信阳毛尖与黄山毛峰及西湖龙井的香气成分分析

摘要:
以信阳、黄山、杭州的茶树鲜叶为原料,分别加工成信阳毛尖、黄山毛峰、西湖龙井,其中信阳的鲜叶由3个公司加工成3个信阳毛尖茶样,共5个茶样,采用同时蒸馏萃取法(SDE法)提取香气物质,结合感官审评和GC–MS,分析3种名优绿茶的香气特点及成分构成。结果表明:3个信阳毛尖茶样香气均表现为清香,黄山毛峰茶样香气馥郁带兰花香,西湖龙井茶样香气浓郁带高火香;5个茶样共检测出香气成分74种,其中共有香气成分24种,5个茶样中的香气成分均以醇类物质的含量最高;信阳毛尖茶样中含量较高的成分有β–芳樟醇、壬醛、环氧芳樟醇、反香叶醇、δ–杜松烯、反–橙花叔醇、棕榈酸、反式植醇;黄山毛峰茶样含量较高的成分有棕榈酸、β–芳樟醇、反香叶醇、壬醛、己醛、反式植醇、环氧芳樟醇、庚醛,且在黄山毛峰茶样中检测出苯乙醛、脱氢芳樟醇2种独有的令人愉悦的香气成分;西湖龙井茶样含量较高的成分有β–芳樟醇、反香叶醇、环氧芳樟醇、壬醛、顺氧化芳樟醇、己醛、δ–杜松烯、δ–杜松醇,西湖龙井茶样中检测出烷烃类和吡嗪类物质的数量较多,但未检测出棕榈酸。

摘要:
Oxidative stress can induce neuronal apoptosis via the production of superoxide and hydroxyl radicals. This process is as a major pathogenic mechanism in neurodegenerative disorders. In this study, we aimed to clarify whether theaflavins protect PC12 cells from oxidative stress damage induced by H 2 O 2 . A cell model of PC12 cells undergoing oxidative stress was created by exposing cells to 200 μM H 2 O 2 in the presence or absence of varying concentrations of theaflavins (5, 10, and 20 μM). Cell viability was monitored using the MTT assay and Hoechst 33258 staining, showing that 10 μM theaflavins enhanced cell survival following 200 μM H 2 O 2 induced toxicity and increased cell viability by approximately 40 %. Additionally, we measured levels of intracellular reactive oxygen species (ROS) and antioxidant enzyme activity. This suggested that the neuroprotective effect of theaflavins against oxidative stress in PC12 cells is derived from suppression of oxidant enzyme activity. Furthermore, Western blot analyses indicated that theaflavins downregulated the ratio of pro-apoptosis/anti-apoptosis proteins Bax/Bcl-2. Theaflavins also downregulated the expression of caspase-3 compared with a H 2 O 2 -treated group that had not been treated with theaflavins. Interestingly, this is the first study to report that the four main components of theaflavins found in black tea can protect neural cells (PC12) from apoptosis induced by H 2 O 2 . These findings provide the foundations for a new field of using theaflavins or its source, black tea, in the treatment of neurodegenerative diseases caused by oxidative stress. © 2016, Springer Science+Business Media New York.

茶叶是一种可以保健和预防人体疾病的健康食品,具有广泛的生物活性成分。整理归纳了近几年国内外茶叶美白功效的文献,从黑色素的合成过程、美白作用调节机理和茶叶提取物的美白作用三方面进行了综述,提出动物实验、病理学及毒理学等方面研究是未来开发茶叶提取物美白产品的研究方向,展望了茶叶美白产品的前景。

摘要:
目的:研究茶多酚等天然抗氧化剂对香烟烟雾致损的 HEK293细胞的保护效能及 JAK2/STAT3代谢调控机制。方法香烟过滤嘴中添加茶多酚、白芦藜醇、维生素C、芦丁、槲皮素、绿原酸、花青素、原花青素8种植物提取物后收集烟气,对 HEK293细胞进行 MTT 对比研究及运用 Western blot 技术分析其Bcl-2、Bax、STAT3蛋白表达量变化。结果香烟烟雾可降低HEK293细胞存活率,呈浓度依赖关系;上调 HEK293细胞中 Bcl-2蛋白相对表达量,下调 Bax、STAT3蛋白相对表达量,且各蛋白变化趋势呈浓度依赖效应;在过滤嘴中添加茶多酚、白芦藜醇、维生素C、芦丁、槲皮素、绿原酸、花青素、原花青素8种抗氧化剂明显抑制了香烟烟雾导致 HEK293细胞死亡,结果具有显著性差异(P<0.05),其中以茶多酚效果最佳。结论8种植物提取物均能对香烟所致HEK293细胞损伤产生一定的保护作用。

