摘要:
In this study, stable water-in-oil emulsions stabilized solely by a naturally occurring steroidal sapogenin was reported for the first time. The results show that a concentrated emulsion with an internal water ratio of up to 60% can be obtained with only 3% of diosgenin concentration. The concentration of diosgenin had a significant effect on the microstructure and rheological properties of the emulsions. More importantly, the emulsion has excellent freeze/thaw stability and thermal stability. The results of polarized light microscopy, CLSM, and XRD indicate that the great structural properties and high stability of the emulsion can be attributed to the combined action of the diosgenin crystal shells on the droplet surface and needle-crystals in the continuous phase. That is, Pickering stabilization and network stabilization acting synergistically on stabilization of the emulsions. This novel food grade water-in-oil emulsions demonstrated great potential for application in food and biomedical-related fields. © 2021 Elsevier Ltd

Buckwheat was processed by solid-state fermentation (SSF) with the probiotic fungal strain Eurotium cristatum YL-1. The effects of SSF on the phytochemical content, as well as the antioxidant and α-glucosidase inhibitory activities, on buckwheat were revealed. Metabolite differences between non-fermented buckwheat (BW) and E. cristatum fermented buckwheat (FBW) were investigated by LC-MS/MS-based untargeted metabolomics. Results showed that 103 and 68 metabolites remarkably differed between BW and FBW in positive and negative ionization modes, respectively. Most phenolic compounds and alkaloids were significantly up-regulated during SSF. Hydrolytic enzymes (i.e., β-glucosidase, α-amylase, protease, and cellulase) were produced by the filamentous fungus E. cristatum during SSF. In vitro spectrophotometric assays demonstrated that the total phenolics content, ferric reducing antioxidant power, reducing power, scavenging activities of DPPH radical and ABTS[rad]+, and α-glucosidase inhibitory activity of buckwheat were considerably enhanced after processing by SSF with E. cristatum. Additionally, solvents with different polarities significantly influenced the antioxidant and α-glucosidase inhibitory activities of buckwheat extracts. Our study indicated that processing by SSF with E. cristatum can greatly improve the phytochemical components of buckwheat and consequently contribute to its antioxidant and α-glucosidase inhibitory activities. SSF with E. cristatum is an innovative method for enhancing the health-promoting components and bioactivities of buckwheat. © 2021 Elsevier Ltd

The impact of nanoemulsions containing triglycerides with different fatty acid chain lengths on the bioavailability of a highly lipophilic bioactive: 5-demethylnobiletin (5-DN) was investigated. 5-DN was encapsulated in nanoemulsions fabricated using either medium-chain triglycerides (MCT) or long-chain triglycerides (LCT). They were then subjected to in vitro digestion, and the resulting mixed micelles was applied to a Caco-2 cell model. Higher 5-DN bioaccessibility was found for the MCT-nanoemulsion (13%) than for the LCT-nanoemulsion (7%). However, only 30% 5-DN in MCT crossed the Caco-2 monolayer and 50% was metabolized, while 60% 5-DN in LCT crossed the monolayer and only 10% was metabolized. More lipid droplets and chylomicrons were also formed for the LCT nanoemulsions, indicating greater 5-DN transported through lymph. Although MCT gave a higher 5-DN bioaccessibility, the final amount of 5-DN absorbed and transported to the lymph was inferior to that of the LCT formulation. © 2020 Elsevier Ltd

摘要:
Mustard (Brassica juncea) is a cruciferous vegetable used as a food spice and folk medicine worldwide. Mustard contains numerous phytochemicals such as: vitamins, minerals, dietary fiber, chlorophylls, glucosinolates (and their degradation products), polyphenols and volatile components(allyl isothiocyanate, 3-butyl isothiocyanate, etc.). The content and exact chemical composition of these phytochemicals is affected by plant variety, growth environment, extraction process and food processing methods. In addition, mustard may possess a plethora of pharmacological activities including anti-oxidation, anti-inflammation, and bacteriostatic and antiviral activity. Mustard has also been used to combat several illnesses such as cancer, obesity, depression, diabetes, and cataracts. This review provides an overview of plant characteristics, types, origins, distribution, and consumption methods of Mustard, as well as its phytochemicals and biological activities. The findings of this paper may serve as references for the development and utilization of Mustard resources. © 2020 The Author(s). Published with license by Taylor & Francis Group, LLC.

