摘要:
Background: To investigated the effect of earthworm extract (EE) on deep second-degree burn wound healing process. Methods: A burn wound model was created on the mice’s skin and was subject to different treatments: the control group received no treatment; the Jingwanhong (JWH: a well-established, widely used external ointment for treating burn wounds) group was treated with 0.1 g of JWH cream and spray it on the wound surface; the EE group was treated with 0.1 mL of EE solution. All the mice were sacrificed at 3, 7, 11, and 15 days after injury (n=6/group/time point). Macroscopic observation, wound healing rate (WHR), wound healing time (WHT), water content (WC), hydroxyproline (Hyp) content, histological, and hematological analyses were performed at the burn wound sites. Results: Better, faster burn wound healing in the JWH and EE groups than the control group at 15 days after injury were detected at the wound sites. Compared to the control group, the EE group had higher WHR, shorter WHT, lower WC, higher Hyp content, more fibroblasts, fibrocytes, and capillary endothelial cells; in addition, they showed greater capillary endothelial cell grouping at the wound sites during the healing process. This group also showed more platelets, white blood cells (WBCs), and neutrophilic granulocytes in serum at the early stages after burn injury. Conclusions: EE could effectively promote skin wound healing by decreasing edema, suppressing fibrosis, activating angiogenesis and epithelial regeneration, inhibiting scar formation, and reducing the risk of infection. Thus, it could be made into a promising healing agent for burn wound. © Annals of Palliative Medicine. All rights reserved.

摘要:
Chronic nonhealing wounds pose a significant challenge to healthcare system because of its tremendous utilization of resources and time to heal. It has a well-deserved reputation for reducing the quality of life for those affected and represent a substantial economic burden to the healthcare system overall. Earthworms are used as a traditional Chinese medicine, and have been applied pharmacologically and clinically since a long time in China. However, there is paucity in data regarding its wound healing effects. Therefore, we investigated the effect of earthworm extract (EE) on skin wound healing process. The obtained data showed that EE has healing effects on local wound of mice. It decreased the wound healing time and reduced the ill-effects of inflammation as determined by macroscopic, histopatho-logic, hematologic, and immunohistochemistry parameters. The potential mechanism could be accelerated hydroxyproline and transforming growth factor-β secretion—thus increasing the synthesis of collagen, promoting blood capillary, and fibroblast proliferation. It could accelerate the removal of necrotic tissue and foreign bodies by speeding up the generation of interleukin-6, white blood cells, and platelets. It thus enhances immunity, reduces the risk of infection, and promotes wound healing. All in all, the obtained data demonstrated that EE improves quality of healing and could be used as a propitious wound healing agent. © 2018 The Author(s).

MicroRNA-32 (miR-32) is associated with tumor progression and poor prognosis in certain malignant tumors. However, the function and clinical relevance of miR-32 in human hepatocellular carcinoma (HCC) has not yet been elucidated. The present study aimed to investigate the expression and prognostic value of miR-32 from liver samples in patients with HCC. The expression of miR-32 was analyzed in HCC and healthy tissues using Gene Expression Omnibus datasets. Reverse transcription-quantitative polymerase chain reaction was used to analyze the levels of miR-32 mRNA in 154 HCC liver samples, 33 of which were paired with adjacent non-tumor tissues. The overall survival (OS) rate in patients with HCC was evaluated using Kaplan-Meier survival analysis, and the factors that may affect the prognosis and survival of patients with HCC were analyzed using univariate (log-rank test) and multivariate Cox proportional hazard models. The present results demonstrated that miR-32 expression levels were significantly upregulated in HCC liver biopsies compared with normal tissues (P<0.05). miR-32 expression was significantly associated with the number of foci and tumor diameter (P<0.05). In addition, Kaplan-Meier analysis revealed that patients with low miR-32 expression had longer OS and disease-free survival compared with those with high miR-32 expression (P<0.01). Altogether, to the best our knowledge, the present study is the first study to indicate the association between increased miR-32 expression with HCC progression and poor prognosis in patients. This suggests that miR-32 may have potential prognostic value and may be used as a tumor biomarker for the diagnosis of patients with HCC. © 2018, Spandidos Publications. All rights reserved.

Previous studies have suggested that dysregulation of microRNA (miR) -124a is associated with various types of human cancer. However, there are few studies reporting the level of miR-124a expression in non-small cell lung cancer (NSCLC). The present study investigated the association between miR-124a and NSCLC by analyzing the differential expression of miR-124a in NSCLC using the GEO database, as well as subsequently performing reverse transcription-quantitative polymerase chain reaction analysis on 160 NSCLC biopsies, 32 of which were paired with adjacent normal tissues. The results indicated that mir-124a expression levels were decreased in NSCLC tumor biopsies compared with adjacent normal tissues. The overall survival (OS) in patients with a high expression of miR-124a was prolonged relative to patients with low expression of miR-124a. The expression levels of miR-124a were associated with clinical characteristics, including lymph-node metastasis, tumor differentiation, tumor node metastasis (TNM) stage and diameter. Frequently, lymph-node metastasis, TNM stage, diameter and lack of chemotherapy have been associated with a worse prognosis in patients. In addition, the present study identified that high expression of miR-124awith chemotherapy may increase OS. In conclusion, the current study demonstrated that miR-124a was downregulated in NSCLC, and miR-124a was a potential prognostic tumor biomarker response to chemotherapy.

