l-Theanine delays d-galactose-induced senescence by regulating the cell cycle and inhibiting apoptosis in rat intestinal cells

BACKGROUND: Intestinal senescence is associated with several aging-related diseases. l-Theanine (LTA) has demonstrated strong potential as an antioxidant and antisenescence agent. This study investigated the regulatory effect of LTA on cellular senescence using an in vitro model of d-galactose (D-Gal)-induced senescence in the rat epithelial cell line, intestinal epithelioid cell-6 (IEC-6). RESULTS: Treatment of IEC-6 cells with 40 mg/mL D-Gal for 48 h resulted in the successful development of the senescent cell model. Compared with D-Gal alone, both LTA preventive and delayed intervention increased cell viability and the ratio of JC-1 monomers to aggregates, increased the antioxidant capacity, and decreased the advanced glycation end product (AGE) levels and the overall number of senescent cells. Preventive and delayed intervention with 1000 μM LTA alleviated the D-Gal-induced cell cycle arrest by regulating p38, p53, CDK4, and CDK6 expression at the mRNA and protein levels, and further induced CycD1 proteins. Moreover, LTA preventive intervention reduced apoptosis to a greater degree than delayed intervention by upregulating the expression of the receptors of AGEs, Bax, Bcl-2, and NF-κB at the mRNA and protein levels. CONCLUSION: Our findings indicate that LTA intervention could attenuate senescence in IEC-6 cells by regulating the cell cycle and inhibiting apoptosis. © 2023 Society of Chemical Industry. © 2023 Society of Chemical Industry.

BACKGROUND: Heat stress (HS) damages the intestines, disrupting gut microbiota and immune balance. l-Theanine (LTA), found in tea, alleviates oxidative stress and cell apoptosis under HS; however, its effects on gut microbiota and immunity under HS remain unclear. To investigate this, we administered LTA doses of 100, 200, and 400 mg·kg−1·d−1 to C57BL/6J mice. On day 44, the model group and LTA intervention group were subjected to continuous 7-day HS treatment for 2 h per day. RESULTS: The results demonstrated that LTA intervention improved food intake, body weight, and intestinal epithelium, and reduced the water intake of heat-stressed mice. It increased the abundance of Turicibacter, Faecalibaculum, Bifidobacterium, and norank_f_Muribaculaceae, while reducing that of Lachnoclostridium and Desulfovibrio. LTA intervention also increased the concentrations of amino acid and lipid metabolites, regulated macrophage differentiation stimulated by gut microbiota and metabolites, reduced the antigen presentation by macrophages to the specific immune system, promoted B-cell differentiation and sIgA secretion, inhibited pro-inflammatory factors, and enhanced intestinal defense. Mechanistically, LTA downregulated heat shock protein 70 expression and the TLR4/NF-κB/p38 MAPK signaling pathway, restoring gut microbiota and immune balance. CONCLUSION: We suggest that LTA can alleviate HS by modulating gut microbiota, metabolites, and immunity, indicating its potential as a natural active ingredient for anti-HS food products. © 2023 Society of Chemical Industry. © 2023 Society of Chemical Industry.

Heat stress caused by heatwaves, extreme temperatures, and other weather can damage the intestinal barrier of organisms. L-Theanine (LTA) attenuates heat stress-induced oxidative stress, inflammatory responses, and impaired immune function, but its protective effect on the intestinal barrier of heat-stressed organisms is unclear. In this study, low (100 mg kg(-1) d(-1)), medium (200 mg kg(-1) d(-1)), and high (400 mg kg(-1) d(-1)) dosages of LTA were used in the gavage of C57BL/6J male mice that were experimented on for 50 d. These mice were subjected to heat stress for 2 h d(-1) at 40 ± 1 °C and 60 ± 5% RH in the last 7 d. LTA attenuated the heat stress-induced decreases in body mass and feed intake, and the destruction of intestinal villi and crypt depth; reduced the serum levels of FITC-dextran and D-LA, as well as the DAO activity; and upregulated the colonic tissues of Occludin, Claudin-1, and ZO-1 mRNA and occludin protein expression. The number of goblet cells in the colon tissue of heat-stressed organisms increased in the presence of LTA, and the expression levels of Muc2, Muc4 mRNA, and Muc2 protein were upregulated. LTA increased the abundance of Bifidobacterium and Turicibacter, and decreased the abundance of Enterorhabdus and Desulfovibrio in the intestinal tract of heat-stressed organisms and restored gut microbiota homeostasis. LTA promoted the secretion of IL-4, IL-10, and sIgA and inhibited the secretion of TNF-α and IFN-γ in the colon of heat-stressed organisms. The expressions of Hsf1, Hsp70, Hsph1, TLR4, P38 MAPK, p-P65 NF-κB, MLCK mRNA, and proteins were downregulated by LTA in the colon of heat-stressed organisms. These results suggest that LTA protects the intestinal barrier in heat-stressed organisms by modulating multiple molecular pathways. Therefore, this study provides evidence on how tea-containing LTA treatments could be used to prevent and relieve intestinal problems related to heat stress.

