Epigallocatechin-3-gallate promotes healthy lifespan through mitohormesis during early-to-mid adulthood in Caenorhabditis elegans

摘要:
The green tea polyphenol epigallocatechin-3-gallate (EGCG) is widely consumed as a dietary supplement. Its potential properties include slowing aging and extending lifespan, although how exactly this is achieved remains unclear. Here, we report that EGCG promoted healthy lifespan in Caenorhabditis elegans when administered throughout or only at early-to-mid adulthood. Specifically, EGCG extended lifespan in an inverted U-shaped dose-response manner. The life-extending mechanism was stimulated by EGCG-induced production of reactive oxygen species (ROS). Additionally, EGCG triggered mitochondrial biogenesis to restore mitochondrial function. The EGCG-induced increase in lifespan depends on known energy sensors such as AMPK/AAK-2, as well as SIRT1/SIR-2.1 and FOXO/DAF-16. Interestingly, aging decreased the response to EGCG and progressively neutralized its beneficial effects on longevity. Collectively, our findings link EGCG to the process of mitohormesis and suggest an inducible, AMPK/SIRT1/FOXO-dependent redox signaling module that could be invoked in different contexts to extend healthy lifespan. Its effectiveness is higher in younger adults and declines with age.

摘要:
对不同年份的27个普洱生茶样品按照国家审评标准从外形、内质对其进行感官评价,并对其汤色的色差值进行测定。结果表明,随着陈化年份的延长,普洱生茶茶饼不同程度地出现疏松,茶叶条索逐渐松散,干茶色泽呈黄绿—褐黄—黄褐—棕褐的变化趋势;茶汤色泽逐渐变深,呈橙黄—黄橙—橙红—棕红的变化趋势;香气中的花香逐渐淡化,由带陈到陈香显露;滋味中的收敛性和粗涩感逐步减弱;色差值a-*值、b-*值增大,茶汤颜色变红变深。普洱生茶感官品质随储藏年份的增加发生一定趋势的变化,但变化的程度及方向受到原料、加工工艺、贮藏环境等影响。试验通过对跨度近20年的不同厂家普洱生茶感官品质进行探讨,为进一步了解普洱茶的陈化过程中品质变化规律提供参考。

摘要:
选取茶叶中三种主要生物碱咖啡碱(Caffeine)、可可碱(Theobromine)、茶碱(Theophylline)作为构建单体-冠突散囊菌共培养液态发酵体系(10 d)的添加底物,考察了冠突散囊菌对茶叶中主要生物碱的发酵特性。结果表明,与对照组相比,咖啡碱(0.2mg/mL)的添加可刺激冠突散囊菌的生长与繁殖,发酵液中葡萄糖的含量下降更为迅速,pH值随着发酵时间的增加持续下降直至第9 d趋于稳定,可溶性蛋白含量呈现先增加后下降的趋势,但在发酵过程中,冠突散囊菌既不能将其作为碳源、氮源消耗来维持生长,也不能代谢分解咖啡碱;可可碱与茶碱(0.08 mg/mL)在发酵体系中的发酵情况与咖啡碱基本相似,两者均能刺激冠突散囊菌的生长与繁殖,但均不能将其作为碳、氮源消耗,且发酵结束后在各自发酵体系中分别检测到0.00091 mg/mL、0.00109 mg/mL的咖啡碱,这说明冠突散囊菌可能具有以可可碱、茶碱作为前体合成咖啡碱的能力。

摘要:
研究铁观音茶提取物对脂多糖(LPS)诱导的RAW264.7细胞炎症反应的抑制作用及机制。用脂多糖作用于RAW264.7细胞,建立炎症模型,并用吲哚美辛和不同浓度铁观音提取物处理,检测NO和IL-6的分泌情况,qPCR检测一氧化氮合酶(iNOS) 、环氧合酶2(COX-2) 、肿瘤坏死因子α(TNF-α) 、单核细胞趋化蛋白1(MCP-1) 、白细胞介素6(IL-6) mRNA相对表达,Western Blot检测炎症相关蛋白激酶(IKKβ),核转录因子κB抑制因子(IκB) 、核转录因子κB p65(NF κB p65)及其磷酸化产物的相对表达。结果显示,铁观音茶提取物能显著抑制炎症介质NO分泌和IL-6蛋白表达量(p < 0.05),抑制炎症相关基因iNOS、COX-2、TNF-α和MCP-1等表达,并极显著抑制NF-κB信号通路相关蛋白IKKβ、IkB和p65的磷酸化(p < 0.01) 。以上结果表明,铁观音茶提取物可明显抑制LPS诱导的RAW264.7细胞炎症反应,其机制可能与抑制NF-κB信号通路激活有关。