摘要:
目的:探讨建立人肝细胞系LO2单纯肝脂肪变性细胞模型的方法。方法:试验分为正常对照组与试验组,用含10%胎牛血清的1640培养基培养LO2细胞,当细胞处于对数生长期时,正常对照组更换新鲜的1640培养基,试验组用含不同浓度游离脂肪酸(油酸∶棕榈酸为2∶1)的1640培养基培养,24 h后油红O染色,观察细胞内脂滴大小,并检测细胞中TG、TC含量的变化及细胞增殖情况。结果:试验组中LO2细胞内有大量的脂滴生成,且甘油三酯水平显著升高,其中,以1.2 mM FFA组脂质沉积最为明显。同时,细胞抑制率小于4%,胆固醇水平无明显升高,均符合单纯脂肪变性模型的特征。结论:采用游离脂肪酸(油酸∶棕榈酸为2∶1)可以成功诱导LO2细胞单纯脂肪变性,其中游离脂肪酸浓度以1.2 m M最为理想。

摘要:
Longlin pig is a native breed of Fujian province in China. It is the first time that the complete mitochondrial genome sequence of Longlin pig is reported in this work, which is determined through the PCR-based method. The total length of the mitognome is 16,699 bp, which contains 1 control region (D-loop region), 2 ribosomal RNA genes, 13 PCGs and 22 tRNA genes. The total base composition of Longlin pig mitochondrial genome is 34.67% for A, 26.21% for C, 25.80% for T and 13.33% for G, in the order A>C>T>G. The complete mitochondrial genome of Longlin pig provides an important data in genetic mechanism and the evolution genomes.

壳寡糖对脂多糖诱导猪空肠上皮细胞氧化损伤的作用

本试验旨在通过脂多糖(LPS)诱导猪空肠上皮细胞(IPEC-J2)来建立氧化应激模型,探讨壳寡糖(COS)的抗氧化作用效果。采用噻唑蓝(MTT)法检测LPS和COS对IPEC-J2作用12、24、48 h后细胞增殖活性的变化;选择适宜浓度LPS(1.0 μg /mL)和COS(200 μg /mL)作用于IPEC-J2,分为对照组、LPS组、COS组、LPS+COS组。采用Western Blot法检测核因子E2相关因子2(Nrf2) 、血红素氧合酶-1(HO-1)蛋白的表达;试剂盒检测超氧化物歧化酶(SOD) 、过氧化氢酶(CAT)活性及丙二醛(MDA)的含量。结果表明: 1.0 μg /mL的LPS对IPEC-J2作用24 h,细胞增殖的抑制率为41.6%,为造模的最适浓度和作用时间; COS在一定浓度范围内对IPEC-J2有增殖作用,其浓度为200 μg /mL时效果最佳,作用时间为24 h时,细胞增殖率达到126.3%。LPS+COS组较对照组的SOD、CAT活性和MDA含量差异不显著(P>0.05), Nrf2和HO-1蛋白相对表达量显著升高(P<0.05); LPS +COS组较LPS组的Nrf2蛋白相对表达量显著升高(P < 0.05), HO-1有升高趋势,但差异不显著(P > 0.05), SOD、CAT活性显著升高(P < 0.05), MDA含量显著降低(P<0.05) 。由此可见,LPS诱导IPEC-J2氧化应激,而COS可进一步促进Nrf2和HO-1蛋白的高表达,并提高抗氧化酶的活性,降低氧化产物含量,从而增强细胞对氧化应激的抵抗力,起到保护作用。

目的:探讨不同浓度蚓激酶(EFE)在体外对LO2细胞株(人正常肝细胞)及软脂酸(PA)诱导的LO2细胞增殖的影响及EFE的最佳作用浓度。方法:用不同浓度(100 u/m L、200 u/m L、400 u/m L、600 u/m L、800 u/m L、1 000 u/m L)EFE作用于LO2细胞,分别在作用6 h、12 h、24 h,通过MTT法检测各组细胞的OD值;用20μg/m L的PA作用LO2细胞24 h,然后用不同浓度(100 u/m L、200 u/m L、400 u/m L、600 u/m L、800 u/m L、1 000 u/m L)的EFE进行干预,并于6 h、12 h、24 h检测细胞增殖情况。结果:200 u/m L、400 u/m L的EFE可促进LO2细胞的增殖,但无统计学意义,高浓度(800 u/m L、1 000 u/m L)的EFE能明显抑制LO2细胞的增殖(P<0.05),且其抑制作用随着时间的延长而增强;低浓度(100 u/m L)的EFE可促进经PA诱导的LO2细胞的增殖,其作用效果显著(P<0.05);200 u/m L、400U/m L的EFE对经PA诱导的LO2细胞的促进作用极显著(P<0.01)。结论:EFE能缓解PA诱导的细胞损伤,对PA诱导的LO2细胞具有保护作用,且其最佳作用浓度为400 u/m L。

