The plant cell mechanics, including turgor pressure and wall mechanical properties, not only determine the growth of plant cells, but also reflect the functional and structural changes of plant cells under biotic and abiotic stresses. However, there are currently no appropriate techniques allowing to monitor the complex mechanical properties of living plant cells non-invasively and continuously. In this work, quartz crystal microbalance with dissipation (QCM-D) monitoring technique with overtones (3–9) was used for the dynamic monitoring of adhesions of living tobacco BY-2 cells onto positively charged N,N-dimethyl-N-propenyl-2-propen-1-aminiumchloride homopolymer (PDADMAC)/SiO2 QCM crystals under different concentrations of mannitol (CM) and the subsequent effects of osmotic stresses. The cell viscoelastic index (CVIn) (CVIn = ΔD·n/ΔF) was used to characterize the viscoelastic properties of BY-2 cells under different osmotic conditions. Our results indicated that lower overtones of QCM could detect both the cell wall and cytoskeleton structures allowing the detection of plasmolysis phenomena; whereas higher overtones could only detect the cell wall’s mechanical properties. The QCM results were further discussed with the morphological changes of the BY-2 cells by an optical microscopy. The dynamic changes of cell’s generated forces or cellular structures of plant cells caused by external stimuli (or stresses) can be traced by non-destructive and dynamic monitoring of cells’ viscoelasticity, which provides a new way for the characterization and study of plant cells. QCM-D could map viscoelastic properties of different cellular structures in living cells and could be used as a new tool to test the mechanical properties of plant cells. © 2021 by the authors. Miscanthus, a perennial rhizomatous C4 grass, has emerged as a promising lignocellulosic crop. To avoid competition with food crops, marginal lands especially saline–alkaline soil are being recommended to grow Miscanthus across China. However, the challenges such as high cost and low reproduction coefficient associated with the propagation methods are impeding its large-scale plantation in saline–alkaline soil. In this study, a “two-steps” propagation method was designed, which was comprised of two phases: (1) seedling cultivation and (2) seedling strengthening. The parameters (substrate composition and plug size) for this method were determined. The optimal substrate composition of the seedling bed for M. sinensis, M. lutarioriparius, and their interspecific hybrids was peat and perlite at a ratio of 3:2, 3:1, and 3:3 by volume, respectively. The seedlings were transplanted into the plugs to deliver robust plantlets. The most appropriate plug size for M. sinensis, M. lutarioriparius, and their interspecific hybrids was 50 plugs (50cm3/plug), 72 plugs (40cm3/plug), and 72 plugs (40cm3/plug), respectively. Three propagation methods, that is, direct sowing of seeds, rhizomes plantation, and “two-steps” seed-derived plugs for M. lutarioriparius were compared in saline–alkaline soil over 3years (2014, 2015, and 2016). The establishment rate of the seeds directly planted was poor (zero). The performance (e.g., biomass yield) of the plants derived from the “two-steps” seed-derived plugs was better than that of the plants derived from rhizomes directly planted. Therefore, the “two-steps” seed-derived plugs can be regarded as an effective propagation method to upscale Miscanthus cultivation in saline–alkaline soil. © 2021 The Authors. A simple, highly sensitive biosensor for S. aureus detection is becoming increasingly important in human health and safety. In this work, a hairpin probe-mediated DNA circuit for the detection of the mecA gene of S. aureus was reported cascading Exo III-assisted cycling signal amplification and the DNAzyme-mediated cleavage reaction. In the presence of the target mecA gene, the recognition and hybridization between HP1 and mecA can trigger Exo III and DNAzyme-mediated signal amplification and further release numerous ATMND, resulting in an enhanced fluorescence response, which serves as a response signal for the fluorescence detection of mecA gene. This biosensor enables the sensitive and specific detection of the mecA gene, showing a linear response ranging from 1 fM to 1 nM with a detection limit of 0.5 fM. Moreover, this fluorescence assay has been applied for the analysis of clinical samples with satisfactory recovery. Importantly, this universal platform can be further extended for the analysis of other targets by alternating the corresponding recognition unit, which holds much promise in point-of-care testing for bacterial analysis.

