Although PCV2 infections generally cause mild disease in pigs, concurrent co-infections with other pathogens can damage the immune system and cause more severe diseases, collectively termed porcine circovirus associated diseases (PCVAD). Involvement of porcine parvovirus (PPV, a common cause of reproductive failure in naïve dams) in PCVAD caused by PCV2, has been reported. As this co-infection can be difficult to eliminate, there is a critical need to develop an effective vaccine to protect against PPV or synergistic effects of PCV2 and PPV under field conditions. In this study, we designed chimeric PCV2 virus-like particles (cVLPs) displaying a B-cell epitope derived from PPV1 structural protein around the surface of the 2-fold axes of PCV2 VLPs, based on 3D-structure analysis of the PCV2 capsid. The cVLPs were successfully prepared, verified by transmission electron microscopy and chromatography, with robust antibody titers against PCV2 and PPV1 produced in mice and guinea pigs. In addition, in guinea pigs challenged with 106 TCID50 PCV2, cVLPs conferred more effective immune protection (based on viral load) than a commercial PCV2 vaccine. Finally, antibody responses and immune protection against PPV were also evaluated. In guinea pigs vaccinated with cVLPs, although PPV antibodies detected by a hemagglutination inhibition (HI) assay appeared later after vaccination in the PCV2 cVLPs group than in the commercial PPV vaccine group, there were fewer PPV genomic DNA copies in the PCV2 cVLPs group than in a PBS group. In conclusion, guinea pigs vaccinated with cVLPs developed effective protective immunity against PCV2 challenge, with some protective immunity against PPV. This study provided valuable research data to pursue molecular design of chimeric epitopes PCV2 VLPs. © 2019

摘要:
Background: Porcine circovirus 2 (PCV2) is the causative agent of porcine circovirus-associated diseases (PCVADs). The infection of PCV2 is widespread and has serious consequence, thereby causing significant economic losses in the swine industry worldwide. Previously, we found that a strain named YiY-3-2-3 has a naturally occurring point mutation (G710 to A710) in ORF1 region, which leads to a shorten product of the rep gene (945 to 660 base pair). Importantly, the Rep protein is responsible for genome replication of PCV2. To explore the effects of this mutation on the PCV2 replication, in the current study we constructed infectious clone of this IF-YiY-3-2-3, as well as those of its two parental strains of IF-YiY-3-2-1 and IF-YiY-3-2-10. Subsequently, these infectious clones which have 1.1 copy of PCV2 genome of their corresponding strains were transfected into PK15 cells to obtain rescued viruses, respectively. Results: Though all of the three infectious clones could be rescued, the copy number and infectivity of these rescued viruses were significantly different, as analyzed by fluorescence quantitative PCR, Tissue culture infectious dose 50 (TCID50), and indirect immunofluorescence assay (IFA). Notably, whether the PCV2 copy number, viral titer or the infectivity of rescued viruses from infectious clone IF-YiY-3-2-3 was significantly less than those of its parental clones. Meanwhile, the spatial structure of the Rep protein from the IF-YiY-3-2-3 displayed an apparent truncation at the C-terminal. Conclusions: These findings therefore suggest that the Rep protein with truncated C-terminal would reduce virus replication and infectivity, and there might also exist both favorable and unfavorable mutations in the ORF1 of PCV2 in the process of its evolution. © 2019 The Author(s).

干扰素诱导跨膜蛋白(interferon-inducing transmembrane proteins,IFITM)是一类宿主限制性因子(host restric-tive factor,HRF),在抗病毒感染过程中发挥重要作用.IFITM作为一类氨基酸高度保守的抗病毒蛋白,在机体内广泛分布.随着对其研究的深入,发现该IFITM以多种通路/机制产生抗病毒感染的作用,使其作为研发抗病毒治疗药物的作用靶点成为可能.现就IFITM拓扑结构,抗病毒机制和抗病毒谱的研究作一概述.