Dark tea has a significant effect on the prevention and treatment of age-related degenerative diseases. At present, further exploration of its functional mechanisms is delayed because of the complexity of post-fermentation microbial metabolites during the production phase. In this study, new isolated microbial metabolites extracted from dark tea were used to explore the neuroprotective effects, and they also helped allow further exploration of the mechanism of dark tea. Taking senescence-accelerated mouse prone 8 (SAMP8) mice as a biological model, we examined the protective effect on brain neurons of post-fermentation microbial metabolites which were extracted from dark tea. The 4-month-old mice were given treatments of the same concentration (10 mg kg−1 d−1) which were l-theanine, 3,3′-azanediylbis(4-hydroxybenzoic acid) (CDT-1) and one of the 8-C N-ethyl-2-pyrrolidinone substituted flavan-3-ols (CDT-2) by gavage for 14 weeks. Relative measurements such as RT-PCR, ELISA, western blotting, and section staining (HE, Nissl and myelin) were carried out. The results showed that l-theanine, CDT-1 and CDT-2 could inhibit the decrease in body weight, and down-regulate the formation of 4-HNE and ubiquitinated protein aggregates and the Aβ metabolic pathway. They could also increase endogenous antioxidant capacity, relieve cell hypoxia, and reduce the rate of neuronal apoptosis. This means that their protective activity regarding SAMP8 neurons was excellent and the activity of CDT-2 was the most significant. © The Royal Society of Chemistry.

Here we identified that BTE (black tea extract), within the studied concentration range, is more effective than GTE (green tea extract) in protecting C. elegans against hypertonic stress, by enhancing survival after exposure to various salts, and alleviating suffered motility loss and body shrinkage. The mechanism of such protection may be due to the ability of black tea to induce the conserved WNK/GCK signaling pathway and down-regulation of the expression levels of nlp-29. Intriguingly, black tea does not relieve hypertonicity-induced protein damage. The findings implicate the potential health benefits of black tea consumed worldwide. © The Royal Society of Chemistry.

摘要:
Fuzhuan brick tea (FBT) possesses various health-promoting functions. However, the available information regarding biological activity of polysaccharides from FBT (FBTPS) is still limited. In this work, the chemical property, cytotoxicity and antioxidant activity in vitro and in vivo of FBTPS were evaluated. It was found that FBTPSs were typical acidic heteropolysaccharides mainly composed of Man, Rha, GalA, Glc, Gal and Ara with little molar content of Rib and GIcA. FBTPS showed little toxicity to human hepatic epithelial (L-02) cell. FBTPS exhibited antioxidant activities, including limited scavenging activity on DPPH free radicals (ranged from 543 +/- 1.9 to 67.8 +/- 2.5%), noticeable scavenging activity on superoxide radicals (over 85%), superior scavenging activity on ABTS radicals (near 100%), and protective effect on H2O2-induced oxidative injury in rat pheochromocytoma line 12 (PC12) cell. Moreover, FBTPS showed significant amelioration of high-fat diet-induced oxidative injury in mice. The results suggest that FBTPS, as natural safe antioxidants, may have potential application in functional foods. (C) 2018 Elsevier B.V. All rights reserved.

Ordered graphene ribbons were grown on the surface of 4 ◦ off-axis 4H-SiC wafers by sublimation epitaxy, and characterized by using scanning electron microscopy (SEM), atomic force microscopy (AFM) and micro-Raman spectroscopy (µ-Raman). SEM showed that there were gray and dark ribbons on the substrate surface, and AFM further revealed that these ordered graphene ribbons had clear stepped morphologies due to surface step-bunching. It was shown by µ-Raman that the numbers of graphene layers of these two types of regions were different. The gray region was composed of mono-or bilayer ordered graphene ribbon, while the dark region was of tri-or few-layer ribbon. Meanwhile, ribbons were all homogeneous and had a width up to 40 µm and a length up to 1000 µm, without micro defects such as grain boundaries, ridges, or mono-and few-layer graphene mixtures. The results of this study are useful for optimized growth of high-quality graphene film on silicon carbide crystal. © 2018 by the authors. Licensee MDPI, Basel, Switzerland.