摘要:
Tea (Camellia sinensis L.) is a perennial woody plant that is widely cultivated to produce a popular non-alcoholic beverage; this beverage has received much attention due to its pleasant flavor and bioactive ingredients, particularly several important secondary metabolites. Due to the significant changes in the metabolite contents of the buds and the young expanding leaves of tea plants, high-performance liquid chromatography (HPLC) analysis and isobaric tags for relative and absolute quantitation (iTRAQ) analysis were performed. A total of 233 differentially expressed proteins were identified. Among these, 116 proteins were up-regulated and 117 proteins were down-regulated in the young expanding leaves compared with the buds. A large array of diverse functions was revealed, including roles in energy and carbohydrate metabolism, secondary metabolite metabolism, nucleic acid and protein metabolism, and photosynthesis- and defense-related processes. These results suggest that polyphenol biosynthesis- and photosynthesis-related proteins regulate the secondary metabolite content of tea plants. The energy and antioxidant metabolism-related proteins may promote tea leaf development. However, reverse transcription quantitative real-time PCR (RT-qPCR) showed that the protein expression levels were not well correlated with the gene expression levels. These findings improve our understanding of the molecular mechanism of the changes in the metabolite content of the buds and the young expanding leaves of tea plants.

摘要:
The Si(0001) face and C(000-1) face dependences on growth pressure of epitaxial graphene (EG) grown on 4H-SiC substrates by ethene chemical vapor deposition (CVD) was studied using atomic force microscopy (AFM) and micro-Raman spectroscopy (μ-Raman). AFM revealed that EGs on Si-faced substrates had clear stepped morphologies due to surface step bunching. However, This EG formation did not occur on C-faced substrates. It was shown by μ-Raman that the properties of EG on both polar faces were different. EGs on Si-faced substrates were relatively thinner and more uniform than on C-faced substrates at low growth pressure. On the other hand, D band related defects always appeared in EGs on Si-faced substrates, but they did not appear in EG on C-faced substrate at an appropriate growth pressure. This was due to the μ-Raman covering the step edges when measurements were performed on Si-faced substrates. The results of this study are useful for optimized growth of EG on polar surfaces of SiC substrates. ©2015 by the authors.

摘要:
The AnJiBaiCha albino mutant tea cultivar has a reversible albino phenotype at low temperatures. Albino AnJiBaiCha leaves contain high levels of amino acids, which are important components affecting the quality of tea as a beverage. To examine the molecular mechanism of albinism and amino acid enrichment in AnJiBaiCha, we used the amplified fragment length polymorphism (cDNA-AFLP) technique to isolate genes that are differentially expressed during periodic albinism in AnJiBaiCha. A total of 127 transcript-derived fragments (TDFs) were successfully sequenced; among those, 60 TDFs showed high similarity to sequences with known functions, but 67 TDFs were not similar to any known genes. The identified transcripts include transcription factors, ubiquitination-related genes, chloroplast biogenesis genes, signal transduction genes, stress-related genes, cell cycle genes, and carbohydrate and energy metabolism genes. To validate the cDNA-AFLP results, quantitative real-time PCR was used to confirm the differential expression of six of the identified genes. The cDNA-AFLP and quantitative real-time PCR results correlated well, indicating that the cDNA-AFLP results are reliable. This study provides insights into the molecular mechanisms by which periodic albinism and amino acid accumulation take place in AnJiBaiCha. © 2015 Elsevier B.V.

摘要:
目的:以电子鼻技术对不同加工工艺的工夫红茶进行香气归类并对各评价方法优劣进行探讨。方法以碧香早为原材料,对工夫红茶传统加工工艺进行研制改进,将萎凋叶进行紫外光照射、做青实验处理,发酵叶添加外源茶汁发酵,对所获得的各实验处理冷冻样及成品茶进行电子鼻分析。结果电子鼻6种传感器中, P40/1、T70/2、PA/2、P30/1等4种传感器对茶叶香气测定的贡献率较大;判别因子分析(DFA)比主成分分析(PCA)更能有效的区分不同工夫红茶样品。结论选用电子鼻不同传感器及相应的统计分析方法,可以对工夫红茶加工不同阶段的香气进行归类,并加以区分。

目的探索湖南农业大学长安教学基地湘妃翠、碧香早和茗丰3个品种制成的黑毛茶经冠突散囊菌发花后其呈味品质成分的差异变化,对黑毛茶进行冠突散囊菌发花优化实验。方法采用国标检测方法对发花前黑毛茶及发花后成品茶其水浸出物、茶多酚、游离氨基酸、可溶性糖等呈味成分进行分析,对所测数据进行组间和组内方差分析。结果湘妃翠经发花后其水浸出物、茶多酚、游离氨基酸和生物碱在发花成品茶中下降幅度最大;碧香早经发花后其成品茶中茶多酚、游离氨基酸与其原料茶相比显著下降;茗丰经发花后其茶多酚、氨基酸和生物碱与其原料茶相比显著下降,而可溶性多糖显著增加。结论在三个发花成品茶中,以湘妃翠化学品质表现最为突出,湘妃翠较碧香早、茗丰更适制发花黑茶,其次为茗丰。

