Heterogeneity of class I and class II MHC sequences in Schistosoma japonicum from different endemic regions in mainland China

摘要:
The present study examined sequence variation in class I and class II major histocompatibility complex (MHC) genes among Schistosoma japonicum isolates from different endemic regions in mainland China and assessed the level of horizontal gene transfer and sequence similarity between parasites and their hosts. S. japonicum cercariae were used to infect male adult rabbits to obtain adult S. japonicum samples. A portion of the class I MHC gene (pMHC I) and class II MHC genes (pMHC II) were amplified separately from individual adult trematodes by polymerase chain reaction and sequenced. Among all the examined isolates of S. japonicum, sequence differences between male and female parasites were 0.0-26.6% for pMHC I and 0.0-7.0% for pMHC II. Sequence variations between male and female parasites among different geographical locations from the mountainous areas were 1.1-26.6% for pMHC I and 1.5-3.0% for pMHC II. Sequence variations between samples from Yunnan and those from Sichuan were 2.7-23.5% for pMHC I and 1.1-3.7% for pMHC II. In the lake/marshland areas, sequence variations between male and female parasites among different geographical locations were 0.0-25.0% for pMHC I and 0.0-7.0% for pMHC II. Sequence variations between S. japonicum isolates from mountainous areas, and those from lake/marshland areas were 0.0-26.1% for pMHC I and 0.4-6.1% for pMHC II. BLASTN analysis indicated that all the pMHC II sequences showed high homology to a portion of exon 3 in rabbit MHC class II DP beta gene with more than 89% similarity, and all the pMHC I sequences except isolates in Yunnan (Eryuan) revealed high homology to the portion of exon 2 in rabbit MHC I gene with more than 81% similarity. Phylogenetic analysis showed no specific clustering comprising parasites from single geographical or endemic regions, and the paired parasites were even found in different clusters. These results demonstrated that pMHC I and II of S. japonicum isolates in mainland China existed heterogeneity, but the pMHC I, II, or combined sequences were not suitable markers for examining genetic relationship among different isolates from endemic regions in mainland China. © 2009 Springer-Verlag.

摘要:
The prevalence of helminths in the Asian water buffaloes (Bubalus bubalis) was investigated in Hunan Province, People's Republic of China between April 2005 and October 2007. A total of 359 adult buffaloes slaughtered at local abattoirs in 12 representative geographical locations in Hunan Province were examined for the presence of helminths. The worms were examined, counted and identified to species according to existing keys and descriptions. A total of 13 helminth species were found representing one phyla, two classes, eight families and nine genera. All buffaloes were infected by more than one helminth species. 61.8% of the examined buffaloes were infected with Haemonchus contortus, 44.7% with Fasciola hepatica, 24.9% with Fasciola hepatica, 23.5% with Homalogaster paloniae and 23.2% with Setaria labiatopapillosa, whereas the infection of adult buffaloes with cestodes was not detected in the present investigation. The results of the present investigation indicated that the prevalence of nematodes and trematodes in buffaloes is quite severe, some of which pose significant zoonotic public health problems (eg., schistosomiasis). It is imperative that integrated strategies and measures be taken to control helminth infections in buffaloes in Hunan Province and elsewhere. © 2008 Springer Science+Business Media B.V.

摘要:
以质粒pMDTLT为模板,用PCR的方法扩增出LTB基因,然后将其插入到pETVP1质粒中VP1基因的上游,构建了含有融合基因LTBVP1的表达质粒pETLTBVP1.转化宿主菌BL21(DE3)LysS后进行诱导表达,诱导菌经SDS-PAGE显示重组蛋白以包涵体的形式表达,分子量约为39 kD;Western blotting分析表明,重组蛋白能与FMDV阳性血清及兔抗霍乱毒素(CT)血清反应,说明融合蛋白保持了LTB和VP1各自的免疫学活性.小鼠免疫实验表明:该融合蛋白通过腹腔接种小鼠能诱导产生较强的免疫应答反应,免疫鼠产生的血清抗体水平高于试验中商品口蹄疫疫苗免疫组.

摘要:
Previous investigations have shown that asymmetric dimethylarginine (ADMA) inhibits nitric oxide (NO) synthases (NOS) and that ADMA is a risk factor for endothelial dysfunction. The objective of this study was to investigate the protective effect of kaempferol, a naturally occurring flavonoid antioxidant agent, against endothelial damage and the mechanisms involved. The experiments were performed in aorta and plasma from C57BL/6J control and apolipoprotein E-deficient (ApoE-/-) mice treated or not with kaempferol (50 or 100 mg/kg, intragastrically) for 4 weeks, and in human umbilical vein endothelial cells (HUVECs) pretreated or not with kaempferol (1, 3 or 10 μM) for 1 h and exposed to lysophosphatidylcholine (LPC) (10 μg/mL) for 24 h. Kaempferol treatment improved endothelium-dependent vasorelaxation, increased the maximal relaxation value, and decreased the half-maximum effective concentration concomitantly with an increase in nitric oxide plasma concentration, a decrease in ADMA and malondialdehyde (MDA) plasma concentrations, and increase in the expression of aortic endothelial NOS (eNOS) as well as dimethylarginine dimethylaminohydrolase II (DDAH II) in ApoE-/- mice. In addition, LPC caused a reduction in NO production, an increase in ADMA concentration concomitantly with a decreased expression of eNOS and DDAH II in HUVECs, and the effect of LPC was abolished by kaempferol. Treatment with kaempferol also significantly decreased reactive oxygen species production in mice aorta and in HUVECs. The present results suggest that the protective effect of kaempferol against endothelial damage may be associated with an improvement in NO production and a decrease in ADMA level. 摘要:
The first dog-associated outbreak of rabies in swine in China (Hunan province) has been diagnosed and the related virus isolated. Sequence analysis showed that the pig isolate was a genotype 1 rabies virus with a very high nucleotide identity to local dog isolates.