黄花蒿中含有大量的香气类物质,在食品和医药行业有广泛的应用前景,但对其生物合成的分子机理研究非常欠缺。本研究采用SMART-RACE-PCR技术从黄花蒿花组织差异文库中获得了1个新的双键还原酶编码基因AaDBR3,其cDNA序列全长为1 279 bp,开放阅读框(ORF)长度为1 038 bp编码345氨基酸,蛋白分子量为38.46 kD,等电点(pI)为6.05。通过生物信息学的分析,AaDBR3属于中链脱氢酶超级家族中leukotriene B4 dehydrogenases类酶。系统发育进化树结果表明,该基因与烟草的烯丙醇脱氢酶(NtADH, GenBank: BAA89423.1)的同源性最高,相似性为77%。体外酶研究发现,重组蛋白酶能催化3-壬烯-2-酮向2-壬酮的转化,表明其属于双键还原酶。本研究为进一步丰富黄花蒿优异基因资源,及黄花蒿的品种改良提供了重要的靶基因。

以大红袍、红茶与茯砖茶原料为供试原材料,利用超高效液相色谱电喷雾四级杆飞行时间串联质谱(UPLC-ESI-Q-TOF-MS)技术的代谢组学对不同茶叶原料茯茶的“发花”前后样品的整体成分组成差异进行了分析.结果表明,不同原料“发花”前、后间差异成分数量均有不同,各组差异代谢化合物个数分别为大红袍49个、红茶40个、茯砖茶原料49个;综合3组茶叶的具体差异代谢化合物,发现其从类别上表现出相似的变化规律,即酯型儿茶素下降的极为显著,而简单儿茶素除没食子儿茶素(GC)变化无明显差异外,表儿茶素(EC)、儿茶素(DL-C)、表没食子儿茶素(EGC)的含量较原料都有所升高,原花青素和咖啡碱、可可碱、茶碱、黄嘌呤与次黄嘌呤也有相同的变化;黄酮类化合物槲皮素、山奈酚、牡荆素和芦丁的含量均有不同程度的升高;而大黄素则是在各原料“发花”后均新增的蒽醌类化合物.

摘要:
In order to make full use of artemisinin production waste and thus to reduce the production cost of artemisinin, we developed an efficient and scalable method to isolate high-purity dihydroartemisinic acid from artemisinin production waste by combining anion-exchange resin with silica-gel column chromatography. The adsorption and desorption characteristics of dihydroartemisinic acid on 10 types of anion-exchange resin were investigated, and the results showed that the 717 anion-exchange resin exhibited the highest capacity of adsorption and desorption to dihydroartemisinic acid. Adsorption isotherms were established for the 717 anion-exchange resin and they fitted well with both Langmuir and Freundlich model. Dynamic adsorption and desorption properties of 717 anion-exchange resin were characterized to optimize the chromatographic conditions. Subsequently, the silica-gel column chromatography was performed and dihydroartemisinic acid with a purity of up to 98% (w/w) was obtained. Finally, the scale-up experiments validated the preparative separation of high-purity dihydroartemisinic acid from industrial waste developed in the present work. This work presented for the first time an isolation of dihydroartemisinic acid with a purity of 98% from Artemisia annua (A. annua) by-product, which adds more value to this crop and has the potential to lower the prices of anti-malarial drugs. © 2018 The Pharmaceutical Society of Japan.

代谢组学是系统生物学研究的重要组成部分,研究生物体在不同条件下所产生的小分子代谢产物的变化,从而揭示生物体的生理状态.近年来代谢组学技术在茶叶相关研究领域得到广泛应用,着重论述了代谢组学在茶叶品质(产地环境、采摘时间、加工工艺对品质的影响以及品质评价等)与药理(茶叶的降脂减肥、延缓衰老、抗辐射、生物利用度与生物转化等)等领域的应用现状,代谢组学技术将在茶叶品质形成及评价、茶叶功效等方面发挥不可替代的作用.

