Koumine promotes ROS production to suppress hepatocellular carcinoma cell proliferation via NF-κB and ERK/p38 MAPK signaling

摘要:
In the past decades, hepatocellular carcinoma (HCC) has been receiving increased attention due to rising morbidity and mortality in both developing and developed countries. Koumine, one of the significant alkaloidal constituents of Gelsemium elegans Benth., has been regarded as a promising anti-inflammation, anxiolytic, and analgesic agent, as well as an anti-tumor agent. In the present study, we attempted to provide a novel mechanism by which koumine suppresses HCC cell proliferation. We demonstrated that koumine might suppress the proliferation of HCC cells and promote apoptosis in HCC cells dose-dependently. Under koumine treatment, the mitochondria membrane potential was significantly decreased while reactive oxygen species (ROS) production was increased in HCC cells; in the meantime, the phosphorylation of ERK, p38, p65, and IκBα could all be inhibited by koumine treatment dose-dependently. More importantly, the effects of koumine upon mitochondria membrane potential, ROS production, and the phosphorylation of ERK, p38, p65, and IκBα could be significantly reversed by ROS inhibitor, indicating that koumine affects HCC cell fate and ERK/p38 MAPK and NF-κB signaling activity through producing excess ROS. In conclusion, koumine could inhibit the proliferation of HCC cells and promote apoptosis in HCC cells; NF-κB and ERK/p38 MAPK pathways could contribute to koumine functions in a ROS-dependent manner. © 2019 by the authors. 摘要:
以雄性昆明小鼠为研究对象,以不同剂量的T-2毒素对小鼠进行腹腔注射,24 h后检测小鼠血清生化指标,判断T-2毒素急性中毒模型是否建立成功。结果显示,随着T-2毒素剂量的增加,小鼠血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性和直接胆红素(DBLI)以及总胆红素(TBLI)水平呈增加趋势,但总蛋白(TP)和白蛋白(ALB)含量以及总胆固醇(T-CHO)和甘油三酯(TG)含量呈降低趋势,与对照组相比均差异显著或极显著(P〈0.05或P〈0.01)。表明T-2毒素对小鼠机体造成了损伤,T-2毒素急性中毒模型建立成功。

摘要:
研究板蓝根多糖(Radix Isatidis polysaccharide,RIPS)对醋酸铅诱导雄性小鼠生长发育的影响.将3周龄60只健康雄性昆明系小鼠随机分为6个组:即对照组,RIPS 100 mg/kg组,铅模型组,RIPS低、中、高剂量(50 mg/kg、100 mg/kg、 200 mg/kg)治疗组,连续7 d腹腔注射醋酸铅20 mg/kg制备小鼠铅中毒模型,造模后对板蓝根多糖治疗组分别饮水给予不同浓度的RIPS,1次/d,连续给药28 d.观测每组小鼠日均采食量、体质量增长、脏器系数.结果显示,模型组日均采食量少于对照组和RIPS组,RIPS治疗组日均采食量随RIPS剂量的增加而明显增加;染毒各组小鼠体质量均显著降低(P<0.01或P<0.05),100 mg/kg RIPS治疗组小鼠自给药第3周开始,体质量显著增加(P<0.05);模型组小鼠除肾脏指数显著降低于对照组(P<0.05),肝脏器指数虽有所增加,但无显著差异(P>0.05),50 mg/kg RIPS治疗组睾丸指数与100 mg/kg RIPS治疗组肝脏指数均显著低于模型组(P<0.05),100 mg/kg RIPS治疗组的肾脏指数则显著增加(P<0.05).提示板蓝根多糖对醋酸铅诱发的小鼠生长发育迟缓具有一定的保护作用,其作用机制有待后续研究.

