that individual theaflavins have differential inhibitory activity in cancer cell lines and that TF2A have promising activity to be considered in anticancer treatment research as a drug or a template for drug synthesis. PRACTICAL APPLICATIONS: Theaflavins have received much attention because of their potential benefits for human health. In this study, we used a modified method for the preparation and isolation of theaflavins and examined the cytotoxic activity of the individual theaflavins against human liver cancer (HepG2) cells, gastric cancer (SGC-7901) cells, lung cancer (A549) cells and acute promyelocytic leukemia (K562) cells. Our results demonstrates that individual theaflavins have differential inhibitory activity in cancer cell lines and has the potential to be used individually for functional food applications and for therapy targeting specific cancer treatments. The most active theaflavin (TF2A) is a promising compound that can be used as a drug or a template for drug synthesis for anticancer research. ©2011 Wiley Periodicals, Inc.

摘要:
建立高效液相色谱同时测定茶叶中8种儿茶素(EGC、DL-C、EC、EGCG、GCG、EGCG4″Me、EGCG3″Me、ECG)、3种生物碱(茶碱、可可碱、咖啡碱)和没食子酸的方法。色谱条件为Welchrom C18(250mm×4.6mm,5μm)色谱柱、磷酸缓冲液-乙腈溶液为流动相、梯度洗脱、波长278nm、流速1.0mL/min、柱温30℃。采用此方法,茶叶中各种成分都能达到基线分离的效果,线性关系良好,具有良好的稳定性和准确度。

White leaf No.1 is a typical albino tea cultivar grown in China and it has received increased attention in recent years due to the fact that white leaves containing a high level of amino acids, which are very important components affecting the quality of tea drink. According to the color of its leaves, the development of this tea cultivar is divided into three stages: the pre-albinistic stage, the albinistic stage and the regreening stage. To understand the intricate mechanism of periodic albinism, a comparative proteomic approach based on two-dimensional electrophoresis (2-DE) and mass spectrometry was adopted first time to identify proteins that changed in abundance during the three developmental periods. The 2-DE results showed that the expression level of 61 protein spots varied markedly during the three developmental stages. To analyze the functions of the significantly differentially expressed protein spots, 30 spots were excised from gels and analyzed by matrix-assisted laser desorption ionization-time of flight-tandem mass spectrometry. Of these, 26 spots were successfully identified. All identified protein spots were involved in metabolism of carbon, nitrogen and sulfur, photosynthesis, protein processing, stress defense and RNA processing, indicating these physiological processes may play crucial roles in the periodic albinism. Quantitative real-time RT-PCR analysis was used to assess the transcriptional level of differentially expressed proteins. In addition, the ultrastructural studies revealed that the etioplast-chloroplast transition in the leaf cell of White leaf No. 1 was inhibited and the grana in the chloroplast was destroyed at the albinistic stage. In this work, the proteomic analysis revealed that some proteins may have important roles in the molecular events involved in periodic albinism of White leaf No. 1 and identificated many attractive candidates for further investigation. In addition, the ultrastructural studies revealed that the change in leaf color of White leaf No. 1 might be a consequence of suppression of the etioplast-chloroplast transition and damage to grana in the chloroplast induced by temperature. These results provide much useful information to improve our understanding of the mechanism of albinism in the albino tea cultivar.