为研究高产新丹系大约克后备母猪背膘厚度与产仔数、弱仔率、初生重的关系,试验随机抽取174头后备母猪采用背膘测定仪活体测定背膘厚度,结合第一胎次产仔数、弱仔率、初生重等数据,分析不同膘情厚度下的母猪产仔性能.结果表明:背膘厚度在13~ 16 mm时产仔数较好,在14~ 15 mm时产仔数最多并与11 mm以下组相比较存在显著差异(P<0.05).在弱仔率方面,背膘厚度在13~ 17 mm时弱仔率较低,和各组别相比较具有显著差异(P<0.05).在窝初生重方面,背膘厚度在14~ 16 mm时窝初生重最大,但与各组比较差异不显著(P>0.05).综合以上数据得出,新丹系大约克后备母猪背膘厚度为14~15 mm时,头胎的产仔性能最佳.

Keap1-Nrf2-ARE动物氧化应激反应信号通路与天然抗氧化产品开发

摘要:
氧化应激与动物机体多种疾病相关联,在动物生产过程中造成了巨大的经济损失。Kelch样环氧氯丙烷相关蛋白-1(Keap1)-核转录因子E2相关因子2(Nrf2)-抗氧化反应序列元件(ARE)信号通路是机体抗氧化反应中的重要调节途径,信号通路的活化受到一系列亲电试剂、蛋白激酶等因子的调控,而活化的通路可以通过调节下游相关抗氧化酶类和抗氧化物的基因表达来稳定机体的氧化还原水平。在研究机体氧化应激和抗氧化防御系统的同时,发现了一系列具有抗氧化活性的天然物质,这对于安全有效的畜牧生产提供了广阔的前景。本文对Keap1-Nrf2-ARE信号通路在抗氧化应激反应中的作用机理、天然抗氧化产品的开发和在动物生产中的应用研究进行了综述。

摘要:
某黑色雪纳瑞犬出现急性发病,突发全身强直收缩,间隔1h后,又出现同样急症,发病后,立即送诊.通过了解患犬基本情况、分析临床症状及对患犬进行临床检查,结果发现犬只严重酸血症,随即做出急诊处置;然后通过连日临床治疗,最终宠物犬痊愈出院.

摘要:
目前发现的与肝脏脂质代谢相关的转录因子主要包括过氧化物酶体增殖物激活受体(PPARs)、肝X受体(LXRs)和固醇调节元件结合蛋白(SREBPs)。这些转录因子通过对脂类代谢基因的调节,从而在维持机体的脂代谢平衡中起着重要作用。随着对转录因子的深入研究,以其为作用靶点的药物相继被开发出来,广泛应用于肥胖、高血脂、糖尿病等脂代谢障碍性疾病的防治,并表现出良好的临床效果和应用前景。论文通过综述肝脏脂质代谢关键转录因子的调控靶基因、调控机制和临床应用,旨在促进肝脏脂质代谢关键转录因子的进一步研究,以及临床新药的开发与应用。

文章就接诊的一例犬因高空坠落而发生骨折的病例,经数码X线摄影检查(CR)确诊为肱骨远端干骺端骨折,然后制定治疗方案:经呼吸麻醉,选择肱骨外侧髁手术通路,采用螺钉加克氏针的内固定方式,将骨折部位进行解剖学复位,经过后期护理,犬恢复良好.

高通量测序技术因其能够一次同时对几十万到几百万条核酸分子进行序列测定,是对传统Sanger测序革命性的改变,因此称为下一代或二代测序技术。随着第二代测序技术的迅猛发展,越来越多的学者也开始应用高通量测序技术来解决临床兽医学中的各类问题。文章从高通量测序技术发展现状简介、动物遗传育种与优良品种改进、基因筛选与基因调节机制、动物疾病预防与诊断、兽药筛选等方面对高通量测序技术在临床兽医学中的应用进行综述。

摘要:
蚯蚓作为一种中药材,在我国应用已经有几千年历史,据《本草纲目》记载:蚯蚓能治疗瘰疬溃烂、口舌糜疮,临床上常用来治疗骨折、创伤、下肢溃烂等疾病[1].蚯蚓中含高度不饱和脂肪酸、核苷酸、酶类、蛋白质、蚯蚓解热碱等多种物质,具有清热定惊、止咳平喘、活血通络、降压、抗溃疡、抗肝纤维化、利尿等功能.此外,现代分离技术发现,蚯蚓中还含有纤溶酶、抗菌肽、多种抗氧化酶等活性成分,因此具有止血镇痛、促进创伤愈合、抗氧化、抗血栓、调节免疫、抗心律失常、抗癌等作用[2].近几年蚯蚓药用价值越来越受到重视,临床研究和应用也日益广泛.国内外对蚯蚓创伤愈合作用的研究成为蚯蚓有效成分分离提纯的新热门.