摘要:
采用PCR技术及DNA序列分析技术对采自湖南省4个不同地区的日本血吸虫的线粒体NADH脱氢酶基因亚基Ⅳ(nad4)部分片段(pnad4)进行克隆及序列分析.获得480bp的pnad4序列。与GenBank上的日本血吸虫相应基因序列进行比对.发现pnad4序列有一定的种内差异(0.4%~1.3%)。

Previous investigations have shown that decreased expression of angiopoietin-like protein 3 (Angptl3) is protective against dyslipidemia in atherosclerosis. The present study was conducted to test the effect of 3,4,5,6-tetrahydroxyxanthone, a xanthone compound, on dyslipidemia in apolipoprotein E-deficient (ApoE-/-) mice. Forty mice were randomly divided into 4 groups (n = 10): control group (C57BL/6J mice), ApoE -/- mice group, and two groups of ApoE-/- mice treated with 3,4,5,6-tetrahydroxyxanthone (10 or 30 mg/kg per day). Eight weeks after treatment, lipid levels in the blood and liver, expression of hepatic Angptl3, and adipose tissue lipoprotein lipase (LPL) were determined. Treatment with 3,4,5,6-tetrahydroxyxanthone (10 or 30 mg/kg) significantly decreased plasma and hepatic total cholesterol and triglyceride concentrations, increased plasma high-density lipoprotein cholesterol, and significantly downregulated expression of Angptl3 mRNA and protein concomitantly with upregulated expression of LPL mRNA. In addition, T0901317 (a liver X receptor ligand) caused elevated expression of hepatic Angptl3 mRNA and protein, and the effect of T0901317 was also abrogated by 3,4,5,6-tetrahydroxyxanthone in vivo and in vitro. The present results suggest that the beneficial effect of 3,4,5,6-tetrahydroxyxanthone on dyslipidemia may be related to reduced expression of Angptl3. © 2008 NRC.

Improvement of animal embryo sexing depends upon high-titer serologically detected male (SDM) antibody fragments. SDM sera collected from isogenic C57BL/7 female mice after inoculation with male spleen cells were characterized and used for construction of a recombinant Fab antibody library against SDM antigen, and used for analysis of the binding capacity and specificity to SDM antigen. The heavy-chain Fd and full-length light-chain K were amplified by RT-PCR from a mouse (#6) that'ed high-titer antiserum. The amplified product was inserted into the pComb3 vector followed by co-infections with the help phage VCSM 13 for construction of the phage library, which gave 1.5 x 10(7) colonies with the titer of 3.2 x 10(11) pfu/ml by a recombination rate of 80%. Sequence analysis of the PCR products of plasmid DNA of E5 clones showed that V-H and V-k had common characteristics shared by other known variable region of antibodies. The Fab antibody libraries against SDM antigen were enriched by three cycles of affinity enrichment with male spleen cells, and two cycles of non-specific absorption with female spleen cells. The ELISA results showed that 9 of 15 clones had binding capacity to the SDM antigen. This is the first report on a phage display library of SDM antigen. The mouse Fab antibody library could be used for identifying SDM antigen, and for the development of sex determination of early embryos in mammals. (C) 2007 Elsevier B.V. All rights reserved.

摘要:
为建立圆弧青霉毒素-青霉酸的免疫学检测方法,研究了青霉酸(PA)的人工抗原合成。通过碳二亚胺法将青霉酸(PA)分别与牛血清白蛋白(BSA)和卵清蛋白(OVA)联结,得到青霉酸人工抗原PA-BSA和PA-OVA。采用紫外扫描光谱法、SDS-PAGE和动物免疫试验对合成的抗原进行鉴定。结果显示联结后的人工抗原特征性吸收峰出现偏移,PA与BSA的偶联比为23.2:1,PA与OVA的偶联比为10.4:1。以PA-BSA为免疫抗原免疫小鼠,PA-OVA为包被抗原,采用间接ELISA检测抗血清,其效价达到1:12800。表明青霉酸的人工抗原已合成,为建立有效的免疫检测方法提供了基础。