A new split aptamer sensing platform is developed for highly sensitive and selective detection of theophylline based on single molecule photobleaching (SMPB) technique. The sensing system contains two probes. One is formed by one streptavidin and four biotinylated RNA fragments labelled with fluorescein isothiocyanate (FITC). Each biotinylated RNA fragment contains two repeating aptamer fragments. The other probe is the complementary aptamer fragment labelled with Cy5 dye. The existence of theophylline can trigger the first probe to bind as many as eight Cy5-labelled probes. The average combined number depends on the theophylline concentration and can be measured by SMPB technique. In the sensing system, the dual-color fluorescence colocalization is performed by the red fluorophore (Cy5) and green fluorophore (FITC), in which the red fluorophore is utilized for quantitative counting of photobleaching steps, while the green fluorophore serves as a counting reference to increase detection efficiency. On basis of the principle, an ultra-sensitive sensing platform of theophylline is created with a low limit of detection (LOD) of 0.092 nM. This work provides not only a highly sensitive method for theophylline detection but also a novel perspective for the applications of SMPB technology to construct biosensors. © 2020 Elsevier B.V.

Controlled-synthesis of carbon nanodots (CNDs) with tunable properties has always been challenging. Here, novel CNDs (blue-emitting CNDs (b-CNDs) and green-emitting CNDs (g-CNDs)) with tunable redox abilities and emission are synthesized via one-pot hydrothermal method. The redox ability of CNDs is regulated by surface state tailoring. The abundant reductive groups on the surfaces endow the b-CNDs appealing reducibility, whereas g-CNDs with fewer surface functional groups are deficient in reducibility. Using HAuCl4 indicator, the distinct reducibility of CNDs is visually identified. Gold salts are reduced to Au nanobelts under reduction and stabilization of b-CNDs, causing plasmons-induced color changes, whereas g-CNDs have no reduction effect on Au3+. The size-evolution process of Au nano-species can guide not only plasmonic absorbance alteration but also fluorescence (FL) and second-order scattering (SOS) changes. Therefore, based on the plasmonic morphology-induced SOS increasement and the Förster resonance energy transfer-induced FL switch off, the b-CNDs contribute to a ratiometric sensor coupled with FL and SOS for Au3+ detection. The ratiometric strategy displays a better sensitivity and selectivity and a broader linear range than single-signal SOS or FL method. The regulation of redox CNDs and ratiometric assay of Au3+ provide a new prospect for the preparation and application of versatile CNDs. © 2020 Elsevier B.V.

Thermal processing may generate toxicants. Carbon dots (CDs) from baked foods are toxic to cells; however, their molecular mechanism is still unexplored to date. The present study investigated the effects of CDs from roasted chicken breasts on normal rat kidney (NRK) and Caco-2 cells. The average size of CDs heated at 200 °C and 300 °C was about 2.8 nm and 1.2 nm, respectively. The element and surface groups of CDs were analyzed via X-ray photoelectron spectroscopy (XPS) and Fourier transform infrared spectroscopy (FTIR), respectively. It was confirmed that the CDs were internalized in lysosomes and induced apoptosis. Furthermore, Z-VAD-FMK did not decrease the rate of apoptosis. The acquired data further confirmed that these internalized CDs enlarged lysosomes, decreased the lysosomal enzyme degradation activity and increased the lysosomal pH value. An increase in the co-localization of RIPK3 in lysosomes in the CD-treated groups was observed. The CD treatment increased the protein level of receptor interaction protein 1 (RIPK1) and receptor interaction protein 3 (RIPK3). Overall, CDs from the baked chicken breast induced lysosomal membrane permeabilization and initiated lysosome-dependent cell death and necroptosis. Our results elucidated the toxic mechanism of CDs from baked chicken breast and implied that food thermal processing at a lower temperature is beneficial to human health. In this study, naturally occurring ingredient diosgenin was utilized as an organogelator for structuring canola oil. Results show that stable diosgenin-based organogel can be obtained at only 2% of diosgenin concentration when the gel preparation temperature is 100 °C. Oil binding capacity and rheological properties of the organogel were investigated. Results demonstrate that these two macroscopic characteristics of the organogels can be significantly modified by simply changing the gel preparation temperature or diosgenin concentration. When the preparation temperature was 120 °C and the diosgenin concentration higher than 4%, oil binding capacity of at least 90% were obtained. Furthermore, higher gel preparation temperature resulted in higher G′values of the diosgenin-based organogels. According to the results analyzed by polarized light microscopy, XRD and FT-IR, it can be found that the possible gelation mechanism of the diosgenin-based organogels is formation of supramolecular structures by self-assembly of diosgenin molecule crystals via hydrogen bonding interaction. Varying gel preparation conditions of the organogels lead to self-assembly of diosgenin molecules to form different microstructures. Therefore, diosgenin can be considered as a good organogelator for producing functional organogel from canola oil. The novel diosgenin-based organogel is expected to be widely used in bio-related fields such as food and pharmaceutical industries. © 2020, Springer Science+Business Media, LLC, part of Springer Nature.