摘要:
为了研究桦木酸(BA)对环磷酰胺(Cy)诱导小鼠免疫器官氧化损伤的影响。将50只健康雄性昆明小鼠随机分为5组,即对照组、Cy组及0.05、0.50、5.00 mg / kg BW BA组。对照组和Cy组灌服1%的可溶性淀粉溶液,其余各组按不同剂量的BA灌服,连续14 d后,除对照组注射生理盐水外,其余4组均腹腔注射Cy(50 mg / kg BW)诱导氧化应激模型。收集血清、脾脏和胸腺,检测血清中谷丙转氨酶(ALT) 、谷草转氨酶(AST)活性及白蛋白(ALB) 、总蛋白(TP)含量,检测脾脏和胸腺超氧化物歧化酶(SOD) 、谷胱甘肽过氧化物酶(GSH-Px) 、过氧化氢酶(CAT)活性及还原型谷胱甘肽(GSH) 、丙二醛(MDA)含量。结果显示:1)对于体重,对照组与Cy组无显著差异(P>0.05),各剂量BA组与Cy组无显著差异(P>0.05) 。2)与对照组相比,Cy极显著提高了血清AST、ALT活性(P < 0.01),而5.00 mg / kg BW BA极显著缓解了这一现象(P<0.01) 。3)与对照组相比, Cy引起了脾脏指数和胸腺指数的显著降低(P < 0.05),而0.05 mg / kg BW BA显著缓解了胸腺指数的降低(P<0.05) 。4)与对照组相比,Cy组胸腺SOD活性、胸腺CAT活性、脾脏GSH含量显著或极显著下降(P<0.05或P<0.01),脾脏MDA含量极显著升高(P<0.01);与Cy组相比,0.50 mg / kg BW BA组显著或极显著提高了脾脏和胸腺GSH-Px和CAT活性及脾脏GSH含量(P <0.05或P <0.01),极显著降低脾脏MDA含量(P < 0.01),但极显著降低了胸腺SOD活性(P<0.01);与Cy组相比,5.00 mg / kg BW BA组显著或极显著提高了脾脏和胸腺GSH-Px和CAT活性(P<0.05或P<0.01),极显著降低脾脏MDA含量(P<0.01) 。由此可见,BA能够改善Cy引起的小鼠免疫器官的氧化应激,对Cy诱导的氧化损伤有预防性的保护作用。

摘要:
Procyanidins B2 (PB2) was found to block T-2 toxin-induced reproductive toxicity and may be used as a dietary preventive agent. We investigated the mechanism of PB2 in blocking the T-2 toxin-induced apoptosis in Leydig cells in conjunction with analysis of T-2 toxin induced proapoptotic responses in Leydig cells including induction of oxidative stresses. The stress inducible proteins ASK, p38 and JNK were increased in response to T-2 toxin and the induction was attenuated by PB2. Furthermore, we showed that T-2 toxin suppressed mitochondrial membrane potential, reduced the levels of the mitochondrial respiratory chain complex I-V, adversely affected the level of mitochondrial proteins CypD, Bax, Bak, Bcl-2, Bcl-xl, Bax/Bcl-2 and cyt c and these adverse effects were counteracted by PB2 in dose-dependent manner. In summary, our results showed great potential of PB2 in preventing T-2 toxin-induced toxicity in Leydig cells through its potent antioxidant property. © 2018

以TM3细胞为研究对象,探讨原花青素B2对青霉酸致TM3细胞氧化损伤的修复作用。结果显示,在给予青霉酸基础上的同时添加原花青素B2,与单独添加青霉酸相比,TM3细胞内的SOD、CAT和GSH-Px的活性显著升高,细胞内MDA的含量显著降低,TM3细胞的活力则显著增加。表明原花青素B2可以通过修复细胞内的氧化损伤来拮抗青霉酸对TM3的细胞毒性。

摘要:
Dexamethasone (Dex), a potent anti-inflammatory/immunosuppressive agent, has been shown to induce oxidative stress. Betulinic acid (BA) is a pentacyclic lupane triterpene with a potent antioxidant activity. The aim of this study was to investigate the ameliorative effect and underlying mechanisms of BA on Dex-induced oxidative damage. Mice were pretreated with BA orally (0, 0.25, 0.5, and 1.0 mg/kg) daily for 14 days, and then a single dose of Dex (25 mg/kg body weight) was administered intraperitoneally 8 h after the last administration of BA to induce oxidative stress. BA pretreatment significantly alleviated Dex-induced changes of blood biochemical indices, increased the total antioxidant capacity (T-AOC), the activity of superoxide dismutase (SOD), and the ability of inhibiting hydroxyl radical (AIHR), reduced the level of malondialdehyde (MDA) in serum. Moreover, BA pretreatment enhanced the T-AOC, AIHR and the activity of peroxidase (POD) in liver, spleen and thymus. Concomitant with these biochemical parameters, BA pretreatment significantly reduced gene and protein expressions of apoptosis signal-regulating kinase 1 (ASK1), c-Jun N-terminal kinase (JNK) and P38 mitogen-activated protein kinase (P38 MAPK) in the lymphatic organs of Dex-treated mice. BA was found to effectively attenuate Dex-induced oxidative damage. These protective effects may be mediated in part through the JNK-P38 MAPK signaling transduction pathway and BA may be a potential therapeutic agent due to its anti-oxidative properties. © 2018 Elsevier Masson SAS