摘要:
The protein levels in a diet are correlated with immunity but the long-term intake of excessive protein can compromise various aspects of health. L-theanine regulates immunity and protein metabolism; however, how its regulatory immunity effects under a high-protein diet are unclear. We used proteomics, metabonomics, and western blotting to analyze the effects of diets with different protein levels on immune function in rats to determine the role of L-theanine in immunity under a high-protein diet. The long-term intake of high-protein diets (>= 40% protein) promoted oxidative imbalance and inflammation. These were alleviated by L-theanine. High-protein diets inhibited peroxisome proliferator-activated receptor (PPAR)alpha expression through the inter-leukin (IL)-6/signal transducer and activator of transcription (STAT)3 pathway and mediated inflammation. L-theanine downregulated anti-fatty acid-binding protein 5 (FABP5), inhibited the IL-6/STAT3 axis, and reduced high-protein diet-induced PPAR alpha inhibition. Therefore, L-theanine alleviates the adverse effects of high-protein diets via the FABP5/IL-6/STAT3/PPAR alpha pathway and regulates the immunity of normally fed rats through the epoxide hydrolase (EPHX)2/nuclear factor-kappa B inhibitor (I kappa B)alpha/triggering receptor expressed on myeloid cells (TREM)1 axis.

Ovalbumin (OVA), a common food protein, can cause deadly allergies with intestine-specific immune reactions. L-Theanine (LTA) shows great potential for regulating intestinal immunity. To investigate the regulatory effect of LTA intervention on intestine-specific immunity, a 41 day experiment was performed on BALB/c OVA-sensitized mice. The results show that injecting female mice intraperitoneally with 50 μg of OVA and administering 30 mg of OVA 4 times can successfully establish an OVA-sensitized mouse model. LTA intervention significantly increased weight gain and thymus index (p < 0.05), decreased allergy and diarrhea scores (p < 0.05), and improved jejunum structure. Meanwhile, the histological score and degranulation of mast cells decreased. LTA intervention increased Clostridiales, Lachnospiraceae, Lactobacillus, Prevotella, and Ruminococcus abundance while decreasing Helicobacter abundance. Flow cytometry and Western blotting results indicated that 200 and 400 mg/kg of LTA upregulated the expression of T-bet and Foxp3 proteins (p < 0.05), thus promoting the differentiation of jejunum CD4+ T cells to Th1 and Tregs and increasing the cytokines IFN-γ, IL-10, and TGF-β (p < 0.05). We found that 200 and 400 mg/kg of LTA downregulated the expression of RORγt and GATA3, thus inhibiting the differentiation of Th2 and Th17 cells and decreasing cytokines IL-4, IL-5, IL-13 TNF-α, IL-6, and IL-17A (p < 0.05). LTA inhibited the degranulation of mast cells and significantly decreased the serum levels of OVA-IgE, HIS, and mouse MCPT-1 (p < 0.05). Therefore, LTA intervention alleviated OVA allergy by improving intestine-specific immunity. © 2022 American Chemical Society. All rights reserved.