摘要:
T-2 and HT-2 toxins can cause cytotoxicity and oxidative stress in animals, while DL-Selenomethionine plays an important role in preventing oxidative stress and improving cell viability. However, the role of DL-Selenomethionine in T-2/HT-2 toxins-induced cell damage is still unknown. In this study, we investigated whether DL-Selenomethionine plays a protective role against T-2/HT-2-induced cytotoxicity and oxidative stress in primary hepatocytes. Our results demonstrated that T-2/HT-2 toxins-exposed broiler hepatocytes exhibited significantly decreased cell viability and intracellular glutathione (GSH) concentration while increased Lacate dehydrogenase (LDH) leakage, intracellular reactive oxygen species (ROS), glutathione peroxidase (GSH-PX), malondialdehyde (MDA) and catalase (CAT) levels, as well as elevated expression levels of genes related to oxidative stress, in a toxin dose-dependent manner (P < 0.05). However, the application of DL-Selenomethionine into T-2/HT-2 treated hepatocytes effectively alleviated the adverse effects of T-2/HT-2, as demonstrated by increased cell viability, decreased LDH leakage, declined intracellular ROS and MDA levels, increased expression of oxidative stress-related genes, as well as accordingly enhanced activities of GSH, GSH-PX, SOD and CAT as compared to the control groups (P < 0.05). Therefore, our in vitro data demonstrate that DL-Selenomethionine can function as an effectively protective agent against T-2/HT-2-induced cytotoxicity and oxidative stress.

摘要:
Porcine circovirus type 3 (PCV3) has recently been isolated from diseased pigs within the USA. The objective was to detect the presence of PCV3 in dogs. Nested polymerase chain reactions (PCR) with PCV3-specific primers for the capsid gene were used to detect PCV3 genomic DNA in serum samples from dogs (n = 44) in China. There was PCV3 DNA detected in 4 of 44 dogs [all were negative for PCV2 and canine circovirus (CanineCV)]. Based on sequence analysis, positive sequences were grouped into PCV3 genotypes. However, these isolates had close evolutionary relationships with FoxCV (KP941114) and CanineCV (JQ821392). Further investigations of the epidemiology, evolutionary biology, and pathobiology of PCV3 to dogs are warranted.

摘要:
为研究褪黑激素对皖系长毛兔毛囊细胞凋亡的影响及相关机制,选择60只皖系长毛兔,随机均分成4组,1组不埋植褪黑激素(对照),另3组每只兔按25 mg(低剂量组)、40 mg(中剂量组)和55 mg(高剂量组)分别埋植褪黑激素,饲养70 d后,进行组织病理学观察和RNA的转录组高通量测序及qPCR表达分析。结果表明:通过组织切片观察,证实55 mg褪黑激素处理能有效抑制毛囊细胞凋亡,维持细胞正常形态;通过对高剂量组和对照组测序,得到25个差异表达基因,其中有14个基因上调,11个基因下调,涉及通路包括嗜T细胞病毒感染、坏死性凋亡、甲状腺癌、铁死亡、I型糖尿病和胰岛素信号通路,其中坏死性凋亡、铁死亡通路与长毛兔毛囊凋亡相关;选取4个与凋亡相关的差异表达基因进行荧光定量验证,得到新基因MSTRG.3561表达显著上调,SLC25A5表达下调,但差异无统计学意义。

短毛兔兔毛生长规律研究

以成年短毛兔为试验对象,通过拔毛构建新的兔毛生长周期,从第4-11周,每周采集一次背部毛样,用直尺测定不同时期粗毛、细毛和两型毛的长度,通过生长曲线模型对兔毛生长情况进行了模拟,并对短毛兔兔毛的生长规律进行了研究。结果表明:试验兔第4、5、6周,3种毛型的长度都依周次显著增长(P〈0.05),而第6-11周,除细毛第7周和第11周、粗毛第6周和第9周毛长差异显著外(P〈0.05),兔毛长度变化均不显著(P〉0.05),这说明短毛兔兔毛在第6周后基本停止生长。生长曲线拟合结果表明,Logistic模型最为符合短毛兔兔毛生长的实际情况,粗毛、细毛和两型毛的拐点周龄分别为3.69、3.82和4.07周,拐点毛长分别为17.58、13.40和16.21 cm,说明短毛兔兔毛生长速度在第4周左右开始变慢。研究结果可为兔毛生长规律和毛囊发育及凋亡规律的研究提供参考。

Porcine circovirus type 2 (PCV2) has been known as a causative agent of reproductive failure in the sow. In the present study mouse model was used to investigate PCV2 infection. In order to investigate whether PCV2 can induce lesions of spermatocytes and oocytes, 6 male and 6 female mice were each inoculated intraperitoneally with PCV2b, and 3 male and 3 female mice mock-infected with cell culture supernatant served as controls. Samples of testes and ovaries from PCV2b-inoculated and mock-infected mice were investigated using PCR, histopathological, ultrastructural and immunofluorescent histochemical methods at 14 and 21 day post infection (dpi). The study revealed that in the virus-inoculated mice leydig cells in testes and granulosa cells in ovaries were degenerated, and a small number of spermatocytes and oocytes showed apoptosis. Positive PCV2b antigen signals were also observed in these apoptotic cells. It can be suggested that PCV2 can cause lesions of spermatocyte and oocyte prior to zygotes formation in its host.