摘要:
Phytoremediation using Miscanthus is a valuable technique for the restoration of saline-alkaline soil, but the potential is not quantified. To assess this potential, a 5years field experiment was conducted on saline-alkaline soils with three genotypes of Miscanthus and an unplanted control. The bulk soil (0–20-cm depth) physicochemical properties were measured annually from 2015 to 2018, while the bacterial and fungal community structures of the rhizosphere soil were investigated by high-throughput sequencing after 5years under Miscanthus (2018). The results showed that Miscanthus cultivation on saline-alkaline soil reduced soil salinization and improved soil fertility on this site. Specifically, compared with the blank control, the five-years Miscanthus cultivation (mean of three different genotypes pooled) decreased the soil pH by 0.33 units (8.78–8.45) and electrical conductivity by 23.4μS/cm (226.7–203.3μS/cm), as well as increasing soil organic matter content from 18.9 to 34.9g/kg and total potassium content from 15.2 to 18.0g/kg. In addition, the soil improvement was more pronounced with a longer duration of growth. Miscanthus remediates saline-alkaline soils by altering the microbial community compositional abundance rather than its diversity. The improvement is due to enrichment of beneficial bacteria leading to soil desalinization (e.g., by Gemmatimonadaceae), litter decomposition (e.g., by Chitinophagaceae), plant growth promotion (e.g., by Micrococcaceae), and the reduction of harmful fungi (e.g., Pleosporales and Chitinophagaceae). Soil pH, electrical conductivity, organic matter, and total potassium were the main physicochemical properties affecting bacterial and fungal communities. Our results quantify the soil improvement potential of Miscanthus and clarify the underlying microbial mechanisms, laying a solid foundation for further implementation of saline-alkaline soil phytoremediation using Miscanthus. In the future, more extensive multi-location trials are required to assess if these improvements are seen across a wide diversity of environments. © 2021 The Authors. The combustion quality of three perennial wild plant species Tanacetum vulgare L., Centaurea nigra L. and Artemisia vulgaris L. was investigated in comparison to the energy yield obtained from anaerobic digestions of these biomasses. Combustion resulted in 1.5–2.8 times higher energy yield compared to anaerobic digestion. All wild plants showed a similar higher heating value to Miscanthus × giganteus Greef et Deuter and Panicum virgatum L. (16.0–17.0 MJ kg−1). The ash-melting behavior of all wild plants was like Sida hermaphrodita L. Rusby, since the ash did not sinter at 1200 °C. However, Artemisia vulgaris L. had highest ash content (5.2–5.7% of dry matter) with a low ash melting behavior (1000 °C) attributed to a high potassium content and calculated phase composition. Therefore, careful consideration should be given to select the wild plants to meet the requirements for their use as solid biofuels in residential and commercial applications. © 2021 Elsevier Ltd