摘要:
高蛋白饮食可导致焦虑或抑郁自主行为,自主行为是判断是否具有焦虑或抑郁病症的重要方法。本研究通过对SPF级6周龄SD雄性大鼠进行40 d灌胃实验,采用旷场与明暗箱实验观察以及测定生理生化指标的方法,探究不同剂量L-茶氨酸(100、200、400 mg/(kg mb·d))对不同蛋白水平饮食(蛋白质供能比分别为20%、30%、40%、50%)SD大鼠行为变化的干预作用,以期为高蛋白饮食的多元化营养干预及L-茶氨酸深层次利用提供科学依据。结果表明:与蛋白质供能比为20%的普通维持饲料组相比,蛋白质供能比为50%的高水平高蛋白饲料组的采食量、体质量显著减少(P<0.05),大鼠在明箱中的停留时间显著缩短(P<0.05),进入明箱的次数显著减少(P<0.05),多巴胺、5-羟色胺、去甲肾上腺素质量浓度减少,但无显著差异,说明高水平高蛋白饮食可诱导大鼠抑郁行为。与高水平高蛋白饲料组相比,低剂量L-茶氨酸干预的高水平高蛋白饲料组大鼠体质量显著升高(P<0.05),且在明箱的停留时间显著延长(P<0.05);低、中剂量L-茶氨酸干预的高水平高蛋白饲料组大鼠在旷场箱的水平移动格数、后肢站立次数显著增加(P<0.05);各剂量L-茶氨酸干预的高水平高蛋白饲料组大鼠进入明箱的次数显著增加(P<0.05),且血清中多巴胺、去甲肾上腺素质量浓度显著升高(P<0.05);同时低剂量L-茶氨酸干预的高水平高蛋白饲料组大鼠血清中5-羟色胺质量浓度显著升高(P<0.05)。综上,L-茶氨酸具有改善高蛋白饮食所致焦虑或抑郁SD大鼠自主行为的作用,其机制可能与单胺类递质的代谢相关。

以茶树品种碧香早夏秋季一芽二叶茶鲜叶为原料,在红茶加工过程中,用微波初干工艺替代传统热风初干工艺,按不同微波强度、微波时间、摊叶厚度设置17组微波初干工艺处理,从感官品质、滋味品质成分和香气品质成分3个方面,探究了微波初干工艺对红茶品质的影响。结果表明,与传统热风初干工艺相比,采用微波强度600 W、微波时间3 min、摊叶厚度2 cm的微波初干工艺技术,所制红茶感官品质较好,红茶中的茶黄素、茶红素、总儿茶素、酯型儿茶素及表没食子儿茶素没食子酸酯(EGCG)、氨基酸、咖啡碱、可溶性糖的含量显著升高(P<0.05),茶多酚、茶褐素含量显著降低(P<0.05),香气品质成分由43种增加到50种,其中芳樟醇、苯乙醇等花果香香气成分的相对含量显著提高(P<0.05),并产生了(Z)-3,7-二甲基-2,6-辛二烯 -1-醇、2,6,6-三甲基-1,3-环己二烯-1-甲醛、2,6,6-三甲基-1-环己烯-1-甲醛、丁香酚等甜香、花香成分。微波初干工艺有利于提高红茶品质。

L-Theanine (LTA), a non-protein-derived amino acid, is widely used as a functional ingredient. Here, we established an immune-stressed mouse model by infecting mice with enterotoxigenic Escherichia coli (ETEC) to study the immunomodulatory effects of LTA with respect to nucleotide-binding oligomerization domain (NOD)-like receptor signaling in vivo. Compared to ETEC control group, LTA alleviated ileal tissue lesions, significantly attenuated interleukin-1 beta (IL-1β) and tumor necrosis alpha (TNFα) overexpression, which was consistent with their protein levels. Additionally, LTA inhibited the ETEC-induced overexpression of NOD2 and receptor-interacting protein 2 (RIP2) proteins and mRNAs, TGE-beta-activated kinase 1 (TAK1) and proteasome subunit alpha type-7 (PSMA7) mRNAs, and partially decreased nuclear factor-κB (NF-κB) p65 protein levels. Furthermore, it suppressed c-Jun NH2-terminal kinase 1/2 (JNK1/2), extracellular signal-regulated kinase 1/2 (ERK1/2), and p38 mitogen-activated protein kinase (p38) phosphorylation. Thus, we demonstrated that LTA regulates the innate immunity of ETEC-induced immune-stressed mice via NOD1/2-NF-κB and NOD1/2-MAPK pathways. © 2019