摘要:
Although mTOR (the mammalian target of rapamycin) can regulate intracellular free Ca2+concentration in normal cultured podocytes, it remains elusive as to how mTORC2/AKT-mediated Ca2+participates in the process of T-2 toxin-induced apoptosis. The potential signaling responsible for intracellular Ca2+ concentration changes was investigated using immunoblot assays in an in vitro model of TM3 cell injury induced by T-2 toxin. Changes in Ca2+ were assessed using the Ca2+-sensitive fluorescent indictor dye Fura 2-AM. The cytotoxicity of TM3 cells was assessed with an MTT bioassay, and apoptosis was measured using Annexin V-FITC staining. Following T-2 toxin treatment, the growth of cells, phospho-mTORSer2481, phospho-mTORSer2448, and phospho-AktSer473 were significantly decreased in a time-dependent manner, whereas Ca2+ and apoptosis were increased. T-2 toxin-induced apoptosis was prevented by BAPTA-AM (a Ca2+chelator) and MHY1485 (an mTOR activator), and the application of mTOR activator MHY1485 also prevented the increase of intracellular free Ca2+concentration in TM3 cells. Our results strongly suggest that T-2 toxin exposure induces apoptosis in TM3 cells by inhibiting mTORC2/AKT to promote Ca2+ production.

本试验旨在研究桦木酸(BA)对地塞米松(Dex)诱导氧化应激小鼠的保护作用和机制。将40只健康雄性昆明小鼠随机分为5组,即对照(NC)组、Dex组、0.25 mg / kg BA组、 0.50 mg / kg BA组、1.00 mg / kg BA组。NC组和Dex组小鼠灌服1 %的可溶性淀粉溶液,其余各组按不同剂量的BA灌服,连续14 d后,除对照组注射生理盐水外,其余4组均腹腔注射Dex(25 mg / kg BW)诱导氧化应激模型。检测各组小鼠肝脏、脾脏和胸腺总抗氧化能力(T-AOC) 、抑制羟自由基能力和过氧化物酶(POD)的活性,反转录(RT) -PCR检测脾脏和胸腺丝裂原活化蛋白激酶(MAPK)信号通路中凋亡信号调节激酶1(ASK1) 、c-Jun氨基末端激酶(JNK)和P38基因的表达量,蛋白质免疫印迹(Western Blot)法检测脾脏MAPK信号通路中ASK1、JNK和P38蛋白的表达量。结果表明:1)与NC组相比,Dex组的肝脏T-AOC、抑制羟自由基能力,脾脏POD活性,胸腺T-AOC、抑制羟自由基能力均极显著下降(P<0.01);与Dex组相比,0.50和1.00 mg / kg BA组肝脏T-AOC、抑制羟自由基能力以及POD活性均显著或极显著的升高(P < 0.05或P<0.01),0.50 mg / kg BA组脾脏T-AOC、0.50和1.00 mg / kg BA组脾脏抑制羟自由基能力、0.25和1.00 mg / kg BA组脾脏POD活性均显著或极显著的升高(P<0.05或P<0.01),0.50和1.00 mg / kg BA组胸腺T-AOC、抑制羟自由基能力和POD的活性均显著或极显著升高(P< 0.05或P<0.01) 。2)与NC组相比,Dex组脾脏和胸腺ASK1、JNK和P38 mRNA表达量均极显著升高(P < 0.01);与Dex组相比,0.50和1.00 mg / kg BA组脾脏和胸腺ASK1、JNK和P38 mRNA表达量均显著或极显著下降(P<0.05或P<0.01) 。3)与NC组相比,Dex组脾脏JNK和P38蛋白的表达量极显著升高(P<0.01);与Dex组相比,0.50 mg / kg BA组脾脏ASK1蛋白的表达量显著降低(P<0.05),0.25、0.50和1.00 mg / kg BA组脾脏JNK和P38蛋白的表达量显著或极显著降低(P<0.05或P<0.01) 。由此可见,BA预处理后,增强了Dex应激小鼠肝脏、脾脏和胸腺的T-AOC、抑制羟自由基能力和POD活性,降低了Dex应激小鼠脾脏和胸腺MAPK信号通路中ASK1、JNK和P38 mRNA表达量以及脾脏ASK1、JNK和P38蛋白的表达量。BA对Dex造成的氧化损伤具有预防性的保护作用,并且这种保护作用与JNK-P38 MAPK信号通路相关。

【目的】探讨T-2毒素诱导人肝L02细胞凋亡与自噬的作用,从而进一步深入探讨T-2毒素的毒性机理。【方法】用不同终浓度的T-2毒素处理L02细胞12h,使用MTT法检测细胞增殖率;用试剂盒提供的方法检测细胞中SOD、GSH-Px、MDA活性和CAT含量,培养上清中ALT、AST和LDH的活性;用Hoechst 33258检测细胞凋亡形态;用流式细胞仪检测细胞凋亡数量;用Western Blot检测