摘要:
Tea O-methylated catechins, especially (-)-epigallocatechin 3-O-(3-O-methyl)gallate (EGCG3″Me), have been attracting much attention as a result of their positive health effects. The transcription regulators of O-methylated catechin biosynthesis remain elusive. In this study, the expression pattern of genes related to O-methylated catechin biosynthesis, including CsLAR, CsANS, CsDFR, CsANR, and CCoAOMT, in three tea cultivars with different contents of EGCG3″Me was investigated. Two WRKY transcription factors (TFs), designated as CsWRKY31 and CsWRKY48, belonging to groups IIb and IIc of the WRKY family, respectively, were further identified. CsWRKY31 and CsWRKY48 were nuclear-localized proteins and possessed transcriptional repression ability. Furthermore, expression of CsWRKY31 and CsWRKY48 showed negative correlation with CsLAR, CsDFR, and CCoAOMT during EGCG3″Me accumulation in tea leaves. More importantly, W-box (C/T)TGAC(T/C) elements were located in the promoter of CsLAR, CsDFR, and CCoAOMT, and further assays revealed that CsWRKY31 and CsWRKY48 were capable of repressing the transcription of CsLAR, CsDFR, and CCoAOMT via the attachment of their promoters to the W-box elements. Collectively, our findings identify two novel negative regulators of O-methylated catechin biosynthesis in tea plants, which might provide a potential strategy to breed high-quality tea cultivar. IntroductionMedicinal plants are gaining increasing attention worldwide due to their empirical therapeutic efficacy and being a huge natural compound pool for new drug discovery and development. The efficacy, safety and quality of medicinal plants are the main concerns, which are highly dependent on the comprehensive analysis of chemical components in the medicinal plants. With the advances in mass spectrometry (MS) techniques, comprehensive analysis and fast identification of complex phytochemical components have become feasible, and may meet the needs, for the analysis of medicinal plants. ObjectiveOur aim is to provide an overview on the latest developments in MS and its hyphenated technique and their applications for the comprehensive analysis of medicinal plants. MethodologyApplication of various MS and its hyphenated techniques for the analysis of medicinal plants, including but not limited to one-dimensional chromatography, multiple-dimensional chromatography coupled to MS, ambient ionisation MS, and mass spectral database, have been reviewed and compared in this work. ResultsRecent advancs in MS and its hyphenated techniques have made MS one of the most powerful tools for the analysis of complex extracts from medicinal plants due to its excellent separation and identification ability, high sensitivity and resolution, and wide detection dynamic range. ConclusionTo achieve high-throughput or multi-dimensional analysis of medicinal plants, the state-of-the-art MS and its hyphenated techniques have played, and will continue to play a great role in being the major platform for their further research in order to obtain insight into both their empirical therapeutic efficacy and quality control. The efficacy, safety and quality of medicinal plants are highly dependent on the comprehensive analysis of their chemical components; The latest developments in MS and its hyphenated technique and their applications for the comprehensive analysis of medicinal plants have been reviewed; To achieve high-throughput or multi-dimensional analysis of medicinal plants, MS and its hyphenated techniques have played, and will continue to play a great role.

摘要:
Chemical vapour deposited graphene on Cu foil should be transferred to target dielectric substrate before being used in various applications. This route often involves preparation of a supporting material on graphene before the transfer process and removal of the material after graphene is transferred to a foreign substrate. Herein, we demonstrate a simple process combining graphene transfer and subsequent graphene-based sensor preparation in one route. By applying a thin flexible polyethylene (PE) film onto graphene and subsequently chemical etching Cu foil, this process enables the high-quality transfer of graphene on PE film. A flexible strain sensor prepared from the as-transferred graphene/PE film shows good detection sensitivity. Therefore, this process could greatly reduce the cost of transfer and device fabrication for mass production.

Determination of dihydroartemisinic acid in Artemisia annua L. by gas chromatography with flame ionization detection

摘要:
Dihydroartemisinic acid (DHAA) is the direct precursor to artemisinin, an effective anti-malaria compound from Artemisia annua L. (A. annua), and it can be transformed to artemisinin without the catalysis of enzyme. A rapid and sensitive analysis of DHAA in A. annua is needed to screen excellent plant resources aimed to improve artemisinin production. In order to develop a rapid and sensitive determination method for DHAA in plant, the extraction and analysis conditions were extensively investigated in the present work. As a result, extraction of powdered A. annua leaves at 55°C for 50 min with chloroform resulted in the highest yield of DHAA, with a recovery of >98%. The precision of this gas chromatographic procedure ranged from 1.22 to 2.94% for intra-day and from 1.69 to 4.31% for inter-day, respectively. The accuracy was 99.55–103.02% for intra-day and 98.86–99.98% for inter-day, respectively. The measured LOQ and LOD values of the proposed method reached 5.00 and 2.00 μg/mL, respectively. Validation indicated the method was robust, quick, sensitive and adequate for DHAA analysis. Copyright © 2016 John Wiley & Sons, Ltd.

代谢组学是系统生物学的一个重要分支,它同时检测生物体系大量的内源性代谢物,从代谢水平上研究其变化差异,对进一步了解代谢网络和新功能基因的发掘具有重要作用,已被广泛应用于多个研究领域。目前,在茶树(Camellia sinensis)上代谢组学主要用于研究原料采摘部位、发育阶段、季节和环境因子、产地、加工等因素对茶叶化学成分和品质的影响。本文综述了代谢组学在茶树研究中的应用进展并作出了展望,为其进一步应用提供参考。