念珠菌属是人、动物和禽类正常消化系统的常住微生物的组成部分,呈全球性分布,念珠菌病是一种内源性的条件性真菌病,当菌群失调或宿主的抵抗力较弱时,就会发生该病,长期或不合理使用抗生素时,破坏机体的微生物生态平衡,可能是导致念珠菌病发生的常见原因.文章就怀化市中方县一个山地放养鸡念珠菌病病例,对该病的临床症状、病理变化、诊断治疗等方面进行了阐述,为山地养鸡在念珠菌病的有效防治上提供参考.

用PCV2感染5周龄昆明系小鼠,分别在感染后7,14,21,28,35,42 d处死小鼠,取脾脏、腹股沟淋巴结、Peyers结做PCR鉴定PCV2感染情况,提取肠道中蛋白检测SIgA及肠道相关细胞因子IL-2、IL-4、IL-10和IFN-γ含量的变化。结果显示:随着感染时间的延长,试验组小鼠肠道SIgA、IL-2和IFN-γ水平均呈先升高后降低的趋势,IL-4和IL-10呈持续升高的趋势。结果表明:小鼠肠道SIgA及相关细胞因子的含量在PCV2的刺激下发生了不同程度的改变。

摘要:
利用一种常规的细胞凋亡检测方法 TUNEL法检测感染PCV2(猪圆环病毒2型)的仔猪淋巴结淋巴细胞的凋亡变化情况,并从组织病理学的角度验证PCV2是否能引发仔猪淋巴结淋巴细胞凋亡。通过HE染色镜下可见仔猪皮质区淋巴结皮质部淋巴小结内坏死的淋巴细胞染色变深,胞核固缩,出现坏死,导致淋巴细胞稀疏,出现细小的细胞缺失区,有的还出现严重的淋巴细胞浸润或弥散性出血,严重者成团的淋巴细胞细胞质溶解以及核碎裂出现溶解坏死,多数淋巴细胞缺失区相连形成空洞,生发中心消失;通过TUNEL法检测可见淋巴小结内出现多量的细胞胞核固缩、凋亡,常堆集在一起呈多量散在分布,细胞凋亡后,在淋巴小结中可以明显地看到细胞凋亡形成的碎片,造成细胞密度疏松,严重者可见大量的细胞碎片,消失形成空洞。阴性对照组仔猪淋巴组织结构比例较正常,细胞形态清晰。通过HE染色和TUNEL法对比观察发现细胞的坏死和凋亡有重叠现象,从病理学的角度非常直观地证明了PCV2能够引起仔猪免疫系统淋巴细胞的凋亡,为阐明PCV2诱导猪的淋巴细胞发生凋亡这一现象奠定基础,并为探讨PCV2导致猪PMWS的致病机制提供基础资料。

MicroRNA(miRNA)是一类广泛存在于人、动植物和病毒的内源性微小RNA,且在不同组织、不同阶段存在特异性表达,能对细胞凋亡、增殖及分化、神经系统发育、疾病发生、免疫调节等众多生命过程起到重要调控作用。文章综述了miRNA对家禽生长与生殖、疾病调控和脂质代谢等方面的研究进展,为进一步研究miRNA在家禽中功能提供理论依据和事实参考。

摘要:
猪繁殖与呼吸综合征(porcine reproductive and respiratory syndrome,PRRS),又称蓝耳病,主要会导致母猪早产、流产、产死胎或木乃伊胎,仔猪和育成猪出现呼吸道疾病,且死亡率高.该病最初于20世纪80年代在美国被首先报道,2006年起在我国大面积爆发.PRRS是由猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)引起的,PRRSV是呈单股正链的RNA病毒,属于动脉炎病毒科,同属该科的还有猴出血热病毒(simian hemorrhagic fever virus,SHFV)、马动脉炎病毒(equine arteritis virus,EAV)、乳酸脱氢酶增高症病毒(lactate-dehydrogenase elevating virus,LDV)和新发现的不稳定负鼠病毒(wobbly possum disease virus,WPDV)[1].根据抗原性和序列的不同,将全球的PRRSV分为以VR-2332为代表的美洲型(North American,NA)和以Lelystad virus (LV)为代表的欧洲型(European,EU).我国流行的PRRSV主要是经典美洲株和变异型美洲株,但近几年欧洲株也屡有发现.此外,PRRSV变异性和重组能力强,不时有新分支出现.