摘要:
In order to investigate the physiological functions and biosynthesis regulation of borneol or comphore in Artermisia annua L., which is the major source of the anti-malaria drug artemisinin, the full length cDNA of the gene encoding a borneol dehydrogenase (AaBDH) was cloned from A. annua for the first time by using RACE (rapid amplification of cDNA ends). The completed open read frame of AaBDH was 1415 bp and it encoded a 885-amino acid protein with a predicted molecular mass of 31.04 kDa and a pI of 6.16. AaBDH showed 68-70% of amino acid identity to alcohol dehydrogenases from Solanum lycopersicum, Ppulus trichocarpa, Morus notabili and Ricinus communis. While it shared 51% and 58% of identity with artemisia alcohol dehydrogenase ADH2 from A. annua and borneol dedrogenase LiBDH from Lavandula x intermedia, respectively. The recombinant protein was obtained by heterogeneous expression of AaBDH in a strain of Escherichia coli BL 21 and purified by affinity chromatography. The function of AaBDH was characterized by using of in vitro enzymatic assays, and the results showed that AaBDH had the ability to specifically convert borneol into camphor in the presence of NAD(+) (nicotinamide adenine dinucleotide) or NADP+ (nicotinamide adenine dinucleotide phosphate). The cloning of AaBDH laid a significant foundation for further investigation on physiological functions and biosynthesis regulation of plant monoterpenoids. (C) 2015 Elsevier B.V. 溶剂法提取藕节中鞣质的工艺优化

摘要:
在利用磷钼钨酸/干酪素法测定藕节中鞣质含量的基础上,通过单因素试验和正交试验从料液比、乙醇浓度、提取温度、提取时间、提取次数等方面对藕节中鞣质类物质的乙醇提取工艺进行了优化,建立了藕节中鞣质的溶剂快速萃取方法。结果表明,在料液比1︰10、乙醇浓度80%、提取温度50℃、提取时间4.0 h的条件下,藕节中鞣质的得率平均可达1.294 8%,最高得率可达1.390 4%。该工艺操作简单方便,成本较低,适合工业化生产,为藕节中鞣质类化合物的进一步开发利用奠定了基础。

摘要:
Despite the biological importance of membrane proteins, their analysis has lagged behind that of soluble proteins and still presents a great challenge mainly because of their highly hydrophobic nature and low abundance. Sodium deoxycholate (SDC)-assisted digestion strategy has been introduced in our previous papers, which cleverly circumvents many of the challenges in shotgun membrane proteomics. However, it is associated with significant sample loss due to the slightly weaker extraction/solubilization ability of 1% SDC. In this study, an enhanced SDC-assisted digestion method (ESDC method) was developed that incorporates the almost strongest ability of SDC with a high concentration (5%) to lyse membrane and extract/solubilize hydrophobic membrane proteins, and then dilution to 1% for more efficient digestion. The comparative study using rat liver membrane-enriched sample showed that, compared with previous SDC-assisted method and the "universal" filter-aided sample preparation (FASP) method, the ESDC method not only increased the identified number of total proteins, membrane proteins, hydrophobic proteins, integral membrane proteins (IMPs) and IMPs with more than 5 transmembrane domains (TMDs) by an average of 10.8%, 13.2%, 17.8%, 17.9% and 52.9%, respectively, but also enhanced the identified number of total peptides and hydrophobic peptides by averagely 12.5% and 14.2%. These results demonstrated that the ESDC method provides a substantial improvement in the recovery and identification of membrane proteins, especially those with high hydrophobicity and multiple TMDs, and thereby displaying more potential for shotgun membrane proteomics. Fuzhuan brick tea has received increasing attention in recent years owing to its benefits for non-alcoholic fatty liver disease (NAFLD) and associated metabolic syndrome. For exploring the ameliorative mechanism, the liver proteomes from three groups of rats fed either a normal control diet (NCD), a high fat diet (HFD), or a HFD supplemented with high-dose FTE (HFD+HFTE) were comprehensively compared by quantitative proteomics using 2DE-LC-MS/MS. This is the first study of the effects of tea aqueous extract on the liver proteome of rats suffering from metabolic syndrome. The results showed that 57 proteins displayed more than 1.5-fold differences in at least one of two comparisons of HFD versus NCD and HFD versus HFD+HFTE due to HFD feeding and FTE treatment, respectively. Of them, over 75% of proteins exhibited a similar tendency of expression in the two comparisons, meaning FTE was able to correct HFD effects on rat livers. By function analyses, an extensive list of proteins were involved in sugar and lipid metabolism. Compared with HFD-fed rats, the reduced lipogenesis and enhanced β-oxidation, tricarboxylic acid cycle and respiratory chain in HFD+HFTE-fed rats, which mainly contributed to ameliorate hepatic fat accumulation and associated NAFLD. 红茶对高脂饮食小鼠血脂的调节作用研究