摘要:
为揭示钩吻素子的体外抗炎作用机理,利用硝酸还原酶法和ELISA法研究了不同质量浓度(100,200,400mg/L)的钩吻素子对一氧化氮(NO)和细胞因子白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)分泌的影响,运用了QPCR的方法检测了钩吻素子对诱导型一氧化氮合成酶(iNOS)、IL-1β、IL-6和TNF-αmRNA表达水平的影响,并运用Western blot法检测了钩吻素子对iNOS蛋白表达的影响。结果显示,100,200,400mg/L质量浓度的钩吻素子能够极显著的抑制RAW264.7细胞NO的产生以及IL-1β、IL-6和TNF-α的分泌(P<0.01),同时能够剂量依赖性的降低iNOS、IL-1β、IL-6和TNF-α的mRNA水平,抑制脂多糖(LPS)引起的iNOS蛋白表达的升高(P < 0.01)。由此推论,钩吻素子可能通过抑制炎症因子的分泌,减少NO的释放,下调iNOS、IL-1β、IL-6和TNF-α的mRNA水平,抑制iNOS蛋白过度表达达到其抗炎作用。

摘要:
研究桦木酸(betulinic acid,BA)对小鼠细胞因子的影响.取健康昆明系小鼠28只,随机分为4组,即空白对照组,BA低、中、高剂量组(0.25 mg/kg、0.5 mg/kg、1 mg/kg),灌胃给药,1次/d,连续14 d.检测BA对小鼠血清白细胞介素-2(IL-2)、IL-6和IL-10以及对腹腔巨噬细胞肿瘤坏死因子(TNF-α)水平的影响.结果表明,BA可降低小鼠血清IL-2和IL-6的分泌量,提高腹腔巨噬细胞TNF-α的分泌量.说明BA在治疗机体肿瘤、感染和炎症的过程中发挥着重要作用.

摘要:
Obesity, which is associated with dietary habits, has become a global social problem and causes many metabolic diseases. In China, both percentages of adult obesity and overweight are far lower compared to western countries. It was designed to increase the two levels of daily intake in human, namely 3.8% and 6.5%, which are recommendatory intake (25 g/d) and Chinese citizens' practical intake (41.4 g/d), respectively. The mice were respectively fed with feeds added with soybean oil, lard or the oil blended by both for 12 weeks. In the mice fed with diet containing 3.8% of the three oils or 6.5% blended oil, their body weight, body fat rate, cross-sectional area of adipocytes, adipogenesis and lipogenesis in adipose were decreased, whereas hydrolysis of triglyserides in adipose was increased. This study demonstrated that the oil mixture containing lard and soybean oil had a remarkable anti-obesity effect. It suggests that the traditional Chinese dietary habits using oils blended with lard and soybean oil, might be one of the factors of lower percentages of overweight and obesity in China, and that the increasing of dietary oil intake and the changing of its component resulted in the increasing of obesity rate in China over the past decades.

摘要:
IRW (Ile-Arg-Trp) is an angiotensin converting enzyme (ACE) inhibitory peptide identified from egg protein ovotransferrin. Given its role also as an antioxidant, the aims of this work were to investigate whether IRW exerts its protective effect against oxidative stress by inhibiting intracellular ROS accumulation and blocking ROS activated mitochondria-mediated cell apoptosis pathway. Adding 200 μM H2O2 to cells led to decreased cell viability while increased (ROS) formation. However, IRW significantly inhibited H2O2-induced cytotoxicity and ROS production. Flow cytometry was used to assess the cell apoptosis and the mitochondrial membrane potential. Pretreatment with IRW decreased cell apoptosis while elevated the mitochondrial membrane potential compared to H2O2 treated cells. Furthermore, IRW reduced the levels of caspase-9 and caspase-3 and thus improved cell survival. The results demonstrate that IRW can significantly protect Caco-2 cells against oxidative stress by inhibiting intracellular ROS accumulation and blocking the ROS activated mitochondria-mediated cell apoptosis pathway. © 2017 Elsevier Ltd

摘要:
目的:探讨桦木酸体外拮抗T-2毒素、减少T-2毒素对细胞的氧化应激作用.方法:选用人肝脏细胞L02作为研究对象,将含0.2、2.0、20.0 μmol/L的桦木酸和5 nmol/L T-2毒素共同暴露在L02中12h,后测定细胞中SOD、MDA、GSH、GSH-Px、CAT、ALT和AST水平以及细胞培养上清液中的LDH活性,检测细胞氧化损伤情况.结果:随着浓度的提高,桦木酸能剂量依赖地改善由T-2毒素引起的氧化损伤.结论:桦木酸能拮抗T-2毒素,从而改善由T-2毒素引起的氧化损伤.

摘要:
钩吻对动物具有良好的促生长作用,本研究以猪小肠上皮细胞IPCE-J2为研究对象,探讨了钩吻中含量最高的生物碱单体钩吻素子对沙门氏杆菌侵染的影响,结果显示钩吻素子能够激活Ahr受体,减少沙门氏杆菌对IPEC-J2细胞的侵染,而添加Ahr受体抑制剂α-NF可以拮抗钩吻素子的作用,表明Ahr受体在这一过程中发挥十分重要的作用。