Lipolysis products released during digestion exert positive metabolic impacts on the nutrition of newborns. However, the lipolysis behavior of yak milk lipids during digestion remains unknown. In this study, the simulated in vitro infant gastrointestinal digestion of cow, yak and standardized yak milk fat globules the same size as those from cow milk (Cow MF, Yak MF and Yak SMF) were compared. Although Cow MF showed a higher lipolysis rate at the beginning of gastric digestion, Yak MF and Yak SMF exhibited a higher lipolysis level during later gastrointestinal digestion. Higher hydrolysis efficiency of yak milk lipids was due to their lipid properties, including their composition and structure. Furthermore, yak milk lipids released more unsaturated fatty acids than Cow MF throughout digestion. This study highlights the crucial role of lipid characteristics in the efficient digestion of milk lipids and provides new insight for the design of yak milk infant diets. © 2020 Elsevier Ltd

绿茶是我国饮用范围最广、最受欢迎的一类茶叶。不同品种绿茶叶外观上差别较小,非专业人员难以直接用肉眼进行辨别。传统化学方法操作复杂、检测费用较高,对样品具有破坏性,无法实现快速无损分析。近红外光谱技术是一种简便、快速、无损、重现性好、可直接用于在线定性定量分析的新型分析技术。由于种植方式以及土壤、气候等生长环境的差异,不同品种绿茶叶中含氢基团有机物的种类和含量也不相同,因此可以通过扫描样品的近红外光谱,得到不同品种绿茶叶的特征信息,实现对不同品种绿茶叶的快速鉴别。研究提出了一种基于近红外光谱与化学计量学技术对不同品种绿茶的快速无损鉴别方法。使用近红外光谱仪得到了八个品种绿茶样品的光谱图,用主成分分析方法对不同品种绿茶样品数据进行了聚类分析。使用连续小波变换方法消除了光谱信号中的基线干扰,从而提升聚类效果。利用基于标准偏差与相对标准偏差的变量筛选方法进一步提高了聚类结果的准确性。结果表明:主成分分析后样品的第一主成分和第二主成分的方差贡献率之和在90%以上,可以选取前两个主成分进行聚类分析。直接采用原始数据进行聚类分析的准确率较低,难以满足应用需要;连续小波变换可以有效地消除光谱信号中的基线干扰。与直接使用原始光谱聚类结果相比,采用连续小波变换后聚类效果有显著提升,但依旧不能实现所有品种茶叶样品的准确鉴别。为了进一步提高方法的稳健性和分类结果的准确性,选取了标准偏差和相对标准偏差较大的波长数据进行聚类分析。在符合平均值大于1%的波长范围内,剔除标准偏差小于5‰的波长,进一步选择较大相对标准偏差值对应的波长点进行聚类分析。采用这种方式,可以仅使用几十个甚至是几个波长即可实现绿茶样品品种的准确聚类分析。波长筛选方法可以大大提高主成分分析结果的准确性,采用近红外光谱分析技术与化学计量学方法可以实现对不同品种绿茶的快速鉴别。经过对各个光谱吸收区域波长所对应官能团分析后,初步得出多酚、酰胺类以及氨基酸类物质的种类不同或含量差异是形成绿茶品种差异的重要原因。所提出的基于近红外光谱与化学计量学技术的方法具有较强的鉴别能力,为绿茶的快速无损分析提供了一种新手段。