摘要:
采用脂代谢紊乱模型法—预防性给予不同地区不同剂量的红茶(剂量为成人每日饮用量的5倍、10倍和20倍 3 种),观察小鼠体重、血脂、相关酶及肝脏形态的变化。结果表明:与高脂模型对照组相比,红茶剂量组能极显著(P<0.01)降低血清总胆固醇(TC)、低密度脂蛋白(LDL-C)、肝脏丙二醛(MDA)、肝脏指数;极显著(P<0.01)升高血清载脂蛋白 A1(apoA1)、脂蛋白脂酶(LPL)、肝脂酶(HL)、总脂酶、肝脏超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GPX),血清高密度脂蛋白(HDL-C)有所上升,小鼠体重也得到了一定的抑制,肝脏病变程度得到了相应的减轻,说明红茶具有一定的调节血脂及减小肝脏因高脂饮食所带来的损伤。

摘要:
选取长势、色泽差异较小的9个茶树品种,使用ASD FieldSpec HandHeld 2光谱分析仪采集茶树冠层的光谱数据,通过对原始数据进行预处理去除噪音干扰,采用主成分分析方法得到茶树冠层特征波段520、765、821、940 nm,提取特征波段的反射率值,运用多元线性回归、一元线性回归、最小二乘法建立了光谱反射率与茶鲜叶茶多酚含量关系的估算模型。结果表明:最小二乘法模型的决定系数达到0.99;另选23个样品对模型进行验证,真实值与预测值的相关系数为0.97,相对误差为2.99%。

摘要:
Functional porous carbon (PC) derived from bio-friendly shaddock peel has been firstly explored as catalyst for vanadium redox flow battery (VRB). The prepared PC is micro-mesoporous with high BET surface area of 882.7 m(2) g(-1), has some surface oxygen-containing functional groups, and is doped with N and P heteroatoms. These three factors greatly favor the electrochemical reactions of VO2+/VO2+ on the PC modified glass carbon (PC-GC). Compared with the naked GC and graphite modified GC, the PC-GC presents a lower peak separation (66 mV), higher anodic current density (17.1 mA cm(-2)) and cathodic current density (15.0 mA cm(-2)). The VRB using PC modified graphite felt (GF) as positive electrode demonstrates an enhanced voltage efficiency of 82.7% at the current density of 60 mA cm(-2), and a better rate performance than that from the virginal GF. 摘要:
采集试验基地鲜茶叶的近红外光谱数据,结合茶叶中的氨基酸、茶多酚含量的差异所呈现出来的不同光谱特性,应用近红外光谱技术在线对鲜茶叶的品质进行检测,并利用多元线性回归、最小二乘法建立了茶多酚和氨基酸的校正模型。结果表明:用最小二乘法检测精度更高,氨基酸和茶多酚测量模型真实值与预测值的决定系数R~2分别为0.92与0.94,相对误差平均值为1.60%与2.99%。

通过PCR方法克隆丰水梨PPO基因全长, 并将其登录Genebank, 登录号JQ861265。基因全长1782 bp, 无内含子, 编码的PPO属于亲水性蛋白质, 无跨膜结构, 含有593个氨基酸, 分子量约为65.8 kDa, 理论等电点为8.4。N端含有一段由47个氨基酸组成的转运肽。去除转运肽的成熟PPO分子量为60.8 kDa, 理论等电点为6.69。PPO中含有两个铜离子结合区, 主要位于PPO二级结构中的α-螺旋区域中。原核诱导目的蛋白在诱导3~6 h后积累量较大。诱导蛋白(PPO前体和成熟PPO)均能氧化儿茶素形成茶黄素。

茶树作为一种多年生常绿叶用木本植物,有着特殊的饮用及经济价值。对植物蛋白质组学的基本概念和茶树蛋白质组学的构建及其在亚细胞水平、逆境胁迫和突变体等方面的研究进展进行了综述,提出简单高效的蛋白样品制备、质谱技术鉴定及完整基因组的构建等是今后茶树蛋白质组学的研究方向。而"紫鹃"和江华苦茶等是研究茶树生育、进化规律和代谢调控的理想茶树品种。