Fatty acids (FAs) in oilseeds are important for human health. Gas chromatography-mass spectrometry (GC-MS) is a popular analytical technology for FAs measurement. FAs need to be derivatized into fatty acid methyl esters (FAMEs), which is essential for GC-MS analysis. The previous methods for methylation of FAs usually include reaction at high temperature and need specific vessels, for which samples with a large scale cannot be analyzed in a short time, leading to low efficiency. Here we report a high-throughput method for profiling fatty acids in plant seeds based on one-step acid-catalyzed methylation followed by GC-MS. This method for FA methylation via derivatization by sulfuric acid with methanol (5%, v/v) and incubation at 64 °C for 4 h in a more convenient 2 mL tube, is efficient. Satisfactory methodological performance was achieved by optimization of the parameters. Only 5 mg seeds were needed as a microscale sample usage by this method. The method is a successful approach to increase the efficiency of sample preparation and analysis for large-scale experiments. © 2021 The Author(s). Biofuel production from crop residues is widely recognized as an essential component of developing a bioeconomy, but the removal of crop residues still raises many questions about the sustainability of the cropping system. Therefore, this study reviews the sustainability effects of crop residues removal for biofuel production in terms of crop production, soil health and greenhouse gas emissions. Most studies found little evidence that residue management had long-term impacts on grain yield unless the available water is limited. In years when water was not limiting, corn and wheat removal rates ≥90% produced similar or greater grain yield than no removal in most studies. Conversely, when water was limiting, corn grain yield decreased up to 21% with stover removal ≥90% in some studies. Changes in soil organic fractions and nutrients depended largely on the amount of residue returned, soil depth and texture, slope and tillage. Reductions in organic fractions occurred primarily with complete stover removal, in the top 15–30cm in fine-textured soils. Soil erosion, water runoff and leaching of nutrients such as total nitrogen (N) and extractable soil potassium decreased when no more than 30% of crop residues were removed. Stover management effects on soil bulk density varied considerably depending on soil layer, and residue and tillage management, with removal rates of less than 50% helping to maintain the soil aggregate stability. Reductions in CO2 and N2O fluxes typically occurred following complete residue removal. The use of wheat straw typically increased CH4 emissions, and above or equal to 8Mg/ha wheat straw led to the largest CO2 and N2O emissions, regardless of N rates. Before using crop residues for biofuel production, it should therefore always be checked whether neutral to positive sustainability effects can be maintained under the site-specific conditions. © 2020 The Authors. The relationship between diversity and productivity (DPR) is a hot topic in ecology, whereas most evidence comes from studies of manipulated terrestrial ecosystems or from two-dimensional space views. In this study, we determined DPR of submerged plants in a natural mesotrophic lake, Lake Erhai, along its water depth in 2013 and 2019. Our results demonstrated that the community productivity was significantly negatively correlated with plant biodiversity from a three-dimensional space view (all of the water depths) in Lake Erhai due to strong competition effects among submerged species. However, DPR patterns at each water depth (from two-dimensional space views) were various, which might be because these DPR patterns were disturbed by many biotic and abiotic factors or their interplays. Both total biomass and biomass of dominant species negatively correlated with species richness. Moreover, total biomass was positively correlated with biomasses of dominant species. Accordingly, we can presume that DPR patterns are largely determined by relationships between biomasses of dominant species and biodiversity in our study. Our results showed that species number in 2019 was generally smaller than that in 2013 and submerged species were migrating from deep water to shallow water with lake eutrophication in such a short time. Accordingly, measurements should be conducted quickly to control the eutrophication process of Lake Erhai to improve its multiple valuable functions and services. © 2020 Elsevier Ltd

The aim of this present research is to enhance the squalene production in Yarrowia lipolytica using pathway engineering and bioprocess engineering. Firstly, to improve the production of squalene, the endogenous HMG-CoA reductase (HMG1) was overexpressed in Y. lipolytica to yield 208.88 mg/L squalene. Secondly, the HMG1 and diacylglycerol acyltranferase (DGA1) were co-overexpressed, the derived recombinant Y. lipolytica SQ-1 strain produced 439.14 mg/L of squalene. Thirdly, by optimizing the fermentation medium, the improved titer of squalene with 514.34 mg/L was obtained by the engineered strain SQ-1 grown on YPD-80 medium. Finally, by optimizing the addition concentrations of acetate, citrate and terbinafine, the 731.18 mg/L squalene was produced in the engineered strain SQ-1 with the addition of 0.5 mg/L terbinafine. This work describes the highest reported squalene titer in Y. lipolytica to date. This study will provide the foundation for further engineering Y. lipolytica capable of cost-efficiently producing squalene. © 2020

Status of the application of exogenous enzyme technology for the development of natural plant resources