1,3-Dioleoyl-2-palmitoylglycerol (OPO), an important structured triacylglycerol, is generally supplemented into infant and toddler formulas as a nutrient fortifier. However, its role on fecal microbiota has not received attention in the last decades. In this study, anaerobic fermentation in vitro and high-throughput sequencing were conducted to investigate effects of standard OPO alone (OPO-A) and OPO-enriched plant-oil formula (OPO-P) on toddler fecal microbiota. The results showed OPO could be efficiently utilized by toddler fecal microbiota. OPO supplementation enhanced the production of short-chain fatty acids and lactic acid by microbiota. Furthermore, OPO promoted the growth of Bifidobacterium whereas inhibited the growth of Enterobacter at different fermentation time when compared with the control group. Besides, OPO supplementation enhanced bacterial diversity. Thus, this study indicated that OPO could modulate the fecal microbiota. The results provided useful information for further exploring the physiological role of OPO on colon, and provided new ideas for application of OPO. © 2019, © 2019 The Author(s). Hexavalent chromium (Cr(VI))is a well-known human carcinogen and a strong oxidizer that causes severe DNA damage. However, the associations between epigenetic dysregulation and DNA damage have not been well-characterized. In this study, we evaluated the effects of short-term and long-term exposure to Cr(VI)in human bronchial epithelial (16HBE)cells. Then, we explored the role of epigenetic modification in Cr(VI)-induced DNA damage. We found that short- and long-term exposure to Cr(VI)induced DNA damage and reduced the expression 53BP1, but increased the expression of other DNA repair mediators. Short- and long-term exposure to Cr(VI)reduced the levels of H3K18ac and H3K27ac and reduced their enrichment at the promoter of 53BP1. Long-term Cr(VI)exposure resulted in multiple malignant characteristics including cell invasion, migration, and tumorgenicity. These data demonstrated that reduced H3K18ac and H3K27ac following Cr(VI)treatment contributed to the suppression of 53BP1. Our study demonstrated that epigenetic changes and DNA damage responses are involved in short-term toxicity and long-term carcinogenesis induced by Cr(VI). © 2019 Elsevier Ltd

The effects of alcohol stress on Caenorhabditis elegans were investigated. Nematodes were exposed to 1–5% alcohol, after which lifespan, movement, reactive oxygen species, antioxidant enzymes, cell apoptosis, and expression of apoptosis-related proteins were evaluated. Low concentrations of alcohol (1–2%) significantly enhanced total antioxidant capacity, along with an observable increase in normal locomotion. Total antioxidant activity was significantly lower at high (4–5%) than at low (1–2%) concentrations of alcohol. These results suggested that alcohol intake leads to an imbalance in oxidative defense in C. elegans. None of the groups administered alcohol displayed signs of apoptosis. Low concentrations of alcohol delayed decrepitude, promoted resistance to oxidative stress, and improved antioxidant capacity. However, nematodes could not withstand the oxidative stress induced by and toxicity of high alcohol concentrations, which eventually shortened their lifespan. 摘要:
通过消除光谱中的冗余信息变量,挑选出代表样品性质的特征变量代替全谱建立定量模型,可以提高近红外分析结果的准确性。基于进化论中适者生存原理的竞争性自适应重加权采样(CARS)算法因具有计算速度快、筛选得到的特征波长少等优点,在近红外特征变量筛选方面得到了广泛的应用。然而该方法在计算过程中容易出现校正集和验证集结果不一致情况。这是因为算法过于强调校正集交叉验证结果,且并未考虑相邻变量之间的协同作用。为了建立更加稳健的变量筛选方法,通过结合“窗口”以及CARS算法的优势,提出了一种基于窗口竞争性自适应重加权采样(WCARS)策略的近红外特征变量筛选方法,并将其应用于复杂植物样品近红外光谱与其化学成分含量之间的建模分析。采用WCARS方法可以实现准确定量分析,且通过与竞争性自适应重加权采样(CARS)方法结果相比较,WCARS方法得到的校正集和预测集结果一致,在一定程度上减少了过拟合问题的出现。该策略能有效增强特征变量选择的稳健性,提高了定量模型的可信度,具有一定的应用价值。

摘要:
A novel lysosome-targetable phthalimide fluorescent probe was designed for detecting palladium based on ESIPT for signal transduction. The fluorescent probe conjugating with allylcarbamate displayed weak fluorescent due to the ESIPT process hinder by allylcarbamate. But with the addition of palladium, the ESIPT emission was recovery though the palladium-catalyzed deallylation reaction and the fluorescence intensity exhibited 40-fold enhancement at 511 nm. In addition, the probe showed excellent selectivity, high sensitivity, fast responds and low limit detection for palladium with a larger Stoke-shift. Moreover, the targetable probe was also successfully applied for detecting palladium in lysosomes of living cells. Hence, the probe though ESIPT modulation is a promising for monitoring palladium in practical samples. (C) 2018 Published by Elsevier B.V.