一株H3N2亚型猪流感病毒广东分离株的基因组序列分析

从广东省疑似流感发病猪分离到1株H3N2亚型猪流感病毒(A/Swine/Guangdong/01/2005(H3N2)),对其各个基因进行克隆与测序,并与GenBank中收录的其它猪流感、禽流感和人流感的相关基因进行比较,结果表明,HA全基因与广东2003~2004 年分离的H3N2猪流感毒株的核苷酸序列同源性在99%以上,与纽约90年代末分离的H3N2人流感毒株同源性在98.5%以上;NA基因与纽约1998~2000 年分离的H3N2人流感毒株的核苷酸序列同源性在99%以上;NS基因、M基因的核苷酸序列与H1N1亚型猪流感毒株A/swine/HongKong/273/1994(H1N1)的核苷酸序列同源性较高,分别为97.9%、98.4%,与美洲A/swine/Iowa/17672/1988(H1N1)的核苷酸序列同源性分别为96.7%、97.1%;其他基因的核苷酸序列与H3N2人流感毒株具有很高的同源性.因此,推测其M和NS基因来源于H1N1亚型猪流感病毒,HA、NA及其他基因均来源于H3N2亚型人流感病毒.表明此H3N2亚型猪流感病毒为H3N2亚型人流感病毒和H1N1亚型猪流感病毒经基因重排而得到的重组病毒.

测定了1996~2005年间在中国分离并保存的395株H3N2亚型人流感病毒神经氨酸酶(NA)基因序列,应用生物信息学工具进行了分析.结果表明:NA基因序列的进化树表现为一主要进化主干伴随多侧分支进化,同一年份的毒株可以分为几个分支存在;疫苗株在NA基因序列进化树上存在明显的滞后现象;NA没有氨基酸的丢失与插入;抗原决定簇大部分位点保守且各抗原决定簇之间的变异情况各有特点,其中197~199位、431~434位和339~347位点的变异频率最高,而153位、328~336位、367~370位、400~403位抗原决定簇的氨基酸变异频率相对比较小;除了抗原决定簇外还有些氨基酸位点的变异频率很高,它们分别是18、23、30、93、143、208、216、221、249、265、267、307、385、437位氨基酸,其中143位和267位这两个位点的变异频率高于抗原决定簇位点,具体生物学意义还需要进一步研究;NA蛋白的酶活性中心位点高度保守;二硫键和糖基化位点保守.NA基因的这些特点为流感的预防、控制以及NA抑制剂药物的应用提供了一定的参考依据.

利用噬菌体抗体库筛选技术获得抗雄性特异性抗原的噬菌体Fab抗体,首次采用雄鼠脾细胞对鼠源性抗雄性特异性抗原噬菌体Fab抗体库进行3轮亲和富集和2轮雌鼠脾细胞吸附,对筛选后特异性噬菌体Fab抗体进行ELISA分析,重组率鉴定及基因测序分析.结果显示,5次筛选后的15个菌落中有9个能产生抗雄性特异性抗原特异性噬菌体抗体,噬菌体Fab抗体的基因重组率为60%,E5克隆的重链、轻链可变区序列分别属于V_H1和VκⅣ基因家族,这为挑选出高亲和力的抗雄性特异性抗原噬菌体Fab抗体奠定了实验基础,将推进雄性特异性抗原及其抗体的研究进程,并为性别控制研究开创新途径.

日本血吸虫SjIR3 DNA疫苗诱导的抗攻击感染保护力

摘要:
目的 探讨日本血吸虫SjIR3核酸疫苗诱导小鼠抗日本血吸虫攻击感染的保护效果.方法 用SjIR3的特异引物构建疫苗SjIR3/pC.54只昆明小鼠随机分为3组:A组(对照组)、B组(空质粒组)和C组(实验组)分别肌注生理盐水(100 μl)、pcDNA3.0和SjIR3/pC(各50 μg),共免疫3次,每次间隔2周.于每次免疫前和感染前尾静脉采血,ELISA检测血清IgG抗体水平.末次免疫后2周,各组剖杀其中6只小鼠取脾细胞,分别用ConA或rSjIR3重组蛋白诱导,用四甲基偶氮唑盐微量酶反应比色法(MTT)检测脾淋巴细胞增殖情况.各组余下小鼠分别经腹部感染日本血吸虫尾蚴40±1条,45 d后宰杀,观察实验组小鼠的减虫率和肝减卵率.结果 ELISA结果显示,A组与B组IgG抗体水平变化不明显(P>0.05),C组于末次免疫后2周(攻击感染前)IgG抗体水平最高为0.78±0.05,且与A、B两组比较差异有统计学意义(P<0.01);末次免疫后,实验组小鼠脾淋巴细胞分别在ConA或rSjIR3重组蛋白诱导下可增殖,分别为0.57±0.02、0.68±0.01,与A、B组相比差异有统计学意义(P<0.01).经SjIR3/pC免疫的实验组小鼠攻击感染后可产生29.4%的减虫率和36.6%的肝减卵率.结论 SjIR3/pC核酸疫苗具有较好的免疫原性,可诱导小鼠产生部分免疫保护力.