摘要:
Exogenous enzymes are extraneous enzymes that are not intrinsic to the subject. The exogenous enzyme industry has been rapidly developing recently. Successful application of recombinant DNA amplification, high-efficiency expression, and immobilization technology to genetically engineered bacteria provides a rich source of enzymes. Amylase, cellulase, protease, pectinase, glycosidase, tannase, and polyphenol oxidase are among the most widely used such enzymes. Currently, the application of exogenous enzyme technology in the development of natural plant resources mainly focuses on improving the taste and flavor of the product, enriching the active ingredient contents, deriving and transforming the structure of a chosen compound, and enhancing the biological activity and utilization of the functional ingredient. In this review, we discuss the application status of exogenous enzyme technology for the development of natural plant resources using typical natural active ingredients from plant, such as resveratrol, steviosides, catechins, mogrosides, and ginsenosides, as examples, to provide basis for further exploitation and utilization of exogenous enzyme technology.

Two fragments of the COVID-19 genome (specific and homologous) were used as two inputs to construct an AND logic gate for COVID-19 detection based on exonuclease III and DNAzyme. The detection sensitivity of the assay can reach fM levels. Satisfactory recovery values were obtained in real sample analysis.

摘要:
As bisphenol A (BPA) is harmful to human health, it is of great significance to develop a new method for BPA detection. Herein, we designed a BPA biosensor by integrating an amplifying DNA circuit with Mg2+-dependent DNAzyme into the sensing system. The BPA-aptamer binding activated a DNA circuit for signal amplification based on toehold-mediated strand displacement. A catalytic Mg2+-dependent DNAzyme was formed through synergistically DNA hybridization, which can cleave the dual-labeled substrate DNA into two segments. The separation of the fluorophore and quencher produces a high fluorescence response for BPA detection. This biosensor exhibited a superior sensitivity with a detection limit of 50 fM. The method is selective and robust, which can work even in milk samples with satisfactory accuracy. The biosensor analytical results were also verified by liquid chromatography coupled with mass spectrometry (LC–MS) and no obvious difference existed between the two methods. © 2021 Elsevier Ltd

摘要:
The interaction between microorganisms and minerals was a hot topic to reveal the transformation of key elements that affecting bioleaching efficiency. Three typical low-grade copper ores, the main copper-bearing components of which were primary sulfide, secondary sulfide and high-oxidative sulfide copper, were obtained from Dexing, Zijinshan and Luanshya copper mine, respectively. Meanwhile, six typical microorganisms were isolated from each of the three habitats, and assembled as communities based on their origins. Cross bioleaching was carried out under identical conditions. The leaching parameters showed that native strains played excellent roles in their corresponding ore bioleaching process, and community structure was greatly determined by mineral composition, indicating that domestication for longitudinal adaption was an effective way to improve microbial leaching performance. Leptospirillum ferriphilum and Acidithiobacillus ferrooxidans promoted copper release by shifting redox potential and pH of the leachate, respectively, indicating that microbial population regulation was another effective way to improve bioleaching efficiency. © 2020 Elsevier Ltd

Fe(II)/persulfate process has been proven to be a promising technique for disintegrating sludge, while during the Fe(II)/persulfate treatment the direct information about the variation of extracellular polymeric substances (EPS) properties, which is a key factor affecting sludge dewatering, is still lacked. In this work, different dosages of Fe(II)/persulfate were employed to treat EPS fractions extracted from waste activated sludge. The experiment results showed that EPS fractions were modified by Fe(II)/persulfate process. An oxidation/flocculation process was raised to unveil how Fe(II)/persulfate modified EPS fractions: Firstly, SO4[rad]– and [rad]OH were formed by chemical reactions of Fe(II) activating persulfate and radical interconversion, respectively. Then these species nonselectively fragmented EPS fractions through decomposing their components, which facilitated decrement of the hydrophilic components and high/mid molecular weight of organics in EPS fractions. Furthermore, these radicals transformed the secondary structure of EPS proteins by affecting the hydrogen bonds at specific positions and inducing the cleavage of the S–S bonds in cysteine residues of proteins, which led to loose layout of protein molecules and thus increased exposure of the hydrophobic groups hidden in EPS protein molecules. Secondly, Fe(III), i.e., the oxidation product of Fe(II), assembled the ruptured colloids particles through lessening electronegativity. Consequently, Fe(II)/persulfate process elevated the flocculability and hydrophobicity of EPS fractions, which would improve physicochemical and rheological properties of sludge to facilitate its dewaterability. © 2020 Elsevier Ltd