摘要:
花色苷是植物中一类具有重要生物活性的次生代谢产物,因其含量较低,使得分离制备花色苷高纯品难度较大.本研究以茶树紫芽为实验材料,以花色苷得率与纯度为考察指标,优化筛选了应用高速逆流色谱结合半制备型液相色谱分离纯化花色苷高纯度的方法.结果表明,茶样经提取、脱脂、阳离子交换树脂分离等前处理后,总花色苷得率28.18 mg/g,纯度为20.38%;以高速逆流色谱分离的最优溶剂体系乙酸乙酯:正丁醇:乙腈:0.1% 三氟乙酸水溶液=8:35:13:60(v/v/v/v),除去部分杂质后,总花色苷得率达到160.59 mg/g,纯度为43.64%,再结合半制备型液相色谱分离出5种花色苷组分,纯度最高达90.61%,其优化色谱条件为C18制备柱、波长280 nm、柱温30℃、流动相1%醋酸水溶液(A)-乙腈(B)、流速5 mL/min、进样量1 mL、梯度洗脱(0~55 min,10%~40%B).研究表明高速逆流色谱结合半制备型液相色谱可有效地分离纯化茶树紫芽花色苷及其组分.

摘要:
Soil microorganisms play important roles in the plant health and agricultural production. However, little is known about the complex responses of microbial communities and interaction networks to different agricultural management practices in tea plantation soils. In the present study, Illumina Miseq high-throughput sequencing technology and molecular ecological network (MEN) analysis were used to investigate the soil microbial diversity, community structure and composition, interaction networks of organic tea plantation (OTP), non-polluted tea plantation (NPTP) and conventional tea plantation (CTP). Alpha-diversity indices, Chao1 and richness, of OTP soil were significantly higher than those of NPTP and CTP soils. The beta-diversity analysis showed there were significant differences among bacterial community structures of OTP, NPTP and CTP soils. Composition analysis showed that Proteobacteria, Acidobacteria and Chloroflexi were the most dominant bacteria in all tea plantation soil samples under different management practices, and the beneficial community compositions of OTP soil were significantly different from NPTP and CTP soils at the phylum and genus levels. Canonical correspondence analysis (CCA) and mantel test revealed that TOC and NO3-N contents as well as pH values were the key soil factors to affect the bacterial community structures of tea plantation soils. Furthermore, network analysis showed that the network of OTP soil possessed more functionally interrelated microbial modules than NPTP and CTP soils, indicating that OTP soil possessed the higher ecosystem multi-functionality. These results provided the theoretical basis and reference for improving soil microbial diversity and enhancing community multi-functionality in tea plantation soil ecosystems through effective agricultural management practices. © 2019 by the authors.

摘要:
为茶园生物多样性保护与利用提供理论依据,采用实地调查法研究湖南省有机茶园、无公害茶园和普通茶园节肢动物群落的差异.结果 表明:有机茶园、无公害茶园和普通茶园生境的节肢动物群落具有一定的差异性;3类茶园皆以同翅目、鳞翅目、鞘翅目、膜翅目、蜱螨目和啮虫目为优势类群,黑刺粉虱、茶橙瘿螨、茶尺蠖和小绿叶蝉的数量分别占有机茶园、无公害茶园和普通茶园节肢动物总数的59.83%、56.17%和71.43%,天敌昆虫的数量分别占总数的10.76%、7.59%和5.79%.有机茶园的物种丰富度和多样性指数分别为102和1.56,显著高于无公害茶园(82和1.31)和普通茶园(79和1.23),害虫发生的数量明显低于其余两类茶园,主要天敌昆虫的发生频度明显高于其余两类茶园.