摘要:
我国是猪肉生产消费大国,猪肉的产量及消费量稳居世界第一位。近年来生猪产业稳中有进,养猪业生产形势较好,效益明显,在给养猪业带来前所未有的经济效益的同时,也给猪场提出了更严峻的挑战,疫病防治需要更严格的要求。目前,由于科学养殖观念的加强、免疫政策的强化,规模养殖中大范围暴发单独性疾病的可能性已不大,猪群中发生的疫病多由病毒、细菌、寄生虫等感染或多病原混合感染所引起,在此基础上,加之病原的变异、病原毒力的增强、机体免疫抑制的产生、疫苗和抗生素的大范围使用等原因,致使猪病的疫情变得越发严重,病情也随之变得更加复杂。因此,在临床上所表现的流行以混合感染、免疫抑制、病原变异等新变化特征。如何防控好猪病,提高经济效益,是饲养管理的重点和难点。

摘要:
Porcine circovirus type 2 (PCV2) is the causative pathogen of porcine circovirus-associated diseases (PCVAD). This virus evolves mostly through point mutations and genome recombination between different PCV2 genotypes (e.g. PCV2a and PCV2b), as has been confirmed in swine herds. In the current work, the complete PCV2 genome sequences of 69 clones derived from various tissues (lymph node, spleen and lung,) of an infected individual, were subjected to phylogenetic and alignment analyses. The results not only demonstrate co-infection with distinct PCV2b subtypes (e.g. 1B and 1C) in the same animal, but also highlight another mechanism of evolution - diverse point mutations acquired during immune evasion by this virus.

摘要:
为构建猪圆环病毒Ⅱ型(PCV2)的感染性克隆及其ORF3缺失的感染性克隆,本研究通过比较分析GenBank中8种亚型的PCV2基因组序列(各3株),设计了PCV2基因全长克隆引物,以从江西省萍乡市某猪场疑似感染断奶仔猪多系统衰竭综合征病死猪的病料中提取的DNA为模板,利用PCR方法扩增PCV2全长基因序列并进行测序。利用分子生物学软件对测序结果分析,结果表明所获得的克隆为PCV2a-2A基因型。采用重叠延伸PCR(SOE-PCR)方法扩增病毒全长基因组DNA,克隆于pSP72载体中构建野生型PCV2a病毒株感染性克隆。此外,采用SOE-PCR方法,缺失野生型PCV2a感染性克隆中的阅读框3 (ORF3),并将这两种重组质粒分别转染PK15细胞,盲传6代后经PCR和间接免疫荧光方法检测显示,构建的PCV2a病毒株与PCV2a-ORF3突变株克隆均具有感染性。本研究为进一步探究ORF3基因对PCV2致病机理和免疫原性的影响奠定了基础。

摘要:
Porcine circovirus type 2-(PCV2-) associated reproductive disorders and enteritis have commonly been observed on PCV2-contaminated pig farms in recent years. In order to investigate disorders of intestinal immunity in piglets infected by PCV2 during the fetal period, 9 PCV2b-infected piglets and 6 noninfected piglets at one day of age were selected and euthanised prior to suckling. Samples of mesenteric lymph nodes (MLNs) and duodena were collected to investigate factors related to intestinal immunity and to detect lymphocytic apoptosis. The results indicated that there were no significant changes in the levels of IL-2, IL-10 and transforming growth factor-β (TGF-β) in the PCV2b-infected piglets but IFN-γ levels were significantly lower (P < 0.01) and IL-4 levels were significantly higher (P < 0.05) in infected piglets than in the controls. Furthermore, lymphocytic apoptosis increased in PCV2b-infected piglets and CD4+ to CD8+ ratios were lower in these piglets than in the controls. These findings suggest vertical transmission of PCV2b to fetuses, leading to an imbalance of intestinal immune function in piglets. © 2017 Akadémiai Kiadó, Budapest.

Porcine circovirus type 2 (PCV2) is the pivotal pathogen causing porcine circovirus-associated diseases. In this study, 62 PCV2 isolates were identified from seven farms in southern China from 2013 to 2015 and phylogenetic trees were reconstructed based on whole-genome sequences or the cap gene. In this investigation, PCV2b was the main genotype in circulation throughout these farms. Furthermore, an emerging mutant (PCV2b-1C), isolated from PCV2-vaccinated farms, was the predominant strain prevalent on these farms. In addition, we isolated a new cluster that may represent evolution of the virus through recombination of PCV2b-1A/1B and PCV2b-1C. Finally, we discuss evidence that antigenicity and surface structure variation of the capsid resulted from mutation of the C-terminal loop (Loop CT) of the PCV2b-1C Cap in silico. © 2016 The Authors.