Rice is a main source of dietary cadmium (Cd), thus, how to reduce the Cd concentration in brown rice has received extensive attention worldwide. In three acidic paddy soils slightly to moderately contaminated with Cd, a series of field experiments were conducted to evaluate the effects of different proportions of nitrogen-phosphorus-potassium (N-P-K) fertilizer (urea, calcium magnesium phosphate, and potassium carbonate, respectively) alone or coupled with a topdressing of manganese (Mn) fertilizer at the tillering stage on reducing Cd bioavailability in soil and uptake in rice. The rational application of NP-K fertilizer not only provided the basic nutrients to promote the normal growth of rice but also increased soil pH and thereby reduced the Cd bioavailability in soil. The Mg(NO3)(2)-extracted Cd concentrations in the three soils were reduced by 26.46-56.53%, while TCLP-extracted Cd were reduced by 19.87-45.41%, with little influence on soil cation exchange capacity (CEC) and organic matter (OM). The application of Mn fertilizer at the tillering stage increased Mn and Cd sequestration in the iron plaque. The Mn content in iron plaque increased by 15.71-58.67% and a significant positive correlation between Cd and Mn was observed at the three sites. Collectively, this combined method of fertilization significantly reduced Cd accumulation in rice tissues, the Cd concentrations in roots of treated plants decreased by 11.18-37.78%, whereas the concentrations in straw decreased by 13.16-41.03%. Particularly to brown rice, in which accumulation decreased by 25.19-44.70%, 37.35-47.84%, and 38.00-60.88% in three typical paddy fields, but no significant effect was observed for the Cd translocation factors (TF) among rice tissues. Thus, the basal application of combined urea and alkaline inorganic fertilizers followed by topdressing of Mn fertilizer may be a promising and cost- effective tactics for the remediation of Cdcontaminated paddy soils. 摘要:
Objective: The anti-malarial drug, artemisinin, is harvested from the leaves of adult Artemisia annua L. plants. As its concentration in juvenile plants is very low, the present study aimed to assess if the airborne signaling molecule, β-ocimene, could be used to enhance artemisinin accumulation in juvenile A. annua plants. Results: Application of exogenous β-ocimene increased artemisinin accumulation in A. annua. Treatment with 10 µM β-ocimene for 4 days resulted in juvenile plants accumulating artemisinin contents of up to 25 mg/g (2.5%) of dry weight. The expression levels of key genes encoding enzymes involved in both precursor biosynthetic pathways and artemisinin biosynthetic pathways induced by β-ocimene were upregulated. Glandular secretory trichome (GST) size and density increased by 49.2% and 38.2%, respectively, along with the upregulation of genes associated with GST development. Conclusion: β-ocimene enhances artemisinin accumulation in juvenile A. annua plants by modulating artemisinin biosynthetic pathways and GST development. © 2020, Springer Nature B.V.