分别选取有机茶园(OTP)、无公害茶园(NPTP)和普通茶园(CTP)的茶树根际土壤,利用Illumina Miseq高通量测序技术研究其细菌群落多样性、结构和组成,并分析茶园土壤理化性质与细菌群落结构的相关性。结果表明:有机茶园土壤的细菌群落多样性指数(Chao1)显著高于无公害和普通茶园土壤;3种茶园土壤细菌群落结构存在显著性差异,变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)和绿弯菌门(Chloroflexi)在不同茶园土壤中都属于优势菌群,但有机茶园土壤的细菌群落组成在门和属水平上与无公害和普通茶园存在显著差异;土壤有效磷(AP)、总磷(TP)和pH值是影响茶园土壤细菌群落结构的关键理化因子。

随着时代的发展和对外交往的日益频繁,礼仪越来越受到人们的重视.当前我国大学生礼仪修养和礼仪教育还存在一些不容忽视的问题,比如在大学校园仍有许多不知礼、不守礼、不文明的行为,还有许多与大学生的礼仪修养、与精神文明建设极不和谐的现象.茶文化是中国传统文化的重要组成部分,传播茶文化可以提升当代大学生的礼仪修养,促进校园生活和谐文明进步.以湖南农业大学校礼仪队为例,就茶文化对大学生礼仪修养的推进作用进行了相关分析.

花青素是一类广泛存在于植物中的水溶性色素,作为一种强自由基清除剂,它具有抗炎,抗氧化,降血压,降血糖等多种保健功能。紫芽茶树作为一种高花青素含量的特异性茶树资源,其研究与利用越来越受人们的关注。为了解茶树紫芽中花青素形成的分子机理,本研究通过对湖南农业大学自选紫芽品种9803与绿芽品种9806进行转录组测序及生物信息学分析,筛选得到42条与花青素代谢相关的unigene,其中比对到参考基因组中的有34条,未在参考基因组中登录的有8条。KEGG富集分析表明这42个基因被富集到5个代谢通路中,包括类黄酮合成途径,木质素合成途径,黄酮和黄酮醇合成途径,油菜素甾醇合成途径,以及参与转录因子的编码。通过荧光定量PCR验证,差异基因荧光定量PCR变化趋势与转录组测序结果一致,转录组测序数据可靠。本次试验在转录组水平上筛选出了茶叶紫芽花青素代谢相关的差异基因,为进一步揭示茶叶紫芽产生的分子机理奠定了基础。

摘要:
L-Theanine (LTA; γ-glutamylethylamide), a peculiar non-protein-derived amino acid isolated from tea, is widely used as a functional ingredient and dietary supplement. L-Theanine has been confirmed to have hepatoprotective effects, but the underlying mechanism remains unknown. This study investigated the protective effect of L-Theanine-in vivo, using an enterotoxigenic Escherichia coli (ETEC)-infected mouse model. L-Theanine significantly decreased the elevated serum activities of both aspartate aminotransferase (AST) and alanine aminotransferase (ALT), two biomarkers of hepatic impairment. This was consistent with histopathological images from the microscopic observation of liver tissue. In addition, L-theanine significantly increased the mRNA and protein expression of Bcl-2 and decreased the expression of Bax, anti- and pro-apoptotic molecules, respectively, compared with levels in the ETEC control group. The expression of cleaved caspase-3 protein in the group pre-treated with L-theanine was significantly lower than that in the ETEC group. Additionally, decreases in extracellular signal-regulated kinase (ERK1/2) and c-Jun NH2-terminal kinase(JNK1/2) MAPK phosphorylation were observed in the L-theanine pre-treated group. Our study demonstrates that L-theanine possesses anti-apoptotic activity, which can be attributed to suppression of the intrinsic mitochondria-mediated apoptosis and MAPK phosphorylation signaling pathways. © 2017 Elsevier B.V.

Polyphenol oxidase (PPO) plays a key role in tea processing, converting catechins into theaflavines. Two PPOs were isolated and purified from tea (Camellia sinensis) leaves. PPO1 and PPO2 exhibited molecular weights of 85 KDa and 42 KDa, respectively. PPO activities were evaluated at different temperatures and pH values to determine thermal stability as well as the optimal temperature and pH. Moreover, PPO1 and PPO2 were allowed to react with catechol, epigallocatechin (EGC), epigallocatechin gallate (EGCG), L-tyrosine, guaiacol, and pyrogallic acid as enzyme substrates. PPO isozymes were more active with the triphenolic substrate, and PPO2 demonstrated no activity toward guaiacol. PPO1 activity only resulted in the synthesis of simple theaflavin (TF), while PPO2 was able to synthesize four types of theaflavins, particularly theaflavin-3-3'-gallate (TFDG). These data provide insights into the optimum manufacturing conditions for the enzymatic synthesis of theaflavins. © 2017 Elsevier Ltd