5-Demethyltengeretin (5DT) is a citrus flavonoid with various potential health benefits. To provide physiologically relevant information on the anti-inflammatory properties of 5DT, we identified the major metabolite of 5DT in the mouse colon and established its anti-inflammatory effects in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. CD-1 mice were fed with a 5DT-containing diet for four weeks, and colonic mucosa samples were collected and subjected to LC-MS analysis. Xanthomicrol (XAN) was identified as the major metabolite of 5DT in the mouse colon. More importantly, the colonic level of XAN was about 3.1-fold higher than that of 5DT. The anti-inflammatory effects of 5DT and XAN were determined in LPS-stimulated macrophages. XAN produced significant inhibitory effects on the production of nitric oxide and PGE2. Western blotting and real-time PCR analyses demonstrated that XAN greatly decreased the protein and mRNA levels of iNOS as well as the protein level of COX-2. Furthermore, XAN also reduced the production of pro-inflammatory cytokine IL-1β and induced the expression of anti-oxidative enzyme HO-1. Conclusion: Our results demonstrated that XAN is a major metabolite of 5DT in the colon of mice fed with 5DT, and XAN may play important roles in the anti-inflammatory effects elicited by orally administered 5DT. © 2018 The Royal Society of Chemistry.

In this work, we demonstrated a single molecule photobleaching-based strategy for the ultrasensitive detection of adenosine. A modified split aptamer was designed to specifically recognize individual adenosine molecules in solution. The specific binding of dye-labeled short strand DNA probes onto the elongated aptamer strand in the presence of adenosine resulted in a concentration-dependent self-aggregation process. The degree-of-aggregation (DOA) of the short DNA probes on the elongated aptamer strand could then be accurately determined based on the single molecule photobleaching measurement. Through statistically analyzing the DOA under different target concentrations, a well-defined curvilinear relationship between the DOA and target molecule concentration (e.g., adenosine) was established. The limit-of-detection (LOD) is down to 44.5 pM, which is lower than those recently reported results with fluorescence-based analysis. Owing to the high sensitivity and excellent selectivity, the sensing strategy described herein would find broad applications in biomolecule analysis under complicated surroundings. © 2018 American Chemical Society.

摘要:
The dynamics of water in sodium alginate film-forming solutions during drying were investigated by low-field nuclear magnetic resonance. At the beginning of drying, three transverse relaxation times at around 1.74, 28.48 and 305.38 ms were assigned to the tightly bound, moderately bound, and free water, respectively. Moreover, the free water was evaporated, followed by the formation of water cluster in the entangled alginate chains network as the drying process continued (beyond 840 min of drying time). Towards the third stage of drying process, water clusters in alginate samples was disappeared, resulting that loosely bonded waters were constrained within the weak-gel network. The observed three relaxation times revealed the multi-exponential relaxation behavior of water in alginate, which indicated that this polymer exhibited spatial heterogeneity during drying. On the basis of diffusive exchange theory, the dimension range of alginate network decreased from 1.94–23.99 to 1.83–6.62 µm as the alginate films solidified. © 2018 Elsevier Ltd

To decrease the drying time and energy consuming, water migration and water–protein interactions of lamb meat being air-dried at 35°C were investigated. Low-field nuclear magnetic resonance T2b indicated the water–protein interactions changed during air-drying. Area of T21 (intramyofibrillar water) decreased, meanwhile the area of T22 (extra-myofibrillar water) increased when the moisture content decreased from 55 to 45%, indicating the water migrated from myofibril to extramyofibril. Drying rate (the rate of water migration from meat to air) could be predicted by the area of T2 populations, and the correlation coefficient was 0.990. Change of water binding and hydration in myofibrils was evident by the increase in hydrophobicity and decrease in solubility of myofibrillar protein. Differential scanning calorimetry showed denaturation of myosin in dried meat which might result in water migration from myofibril to extramyofibril space. In conclusion, water–protein interactions changed, and then influenced the drying rate during drying. © 2017 Taylor & Francis.

摘要:
A novel mitochondria-targeted fluorescent probe compound 1 for H2O2 has constructed based on a coumarin as the fluorophore and a boronate moiety as the reaction site, which showed colorimetric and ratiometric fluorescence dual response to H2O2. This probe displayed a large blue-shifted in emission wavelength (105 nm) with two well-resolved emission bands toward hydrogen peroxide. The compound 1 can be quantitative, good selective and sensitive recognition to H2O2 with low detection limit (10 nM). The sensing mechanism was confirmed by H NMR and HRMS spectroscopy. The mechanism regarding the optical responses of the compound 1 toward H2O2 was also determined by density function theory (DFT) calculations. Moreover, the probe could be used for determining H2O2 in living cells by ratiometric fluorescence imaging with excellent mitochondrial targeting. (C) 2017 Published by Elsevier B.V.