Oncolytic therapy is a fast-developing cancer treatment field based on the promising clinical performance from the selective tumor cell killing and induction of systemic antitumor immunity. The virotherapy efficacy, however, is strongly hindered by the limited virus propagation and negative immune regulation in the tumor microenvironments. To enhance the antitumor activity, we developed injectable pH-degradable PVA microgels encapsulated with oncolytic adenovirus (OA) by microfluidics for localized OA delivery and cancer treatments. PVA microgels were tailored with an OA encapsulation efficiency of 68% and exhibited a pH-dependent OA release as the microgel degradation at mildly acidic conditions. PVA microgels mediated fast viral release and increased replication in HEK293T and A549 cells at a lower pH, and the replication efficiency could be further reinforced by co-loading with one BET bromodomain inhibitor JQ1, inducing significant cytotoxicity against A549 cells. An in vivo study revealed that OA release was highly located at the tumor tissue assisted by PVA microgels, and the OA infection was also enhanced with the addition of JQ1 treatment, meanwhile greatly inhibiting the PD-L1 expression to overcome the immune suppression. OA/JQ1 co-encapsulated injectable microgels exhibited a superior in vivo antitumor activity on the A549 lung tumor-bearing mice by the combination of inhibited proliferation, amplified oncolysis, and potential immune regulation. © 2020 The Royal Society of Chemistry.

Highly photoactive Z–scheme of graphene layers anchored TiO2/g–C3N4 (GTOCN) photocatalyst was synthesized from MXene Ti3C2 through one–step in–situ calcination, and used for tetracycline (TC) and ciprofloxacin (CIP), bisphenol A (BPA) and rhodamine B (RhB) degradation under visible light irradiation. The crystal phases, chemical compositions, morphological structures and photoelectrochemical properties were characterized. The results demonstrated that hierarchical structured GTOCN photocatalysts exhibited significant improvement in visible light adsorption and photogenerated carriers separation. The GTOCN3 presented the best visible light photocatalytic synergism of degradation rate towards TC (0.02442 min−1), CIP (0.01675 min−1), BPA (0.01935 min−1) and RhB (0.05586 min−1), which was much higher than that of graphene layers anchored TiO2, g–C3N4 and g–C3N4/TiO2(P25), benefitting from solid interactions among graphene, TiO2 and g–C3N4 for separation and accumulation of g–C3N4 electrons with high reduction capability and TiO2 holes with high oxidation capability. Furthermore, the main species of [rad]O2− and [rad]OH for pollutants decomposition was confirmed by free radical capture and electron spin resonance experiments. Besides, the acceptable reusability and stability indicated the great application potential of GTOCN in pharmaceuticals, endocrine disruptors and colourants contained wastewater disposal and environmental protection. © 2020 Elsevier B.V.

A piscicide, rotenone (RT), is frequently used for clear and management of aquatic systems such as fish pond, and even for illegal fishing throughout the world. The effects of RT on submerged macrophytes remain elusive although the effects of RT on many kinds of animals are well documented. We wanted to determine the effects of RT on the growth and metabolism of three submerged plants (Vallisneria natans, Myriophyllum spicatum, Potamogeton maackianus) and try to find the reasons of these effects. The results showed that the shoot height, shoot dry weight, root dry weight, root:shoot ratios, contents of soluble protein and soluble carbohydrate of the three tested submerged plants were significantly negatively affected by RT and the effects were different among the studied species. Furthermore, pH rised a little and light transmission was greatly reduced in the water with RT treatment. We think that the negative effects of RT on the growth and metabolism of submerged species is partially attributing to the lower light caused by RT application. Accordingly, we highlight that submerged species may be greatly suppressed by RT, and we should apply RT in water ecosystems with great caution. © 2020 Elsevier Ltd

Muscle-related disorders, such as sarcopenia and cachexia, caused by aging and chronic diseases can lead to the loss of muscle mass and strength to different degrees, severely affecting human health. Globally, tea is one of the three most popular beverages, and its major active ingredient catechins have been reported to delay muscular atrophy and enhance movement. However, currently, there is no systematic review to elaborate its roles and the associated mechanisms. This article reviews the (1) functions and mechanisms of catechins in the differentiation of myogenic stem cells, biogenesis of mitochondria, synthesis and degradation of proteins, regulation of glucose level, and metabolism of lipids in muscle cells; and (2) effect of catechins on the blood vessels, bones, and nerves that are closely related to the skeletal muscles. Catechins could prevent, mitigate, delay, and even treat muscle-related disorders caused by aging and diseases. © 2019, © 2019 Taylor & Francis Group, LLC.