大黄鱼源气单胞菌的分离鉴定及致腐表型

摘要:
为鉴别大黄鱼腐败菌气单胞菌的致腐能力,采用生理生化和分子方法(16S rRNA)鉴定冷藏大黄鱼货架期终点不产H_2S的菌株。分析菌株在体外培养条件下的生长、胞外酶活性、三甲胺(TMA)和生物胺形成的腐败表型,将菌株接种在灭菌鱼汁中测定其致腐能力。结果显示,分离株AE03和AE04在0~37 °C及0~60的盐度下生长良好,在4和25 °C条件下AE03菌株的生长速率显著高于AE04菌株。2株细菌不产H_2S,能还原氧化三甲胺(TMAO),分解尿素和液化明胶,氧化酶、赖氨酸、鸟氨酸脱羧酶、精氨酸水解酶等都呈阳性,能利用葡萄糖。结合16S rRNA鉴定发现分离株AE03为杀鲑气单胞菌,AE04为软体动物气单胞菌。在体外培养中,2种气单胞菌均能产生蛋白酶、形成TMA和生物被膜,其中AE03菌株生成各指标的能力高于AE04菌株,同时AE03菌株还具有脂酶活性,产生生物胺,其中尸胺含量较高。气单胞菌AE03在灭菌鱼汁中产生的挥发性盐基氮(TVB-N)显著高于AE04。通过紫色杆菌CV026和根癌农杆菌A136 2种报告菌检测发现,2株气单胞菌是高丝氨酸内酯(AHLs)产生菌,其中AE03菌株产生的AHLs含量显著高于AE04菌株。研究表明,杀鲑气单胞菌AE03为冷藏大黄鱼的强致腐菌,且能分泌高含量的AHLs,研究为从群体感应角度阐明气单胞菌的致腐机理奠定了良好的基础。

L-Theanine (LTA), a non-protein-derived amino acid, is isolated from tea and is used as dietary supplement or beverage ingredient. This study established an enterotoxigenic Escherichia coli (ETEC)-infected mouse model of oxidative stress to examine the antioxidative mechanisms of LTA in vivo. LTA regulated non-enzymatic antioxidative activity and increased catalase (CAT) mRNA expression in non-ETEC-infected mice. In addition, LTA decreased oxidative parameters and the activity and mRNA levels of CAT in ETEC-infected mice. LTA increased superoxide dismutase (SOD) activity and mRNA expression of CuZn-SOD, ECSOD and glutathione peroxidase 1 (Gpx1); it restored MnSOD mRNA levels as well. Our results demonstrate that LTA has antioxidative properties via regulation of non-enzymatic pathways. In addition, LTA protects against superoxide and hydrogen peroxide via SOD or Gpx1 as the first and second defence line. Thus, LTA differentially regulates antioxidative enzymes depending on the dose and pathophysiological conditions through transcription, post-transcriptional and post-translational mechanisms in mice. © 2016 Published by Elsevier Ltd.

以适制红茶茶树品种政和大白茶春季一芽二叶新梢为原料,通过分析其红条茶加工中多酚氧化酶(PPO)、过氧化物酶(POD)酶活性及其比值,探讨儿茶素组分与茶黄素组分动态变化。结果表明,萎凋期间,PPO、POD酶活性在萎凋末期达到最大值,儿茶素总量小幅下降,茶黄素组分TF小幅上升,PPO/POD比值从0.78下降到0.33;揉捻阶段,儿茶素组分含量下降加快,ECG、GCG、EC下降幅度较大,茶黄素组分快速上升,且TFDG增幅最大,PPO/POD比值降至0.28;发酵期间,儿茶素组分含量下降较揉捻时慢,茶黄素组分及总量在发酵4 h时达到最大值,PPO/POD比值在发酵3 h时下降至最小值0.27,此后上升;干燥阶段,儿茶素组分含量略减,其中EGCG降幅最大,茶黄素组分下降,其总量降至干重的0.47%,PPO/POD比值上升至0.47。认为红条茶加工中儿茶素组分与茶黄素组分动态变化是PPO、POD酶活性及其比值驱动的结果,且可根据PPO/POD比值确定各工艺适度的标准。
